Objective To establish a method for the qualitative and quantitative determination of a new illegal additive in foods, vardenafil impurity 31 by ultra performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS). Methods The UPLC-MS/MS was used to screen 92 kinds of substances of nafils illegally added in foods. The ultra performance liquid chromatography-high resolution mass spectrometry (UPLC-HRMS) was used to analyze the structure of the test sample. After the structure was inferred, the standard substance was purchased and verified. The target substance in the test sample was confirmed to be vardenafil impurity 31. The multiple reaction monitoring (MRM) method was established by UPLC-MS/MS, which was suitable for the qualitative and quantitative determination of vardenafil impurity 31 in foods. Results The limit of detection of vardenafil impurity 31 was 5 μg/kg, and the limit of quantitation was 10 μg/kg. The linear relationship was good in the linear range of 0.1-50.0 ng/mL, the correlation coefficient (r) was 0.9976, the average recovery rate was 85.33%-89.41%, and the relative standard deviation (RSD) was less than 10%. Conclusion The established method is simple and accurate, and can be used for the qualitative and quantitative determination of the new illegal additive vardenafil impurity 31 in foods.

目的 建立超高效液相色谱-串联质谱法(ultra performance liquid chromatography-tandem mass spectrometry, UPLC-MS/MS)测定食品中一种新型非法添加物伐地那非杂质31的定性定量检测方法。方法 运用UPLC-MS/MS对待测试样中92种那非类非法添加物质进行筛查, 运用超高效液相色谱-高分辨质谱法(ultra performance liquid chromatography-high resolution mass spectrometry, UPLC-HRMS)对待测试样进行结构解析, 推断出结构, 购入标准品经验证后, 确认待测试样中目标物为伐地那非杂质31, 采用UPLC-MS/MS建立多反应监测(multiple reaction monitoring, MRM)方法, 定性定量测定食品中伐地那非杂质31。结果 本方法伐地那非杂质31的检出限为5 μg/kg, 定量限为10 μg/kg。在0.1~50.0 ng/mL的线性范围内线性关系良好, 相关系数(r)为0.9976, 平均加标回收率在85.33%~89.41%之间, 相对标准偏差(relative standard deviation, RSD)小于10%。结论 所建立的检测方法简便准确, 可用于食品中新型非法添加物伐地那非杂质31的定性定量测定。

目的 建立超高效液相色谱-串联质谱法(ultra performance liquid chromatography-tandem mass spectrometry, UPLC-MS/MS)测定食品中一种新型非法添加物伐地那非杂质31的定性定量检测方法。方法 运用UPLC-MS/MS对待测试样中92种那非类非法添加物质进行筛查, 运用超高效液相色谱-高分辨质谱法(ultra performance liquid chromatography-high resolution mass spectrometry, UPLC-HRMS)对待测试样进行结构解析, 推断出结构, 购入标准品经验证后, 确认待测试样中目标物为伐地那非杂质31, 采用UPLC-MS/MS建立多反应监测(multiple reaction monitoring, MRM)方法, 定性定量测定食品中伐地那非杂质31。结果 本方法伐地那非杂质31的检出限为5 μg/kg, 定量限为10 μg/kg。在0.1~50.0 ng/mL的线性范围内线性关系良好, 相关系数(r)为0.9976, 平均加标回收率在85.33%~89.41%之间, 相对标准偏差(relative standard deviation, RSD)小于10%。结论 所建立的检测方法简便准确, 可用于食品中新型非法添加物伐地那非杂质31的定性定量测定。

Objective To establish a method for the qualitative and quantitative determination of a new illegal additive in foods, vardenafil impurity 31 by ultra performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS). Methods The UPLC-MS/MS was used to screen 92 kinds of substances of nafils illegally added in foods. The ultra performance liquid chromatography-high resolution mass spectrometry (UPLC-HRMS) was used to analyze the structure of the test sample. After the structure was inferred, the standard substance was purchased and verified. The target substance in the test sample was confirmed to be vardenafil impurity 31. The multiple reaction monitoring (MRM) method was established by UPLC-MS/MS, which was suitable for the qualitative and quantitative determination of vardenafil impurity 31 in foods. Results The limit of detection of vardenafil impurity 31 was 5 μg/kg, and the limit of quantitation was 10 μg/kg. The linear relationship was good in the linear range of 0.1-50.0 ng/mL, the correlation coefficient (r) was 0.9976, the average recovery rate was 85.33%-89.41%, and the relative standard deviation (RSD) was less than 10%. Conclusion The established method is simple and accurate, and can be used for the qualitative and quantitative determination of the new illegal additive vardenafil impurity 31 in foods.

