Objective To investigate the effects of different storage conditions on the water migration and volatile component changes of Zea mays, and to screen out characteristic volatile components. Methods Low field-nuclear magnetic resonance (LF-NMR) and gas chromatography-ion mobility spectrometry (GC-IMS) technology were used to study the changes in micro moisture, volatile component composition, and relative content of Zea mays under controlled temperature and humidity conditions. Results With the extension of storage time, moisture, fatty acid value, starch, and alcohol soluble protein indicators all undergo certain changes. When stored for 30 days, the moisture content reached equilibrium; the maximum value of fatty acid value was 46.11 mgKOH/100 g when stored at 65% RH for 90 days at 30 ℃, and the fatty acid value was still suitable for storage after 120 days; storing at 20 ℃ was beneficial for the accumulation of starch content; alcohol soluble proteins were significantly affected by temperature; under 65% RH conditions, as the storage temperature increased, the bound moisture content gradually decreased, resulting in the transformation of T20 strongly bound water to T21 weakly bound water and T22 strongly bound water to T23 weakly bound water; under 55% RH conditions, as the storage temperature increased, the bound water content gradually decreased, and the changes in the peaks of non flowing water and free water were consistent with those of bound water; the volatile components in the sample were classified as acids (2 types), aldehydes (20 types), ketones (15 types), alcohols (15 types), esters (12 types), furans (2 types), olefins (3 types), and amines (1 type). Conclusion As the storage temperature increases, the ability of Zea mays to retain moisture gradually weakens, while as the humidity increases, its ability to retain moisture increases. The high temperature and high humidity environment promotes the release of volatile components such as aldehydes and alcohols, while ketones and esters decrease. The 3-pentanone, 1-octen-3-one-M, 1-butanol, 3-methyl-D, 1-hexanol-D, isovaleric acid, methyl ester can be used as characteristic volatile components to distinguish mold growth in high moisture Zea mays, providing reference for the identification of mold growth in high moisture Zea mays.

目的 探究不同储藏条件对玉米水分迁移规律及挥发性成分变化的影响, 进而筛选出特征性挥发性成分。方法 采用低场核磁共振技术(low field-nuclear magnetic resonance, LF-NMR)和气相离子迁移谱技术(gas chromatography-ion mobility spectrometry, GC-IMS)研究在解除控温控湿条件下玉米的微观水分、挥发性成分组成及其相对含量的变化情况。结果 随着储藏时间的延长, 水分、脂肪酸值、淀粉、醇溶蛋白指标均发生一定变化。储藏30 d时水分含量达到平衡; 脂肪酸值在30 ℃ 65% RH储藏90 d时数值最大为46.11 mgKOH/100 g, 在储藏120 d脂肪酸值仍为宜存; 20 ℃储藏条件有利于淀粉含量积累; 醇溶蛋白受温度影响较明显; 在65% RH条件下, 随着储藏温度的升高, 结合水分含量逐渐降低, 出现了T20强结合水向T21弱结合水转化, T22强结合水向T23弱结合水转化; 在55% RH条件下, 随着储藏温度的升高, 结合水分含量逐渐降低, 不易流动水和自由水峰与结合水峰变化趋势一致; 样本中的挥发性成分归类为酸类(2种)、醛类(20种)、酮类(15种)、醇类(15种)、酯类(12种)、呋喃类(2种)、烯烃类(3种)、胺类(1种)。结论 随着储藏温度升高, 玉米保持水分的能力逐渐减弱, 而随湿度增加, 其保持水分能力则增强, 高温高湿环境促进醛类和醇类挥发性成分的释放, 而酮类与酯类物质则减少。3-戊酮、1-辛烯-3-酮-M、异戊醇-D、正己醇-D、异戊酸甲酯可作为区分偏高水分玉米霉变的特征性挥发性成分, 为偏高水分玉米霉变鉴定提供参考。

