Objective To develop a solid beverage with Polygonatum sibiricum-Lycium barbarum as the main raw materials. Methods Using polysaccharides as the indicator, the total polysaccharides were extracted using the ultrasonic method. The extraction process of Polygonatum sibiricum and Lycium barbarum was determined through single-factor and orthogonal tests. The extract was concentrated, and the extract powder was obtained using wet granulation and drying. The appearance and quality of the granules were assessed as the evaluation indicator, and the effects of different amounts of main raw materials and excipients on the granule characteristics were studied through single-factor and orthogonal tests. Excipients such as sucrose powder, DL-malic acid and β-cyclodextrin were added. Using sensory evaluation as the indicator, the optimal formulation for Polygonatum sibiricum-Lycium barbarum solid beverage was determined through single-factor and orthogonal tests, ultimately obtaining the optimal preparation process. Results The optimal extraction conditions were as follows: Took Polygonatum sibiricum and Lycium barbarum medicinal materials 70 g with the ratio of 4:1 (m:m), then added 80 ℃ purified water. With an extraction time of 30 minutes, a liquid-to-material ratio of 30:1 (V:m), and an ultrasonic power of 250 W. The extract was concentrated to a thick paste, each 1 mL of which was equivalent to 1 g of the crude drug. The 70 mL extract paste was taken, and 15 mL of 95% ethanol and 250 g of maltodextrin were added. The mixture was granulated using the wet granulation method, and the granules were dried at 60 ℃ for 4 hours, 20 g extract particles were taken. The final extract granules were mixed with 0.2% DL-malic acid, 5 g (25%) sucrose powder, and 0.06% β-cyclodextrin, then divided and packed into 12 bags, each containing 25 g. Under this preparation process, the validation test was conducted. The average extraction rate of polysaccharides was 10.633%, with a relative standard deviation (RSD) of 0.235%. The average score for granule evaluation was 87.200, with an RSD of 0.344%. The average score for sensory evaluation was 86.133, with an RSD of 0.408%. Conclusion The entire preparation process demonstrates high repeatability and excellent stability. The solid beverage prepared by this process, when brewed, exhibits a uniform color, a sweet and sour taste, and a refreshing aroma of Polygonatum sibiricum and Lycium barbarum. It is a highly nutritious functional product.

目的 以黄精、枸杞子为主要原料, 研制黄精枸杞固体饮料。方法 以多糖为指标, 采用超声法提取总多糖, 通过单因素及正交实验确定黄精、枸杞子原料的提取工艺, 将提取液浓缩后利用湿法制粒并干燥获得提取物, 以颗粒性状评分为指标, 通过单因素和正交实验研究主要原、辅料的不同添加量对提取物颗粒性状的影响。添加蔗糖粉、DL-苹果酸和β-环状糊精等辅料, 再以感官评分为指标, 利用单因素和正交实验确定黄精枸杞固体饮料调配的最佳配方, 最终得到最佳制备工艺。结果 最佳制备工艺条件为: 黄精和枸杞子(药材比例4:1, m:m) 70 g, 加入80 ℃纯净水超声提取30 min, 液料比30:1 (V:m), 超声功率250 W, 将提取液浓缩至每1 mL相当于原药材1 g的稠膏。取提取液稠膏70 mL, 加入95%乙醇15 mL、麦芽糊精250 g, 采用湿法制粒, 颗粒60 ℃干燥4 h, 取提取物颗粒20 g, 加入DL-苹果酸0.2%, 蔗糖粉5 g (25%), β-环状糊精0.06%, 混匀后分装成25 g/袋(12袋)。该制备工艺下进行验证实验, 多糖平均提取率为10.633%, 相对标准偏差(relative standard deviation, RSD)为0.235%; 颗粒评价平均得分为87.200分, RSD为0.344%; 感官评价平均得分为86.133分, RSD为0.408%。结论 整个制备工艺重复性高、稳定性好, 该工艺制备的固体饮料冲泡后颜色均匀、酸甜可口并且伴有黄精、枸杞子的清香, 是一款具有较高营养价值的功能性产品。

Research on the preparation process of Polygonatum sibiricum-Lycium barbarum solid beverage

目的 以黄精、枸杞子为主要原料, 研制黄精枸杞固体饮料。方法 以多糖为指标, 采用超声法提取总多糖, 通过单因素及正交实验确定黄精、枸杞子原料的提取工艺, 将提取液浓缩后利用湿法制粒并干燥获得提取物, 以颗粒性状评分为指标, 通过单因素和正交实验研究主要原、辅料的不同添加量对提取物颗粒性状的影响。添加蔗糖粉、DL-苹果酸和β-环状糊精等辅料, 再以感官评分为指标, 利用单因素和正交实验确定黄精枸杞固体饮料调配的最佳配方, 最终得到最佳制备工艺。结果 最佳制备工艺条件为: 黄精和枸杞子(药材比例4:1, m:m) 70 g, 加入80 ℃纯净水超声提取30 min, 液料比30:1 (V:m), 超声功率250 W, 将提取液浓缩至每1 mL相当于原药材1 g的稠膏。取提取液稠膏70 mL, 加入95%乙醇15 mL、麦芽糊精250 g, 采用湿法制粒, 颗粒60 ℃干燥4 h, 取提取物颗粒20 g, 加入DL-苹果酸0.2%, 蔗糖粉5 g (25%), β-环状糊精0.06%, 混匀后分装成25 g/袋(12袋)。该制备工艺下进行验证实验, 多糖平均提取率为10.633%, 相对标准偏差(relative standard deviation, RSD)为0.235%; 颗粒评价平均得分为87.200分, RSD为0.344%; 感官评价平均得分为86.133分, RSD为0.408%。结论 整个制备工艺重复性高、稳定性好, 该工艺制备的固体饮料冲泡后颜色均匀、酸甜可口并且伴有黄精、枸杞子的清香, 是一款具有较高营养价值的功能性产品。

