Objective To evaluate the degradation dynamics of the combined use of bifenazate and etoxazole on Vigna unguiculata and assess the dietary exposure risks for different population groups. Methods Bifenazate (sum of bifenazate and bifenazate-diazene) in samples of Vigna unguiculata collected from eight regions of China were detected by liquid chromatography-tandem mass spectrometry. Dietary exposure risk assessment was conducted based on the test results, dietary consumption data and body weight data of different population groups in China. Results The dissipation behaviors of bifenazate in Vigna unguiculata from Shandong, Anhui, Hunan and Guangxi all did not follow first-order kinetics. The dissipation behaviors of etoxazole in Vigna unguiculata from Shandong, Anhui, Hunan, and Guangxi followed first-order kinetics, and their half-lives were 1.93-2.54 d. Using a suspension concentrate containing 45% bifenazate and etoxazole with 8 mL formulation per mu (Chinese acre) to control red mites on Vigna unguiculata, spraying once, at the harvest interval of 10 d, terminal concentrations of bifenazate and etoxazole in Vigna unguiculata were <0.0100 mg/kg and <0.0020-0.0042 mg/kg, respectively. The contribution rate of the long-term dietary exposure risk of these 2 kinds of pesticides were all lower than 100% for different population groups, indicating that there was no unacceptable risk. Conclusion 45% bifenazate·etoxazole suspension concentrate is applied at a dosage of 8 mL per mu for controlling spider mites on Vigna unguiculata, with one spray application and a recommended pre-harvest interval of 10 days.

目的 评估联苯肼酯和乙螨唑复配使用在豇豆上的消解动态及对不同人群的膳食暴露风险。方法 采用液相色谱-串联质谱法对全国8个地区露地条件下豇豆上联苯肼酯(联苯肼酯和联苯肼酯-二氮烯的总和)和乙螨唑残留进行检测分析。根据检测结果和我国不同人群的膳食消费量与体重数据进行了膳食暴露风险评估。结果 联苯肼酯在山东、安徽、湖南和广西豇豆上的消解动态过程均不符合一级动力学方程。乙螨唑在山东、安徽和广西豇豆上的消解动态过程均符合一级动力学方程, 半衰期为1.93~2.54 d; 乙螨唑在湖南豇豆上的消解动态不符合一级动力学方程。在豇豆上施用45%联苯肼酯·乙螨唑悬浮剂, 制剂用量为8 mL/亩, 喷雾施药1次, 在10 d采收间隔下, 联苯肼酯和乙螨唑在豇豆上的最终残留量分别为<0.0100 mg/kg和<0.0020~0.0042 mg/kg。这两种农药对各人群的长期膳食暴露风险贡献值均远低于100%, 表示没有不可接受的风险。结论 45%联苯肼酯·乙螨唑悬浮剂, 在豇豆上防治红蜘蛛, 制剂用药量8 mL/亩剂量, 喷雾施药1次, 安全间隔期推荐为10 d。

