Objective To establish a method for the simultaneous determination of 26 kinds of veterinary drug residues in 3 categories including sulfonamide, quinolones and tetracyclines based on duck meat by ultra performance liquid chromatography-tandem triple quadrupole mass spectrometry. Methods The 0.1% formic acid acetonitrile extraction was used, QuEChERS purification agent was used to remove impurities. After nitrogen blowing in a 45 ℃ water bath to near dryness, the mixture was diluted with 0.1% formic acid water acetonitrile (1:1, V:V) and analyzed qualitatively and quantitatively using ultra performance liquid chromatography-tandem triple quadrupole mass spectrometry. Results The 26 kinds of veterinary drugs showed good linearity in the range of 2.0-300.0 ng/mL (r2≥0.9996), the limits of detection were 0.28-6.02 μg/kg, the limits of quantification were 11.28-15.03 μg/kg. The average recovery rates of 26 kinds of veterinary drugs at different concentration levels (5.0, 50.0, 300.0 μg/kg) were 75.08%-112.35% (n=15), and the relative standard deviations (RSDs) were 1.25%-7.88% (n=15). Conclusion This method features simple pretreatment operations, high efficiency, and low consumption of reagents and materials. It is suitable for the preliminary screening and detection of 26 kinds of veterinary drug residues in 3 categories for large quantities of duck meat samples.

目的 建立超高效液相色谱-串联三重四极杆质谱法同时测定鸭肉中磺胺类、四环素类、氟喹诺酮类, 共3类26种兽药残留的分析方法。方法 采用0.1%的甲酸-乙腈提取, QuEChERS净化剂除杂, 经45 ℃水浴氮吹至近干后0.1%甲酸水-乙腈(1:1, V:V)定容, 通过超高效液相色谱-串联三重四极杆质谱仪进行定性及定量检测分析。结果 26种兽药在2.0~300.0 ng/mL质量浓度范围内线性关系良好(r2≥0.9996), 检出限为0.28~6.02 μg/kg, 定量限为11.28~15.03 μg/kg。在不同浓度添加水平(5.0、50.0、300.0 μg/kg)下26种兽药的平均回收率为75.08%~112.35% (n=15), 相对标准偏差(relative standard deviations, RSDs)为1.25%~7.88% (n=15)。结论 该方法前处理操作简便, 效率高, 试剂耗材用量小, 适用于大批量鸭肉样品中此3类26种兽药残留的初筛检测。

目的 建立超高效液相色谱-串联三重四极杆质谱法同时测定鸭肉中磺胺类、四环素类、氟喹诺酮类, 共3类26种兽药残留的分析方法。方法 采用0.1%的甲酸-乙腈提取, QuEChERS净化剂除杂, 经45 ℃水浴氮吹至近干后0.1%甲酸水-乙腈(1:1, V:V)定容, 通过超高效液相色谱-串联三重四极杆质谱仪进行定性及定量检测分析。结果 26种兽药在2.0~300.0 ng/mL质量浓度范围内线性关系良好(r2≥0.9996), 检出限为0.28~6.02 μg/kg, 定量限为11.28~15.03 μg/kg。在不同浓度添加水平(5.0、50.0、300.0 μg/kg)下26种兽药的平均回收率为75.08%~112.35% (n=15), 相对标准偏差(relative standard deviations, RSDs)为1.25%~7.88% (n=15)。结论 该方法前处理操作简便, 效率高, 试剂耗材用量小, 适用于大批量鸭肉样品中此3类26种兽药残留的初筛检测。

Objective To establish a method for the simultaneous determination of 26 kinds of veterinary drug residues in 3 categories including sulfonamide, quinolones and tetracyclines based on duck meat by ultra performance liquid chromatography-tandem triple quadrupole mass spectrometry. Methods The 0.1% formic acid acetonitrile extraction was used, QuEChERS purification agent was used to remove impurities. After nitrogen blowing in a 45 ℃ water bath to near dryness, the mixture was diluted with 0.1% formic acid water acetonitrile (1:1, V:V) and analyzed qualitatively and quantitatively using ultra performance liquid chromatography-tandem triple quadrupole mass spectrometry. Results The 26 kinds of veterinary drugs showed good linearity in the range of 2.0-300.0 ng/mL (r2≥0.9996), the limits of detection were 0.28-6.02 μg/kg, the limits of quantification were 11.28-15.03 μg/kg. The average recovery rates of 26 kinds of veterinary drugs at different concentration levels (5.0, 50.0, 300.0 μg/kg) were 75.08%-112.35% (n=15), and the relative standard deviations (RSDs) were 1.25%-7.88% (n=15). Conclusion This method features simple pretreatment operations, high efficiency, and low consumption of reagents and materials. It is suitable for the preliminary screening and detection of 26 kinds of veterinary drug residues in 3 categories for large quantities of duck meat samples.

