Objective To establish a detection method for rapid determination of chlorate and perchlorate in Capsicum annuum L. powder by automatic QuEChERS-ultra performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS). Methods After the sample was centrally weighed, the sample was extracted with a mix-ture of 1% avetic acid-acetonitrile-water (1:1, V:V), and automatic pre-treatment with automatic QuEChERS instrument, the gradient elution was performed by 0.5% formic acid methanol-0.5% formic acid solution, Thermo HYPERCARB as a separation column, gradient elution, series mass spectra electrospray ionization negative mode ionization, multiresponse monitoring pattern detection, qualitative research by the ratio of retention time and ion, the internal standard method was used to qualitative experiment. Results Automatic QuEChERS instrument was used to determine the chlorate and perchlorate of Capsicum annuum L. powder and the pretreatment efficiency was 6 times that of manual. The limits of quantification (S/N=10) were 10 μg/kg, when the spiked level was 10-50 μg/kg (n=6), the sample recoveries for chlorate and perchlorate were 94.0%-96.4%, the relative standard deviations were less than 5%. The calibration curves were linear in the range of 1-100 μg/L with the correlation coefficients above 0.999. Conclusion The method has high automation degree, good accuracy and stability, and the limit of quantitation also meets the requirements of European Union export, can be applied for the determination of chlorate and perchlorate in Capsicum annuum L. powder.

目的 建立全自动QuEChERS前处理结合超高效液相色谱-串联质谱法(ultra performance liquid chromatography-tandem mass spectrometry, UPLC-MS/MS)快速测定辣椒粉中氯酸盐和高氯酸盐的检测方法。方法 样品经集中称量后, 用1%乙酸乙腈-水(1:1, V:V)作为提取溶剂, 利用全自动QuEChERS仪编程后自动前处理, 采用0.5%甲酸甲醇-0.5%甲酸水溶液为流动相, Thermo HYPERCARB为分离柱, 进行梯度洗脱, 串联质谱电喷雾电离源负模式电离, 多反应离子监测模式检测, 以子离子比和保留时间定性, 用内标法定量。结果 采用全自动QuEChERS仪来进行辣椒粉氯酸盐及高氯酸盐的测定前处理效率是人工的6倍, 定量限(S/N=10)为10.0 μg/kg, 加标水平为10~50 μg/kg (n=6)时, 平均回收率为94.0%~96.4%, 相对标准偏差小于5%, 在1~100 μg/L范围内线性关系良好, 相关系数均大于0.999。结论 本方法自动化程度高、准确性及稳定性好, 同时定量限也满足欧盟出口的要求, 适用于出口辣椒粉中氯酸盐及高氯酸盐的测定。

目的 建立全自动QuEChERS前处理结合超高效液相色谱-串联质谱法(ultra performance liquid chromatography-tandem mass spectrometry, UPLC-MS/MS)快速测定辣椒粉中氯酸盐和高氯酸盐的检测方法。方法 样品经集中称量后, 用1%乙酸乙腈-水(1:1, V:V)作为提取溶剂, 利用全自动QuEChERS仪编程后自动前处理, 采用0.5%甲酸甲醇-0.5%甲酸水溶液为流动相, Thermo HYPERCARB为分离柱, 进行梯度洗脱, 串联质谱电喷雾电离源负模式电离, 多反应离子监测模式检测, 以子离子比和保留时间定性, 用内标法定量。结果 采用全自动QuEChERS仪来进行辣椒粉氯酸盐及高氯酸盐的测定前处理效率是人工的6倍, 定量限(S/N=10)为10.0 μg/kg, 加标水平为10~50 μg/kg (n=6)时, 平均回收率为94.0%~96.4%, 相对标准偏差小于5%, 在1~100 μg/L范围内线性关系良好, 相关系数均大于0.999。结论 本方法自动化程度高、准确性及稳定性好, 同时定量限也满足欧盟出口的要求, 适用于出口辣椒粉中氯酸盐及高氯酸盐的测定。

Objective To establish a detection method for rapid determination of chlorate and perchlorate in Capsicum annuum L. powder by automatic QuEChERS-ultra performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS). Methods After the sample was centrally weighed, the sample was extracted with a mix-ture of 1% avetic acid-acetonitrile-water (1:1, V:V), and automatic pre-treatment with automatic QuEChERS instrument, the gradient elution was performed by 0.5% formic acid methanol-0.5% formic acid solution, Thermo HYPERCARB as a separation column, gradient elution, series mass spectra electrospray ionization negative mode ionization, multiresponse monitoring pattern detection, qualitative research by the ratio of retention time and ion, the internal standard method was used to qualitative experiment. Results Automatic QuEChERS instrument was used to determine the chlorate and perchlorate of Capsicum annuum L. powder and the pretreatment efficiency was 6 times that of manual. The limits of quantification (S/N=10) were 10 μg/kg, when the spiked level was 10-50 μg/kg (n=6), the sample recoveries for chlorate and perchlorate were 94.0%-96.4%, the relative standard deviations were less than 5%. The calibration curves were linear in the range of 1-100 μg/L with the correlation coefficients above 0.999. Conclusion The method has high automation degree, good accuracy and stability, and the limit of quantitation also meets the requirements of European Union export, can be applied for the determination of chlorate and perchlorate in Capsicum annuum L. powder.