近年来, 食品中非法添加药物的现象趋于多元化、隐蔽化, 许多不法商贩为了使产品达到宣传的功效, 会在食品、保健品中添加危害身体健康的药物成分, 从而牟取暴利[1-2], 并且添加药物的种类逐渐从广为熟知的化合物变成相关衍生物等[3-5]。食品中非法添加药物的检测手段有光谱法[6-8]、色谱法[9-10]以及质谱法[11-15], 目前以液相色谱法-串联质谱法应用为主[16], 另外还有高分辨质谱法(high resolution mass spectrometry, HRMS)[17-18]及定量核磁共振技术等用于新型非法添加的筛查和确证[19-25]。以宣传壮阳、缓解疲劳等功效的食品及保健品为例, 常见非法添加物质为那非类、拉非类物质, 是5型磷酸二酯酶的抑制剂化合物(5-type phosphodiesterase inhibitors, PDE-5)[26], 主要代表物质为西地那非、他达拉非、伐地那非、红地那非等, 这类物质在医学上可用于治疗男性的勃起功能障碍, 但将药品或其他非食用物质添加在食品中已经违反了我国的《食品安全法》[27]。目前国家市场监督管理总局于2018年发布了食品补充检验方法BJS 201805《食品中那非类物质的测定》, 涵盖了90种那非、拉非类物质, 适用于饮料、果冻、蛋白粉、牡蛎粉、饼干、糖果、酒类及咖啡等食品中那非类物质的测定, 但是那非类衍生物可能远不止这90种, 国家市场监督管理总局于2022年又发布了《市场监管总局办公厅关于打击食品中非法添加物那非拉非类物质及其系列衍生物违法行为的意见》并发布了新型非法添加物苯丙代卡巴地那非的测定方法, 2023年国家市场监督管理总局再次发布了食品中伐地那非杂质30的测定方法, 但迄今为止, 官方还未发布关于食品中伐地那非杂质31的测定方法, 伐地那非杂质30或31是指在伐地那非这种药物的生产和存储过程中, 由于化学反应的副产品、原料药的不纯或者其他原因而产生的特定杂质, 其化学结构与伐地那非相似, 根据《市场监管总局办公厅关于打击食品中非法添加物那非拉非类物质及其系列衍生物违法行为的意见》, 伐地那非杂质与伐地那非核心药效团一致, 具有等同属性和等同危害, 食用添加有那非、拉非类物质及其衍生物的食品对人体有毒副作用的风险, 影响人体健康甚至危害生命。在实际食品非法添加案件中, 那非类结构类似物不断出现, 其结构未知且缺乏标准品, 亟需建立快速、准确的定性鉴定和定量分析方法, 为食品安全监管提供有力的技术支持。因而, 食品和保健食品中非法添加物筛查策略[28-30]显得尤为重要。
笔者在日常检验鉴定工作中发现公安送检的某产品经过上述92种那非类物质筛查后, 有疑似检出但保留时间无法匹配, 因此对该疑似那非类目标物进行结构确认, 推断其结构式疑似伐地那非杂质31, 并进一步购买伐地那非杂质31标准品进行确证后建立超高效液相色谱-串联质谱法(ultra performance liquid chromatography-tandem mass spectrometry, UPLC-MS/MS)定性定量检测, 经方法学验证后能够适用于食品中伐地那非杂质31的定性定量检验工作, 以期完善那非类非法添加谱库, 并为公安的非法添加案件检验鉴定问题提供参考。

