Objective To establish a qualitative screening method for 29 kinds of mycotoxins in grains based on QuEChERS dSPE EMR-Lipid and liquid chromatography-quadrupole time of flight-mass spectrometry. Methods The sample was extracted by 20 mL 70% acetonitrile (containing 1% formic acid), purified by modified QuEChERS method, electrospray ionization, positive ion sweeping, sequential window acquisition of all theoretical mass spectra. Results The established method included a database of primary precision mass, secondary mass spectrometry, and retention time. This method enabled rapid screening of 29 kinds of mycotoxins in soybean, corn, rice, and wheat. The screening limits of detection of ranged from 0.005 to 0.050 mg/kg. The method met the basic screening requirements. Conclusion The method is simple, specific, stable and reliable. The method enables the processing and qualitative screening of large volumes of samples from soyabean, corn, rice and wheat in a short time. It has high application value.

目的 建立QuEChERS dSPE EMR-Lipid结合液相色谱-四极杆飞行时间质谱法定性筛查粮谷中29种真菌毒素残留的方法。方法 样品经20 mL 70%乙腈(含1%甲酸)水溶液提取, 改良QuEChERS法净化, 电喷雾电离, 正离子扫猫, 顺序窗口采集所有理论质谱模式。结果 建立了包含一级精确质量、二级质谱和保留时间的数据库, 该方法实现了黄大豆、玉米、大米和小麦中29种真菌毒素的快速筛查, 筛查检出限分布在0.005~0.050 mg/kg之间, 均能满足筛查基本要求。结论 该方法操作简便、特异性好、稳定可靠, 较短时间内完成黄大豆、玉米、大米和小麦中大批量样品的处理及定性筛查, 具有较高的应用价值。

Objective To establish a qualitative screening method for 29 kinds of mycotoxins in grains based on QuEChERS dSPE EMR-Lipid and liquid chromatography-quadrupole time of flight-mass spectrometry. Methods The sample was extracted by 20 mL 70% acetonitrile (containing 1% formic acid), purified by modified QuEChERS method, electrospray ionization, positive ion sweeping, sequential window acquisition of all theoretical mass spectra. Results The established method included a database of primary precision mass, secondary mass spectrometry, and