我国是世界养禽大国, 从“十一五”期间开始, 水禽业多样化、差异化发展明显, 鸭肉产量得到大幅增加, 成为全球禽肉生产大国之一[1-2]。随着人们消费口味的多样化, 中国禽肉消费市场也不断扩大, 禽业产值逐年增加[3-5]。尽管中国禽业得到了长足的发展, 但仍然面临着一些挑战, 制约禽产品质量安全的不利因素横跨整个产业链, 牵涉众多要素, 而家禽饲养过程中, 饲料添加剂、兽药和相关生物制品等对产品质量安全影响较大, 主要表现在用药不规范以及药品质量导致的兽药残留超标、违禁药物检出等问题[6-10]。应用较为广泛的禽肉产品检测国家标准既受限于检测范围无法同时囊括所有禽产品种类, 又受限于只能检测单一兽药。传统的禽产品检测方法前处理过程极其烦琐, 效率不高且无法保证回收率。对于禽产品消费比例日益增加的鸭肉产品来说, 缺乏简单高效且能同时满足多种类兽药残留的检测方法。因此, 对同时检测除鸡肉外的禽肉产品中多种常规兽药残留的技术进行研究, 十分必要。本研究建立采用超高效液相色谱-串联三重四极杆质谱法(ultra performance liquid chromatography-triple quadrupole mass spectrometry, UPLC-TQMS)同时检测鸭肉中磺胺类、四环素类、氟喹诺酮类共3类26种兽药残留的方法, 可对日常大批量鸭肉样品进行此26种兽药残留的筛查, 为鸭肉产品中相关限用药物快速、准确的监督检测发挥积极的作用。
鸭肉(去羽毛、去皮、去筋膜, 取胸脯处新鲜肉样)样品(1000±1) g/个, 均需现场制样, 样品要充分混匀, 等分3份, 每份不得少于200 g, 共50份, -20 ℃保存, 均为本实验室日常抽检样品。
乙腈、甲酸(质谱纯, 赛默飞世尔科技中国有限公司); 正己烷(分析纯, 天津市科密欧化学试剂有限公司); 四环素类、氟喹诺酮类、磺胺类标准品(含量≥95.0%, 北京坛墨质检科技股份有限公司); 自配QuEChERS净化剂[乙二胺-N-丙基硅烷:C18:无水硫酸钠:乙二胺-N-丙基硅烷(primary secondary amine, PSA)=3:5:10:60, 质量比](分析纯, 阿拉丁试剂上海有限公司); 实验室用水均为一级水。
LC-JA1003千分之一天平(感量0.001 g)、LC-LX-H185C普通高速离心机(浙江力辰仪器科技有限公司); KZ-GDZ-60L超纯水机(上海科治环保设备有限公司); XU-DCY24Y圆形水浴氮吹仪(上海析牛仪器科技有限公司); MK-16R高速冷冻离心机(湖南迈克尔实验室仪器有限公司); H-Class/TQ-S Micro超高效液相色谱-串联三重四极杆质谱联用仪、ACQUITY UPLC BEH C18柱(2.1 mm×100 mm, 1.7 μm)(美国沃特斯公司)。
精密称取鸭肉试样(1±0.02) g, 置于50 mL离心管中, 加入10.0 mL 0.1%甲酸-乙腈, 均质提取5 min, -4 ℃冷冻条件下10000 r/min离心10 min, 取全部上清液。上清液中加入自配QuEChERS净化剂5~8 g, 涡旋振荡3~5 min, 4500 r/min离心10 min, 精密移取备用液10.0 mL, 45 ℃氮吹至近干, 准确加入流动相1.0 mL (0.1%甲酸-水:乙腈=1:1, V:V)溶解残渣, 涡旋混合20~30 s, 过0.22 μm滤膜, 采用UPLC H-Class/TQ-S Micro仪器进行检测(按此操作步骤制备不含本研究中26种兽药残留的阴性鸭肉空白样品基质溶液)。
(1)色谱条件
