Objective To optimize the acid hydrolysis-Fehling reagent titration method and the acid hydrolysis-3,5-dinitrosalicylic acid (DNS) colorimetric method to compare the consistency of different methods for measuring starch content in grain crop samples. Methods Experimental conditions, including ethanol washing volume, acid addition volume, acid hydrolysis time, sample solution pH, detection wavelength, chromogenic agent volume, color development time and color development temperature, were optimized to determine the optimal processing conditions for determining the starch content in such products. Results The optimal conditions for the acid hydrolysis-Fehling reagent titration method were as follows: After weighing the sample, fats and soluble sugars were removed using an appropriate amount of petroleum ether and ethanol; 25 mL of hydrochloric acid was added, followed by condensation reflux for 120 minutes; the hydrolyzed sample solution was neutralized with sodium hydroxide, filtered, and the filtrate was titrated with alkaline copper tartrate methyl and ethyl solution at a rate of one drop every 2 seconds. The optimal conditions for the acid hydrolysis-DNS colorimetric method were as follows: After removing fat, sugar and acid hydrolysis, the sample solution pH was adjusted to 8-9; for samples with color and high protein content, 10 mL of lead acetate was added to reduce or eliminate pigment interference; after filtration, 2 mL of DNS was added to the filtrate, followed by color development in a water bath at 90-100 °C for 12 minutes; the absorbance was measured at 540 nm using a spectrophotometer, and the determination was completed within 4 hours. Both methods had a relative standard deviation (RSD) of less than 2% and a relative error (RE) of less than 4%. The measurement results had passed mean consistency test and paired samples t-test. Conclusion The 2 kinds of optimized methods are more efficient and accurate compared to the national standard method, with good accuracy and precision making them suitable for starch content determination in grain crops.