目的 探究不同储藏条件对玉米水分迁移规律及挥发性成分变化的影响, 进而筛选出特征性挥发性成分。方法 采用低场核磁共振技术(low field-nuclear magnetic resonance, LF-NMR)和气相离子迁移谱技术(gas chromatography-ion mobility spectrometry, GC-IMS)研究在解除控温控湿条件下玉米的微观水分、挥发性成分组成及其相对含量的变化情况。结果 随着储藏时间的延长, 水分、脂肪酸值、淀粉、醇溶蛋白指标均发生一定变化。储藏30 d时水分含量达到平衡; 脂肪酸值在30 ℃ 65% RH储藏90 d时数值最大为46.11 mgKOH/100 g, 在储藏120 d脂肪酸值仍为宜存; 20 ℃储藏条件有利于淀粉含量积累; 醇溶蛋白受温度影响较明显; 在65% RH条件下, 随着储藏温度的升高, 结合水分含量逐渐降低, 出现了T20强结合水向T21弱结合水转化, T22强结合水向T23弱结合水转化; 在55% RH条件下, 随着储藏温度的升高, 结合水分含量逐渐降低, 不易流动水和自由水峰与结合水峰变化趋势一致; 样本中的挥发性成分归类为酸类(2种)、醛类(20种)、酮类(15种)、醇类(15种)、酯类(12种)、呋喃类(2种)、烯烃类(3种)、胺类(1种)。结论 随着储藏温度升高, 玉米保持水分的能力逐渐减弱, 而随湿度增加, 其保持水分能力则增强, 高温高湿环境促进醛类和醇类挥发性成分的释放, 而酮类与酯类物质则减少。3-戊酮、1-辛烯-3-酮-M、异戊醇-D、正己醇-D、异戊酸甲酯可作为区分偏高水分玉米霉变的特征性挥发性成分, 为偏高水分玉米霉变鉴定提供参考。

Objective To investigate the effects of different storage conditions on the water migration and volatile component changes of Zea mays, and to screen out characteristic volatile components. Methods Low field-nuclear magnetic resonance (LF-NMR) and gas chromatography-ion mobility spectrometry (GC-IMS) technology were used to study the changes in micro moisture, volatile component composition, and relative content of Zea mays under controlled temperature and humidity conditions. Results With the extension of storage time, moisture, fatty acid value, starch, and alcohol soluble protein indicators all undergo certain changes. When stored for 30 days, the moisture content reached equilibrium; the maximum value of fatty acid value was 46.11 mgKOH/100 g when stored at 65% RH for 90 days at 30 ℃, and the fatty acid value was still suitable for storage after 120 days; storing at 20 ℃ was beneficial for the accumulation of starch content; alcohol soluble proteins were significantly affected by temperature; under 65% RH conditions, as the storage temperature increased, the bound moisture content gradually decreased, resulting in the transformation of T20 strongly bound water to T21 weakly bound water and T22 strongly bound water to T23 weakly bound water; under 55% RH conditions, as the storage temperature increased, the bound water content gradually decreased, and the changes in the peaks of non flowing water and free water were consistent with those of bound water; the volatile components in the sample were classified as acids (2 types), aldehydes (20 types), ketones (15 types), alcohols (15 types), esters (12 types), furans (2 types), olefins (3 types), and amines (1 type). Conclusion As the storage temperature increases, the ability of Zea mays to retain moisture gradually weakens, while as the humidity increases, its ability to retain moisture increases. The high temperature and high humidity environment promotes the release of volatile components such as aldehydes and alcohols, while ketones and esters decrease. The 3-pentanone, 1-octen-3-one-M, 1-butanol, 3-methyl-D, 1-hexanol-D, isovaleric acid, methyl ester can be used as characteristic volatile components to distinguish mold growth in high moisture Zea mays, providing reference for the identification of mold growth in high moisture Zea mays.