氯酸盐和高氯酸盐, 作为一类新型污染物引起大家的普遍关注, 氯酸盐会造成人的急性及慢性毒性, 诱发溶血性贫血及肾功能损害, 高铁血红蛋白血症, 抑制人甲状腺对碘元素的吸收。人类主要通过食品接触和饮用水来接触氯酸盐, 其中食品主要是生产线上消毒水的消毒, 易导致氯酸盐迁移至食品中[1], 高氯酸盐是一种易溶于水、难降解的环境污染物。环境中高氯酸盐的来源包括自然形成和人工合成两部分。自然形成的高氯酸盐主要是在大气中自然形成和天然存在于硝石矿; 人工合成主要是由于高氯酸盐具有强氧化性, 使其广泛应用于火箭推进剂、烟花、道路照明弹和炸药等生产中。高氯酸盐通过土壤和水源进入食品链, 人食用后进会抑制碘含量的摄取损害人体的甲状腺功能, 进而会使婴幼儿的生长及发育出现问题[2-3]。对于成年人来说, 如果在一段时间内摄入高氯酸盐, 会发生机体的骨髓造血细胞再生障碍[4-5], 我国作为辣椒粉大国, 每年有上亿元以上的市场, 而因氯酸盐、高氯酸盐的超标影响出口的案例不在少数, 所以准确高通量、快速监控辣椒粉中氯酸盐、高氯酸盐的含量时迫在眉睫的事情。
目前国家及相关文献对氯酸盐和高氯酸盐检测方法通常为液相色谱-串联质谱法[6-11]、液相色谱法、离子色谱法等[12-15], 测定时间长、自动化程度不高等的缺点, 随着科技的发展, 检测技术也慢慢由劳动密集型逐步向自动化、智能化发展[15-16], 因此开展全自动QuEChERS-超高效液相色谱-串联质谱法测定残留量的检测技术迫在眉睫[17-19]。本研究建立了辣椒粉中氯酸盐和高氯酸盐的全自动QuEChERS-超高效液相色谱-串联质谱(ultra performance liquid chromatography-tandem mass spectrometry, UPLC-MS/MS)检测方法, 拓展了自动化对食品检测的方向的影响, 更对其他项目如农药残留、兽药残留、污染物等的检测提供相应的参考。
STX1202ZH电子天平[精度0.01 g, 奥豪斯仪器(常州)有限公司]; 1290-6495C超高效液相色谱-串联质谱仪(美国Agilent公司); HRH-Q60全自动QuEChERS仪(北京慧荣和科技有限公司); PTFE-Q亲水性滤膜(0.22 μm)(上海安谱实验科技股份有限公司); Thermo HYPERCARB色谱柱(3.0 mm× 100 mm, 5.0 μm)(美国Thermo Fisher Scientific公司)。
辣椒粉购买于市场。
氯酸盐标准品、高氯酸盐标准品(质量浓度1000 mg/L)、氯酸盐内标(质量浓度200 mg/L)、高氯酸盐内标(质量浓度100 mg/L)(上海安谱实验科技股份有限公司)。
甲酸、乙酸(色谱纯, 上海麦克林生化科技股份有限公司); 氯化钠(优级纯, 国药集团化学试剂有限公司); 甲醇、乙腈(色谱纯, 德国Merck公司); 一级水(蒸馏水, 杭州娃哈哈集团有限公司)。
将辣椒去蒂, 用四分法制样方法取500 g, 用不锈钢均质器粉碎后, 全部装入样品杯中, 冷冻, 备用[20]
用容量瓶准确配制1.0 mg/L各标准液, 存储于棕色标准品储液瓶中, 放置4 ℃冷藏保存[21-22]
准确称取2.00 g试样于50 mL离心管放置于全自动QuEChERS仪中, 编辑程序, 按照如表1运行。
取离心后样液过0.22 μm滤膜后, 供UPLC-MS/MS分析。
(1)液相色谱条件
流动相: 0.5%甲酸水(A), 0.5%甲酸甲醇(B); 色谱柱: Thermo HYPERCARB色谱柱(3.0 mm×100 mm, 5.0 μm); 流动相洗脱程序: 0.0~0.5 min, 100% A; 0.5~5.0 min, 100%~90% A; 5.0~5.5 min, 90%~5% A; 5.5~6.5 min, 5% A; 6.5~7.0 min, 5%~100% A; 7.0~8.5 min, 100% A, 流速: 0.50 mL/min; 柱温: 40 ℃; 进样量: 10 μL[23]
(2)质谱条件
电离模式: 电喷雾正离子扫描(electrospray ionization, ESI)负模式; 离子源参数: 雾化气温度150 ℃; 雾化气流速: 15 L/min; 雾化器压力: 40 psi; 鞘气温度325 ℃; 鞘气流速11 L/min; 毛细管电压: 3500 V; 喷嘴电压: 500 V; 扫描模式: 多反应监测(multiple reaction monitoring, MRM); 化合物的离子(m/z)及质谱参数(去簇电压、碰撞能、加速电压等)见表2, 离子对的驻留时间均为20 ms[7-11,24]