目的 优化酸水解-斐林试剂滴定法和酸水解-3,5-二硝基水杨酸(3,5-dinitrosalicylic acid, DNS)比色法, 对比粮食作物样品中淀粉含量不同测定方法的一致性。方法 通过对乙醇洗涤量、加酸量、酸解时间、样液pH、检测波长、显色剂加入量、显色时间、显色温度等实验条件的优化, 确定测定该类产品淀粉含量的最佳条件。结果 酸水解-斐林试剂滴定法最优条件为称取样品后用适量石油醚、乙醇洗去脂肪及可溶性糖, 加入25 mL盐酸, 冷凝回流120 min, 用氢氧化钠校正试样水解液至中性, 过滤后取滤液以2 s/滴的速度滴定碱性酒石酸铜甲乙液。酸水解-DNS比色法最优条件为脱脂除糖酸解后, 校正试样pH为8~9, 有颜色且蛋白质含量高的样品加入10 mL乙酸铅沉淀蛋白以减少或消除色素干扰, 过滤后取滤液加入2.0 mL DNS, 90~100 ℃水浴12 min进行显色, 分光光度计540 nm波长下进行比色, 4 h内完成测定。两种方法相对标准偏差(relative standard deviation, RSD)均小于2%, 相对误差(relative error, RE)均小于4%, 测定结果通过平均值一致性检验与配对样本t检验, 具有一致性。结论 优化的两种方法相较国标法更加高效、准确, 方法正确度、精密度良好, 可适用于粮食作物中淀粉含量的检测。

目的 优化酸水解-斐林试剂滴定法和酸水解-3,5-二硝基水杨酸(3,5-dinitrosalicylic acid, DNS)比色法, 对比粮食作物样品中淀粉含量不同测定方法的一致性。方法 通过对乙醇洗涤量、加酸量、酸解时间、样液pH、检测波长、显色剂加入量、显色时间、显色温度等实验条件的优化, 确定测定该类产品淀粉含量的最佳条件。结果 酸水解-斐林试剂滴定法最优条件为称取样品后用适量石油醚、乙醇洗去脂肪及可溶性糖, 加入25 mL盐酸, 冷凝回流120 min, 用氢氧化钠校正试样水解液至中性, 过滤后取滤液以2 s/滴的速度滴定碱性酒石酸铜甲乙液。酸水解-DNS比色法最优条件为脱脂除糖酸解后, 校正试样pH为8~9, 有颜色且蛋白质含量高的样品加入10 mL乙酸铅沉淀蛋白以减少或消除色素干扰, 过滤后取滤液加入2.0 mL DNS, 90~100 ℃水浴12 min进行显色, 分光光度计540 nm波长下进行比色, 4 h内完成测定。两种方法相对标准偏差(relative standard deviation, RSD)均小于2%, 相对误差(relative error, RE)均小于4%, 测定结果通过平均值一致性检验与配对样本t检验, 具有一致性。结论 优化的两种方法相较国标法更加高效、准确, 方法正确度、精密度良好, 可适用于粮食作物中淀粉含量的检测。

Objective To optimize the acid hydrolysis-Fehling reagent titration method and the acid hydrolysis-3,5-dinitrosalicylic acid (DNS) colorimetric method to compare the consistency of different methods for measuring starch content in grain crop samples. Methods Experimental conditions, including ethanol washing volume, acid addition volume, acid hydrolysis time, sample solution pH, detection wavelength, chromogenic agent volume, color development time and color development temperature, were optimized to determine the optimal processing conditions for determining the starch content in such products. Results The optimal conditions for the acid hydrolysis-Fehling reagent titration method were as follows: After weighing