Objective To investigate the pollution of 5 kinds of heavy metals in 4 types of food in Hubei Province from 2022 to 2023, and to assess the health risks caused by heavy metal intake in the population. Methods From 2022 to 2023, a total of 316 samples of 4 types 6 kinds of food (soybean, peanut, mushroom, dictyophora, rice and wheat) were randomly collected in Hubei Province. The content of 5 kinds of heavy metals were detected by inductively coupled plasma-mass spectrometry (ICP-MS), and the contamination of food was analyzed by single factor pollution index (Pi) and comprehensive factor pollution index (PM). According to the health risk assessment model recommended by U.S. Environmental Protection Agency, the target hazard quotient (THQ) and target cancer risk assessment (TCR) were used to assess the non-carcinogenic and carcinogenic health risks caused by heavy metal exposure in the population. Results The overall qualified rate of food was close to 95%, the detection rate of Cd in heavy metals was the highest (100.0%, 14.2%). There were statistically significant differences in the detection rate and excess rate of the 5 kinds of heavy metals (χ2=179.320, 21.849, P<0.001). The 6 kinds of food contamination levels were dictyophora(severe), shiitake mushroom (mild), soybean (safe), peanut (safe), rice (safe), and wheat (safe). The individual heavy metal THQ of different regions and different genders was less than 1, but the comprehensive heavy metal THQ was greater than 1, and the carcinogenic risk of heavy metal in different regions and different genders was greater than 10-4. In Hubei, the non-carcinogenic and carcinogenic health risks caused by dietary heavy metals intake were higher in rural areas than in urban areas, and higher in women than in men. Non-carcinogenic health risks mainly come from Cd and Cr, while carcinogenic health risks mainly come from Cr. Conclusion There is a certain degree of contamination of edible fungi in the four types of food sold in Hubei Province from 2022 to 2023, especially the heavy pollution of dictyophora. The non-carcinogenic health risk caused by the intake of heavy metals through dietary means is high, and the carcinogenic risk is within the acceptable range. Relevant departments should strengthen supervision and pay attention to the health risks of Cd and Cr exposure.

目的 了解2022—2023年湖北省4类市售食品中5种重金属污染情况, 评估人群经食品摄入重金属所致健康风险。方法 2022—2023年, 在湖北省随机采集4类6种市售食品(黄豆、花生、香菇、竹荪、大米、小麦)共316份。采用电感耦合等离子体质谱法(inductively coupled plasma-mass spectrometry, ICP-MS)检测5种重金属含量, 采用单因子污染指数(Pi)、综合因子污染指数(PM)分析食品污染情况。根据美国国家环保局推荐的健康风险评价模型, 以目标危害指数法(target hazard quotient, THQ)、目标致癌风险评价法(target cancer risk, TCR)评估人群经重金属暴露所致的非致癌和致癌健康风险。结果 食品整体合格率接近95%, 重金属中Cd检出率、超标率均为最高(100.0%、14.2%)。5种重金属检出率和超标率差异均具有统计学意义(χ2=179.320、21.849, P<0.001)。6种食品污染等级依次为竹荪(重度)、香菇(轻度)、黄豆(安全)、花生(安全)、大米(安全)、小麦(安全)。不同地区、不同性别人群单项重金属THQ均小于1, 但综合重金属THQ均大于1, 不同地区、不同性别人群重金属致癌风险均大于10-4。湖北地区人群膳食摄入重金属所致非致癌和致癌健康风险, 农村高于城市, 女性高于男性。非致癌健康风险主要来源于Cd和Cr, 致癌健康风险主要来源于Cr。结论 2022—2023年湖北省4类市售食品中食用菌类存在一定程度的污染, 尤其是竹荪为重度。居民经膳食途径摄入重金属所致的非致癌健康风险较高, 致癌风险在可接受范围内。有关部门应加强监管, 关注Cd、Cr的暴露健康风险。