retention time. This method enabled rapid screening of 29 kinds of mycotoxins in soybean, corn, rice, and wheat. The screening limits of detection of ranged from 0.005 to 0.050 mg/kg. The method met the basic screening requirements. Conclusion The method is simple, specific, stable and reliable. The method enables the processing and qualitative screening of large volumes of samples from soyabean, corn, rice and wheat in a short time. It has high application value.

真菌毒素是真菌产生的有毒次级代谢产物, 是粮谷的主要污染源之一, 对人和动物具有致癌、致畸、致突变、免疫毒性和雌激素效应等[1]。我国是粮谷进口大国, 主要进口黄大豆、玉米、大米和小麦四大类粮谷[2]。真菌毒素是粮食安全的主要指标, 监测粮谷中的真菌毒素具有重要意义。GB 2761—2017《食品安全国家标准 食品中真菌毒素限量》规定了粮谷中黄曲霉毒素B1 (aflatoxin B1, AFB1)、黄曲霉毒素M1 (aflatoxin M1, AFM1)、脱氧雪腐镰刀菌烯醇(deoxynivalenol, DON)、赭曲霉毒素A (ochratoxin A, OTA)及玉米赤霉烯酮等限量。
近年来, 真菌毒素检测方法主要包括酶联免疫吸附法、高效液相色谱法、液相色谱-串联质谱法、胶体金免疫层析法、近红外光谱分析技术等[3-5]。高效液相色谱-串联质谱法作为主流的检测方法, 虽有较好的灵敏度和特异性, 但是其分辨率较低, 遇到样品基质复杂, 容易出现假阳性结果。诸多文献表明, 基于高效液相色谱-串联质谱法的真菌毒素检测需依赖同位素内标法解决回收率低的缺陷, 其检测成本高昂, 对检测人员要求较高[6-8]。随着质谱技术逐渐发展成熟, 液相色谱-四极杆飞行时间质谱法(liquid chromatography quadrupole time of flight mass spectrometry, LC-Q/TOF-MS)可以在不降低灵敏度的前提下大幅提高分辨率, 抗复杂基质干扰能力更强, 对目标化合物具有更好的鉴别能力[9], 其在定性鉴别真菌毒素领域具有应用前景, 但目前鲜少有相关研究报道。
本研究采用QuEChERS dSPE EMR-Lipid为前处理方法技术, 该技术无复杂的前处理步骤, 无需采用内标检测, 依赖LC-Q/TOF-MS的高选择性, 结合顺序窗口采集所有理论质谱模式(sequential window acquisition of all theoretical mass spectra, SWATH), 构建了29种真菌毒素的质谱库, 建立的黄大豆、玉米、大米和小麦等中29种真菌毒素的快速筛查方法可用于进口大宗粮谷的真菌毒素监测, 为防范风险提供更多有力选择。
黄大豆、玉米、大米、小麦为随机抽检样品。
甲醇、乙腈、甲酸、乙酸铵(色谱纯, 德国Merck公司); 萃取盐包(4 g MgSO4、1 g NaCl、1 g柠檬酸钠、0.5 g三水合二柠檬酸二钠)(分析纯, 上海安谱实验科技股份有限公司); 15 mL Bond Elut EMR-Lipid dSPE净化管(美国Agilent公司)。
29种真菌毒素标准: 15-乙酰基脱氧雪腐镰刀菌烯醇(15-acetyl-deoxynivalenol, 15-A-DON)、3-乙酰基脱氧雪腐镰刀菌烯醇(3-acetyl-deoxynivalenol, 3-A-DON)、AFB1、黄曲霉素B2 (aflatoxin B2, AFB2)、黄曲霉素G1 (aflatoxin G1, AFG1)、黄曲霉素G2 (aflatoxin G2, AFG2)、AFM1、交链孢酚(alternariol, AOH)、链孢酚单甲醚(alternariol monomethyl