Objective To establish a method for the simultaneous determination of 10 kinds of phytotoxins, including tripterygium wilfordii and aconite in honey by ultra performance liquid chromatography-tandem mass spectrometry. Methods The samples were extracted by vortexing with 1% formic acid water-70% acetonitrile water solution. After high-speed centrifuged, added appropriate amount of primary secondary amine (PSA) to the supernatant, and shaked and mixed evenly, passed through 0.22 μm filter membrane, then determined and analyzed by the instrument. The matrix working curve was used for quantification by the external standard method. Results The limits of detection of 10 kinds of phytotoxins were 0.03-1.90 μg/kg. The 10 kinds of phytotoxins showed good linearity in a certain range of mass concentration with the correlation coefficients were all greater than 0.999. The average recovery rates of toxins ranged from 89.5% to 113.8% at 3 kinds of different concentration spiked levels, and the relative standard deviations of 0.5%-8.7%. Conclusion The method is convenient, sensitive and accurate, and provides technical support for rapid screening of phytotoxins in emergencies. It is suitable for simultaneous determination of various phytotoxins in honey samples, especially in batch samples.

目的 建立超高效液相色谱-串联质谱法同时测定蜂蜜中雷公藤类、乌头类10种植物毒素的方法。方法 样品采用1%甲酸水-70%乙腈水溶液涡旋提取, 经高速离心后, 取上清液加入适量N-丙基乙二胺(primary secondary amine, PSA), 振荡混匀, 过0.22 μm滤膜, 上机测定分析, 采取基质工作曲线, 外标法定量。结果 10种植物毒素的检出限为0.03~1.90 μg/kg。10种植物毒素在一定的质量浓度范围内线性关系良好, 相关系数均大于0.999。3种不同浓度加标水平下, 植物毒素的平均加标回收率范围为89.5%~113.8%, 相对标准偏差为0.5%~8.7%。结论 该测定方法操作便捷、灵敏度高, 准确性强, 为应急事件中植物毒素快速筛查提供技术支持, 适用于实验室对蜂蜜样品特别是批量样品中多种植物毒素的同时测定分析。

Objective To establish a method for the simultaneous determination of 10 kinds of phytotoxins, including tripterygium wilfordii and aconite in honey by ultra performance liquid chromatography-tandem mass spectrometry. Methods The samples were extracted by vortexing with 1% formic acid water-70% acetonitrile water solution. After high-speed centrifuged, added appropriate amount of primary secondary amine (PSA) to the supernatant, and shaked and mixed evenly, passed through 0.22 μm filter membrane, then determined and analyzed by the instrument. The matrix working