Objective To establish a rapid analysis method based on solid phase extraction-high performance liquid chromatography-evaporative light scattering detection (SPE-HPLC-ELSD) for determining the γ-aminobutyric acid (GABA) content in edible enzymes from medicinal and edible homologous sources. Methods Fermented panacis quinquefolii radix liquid was selected as a representative sample. After centrifugation (8000 r/min, 10 min) and ultrasonic extraction (10 min, 25 ℃), 3 kinds of different sample pretreatment methods—non-purification, dispersive solid phase extraction purification, and hydrophilic-lipophilic balance (HLB)-based solid phase extraction purification were compared and optimized. An optimized HPLC-ELSD instrumental method was developed to achieve accurate content determination. The method was then applied to determine the GABA content in other medicinal and edible homologous enzyme products. Results GABA was determined by optimized SPE-HPLC-ELSD. GABA showed a good linear relationship (r=0.999) in the mass concentration range of 50-2000 μg/mL. The recovery rate ranged from 85.93% to 96.30%, with a repeatability relative standard deviation of 0.87% (n=6). Conclusion This method is efficient and accurate, suitable for the quantitative analysis of GABA in various medicinal and edible homologous enzymes. It also provides a new approach and reference for GABA determination in complex matrices and for detecting small molecules with strong polarity and low ultraviolet absorption.

目的 建立一种基于固相萃取-高效液相色谱-蒸发光散射法(solid phase extraction-high performance liquid chromatography-evaporative light scattering detection, SPE-HPLC-ELSD)快速检测药食同源食用酵素中γ-氨基丁酸(γ-aminobutyric acid, GABA)含量的分析方法。方法 选取西洋参发酵液为代表样品, 经离心(8000 r/min, 10 min)、超声提取(10 min, 25 ℃)后, 对比不净化、分散固相萃取净化、亲水-亲脂平衡(hydrophilic-lipophilic balance, HLB)通过式固相萃取净化3种不同的处理方式优化样品前处理过程, 开发优化HPLC-ELSD仪器方法实现含量准确测定, 并推广应用于其他多种药食同源食用酵素产品中的GABA含量测定。结果 选用优化后的SPE-HPLC-ELSD测定GABA, 在50~2000 μg/mL质量浓度范围内线性指标良好(r=0.999), 加标回收率在85.93%~96.30%, 重复性相对标准偏差为0.87% (n=6)。结论 本方法高效、准确, 适用于多种药食同源食用酵素中GABA定量分析, 也可为复杂基质下的GABA测定及强极性、低紫外吸收小分子化合物检测提供新的途径和参考。

目的 建立一种基于固相萃取-高效液相色谱-蒸发光散射法(solid phase extraction-high performance liquid chromatography-evaporative light scattering detection, SPE-HPLC-ELSD)快速检测药食同源食用酵素中γ-氨基丁酸(γ-aminobutyric acid, GABA)含量的分析方法。方法 选取西洋参发酵液为代表样品, 经离心(8000 r/min, 10 min)、超声提取(10 min, 25 ℃)后, 对比不净化、分散固相萃取净化、亲水-亲脂平衡(hydrophilic-lipophilic balance, HLB)通过式固相萃取净化3种不同的处理方式优化样品前处理过程, 开发优化HPLC-ELSD仪器方法实现含量准确测定, 并推广应用于其他多种药食同源食用酵素产品中的GABA含量测定。结果 选用优化后的SPE-HPLC-ELSD测定GABA, 在50~2000 μg/mL质量浓度范围内线性指标良好(r=0.999), 加标回收率在85.93%~96.30%, 重复性相对标准偏差为0.87% (n=6)。结论 本方法高效、准确, 适用于多种药食同源食用酵素中GABA定量分析, 也可为复杂基质下的GABA测定及强极性、低紫外吸收小分子化合物检测提供新的途径和参考。

Objective To establish a rapid analysis method based on solid phase extraction-high performance liquid chromatography-evaporative light scattering detection (SPE-HPLC-ELSD) for determining the γ-aminobutyric acid (GABA) content in edible enzymes from medicinal and edible homologous sources. Methods Fermented panacis quinquefolii radix liquid was selected as a representative sample. After centrifugation (8000 r/min, 10 min) and ultrasonic extraction (10 min, 25 ℃), 3 kinds of different sample pretreatment methods—non-purification, dispersive solid phase extraction purification, and hydrophilic-lipophilic balance (HLB)-based solid phase extraction purification were compared and optimized. An optimized HPLC-ELSD instrumental method was developed to achieve accurate content determination. The method was then applied to determine the GABA content in other medicinal and edible homologous enzyme products. Results GABA was determined by optimized SPE-HPLC-ELSD. GABA showed a good linear relationship (r=0.999) in the mass concentration range of 50-2000 μg/mL. The recovery rate ranged from 85.93% to 96.30%, with a repeatability relative standard deviation of 0.87% (n=6). Conclusion This method is efficient and accurate, suitable for the quantitative analysis of GABA in various medicinal and edible homologous enzymes. It also provides a new approach and reference for GABA determination in complex matrices and for detecting small molecules with strong polarity and low ultraviolet absorption.