目的 评估联苯肼酯和乙螨唑复配使用在豇豆上的消解动态及对不同人群的膳食暴露风险。方法 采用液相色谱-串联质谱法对全国8个地区露地条件下豇豆上联苯肼酯(联苯肼酯和联苯肼酯-二氮烯的总和)和乙螨唑残留进行检测分析。根据检测结果和我国不同人群的膳食消费量与体重数据进行了膳食暴露风险评估。结果 联苯肼酯在山东、安徽、湖南和广西豇豆上的消解动态过程均不符合一级动力学方程。乙螨唑在山东、安徽和广西豇豆上的消解动态过程均符合一级动力学方程, 半衰期为1.93~2.54 d; 乙螨唑在湖南豇豆上的消解动态不符合一级动力学方程。在豇豆上施用45%联苯肼酯·乙螨唑悬浮剂, 制剂用量为8 mL/亩, 喷雾施药1次, 在10 d采收间隔下, 联苯肼酯和乙螨唑在豇豆上的最终残留量分别为<0.0100 mg/kg和<0.0020~0.0042 mg/kg。这两种农药对各人群的长期膳食暴露风险贡献值均远低于100%, 表示没有不可接受的风险。结论 45%联苯肼酯·乙螨唑悬浮剂, 在豇豆上防治红蜘蛛, 制剂用药量8 mL/亩剂量, 喷雾施药1次, 安全间隔期推荐为10 d。

Objective To evaluate the degradation dynamics of the combined use of bifenazate and etoxazole on Vigna unguiculata and assess the dietary exposure risks for different population groups. Methods Bifenazate (sum of bifenazate and bifenazate-diazene) in samples of Vigna unguiculata collected from eight regions of China were detected by liquid chromatography-tandem mass spectrometry. Dietary exposure risk assessment was conducted based on the test results, dietary consumption data and body weight data of different population groups in China. Results The dissipation behaviors of bifenazate in Vigna unguiculata from Shandong, Anhui, Hunan and Guangxi all did not follow first-order kinetics. The dissipation behaviors of etoxazole in Vigna unguiculata from Shandong, Anhui, Hunan, and Guangxi followed first-order kinetics, and their half-lives were 1.93-2.54 d. Using a suspension concentrate containing 45% bifenazate and etoxazole with 8 mL formulation per mu (Chinese acre) to control red mites on Vigna unguiculata, spraying once, at the harvest interval of 10 d, terminal concentrations of bifenazate and etoxazole in Vigna unguiculata were <0.0100 mg/kg and <0.0020-0.0042 mg/kg, respectively. The contribution rate of the long-term dietary exposure risk of these 2 kinds of pesticides were all lower than 100% for different population groups, indicating that there was no unacceptable risk. Conclusion 45% bifenazate·etoxazole suspension concentrate is applied at a dosage of 8 mL per mu for controlling spider mites on Vigna unguiculata, with one spray application and a recommended pre-harvest interval of 10 days.

