Objective To establish a method for the determination of ethylenediaminetetraacetic acid ferric sodium salt (NaFeEDTA) by microemulsion liquid chromatography. Methods After simple dilution and centrifugation of the sample, it could be filtered through a 0.22 μm microporous membrane and directly loaded onto a Waters BEH C18 column (2.1 mm×150 mm, 1.7 µm) was the chromatographic column. The microemulsion mobile phase was 3.64 g/L cetyltrimethylammonium bromide-50 g/L acetonitrile-8 g/L ethyl acetate-6.8 g/L potassium dihydrogen phosphate. The flow rate was 0.30 mL/min and the detection wavelength was 254 nm. The column temperature was 30 ℃ temperature. External standard method was used for quantitative determination. Results Under the selected experimental conditions, the mass concentration of NaFeEDTA had a good linear relationship (r≥0.999) within the range of 0.50-35.00 µg/mL, the limits of detection were 50 mg/kg. The recovery rates of NaFeEDTA were investigated, with average recovery rates ranging from 92.7% to 97.7%. Conclusion This method is easy to operate and has good reproducibility. The method can be used for the detection of NaFeEDTA content in soy sauce.

目的 建立微乳液相色谱法测定酱油中乙二胺四乙酸铁钠(ethylenediaminetetraacetic acid ferric sodium salt, NaFeEDTA)含量的分析方法。方法 样品经简单的稀释和离心, 通过0.22 μm微孔滤膜进行过滤, 即可直接上机, 以Waters BEH C18柱(2.1 mm×150 mm, 1.7 µm)为色谱柱, 以3.64 g/L十六烷基三甲基溴化铵水溶液-50 g/L乙腈-8 g/L乙酸乙酯-6.8 g/L磷酸二氢钾水溶液为流动相, 流速为0.30 mL/min, 检测波长为254 nm, 柱温为30 ℃, 使用外标法进行定量测定。结果 在选定实验条件下, NaFeEDTA质量浓度在0.50~35.00 µg/mL范围内具有良好的线性关系(r≥0.999), 检出限为50 mg/kg。考察了NaFeEDTA回收率, 其平均回收率分别为92.7%~97.7%。结论 该方法操作简单、重现性好, 可用于酱油中NaFeEDTA含量的检测。

Objective To establish a method for the determination of ethylenediaminetetraacetic acid ferric sodium salt (NaFeEDTA) by microemulsion liquid chromatography. Methods After simple dilution and centrifugation of the sample, it could be filtered through a 0.22 μm microporous membrane and directly loaded onto a Waters BEH C18 column (2.1 mm×150 mm, 1.7 µm) was the chromatographic column. The microemulsion mobile phase was 3.64 g/L cetyltrimethylammonium bromide-50 g/L acetonitrile-8 g/L ethyl acetate-6.8 g/L potassium dihydrogen phosphate. The flow rate was 0.30 mL/min and the detection wavelength was 254 nm. The column temperature was 30 ℃ temperature. External standard method was used for quantitative determination. Results Under the selected experimental conditions, the mass concentration of NaFeEDTA had a good linear relationship (r≥0.999) within the range of 0.50-35.00 µg/mL, the limits of detection were 50 mg/kg. The recovery rates of NaFeEDTA were investigated, with average recovery rates ranging from 92.7% to 97.7%. Conclusion This method is easy to operate and has good reproducibility. The method can be used for the detection of NaFeEDTA content in soy sauce.