采用MassHunter Workstation 10.1软件对样品进行采集分析, Quantitative Analysis 10.2软件对采集和检测结果进行数据处理分析。
氯酸盐和高氯酸盐属于强极性化合物, 取1 mL标准工作液到2 mL进样小瓶中, 使用两通连接液相色谱仪与质谱仪, 在负离子模式下进行全扫描, 确定化合物的分子离子, 用软件自带的自动优化软件(Optimizer)自动优化出两个子离子去簇电压和碰撞能量等参数, 2个子离子根据仪器响应分别作为定性及定量离子(内标1个响应最高的离子作为定量离子), 也可用手动优化的模式, 按照全扫描→子离子扫描→MRM模式逐步优化化合物的仪器参数[24-25], 氯酸盐和高氯酸盐质谱参数优化结果见表2
选用专门针对极性化合物的色谱柱Thermo HYPERCARB (3.0 mm×100 mm, 5.0 μm), 在酸性体系下, 氯酸盐和高氯酸成分子态, 分别验证0.1%甲酸水-甲醇、0.1%甲酸水-乙腈、0.1%甲酸-5 mmol/L乙酸铵水-甲醇、0.5%甲酸水-0.5%甲酸甲醇4种流动相体系对分析效果的影响[26], 通过色谱峰响应及分离效果来看, 采用0.5%甲酸水-0.5%甲酸甲醇作为流动相时, 氯酸盐及高氯酸盐的离子化效率高, 峰型好, 灵敏度高, 目标物标准溶液(10 μg/L)的总离子流图和提取离子色谱图见图1
氯酸盐和高氯酸盐为强极性化合物, 易溶于水, 不溶于三氯甲烷、正己烷等试剂, 本研究用阳性辣椒粉样品, 对比了以1%乙酸乙腈-水、1%乙酸-水、1%甲醇-水作为提取溶剂的提取效果。结果显示, 以水或者甲醇-水提取时, 辣椒粉会分散浮在上中下层, 提取液不澄清, 难以离心分离或过滤膜, 以乙腈-水混合液提取时提取时, 提取液较澄清, 阳性样品的稳定性及结果最佳, 但不同比例1%乙酸乙腈-水混合提取液, 阳性样品的结果也有差距, 主要跟1%乙酸乙腈-水提取效果有关系, 比较不同比例的1%乙酸乙腈-水的提取效果见表3, 1%乙酸乙腈-水=1:1 (V:V)时回收率及提取效果最好。
本研究先以每月1000个样品为工作量, 每天工作8 h, 考察人工操作及全自动QuEChERS仪效率及精密度(结果见表4), 由表4可以看出, 仪器折旧成本与1位员工工资相当, 但是效率是人工的6倍, 若样品量增多, 其效率会倍数增加, 同时所测试数据的精密度要大于人工操作。
用1%乙酸乙腈-水=1:1 (V:V)流动相配制质量浓度为1、5、10、20、50、100 ng/mL氯酸盐及高氯酸盐标准溶液进样分析, 记录色谱图, 氯酸盐线性方程为Y=0.0715X+0.139, 线性相关系数(r2)为0.9996, 高氯酸盐线性方程为Y=0.0473X+0.0032, 线性相关系数(r2)为0.9994, 线性关系良好。在空白样品中添一定浓度的混合标准溶液, 按照本方法用全自动QuEChERS仪处理后分析, 分别以3倍信噪比、10倍信噪比计算氯酸盐和高氯酸盐的检出限和定量限(limit of quantitation, LOQ)。得到方法检出限为3.0 μg/kg, LOQ为10.0 μg/kg, 可以满足方法灵敏度及出口欧盟限量要求。
选取辣椒粉样品, 按本研究方法进行3个加标水平分别测定6次(LOQ、2倍LOQ、5倍LOQ)的回收率和精密度实验, 计算其回收率和RSDs, 实验结果见表5。目标物的平均回收率为94.0%~96.4%, 日内RSDs均不高于5%, 表明方法的准确度和精密度良好[24,27-32]
采用用本方法对山东地区10批次辣椒粉样品进行检测, 发现阳性样品8例, 检出范围氯酸盐10~80 μg/kg之间, 高氯酸盐在10~200 μg/kg之间, 同时将10批次样本, 用国家标准检测氯酸盐高氯酸盐的方法进行检测, 所得到的的检测结果与全自动QuEChERS仪测定结果精密度均小于10%, 说明本方法对样品中目标物的定量分析可信度较高。
本研究建立了全自动QuEChERS-UPLC-MS/MS快速测定辣椒粉中的氯酸盐及高氯酸盐的方法, 与传统的人工操作相比, 全自动QuEChERS仪替代人工更加符合现在科技高速发展的现状, 经优化前处理、色谱等条件, 目标化合物分离度好, 结果准确可靠, 有较高的精密度及回收率, 更加适用于辣椒粉中氯酸盐和高氯酸盐的检测。