水分是影响玉米储藏品质的主要因素之一[1]。玉米内部水分含量和分布状态决定了玉米储藏过程中的生理、生化反应和品质变化情况。在玉米储藏过程中其内部不同强度的水分之间会发生迁移和含量的变化, 水分会参与糖类、脂类和蛋白质等生物大分子物质在粮食内部的代谢过程, 导致水分的移动性降低。如BAKHTAVAR等[2]研究发现玉米的原始水分含量越高, 随着储藏时间的延长, 脂肪和淀粉含量下降得越快; 边英霞[3]确定了在相同条件下, 低水分玉米的脂肪酸值增加速度明显低于高水分含量玉米的脂肪酸值增加速度。关于玉米籽粒水分迁移和含量变化的研究主要针对玉米干燥及种子发芽过程中的水分变化[4-5]。粮食中的挥发性成分也是目前学者关注的重点, 陈怡岑等[6]以温度为响应条件对偏高水分玉米的挥发性化合物进行表征, 确定了样品中的挥发性化合物, 主要包括醛类、酯类、酮类、烯醛类和醇类等; 钱佳成等[7]发现随着玉米储藏周期的变化, 不同水分、温度条件下玉米的挥发性物质差异较大, 气味变化明显; 徐瑞等[8]通过顶空-固相微萃取(headspace solid phase microextraction, HS-SPMC)结合气相色谱-质谱法(gas chromatography-mass spectrometry, GS-MS), 对不同冻藏温度下储藏期间玉米挥发性风味成分的种类和相对含量进行分析, 共检出烃类、醇类、醛类、酮类、酸类、酯类和少量其他类7大类物质, 共259种, 基本反映了玉米挥发性成分的整体信息。
粮食中水分检验主要参考GB 5009.3—2016《食品安全国家标准 食品中水分的测定》直接干燥法和GB 5497—1985《粮食、油料检验 水分测定法》定温定时烘干法等, 这些化学方法主要指示宏观水分的变化, 对于微观水分的研究需要依靠低场核磁共振技术(low-field nuclear magnetic resonance, LF-NMR)。LF-NMR可以提供试样内部水分在空间和时间上的信息、底物结合的程度以及有机物组分的变化情况, 显示水分的变化情况与迁移状态[9], 广泛用于试样中微观水分变化的测定[10-11]。常见的粮食挥发性成分检测方法有GC-MS[12-16]、电子鼻[17-20]和气相离子迁移谱法(gas chromatography-ion mobility spectrometry, GC-IMS)[21-23], GC-IMS作为新兴测试手段, 对粮食一些挥发性成分的小分子物质检测效果非常好。目前, 关于玉米储藏过程中解除控温控湿变化的水分迁移情况以及不同储藏条件对玉米挥发性差异性的分析鲜见报道。
本研究通过确定控温控湿储藏条件下玉米品质变化, 进一步采用LF-NMR和GC-IMS探讨解除控温控湿条件下玉米的微观水分、挥发性成分组成及相对含量的变化规律, 确保粮食仓储企业实现偏高水分玉米的安全储藏, 进而为节约干燥费用、降低水分减量损耗等方面提供理论依据。
选用2021年10月收获的M81玉米, 产地辽宁省开原市, 初始水分含量约27%, 室温干燥至水分含量15.0%±0.5%(偏高水分玉米), 实验开始于2021年11月, 完成于2023年12月。
无水乙醇、95%乙醇、0.1 mol/L的氢氧化钾乙醇溶液、酚酞指示剂、硫酸铜、硫酸钾、浓硫酸(分析纯, 国药集团化学试剂有限公司)。
