Objective To explore the effects of harvesting periods on nutritional quality, processing properties and antioxidant activities of different parts of Fagopyrum tataricum. Methods The harvesting period was determined based on the first flowering time, and the flavonoid content, protein content, dietary fiber content, water holding capacity, oil holding capacity and antioxidant activity of seeds, stems, flowers and leaves of Fagopyrum tataricum at different harvest periods were determined measured and analyzed. Results The seeds, stems, leaves, and flowers of Fagopyrum tataricum all include the rich nutrients and the biological activity ingredient, with excellent processing properties. The flavonoid content, protein content, dietary fiber content, water and oil holding capacity of Fagopyrum tataricum seeds showed a downward trend, while the dietary fiber content of Fagopyrum tataricum stems and flowers generally showed an upward trend. The flavonoid and protein content of Fagopyrum tataricum stems and leaves decreased. The dietary fiber content of Fagopyrum tataricum leaves, flavonoid content and total antioxidant activity of Fagopyrum tataricum flowers increased first and then decreased. The total antioxidant capacity and flavonoid content of Fagopyrum tataricum flowers were significantly higher than other parts of tartary buckwheat. The highest total antioxidant capacity and flavonoid content were found 14 days after flowering, at 369.42 μmol/g and 7.83%, respectively, which were 6.2 and 8.8 times that of Fagopyrum tataricum seeds. Conclusion As food processing raw materials, different parts of Fagopyrum tataricum have the potential and value for food development and processing, and can be fully utilized. The nutrition and functional components of different parts of Fagopyrum tataricum are directly affected by harvest period, and the reasonable harvest period shall be determined based on the purpose in production.