the sample, fats and soluble sugars were removed using an appropriate amount of petroleum ether and ethanol; 25 mL of hydrochloric acid was added, followed by condensation reflux for 120 minutes; the hydrolyzed sample solution was neutralized with sodium hydroxide, filtered, and the filtrate was titrated with alkaline copper tartrate methyl and ethyl solution at a rate of one drop every 2 seconds. The optimal conditions for the acid hydrolysis-DNS colorimetric method were as follows: After removing fat, sugar and acid hydrolysis, the sample solution pH was adjusted to 8-9; for samples with color and high protein content, 10 mL of lead acetate was added to reduce or eliminate pigment interference; after filtration, 2 mL of DNS was added to the filtrate, followed by color development in a water bath at 90-100 °C for 12 minutes; the absorbance was measured at 540 nm using a spectrophotometer, and the determination was completed within 4 hours. Both methods had a relative standard deviation (RSD) of less than 2% and a relative error (RE) of less than 4%. The measurement results had passed mean consistency test and paired samples t-test. Conclusion The 2 kinds of optimized methods are more efficient and accurate compared to the national standard method, with good accuracy and precision making them suitable for starch content determination in grain crops.

淀粉是葡萄糖分子通过α-1,4和α-1,6糖苷键聚合而成的多糖, 由直链淀粉和支链淀粉组成, 广泛存在于谷类、豆类、根茎类等粮食作物中, 如小麦、大米、板栗、土豆等[1], 是植物中最关键的贮藏多糖[2]。淀粉一般储存在果实、种子和根中, 是人类饮食中主要的碳水化合物成分[3], 其消化和水解后产生的麦芽糊精、葡萄糖和其他物质是人体能量来源的主要途径[4]
国内外淀粉含量的前处理与测定方法有酸水解法[5]、酶水解法[6]、滴定法[7-8]、比色法[9]和旋光法[10-13]等。在实际应用中, 不同前处理与测定方法因其原理和操作步骤不同, 在测定相同样品时, 其淀粉含量存在较大差异[14-15]。实验室检测淀粉的主要方法为GB/T 5009.9—2023《食品安全国家标准 食品中淀粉的测定》, 但该标准所涉及的方法所用试剂较多[16]、实验过程烦琐, 实验操作复杂, 而且对实验结果的影响因素较多, 易导致结果不准确, 亟需通过优化实验条件加以改进[17]。近年来, 还原糖相关测定方法的优化研究取得显著进展。如3,5-二硝基水杨酸(3,5-dinitrosalicylic acid, DNS)比色法通过优化DNS试剂配方, 添加脱色试剂提高该方法的稳定性与灵敏度, 并使其适用于测定颜色样品[18-19]。电位滴定法通过采用自动电位滴定仪代替传统手工滴定提高该方法的分析效率和准确度[20]。本研究参考GB/T 5009.9—2023标准第二法, 通过对酸水解-斐林试剂滴定法存在的问题进行分析探讨, 确定了适合于粮食作物的最佳前处理与测定条件, 旨在提高该方法的准确性、稳定性和实用性; 同时探索采用酸水解法联合DNS比色法测定淀粉含量, 缩短检测时间, 提高检测灵敏度[21-22]。为淀粉含量的准确测定提供了可靠的多方法一致性验证。