药食同源食用酵素是指以国家发布的“药食同源”目录品种为原料, 经过可控的微生物定向发酵过程为途径, 制得含有特定生物活性成分的食用酵素类产品[1-3]。药食同源经过微生物的定向发酵, 相比于传统的炮制或水提工艺, 不仅可以改善口感风味, 同时在减毒增效、提高生物活性和利用率等方面均具有不可替代的优势[4-8]γ-氨基丁酸(γ-aminobutyric acid, GABA)是一种非蛋白类氨基酸近年来逐渐成为研究热点, 尤其是在其生物合成、代谢途径、生理功能及食品应用等方面[9-12]。其结构简单并无明显的紫外吸收, 因此常规的分光光度法和液相色谱紫外检测法具有较大的局限, 往往只能通过化学衍生、氨基酸分析仪或者液相色谱-质谱法等方法进行分析[13-15], 并常会遇到衍生效果不佳、实验重复性差、耗时及设备要求高等问题。建立科学的GABA检测方法对于提高研发效率和质量, 确保其真实含量和有效性、安全性具有重要的现实意义和应用价值。
亲水-亲脂平衡(hydrophilic-lipophilic balance, HLB)通过式固相萃取柱, 使用时直接将样品提取液加载到HLB固相萃取柱上, 通过液即为净化后的样品溶液, 操作简便快捷, 不需要换相操作过程及额外的有机溶剂已逐渐被广泛使用[16-18]。蒸发光散射检测器能够针对低紫外吸收化合物的灵敏检测, 作为一种通用紫外检测器的补充应用于液相色谱分析中, 而且相比示差折光检测器, 具有平衡时间短, 可以采用梯度洗脱等优势; 相比于电喷雾式检测器也具有测试成本低、稳定性好等优点[19-24]
本研究创新结合采用固相萃取-高效液相色谱-蒸发光散射法检测(solid phase extraction-high performance liquid chromatography-evaporative light scattering detection, SPE- HPLC-ELSD)的方式, 针对基质较为复杂的药食同源食用酵素中的GABA进行分析检测, 便于药食同源微生物发酵过程中的过程监控和成品成分分析。不仅可以为复杂基质状态下的GABA检测提供一条新的出路, 同时也可以为药食同源质量研究及产品控制过程中低紫外吸收小分子化合物的定性定量提供一种新的检测途径。

多种药食同源食用酵素样品由浙江大医德美有限公司研发制备。
三氟乙酸(色谱纯, 北京沃凯生物科技有限公司); 甲醇(色谱纯, 美国TEDIA公司); 水(超纯水, 电阻率≥18.2 MΩ·cm); GABA标准品[含量为99.0%, 安谱云实验用品(上海)有限公司]
本研究所用的药食同源食用酵素工艺流程图见图1
色谱柱: Waters XSelect HSS T3柱(4.6 mm×250 mm, 3.5 μm); 进样体积2 μL; 柱温: 40 ℃; 流动相流速0.7 mL/min; ELSD气化温度: 95 ℃; 氮气流量: 1.8 L/min; 增益: 4; 运行时间: 35 min; 数据结果以外标对数法计算; 液相梯度洗脱程序和流动相如表1所示。
精密称取GABA标准品50.0 mg(精确至0.1 mg)至25 mL容量瓶中, 加纯水溶解稀释至刻度定容。以此溶液为标准品储备溶液, 并逐级用纯水进行稀释至不同浓度, 4 ℃可保存24 h, 备用。
取西洋参发酵液或其他食用酵素原液, 8000 r/min离心10 min去除残留药材及沉淀, 混合均匀后, 准确移取1.0 mL至10 mL容量瓶中, 加水8 mL, 25 ℃超声提取10 min。静置至室温后定容, 摇匀, 取3 mL上清液, 分别先采用不净化、分散固相萃取净化和HLB通过式固相萃取3种不同形式的净化方式, 再过水相滤膜(0.22 μm), 4 ℃可保存20 h, 备用待测。
分别吸取2 μL样品溶液注入HPLC-ELSD仪器中, 以保留时间定性, 外标对数法定量(ELSD的响应值与样品浓度的对数呈线性关系), 测定不同药食同源食用酵素样品中的GABA含量。
本研究采用Agilent OpenLab CDS软件进行色谱数据的采集和数据处理。其中, 实验平行重复测定3次, 结果以平均值标识, 并通过Mocrosoft Excel 2010和Origin 2016软件进行数据的再分析和图表的绘制。