目的 研究采收期对苦荞不同部位营养品质、加工特性与抗氧化活性影响。方法 采收期根据首次开花时间来确定, 对苦荞籽粒、茎、花、叶在花后不同采收期的总黄酮含量、蛋白含量、膳食纤维含量、持水性、持油性以及抗氧化活性等进行测定分析。结果 苦荞籽粒、茎、叶与花均含有丰富的营养物质与活性成分, 加工特性优良。随着采收期的延长, 苦荞籽粒的总黄酮含量、蛋白含量、膳食纤维含量、持水性和持油性总体呈下降趋势, 苦荞茎、花的膳食纤维含量总体呈上升趋势, 苦荞茎、叶的总黄酮含量、蛋白含量减少, 苦荞叶的膳食纤维含量、苦荞花总黄酮含量、总抗氧化活性则先增加后减少。苦荞花总抗氧化能力要显著高于苦荞的其他部位, 花后14 d总抗氧化能力与黄酮含量最高, 分别为369.42 μmol/g与7.83%, 是苦荞籽粒的6.2倍和8.8倍。结论 不同部位的苦荞都有作为食品开发加工原料的潜能与应有价值, 可充分利用; 采收期对苦荞不同部位的营养与功能成分有直接影响, 在实际生产应用中, 应根据目的来确定合理的采收期。

目的 研究采收期对苦荞不同部位营养品质、加工特性与抗氧化活性影响。方法 采收期根据首次开花时间来确定, 对苦荞籽粒、茎、花、叶在花后不同采收期的总黄酮含量、蛋白含量、膳食纤维含量、持水性、持油性以及抗氧化活性等进行测定分析。结果 苦荞籽粒、茎、叶与花均含有丰富的营养物质与活性成分, 加工特性优良。随着采收期的延长, 苦荞籽粒的总黄酮含量、蛋白含量、膳食纤维含量、持水性和持油性总体呈下降趋势, 苦荞茎、花的膳食纤维含量总体呈上升趋势, 苦荞茎、叶的总黄酮含量、蛋白含量减少, 苦荞叶的膳食纤维含量、苦荞花总黄酮含量、总抗氧化活性则先增加后减少。苦荞花总抗氧化能力要显著高于苦荞的其他部位, 花后14 d总抗氧化能力与黄酮含量最高, 分别为369.42 μmol/g与7.83%, 是苦荞籽粒的6.2倍和8.8倍。结论 不同部位的苦荞都有作为食品开发加工原料的潜能与应有价值, 可充分利用; 采收期对苦荞不同部位的营养与功能成分有直接影响, 在实际生产应用中, 应根据目的来确定合理的采收期。

Objective To explore the effects of harvesting periods on nutritional quality, processing properties and antioxidant activities of different parts of Fagopyrum tataricum. Methods The harvesting period was determined based on the first flowering time, and the flavonoid content, protein content, dietary fiber content, water holding capacity, oil holding capacity and antioxidant activity of seeds, stems, flowers and leaves of Fagopyrum tataricum at different harvest periods were determined measured and analyzed. Results The seeds, stems, leaves, and flowers of Fagopyrum tataricum all include the rich nutrients and the biological activity ingredient, with excellent processing properties. The flavonoid content, protein content, dietary fiber content, water and oil holding capacity of Fagopyrum tataricum seeds showed a downward trend, while the dietary fiber content of Fagopyrum tataricum stems and flowers generally showed an upward trend. The flavonoid and protein content of Fagopyrum tataricum stems and leaves decreased. The dietary fiber content of Fagopyrum tataricum leaves, flavonoid content and total antioxidant activity of Fagopyrum tataricum flowers increased first and then decreased. The total antioxidant capacity and flavonoid content of Fagopyrum tataricum flowers were significantly higher than other parts of tartary buckwheat. The highest total antioxidant capacity and flavonoid content were found 14 days after flowering, at 369.42 