所选小麦、大米、玉米、小米、高粱、土豆、板栗样品为研制标准物质的候选物样品, 原料经过大型球磨机粉碎后混匀, Co60辐照灭菌, 样品粒度为80目(0.198 mm)左右, 已经过均匀性检验确认其均匀性良好。
D-无水葡萄糖纯度标准物质(标准号GBW10062, 中国计量科学研究院); 石油醚、乙醇、氢氧化钠、α-萘酚、乙酸铅、硫酸钠、酒石酸钾钠、苯酚、DNS(分析纯, 上海迈瑞尔生化科技有限公司); 盐酸、浓硫酸(分析纯, 天津市大茂化学试剂厂); 甲基红、亚甲蓝、硫酸铜(分析纯, 天津市科密欧化学试剂有限公司); 亚铁氰化钾、无水亚硫酸钠(分析纯, 天津渤化化学试剂有限公司)。
ME204E电子天平(精度0.0001 g, 瑞士Mettler Toledo公司); HH-S6A型电热恒温水浴锅(北京科伟仪器有限公司); DK-II型电子调温万用电炉(天津天泰仪器有限公司); UV1901PCS型双光束紫外可见分光光度计(上海佑科仪器仪表有限公司); SC-6型高精度恒温槽(河南佰年仪器有限公司)。
参考GB/T 5009.9—2023标准第二法酸水解法与赵甲慧等[13]的方法并加以改进: (1)脱脂。称取2 g试样(精确至0.0001 g), 置于放有慢速滤纸的漏斗中, 用适量石油醚少量多次洗涤样品, 脱去脂肪; (2)洗糖。用适量乙醇溶液(85%, V/V)分次洗去可溶性糖至微糖检验结果为阴性, 通过莫利施反应确认无可溶性糖残留; (3)酸解。样品残渣用100 mL左右蒸馏水转移至250 mL锥形瓶中, 加入25 mL盐酸(1:1=V:V), 沸水浴中冷凝回流2 h; (4)除杂。回流完毕将试样水解液迅速放进凉水冷却, 待样液冷却后, 加入1滴甲基红指示剂, 用氢氧化钠溶液(400 g/L)调至黄色, 试样水解液颜色较深的样品用精密pH试纸调至试样水解液的pH为中性, 然后加10 mL乙酸铅溶液(200 g/L), 摇匀, 放置10 min后加入10 mL硫酸钠溶液(100 g/L), 以除去过多的铅, 摇匀后将全部溶液及残渣转入500 mL容量瓶中, 用蒸馏水润洗锥形瓶, 洗液合并入容量瓶中, 加蒸馏水定容至刻度, 混匀, 过滤, 弃去初滤液; (5)滤液按直接滴定法滴定, 试剂空白按反滴定法滴定。
称取0.3 g试样(精确至0.0001 g), 脱脂、洗糖、酸解操作步骤同斐林试剂滴定法的酸水解前处理。试样水解液冷却后, 不加甲基红, 用精密pH试纸调至试样水解液的pH约为8~9后, 加10 mL乙酸铅溶液(200 g/L), 摇匀, 放置10 min后加入10 mL硫酸钠溶液(100 g/L), 以除去过多的铅。摇匀后将全部溶液及残渣转入250 mL容量瓶中, 用蒸馏水润洗锥形瓶, 洗液合并入容量瓶中, 加蒸馏水定容至刻度。过滤, 弃去初滤液, 滤液供测定用。
用移液管准确吸取0.00、0.40、0.80、1.00、1.20、1.60、2.00 mL葡萄糖标准溶液(1.0 mg/mL)于25 mL比色管中, 加蒸馏水补足至2.0 mL, 加入DNS试剂2.0 mL, 沸水中加热12 min, 凉水冷却, 加蒸馏水定容至25 mL; 在2.0 mL去离子水中加入2.0 mL DNS试剂作相同加热处理后用于分光光度计的调零。540 nm处测定葡萄糖标准溶液吸光度。以葡萄糖含量(X, mg)为横坐标, 吸光度为纵坐标(Y)绘出标准曲线。吸取1.0 mL样液与1.0 mL蒸馏水, 加2.0 mL DNS试剂, 按上述处理方法测吸光度, 由葡萄糖含量折算出淀粉含量。
实验数据采用Excel 2021进行处理; 采用Origin 2019软件绘制图形。
通过莫利施反应得出小麦粉、大米粉、玉米粉、小米粉、高粱粉乙醇洗涤量为100 mL时出现黄绿色环; 土豆粉、板栗粉乙醇洗涤量为150 mL时出现黄绿色环。选取大米粉和板栗粉, 其他条件不变, 将乙醇洗涤量选定为0、50、100、150、200、250 mL分别洗涤样品, 测得的淀粉含量如图12所示。实验结果表明, 不同乙醇体积对不同样品洗涤效果不同, 大米粉洗涤量由0 mL增加至100 mL时, 测得的淀粉含量显著下降; 洗涤量为100 mL时, 洗去可溶性糖的效果最好; 洗涤量超过100 mL后, 洗涤效果基本无差别。板栗粉洗涤量由0 mL增加至150 mL时, 测得的淀粉含量显著下降; 洗涤量为150 mL时, 洗去可溶性糖的效果最好; 洗涤量超过150 mL后, 洗涤效果基本无差别。因此, 在其他条件不变的情况下, 同时考虑到节约成本及减少试剂对环境的污染, 小麦粉、大米粉、玉米粉、小米粉、高粱粉用100 mL乙醇洗涤; 土豆粉、板栗粉用150 mL乙醇洗涤, 与莫利施反应验证出的洗涤量相符合。