Xevo TQ-XS超高效液相色谱-串联质谱仪、Waters ACQUITY UPLC C18色谱柱(2.1 mm×100 mm, 1.7 μm)(美国Waters公司); Agilent 1290 Infinity II-6546 QTOF液质联用系统、Agilent Eclipse C18 120色谱柱(3.0 mm×150 mm, 1.8 μm)(美国安捷伦科技有限公司); MS105十万分之一天平(瑞士梅特勒-托利多仪器有限公司); TG16-WS台式离心机(湖南湘仪实验室仪器开发有限公司); CR-020S超声清洗器(深圳市春霖清洗设备有限公司); MX-F涡旋振荡器(大龙兴创实验仪器股份公司)。
用甲醇将90种那非混合标准溶液(质量20 μg/mL)(组分为BJS 201805标准中涉及的90种那非类物质)、甲醇中伐地那非杂质30溶液(100 μg/mL)和甲醇中苯丙代卡巴地那非溶液(100 μg/mL)分别稀释至1 μg/mL, 备用; 将1 mg伐地那非杂质31对照品全部溶解于1 mL甲醇中, 得到1 mg/mL的甲醇中伐地那非杂质31标准储备溶液, 并用甲醇逐级稀释至1 μg/mL, 备用。
称取待测样品(某品牌咖啡)0.2 g(精确到0.0001 g)于10 mL具塞离心管中, 先加入8 mL乙腈, 超声提取15 min后冷却至室温, 4500 r/min下离心5 min后将上清液转移至10 mL容量瓶中, 用乙腈定容至刻度, 取适量上清液过0.22 μm滤膜, 取续滤液上机。
称取空白速溶咖啡样品适量, 添加已知浓度的目标物, 与待测试样同法处理, 制得质控样品。
ACQUITY UPLC I-Class液相色谱条件: 流动相A为含5 mmol/L乙酸铵的0.1%甲酸水溶液; 流动相B为质谱纯乙腈; 流速为0.3 mL/min; 色谱柱型号为Waters ACQUITY UPLC C18 (2.1 mm×100 mm, 1.7 μm); 柱温设置为40 ℃; 梯度洗脱程序: 0~0.5 min, 70% A; 0.5~3.5 min, 70%~35% A; 3.5~4.0 min, 35%~5% A; 4.0~5.0 min, 5% A; 5.0~5.1 min, 5%~70% A; 5.1~6.5 min, 70% A。
TQ-XS质谱条件: 电喷雾离子源(electron spray ionization, ESI); 正离子扫描模式; 多重反应监测(multiple reaction monitoring, MRM)模式, 母离子m/z 519.3, 锥孔电压为25 V; 定量离子m/z 151.1, 碰撞能量为30 eV; 定性离子m/z 299.1, 碰撞能量为32 eV。
Agilent 1290 Infinity II-6546 QTOF液相色谱条件: 流动相A为0.1%甲酸水溶液; 流动相B为质谱纯甲醇; 流速为0.4 mL/min; 色谱柱型号为Agilent Eclipse C18 120 (3.0 mm×150 mm, 1.8 μm); 柱温设置为35 ℃; 梯度洗脱程序: 0~1 min, 90%~50% A; 1~16 min, 50%~35% A; 16~19 min, 35%~2%; 19~22 min, 2% A; 22~22.1 min, 2% A; 22.1~26 min, 90% A。
6546 QTOF质谱条件: 增强型喷射流电喷雾离子源(advanced jet stream electron spray ionization, AJS ESI)离子源; 正离子扫描模式; 干燥气温度: 300 ℃; 干燥气流速: 10 L/min; 雾化器压力: 45 psi; 鞘气温度: 350 ℃; 鞘气流速: 11 L/min; 毛细管电压: 4000 V; 喷嘴电压: 0 V; 全扫描一级质谱, 质量采集范围为50~1100 Da; 全扫描二级质谱, 质量采集范围为50~1100 Da。