μmol/g and 7.83%, respectively, which were 6.2 and 8.8 times that of Fagopyrum tataricum seeds. Conclusion As food processing raw materials, different parts of Fagopyrum tataricum have the potential and value for food development and processing, and can be fully utilized. The nutrition and functional components of different parts of Fagopyrum tataricum are directly affected by harvest period, and the reasonable harvest period shall be determined based on the purpose in production.

苦荞(Fagopyrum tataricum)属蓼科(Polygonaceae), 是一种重要的药食同源杂粮作物, 既可做食品原料也可作为饲草原料, 又名鞑鞭荞麦、荞叶七、野兰荞、万年荞、菠麦、乌麦、花荞, 为双子叶草本植物, 性喜阴湿冷, 多种植于高山地域, 我国80%的苦荞集中种植在云南、贵州、四川等地[1-3]。苦荞被誉为“五谷之王”, 有很高的营养价值和多种活性功能[4], 不仅含有蛋白质、维生素、纤维素、氨基酸、叶绿素及镁、钾、钙、铁、锌、铬、铜、硒等多种营养成分, 还含有黄酮、酚酸、植物甾醇和荞麦碱等生物活性成分, 具有抗氧化、抗炎、抗癌等生物活性[5-11], 除苦荞籽粒外, 苦荞花、叶、茎等组织中均含有大量的芦丁、榭皮素、儿茶素等酚类化合物及其他营养物质, 也具有一定的保健功能[1,12]
植物采收期与其营养品质与功能活性成分有着直接密切的关系[13-14], 近年来, 不同采收期对作物品质的影响研究已成为许多学者关注的热点[15]。戴胜等[16]利用高效液相色谱法测定野菊花不同花期3种代表性有机酸类成分的含量, 以确定采收期对有机酸含量的影响。李欢等[17]测定了不同产地、采收期、加工方式及不同部位的槐角的槐角苷含量, 发现从9月初到12月初, 槐角苷含量逐渐降低。刘八斤等[18]通过分析萍乡安源地区不同采收期吴茱萸中吴茱萸碱、吴茱萸次碱、去氢吴茱萸碱3个有效成分的含量及体外抗氧化活性, 确定该地区吴茱萸的最佳采收期, 发现抗氧化活性与其有效成分含量呈正相关。
然而, 鲜见探讨不同采收期和部位对苦荞营养品质、加工特性与抗氧化活性影响的相关研究报道, 本研究拟通过分析不同采收期和不同部位苦荞淀粉含量、蛋白质含量、总黄酮含量和膳食纤维含量、持水性、持油性以及抗氧化活性的变化及相关性, 以期阐明不同采收期苦荞营养加工品质及其抗氧化活性变化规律, 为苦荞不同部位的综合利用提供科学依据。
苦荞, 采自会泽实验基地。
芦丁(纯度≥98.0%, 上海源叶生物科技有限公司); 乙二胺四乙酸(分析纯, 丹麦福斯集团); 乙醇、氢氧化钠、硝酸铝、亚硝酸钠、酒石酸钾钠、硫酸铁、乙酸铅、硫酸纳(分析纯, 天津市风船化学试剂科技有限公司); 总抗氧化活性(total antioxidant activity, TOA-C)、1,1-二苯基-2-三硝基苯肼(1,1-diphenyl-2-picrylhydrazyl, DPPH)检测试剂盒(北京索莱宝科技有限公司)。
ME204万分之一电子天平[梅特勒-托利多仪器(上海)有限公司]; UV-1800PC紫外可见分光光度计(上海美谱达仪器有限公司); DUMATECTM8000杜马斯定氮仪、Fibertec System E膳食纤维分析仪(丹麦福斯集团)。
苦荞开花后每隔7 d采收一次, 苦荞全株采收, 将籽粒、茎、叶、花分开, 105 ℃杀青10 min, 60 ℃烘干后, 打粉, 过80目网筛后, 并测定相关指标。籽粒、茎、叶、花采收编号根据时间先后顺序分别用Z、J、Y、H加采收批次数字进行命名。
膳食纤维采用GB 5009.88—2014《食品中膳食纤维的测定》, 淀粉含量按GB 5009.9—2016《食品中淀粉的测定》, 蛋白质含量采用杜马斯定氮法[19]测定, 总黄酮含量采用亚硝酸钠-硝酸铝比色法[20]测定。
持水力测定: 取1.5 g样品加入离心管中, 记录离心管和样品质量W1, 逐步加水, 每加1次水, 即用玻璃棒将样品混匀, 直至样品呈浆状但无水析出为止。在管壁上刮干玻棒, 在25 ℃, 4000 r/min转速下离心10 min。倒去上清液并用滤纸吸净多余的水分, 称量此时物料与离心管的质量W2。若离心后没有水析出, 则应继续加水、搅匀并再离心, 直至离心后有少量水析出为止。根据离心前后的质量变化计算青稞全粉的持水力。持水力(water holding capacity, WHC)根据式(1)计算:

持油特性的测定: 将5.0 g样品加入试管中, 称管料总质量M1, 用移液管准确加入30 mL花生油, 在沸水中加热20 min, 3000 r/min离心15 min, 小心倾倒出上层游离油, 然后将离心管倒置15 min, 沥尽油后称管料总质量M2。持油能力(oil holding capacity, OHC)用1.0 g样品所吸油的质量来表示, 计算如公式(2):

TOA-C、DPPH自由基清除能力采用试剂盒进行检测, 实验方法参照该试剂盒说明书。
所有实验均重复3次, 采用SPSS 22.0软件进行显著性分析(Duncan多重检验, P<0.05代表具有显著性差异); 采用Origin 2019软件绘制图形。
苦荞不同采收期籽粒的营养品质变化趋势如表1所示, 苦荞开花后35 d (Z5)内, 苦荞籽粒一直处于灌浆期, 淀粉含量随着采收期的延长不断上升, 花后21 d (Z3), 淀粉含量较花后7 d (Z1)增加了266.12%, 淀粉大量积累, 而苦荞籽粒蛋白含量仅增加了2.42%。花后28 d (Z4)后, 苦荞籽粒蛋白质含量较花后21 d (Z3)显著降低, 这与蔡芳丽[21]的研究结果相似。随着采收期的延长, 膳食纤维含量与总黄酮含量逐渐降低。开花时, 苦荞籽粒中, 膳食纤维含量较高, 可以达到48.40%, 这是由于苦荞籽粒和种皮难以实现分离, 带壳采收, 种皮占比较大。花后35 d (Z5), 苦荞籽粒总黄酮含量和膳食纤维含量已分别下降至0.33%和16.95%。苦荞开花后42~49 d (Z6~Z7), 籽粒基本成熟, 淀粉、黄酮、膳食纤维含量基本稳定, 无显著差异。
苦荞不同采收期茎的营养品质变化如表2所示, 随着采收期的延长, 苦荞茎总黄酮含量与蛋白含量总体呈下降趋势, 膳食纤维含量呈上升趋势, 且相较于苦荞籽粒、叶和花, 苦荞茎富含膳食纤维, 膳食纤维含量最高, 蛋白含量最低。花后35 d (J5), 总黄酮含量与蛋白含量分别较花后7 d (J1)分别下降了42.31%和44.06%, 膳食纤维含量则增加了51.21%。
苦荞不同采收期叶的营养品质变化如表3所示。随着采收期的延长, 苦荞叶总黄酮含量与蛋白含量总体呈下降趋势, 膳食纤维含量呈先升高后又下降的趋势, 开花后35 d (Y5), 苦荞叶中膳食纤维累积, 含量达58.57%。苦荞开花后42~49 d (Z6~Z7), 苦荞叶膳食纤维含量开始下降, 花后49 d (Z7)苦荞叶膳食纤维含量下降至48.01%, 仍比开花时高15.24%。在7个采收批次中, 苦荞叶的蛋白、总黄酮和膳食纤维含量均较高, 营养成分丰富, 可作为营养膳食补充添加剂添加至苦荞产品中, 提高产品营养功能性。
苦荞不同采收期花的营养品质变化如表4所示。苦荞首次开花后, 仍在不断开花, 后续花变成籽粒。随着采收期的延长, 苦荞花中蛋白质含量逐渐降低, 黄酮含量呈先增加后减少的趋势。其中, 苦荞花蛋白含量呈下降趋势与叶的变化规律一致, 花后21 d (H3), 苦荞花的蛋白质含量下降至16.21%。
采收期内, 苦荞花的蛋白、总黄酮、膳食纤维含量都比较高, 黄酮含量最高可达7.83% (H2), 膳食纤维高达51.44% (H3), 苦荞花也可以作为营养膳食补充添加剂, 以提高苦荞产品的营养功能性。
持油性与持水性是评价谷物类物质的重要特性, 持水性可评价谷物淀粉稳定性, 能反映谷物与水分子结合的能力, 持油性反映谷物蛋白质乳化的能力[22-24], 持水性和持油性在一定程度上反映了苦荞加工品质。不同采收期籽粒的持水性和持油性如图1所示, 随着采收期的延长, 苦荞籽粒的持水性和持油性总体呈下降趋势。开花后21~49 d (Z3~Z7), 采收的苦荞籽粒持油性无显著差异, 持水性在前6个采收批次内均有极显著差异。总体而言, 花后14 d (Z2)采收的苦荞籽粒持水性和持油性最优, 分别为3.86 g/g和2.61 g/g, 与其他几个批次有极显著差异(P<0.01)(除持水性与Z1无显著性差异), 此时, 苦荞籽粒加工特性最佳。
不同采收期苦荞茎的持水性和持油性如图2所示, 苦荞茎的持水性与持油性要高于苦荞籽粒、苦荞叶和苦荞花, 这可能与苦荞茎中膳食纤维含量较高有关, 说明苦荞茎有良好的加工性能, 可作为较优的生产原料添加到苦荞产品中。随着采收期的延长, 苦荞茎的持水性和持油性在总体呈上升趋势。花后35 d (J5), 苦荞茎的持水性达到最高值6.77 g/g, 但相较后2个采收批次(J6, J7)无显著差异。花后49 d (J7), 苦荞茎的持油性达到最高值5.66 g/g, 极显著高于其他采收批次(P<0.01)。因此, 可在花后35~49 d时采收苦荞茎, 此时, 苦荞茎加工特性较佳。
不同采收期苦荞叶的持水性和持油性如图3所示, 随着采收期的延长, 苦荞叶的持水性总体呈先升高再降低的趋势, 持油性无明显规律性。花后28 d内(Y1~Y4), 苦荞叶持水性无显著差异(P>0.05), 花后35 d (Y5), 苦荞叶的持水性达到最大5.41 g/g, 较花后7 d (Y1)持水性增加了24.08%, 在7个采收批次中, 花后35 d (Y5), 苦荞叶的持水性和持油性最好。