目的 了解2022—2023年湖北省4类市售食品中5种重金属污染情况, 评估人群经食品摄入重金属所致健康风险。方法 2022—2023年, 在湖北省随机采集4类6种市售食品(黄豆、花生、香菇、竹荪、大米、小麦)共316份。采用电感耦合等离子体质谱法(inductively coupled plasma-mass spectrometry, ICP-MS)检测5种重金属含量, 采用单因子污染指数(Pi)、综合因子污染指数(PM)分析食品污染情况。根据美国国家环保局推荐的健康风险评价模型, 以目标危害指数法(target hazard quotient, THQ)、目标致癌风险评价法(target cancer risk, TCR)评估人群经重金属暴露所致的非致癌和致癌健康风险。结果 食品整体合格率接近95%, 重金属中Cd检出率、超标率均为最高(100.0%、14.2%)。5种重金属检出率和超标率差异均具有统计学意义(χ2=179.320、21.849, P<0.001)。6种食品污染等级依次为竹荪(重度)、香菇(轻度)、黄豆(安全)、花生(安全)、大米(安全)、小麦(安全)。不同地区、不同性别人群单项重金属THQ均小于1, 但综合重金属THQ均大于1, 不同地区、不同性别人群重金属致癌风险均大于10-4。湖北地区人群膳食摄入重金属所致非致癌和致癌健康风险, 农村高于城市, 女性高于男性。非致癌健康风险主要来源于Cd和Cr, 致癌健康风险主要来源于Cr。结论 2022—2023年湖北省4类市售食品中食用菌类存在一定程度的污染, 尤其是竹荪为重度。居民经膳食途径摄入重金属所致的非致癌健康风险较高, 致癌风险在可接受范围内。有关部门应加强监管, 关注Cd、Cr的暴露健康风险。

Objective To investigate the pollution of 5 kinds of heavy metals in 4 types of food in Hubei Province from 2022 to 2023, and to assess the health risks caused by heavy metal intake in the population. Methods From 2022 to 2023, a total of 316 samples of 4 types 6 kinds of food (soybean, peanut, mushroom, dictyophora, rice and wheat) were randomly collected in Hubei Province. The content of 5 kinds of heavy metals were detected by inductively coupled plasma-mass spectrometry (ICP-MS), and the contamination of food was analyzed by single factor pollution index (Pi) and comprehensive factor pollution index (PM). According to the health risk assessment model recommended by U.S. Environmental Protection Agency, the target hazard quotient (THQ) and target cancer risk assessment (TCR) were used to assess the non-carcinogenic and carcinogenic health risks caused by heavy metal exposure in the population. Results The overall qualified rate of food was close to 95%, the detection rate of Cd in heavy metals was the highest (100.0%, 14.2%). There were statistically significant differences in the detection rate and excess rate of the 5 kinds of heavy metals (χ2=179.320, 21.849, P<0.001). The 6 kinds of food contamination levels were dictyophora(severe), shiitake mushroom (mild), soybean (safe), peanut (safe), rice (safe), and wheat (safe). The individual heavy metal THQ of different regions and different genders was less than 1, but the comprehensive heavy metal THQ was greater than 1, and the carcinogenic risk of heavy metal in different regions and different genders was greater than 10-4. In Hubei, the non-carcinogenic and carcinogenic health risks caused by dietary heavy metals intake were higher in rural areas than in urban areas, and higher in women than in men. Non-carcinogenic health risks mainly come from Cd and Cr, while carcinogenic health risks mainly come from Cr. Conclusion There is a certain degree of contamination of edible fungi in the four types of food sold in Hubei Province from 2022 to 2023, especially the heavy pollution of dictyophora. The non-carcinogenic health risk caused by the intake of heavy metals through dietary means is high, and the carcinogenic risk is within the acceptable range. Relevant departments should strengthen supervision and pay attention to the health risks of Cd and Cr exposure.