curve was used for quantification by the external standard method. Results The limits of detection of 10 kinds of phytotoxins were 0.03-1.90 μg/kg. The 10 kinds of phytotoxins showed good linearity in a certain range of mass concentration with the correlation coefficients were all greater than 0.999. The average recovery rates of toxins ranged from 89.5% to 113.8% at 3 kinds of different concentration spiked levels, and the relative standard deviations of 0.5%-8.7%. Conclusion The method is convenient, sensitive and accurate, and provides technical support for rapid screening of phytotoxins in emergencies. It is suitable for simultaneous determination of various phytotoxins in honey samples, especially in batch samples.

蜂蜜是一种深受广大人民群众喜爱的天然甜味剂, 包含丰富的营养物质, 如葡萄糖、抗坏血酸、类胡萝卜素和葡萄糖氧化酶等, 不仅为机体提供能量, 还具有抗炎和抗氧化功能[1-2]。然而, 蜜蜂在采集花蜜过程中极有可能将部分有毒植物毒素带入蜂蜜中, 从而存在引发食物中毒应急事件的风险。若误食有毒蜂蜜可能会造成腹痛腹泻、恶心呕吐等症状, 严重者甚至会导致休克或死亡[3], 其中毒症状及严重程度与蜂蜜中所含毒素的种类息息相关。
蜂蜜中常见的植物毒素有雷公藤类、马桑类、乌头类及其代谢产物等毒素。雷公藤作为一种传统的中药材, 适当摄入具有良好的保健功能[4-5], 过量摄入易对肝肾等内脏系统造成严重损害[6-7], 还可能引起女性生理期紊乱甚至不孕以及男性生殖障碍等症状[8-10]。误食马桑类植物通常会导致兴奋、痉挛等症状[11-12]。乌头类毒素是常存在于乌头属植物中的生物碱, 适当的摄入可为机体带来良好的调理作用, 例如祛风除湿、抗炎镇痛、抗肿瘤等功能[13], 一旦摄入过量则会造成消化系统、神经系统以及心血管系统等器官严重损害[13]。2004—2020年, 云南省蜂蜜中毒事件共19起, 其中84人发病, 27人死亡, 死亡率高达32.14%[14-15], 野生蜂蜜中毒的死亡率在20%以上[16]
目前, GB 14963—2011《食品安全国家标准 蜂蜜》中明确规定: 蜜蜂采集植物的花蜜、分泌物或蜜露应安全无毒, 不得来源于雷公藤、博落回、狼毒等有毒蜜源植物。但是, 目前针对蜂蜜中植物毒素的检测方法尚无相应的国家标准, 检测方法主要有高效液相色谱法[17-19]、液相色谱-串联质谱法[20-25]及液相色谱-串联飞行时间质谱法[26-29]。高效液相色谱法易受样品中各种基质的干扰; 目前大部分的研究均采用高效液相色谱-串联质谱法, 其检测方便, 灵敏度高, 准确度好, 适用于多种毒素的同时检测。
近年来, 主要偏向于研究蜂蜜中农药残留、抗生素以及营养成分等的测定[30-33], 蜂蜜中植物毒素的测定则多数为吡咯里西啶类生物碱, 有关雷公藤类和乌头类毒素的测定相对较少[26,34-35], 云南由于其独特的气候条件赋予了植物王国称号, 同时也增加了蜂蜜中存在多种植物毒素的风险, 结合云南常见引发食物中毒事件的植物毒素, 蜂蜜中所含雷公藤和乌头类毒素亦不容忽视[36-37], 本研究通过蜂蜜样品前处理条件、质谱参数及色谱条件的优化, 并对方法进行验证, 以期建立科学、高效、准确的同时检测蜂蜜中10种植物毒素的测定方法, 旨在保障蜂蜜的食用安全以及实现快速应对公共卫生应急中毒事件中植物毒素的快速筛查, 为植物毒素中毒事件的防控及医疗诊断提供技术支持。
蜂蜜样品购自超市、农贸市场、商店及网络商店, 共计61件, 产地为云南22件, 江西12件, 福建2件, 湖南1件, 湖北2件, 广西2件, 浙江2件, 上海1件, 山东1件, 河北1件, 产地不详15件, 散装为12件, 预包装49件。
QTRAP 4500质谱仪(美国AB SCIEX公司); 1290 Infinity Ⅱ型高效液相色谱(美国Agilent Technologies公司); Multi Reax Fu多位试管振荡器(德国Heidolph公司); Allegra X-30R Centrifuge高速冷冻离心机(美国Beckman Coulter公司); XS205DU十万分之一分析天平(瑞士Mettler Toledo公司); BEH C18色谱柱(2.1 mm×100 mm, 1.7 μm, 美国Waters公司)。
