In recent years, mycotoxin contamination in food has become an important food safety issue with global concern. Mycotoxins are a type of secondary metabolites produced by fungi with characteristics of high toxicity, difficult decomposition and chemical stability. It is of great significance to establish an efficient and safe mycotoxin detoxification technology applicable to food. Based onenzyme catalysis, the biological detoxification technology has the advantages of high substrate specificity, mild conditions, green environment, which can overcome the damage of traditional physical and chemical detoxification technology to the quality and nutritional value of food. Immobilized enzyme technology can improve the catalytic performance of enzymes, endow them with values such as stability, reusability, and high throughput, as well as overcome the limitations of biological enzymes in the application process. This article reviewed the application of immobilized enzyme technology in food mycotoxin removal, analyzed the deficiencies of the current immobilized enzyme technology applied in food detoxification and possible future research directions, with a view to providing research ideas for the construction of more efficient immobilized enzyme catalysts.

近年来, 食品中真菌毒素的污染已成为全球关注的重要食品安全问题。真菌毒素是一类由真菌产生的次生代谢产物, 具有毒性强、难分解、化学性质稳定的特点。建立适用于食品体系的高效安全的真菌毒素脱毒技术具有重要意义。基于酶催化的生物脱毒技术具有底物特异性强、条件温和、绿色环保等优点, 可以克服传统物理化学脱毒技术对于食品品质和营养价值的破坏。固定化酶技术可以提高酶催化性能, 赋予酶稳定性、可重复使用性、高通量性等价值, 克服生物酶在应用过程中的局限性。本文综述了固定化酶技术在食品真菌毒素脱除中的应用, 分析了目前固定化酶技术应用在食品脱毒过程中的不足及未来可能的研究方向, 以期为构建更加高效的固定化酶催化剂提供研究思路。

近年来, 食品中真菌毒素的污染已成为全球关注的重要食品安全问题。真菌毒素是一类由真菌产生的次生代谢产物, 具有毒性强、难分解、化学性质稳定的特点。建立适用于食品体系的高效安全的真菌毒素脱毒技术具有重要意义。基于酶催化的生物脱毒技术具有底物特异性强、条件温和、绿色环保等优点, 可以克服传统物理化学脱毒技术对于食品品质和营养价值的破坏。固定化酶技术可以提高酶催化性能, 赋予酶稳定性、可重复使用性、高通量性等价值, 克服生物酶在应用过程中的局限性。本文综述了固定化酶技术在食品真菌毒素脱除中的应用, 分析了目前固定化酶技术应用在食品脱毒过程中的不足及未来可能的研究方向, 以期为构建更加高效的固定化酶催化剂提供研究思路。

In recent years, mycotoxin contamination in food has become an important food safety issue with global concern. Mycotoxins are a type of secondary metabolites produced by fungi with characteristics of high toxicity, difficult decomposition and chemical stability. It is of great significance to establish an efficient and safe mycotoxin detoxification technology applicable to food. Based onenzyme catalysis, the biological detoxification technology has the advantages of high substrate specificity, mild conditions, green environment, which can overcome the damage of traditional physical and chemical detoxification technology to the quality and nutritional value of food. Immobilized enzyme technology can improve the catalytic performance of enzymes, endow them with values such as stability, reusability, and high throughput, as well as overcome the limitations of biological enzymes in the application process. This article reviewed the application of immobilized enzyme technology in food mycotoxin removal, analyzed the deficiencies of the current immobilized enzyme technology applied in food detoxification and possible future research directions, with a view to providing research ideas for the construction of more efficient immobilized enzyme catalysts.