铁是以功能铁和贮存铁的形式存在于人体内, 主要有血红蛋白铁、肌红蛋白铁、含铁酶、铁蛋白、含铁血黄素等, 成年人体内约有4~5 g铁, 它影响人体的代谢、免疫和白细胞的杀伤力。铁缺乏症是指人体缺乏足够的铁元素, 从而导致身体无法正常运作的一种疾病, 是全球性的营养缺乏性疾病。全球共有20多亿人口缺铁, 摄食铁强化食品通常被认为是改善铁营养状况有效且低成本的路线, 是目前解决这一问题最有效可行的办法。传统铁营养强化剂的应用难题在于生物利用度和稳定性不能兼顾, 易引起不良感官变化等问题, 而乙二胺四乙酸铁钠(ethylenediaminetetraacetic acid ferric sodium salt, NaFeEDTA)是一种性质稳定的络合型铁营养强化剂, 易溶于水、铁锈味较小、胃肠刺激小[1-2]且人体吸收率高[3-5], 还可以促进内源性铁源的吸收。NaFeEDTA安全无毒副作用, 被认为是目前最具发展前景的铁营养补充剂[6], 2002年被批准在酱油中使用[7-9], 食用铁强化酱油成为改善铁营养状况的有效途径[10]
GB 14880—2012《食品安全国家标准 食品营养强化剂使用标准》标准中规定: 营养强化剂铁在酱油中使用量为180~260 mg/kg, 折算成NaFeEDTA为1357~1960 mg/kg。食品中元素铁的检测方法如原子吸收法、原子发射光谱法、质谱法、溶出伏安法等, 无法对NaFeEDTA和焦磷酸铁、硫酸亚铁等各种铁强化剂进行直接检测各化合物。NaFeEDTA是Fe3+与EDTA所形成络合物的钠盐, 在中性和弱酸性溶液中通常以Na+和FeEDTA-形式存在[11]。目前, 强化酱油中NaFeEDTA的定量测定的方法报道较多, 主要包括分光光度法[12-13]、毛细管区带电泳法[14-15]、极谱法[16-17]、离子色谱法[18-19]、高效液相色谱法[20-24]等, 其中高效液相色谱法目前存在不足: (1)在水相中添加离子配体, 而添加离子对试剂的流动相平衡较慢, 稳定时间长, 否则化合物保留时间会有偏移; (2)使用离子对试剂不适合使用梯度洗脱。
微乳液相色谱法(microemulsion liquid chromatography, MELC)是一种新的色谱形式, 是使用微乳为流动相的一种反相液相色谱法。微乳是由表面活性剂、助表面活性剂、油相和水相按适当比例混合而成的, 具有无色透明、各向同性、低黏度等特点, 属于热力学稳定体系。与传统高效液相色谱法方法相比, MELC流动相含有机物少, 对环境比较友好。常使用的表面活性剂有十二烷基硫酸钠(sodium dodecyl sulfate, SDS)、十六烷基三甲基溴化铵(cetyltrimethylammonium bromide, CTAB)、聚氧乙烯月桂醚和十二烷基二甲基甜菜碱[25], 且所使用的表面活性剂CTAB属于季铵化合物, 具有生物降解性, 可以通过不同途径进行降解[26]。在MELC中具有固定相、水相和微乳流支相3个界面和3个分配系数, 因此可以获得较高的灵敏度和选择性, 直接进样就可以定量测定不同复杂基质的样品, 解决了许多难以解决的分离测定问题。近年来, MELC已应用于多组分的分离、药物及杂质的分离与生物样品分析检测[27-29], 但关于MELC在复杂基质酱油检测中的应用却鲜有报道。