ether, AME)、黄绿青霉素(citreoviridin, CIT)、桔霉素(citrinin, CIN)、环匹阿尼酸(cyclopiazonic acid, CPA)、DON、二乙酰氧基草镰刀菌烯醇(diacetoxyscirpenol, DAS)、伏马毒素B1 (fumonisin B1, FB1)、伏马毒素B2 (fumonisin B2, FB2)、伏马毒素B3 (fumonisin B3, FB3)、镰刀菌酮X (fusarenon X, FU-X)、茄病镰刀菌烯醇(neosolaniol, NEO)、雪腐镰刀菌烯醇(nivalenol, NIV)、OTA、赭曲霉毒素B (ochratoxin B, OTB)、青霉酸(penicillic acid, PA)、青霉震颤素A (penitrem A, PEA)、树脂毒素(RSF-resiniferatoxin, RTX)、杂色曲霉素(sterigmatocystin, ST)、T-2毒素(T-2 toxin, T-2)、腾毒素(tentoxin, TEN)、细交链孢菌酮酸(tenuazonic acid, TA)(质量浓度均为50 mg/L, 天津阿尔塔科技有限公司)。
X500R液相色谱-四极杆飞行时间质谱仪(美国AB SCIEX公司); AL204精密电子天平(精密度0.1 mg, 瑞士梅特勒托利多公司); SDC-4000涡旋振荡仪(深圳逗点生物技术有限公司); Allegra X-30R高速冷冻离心机(美国贝克曼库尔特公司); Milli-Q高纯水发生器(德国默克公司); AUTO EVA-60氮吹浓缩仪(睿科集团股份有限公司); 0.22 µm亲水PTFE有机滤膜(上海安谱实验科技股份有限公司); ACQUITY UPLC HSS T3 色谱柱(150 mm×2.1 mm, 1.8 μm, 美国Waters公司)。
准确称取5 g(精确至0.01 g)粮谷(黄大豆、玉米、大米、小麦)试样于50 mL离心管中, 放入一颗陶瓷均质子, 加入20 mL含1%甲酸的70%乙腈水溶液(V:V), 涡旋振荡3 min, 9500 r/min离心5 min。吸取上清液至50 mL离心管中, 放置-30 ℃冰箱冷冻1 h后加入萃取盐包, 涡旋振荡3 min, 9500 r/min离心5 min, 移取全部上清液至15 mL离心管中, 40 ℃氮吹浓缩至略低于5 mL, 乙腈定容为5.00 mL, 混匀, 待净化。
移取待净化液至15 mL Bond Elut EMR-Lipid dSPE净化管(使用前, 加入5.00 mL水, 涡旋混匀30 s活化), 涡旋混匀2 min, 迅速在10 °C和9500 r/min下离心5 min, 吸取上清液过0.22 μm有机滤膜, 供LC-Q/TOF-MS检测。
色谱柱: ACQUITY UPLC HSS T3 (150 mm×2.1 mm, 1.8 μm)。流动相A: 含0.1%甲酸水溶液; 流动相B: 含0.1%甲酸的50%乙腈甲醇溶液(V:V)。梯度洗脱程序: 0~3.0 min, 5% B; 3.0~5.0 min, 5%~40% B; 5.0~9.0 min, 40%~99% B; 9.0~15.0 min, 99% B; 15.1~20.0 min, 5% B; 流速: 0.3 mL/min; 进样量: 10 µL。
采集模式: 正离子; 喷雾电压: 5500 V; 离子源温度: 550 °C; 雾化气压: 0.379 MPa; 辅助气压: 0.414 MPa; 气帘气压: 0.241 MPa; 扫描模式: SWATH/TOF- MS/TOF- MS/MS。TOF-MS参数: 采集范围150~785 m/z; 去簇电压: 80 V; 碰撞能: 20、35、50 eV。TOF-MS/MS参数: 采集范围50~785 m/z, 根据实际分布, 平均拆分成10段。每5针进样进行一次自动校准。
质谱数据、方法学考察数据处理采用SCIEX OS 2.0.1软件分析, 样品含量计算采用WPS 2024, 绘图采用Origin 2022。每组样品均设置平行重复6次。