近年来, 食品中真菌毒素的污染日益严重, 已成为全球广泛关注的重要问题。真菌毒素是一类由真菌产生的强毒性次生代谢产物[1]。青霉、曲霉和镰刀菌等特定种属的真菌易侵染谷类、豆类和水果等农作物, 并在适宜的环境条件下快速繁殖产生和积累真菌毒素。根据联合国粮食及农业组织(Food and Agriculture Organization of the United Nations, FAO)的统计数据, 世界上每年约25%的农产品受到真菌毒素的污染[2-3]。真菌毒素具有毒性强、难分解、化学性质稳定的特点, 常规的食品加工和烹饪过程难以将其彻底消除。真菌毒素通过直接或间接的途径进入人体后, 具有强致癌、致畸、致突变的危害, 对人类健康构成严重威胁[4-5]
当前, 食品工业中应用最广泛的真菌毒素脱除技术主要包括吸附法、热处理、辐射处理等的物理脱毒技术和利用酸、碱、氧化剂等化学试剂将真菌毒素转化成低毒甚至无毒分子的化学脱毒技术[6-8]。这两种技术虽然可以解决传统食品加工方法无法彻底有效脱除真菌毒素的问题, 但是它们缺乏特异性且过程不可控, 在处理真菌毒素的同时, 会降低食品营养价值, 破坏食品品质[9]。基于酶催化的生物脱毒技术具有特异性强、反应条件温和、绿色环保等优点, 对食品营养成分和风味物质破坏少, 是一种具有广阔应用前景的食品真菌毒素脱毒技术[10-11]。然而, 游离的生物酶, 活性不稳定、保质期短、分离困难、不可重复使用且对于多种生产工艺条件极为敏感, 限制了其工业应用[12]
固定化酶技术, 通过吸附、共价结合等方式, 将酶固定于惰性或不溶性载体上, 可以提高酶催化性能、增强稳定性并实现重复使用, 有效克服生物酶在应用过程中的局限性, 已广泛用于食品加工过程, 为生物酶技术在食品脱毒中的有效应用提供了一种策略[13-14]。因此, 本文综述了固定化酶技术在食品真菌毒素脱除方向中的应用, 分析了目前固定化酶技术应用在食品脱毒过程中的不足及未来可能的研究方向, 以期为更加高效的固定化酶催化剂的开发及在食品中脱除真菌毒素的应用提供研究思路。
固定化酶技术中, 载体的选择对酶催化性能具有重要影响。载体一般具有如下性质: 高比表面积、大孔隙、优异的生物相容性、高稳定性、不与酶活性位点作用、成本低廉、绿色环保等[15]。常见的酶固定化载体一般多为天然存在或人工合成的多孔材料、磁性纳米材料和天然生物聚合物材料等, 此外, 还包括兼具多种性能的复合材料[12-16]
多孔材料是一种由相互贯通或封闭的孔洞构成网络结构的材料, 按照孔的直径分类, 主要分为: 微孔材料(小于2 nm), 介孔材料(2~50 nm)和大孔材料(大于50 nm)[17-18]。多孔材料孔道丰富, 比表面积大, 为酶的固定化提供了更多的结合位点, 此外, 其较强的吸附能力, 能够在一定空间范围内聚集高浓度的底物以加速酶促反应。同时, 为增强酶与载体的相容性和亲和力, 研究者对载体进行适当的改性, 例如表面用不同的分子功能化, 从而增强固定化酶的活性和稳定性。
介孔材料的孔道分布有序且可连续调节, 相比于微孔材料, 比表面积较大, 孔隙分布窄, 稳定性好, 吸附能力强, 因此被广泛开发用于酶固定化技术。其中, 硅基介孔材料应用广泛, 主要包括KIT-6、MCF和MCM-41等[19-20]。最近研究报道中, 二氧化硅可以从天然来源的农业废弃物如稻壳中混合碱液煅烧提取, 并用于形成二氧化硅基介孔纳米颗粒, 进一步作为载体固定酶用于催化反应, 具有成本较低、环保和可持续的优势。通过在硅表面修饰—NH2、—SH、—CN、—Cl、—C6H5及不同链长烷基等有机功能基团进行硅烷改性, 利用其提供的大量反应位点, 与酶形成更加稳定的共价连接或次级键连接, 提高固载效率, 防止重复使用过程中酶的泄露[21-22]。