酱油是中国传统的调味品, 由粮食发酵制成, 制作时用大豆、小麦或麸皮, 加入水、食盐酿造, 经细菌发酵而生成, 其色泽呈红褐色, 有独特酱香, 滋味鲜美, 其成分也极为复杂, 有食盐、氨基酸、糖类、香料、色素、防腐剂和有机酸等。在传统高效液相色谱法中, 样品不经过前处理, 易导致严重的干扰并损害色谱柱。而在MLEC中的表面活性剂具有极强的增溶能力, 支持各类样品直接进样测定, 从而简化样品制备过程[30]。MLEC允许生物样品不经过除蛋白质、富集、浓缩等样品前处理过程, 用流动相稀释过滤后直接注入色谱系统[31]。这是由于溶质、固定相、水相和微乳流动相之间的多种相互作用对许多化合物产生了独特的选择性。一方面, 微乳流动相的增溶作用可以使溶质以更快的速度在更短的时间洗脱; 另一方面, 在MELC中, 溶质之间的分离能力主要取决于溶质在3相之间的分配差异大小[32]和微乳流动相的极性大小, 这种相互作用使保留机制复杂化, 提高了分离效率, 对于复杂样品可以使用MELC简化样品的前处理过程。因此, 本研究采用流动相直接稀释酱油样品, 运用MELC测定酱油中NaFeEDTA含量, 以期为酱油中NaFeEDTA的检测提供简单有效手段。
酱油样品全部为市售产品。
NaFeEDTA标准品(纯度98.4%, 北京坛墨质检科技股份有限公司); 甲醇、乙睛(色谱纯, 德国Merck公司); 乙酸乙酯、正己烷、正庚烷(色谱纯, 天津市四友精细化学品有限公司); 磷酸二氢钾(分析纯, 西陇科学股份有限公司); CTAB、SDS、磷酸、氢氧化钠(分析纯, 国药集团化学试剂有限公司)。
ACQUITY UPLC H-Class超高效液相色谱仪(配备二极管阵列检测器)、Waters BEH C18柱(2.1 mm×150 mm, 1.7 µm)(美国Waters公司); BSA223S电子天平(精度1 mg, 德国赛多利斯公司); TG16-WS台式高速离心机(上海卢湘仪离心机仪器有限公司); 0.22 μm微孔过滤膜(上海安谱实验科技股份有限公司)。
精确称取约1 g的酱油(精确到1 mg), 置于100 mL容量瓶中, 加入流动相溶液, 混匀, 转移至离心管中, 6000 r/min离心15 min后, 上清液经0.22 µm的微孔滤膜过滤后进样。
按照GB 5009.249—2016《食品安全国家标准 铁强化酱油中乙二胺四乙酸铁钠的测定》样品前处理, 称取混匀后试样2.50 g(精确到1 mg), 置于小烧杯中加甲醇水溶液(3:1, V:V)溶解, 多次洗涤烧杯并移入50 mL棕色容量瓶中, 加甲醇水溶液至刻度, 摇匀。于避光处静置50 min后, 滤纸过滤。吸取滤液5.00 mL, 置于50 mL棕色容量瓶中, 用水稀释至刻度, 摇匀, 过0.22 µm微孔滤膜过滤后备用。
1000 mg/L NaFeEDTA标准溶液的制备: 准确称取标准物质NaFeEDTA 100.0 mg于100 mL棕色容量瓶内, 加水溶解并定容至刻度, 混匀, 制成质量浓度为1000 mg/L的标准储备液, 备用。
标准工作溶液的制备: 吸取一定量的标准储备液于10 mL容量瓶中, 用纯水定容, 逐级稀释配制成质量浓度为0.5、1.0、5.0、10.0、20.0、35.0 μg/mL的混合标准工作溶液。
色谱柱: Waters BEH C18柱(2.1 mm×150 mm, 1.7 µm); 柱温: 30 ℃; 流速: 0.30 mL/min; 流动相: 3.64 g/L CTAB水溶液-50 g/L乙腈-8 g/L乙酸乙酯-6.8 g/L磷酸二氢钾水溶液; 进样量为10 μL; 检测器: 二极管阵列检测器(photo-diode array, PDA); 检测波长: 254 nm。
数据采用Origin pro 7.5软件进行分析, 并制表、绘制图形, 样品检测与加标实验, 每组实验设3个平行, 实验结果均以平均值表示。