豇豆(Vigna unguiculata L. Walp.)是我国重要的豆类蔬菜, 但蓟马、叶螨、豆荚螟、美洲斑潜蝇、锈病等病虫害发生较为普遍[1], 目前主要采用化学药剂进行防治。豇豆整个生长期均有病虫害发生, 进入开花结荚期后病虫害发生逐渐严重, 发生的病虫害种类多, 农药使用种类多、用量大, 花果同期、采摘间隔短, 导致了豇豆农药残留水平较高, 增加豇豆农药残留超标的风险, 可能对人体健康形成安全隐患, 因此其农药残留膳食暴露风险需要引起格外关注。
目前国内登记在豇豆上防治叶螨病虫害的农药只有43%联苯肼酯悬浮剂的一种制剂, 防治对象为二斑叶螨[2]。联苯肼酯对螨类的各个时期均有影响[3], 通过触杀和胃毒作用快速高效防治叶螨, 与其他杀螨剂无交互抗性[3-4]。乙螨唑抑制蜕皮, 高效防治螨类卵和若螨、幼螨, 但对成螨无效[3,5]。然而长期使用同一有效成分, 叶螨可能对其产生抗性, 联苯肼酯和乙螨唑共同使用, 可延缓叶螨抗药性发展。联苯肼酯与乙螨唑复配制剂目前仅登记在柑橘和苹果的病虫害防控上[2]。前期药效结果表明, 45%联苯肼酯·乙螨唑悬浮剂对豇豆红蜘蛛有较好的防治效果(未发表数据)。目前已有在草莓[6]、柑橘[7]和葡萄干[8]上开展了联苯肼酯残留水平和膳食风险评估的相关报道, 但以上研究仅聚焦于联苯肼酯母体, 忽视了联苯肼酯的代谢物联苯肼酯-二氮烯, 低估了联苯肼酯的膳食风险。另外, SATHESHKUMAR等[9]研究了联苯肼酯和联苯肼酯-二氮烯在茶叶上的消解动态, 赵翔[10]和凌淑萍等[11]报道了联苯肼酯及联苯肼酯-二氮烯在柑橘中的残留消解动态, HOU等[12]对柑橘中的联苯肼酯、联苯肼酯-二氮烯和乙螨唑进行了残留水平分析和膳食风险评估, 杜月梅等[13]研究了联苯肼酯和联苯肼酯-二氮烯在茄子上的最终残留量, 付岩等[14]报道了联苯肼酯及联苯肼酯-二氮烯在草莓上的残留消解情况, 孙瑞卿等[15]对苹果中联苯肼酯及联苯肼酯-二氮烯的储藏稳定性研究。目前鲜少见在豇豆上复配使用联苯肼酯和乙螨唑消解行为的相关报道。本研究以豇豆为对象, 研究复配使用联苯肼酯和乙螨唑在豇豆上的残留动态。通过计算半衰期和残留量, 确定它们在豇豆中的残留消解动态和最终残留量, 并采用点评估方法进行了短期和长期膳食暴露风险分析。本研究旨在为多种杀螨剂在豇豆中的合理使用和风险评价提供理论依据。
45%联苯肼酯·乙螨唑悬浮剂(联苯肼酯和乙螨唑的有效成分的质量分数分别为30%和15%)(福建省德盛生物工程有限责任公司); 联苯肼酯(纯度97.9%)、联苯肼酯-二氮烯(纯度99.3%)、乙螨唑(纯度98.6%)(北京曼哈格生物科技有限公司); 无水硫酸镁(纯度99%)、甲酸(纯度99%)、甲酸铵(纯度99%)、氯化钠(纯度99.5%)(上海麦克林生化科技股份有限公司); 抗坏血酸(纯度99%)(上海阿拉丁生化科技股份有限公司); 甲醇、乙腈(色谱纯, 德国达姆施塔特默克集团); N-丙基乙二胺(primary secondary amine, PSA)固相吸附剂(美国Sigma-Aldrich公司)。
LCMS-8050液相色谱串联质谱仪、AUD220D电子天平(精度十万分之一, 日本岛津公司); ACQUITY UPLC® HSS T3色谱柱(100 mm×2.1 mm., 1.8 µm)(美国沃特世科技有限公司)。
田间试验于2023年7—12月, 根据农业农村部农药检定所发布的《农药登记残留试验区域指南》(农药检(残留)[2018] 18号)在山西晋中、北京大兴、山东泰安、安徽合肥、湖南长沙、广西南宁、福建南平和海南澄迈进行最终残留量试验, 其中山东泰安、安徽合肥、湖南长沙和广西南宁进行残留消解试验。施药剂量如下: 45%联苯肼酯·乙螨唑悬浮剂(施药剂量为8 mL/亩)。在露地条件下, 设置药剂单独处理小区并设置清水对照小区, 每个小区50 m2, 在豇豆盛产期(开花结荚至采收期)对豇豆植株均匀喷雾1次。残留消解试验: 施药后2 h, 0、3、7、10、14 d分别采集处理小区的豇豆样本, 0 d和14 d采集对照区豇豆样本。最终残留量试验: 施药后7 d、10 d分别采集处理小区和对照小区的豇豆样本, 各小区每次采集样本2 kg。把样本切碎混匀后, 按四分法缩分, 用组织捣碎机或匀浆器将样品均匀制成泥, 分取两份, 每份至少250 g待测, 于-18 ℃以下冰柜中保存。
(1)联苯肼酯和联苯肼酯-二氮烯
称取(5.00±0.02) g豇豆于50 mL离心管中, 加入10 mL 0.25%的乙酸乙腈, 涡旋1 min后超声提取10 min, 4000 r/min离心5 min。取5 mL上清液, 加入5 mL 0.25%的抗坏血酸和4 g氯化钠于50 mL离心管中, 而后涡旋1 min, 超声提取30 min, 4000 r/min离心5 min。取1 mL上清液置于10 mL离心管中, 加入50 mg PSA, 50 mg C18和100 mg无水硫酸镁, 4000 r/min离心5 min。取上清液过0.22 μm有机滤膜, 待检测。