MUÑOZ-PINA等[23]应用胺、羧酸、异氰酸酯、烷烃和吡啶5种官能团分别改性介孔二氧化硅材料UVM-7, 作为固定多酚氧化酶(polyphenol oxidase, PPO)的载体, 探究了其对于酶促褐变过程的影响。结果表明, 载体的功能化强烈影响酶的固定化, 除羧酸功能化外, 其余功能化的载体均可提供高PPO的负载能力并且固定速率随功能化而提高。在新鲜苹果汁中的验证表明, 胺功能化的固定化酶纳米材料兼具捕获氧化产物及PPO的能力, 能够在室温含氧的90 min内显著避免褐变的产生。
由金属离子和有机配体桥接构成的金属有机框架(metal organic frameworks, MOFs), 具有比表面积大、孔隙规则、结构可调等优点, 是固定化酶的一种重要载体[24-25]。KUANG等[26]在油酸与水的乳液体系中, 通过一锅法合成具有长链脂肪酸修饰的疏水性UiO-66 (UiO-66-NH2-OA), 用作米曲霉脂肪酶的固定化载体。合成的疏水性固定化脂肪酶材料(AOL/UiO-66-NH2-OA)展现出出色的酶催化活性。
磁性材料主要由磁性成分(如铁)和化学功能剂组成, 具有比表面积大、机械强度高和易化学改性的特点。利用磁性纳米材料作为酶的固定化载体, 可以通过外部磁场作用, 可控地分离和回收酶催化剂[27-28]。在实际应用过程中, 磁性材料表面功能化改性或包覆涂层, 可以解决纳米粒子表面能大、表面活性高, 以及粒子之间磁偶极矩作用导致的团聚等问题, 进而提高分散性和稳定性[29]。常用的改性和包覆材料主要包括高分子聚合物(如壳聚糖、麦芽糖、纤维素、聚合多巴胺、淀粉、透明质酸等)、硅烷等有机硅化物和一些小分子金属离子(如Co2+、Cu2 +等)[29-31]。VITOR A等[32]首次将嗜热菌蛋白酶(thermolysin, TLN)固定在胺化超顺磁性纳米颗粒(Fe3O4@silica-NH2)上, 可高效蛋白水解牛奶酪蛋白和牛血清白蛋白(bovine serum albumin, BSA)。开发的纳米生物催化剂(Fe3O4@silica-TLN)具有良好的稳定性、可恢复性、可重复使用性和适用性。Fe3O4@silica-TLN在60 °C下暴露48 h, 在4 °C下储存80 d, 在室温下保存42 d后, 仍保持较好的活性。此外, Fe3O4@silica-TLN可作为一种稳定且可回收的牛奶凝固微生物酶, 在应用中重复使用长达11次循环, 并且在24 h内对牛乳酪蛋白的蛋白水解活性提供了84种肽, 其中29种具有潜在的生物活性。
淀粉、甲壳素、藻酸盐、角叉菜胶、壳聚糖和聚丙烯酰胺等天然来源的高分子聚合物, 与其他载体材料相比, 具有良好的生物相容性、生物稳定性等生物功能优势, 并且资源丰富, 易于降解回收, 具有绿色环保和成本低廉的经济效益, 是固定化酶的优良载体材料[33-36]。DE-OLIVEIRA等[37]将来自棘曲霉的果胶酶掺入藻酸盐微胶囊中, 并用于在填充床反应器中连续工艺下澄清苹果汁和巴西梅汁。固定化酶催化剂具有高催化效率和操作稳定性, 在生物反应器中重复使用循环3次后, 仍保留80%以上催化活性。DAI-MAGRO等[38]通过将商业酶制剂固定在戊二醛官能化的壳聚糖微球, 建立连续填充床和流化床反应器, 应用于橙汁的澄清, 连续使用72 h后保留了60%的催化活性。