MGC-HP人工气候箱、PH-240(A)干燥箱(上海一恒科学仪器有限公司); K9860全自动凯氏定氮仪、FlavourSpec风味分析仪(济南海能仪器股份有限公司); XFM110锤式旋风磨(上海嘉定粮油仪器有限公司); TCZ-H低场核磁共振成像分析仪(上海纽迈电子科技有限公司); LD510-2电子天平(精度0.01 g, 沈阳龙腾电子有限公司); MXT-WAX色谱柱(30 m×0.53 mm, 1.0 μm, 美国RESTEK公司)。
设定温度为20、25、30 ℃, 湿度为65% RH, 在人工气候箱中储藏0、30、60、90、120 d, 测定营养指标(可溶性糖、淀粉、醇溶蛋白)和储存指标(脂肪酸值、水分), 确定偏高水分玉米营养品质与储存品质间的差异。
解除控温控湿, 分别对20 ℃ 55% RH、25 ℃ 55% RH、30 ℃ 55% RH和20 ℃ 65% RH、25 ℃ 65% RH、30 ℃ 65% RH储藏120 d条件下的玉米为样本, 以原粮(YL)作为对照, 检测分析玉米样品中的水分变化。
解除控温控湿, 将样本放入搪瓷盘, 后用保鲜膜包裹, 模拟覆膜状态, 以20 ℃ 55% RH、30 ℃ 55% RH和20 ℃ 65% RH、30 ℃ 65% RH储存120 d条件下的玉米为样本, 以自然条件发霉玉米为对照, 在室温储藏放置30 d进行玉米挥发性有机物的检测分析。
(1)营养指标测定
醇溶蛋白: 样品粉碎后过60目筛, 取1 g样品加少量75%乙醇溶液, 置于研钵中研磨至匀浆。将研磨好的样品转移到离心管中。加10 mL 75%乙醇溶液, 将装有混合液的试管置于65 ℃水浴锅中5 min, 在此期间用玻璃棒不断搅拌。取出离心管, 以4000 r/min离心5 min, 将上清液倒入烧杯中。向离心管中再加入10 mL 75%乙醇溶液, 以上操作重复4次。将提取液以75%乙醇溶液定容至50 mL容量瓶中。取10 mL放入消化瓶中, 用凯氏定氮法进行测定。
淀粉: 按照GB 5009.9—2016《食品安全国家标准 食品中淀粉的测定》酸水解法进行测定。
(2)储藏指标测定
脂肪酸值: 按照GB/T 20570—2015《玉米储存品质判定规则》附录A中规定的方法进行测定。
水分: 按照GB 5009.3—2016进行测定。
(3)水分迁移分析
系统条件设置: 线圈直径: 40 mm, 主频SF=20 MHz, 偏移频率O1=994 kHz, 90度脉冲时间P1=7.52 μs, 180度脉冲时间P2=17.52 μs, 累加采样次数NS=4, 回波时间TE=0.350 ms, 回波个数NECH=18000, 信号接收带宽SW=100 kHz, 磁体强度0.55 T, 磁体温度32 ℃。
(4)挥发性成分分析
自动顶空进样条件: 顶空孵化温度80 ℃; 孵化时间15 min; 顶空进样针温度85 ℃; 进样量200 μL; 孵化转速500 r/min。
GC-IMS条件: MXT-WAX色谱柱(30 m×0.53 mm, 1.0 μm); 柱温60 ℃; 漂移气为高纯N2, 纯度≥99.999%; IMS探测器温度45 ℃。样品前处理: 称取1 g样品, 置于20 mL顶空瓶中, 80 ℃孵育15 min后进样。
GC-IMS数据使用G.A.S.公司开发的LAV 2.2.1软件的Gallery Plot插件、Dynamic PCA插件, 采用Excel 2007进行数据处理和制图; 采用SPSS 20.0软件进行统计学分析。