用于制备微乳流动相的表面活性剂, 根据其种类可分为阴离子、阳离子、非离子和两性离子等类型, 最常用的阴离子表面活性剂是SDS, 阳离子表面活性剂是CTAB。在MELC中, 表面活性剂对固定相的吸附改变了固定相的理化性质, 可引起极性改变, 固定相与流动相、固定相与分析物之间的分配系数改变等。本研究在色谱条件一样的情况下, 分别考察了SDS、CTAB表面活性剂对色谱峰分离的影响。从图1中可以看出, 当使用SDS时, 目标物色谱峰几乎无保留, 很快被洗脱下来; 当使用CTAB作为表面活性剂时, 目标物色谱峰有保留, 分离较好。这可能的原因是在使用阳离子表面活性剂时, 带负电荷的溶质分子将会与带正电荷的固定相发生静电相互作用而被保留, 分析物NaFeEDTA是Fe3+与EDTA所形成络合物的钠盐, 在中性和弱酸性溶液中通常以FeEDTA-形式存在, 带负电荷, 故能发生静电相互作用而被保留, 所以选择CTAB进行后续实验。
在MELC中, 物质在微乳流动相、固定相和水三者间进行分配, 表面活性剂的浓度一定要大于其临界胶束浓度, 临界胶束浓度才能形成微乳液, 在较低浓度下, 分析目标物的保留时间过大; 浓度过高时, 将使流动相的黏度增加, 导致黏度效应, 柱压增高。CTAB临界胶束浓度为0.0013 mol/L, 因此, 选择超过临界胶束浓度以上的0.005、0.010、0.020和0.030 mol/L CTAB进行实验。从图2可以看出, 随着CTAB浓度的增加, 分析物的保留时间明显缩短, 这可能是随着CTAB浓度的增加, 被测物分配到微乳流动相的程度也增加, 化合物迅速从固定相中释放出来, 从而减少了溶质的保留时间, 因此在柱上的保留随之降低。综合考虑柱压力和样品分析时间, 选定CTAB浓度为0.010 mol/L, 即为3.64 g/L CTAB水溶液。
在MELC中, 因表面活性剂在固定相上的吸附减缓了传质速率, 降低了色谱柱柱效。故MELC的分离效率一般较小, 微乳流动相的溶解能力一般低于等体积有机溶剂, 甚至有些溶质在微乳液存在下不溶, 这时需用添加助表面活性剂。为了提高传质速率, 减少表面活性剂在固定相上的吸附, 提高色谱柱柱效, 常添加的助表面活性剂有正丁醇、异戊醇和乙腈等。本研究中选择了乙腈、正丁醇、异戊醇作为助表面活性剂。图3显示了不同助表面活性剂对色谱峰的影响, 从图3中可以看出, 当使用极性较小的助表面活性剂(异戊醇和正丁醇)时, 色谱峰拖尾, 分离效果不佳。当使用极性较大的助表面活性剂(乙腈)时, 目标分析物峰高而尖, 分析时间合适, 因此, 选择乙腈作为助表面活性剂。
为了得到助表面活性剂的合适浓度, 分别选择5%、10%乙腈为流动相进行实验。图4显示了流动相中不同浓度的乙腈对色谱峰分离的影响。结果显示, 乙腈浓度越高, 目标分析物出峰越快, 当乙腈质量分数为10%时, 目标分析物色谱峰与样品基质干扰峰接近; 当乙腈的体积分数为5%时, 此时与样品基质干扰峰分离较好, 分离度大于1.5。综合考虑样品分析时间及与干扰物的分离情况, 最终确定助表面活性剂的比例为5%, 即为50 g/L乙腈水溶液。
在色谱条件的优化中, 流动相的pH必须首先考虑。这是因为不同分析物的理化性质不同, 在不同pH的流动相中存在的形式不同。在中性条件下, 有部分FeEDTA-在流动相中发生水解反应以阴离子FeEDTAOH2-的形式存在, 引起组分的保留性质发生变化。因此, NaFeEDTA的分离需要在酸性条件下进行。在MELC中, 由于色谱柱固定相表面被阳离子表面活性剂CTAB吸附而带正电荷, 而NaFeEDTA在酸性条件下主要以FeEDTA-离子形式存在, 由于正负电荷之间相互吸引作用而被保留。在pH的优化中, 本研究选择pH范围为3.0~7.0, 微乳流动相按1.3.3进行配制, 配制好的微乳流动相用20%磷酸调节pH, 设置了5个梯度, 分别为3.0、5.0、6.0、7.0和4.3(不调pH)。色谱分离结果如图5所示, 在pH为4.3时, 目标分析物峰形较好且分析保留时间适宜。因此, 选择流动相pH为4.3。