复合材料是由两种或更多种不同的材料通过物理、化学或机械方法结合而成的多组分材料, 通常会表现出单一组分的材料所无法具备的优异性能[39]。一方面, 复合材料通过内部不同组分的相互作用, 导致物理、化学和机械性能, 包括力学性能、导电性与导热性和光学性能等更加出色。另一方面, 复合材料可以将不同材料的优势特性结合在一起, 使多种功能集成于一个材料内[40]。此外, 复合材料通常在化学稳定性、热稳定性和机械稳定性方面优于单一组分材料, 并且通过精确调控不同组分的比例、形态、分布, 可以实现其性能的调节, 以满足特定应用的需求。
在固定化酶的应用中, 通过将不同类型的催化剂材料(如贵金属纳米颗粒、金属氧化物、生物聚合物和碳基材料等)组合, 可以基于协同催化效应而显著提高催化反应的效率、选择性和稳定性。并且扩展催化反应在高温、高压或极端pH条件下仍能维持高催化活性, 扩展催化范围[39,41-42]
合适的固定化酶方法是实现最终固定化酶材料高效催化效果的另一重要方面。常用的固定化酶方法包括吸附、包埋、交联和共价结合[43]。其中, 物理吸附法是最简单的固定策略, 涉及各种非共价相互作用, 包括范德华力、疏水相互作用、氢键和盐键。这种方法不仅成本较低, 操作简单, 反应时间短, 并且不需要活化剂和交联剂, 避免了酶的构象被破坏, 防止酶的团聚[44-45]。但是, 在应用过程中可能会面临酶浸出以及容易接触环境等问题, 固定效率低, 酶构象变化严重。共价结合是目前研究和应用最广泛的固定策略[43]。酶的氨基酸残基与载体表面的巯基(-SH)、羧基(-COOH)、氨基(-NH2)等官能团通过共价键结合, 从而实现酶的稳定固定。与物理吸附法相比, 共价结合法的酶与载体相互作用力更强、固定化效率更高。然而, 一般的共价桥接过程操作步骤相对复杂, 所需孵育时间较长, 且酶表面化学修饰可能破坏酶的结构、降低酶的活性[43-47]
目前, 固定效率低、固定程序复杂和可重复性低是食品中固定化酶策略亟待解决的严重问题[43]。因此, 开发新的固定化酶载体和固定化策略以适应食品反应环境具有十分重要的意义。静电纺丝、电喷雾、增材制造技术(3D打印技术)已成为新型固定化酶的合成方法, 具有简单、经济高效等优点, 在食品工业的固定化酶催化剂制备中具有广阔应用前景[48-49]
真菌毒素作为食品污染的主要来源之一, 已成为影响食品安全的重要因素, 引起全世界的广泛关注[1-2]。农作物及其食品制品中, 主要存在的5种真菌毒素为: 黄曲霉毒素(aflatoxins, AFs)、玉米赤霉烯酮毒素(zearalenone, ZEN)、脱氧雪腐镰刀菌烯醇(deoxynivalenol, DON)、赭曲霉毒素(ochratoxin, OTA)和展青霉毒素(patulin, PAT)[50-51]。随着生物法在食品真菌毒素脱除领域的兴起, 利用酶去除食品中的真菌毒素具有特异性强, 安全性高的优势。然而, 在实际应用中, 天然酶不易回收、不便于连续化操作, 成本较高。此外, 天然酶易受食品复杂基质的影响, 稳定性低。因此, 提高酶的稳定性和回收性、增强酶催化效率的固定化酶技术在食品真菌毒素脱毒中的应用逐渐开始受到研究者的广泛关注。目前, 将固定化酶应用于食品真菌毒素脱毒的研究多集中于AFB1、ZEN、PAT等毒素, 对于OTA和DON的研究较少。