储藏条件及水分含量对玉米品质的影响较关键, 偏高水分玉米在一定条件下控温控湿, 可使其水分含量达到平衡[24], 如图1所示, 随着储藏时间的延长, 玉米水分含量整体呈现下降趋势, 并在储藏30~120 d保持水分近似恒定状态, 在储藏第120 d时水分含量分别为12.9%、12.9%和12.0%, 主要原因是由于环境温度升高, 相对湿度变小, 从而使玉米籽粒的平衡水分减少; 当湿度一致时, 籽粒的平衡水分随温度的升高而减少[25]。脂肪酸值通常表示谷物中脂质的水解程度, 是判断粮食品质的重要标准指标[26], 如图2所示, 脂肪酸值整体呈现上升趋势, 且变化趋势一致, 在储藏第120 d时脂肪酸值含量分别为40.11、43.73和43.31 mgKOH/100 g, 均为适宜储藏, 且在储藏第60~90 d发现随着储藏温度越高, 脂肪酸值上升速度越快, 这与万凡杰等[27]的实验结果一致。淀粉含量是玉米储藏过程中的主要品质指标[28], 如图3所示, 不同温度条件下, 偏高水分玉米的淀粉含量变化趋势较为一致, 整体上呈现先上升后下降再上升趋势, 在储藏第120 d时25 ℃ 65% RH条件下淀粉含量要高于其他条件, 说明25 ℃的储藏条件有利于淀粉含量的积累。玉米中的储藏蛋白以醇溶蛋白为主, 占储藏蛋白总量的60%以上[29], 如图4所示, 25 ℃ 65% RH、30 ℃ 65% RH条件下醇溶蛋白含量的变化趋势均为先下降后上升再下降, 20 ℃ 65% RH条件下是先下降后上升, 3个条件下醇溶蛋白含量均比储藏初期含量有所下降, 说明不同温度条件使得偏高水分玉米的醇溶蛋白含量发生变化, 这对玉米的功能特性有重要影响[30]
以每个组分峰值对应的时间作为弛豫时间常数, 弛豫时间常数的大小可以反映水分的自由度, 用T2表示, 弛豫时间常数分为强结合水T20 (0.1~1 ms)、弱结合水T21 (1~10 ms)、不易流动水T22 (10~100 ms)、自由水T23 (>100 ms)[31]。如图5所示, 体系包括4个T2弛豫组分, 出现两个峰, 第一个为结合水峰(T20、T21), 包括强结合水和弱结合水, 为体系水分的主要状态; 第二个为不易流动水和自由水峰, 由图5a可以看出, 65% RH条件下, 20 ℃结合水峰>25 ℃结合水峰>30 ℃结合水峰, 说明结合水峰随着储藏温度的升高, 结合水分含量在逐渐降低, 出现了T20强结合水向T21弱结合水转化, T22强结合水向T23弱结合水转化, 说明玉米体系中结合水紧密附着于大分子的极性基团上, 另外低温条件下水分子运动也受到周围环境的限制。由图5b可以看出, 55% RH条件下结合水峰随着储藏温度的升高, 水分含量逐渐降低, 不易流动水和自由水峰与结合水峰变化趋势一致, 两种水分含量均低于原粮(YL), 说明偏高水分玉米的结合水峰会向不易流动水和自由水峰迁移, 并且温度越高, 偏高水分玉米水分流失越快。
图6为相同温度不同湿度条件下水分变化图。由图6c可以看出结合水峰(第一个峰)湿度越高, 水分含量越高, 但均低于原粮(YL), 不易流动水和自由水峰(第二个峰)变化不明显; 由图6b可以看出其变化趋势与图6a一致; 图6a可以看出结合水峰(第一个峰)湿度越高, 水分含量越高, 且20 ℃ 65% RH条件下该部分水分高于原粮(YL)。
图7为二维俯视谱图, 不同样品中的挥发性物质存在差异, 为了更加明显地对比这种差异, 采用差异对比模式, 选取20 ℃ 65% RH 30 d的样品谱图作为参比, 其他样品的谱图扣减参比。如果二者挥发性有机物一致, 则扣减后的背景为白色, 而红色代表该物质的浓度高于参比, 蓝色代表该物质的浓度低于参比。从图7可以看出DZ(对照)发霉玉米样本的整体挥发性成分的挥发量要大于实验条件下的样本挥发量, 大部分挥发性物质的挥发量要高于参比; 30 ℃条件下样本的挥发量要高于20 ℃条件下的样本, 说明高温条件在一定程度上会促进挥发性成分的释放。