采用Waters ACQUITY UPLC HSS T3 (150 mm×2.1 mm, 1.8 μm)、Agilent Poroshell 120 EC-C18 (100 mm×2.1 mm, 2.7 μm)、Shimadzu Shim-pack Velox C18 (100 mm×2.1 mm, 2.7 μm)测试, 29种真菌毒素在3种C18柱均有较好的保留和分离, 峰形尖锐对称, 且稳定, 满足检测需求。29种真菌毒素大部分为弱极性化合物, 个别化合物中等极性, 考虑到Waters ACQUITY UPLC HSS T3 (150 mm×2.1 mm, 1.8 μm)是在C18键合极性修饰基团, 增强对极性分子的反相保留能力, 延长目标物的保留时间, 可以排除更多极性干扰物, 故最终选择采用此款色谱柱在正模式下进行检测。
本研究对29种真菌毒素在乙腈-水、甲醇-水和50%乙腈/甲醇-水流动相体系[水相和有机相中均分别添加0.1%甲酸、0.005 mol/L乙酸铵(含0.1%甲酸, pH约为3.2)和0.005 mol/L乙酸铵(含0.01%甲酸, pH约为4.5)]的色谱分离效果进行对比研究。在不同流动相条件下目标化合物的分离度存在差异, 且质谱响应强弱不同。在甲醇流动相体系中, 大多化合物响应较强, 分离度相对较好, 但个别化合物峰型稍差。在乙腈流动相条件下, 较多化合物峰型较好, 但部分结构相似物质的分离度较差。50%乙腈/甲醇混合流动相的优点是结构相似物质分离更好, 兼顾峰形和分离度。结果表明, 0.1%甲酸水-50%甲醇/乙腈溶液(含0.1%甲酸)混合溶液为流动相时获得了最优的色谱峰形及质谱响应, 由图1可见, 29种真菌毒素在色谱柱中保留分布较好, 分离效果达到理想状态。
本研究29种真菌毒素的极性差异较大, 选择提取溶剂时需考虑溶剂性质和样品基质特点。常用提取试剂有乙腈、甲醇、乙酸乙酯和水等。乙腈的极性适中, 提取目标化合物的极性范围更宽。通过适当酸化可促进真菌毒素从组织中溶出, 改善提取效率, 乙腈水可增加极性化合物的提取效率[10-12]
本研究分别考察了6种提取溶剂: 乙腈/水(50:50, V:V)、乙腈/水/甲酸(50:49:1, V:V:V)、乙腈/水(70:30, V:V)、乙腈/水/甲酸(70:29:1, V:V:V)、乙腈/水(84:16, V:V)、乙腈/水/甲酸(84:15:1, V:V:V), 对29种真菌毒素在黄豆中的加标(0.100 mg/kg)回收率作为比较指标。如图2, 结果表明, 乙腈占比越大, 越适用于大多数毒素的提取(乙腈体积比为70%和84%的提取试剂好于体积比50%的乙腈提取试剂)。酸性条件下能较好的提取大多数毒素, 除FB1、FB2、FB3、DON、AME、DAS、PA、AOH的提取回收率较低, 其余19种毒素的回收率均超过了60%。最终选定提取溶剂为乙腈/水/甲酸(70:29:1, V:V:V)。
免疫亲和柱分析多种包括黄曲霉毒素在内的多种真菌毒素[13-16], 但免疫亲和柱价格昂贵, 作为定性筛查不是最好的选项。QuEChERS方法针对样品采用简单的3步流程(萃取、净化和分析), 该方法在谷物和乳制品中包括黄曲霉毒素在内的多种不同分析物和基质的前处理方面极具前景[17-19]
QuEChERS常用净化剂十八烷基硅胶(C18)和乙二胺基-N-丙基(ethylenediamine-N-propyl, PSA)在处理高脂质样品时有一定限制[20-23], 净化剂本身与目标物非选择性作用, 脂类物质去除能力不足, 加大了污染检测仪器的可能性, 影响数据的准确度和精密度。增强型脂质去除产品EMR-Lipid是一种新型吸附剂, 它结合了体积排阻和吸附剂与长脂肪链官能团之间的疏水作用, 可选择性吸附样液中的直链烃类结构化合物(包括游离脂肪酸、甘油三酯和磷脂等), 有助于实现简单高效的净化[24-25]
本研究的样品基质为富含脂质的粮谷样品, 如前处理净化不当会影响仪器检测。考察了提取液经EMR-Lipid dSPE新型吸附剂净化(第一组), 未经净化(第二组), 经传统QuEChERS(提取液加入900 mg MgSO4、150 mg PSA和150 mg C18 dSPE净化粉末)净化(第三组)后, 采用LC-MS/TOF进行全扫描得到的色谱图进行比较。结果显示(见图3), 第一组可获得出色的基质去除效果。
在上述仪器条件下进标准溶液, 对29种真菌毒素逐一进行Q1全扫描, 记录保留时间、母离子质荷比、质量偏差以及离子化形式等主要质谱信息[26-27]。在软件中输入化合物的名称、分子式, 选择加荷方式, 软件自动计算母离子理论质量质荷比, 建立29种真菌毒素的一级精确质量数据库(见表1)。