黄曲霉、寄生曲霉和集峰曲霉等真菌易侵染花生, 小麦, 豆类等农作物, 在温暖潮湿的环境下, 快速繁殖并分泌积累次级代谢产物AFs[52-53]。其中, 黄曲霉毒素B1 (aflatoxins B1, AFB1)分布最广且毒性最强, 可以随各种食品加工链条进入食用油、面粉等制品中[54-55]。为了增强酶在油水体系中的分散性和稳定性, 实现酶在食用油中高效去除AFB1, LU等[56]制备了一种具有双亲性结构的漆酶-无机杂化纳米花(laccase-inorganic hybrid nanoflowers, Lac NF-P), 用于花生油中AFB1的去除。首先, 利用五水合硫酸铜和含有漆酶的磷酸盐缓冲液搅拌形成无机纳米花Lac NF, 通过吸附和共价结合将漆酶固定在无机纳米花NF的内部和表面。随后利用交联剂聚合物伴刀豆球蛋白A (concanavalin A, Con A), 将双亲性聚合物(PEO−PPO−PEO, Pluronic F127)偶联在Lac NF上以赋予漆酶在油水两相中的高分散性和强稳定性。同时, 单宁酸(tannic acid, TA)作为氧化还原介质, 被漆酶氧化形成的阳离子自由基进一步氧化AFB1, 提升漆酶的降解效率。研究表明, Lac NF-P对于AFB1的降解效率分别比laccase和Lac NF提高了134倍和3.2倍, 并且在3 h内可将花生油中的AFB1去除至0.96 μg/kg以下, AFB1可去除的浓度范围为50~150 μg/kg。此外, 花生油的品质没有明显变化, 处理Lac NF-P后未观察到催化剂泄漏。因此, 两亲性固定化酶催化剂的设计可以保留天然酶较高的活性, 实现酶在食用油及乳类食品中去除真菌毒素。同时, 漆酶-氧化还原介体体系的构建可以提高漆酶对于AFB1的降解效率。
结合磁性固定化酶材料便于回收利用的优点, FU等[57]提出了一种利用磁性固定化酶靶向去除花生油中AFB1的新策略。通过制备两亲性共价有机框架(covalent organic frameworks, COFs)作为限制性纳米反应器, 用于区隔化共固定葡萄糖氧化酶(glucose oxidase, GOx)和磁性纳米颗粒(Fe3O4), 实现了非均相类芬顿反应与葡萄糖氧化酶催化反应相结合级联去除花生油中的AFB1。通过化学-生物级联反应, 克服了传统芬顿反应的限制, 一方面, H2O2由GOx原位持续生成, 防止羟基自由基(•OH)淬灭, 提高H2O2利用率; 另一方面, 酶促反应生成的葡萄糖酸, 调控反应微环境的pH以增强金属催化的活性。两亲性生物催化剂(PL-GOx-Fe3O4@COF)的GOx活性是游离型GOx的300%, 催化效率是游离型GOx和Fe3O4混合体系的6倍, 并且重复利用8次后, 剩余活性依然保持68%, GOx无明显泄漏。此外, PL-GOx-Fe3O4@COF具有良好的储藏稳定性, 在pH=7.4的磷酸盐缓冲液中储存30 d后, PL-GOx- Fe3O4@COF的活性保留了78%, 而游离GOx的活性仅保留32%。反应后, 花生油的品质无明显变化, 未观察到催化剂的泄露。构建的PL-GOx-Fe3O4@COF可以高效、绿色地脱除植物油的霉菌毒素, 采用化学-生物耦合催化, 提高了真菌毒素降解效率。此外, 磁性颗粒的负载解决了固定化酶催化剂在食用油等食品体系中难以分离及回收利用困难的问题。
RASHEED等[58]在废蘑菇基质中(spent-mushroom- substrate, SMS)衍生出生物炭(biochar, BF), 通过共价交联的方法将漆酶固定化在NH2活化的磁性生物炭(BF-NH2)上, 设计了一种可磁分离的新型固定漆酶的生物催化剂(BF-NH2-Lac)。在4 °C下保存30 d后, 固定化漆酶保持95%的初始活性。在香豆酸作为介体激活漆酶降解AFB1的测试中, BF-NH2-Lac在5 h内, 对AFB1的去除率大于90%。玉米油和缓冲液中的降解效率相当, 多种霉菌毒素的共存对AFB1的降解没有明显影响, 并且处理后的玉米油质量未受影响。因此, 基于天然生物来源的固定化酶支持材料结合磁性支持材料可分离的优势, 可以在实现天然酶回收利用的同时, 提高固定化酶材料在食品体系脱除真菌毒素过程中的安全性, 并且降低催化剂成本, 便于后续工业化生产。