选取大米粉, 其他条件不变, 分别以同一浓度(1:1=V:V)不同体积(5、10、15、20、25、30、35、40、50、60 mL)盐酸对大米粉进行水解, 每个体积下平行3次, 所得淀粉含量如图3所示。实验结果表明, 盐酸用量由5 mL增加至15 mL时, 测得淀粉含量逐渐增加, RSD均大于2%; 由20 mL增加至30 mL时, 淀粉含量几乎不变, RSD均在1%左右; 由30 mL增加至50 mL时, 淀粉含量呈下降趋势, RSD逐渐增高且超过2%。其原因可能是盐酸用量小于20 mL时, 样品不能充分被酸解, 形成不稳定的糊状物和凝胶[23-24], 造成淀粉含量偏低; 盐酸用量大于30 mL时, 水解液色泽随着盐酸用量的增加而逐渐加深, 不利于pH调节, 造成RSD逐渐增加; 盐酸用量为20~30 mL时, 盐酸用量适中, 水解完全, 水解液色泽较浅, 便于pH调节时观察终点[25]。当盐酸用量为25 mL时, RSD最小, 故盐酸用量25 mL为最佳。
选取大米粉, 其他条件不变, 将冷凝回流时间选定为0、30、60、90、120、180、210 min, 每个时间下平行3次, 所得淀粉含量如图4所示。实验结果表明, 冷凝回流时间由0 min增加至120 min时, 测得淀粉含量逐渐增加, RSD逐渐降低; 由120 min增加至210 min时, 测得淀粉含量逐渐下降, RSD逐渐增高。其原因可能是冷凝回流时间小于120 min时, 酸水解时间过短, 淀粉可能刚只水解成糊精和麦芽糖, 还未完全水解成葡萄糖, 造成淀粉含量较低; 冷凝回流时间大于120 min时, 酸水解时间过长, 可能会发生副反应消耗掉部分葡萄糖, 造成淀粉含量持续减少。当冷凝回流时间为120 min时, 淀粉含量最高, RSD最小, 故冷凝回流时间120 min为最佳。
选取大米粉, 其他条件不变, 将试样水解液的pH调至为1~2、3~4、5~6、7~8、9~10、11~12, 每个pH下平行3次, 所得淀粉含量如图5所示。实验结果表明, 过酸过碱都会导致测得淀粉含量降低, pH为7~8时, 测得淀粉含量达到最大值, RSD较低, 故pH 7~8为最佳。
选取大米粉, 其他条件不变, 将碱性酒石酸铜溶液沸腾时间控制在1~2、2~3、3~4、4~5、5~6、6~7 min, 每个沸腾时间下平行3次。所得淀粉含量如图6所示。实验结果表明, 沸腾时间超过2 min, 随着沸腾时间的延长, 淀粉含量逐渐降低且RSD逐渐升高。其原因可能是滴定过程中稍过量的还原糖把次甲基蓝还原, 使蓝色的氧化型次甲基蓝还原为无色的还原型次甲基蓝, 即达滴定终点[26]。沸腾时间过长, 次甲基蓝、葡萄糖可能会与空气发生氧化还原反应, 此时就需要更大量的葡萄糖去还原次甲基蓝, 导致淀粉含量降低。因此在使用斐林试剂滴定法测定淀粉含量时, 要严格控制酒石酸铜溶液在2 min内加热至沸。
选取大米粉, 其他条件不变, 将滴定速度控制在每滴0~1、1~2、2~3、3~4、4~5 s, 每个滴定速度下平行3次。所得淀粉含量如表1所示。实验结果表明, 滴定速度过慢会导致测得淀粉含量偏低, RSD高于3%; 滴定速度过快会导致测得淀粉含量偏高, RSD高于2%; 滴定速度在2 s/滴左右时, 淀粉含量平稳, 且RSD低于1%。其原因可能是滴定速度过慢, 次甲基蓝、葡萄糖可能会与空气发生氧化还原反应, 此时就需要更大量的葡萄糖去还原次甲基蓝, 导致淀粉含量降低; 滴定速度过快, 次甲基蓝快速变成无色, 但一段时间后, 又会慢慢变回蓝色, 说明次甲基蓝没有完全被葡萄糖还原。因此在滴定过程中用秒表计时, 严格控制以2 s/滴的速度滴定。
选取大米粉, 其他条件不变, 将试样水解液的pH调至为5~6、6~7、7~8、8~9、9~10、10~11、11~12、12~13、13~14, 每个pH下平行3次, 所得淀粉含量如图7所示。实验结果表明, 样液pH为5~8时, 测得淀粉含量逐渐增加; 样液pH为8~10时, 测得淀粉含量趋势保持平稳; 样液pH为10~14时, 测得淀粉含量逐渐降低。其原因是还原糖与DNS的反应需要碱性条件, 在氢氧化钠存在下, 还原糖能将DNS中的硝基还原为氨基, 生成氨基化合物, 此化合物在碱性溶液中呈橘红色, 在540 nm波长处有最大吸收[27]。当样液pH为8~9时, 测得淀粉含量最大, RSD最低, 故pH 8~9最佳。