UPLC-MS/MS数据采用MassLynx V4.2软件收集和处理, HRMS数据使用Agilent MassHunter Acquisition V10.0软件采集, 并使用Agilent MassHunter Qualitative Analysis, V10.0分析处理, 同时使用WPS Office 2019进行图表的绘制。
待测样品(某品牌咖啡)来自于某公安局, 怀疑其中含有那非类物质, 首先按照BJS 201805对待测样品进行处理后上机检测, 未检出标准中涵盖的90种那非类物质、伐地那非杂质30和苯丙代卡巴地那非, 但样品在丙氧苯基羟基豪莫西地那非离子通道中出现了1个响应很高的色谱峰, 保留时间为2.22 min, 其特征碎片与丙氧苯基羟基豪莫西地那非符合, 但标准品丙氧苯基羟基豪莫西地那非的保留时间为2.57 min, 两者不匹配(见图1)。经分析推断待测样品中可能含有一种类似物或衍生物, 需进一步确认。
按照1.5.2条件进行定性筛查, 在咖啡样品的总离子流色谱图的7.454 min处出现1个色谱峰(见图2), 根据二级质谱图(见图3), 其准分子离子峰为m/z 519.2390([M+H]+), 利用安捷伦定性软件分析匹配m/z 519.2389的分子式为C24H34N6O5S, 推测该化合物的分子式(C24H34N6O5S)与丙氧苯基羟基豪莫西地那非分子式相同, 利用Agilent已有PCDL (personal compound database and library)数据库中化合物二级谱库, 对目标物进行比对发现, 目标物分子式虽与丙氧苯基羟基豪莫西地那非分子式相同, 但二者的二级质谱匹配得分不高, 可排除目标物为丙氧苯基羟基豪莫西地那非的可能性。
图3可见, 目标物存在一个主要碎片离子, m/z为151.0868, 分析那非类主要代表物西地那非和伐地那非的二级质谱图发现, 伐地那非的二级碎片离子包含m/z为151.0867, 与目标物非常接近, 因此本研究比较了伐地那非、羟基伐地那非和目标物的二级质谱图, 均包含特征碎片离子m/z为151.0868和m/z为376.1079, 推测目标物可能为伐地那非类似物, 利用安捷伦MSC (molecular structure correlator)软件和Sirus质谱数据分析软件结合化合物二级质谱图推测出化合物的可能结构式(图4a), 与伐地那非结构(图4b)相比丙氧基替代了乙氧基, 另外多一个-OH取代, 本研究根据分子式和推断的结构式查找化源网, 发现可能为伐地那非杂质31, 因此立即购置了标准品进行确认。
根据分子式为C24H34N6O5S及推断出的结构式, 本研究购置了伐地那非杂质31对照品, 并用甲醇作为溶剂配制了伐地那非杂质31对照品, 蠕动泵进样优化质谱参数, 得到母离子碎片为m/z 519.3, 子离子碎片为m/z 151.1和m/z 299.1, 进一步优化液相色谱参数、质谱参数得到1.5.1条件。用10 mL甲醇浸提待测样品, 按照上述仪器条件测定, 与伐地那非杂质31对照品比对, 从图5可见, 伐地那非杂质31(图5a)和待测样品(图5b)均在2.27 min左右处出现了特征峰, 离子丰度比误差<20%, 满足定性条件, 可确认待测样品中含有伐地那非杂质31, 且从峰高可看出待测样品中伐地那非杂质31含量较高。