ZEN由镰刀菌产生, 常发生在发霉的大麦、玉米和小麦等谷物及制品中的非甾体雌激素霉菌毒素, 此外, ZEN可以从受污染的饲料转移到动物源性食品中, 如牛奶、肉类和鸡蛋[57-62]。研究表明, 来源于白腐菌和细菌的漆酶可以降解ZEN。GUO等[63]发现来自B. licheniformis的孢子CotA漆酶具有ZEN氧化酶活性, 并且2,2’-联氮基双-(3-乙基苯并噻唑啉-6-磺酸)[ABTS]、乙酰丁香酮、阿魏酸、咖啡酸和丁香酸甲酯等氧化还原介质的存在可以增强CotA漆酶对ZEN的降解。CotA漆酶表面的氨基与戊二醛功能化的壳聚糖微球共价结合后, 得到CotA漆酶-壳聚糖微球。固定化漆酶的热稳定性和对ZEN的催化效率明显提高, 在80 °C热处理30 min后仍保持87%的初始活性, 对玉米粉中ZEN的去除率由70%提升至90%。同时, 固定化CotA漆酶生物催化剂具有良好的可重复利用性, 连续3次循环后, 在玉米粉中仍可实现54%的ZEN降解。
花状多孔纳米材料可以提高固定化酶催化剂与真菌毒素的接触面积, 提供更多酶吸附位点, 可以显著提高降解效率。ZHOU等[64]构建了具有花状结构的玉米赤霉烯酮内酯酶-无机杂交纳米花(zearalenone lactase-inorganic hybrid nanoflowers, InHNF-ZHD518)。研究发现, 与游离酶相比, 固定化的玉米赤霉烯酮内酯酶比活性提高了40%~60%, 即使在极端pH条件下(pH 3~11)也能防止酶变性, 并且在8个反应循环后, 其残余活性超过70%。此外, 在啤酒样品的ZEN解毒过程中, 游离ZHD518的活性基本丧失, 但InHNF-ZHD518仍实现了50%以上的ZEN降解。由此可见, 通过该固定化酶技术负载的玉米赤霉烯酮内酯酶ZHD518相比于游离酶表现出显著提高的pH稳定性、增强的比活性和优异的可重复使用性, InHNF可以实现食品酶的环保、无纯化和定点固定, 并增强其催化性能, 适用于广泛的工业应用。
水果污染的主要因素之一是腐烂水果产生的PAT的积累。由于PAT的酸稳定性强, 极易通过食品加工进入果汁产品[65]。食用受PAT污染的果汁, 会导致肠道黏膜炎症和器官损伤等严重的健康后果[66-67]。LIU等[68]制备了磁性纳米晶纤维素(microcrystalline cellulose, MNCC), 用作猪肝酯酶(porcine liver, PLE)固定化的支持载体, 合成了一种新型基于纤维素的磁性棒曲霉素解毒剂(PLE@MNCC)。研究表明, 相比于游离PLE, 固定化后的PLE具有更好的稳定性, 45~55 °C的温度下, 棒曲霉素的解毒率仍保持在65%。PLE@MNCC可以通过生物降解和吸附有效地从苹果汁中解毒棒曲霉素。棒曲霉素的解毒率随PLE@MNCC含量的增加而显著增加。当PLE@MNCC含量从5 mg/mL增加到15 mg/mL时, 解毒率从8.85%显著提高到43.73%, 处理72 h后, 棒曲霉素的解毒率达到81.20%。PLE@MNCC重复使用8次后, PLE仍可以保留大约50%的初始活性。并且具有良好的磁回收性, 实现了应用后的快速分离。