药食同源食用酵素经乳酸菌发酵后, 发酵液经过离心分离, 仍会有发酵产物、药渣、菌体、培养基、生物酶等各类基质的干扰[25-28], 而GABA化学性质又是极性较大, 出峰靠前。本研究以西洋参发酵液为样本, 考察了前处理提取液的净化方式, 分别采用不净化、分散固相萃取净化和HLB通过式固相萃取3种不同形式的净化方式, 以加标回收率作为结果表征手段。具体回收率结果见图2
样品不净化时, 目标峰存在一定的基质干扰, 基线明显整体抬高, 影响色谱峰分离, 加标回收率受积分方式和程度影响较大, 准确性由此必受影响; 传统的分散固相萃取净化能够通过去除样品中的磷脂、脂肪、盐、蛋白质等干扰降低基线问题, 然而相比于HLB通过式固相萃取回收率结果不具有优势。同时HLB通过式固相萃取净化在样品前处理的时间和操作复杂程度上更优, 无需活化步骤、不消耗额外的有机溶剂、不用换相。故对于基质复杂的酵素样品或其他含有GABA的复杂样品而言, HLB通过式固相萃取具有显著的优势。不同前处理的色谱图见图3。因此, 选择HLB通过式固相萃取作为样品净化方法。
基于药食同源发酵基质复杂的特性, 及GABA水溶性好, 极性强的特性, 在液相分离过程中往往会遇到基质干扰、出峰过于靠前等问题[29-30]。因此在色谱柱的选择上进行了优化, 实验比对不同类型色谱柱[XSelect HSS T3柱(4.6 mm×250 mm, 3.5 μm)、XSelect HSS T3柱(4.6 mm×250 mm, 5 μm)、XBridge C18 (4.6 mm×250 mm, 5 μm)、XBridge C18 (4.6 mm× 150 mm, 5 μm)、Atlantis T3 (4.6 mm×250 mm, 5 μm)]。尝试多种药食同源食用酵素样品, 对比主峰分离度和峰型。发现XBridge C18和Atlantis T3填料类型的色谱柱对于GABA的保留效果不及亲水性保留效果更好的XSelect HSS T3柱, GABA会和溶剂峰及杂质峰部分重合, 影响定量。而3.5 μm小粒径的XSelect HSS T3柱相比于5 μm分离效果也会更佳。在优化后的0.7 mL/min的流速条件下, 使用3.5 μm小粒径的也不会超过液相仪器的最高使用压力。各食用酵素样品中GABA主峰与杂质峰、溶剂峰分离度≥1.5, 理论塔板数>8000, 满足色谱定量要求。最终选用XSelect HSS T3柱(4.6 mm×250 mm, 3.5 μm), 可以满足不同药食同源食用酵素样品中GABA的有效保留和高效分离。
以下参照《中华人民共和国药典》2020版四部通则9101“分析方法验证指导原则”和GB 5009.295—2023《食品安全国家标准 化学分析方法验证通则》中规定的内容进行本分析方法的验证研究。
根据上述优化后的样品前处理及色谱条件, 以西洋参发酵液为典型, 考察色谱条件的专属性, 分别针对空白溶液、西洋参发酵液样品溶液、GABA标准品溶液进行仪器分析, 结果表明在此方法下GABA得到良好分离和保留, 具体详见图4。标准品和样品溶液中的GABA出峰保留时间完全一致, 色谱峰型良好对称, 理论塔板数大于9300, 拖尾因子为1.07, 与前后杂质峰的分离度分别为1.84和2.33, 达到分离要求。表明方法专属性良好, 能够满足样品中低含量GABA的准确定量。
分别配制50、150、300、600、1000、2000 μg/mL不同质量浓度的6个GABA标准品溶液, 依次由低到高注入HPLC中, 通过ELSD检测色谱峰面积, 以质量浓度的对数为横坐标(X, μg/mL), 峰面积的对数为纵坐标(Y), 重复平行操作3次取平均值, 进行线性回归曲线的绘制。最终得到线性回归方程Y=1.6245X+3.1926, 相关系数r=0.999, 表明方法线性较好, 范围合适, 适用于各类不同GABA含量水平样品的定量分析。
仪器检出限和定量限: 通过标准品溶液的逐级稀释进样, 以色谱峰主峰信噪比S/N≥10.0时对应的GABA浓度作为定量限, 以S/N≥3.0时对应的GABA浓度作为检出限。最终得到本方法在标准品溶液质量浓度为15 μg/mL时, 检出限为4.6, 满足检出限要求; 标准品溶液质量浓度为30 μg/mL时, 定量限为11.1, 满足定量限要求。