考虑到以往发布的关于那非类物质的检测方法均采用甲醇作为提取溶剂, 本研究选择速溶咖啡粉作为空白样品, 按照1.4.2操作, 制得伐地那非杂质31含量为250 μg/kg的质控样品, 然后先加入8 mL甲醇超声提取15 min后冷却至室温, 4500 r/min下离心5 min后将上清液转移至10 mL容量瓶中, 用甲醇定容至刻度, 取适量上清液过0.22 μm滤膜, 取续滤液上机。重复6次测定, 计算得到回收率在30%~55%之间。
本研究首先考察了不同提取溶剂和提取时间对提取效果的影响, 在4组实验条件(见表1)下各重复测定质控样品6次, 通过分析对比平均加标回收率发现, 乙腈作为提取溶剂时, 加标回收率在100%以上, 而甲醇作为提取溶剂时, 加标回收率约为30%~40%。可见使用乙腈的提取效果更好。
同时也发现, 超声提取时间15 min和30 min两种条件下, 加标回收率差异不明显。因此最终确定乙腈作为提取溶剂, 超声提取时间为15 min。
取空白咖啡样品, 按照1.4.1进行提取, 得到空白基质提取液过0.22 μm滤膜, 然后用于稀释系列标准曲线溶液, 得到伐地那非杂质31质量浓度为0.1、0.2、1.0、5.0、20.0、50.0 ng/mL的标准曲线, 上机测定, 同步测定空白提取液。外标法定量, 以质量浓度(X, ng/mL)为横坐标, 峰面积(Y)为纵坐标, 用MassLynx V4.2处理数据绘制标准曲线, 得到线性方程为Y=105225X+4771.89, 线性系数r=0.9976, 目标物在0.1~50.0 ng/mL的线性范围内线性关系良好。另外按照1.4.2制备伐地那非杂质31含量分别为5 μg/kg和10 μg/kg的质控样品, 按照1.5.1平行测定7次, 上机测定响应分别满足S/N≥3、S/N≥10的要求, 因而以5 μg/kg为检出限, 以10 μg/kg为定量限。
按照1.4.2制得伐地那非杂质31含量分别为10、25、250 μg/kg的低中高3个水平的质控样品, 按照1.5.1平行测定6次, 计算平均回收率和精密度。由表2可见, 不同加标水平下, 回收率在85.33%~89.41%之间, RSDs在1.27%~3.93%之间, 满足分析要求。
采用本研究建立方法对以往检测过那非类非法添加的40个样品进行复测, 发现3款咖啡中出现了伐地那非杂质31的特征峰, 保留时间、离子丰度比与对照品相匹配, 可认定在该3款产品中检出了伐地那非杂质31成分, 含量在1.0×104~5.0×104 mg/kg之间。
本研究通过UPLC-MS/MS筛查、HRMS结构解析、标准品确证, 发现并鉴定了一种新型的非法添加物伐地那非杂质31, 该化合物尚无相关检验标准, 因此本研究建立了食品中伐地那非杂质31的UPLC-MS/MS检测方法, 考察了乙腈和甲醇分别作为提取溶剂以及超声时间对提取效果的影响, 以加标回收率和精密度作为考察指标, 最终确定乙腈的提取效果优于甲醇, 同时发现超声时间15 min和30 min的差异不大, 因此确定乙腈作为提取溶剂, 超声提取时间为15 min。同时对方法的检出限、定量限、标准曲线相关性进行验证实验, 实验结果表明在伐地那非杂质31质量浓度在0.1~50.0 ng/mL范围内线性关系良好, 相关系数(r)为0.9976, 检出限为5 μg/kg, 定量限为10 μg/kg。并对空白样品进行10、25、250 μg/kg 3个添加水平的加标回收率实验, 回收率在85.33%~89.41%之间, 精密度良好, RSDs均小于10%。该方法提取简单, 回收率稳定, 灵敏度高, 能够实现食品中伐地那非杂质31的鉴定检测工作。笔者将该方法用于食品非法添加案件样品检测, 发现有3款咖啡中检出了伐地那非杂质31成分, 本检测方法可以助力食品安全检测工作的开展。