甲醇、乙腈[纯度≥99.9%, 西格玛奥德里奇(上海)贸易有限公司]; 甲酸(色谱纯, 美国ROE SCIENTIFIC INC公司); Bondesil C18散装净化剂、N-丙基乙二胺(primary secondary amine, PSA)净化剂(美国Agilent公司)。
雷公藤吉碱、雷公藤春碱(纯度≥95%)、雷公藤次碱、雷公藤定碱、雷酚内酯、雷公藤内酯酮、雷公藤甲素、乌头碱(纯度≥98%)(乐美天医药/德斯特生物公司); 马桑宁、新乌头碱、次乌头碱(纯度≥98%)(武汉天植生物技术有限公司)。
色谱柱: BEH C18柱(2.1 mm×100 mm, 1.7 μm); 流动相: A相为甲醇, B相为0.1%甲酸水溶液; 柱温: 40 ℃; 流速0.3 mL/min; 进样体积: 5 μL。
电喷雾电离(electrospray ionization, ESI)正离子模式(ESI+)扫描; 多反应监测(multiple reaction monitoring MRM)模式检测; 离子源温度为550 ℃; 离子喷雾电压5500 V。
分别用甲醇溶解10种固体标准物质得标准储备液, 根据各植物毒素的响应强度用15%甲醇水溶液稀释得混合标准使用液, 采用未检出目标化合物的蜂蜜样品配制基质标准工作系列浓度。
精确称取蜂蜜样品1.0 g于10 mL离心管中, 加入10 mL 1%甲酸水-70%乙腈水溶液涡旋提取20 min, 离心机8000 r/min离心5 min, 取上清液3 mL, 加入10 mg PSA, 振荡混匀, 经0.22 μm滤膜过滤, 备测。
仪器数据处理分析软件为MultiQuant 3.0, 再采用Excel 2016进行数据整理和分析, 实验重复3次测定, 结果用平均值±标准偏差表示。
采用针泵恒流注入方式注入10种植物毒素的单标溶液进行质谱参数确认。ESI+扫描, 利用Q1 MS扫描模式寻找一级质谱母离子, 通过Product Ion Scan (MS2)扫描模式寻找二级质谱碎片离子信息, 选择响应程度稳定和最强的前2个碎片离子分别作为定量定性离子, 最后用MRM扫描模式优化质谱参数去簇电压及碰撞能量, 详见表1
10种植物毒素易溶于甲醇和乙腈, 通过对流动相组成和配比的考察, B相为纯水时目标色谱峰的响应强度较低甚至部分目标峰未出现, 加入甲酸后能显著提高目标峰的响应强度, A相为乙腈时各植物毒素的响应强度差别太大, 如图1所示, 色谱峰分离度欠缺, 而在甲醇的洗脱下可获得较佳的响应值和分离效果, 因此确定流动相为甲醇-0.1%甲酸水并进行梯度洗脱, 洗脱程序为: 0~2.0 min, 10%~15% A; 2.0~3.0 min, 15%~25% A; 3.0~4.0 min, 25% A; 4.0~4.1 min, 25%~45% A; 4.1~5.0 min, 45%~80% A; 5.0~6.8 min, 80%~95% A; 6.8~6.9 min, 95%~100% A; 6.9~8.5 min, 100% A; 8.5~8.6 min, 100%~10% A; 8.6~10.0 min, 10% A; 分离效果如图2所示, 植物毒素离子色谱图见图3
通过不同比例的甲醇水溶液及乙腈水溶液对目标化合物进行提取条件考察, 采用50%以上浓度的乙腈水溶液进行毒素提取时, 离心后有机相和水相分层情况良好, 但马桑宁的提取率均低于70%, 而未出现分层的甲醇水和30%乙腈水提取溶液所得马桑宁提取率均在合理范围内(如表23), 推测马桑宁主要存在于水相中, 因此, 后续实验中可针对马桑宁进行单独实验。除此之外, 由于不同比例的甲醇溶液及30%乙腈水溶液经提取离心后均不分层, 进样时其中的糖分可能对色谱柱及最终结果产生影响, 因此不适用于蜂蜜中10种雷公藤毒素的提取。
通过不同比例乙腈水提取液的提取效果分析, 如表3所示, 100%乙腈水对10种毒素的提取率大部分均在80%以下; 80%乙腈水对雷公藤次碱的提取率高达122.86%±0.07%; 50%乙腈水仅对4种毒素提取效果较好, 结合成本考虑, 最终选择70%乙腈水作为最佳提取试剂。
为了更好确保实验结果的准确性, 本研究还对70%乙腈水的提取体积进行了考察, 结果如图4所示。3 mL提取试剂由于体积较少, 可能存在提取不充分的情况, 仅对5种毒素的提取效果较为理想。随着提取试剂体积量的逐步增加, 10种毒素的提取效果也在逐渐优化, 5 mL的提取试剂仅对雷公藤春碱、雷公藤定碱、新乌头碱3种毒素的提取效果不佳, 8 mL的提取试剂仅对雷公藤春碱的提取率较差, 而10 mL和15 mL的提取试剂所得10种毒素的提取率则均在80%~120%范围内。而20 mL提取试剂时所得提取效果则有所欠佳, 因此, 选择10 mL作为提取试剂最佳体积。
考虑蜂蜜中植物毒素大多为偏弱碱性的生物碱, 使用弱酸性溶液能更容易提取。通过对不同体积分数甲酸水进行考察, 结果如图5所示, 0.1%、0.2%、0.5% 3种体积分数的甲酸水对10种毒素的总体提取效果均不理想, 其中均有部分毒素的提取率超出120%, 不符合实验室检测标准。剩下4种体积分数的甲酸水, 其对10种毒素的总体提取率则随着甲酸水体积分数的增加而呈现出下降的趋势, 因此, 最终选择1%的甲酸水-70%乙腈水作为提取剂。