方法检出限和定量限: 按照上述方法, 通过对于西洋参发酵液样品经逐级稀释后, 再经过样品前处理和色谱检测, 得到西洋参发酵液样品中GABA质量浓度分别为20 μg/mL和40 μg/mL时, 满足S/N≥3.0和S/N≥10.0要求, 即为本方法的检出限和定量限。
取已制备西洋参发酵液样品提取溶液和标准品溶液(0.6 mg/mL)按上述色谱分析条件采取连续进样6次, 记录色谱峰面积响应值, 计算相对标准偏差(relative standard deviation, RSD)。结果样品溶液连续进样RSD为0.73%; 标准品溶液连续进样RSD为0.39%, 表明本方法仪器精密度考察结果较好。
选取同批次的西洋参发酵液, 重复6次样品前处理和仪器检测过程, 考察本分析方法的重复性, 计算得到GABA含量的RSD值为0.87%, 证明方法重复性良好。
为考察随机变动因素, 在同个实验室, 不同研究人员、不同日期及不同仪器水平上进行同批次实际样品进行检测, 连续平行实验3次, 考察方法中间精密度。结果2位实验人员在不同实验日期6组的实验数据中表现基本一致, 6次实验结果的RSD为2.73%, 符合中间精密度考察要求。
基于室温、避光的贮存条件下放置的已通过样品前处理后的西洋参发酵液样品溶液, 进行样品溶液稳定性考察, 通过一定的色谱自动进样程序, 分别在0、5、10、15、20、25、30 h的时间段进行进样分析, 记录相应的GABA色谱峰面积, 考察样品溶液在一定存放时间内的稳定性。结果发现在0~20 h时间段内, GBAB峰面积基本无显著变化, 前5次进样峰面积的RSD在0.97%范围中, 而后续25 h、30 h后逐渐出现缓慢分解下降趋势。同个样品在室温、避光的贮存条件下25 h和30 h后峰面积将分别下降1.4 %和3.6%。由此, 本样品前处理后的样品溶液, 在室温、避光的贮存条件下建议存放20 h内进行色谱分析, 以达到最佳的定量检测结果。
改变一定区间内的色谱条件参数, 以考察分析方法的耐用性及不同实验室间的重现性, 结果发现在流速变化±0.05 mL/min, 氮气流量0.3 mL/min以及柱温, ELSD气化温度变动±3 ℃的条件下, 样品提取溶液及标准品峰型依旧保持对称良好, GABA色谱峰理论塔板数在8642~9314区间内波动; 与主峰最近杂质的分离度在1.78~2.11区间内波动; 以峰面积定量结果恒定, 不将显著影响检测结果, 证明方法耐用性较佳。
选取已测样品西洋参发酵液, 分别加入不同浓度的低、中、高3种标准品, 按优化后的样品前处理稀释、超声提取、净化后, 同样进行仪器测定, 计算加标回收率结果, 以考察本方法的准确程度。具体数据见表2, 不同浓度下的加标回收率在范围85.93%~96.30%内, 平均回收率范围在89.40%~94.35%之间, RSDs为1.86%~3.90%, 说明本分析方法准确性良好。
选取除西洋参发酵液之外的10款已研究开发的药食同源食用酵素产品进行适用性研究, 结果表明本方法可以适用于多种不同品类的药食同源食用酵素产品中GABA的低含量分析, 色谱分离良好, 检测稳定, 且适用于多品类不同浓度水平的样品检测。具体数据详见表3。相比于传统衍生剂衍生紫外光谱检测的标准方法, 尤其是在基质复杂、含量较低的终产品检测过程中, 改进后的方法准确度和检测效率具有明显的优势。同时, 针对已研究开发下述10款食用酵素产品中的GABA含量进行分析, 发现实测数据远高于QB/T 5323《植物酵素》中关于GABA的限量要求, 也证明本研究设计开发的食用酵素工艺及配方较为成功。
本研究建立了一种基于SPE-HPLC-ELSD的色谱分析方法, 可以简便、快速、高准确度地针对药食同源食用酵素产品中的GABA进行分析检测。相比于传统的化学衍生或质谱分析等技术手段, 本方法不仅具有良好的抗基质干扰能力, 同时效率更高、操作更易。同时通过优化样品称样量及稀释倍数, 本方法同样适用于其他功能果蔬食用酵素、天然食品、GABA原料及复杂基质下含GABA的功能保健食品产品中GABA的定量分析, 可为复杂样品中强极性、低紫外吸收化学小分子的定性定量分析提供一种新的途径和方向。