色谱柱为ACQUITY UPLC BEH C18柱(2.1 mm×100 mm, 1.7 μm), 柱温35 ℃, 样品温度25 ℃, 流动相: A为0.1%甲酸水溶液; B为乙腈, 流速0.3 mL/min, 进样量2.0 μL。
(2)质谱条件
采用电喷雾正离子源(electro spray ionization, ESI+)在多反应监测(multiple reaction monitoring, MRM)模式下进行检测。其中, 脱溶剂气流速1000 L/h(脱溶剂气为氮气), 脱溶剂温度500 ℃, 锥孔气流量50 L/h, 毛细管电压3.0 kV(碰撞气体为氩气), 离子源温度150 ℃。
(3)检出限及定量限
将混合标准溶液加入空白式样中, 配制成的阳性样品残留量分别为0.1、0.5、1.0、5.0、10.0、20.0、50.0、100.0、200.0 μg/kg, 按1.4.1的方法进行前处理, 1.4.2中(1)与(2)的方法进行上机检测, 以信噪比等于3 (S/N=3)时的检测浓度为检出限, 以信噪比等于10 (S/N=10)时的检测浓度为定量限[11-16]
(4)线性回归关系
用初始流动相(0.1%甲酸水:乙腈=95:5, V:V)将混合标准溶液逐步稀释配制成2.0、5.0、10.0、50.0、100.0、200.0、300.0 ng/mL的混合标准工作液。以标准溶液质量浓度(X, ng/mL)为横坐标, 对应峰面积(Y)为纵坐标绘制梯度标准曲线, 计算回归方程与相关系数。
(5)准确度与精密度
向空白式样中分别添加5.0、50.0、300.0 μg/kg不同浓度水平的标准溶液, 各浓度重复15次, 按1.4.1的方法进行前处理, 1.4.2中(1)与(2)的方法进行上机检测, 计算回收率与相对标准偏差(relative standard deviation, RSD), 以此验证本方法的准确度和精密度。
(6)鸭肉中26种兽药残留样品的测定
2023年1—12月在黔东南自治州12个区域共20个采样点随机采取鸭肉样品50份。经上述1.4.1方法进行前处理, 采用UPLC H-Class/TQ-S Micro仪器进行检测, 外标法定量。
本研究目标兽药离子对定性识别及图谱定量积分均在Waters masslynx软件中完成。所有检测结果均通过至少3次测定后取其平均值(加标回收实验为15次平行)进行分析。采用WPS office Excel 2024进行数据整理与统计学分析并结合Powerpoint 2017绘制图表。
(1)提取试剂的选择
乙腈与水和醇都能无限互溶, 是前处理步骤中理想提取溶剂的首选。在酸化乙腈中正离子物质更容易溶解, 提取效果更好, 回收率更高[17-18]。乙二胺四乙酸(ethylene diamine tetraacetic acid, EDTA)缓冲溶液能够在一定范围内稳定溶液的pH, 这对于维持生物分子的活性和稳定性至关重要。通过查阅GB/T 20759—2006《畜禽肉中十六种磺胺类药物残留量的测定 液相色谱-串联质谱法》、GB/T 21312—2007《动物源性食品中14种喹诺酮药物残留检测方法 液相色谱-质谱/质谱法》、GB/T 21317—2007《动物源性食品中四环素类兽药残留量检测方法 液相色谱-质谱/质谱法》及相关文献[19-20], 在预实验中选用此3种溶液(EDTA-Mcllvaine溶液、乙腈、0.1%甲酸乙腈)作为提取溶剂进行比较, 发现0.1%甲酸乙腈提取率最高, 故选择0.1%甲酸乙腈作为提取溶剂, 详见图1
(2)净化试剂的选择