Objective To develop a solid beverage with Polygonatum sibiricum-Lycium barbarum as the main raw materials. Methods Using polysaccharides as the indicator, the total polysaccharides were extracted using the ultrasonic method. The extraction process of Polygonatum sibiricum and Lycium barbarum was determined through single-factor and orthogonal tests. The extract was concentrated, and the extract powder was obtained using wet granulation and drying. The appearance and quality of the granules were assessed as the evaluation indicator, and the effects of different amounts of main raw materials and excipients on the granule characteristics were studied through single-factor and orthogonal tests. Excipients such as sucrose powder, DL-malic acid and β-cyclodextrin were added. Using sensory evaluation as the indicator, the optimal formulation for Polygonatum sibiricum-Lycium barbarum solid beverage was determined through single-factor and orthogonal tests, ultimately obtaining the optimal preparation process. Results The optimal extraction conditions were as follows: Took Polygonatum sibiricum and Lycium barbarum medicinal materials 70 g with the ratio of 4:1 (m:m), then added 80 ℃ purified water. With an extraction time of 30 minutes, a liquid-to-material ratio of 30:1 (V:m), and an ultrasonic power of 250 W. The extract was concentrated to a thick paste, each 1 mL of which was equivalent to 1 g of the crude drug. The 70 mL extract paste was taken, and 15 mL of 95% ethanol and 250 g of maltodextrin were added. The mixture was granulated using the wet granulation method, and the granules were dried at 60 ℃ for 4 hours, 20 g extract particles were taken. The final extract granules were mixed with 0.2% DL-malic acid, 5 g (25%) sucrose powder, and 0.06% β-cyclodextrin, then divided and packed into 12 bags, each containing 25 g. Under this preparation process, the validation test was conducted. The average extraction rate of polysaccharides was 10.633%, with a relative standard deviation (RSD) of 0.235%. The average score for granule evaluation was 87.200, with an RSD of 0.344%. The average score for sensory evaluation was 86.133, with an RSD of 0.408%. Conclusion The entire preparation process demonstrates high repeatability and excellent stability. The solid beverage prepared by this process, when brewed, exhibits a uniform color, a sweet and sour taste, and a refreshing aroma of Polygonatum sibiricum and Lycium barbarum. It is a highly nutritious functional product.