选取大米粉、板栗粉, 其他条件不变, 将乙酸铅用量选定为0、5、10、15、20、30、50 mL, 每个体积下平行3次。所得淀粉含量如图89所示。结果表明, 不同体积的乙酸铅用量对不同样品作用效果不同。大米粉乙酸铅用量由0 mL增加至15 mL时, 测得淀粉含量基本没有变化; 由15 mL增加至50 mL时, 测得淀粉含量逐渐降低。板栗粉乙酸铅用量由0 mL增加至10 mL时, 测得淀粉含量逐渐增加; 由10 mL增加至50 mL时, 测得淀粉含量逐渐降低。其原因可能是乙酸铅可以吸附颜色, 沉淀蛋白质、果胶等杂质。大米粉本身没有颜色且蛋白质含量较低, 故是否加入乙酸铅对淀粉含量没有影响, 相反加入过多乙酸铅, 沉淀较多, 后续无法过滤全部滤液, 造成淀粉含量降低。板栗粉本身有颜色且蛋白质含量较高, 加入10 mL乙酸铅溶液时, 板栗粉中蛋白质的沉淀效果较好, 得到的样品溶液透明澄清, 淀粉含量最高, RSD值最小; 少于10 mL时, 板栗粉样液色泽较深, 蛋白质沉淀不完全, 造成淀粉含量偏低; 大于10 mL时, 沉淀较多, 后续无法过滤, 造成淀粉含量降低。因此, 在其他条件不变的情况下, 同时考虑到节约成本及减少试剂对环境的污染, 大米粉不添加乙酸铅、板栗粉加入10 mL乙酸铅为最佳。
在碱性条件下, DNS与还原糖沸水浴中共热后生成棕红色的3-氨基-5硝基水杨酸[28]。在一定波长范围内, 棕红色物质颜色深浅与还原糖含量成正比。DNS比色法测定波长多集中在400~800 nm[29-32], 其最佳波长的确定受到仪器性能、实验样品和DNS试剂等因素的影响[33]。为了确定测定淀粉的最佳检测波长, 其他条件不变, 改变吸收波长, 在400、420、440、460、480、500、520、540、560、580、600 nm波长处进行测定, 结果如图10所示。实验结果表明, 尽管葡萄糖显色液在440~480 nm达到吸收最大值, 但是由于DNS试剂本身对该波段具有较高的吸收, 因此会干扰样品的检测结果。根据“吸收最大, 干扰最小”原则, 选择DNS本底吸收最小, DNS加葡萄糖吸收较高的波长作为最适吸收波长。当波长不短于540 nm时, DNS试剂自身的吸收对葡萄糖显色液的干扰可忽略, 同时DNS加葡萄糖的吸收强度较540 nm后高, 因此将540 nm作为检测波长。
选取大米粉, 其他条件不变, 将显色好的样液放入高精度恒温槽, 将温度分别设置成20、30、40 ℃, 模拟环境温度, 每个温度下平行3次。所得淀粉含量如表2所示。实验结果表明, 当温度为20 ℃或30 ℃时, 测得淀粉含量稳定, 但30 ℃下的RSD高于20 ℃; 当温度上升到40 ℃时, 吸光度值剧增且极不稳定。可以看出, 环境温度对DNS比色法测定淀粉含量具有较大的影响。因此, 在其他条件不变的情况下, 同时考虑到显色液稳定性与检测精密度, 选择环境温度为20 ℃最佳。
选取大米粉, 其他条件不变, 将DNS试剂加入量选定为0.5、1.0、1.5、2.0、2.5、3.0、4.0、5.0、6.0、7.0、8.0 mL, 每个加入量下平行3次。所得淀粉含量如图11所示。实验结果表明, 当加入量少于1.5 mL时, 测得淀粉含量超过100%; 当加入量为1.5~2.5 mL时, 测得淀粉含量趋于平稳; 当加入量多于2.5 mL时, 测得淀粉含量随着加入量的增加而逐渐降低。其原因可能是当DNS试剂加入量过少, 标准曲线灵敏度低, RSD高; 当加入量多于2.5 mL时, 样品中的葡萄糖已全部与DNS发生氧化还原反应生成3-氨基-5-硝基水杨酸, 吸光度值趋于平稳, 但过量的DNS导致灵敏度降低, 造成测得淀粉含量逐渐降低。因此, 在其他条件不变的情况下, 同时考虑到节约试剂与方法精密度, 选择DNS试剂加入量为2.0 mL最佳。
选取大米粉, 其他条件不变, 分别测定了加入DNS试剂摇匀待测液放进沸水浴后1、2、3、4、5、6、7、8、9、10、12、15、20 min的淀粉含量, 每个时间下平行3次。所得淀粉含量如图12所示。实验结果表明, 淀粉含量随着煮沸反应时间的延长先增加后趋于平稳, 在显色时间大于10 min后, 淀粉含量趋于稳定, 表明在10 min后DNS与样液中的葡萄糖完全反应, 但在12 min下RSD值最低。因此, 在其他条件不变的情况下, 同时考虑到淀粉含量稳定性与检测精密度, 选择显色时间12 min为最佳。
选取大米粉, 其他条件不变, 分别在50、60、70、80、85、90、95、100 ℃水浴中加热12 min, 每个温度下平行3次。所得淀粉含量如图13所示。实验结果表明, 显色温度达90~100 ℃时, 淀粉含量趋于稳定。因此, 在其他条件不变的情况下, 选择显色温度90~100 ℃为最佳。