微乳油相通常有乙酸乙酯、正己烷、正庚烷等, 增加油的含量, 流动相中有机相的比例增加, 溶质的保留时间降低, 油相的类型改变也能改变溶质的保留时间。油相的含量一般在1.2%(体积分数)内, 油相其最佳含量约为0.8%[33]。本研究使用正庚烷、正己烷时, 不能很好分离复杂样品中目标分析物, 分离效果不好, 尝试采用乙酸乙酯时, 取得较好结果, 因此选用常用浓度为0.8%的乙酸乙酯, 即为8 g/L乙酸乙酯。
MELC一般会使用0.005~0.050 mol/L的缓冲盐, 来达到改善峰形和保留的目的, 磷酸二氢钾可以控制流动相的pH并增大离子强度。本研究在流动相中分别添加了0.005、0.015、0.025 mol/L的磷酸二氢钾(图6), 当其浓度增加时, 分析物的峰高略有增加, 这可能是由于磷酸二氢钾浓度的增加提高了溶液的离子强度, 降低了微乳液表面的电势, 增强了溶质和微乳液的作用。磷酸二氢钾含量过大时, 柱压增高明显, 经综合考虑, 本研究中磷酸二氢钾的浓度选择0.025 mol/L, 即为6.8 g/L磷酸二氢钾水溶液。
综上所述, 通过实验优化, 得到了最佳的色谱条件: 3.64 g/L CTAB水溶液-50 g/L乙腈-8 g/L乙酸乙酯-6.8 g/L磷酸二氢钾水溶液为流动相、流动相pH=4.3, 柱温30 ℃。在最佳色谱条件下, 注入10 µL用流动相溶液配制的质量浓度为2.5 µg/mL标准溶液, 在0.30 mL/min的流速下, 在254 nm处得到的标准溶液色谱图, 如图7所示。
称取6份样品, 按1.3处理后进样检测, NaFeEDTA测得平均值为92 mg/kg, 相对标准偏差(relative standard deviation, RSD)为2.5%, 保留时间均在5 min左右, 保留时间RSD为0.10%, 说明该方法的重复性良好。分别吸取NaFeEDTA标准液(100 mg/L) 0.50、1.00、2.00 mL加入样品中, 按方法进行前处理、检测, 计算回收率和RSD。经检测计算, 样品回收率在88.0%~99.0%之间, 平均回收率为92.7%~97.7%。结果表明建立的方法回收率结果满意, 详见表1
在该方法的实验条件下, NaFeEDTA在0.50~35.00 µg/mL范围内呈良好线性关系, 通过计算峰面积(Y)与质量浓度(X, µg/mL)得线性方程为Y=2132.476X-994.957, 相关系数r=0.9997。同时, 根据标准得出最低检出浓度为0.50 µg/mL, 计算得出方法的检出限为50 mg/kg, 符合检测的要求。
购买不同品牌的市售酱油, 按GB 5009.249—2016和本研究方法进行样品前处理, 对不同品牌酱油中强化铁进行测定, 实验结果与国标法结果基本相符, 实验结果见表2, 样品色谱图见图8
MELC的优点是样品处理简单, 仅需将样品稀释后经微孔滤膜过滤便可进样分析, 这样既可节省分析时间又可减少样品的用量。经实验优化选择, 最终确定3.64 g/L CTAB水溶液-50 g/L乙腈-8 g/L乙酸乙酯-6.8 g/L磷酸二氢钾水溶液为流动相, 确保NaFeEDTA与酱油中复杂成分完全分开, 分析时间短。MELC直接进样的优点显而易见, 但对分析物无富集浓缩过程, 仅适用于浓度较高的目标物分析; 同时酱油样品可直接进样的机制还不是完全清楚, 推测是由于微乳流动相中较高浓度的表面活性剂分子, 吸附于固定相表面, 阻止了蛋白与固定相的结合, 确切的机制有待进一步研究。总的来说, 本方法具有绿色环保、成本低、样品处理简单等优点, 是一种值得推广的方法。