由于蜂蜜样品具有较高的水溶性, 先用水进行蜂蜜样品溶解促使具有较高分散性, 再添加乙腈来进行目标化合物的提取。通过不同水量的加入进行加标回收实验分析, 考察最佳的加水量, 如表4所示, 加水量的变化对3种乌头类生物碱的提取率未产生较大影响, 均未有较好的提取效果, 3种乌头类生物碱提取率范围为29.20%± 1.27%~53.92%±2.09%, 3种乌头类生物碱可能溶于水相时较难提取到有机相中; 对雷公藤毒素均具有较强的基质增强效应, 提取率范围为92.41%±4.06%~266.14%±3.95%, 随着添加水量的增加, 样品表现出的基质效应增强越加突出, 添加0.5 mL水量时平均提取率为87.28%, 添加4.0 mL水量时的平均提取率为154.02%。通过不同添加水量对植物毒素提取率的考察, 通过添加水量的变化进行蜂蜜中植物毒素的提取未能达到良好的提取效果, 采用直接配制好的乙腈水溶液进行提取。
部分蜂蜜样品在离心后仍可能存在乳化离心后分层不理想的现象, 通过加入氯化钠盐析剂可以使有机相和水相较好分层, 方便样品的后续处理。通过对加入不同量的氯化钠进行加标回收率的考察, 从图6可知, 氯化钠的加入对3种乌头类生物碱的损失较大, 回收率范围仅为24.79%~33.09%; 对雷公藤内酯酮均存在基质增强效应, 且随着氯化钠添加量的增多, 其基质增强效应越发凸显, 因此进行蜂蜜样品植物毒素的提取可不加盐析剂, 直接采用0.22 μm滤膜过滤即可。
PSA和C18作为常用的两种吸附剂, 具有吸附样品基质中多种杂质的功能, 能够得到准确提取。本研究对PSA和C18剂量分别进行了考察, 结果如图7图8所示, 其中, 5、40、80 mg PSA和5、10、20、40 mg C18均对部分毒素的提取率过高或较低, 不符合实验室检测标准。另外, 与C18相比, PSA吸附色素及糖类的效果更好, 更适用于蜂蜜中各类杂质的吸附, 因此, 最终采用PSA作为吸附剂。10、20、100 mg PSA相比, 10 mg PSA所得10种毒素的提取率更接近于100%, 更符合检测要求, 因此最终选择10 mg PSA作为吸附剂。
目标植物毒素在离子化反应过程中受到样品基质影响, 响应信号可发生抑制或增强的现象叫做基质效应。将检测样品中有样品基质的响应信号与不含样品基质的响应信号比较, 响应信号减小则为基质抑制效应, 反之则为基质增强效应。按照样品前处理条件分别处理加入C18净化剂和PSA净化剂以及未加净化剂的蜂蜜样品基质, 样品基质处理前及纯溶剂均加入等量的10种植物毒素混合标准溶液, 测定分析后计算样品基质中各植物毒素与纯溶剂中目标植物毒素的峰面积比值, 比值越接近100%则表示无基质效应。由图9可知, 加入C18后, 对雷公藤吉碱和雷公藤次碱均存在明显的基质增强效应, 对雷公藤甲素存在明显基质抑制效应, 而未加吸附剂和加入PSA吸附剂的样品则未出现明显的基质增强和抑制效应, 二者相比较, 加入PSA吸附剂的样品呈现出的基质增强或抑制效应更弱, 因此, 可加入适量PSA吸附剂进行实验检测。
用配制好的系列质量浓度混合标准溶液, 采取优化好的质谱参数、色谱条件进行标准曲线测定, 以标准物质质量浓度为横坐标(X, µg/L), 各组分对应峰面积为纵坐标(Y)绘制标准曲线, 得到线性范围和相关系数。以3倍信噪比(S/N)和10倍(S/N)分别计算各植物毒素的方法检出限(limit of detection, LOD)和定量限(limit of quantity, LOQ)。从表5结果可知, 相关系数r>0.999, 10种植物毒素在给定的浓度范围内有良好的线性关系。LODs范围为0.03~1.90 μg/kg, LOQs范围为0.1~6.2 μg/kg, 满足实验要求。
随机选取蜂蜜样品进行加标回收实验, 各称取1.0 g, 加入低、中、高3个浓度水平的各植物毒素标准溶液, 按照上述条件处理及上机测定, 通过外标法定量计算加标回收率, 每个加标样品平行测定6次, 计算相对标准偏差(relative standard deviation, RSD)。由表6可知, 10种植物毒素的平均加标回收率均在89.5%~113.8%之间, 相对标准偏差为0.5%~8.7% (n=6)。实验数据表明, 该方法具有良好的稳定性与准确性, 适用于蜂蜜样品中植物毒素的测定分析。
采用建立的方法对61件蜂蜜样品进行了检测分析, 其中乌头碱检出1件, 含量为4.76 µg/kg, 检出率1.64%, 蜂蜜中毒事件多发于每年的6—8月份, 高发期为8月, 此时正值雷公藤类和乌头类开花泌蜜季节[38], 其余样品及其他检测项目都小于检出限均为未检出, 说明市场所售蜂蜜产品有安全保障。
植物毒素种类多样, 对人体的危害也各不相同, 在多毒素存在的情况下毒性有叠加的风险。蜂蜜在采花粉或采蜜、加工环节均有被植物毒素污染的风险, 且存在多种植物毒素污染可能性, 目前国家还未制定植物毒素相关检测标准方法, 但通过实际样品的测定分析, 已有低浓度水平的植物毒素检出, 因此需要加强蜂蜜中植物毒素的监测, 保障国家的食品安全。该优化的方法前处理简单便捷, 具有较高的回收率及较好的重现性, 满足日常的实验室检测需求, 为蜂蜜质量安全监管提供技术支持, 为蜂蜜产业的发展保驾护航。