随着我国工业化进程加快, 城市化程度不断加深, 工业三废对水源、土地的金属污染不断累积[1]。据调查, 全国有16%左右土地点位重金属含量检测超标[2], 导致农作物对重金属的过度富集[3-5]。膳食途径是人群摄入重金属的主要途径, 可达60%~99%[6], 重金属通过食物链逐级积累, 最终在人群引起神经系统、血液系统、消化系统、泌尿系统等多重损害, 严重威胁人体健康[7-10]
我国长期开展食品有害因素监测, 苏州、湖南温州等地发现大米、水生蔬菜等Pb、Cd、As均存在超标情况[11-13], 西南矿区、南宁、阜阳等地谷物、黄豆重金属含量存在不同程度的超标情况, 部分样品可达国家标准限值3.8倍[14-16], 可见, 食品中的重金属污染已经成为普遍问题。
湖北省作为华中地区重要农作物种植大省[17]、全国食用菌产业大省, 其中孝感、十堰、黄冈等地已经有规模化的食用菌、花生、黄豆、大米生产集群, 因农作物生长环境易受到重金属污染[18], 监管湖北省此4类食品重金属污染情况有助于保障本地甚至全国居民食品安全。谷物、豆类、食用菌类等是湖北省居民膳食的主要部分, 目前, 仅有恩施州报道食品中重金属赋存情况[19-20], 其他地市报道较少。为了解湖北省食品重金属污染状况, 本研究对湖北省市售4类6种食品在多个环节进行随机取样, 分析5种重金属(Pb、Cd、Hg、As、Cr)含量, 结合膳食消费数据, 评估重金属污染情况与健康风险, 为湖北省重点食品重金属污染风险评估、开展食品监管专项工作、指导居民选择安全膳食提供数据支撑。
依托2022和2023年湖北省食品安全风险监测工作, 在湖北省各地市州(种植地、农贸市场、商店超市、网店)随机采集黄豆、花生、食用菌、谷物样品共计316份, 其中黄豆48份、花生108份、食用菌31份(竹荪11份, 香菇20份)、谷物129份(大米63份、小麦粉66份)。
NexION 1000G 电感耦合等离子体质谱仪(inductively coupled plasma-mass spectrometry, ICP-MS)(美国珀金埃尔默公司); M6型微波消解仪、ME203/02千分之一电子分析天平(瑞士梅特勒-托利多仪器公司); Milli-Q EQ 7000超纯水系统(德国默克公司)。
多元素标准溶液(编号GNM-M321686-2013, 质量浓度100 μg/mL)、多元素内标溶液(编号GSB 04-2826-2011, 质量浓度10 μg/mL)(国家有色金属及电子材料分析测试中心); 硝酸(超纯级, 江苏晶瑞公司); 菠菜质控品(编号GBW10015a GSB-6a, 中国地质科学院地球物理地球化学勘查研究所)。
按照GB 5009.268—2016《食品安全国家标准 食品中多元素的测定》方法, 采用ICP-MS法对316份样品进行5种重金属检测与复核。
实验室每年参加并通过国家食品安全风险评估中心、湖北省疾病预防控制中心等食品中多元素检测能力验证。检测仪器均在检定有效期内, 每批次样品均使用质控品进行质量控制, 随机抽取10%样品复测。超标样品复检两次, 取平均值上报系统。
食品中重金属限值参照GB 2762—2022《食品安全国家标准 食品中污染物限量》。采用单因子污染指数法和综合因子污染指数法对食品重金属污染程度进行评价, 评价标准见表1。单因子污染指数计算见公式(1)。

人体重金属暴露的健康风险根据美国国家环保局(U.S. Environmental Protection Agency, US EPA)中推荐的“四步法”健康风险评价模型。人体每天平均重金属摄入量(acceptable daily intake, ADI)的计算见公式(3)。

式中: ADIi为重金属i的日均估计暴露量, mg/(kg·d); Ci 为食品中重金属i的实测浓度, mg/kg; mIR为食品日摄入量, kg/d, 根据《第五次中国总膳食研究》对居民膳食消费量进行取值; tEF为暴露频率, 取值365 d/a; tED为持续暴露时间; tAT为平均作用时间, 取值tED×365 d; mBW为平均体重, 根据国家卫健委《中国居民营养与慢性病状况报告(2020年)》取值。具体取值见表2
非致癌风险评价采用目标危害指数法(target hazard quotient, THQ), 包括单项重金属THQ与综合重金属目标危害指数(total target hazard quotient, TTHQ), 计算见公式(4)~(5)。

式中: THQi为重金属i的非致癌健康风险值; ADIi为重金属i的日均估计暴露量, mg/(kg·d); RfDi代为重金属i的暴露参考剂量, 取值参考US EPA, Cr、As、Cd、Hg、Pb分别取值0.003、0.0003、0.001、0.0003和0.0035 mg/(kg·d)。THQ或TTHQ<1表明暴露人群没有明显健康风险; THQ或TTHQ≥1表明暴露人群存在显著健康风险; THQ或TTHQ>10表明存在慢性毒性作用。THQ值越大, 暴露人群健康风险越高。
致癌风险评价采用目标致癌风险(target cancer risk, TCR)评价法。包括单项重金属TCR与综合重金属目标致癌风险(total target cancer risk, TTCR), 参考US EPA的致癌斜率系数(cancer slope factor, CSFi), Cr、As分别取值0.5 kg·d/mg、1.5 kg·d/mg, 计算见公式(6)~(7)。