醛类物质是脂肪氧化的主要产物, 对玉米整体风味具有重要的影响[32], 醇类是另一类重要的谷物气味物质, 常被认为是不饱和脂肪酸的次生代谢产物, 由醛类物质进一步分解形成[33], 图8为30 ℃下不同湿度样本挥发性物质的对比, 其中红色框区域中成分在30 ℃ 55% RH 30 d样品组中峰点亮度较大, 说明这些成分在该样品组中挥发量较高, 包含壬醛(柑橘、肥皂味)、正丁醇-M(酒气味)等化合物以及未鉴别出的成分ID-4; 黄色框区域中成分在30 ℃ 65% RH 30 d样品组中峰点亮度较大, 说明这些成分在该样品组中挥发量较高, 主要包含β-蒎烯(松香树脂香)等化合物以及未鉴别出的成分ID-2、ID-5; 绿色框区域中成分在DZ样品组中峰点亮度较大, 说明这些成分在该样品组中挥发量较高, 主要包含2-丁醇(酒气味)、正丙醇-M(酒气味)等化合物以及未鉴别出的成分ID-6、ID-7等成分。
图9是65% RH条件下不同温度的样本挥发性物质对比, 其中红色框区域中成分在20 ℃ 65% RH-30 d样品组中峰点亮度较大, 说明这些成分在该样品组中挥发量较高, 包含丁酸乙酯(果香味)、正丙醇-M(酒气味)等化合物以及未鉴别出的成分ID-4; 黄色框区域中成分在30 ℃ 65% RH 30 d样品组中峰点亮度较大, 说明这些成分在该样品组中挥发量较高, 主要包含正己醛(生油脂、青草味)、2-丁酮-D(香味)等化合物以及未鉴别出的成分ID-2、ID-5; 绿色框区域中成分在DZ样品组中峰点亮度较大, 说明这些成分在该样品组中挥发量较高, 主要包含乙酸乙酯-D(果香味)等化合物以及未鉴别出的成分ID-6、ID-7等成分。
采用相对定量的方法, 利用不同化合物在不同样本中的峰体积, 对比不同化合物在不同样本中的挥发量, 具体数值见表1。样本中的挥发性成分归类为酸类(2种)、醛类(20种)、酮类(15种)、醇类(15种)、酯类(12种)、呋喃类(2种)、烯烃类(3种)、胺类(1种); DZ(对照)发霉玉米样本是自然条件下发霉的玉米, 通过数据可知各样本乙酸挥发量高, 发现20 ℃ 55% RH 30 d偏高水分玉米的乙酸挥发量高于其他条件但小于对照样本乙酸挥发量, 说明温度和湿度环境条件不能影响乙酸的挥发量, 醛类、醇类物质的挥发量受温度和湿度环境条件影响大, 此外样本中酯类挥发量较多, 主要包括乙酸乙酯-D(果香味)等。
偏高水分玉米在实验条件(20 ℃ 65% RH、25 ℃ 65% RH、30 ℃ 65% RH)下水分含量和醇溶蛋白含量均比储藏初期有所下降, 偏高水分玉米在20 ℃ 65% RH条件下储藏120 d, 反演峰图谱面积最大, 结合水保持的最好, 25 ℃ 65% RH条件次之; 此外, 发现储藏温度越高, 籽粒保持水分的能力越弱, 储藏湿度越高, 籽粒保持水分的能力越强; 不同储藏条件下的玉米籽粒在挥发性成分上具有差异, 主要挥发性有机物包括醛类、酯类、烯萜类、酮类和醇类等, 在30 ℃ 65% RH 30 d实验条件下, 醛类和醇类物质的挥发量增大, 酮类、酯类物质减少, 通过定量分析发现DZ(对照)发霉玉米样本含有的3-戊酮、1-辛烯-3-酮-M、异戊醇-D、正己醇-D、异戊酸甲酯等物质的挥发量较实验样本的挥发量明显增加且具有刺激性气味[34], 因此, 可作为区分偏高水分玉米霉变的标志性化合物, 为偏高水分玉米霉变鉴定提供参考。