选取大米粉, 其他条件不变, 试样水解液经显色反应后, 立即放置于自然环境、冷水、冰水中阻断反应, 每个条件下平行3次。所得淀粉含量如表3所示。实验结果表明, 自然凉或者凉水阻断显色反应, 测得淀粉含量基本没有变化且RSD值低于1%; 冰水阻断反应, 测得淀粉含量降低且RSD高于1%。其原因可能是冰水使反应温度突然骤降, 破坏反应体系, 造成淀粉含量降低, RSD提高。因此, 样品经DNS试剂显色完全后, 可放在通风橱中自然冷却或用流动自来水浸泡冷却。
选取大米粉, 其他条件不变, 分别将煮沸后的样液放置0、0.5、1、1.5、2、2.5、3、3.5、4、6、8 h后测其淀粉含量, 每个时间下平行3次。所得淀粉含量如图14所示。实验结果表明, 葡萄糖与DNS试剂反应生成的棕红色溶液在4 h内保持稳定。因此, 样品经DNS试剂显色后需在4 h内完成测定。
按照样品处理步骤, 进行标准曲线的绘制, 曲线回归方程为: Y=0.5911X-0.0003, r2=0.9998, 线性相关性良好。
按照优化后的条件制备空白样液12组并进行滴定, 以3倍标准偏差计算检出限。酸水解-斐林试剂滴定法检出限为1.31%, 酸水解-DNS比色法检出限为0.08%。
参照优化后的酸水解-斐林试剂滴定法和酸水解-DNS比色法, 每个样品平行测定6次。测定结果如表4所示, RSDs小于2%, 相对误差小于4%(该7个样品作为标准物质候选物已进行了多家实验室协作定值并经统计分析得到了标准值与不确定度, 相对误差是与定值所得标准值比较所得), 表明两种方法的精密度和正确度均良好。
参照优化后的酸水解-斐林试剂滴定法和酸水解-DNS比色法, 将7个样品每个方法平行测定6次, 计算平均值。将数据进行平均值一致性检验—t检验, 两种方法所测结果与∣t∣值见表5。结果表明∣t∣值小于t0.05, 证明两种测定方法的结果具有一致性。
将酸水解-斐林试剂滴定法与酸水解-DNS比色法测定7个样品数据进行配对样本t检验, 结果见表6, 所得t值均小于t0.95(5), 故滴定法与比色法测定结果之间无显著性差异。
通过对GB/T 5009.9—2023标准方法第二法酸水解法测定淀粉含量的实验研究, 本研究确定了影响测定准确性的关键条件并进行了有效优化。结果表明, 将样品用100 mL乙醇溶液(85%, V:V)洗除可溶性糖(土豆粉和板栗粉150 mL), 加入25 mL盐酸溶液于沸水浴中冷凝回流2 h, 于样液中滴1滴甲基红指示液, 利用氢氧化钠溶液(400 g/L)调至水解液刚好变成黄色, 定容至500 mL, 混匀, 过滤。将碱性酒石酸铜溶液在2 min内加热至沸, 保持沸腾状态以2 s/滴的速度滴定。该条件下的酸水解-斐林试剂滴定法检出限为1.31%, 方法相对标准偏差小于2%, 相对误差小于3%。本方法因所测结果RSD较低且检测过程中不易受各种因素影响使数据重现性好、稳定性高, 故从准确、精确、稳定等方面考虑, 该方法具有较强优势。但该方法操作较为复杂, 大批量测定样品时效率低, 人力物力耗费大, 而且该方法对实验人员操作水平要求较高, 滴定速度和反应液起沸时间需要操作人员严格控制, 滴定终点的判断依赖操作人员的丰富经验。
将DNS比色法和传统化学方法酸水解法相结合, 建立了采用DNS比色法测定粮食作物中淀粉含量的方法。结果表明: 洗糖、酸解条件同酸水解-斐林试剂滴定法, 回流完毕, 利用精密pH试纸调至水解液pH 8~9, 于土豆、板栗粉试样水解液中加入10 mL乙酸铅溶液和10 mL硫酸钠溶液, 定容至250 mL, 混匀, 过滤。加入DNS显色剂2.0 mL, 于90~100 ℃水浴加热12 min进行显色, 凉水阻断显色反应, 在吸收波长为540 nm处, 于20 ℃环境中, 在4 h内完成上机测定, 以葡萄糖标准溶液为溶剂制作标准曲线, 标曲线性相关性良好, 检出限为0.08%, 方法相对标准偏差小于2%, 相对误差小于4%。本方法适用范围广泛, 检出限低, 操作步骤简单, 反应快速, 适用于大规模样品的检测。但该方法测定结果容易被样品中的复杂基质干扰, 某些样品中天然存在的色素成分可能与显色剂发生竞争性反应, 从而影响测定结果的准确性和可靠性。因此, 结合具体样品优化检测条件是研究的重点。
斐林试剂滴定法属于常量分析法, DNS比色法属于微量分析法。通过平均值一致性检验与配对样本t检验, 得出优化后的两种方法测定结果之间没有显著性差异。本研究通过多方法一致性验证, 为淀粉含量检测工作提供了科学、可靠的技术方法。