(2)乙螨唑
称取(5.00±0.02) g豇豆于50 mL离心管中, 加入10 mL 0.25%的乙酸乙腈, 涡旋1 min, 加入4 g氯化钠, 涡旋1 min, 后超声提取10 min, 4000 r/min离心5 min。取1 mL上清液置于10 mL离心管中, 加入50 mg PSA, 50 mg C18和100 mg无水硫酸镁, 4000 r/min离心5 min。取上清液过0.22 μm有机滤膜, 待检测。
色谱条件: ACQUITY UPLC® HSS T3色谱柱(100 mm× 2.1 mm, 1.8 µm)。流动相: A为2 mmol/L甲酸铵0.01%甲酸水溶液, B为2 mmol/L甲酸铵0.01%甲酸甲醇溶液, 流速为0.3 mL/min。进样体积1 μL, 柱温40 ℃。梯度洗脱程序见表1
质谱条件: 电喷雾正离子源(+); 多反应监测模式; 雾化气流速3 L/min, 干燥气流速10 L/min, 加热气流速10 L/min, 接口温度300 ℃, 脱溶剂管线温度250 ℃, 加热模块温度400 ℃。其他质谱条件如表2所示。
(1)联苯肼酯和联苯肼酯-二氮烯
称取联苯肼酯标准品0.00324 g, 用乙腈定容至5 mL容量瓶中, 得到质量浓度为634.39 mg/L联苯肼酯标准溶液储备液。称取联苯肼酯-二氮烯标准品0.00341 g, 用乙腈定容至5 mL容量瓶中, 得到质量浓度为677.23 mg/L联苯肼酯标准溶液储备液。用乙腈将联苯肼酯和联苯肼酯-二氮烯标准溶液储备液逐级稀释成100、10、1 mg/L的标准中间溶液, 再稀释成质量浓度为0.001、0.005、0.010、0.020、0.050、0.100 mg/L的联苯肼酯和联苯肼酯-二氮烯标准混合溶液。按1.3.2节的条件测定, 以质量浓度为横坐标, 对应的峰面积为纵坐标绘制标准曲线。外标法定量。
(2)乙螨唑
称取乙螨唑标准品0.00310 g, 用乙腈定容至5 mL容量瓶中, 得到质量浓度为611.32 mg/L乙螨唑标准溶液储备液。用乙腈将乙螨唑标准溶液储备液逐级稀释成100、10、1 mg/L的标准中间溶液, 再稀释成质量浓度为0.0005、0.0010、0.0025、0.0050、0.0100、0.0200 mg/L的标准工作溶液。按1.3.2节的条件测定, 以质量浓度为横坐标, 对应的峰面积为纵坐标绘制标准曲线。外标法定量。
在空白豇豆中添加3个水平浓度的联苯肼酯和联苯肼酯-二氮烯标准溶液, 添加水平分别为0.01、0.02、7.00 mg/kg。乙螨唑添加水平分别为0.002、0.010、0.020 mg/kg。每个添加水平浓度重复5次, 涡旋混匀静置30 min后, 按1.3.1和1.3.2节进行样本前处理和分析测定, 分别计算平均回收率和相对标准偏差。其中联苯肼酯在豇豆中添加浓度为7 mg/kg的样本提取液, 上机前稀释100倍后测定。
短期膳食暴露量(international estimated short-term intake, IESTI, mg/kg bw)基于联合国粮农组织/世界卫生组织(food and agriculture organization of united nations/world health organization, FAO/WHO)农药残留专家联席会议(joint meeting on pesticide residues, JMPR)的相关公式计算得到[16]。因此其IESTI计算公式见式(1):

式中: Ue为可食部分的单重(edible portion of the unit weight), kg; HR为农药的最高残留量(highest residue), mg/kg; ν为个体之间的变异因子, 豇豆取3; LP为大份餐重量(large portion), kg; bw为体重(body weight), kg, 中国不同人群体重见表3
短期膳食暴露风险(%acute reference dose, %ARfD)计算公式见式(2):

式中: ARfD为各农药的急性参考剂量, mg/kg bw。
具体数值参考JMPR数据库[17], 当ARfD≤100%时, 表示风险可接受; 当ARfD>100%时, 表示存在不可接受的短期膳食暴露风险[17]
采用国家农药残留标准审评委员会推荐的点评估方法[18]进行, 计算公式见式(3)和式(4):

式中: NEDI为国家估算每日摄入量(national estimated daily intake), mg/kg bw; STMRi为农药残留中值, mg/kg, 本研究中用各农药的残留平均值代替; Fi为第i种食物的每日消费量, kg/d。福建省不同人群豇豆每日消费量见表3