式中: TCRi为重金属i的致癌健康风险值; ADIi为重金属i的日均估计暴露量, mg/(kg·d); CSFi为重金属i致癌强度指数, kg·d/mg。根据US EPA推荐的致癌风险参考值, 当≥1.00×10-4时, 表明该重金属在该暴露途径下有致癌风险。
采用Excel 2010及SPSS 20.0软件进行数据的统计和分析, 对率或构成比的计数资料进行比较采用χ2检验, 检验水准α=0.05。对于低于检出限的数据均以检出限的半量参与统计。
316份样品整体合格率为94.7%。5种重金属中Cr平均含量最高, 为0.3770 mg/kg, 其次是Cd, 平均含量为0.1892 mg/kg, 其余重金属平均含量为0.0077~0.0526 mg/kg。5种重金属中Cd检出率最高, 达100.0%, 其余重金属检出率依次为Cr (88.9%)>As (79.1%)>Pb (69.3%)>Hg (21.8%), 不同重金属的检出率差异有统计学意义(χ2=179.320, P<0.001)。5种重金属中Cd超标率最高, 为14.2%, Hg无超标, 其余重金属超标率依次为Pb (5.7%)>Cr (4.1%)>As (2.5%)。不同重金属的超标率差异有统计学意义(χ2=21.849, P<0.001)。见表3
不同重金属在不同种类食品中的含量不同, Pb含量: 食用菌类>谷类>坚果类>豆类, 最高为1.3205 mg/kg; Cd含量: 食用菌类>坚果类>谷类>豆类, 为最高5.8805 mg/kg; Hg在所有食品中含量均较低; As在竹荪上含量较高, 最高为1.1075 mg/kg, 其他食品As含量较低; Cr含量: 坚果类、食用菌类>豆类>谷类, 最高为4.3100 mg/kg。不同重金属在不同种类食品中的检出率不同, Pb检出率: 食用菌类>坚果类>豆类、谷类, 最高为100.0%; Cd在所有种类食品中的检出率均为100.0%; Hg在大米中检出率为100.0%, 食用菌中为20.0%以上, 其他种类食品中未检出; As检出率: 食用菌类>谷类、坚果类>豆类, 最高为100.0%; Cr检出率: 食用菌类>坚果类>豆类>谷类, 最高为100.0%。具体见表4
不同类别种类的食品中对不同重金属的富集能力差异显著。竹荪中Pb、Cd以及As的贡献比最高, 分别为65.8%、84.29%和63.37%。大米中Hg贡献比最高为43.65%, 黄豆中铬的贡献比最高为44.59%。花生、小麦各重金属贡献比均较低。不同食品的Pb、Cd、Hg、As、Cr 5种重金属贡献比差异具有统计学意义(χ2=22217.731、39825.669、7029.533、20819.708、9646.970, P<0.001)。结果见图1
单因子污染指数评价显示, Pb、Cd、As在竹荪中的Pi值超标, 分别为2.173、14.250、2.000, Cd在花生、香菇中的Pi值超标为0.766、1.388, 其余食品的Pi 值均小于0.7。综合污染因子评价显示, 竹荪、香菇PM值分别为10.601、1.089; 黄豆、花生、大米、小麦PM值均小于0.7, 为安全食品。结果见表5
对不同地区、不同性别重金属暴露所致的非致癌健康风险和致癌健康风险进行评估, 结果如表6所示。不同地区、不同性别人群THQ值均小于1, 但TTHQ值均大于1,主要来源均为Cd和Cr。不同地区、不同性别人群TCR、TTCR值均大于1×10-4, 主要来源均为Cr。非致癌及致癌健康风险农村高于城市, 女性高于男性。
湖北省是鱼米之乡, 重要的食品基地, 关注湖北地区食品的重金属污染现状有利于保护市民身体健康。2022—2023年湖北省4类市售食品整体合格率接近95%, 其中谷类、豆类、坚果类食品5种重金属污染程度均处于安全范围内, 而食用菌类存在轻至重度的污染, 与河南、无锡、陕西、上海、贵州等地区近些年的监测结果一致[21-25]。监测结果表明, Cd、As、Cr在市售食品中的污染比较普遍, 检出率在80%~100%, 同时Cd的超标率也达到14.2%。Pb、Cd、As对竹荪的重金属污染贡献较高。竹荪Cd超标报道较多[26-29], 其高污染现状可能与其生长特性、生产流程有关, 有关部门应该积极开展竹荪食品的Cd污染风险防治宣传教育活动。与青岛地区调查结果相比[30], 香菇中Cd污染水平有所不同, 但与刘哲等[31]、李静等[32]研究结果一致, 湖北省香菇对Cd富集程度较高, 且存在一定非致癌健康风险, 应引起关注。
监测表明, 湖北地区人群均存在5种重金属暴露所致的非致癌风险和致癌风险, 其中农村居民高于城市居民, 女性高于男性。TTHQ主要来源于Cd和Cr, Pb和Hg贡献较低, 这与各类食品中Cd浓度均较高相符合。TTCR主要来源于Cr。湖北地区人群Cd和Cr所致的健康风险需要引起关注。
本研究也存在一些局限性, 竹荪样本的采样量不高, 可能会产生偏差; 竹荪食品的产地来源比较单一, 没能开展大范围多样本多来源的集中检测。此外, 竹荪食品在居民日常生活中食用制作前, 会进行大量冲洗, 其实际产生的健康风险可能会比本研究所评估的结论要低。在目前的国家相关标准下, 除食用菌外其他食品处于安全状况, 但仍要关注重金属非致癌风险, 建议监管部门开展食用菌专项监测工作, 保护居民身体健康。