29种真菌毒素在保留时间窗口的响应值超过阈值, 则自动触发在不同碰撞能量下采集二级碎片谱图, 由此建立二级信息谱库, 包括母离子提取离子色谱图、母离子同位素分布图、不同碰撞能量下的叠加二级质谱图等信息[28-29], 如图4
欧盟最新版SANTE/11945/2015《食品和饲料中农残分析的质量控制和方法确认的指导文件》的决议, 使用高分辨质谱确证时: 至少需要1个高分辨母离子及1个特征碎片离子, 且质量偏差≤5 ppm, 一级母离子m/z<200时, 其绝对偏差应<1 mDa。《农业农村部公告第312号饲料中风险物质的筛查与确认导则液相色谱-高分辨质谱法(LC-HRMS)》目标物筛查判定条件为采样点数: 单个目标物峰不少于5个采样点; 信号响应要求: 信噪比(S/N)存在时, 应使S/N≥3; 质量准确性: 一级母离子m/z≥200时, 其相对偏差应≤10 ppm; 一级母离子m/z<200时, 其绝对偏差应<1 mDa。
参考上述文件要求, 筛查规则定为: 母离子和一个特征碎片离子的质量偏差≤5 ppm(母离子m/z<200, 绝对偏差<1 mDa), 保留时间≤2.5%(最大不超过0.5 min), 母离子的同位素分布差异≤10%, 二级谱库相似度≥80%, 4个关键因子占总得分的30%、30%、10%、30%, 且仪器自动综合打分不低于90分。为考察方法的可靠性, 添加了29种真菌毒素标准品的样品按1.3、1.4进行检测, 目标化合物的综合得分为91.4~99.7分, 所得结果均大于定性筛查的最低分(90分)。
质谱的基质效应主要由分析物共流组分影响离子化效率所致, 表现为离子增强或抑制。试验考察了黄豆、大米、玉米、小麦的基质效应, 在纯试剂和不同基质提取液中添加标准溶液, 比较目标物在纯试剂和基质液中的信号峰面积, 可按公式对基质效应进行量化评估: 基质效应(matrix effects, ME)/%=[(基质匹配校准曲线斜率/纯溶剂标准曲线斜率)-1]×100%。|ME|<20%为弱基质效应, 可忽略而无需采取补偿措施; 20%≤|ME|≤50%为中等程度基质效应, |ME|>50%为强基质效应, 须采取措施补偿基质效应[30-32]。DAS、NEO、RTX、PEA表现为基质增强效应, 其他项目表现为基质抑制效应。29种真菌毒素在4种基质中的基质效应情况见表2(用|ME|表示), 尽管采用优化的净化方法能有效减少样品提取液中的干扰组分, 但大部分|ME|均大于20%, 故本研究以基质匹配法检测。
本研究选择了黄大豆、玉米、大米、小麦来进行方法适用性验证, 按1.3前处理方法操作, 质谱数据采集后分析, 结果表明: 4种基质不存在对29种真菌毒素产生影响的假阳性化合物。
分别取黄大豆、玉米、大米、小麦空白基质样品, 添加不同体积的混合标准溶液, 按1.3样品前处理方法制备加样溶液, 以符合定性结果判定方法所述检出结果的最低质量浓度作为检出限。结果表明, NIV在4种基质中检出限0.050 mg/kg, 15-A-DON和3-A-DON在4种基质中检出限0.025 mg/kg, AFB1、AFB2、AFG1、AFG2、AFM1在4种基质中检出限0.005 mg/kg, 其他真菌毒素在4种基质中检出限0.010 mg/kg。
采集深圳地区市售黄大豆、玉米、大米、小麦各10批次, 采用本研究建立的方法对其进行29种真菌毒素的筛查分析, 结果表明: 22批次粮谷中检出DON, 检出值0.02~0.10 mg/kg; 7批次粮谷中检出FB1, 检出值0.02~0.06 mg/kg; 5批次粮谷中检出FB2, 检出值0.03~ 0.05 mg/kg; 6批次粮谷中检出FB3, 检出值0.01~0.05 mg/kg。
本研究利用LC-Q/TOF 20 min内可完成黄大豆、玉米、大米、小麦中29种真菌毒素的快速筛查。样品提取液经QuEChERS dSPE EMR-Lipid 简单快速的方法净化, 有效地消减了基质干扰。以精确相对分子质量、二级质谱图和相对保留时间为基础, 构建了筛查数据库, 建立快速筛查方法, 利用高分辨质谱的检测, 极大提高了方法定性的准确性。该方法在粮谷的真菌毒素快速筛查中可以得到广泛应用。
本研究覆盖的真菌毒素度尚小, 可通过此研究路线构建更大的本地筛查数据库, 通过优化色谱分离技术, 更加准确地定性部分同分异构体真菌毒素, 使此研究方法更具使用价值。