兽药检测中脂肪等大分子杂质的去除至关重要, 一方面杂质去除不完全会影响目标物在仪器上的响应值, 另一方面过度除杂会降低回收率[21-22]。现行有效的净化方法, 一是液液萃取: 采用如正己烷、乙酸乙酯以及其水饱和溶剂等进行除杂; 二是固相萃取: 采用固相小柱及固相萃取仪进行除杂, 前者操作费时, 易乳化, 有机溶剂消耗量大, 对环境不友好; 后者使用固相萃取小柱, 成本较高, 批次间的重复性难以保证。因此, 本研究针对鸭肉样品中杂质的去除, 探索自配一种QuEChERS净化剂, 以“试剂包”的形式进行样品净化处理, 详见图2
(1)色谱条件的选择
参考GB 31658.17—2021《动物性食品中四环素类、磺胺类和喹诺酮类药物残留量的测定 液相色谱-串联质谱法》、农业部1077号公告-1-2008《水产品中17种磺胺类及15种喹诺酮类药物残留量的测定 液相色谱-串联质谱法》等方法的色谱条件, 配制上机质量浓度为100 ng/mL的混合标准品溶液进行液相梯度测试, 选取目标药物全部出峰、峰型好、基线稳定的条件, 最终形成本研究中26种兽药检测的色谱梯度, 见表1
(2)质谱条件的选择
参考GB 31658.17—2021, 配制上机质量浓度为100 ng/mL的混合标准品溶液进行质谱调谐, 不断优化离子源各项参数, 选取各个目标药物同一液相条件下响应最高的离子对, 最终形成26种兽药检测的质谱离子对扫描参数, 见表2
在选定的色谱及质谱条件下, 对配制好的2.0~300.0 ng/mL混合标准工作液进行检测, 得出目标物浓度在范围内与其峰面积的线性关系。由表3可见: 该方法在2.0~300.0 ng/mL质量浓度范围内线性关系及重现性优异(相关系数r2≥0.9996), 符合检测要求。
按1.4.1的方法进行前处理, 1.4.2中(1)与(2)的方法进行上机检测, 根据1.4.2中(3)方法测得26种兽药在鸭肉中的检出限为0.28~6.02 μg/kg, 定量限为11.28~15.03 μg/kg。本方法检出限与定量限均低于相关指标的传统检测标准, 方法的检出限与定量限不仅与仪器噪音有关, 而且还取决于前处理流程的各个环节, 说明本方法采用的仪器性能稳定, 前处理步骤与分析方法具备灵敏的定量检测能力。
分别向空白试样中添加低(5.0 μg/kg)、中(50.0 μg/kg)、高(300.0 μg/kg)不同浓度的26种兽药进行回收率实验。由表4可见: 不同浓度的平均回收率为75.08%~112.35% (n=15), RSDs为1.25%~7.88% (n=15)。对比相关兽药多残留检测方法, 本方法回收率较高, 表明本方法准确性好; RSD较低, 表明数据重复性及精密度高, 由此看来, 本研究优化过程合理, 实验检测结果可靠。
50份鸭肉样品中共检出1份阳性样品, 检出指标为四环素, 残留量为52.4 μg/kg, 未超过国家规定的最高残留限量(200 μg/kg)。同时采用GB/T 21317—2007对此份阳性样品进行复验, 结果均无明显差异(初验与复验组间RSD<3.29%)。
本研究综合国家现行标准及相关文献研究[23-28], 对鸭肉中26种限用兽药残留的检测方法进行开发: (1)针对正离子指标, 将0.1%甲酸乙腈作为提取试剂, 采用QuEChERS试剂包净化大分子杂质, 替代了传统单一提取试剂, 简化了净化步骤, 保证回收率的同时也提高了操作效率; (2)针对三重四极杆质谱仪在酸性流动相的情况下对正离子物质扫描精确度更高的特性[29-32], 流动相由0.1%甲酸水与乙腈组成, 梯度洗脱, 同时采用针对兽药分离度较好的ACQUITY UPLC BEH C18色谱柱, 对本研究中26种兽药指标同时进行离子对扫描检测, 重现性佳。本方法可同时检测鸭肉中共3类26种兽药, 准确度高、精密性好, 适用于大批量鸭肉中限用常规药物的同时快速筛查, 对于承担大批量禽肉产品风险监测任务的机构来说, 在提高检测效率与节约实验成本方面均提供了有效帮助。