锆-金属有机框架材料(Zr-MOF)具有酸稳定性高、比表面积大、官能团丰富、孔径可调等特点, 作为固定化酶支持材料具有显著优势。因此, YAN等[67]使用非原位方法和自组装策略, 定制了一种掺入Zr-MOF, 并由半胱氨酸(cysteine, Cys)和猪胰脂肪酶(pancreatic lipase, PPL)修饰的分级介孔结构的羧甲基纤维素气凝胶(CMC@HMMOF-Cys/PPL)。最佳CMC@HMMOF- Cys/PPL气凝胶表现出高吸附容量(38.41 μg/mg)、优异的降解能力(10 μg/mL PAT时为28.68 μg/mg)、良好的可重用性和储存稳定性以及更高的选择性。相比于磁性固定化酶催化剂, CMC@HMMOF-Cys/PPL气凝胶不仅可以提高回收利用效率, 并且丰富的通道可以提高天然酶的装载率, 提供了巨大的反应面积, 此外, 气凝胶类固定化酶催化剂可以定制为流动反应器的填充剂, 实现高通量地去除苹果汁中的PAT。
OTA是由曲霉属和青霉属产生的主要真菌毒素。OTA污染常见于葡萄酒和葡萄产品, 并可能导致营养摄入障碍和经济损失。在最近的研究中, 胰酶因其卓越的解毒效率和对葡萄酒质量的影响最小而被确定为一种重要的酶[69-70]。WANG等[71]将胰酶固定在聚多巴胺功能化的磁性多孔壳聚糖(MPCTS@PDA)表面以降解OTA。壳聚糖(chitosan, CTS)是一种天然多糖, 具有优异的生物相容性、生物降解性和抗菌性能, 具有多孔结构的壳聚糖, 孔隙率高, 比表面积大, 提供了对酶分子的强亲和力和高负载能力。同时, 聚多巴胺(polydopamine, PDA)涂层膜中存在邻苯二酚和醌基, 易与含有巯基或胺基的分子反应。通过PDA功能化CTS, 无需额外的交联剂即可实现酶的固定, 简化了固定酶的过程。经研究发现, 超声辅助方法能够显著改善胰酶的固定化。在优化的固定化条件下, 胰酶负载量为94 mg/g, 活性恢复率为87%。与游离胰酶相比, MPCTS@PDA@胰酶在17~47 °C之间的热稳定性显著提高, 在3.0~7.0的宽pH范围内表现出更高的稳定性。在4 °C和25 °C下储存21 d后, 其初始活性分别保持在约94%和43%, 在8个回收周期中始终保持较高的相对活性, 可重复利用性能优异, 表明MPCTS@PDA@胰酶在稳定性和性能方面明显优于游离胰酶。此外, MPCTS@PDA@胰酶在葡萄酒中的解毒率达到58%, 并且对于葡萄酒的质量影响小。表面涂覆聚多巴胺的固定化酶技术, 可以简化天然酶负载过程, 减少酶固定化的交联步骤对酶催化活性的影响, 对于固定化酶催化剂更高效地降解食品中真菌毒素具有重要意义。
固定化酶技术已在食品加工领域广泛应用, 并逐渐在食品真菌毒素的脱除方向兴起。生物酶依托吸附、包埋、交联和共价连接等方式固定在各种具有优异性能的支撑载体上, 一方面, 可以提高自身的生物催化活性, 另一方面, 借助载体的各种独特性能, 能够保持较高的稳定性、可重复使用性、高通量性等优势。目前, 用于食品加工中的固定化酶载体主要是天然存在或人工合成的多孔材料, 磁性纳米材料、天然生物聚合物材料和兼具多种性能的复合材料。在脱除食品真菌毒素的方向, 其中, 固定化酶技术已可以安全高效地去除部分食品中污染最严重的AFB1、ZEN、PAT 3种毒素。然而, 对于OTA和DON的研究较少。此外, 部分固定化酶载体存在生物相容性较低、合成步骤烦琐, 以及无法满足工业生产的需求的问题, 是提高其在食品中脱毒效率的关键。未来, 考虑到在食品体系应用固定化酶技术的局限性, 可能的研究方向如下: (1)开发生物相容性好、对人体无毒副作用、绿色经济的新型酶固定化载体。(2)选择合适的酶固定化策略, 探究酶与载体间相互作用的机制。(3)结合各种载体的优势。在提高酶负载率的同时, 寻求双载体或多载体、双酶或多酶的共固定方式以发挥协同作用, 同时去除多种毒素或提高食品加工过程中的催化效率。