不同采收期苦荞花的持水性和持油性如图4所示, 采收期间, 苦荞花持水性和持油性无显著差异(P>0.05), 平均持水性和持油性分别为4.46 g/g和2.91 g/g, 要优于苦荞籽粒, 这可能与苦荞花的蛋白、总黄酮、膳食纤维含量均较高有关。综合来讲, 苦荞花具有良好的加工特性和营养功能性, 可作为原料添加至苦荞产品中。
铁离子还原/抗氧化能力(ferric ion reducing antioxidant power, FRAP)可以用于测定总抗氧化能力, 反应物质抗氧化物质和抗氧化酶等构成总抗氧化水平。不同采收批次及部位对苦荞总抗氧化能力的影响如图5所示。采收期间, 苦荞不同部位的总抗氧化能力由强到弱为: 花>叶>茎>籽粒, 苦荞花的总抗氧化能力高于籽粒、茎和叶, 这可能与苦荞花中黄酮等活性物质的含量较高有关[25]。采收期间, 苦荞花的总抗氧化能力呈先升高后降低的趋势, 花后14 d (H2), 苦荞花的总抗氧化能力最强, 为369.42 μmol/g, 这与苦荞花采收期总黄酮含量的变化趋势一致。随着采收期延长, 苦荞叶与苦荞籽粒的总抗氧化能力总体均呈先升高后降低的趋势, 花后42 d (Y6), 苦荞叶的FRAP值达到最高140.38 μmol/L; 花后14 d (Z2), 苦荞籽粒的FRAP值已达最高值, 此后(花后14~49 d, Z2~Z7)苦荞籽粒的总抗氧化能力持续下降。苦荞茎的抗氧化能力随采收期的延长不断下降, 花后49 d (J7)较花后7 d (J1)抗氧化能力下降了61.34%。
DPPH自由基清除评价方法是一种常用的评价植物抗氧化活性的方法, 主要通过抗氧化物质和DPPH自由基组合后改变溶液颜色, 从而影响吸光度来衡量样品的抗氧化活性[26]。由图6可知, 不同采收批次苦荞籽粒、苦荞叶与苦荞花的DPPH自由基清除能力没有明显的变化趋势, DPPH自由基清除率有轻微的波动, 但整体呈平稳趋势, 这说明开花后不同采收时间对苦荞籽粒、叶和花的DPPH自由基清除能力影响较小。采收初期(J1、J2)苦荞茎的DPPH自由基清除率著低于其他采收期, 花后21~42 d (J3~J6), 苦荞茎的DPPH自由基清除能力趋于平稳趋势, 花后49 d (J7), DPPH自由基清除率相较于花后42 d (J6)显著下降, 但仍高于采收初期(J1、J2)。
采收期间, 苦荞不同部位营养加工特性及抗氧化活性差别较大。苦荞叶与苦荞花中的总黄酮含量要高于苦荞籽粒与苦荞茎, 苦荞叶与苦荞花的总抗氧化能力同样要显著高于苦荞籽粒与苦荞茎; 苦荞茎蛋白含量显著低于苦荞其他部位, 采收期间苦荞茎平均蛋白含量仅为苦荞籽粒的38.32%, 而苦荞茎膳食纤维含量、持水性与持油性则明显高于其他部位, 苦荞茎持水性与持油性较高的原因可能与其膳食纤维含量较高有关。目前大部分研究与苦荞加工产品的研发主要集中于苦荞籽粒, 对苦荞茎、叶、花等组织开发利用较为局限, 造成了苦荞副产品资源的浪费[1,12], 可深入研究苦荞各部位功能性成分作用, 发挥苦荞茎、叶、花在营养、加工与保健功能等方面的优势。
采收期对苦荞不同部位的营养加工特性及抗氧化活性有重要影响。随着采收期的延长, 苦荞籽粒的黄酮含量、蛋白含量、膳食纤维含量、持水性、持油性与抗氧化能力总体呈下降趋势, 淀粉则逐渐增加。近年来, 苦荞醋、苦荞面、苦荞饮料、苦荞饭等新型苦荞产品被开发, 可根据苦荞籽粒的加工用途来确定采收时间[27-29], 如需要高纤维原料生产产品时可在前期采收苦荞籽粒, 生产苦荞曲奇饼干等淀粉类产品时可在采收后期采收苦荞籽粒[30]。苦荞茎富含膳食纤维, 随着采收期的延长, 其总黄酮含量、蛋白含量、总抗氧化活性总体呈下降趋势, 膳食纤维含量、持水性、持油性与DPPH自由基清除率总体呈上升趋势, 苦荞茎在采收前期已展现较优的加工特性, 而在采收后期其膳食纤维已高达65%~70%, 综合来看, 苦荞茎更宜在前期采收。苦荞叶黄酮含量与蛋白含量会随采收期延长而降低, 膳食纤维含量在花后35 d达到最大值, 在持水性和持油性在花后35 d达到最佳, 总抗氧化能力在花后42 d达到最大值。较苦荞其他部位, 苦荞花采收期较短, 但在保健功能方面有较大的开发潜能, 苦荞花总抗氧化能力与黄酮含量要明显高于苦荞苦荞籽粒与苦荞茎, 其变化趋势为先上升后下降, 花后14 d总抗氧化能力与黄酮含量最高, 此时为苦荞花采收的最佳时期。
苦荞籽粒、茎、叶与花富含营养物质与生物活性成分, 不同部位苦荞都有作为食品开发加工原料和饲草原料的潜能与应用价值, 应充分利用苦荞籽粒、茎、叶, 开发不同类型或保健功能的食品, 不可用于食品加工原料的部分也可加工成饲草, 实现苦荞的全株利用和综合利用。采收期对苦荞不同部位的营养与功能成分有直接影响, 总体来讲, 采收前期苦荞黄酮含量较高, 抗氧化能力较强; 采收后期苦荞淀粉、膳食纤维含量较高, 持水性、持油性较佳, 在实际生产应用中, 应根据产品的类型和目的可以选择不同的采收期。