鉴于目前仅有豇豆一种食用农产品上的农药残留数据, 并无其他食用农产品上的相关农药残留数据, 因此不能计算包含所有登记作物在内的农药残留膳食暴露NEDI值, 故只计算豇豆中各农药残留膳食暴露占NEDI的份额以及对长期暴露风险商(long-term dietary exposure risk, RQc)的贡献率(RQc%), 具体计算公式见式(4):
RQc%=NEDI/ADI×100%
式中: ADI为各农药的每日允许摄入量(acceptable daily intake), mg/(kg bw·d)。
利用Microsoft office LTSC 2024中的Excel制表, 并进行拟合联苯肼酯和乙螨唑在豇豆中的消解动态方程, 分析检测重复3次。
联苯肼酯、联苯肼酯-二氮烯和乙螨唑的线性方程见表4, 相关系数(r)均大于0.99。联苯肼酯和联苯肼酯-二氮烯的方法定量限均为0.010 mg/kg; 乙螨唑的方法定量限为0.002 mg/kg。
添加回收试验结果如表5所示, 在0.010~7.000 mg/kg添加水平下, 联苯肼酯、联苯肼酯-二氮烯和乙螨唑在豇豆中的平均回收率分别为联苯肼酯92%~100%, 相对标准偏差小于等于9.90%; 联苯肼酯-二氮烯85%~89%, 相对标准偏差小于等于10。00%; 在0.002~0.020 mg/kg添加水平下, 豇豆中的乙螨唑平均回收率为92%~116%, 相对标准偏差小于等于6.90%。均符合农药残留分析要求。
表6所示, 联苯肼酯在山东、安徽、湖南和广西豇豆上的原始沉积量分别为0.110、0.026、0.420、0.043 mg/kg, 其消解动态过程均不符合一级动力学方程。乙螨唑在山东、安徽、湖南和广西豇豆上的原始沉积量分别为0.210、0.084、0.098、0.098 mg/kg, 山东、安徽和广西的乙螨唑消解动态过程均符合一级动力学方程, 消解半衰期分别为1.93、2.54、2.44 d; 湖南的乙螨唑消解动态不符合一级动力学方程。
表7所示, 联苯肼酯在豇豆上采收间隔7 d和10 d八地的最终残留量均为小于0.0100 mg/kg, 低于GB 2763—2021《食品安全国家标准 食品中农药最大残留限量》在豇豆上制定的最大残留限量7 mg/kg。乙螨唑在豇豆上7 d和10 d的最终残留量分别为<0.0020~0.0240 mg/kg和<0.0020~0.0042 mg/kg, 其中10 d八地的最终残留量均低于欧盟在豆类蔬菜上制定的临时残留限量0.01 mg/kg[20]
根据FAO数据库[17], 联苯肼酯和乙螨唑没有必要制定ARfD, 因此针对以上两种农药未进行短期膳食暴露风险评估。
根据联苯肼酯和乙螨唑在豇豆样本中检出的残留平均值及其ADI和福建省豇豆膳食消费数据[19]进行长期膳食暴露风险评估, 结果见表8。联苯肼酯和乙螨唑在豇豆上采收间隔10 d的RQc%均远小于100%, 表明其长期膳食暴露风险在可接受范围内。
本研究中联苯肼酯在豇豆上的消解动态不符合一级动力学方程, 无法计算半衰期, 但其在草莓上的半衰期3.15 d[6]、6.4~10.4 d[14], 在柑橘上半衰期为0.23~1.54 d[21]、1.92~2.66 d[7]、4.86 d[22]、5.1~7.9 d[10]、20.7 d[11], 消解较快。本研究中乙螨唑在豇豆上的消解半衰期为1.93~2.54 d, 与其在以下农产品中的半衰期: 芸豆荚3.13 d[23]、芸豆叶2.73 d [23]、草莓2.20~3.00 d[24]和6.4~8.7 d[25]、琯溪蜜柚4.34~4.54 d[26]、枸杞5.77~7.13 d[27]、枇杷6.9~8.9 d[28]相似。CHEN等[29]报道了降雨冲刷率与农药的水溶性非常相关, 高水溶性的农药受施药后降雨的冲刷影响较大。联苯肼酯和乙螨唑的正辛醇-水分配系数(octanol-water partition coefficient, Kow) lgP分别为3.4 (40 ℃, pH 7)和5.52 (20 ℃)[30], 亲水性的理化特点可能影响了这两种杀螨剂的消解速率。
本研究探究联苯肼酯和乙螨唑复配在豇豆中的消解行为及风险评估。45%联苯肼酯·乙螨唑悬浮剂在在豇豆上施药制剂量8 mL/亩, 施药1次, 采收间隔为10 d时, 在露地条件下这两种农药的残留量均低于最大残留限量。然而, 考虑到多种膳食来源和农药的积累, 复配使用的风险增加应引起广泛关注。由于我国人均膳食结构中蔬菜的每日摄入量包括了各种蔬菜的摄入量, 本研究以其作为豇豆的摄入量进行计算, 可能增大豇豆长期膳食暴露风险的评估结果。本研究中仅针对单一农药残留进行点评估, 未对多种农药残留进行累积膳食暴露风险评估, 而多种农药残留联合暴露可能会增大居民的膳食风险, 因此还需要进一步开展农药多残留的联合膳食暴露风险评估研究, 更好地指导农业生产和提高农产品质量安全水平。