黄精枸杞固体饮料是一种以黄精和枸杞子为主要原料开发的功能性食品。黄精和枸杞子作为传统中药材, 于2012年被国家卫生健康委员会列入“既是食品又是药品”的中药名单[1-2], 兼具药用与食用价值, 为开发功能性食品提供了重要理论基础。
黄精为百合科植物, 其化学成分丰富, 主要包括多糖、黄酮类、皂苷类、生物碱、蒽醌类和木质素等[3-5]。其中, 黄精多糖是其主要活性成分, 具有抗氧化、抗衰老、免疫调节、抗肿瘤、抗骨质疏松及预防老年痴呆等多种生物活性[6-11]。目前, 黄精已被广泛应用于保健饮料、压片糖果、酸奶等多种功能性产品中[12-13], 显示出广阔的市场前景。
枸杞子为茄科植物, 富含多种药理活性成分, 具有保护肝肾、抗衰老、抗疲劳、免疫调节及保护视力[14-19]等多种功效。其主要化学成分包括生物碱、多糖、黄酮、氨基酸和维生素[20-24]等, 其中枸杞多糖是其核心活性成分, 具有显著的抗氧化和增强免疫活性的作用[25-28], 同时对枸杞子的品质、口感及加工性能具有重要影响[29]。枸杞子在功能食品领域应用广泛, 例如李金洋等[30]开发的枸杞决明子功能性酸奶便是一种典型的应用案例。
本研究基于“药食同源”理论, 结合黄精和枸杞子在抗氧化、滋补肝肾等方面的协同功效, 通过超声提取多糖、含量测定、干燥制粒及感官评价等综合技术, 开发出一款兼具中药清香与酸甜口感的功能性固体饮料。该产品不仅拓展了黄精和枸杞子的市场应用形式, 提升了其经济价值, 同时也为其规模化生产提供了技术参考, 对推动中药功能性食品的产业化发展具有重要意义。
黄精饮片(陕西步长制药有限公司略阳黄精基地); 枸杞子饮片(西安贝斯特药材市场); D-葡萄糖标准品(批号100082-201803, 纯度≥99%, 中国食品药品检定研究院); 95%乙醇、苯酚、浓硫酸(分析纯, 北京北化精细化学品有限责任公司); 麦芽糊精(食品级, 东营市晨发商贸有限公司); 蔗糖粉、DL-苹果酸、β-环状糊精(食品级, 河南万邦实业有限公司); 水为娃哈哈纯净水(杭州娃哈哈集团有限公司)。
TU-1900双光束紫外分光光度计(北京普析通用仪器有限公司); LC-JY92-ⅡN超声波细胞破碎仪(力辰仪器科技有限公司); KQ5200DE高功率数控超声波清洗器(昆山市超声仪器有限公司); SQP电子分析天平(精度0.01 g, 瑞安市乐祺贸易有限公司); DHG-9625A电热恒温鼓风干燥箱(上海一恒科学仪器有限公司)。
称取黄精碎块7.5 g、枸杞子碎块2.5 g于锥形瓶中, 加入10倍80 ℃的热水, 在300 W的频率下超声30 min, 过滤, 取1 mL滤液稀释250倍, 备用。
精密称取无水葡萄糖标准品10 mg, 加入适量的纯化水溶解, 移入100 mL的容量瓶中, 稀释至刻度后, 摇匀, 即得到质量浓度为0.1 mg/mL的葡萄糖对照品溶液。
精密吸取对照品溶液和供试品溶液各1.0 mL于具塞试管中, 加入蒸馏水至2.0 mL, 摇匀。加入5%苯酚试剂(现用现配)1 mL, 摇匀, 再加入浓硫酸5 mL, 摇匀, 放置10 min, 置40 ℃水浴中保温15 min, 取出, 迅速冷却至室温。取经苯酚-硫酸显色后的葡萄糖供试品溶液和经苯酚硫酸显色后无水葡萄糖对照品液[31], 用紫外分光光度计在400~700 nm的区间进行扫描, 选择最佳吸收波长。结果显示在492 nm处呈最大吸收值, 最终选取490 nm最为合适。
精密吸取葡萄糖对照品溶液0、0.2、0.4、0.6、0.8、1.0 mL, 分别置于25 mL具塞试管中, 加入纯化水至2.0 mL, 摇匀。加入5%苯酚试剂(现用现配)1 mL, 摇匀, 再加入浓硫酸5 mL, 摇匀, 放置10 min, 置40 ℃水浴中保温15 min, 取出, 迅速冷却至室温[32]。以相应的空白溶液为参比, 在1.3.3项中测得的最佳波长490 nm处测定吸光度值。以吸光度值(Y)为纵坐标, 质量浓度(X, mg/mL)为横坐标, 借助Excel 2010软件绘制标准曲线, 得到标准曲线回归方程为: Y=39.76X-0.009, 相关系数r2=0.9943。
将各组提取液稀释250倍后测定吸光度, 依据回归方程计算出各组供试品溶液中多糖的质量浓度C (mg/mL), 根据公式(1)计算多糖的提取率。

式中: C为供试品溶液中多糖的质量浓度, mg/mL; V为滤液体积, mL; D为稀释倍数; m为称取混合药材碎块的质量, g。
每组实验取药材碎块共10 g, 利用热水超声提取, 以黄精和枸杞子的比例、提取时间、液料比及超声功率为考察因素, 以多糖含量为指标, 采用单因素实验法结合L9(34)正交实验进行工艺优化。实验因素水平见表1
按药材比例4:1 (m:m), 取黄精、枸杞子适量混合, 加入30倍水, 250 W超声提取30 min, 将提取液浓缩至每1 mL相当于原药材1 g的稠膏, 备用。
因多糖具有一定黏性, 且实验量较小, 采用喷雾干燥, 多糖容易糊化且挂壁, 损耗较大。因此, 本研究采用湿法制粒制备固体颗粒。以95%乙醇添加量、麦芽糊精添加量、提取液稠膏添加量为因素, 设计单因素实验, 每个因素选取3个水平, 按照L9(33)进行正交实验, 因素水平表见表2。综合考虑颗粒性状以及溶解时间, 利用评分表进行判定, 确定最佳制粒条件。评分标准见表3
随机挑选10名专业人员组成评定小组, 对黄精枸杞固体饮料进行感官评价[33], 如表4。以提取物颗粒、DL-苹果酸、蔗糖粉和β-环状糊精等辅料的添加量作为因素, 设计单因素实验, 以感官评分为指标, 对比不同添加量对产品口感的影响, 并在单因素实验结果的基础上进行L9(34)正交实验, 确定黄精枸杞固体饮料的最佳调配工艺。因素水平表见表5
称取25 g制作的黄精枸杞固体饮料样品, 倒入盛有500 mL纯净水的烧杯中, 搅拌均匀后, 倒入100 mL量筒中, 加水至刻度线, 静置沉降20 min, 读取量筒上清液的下刻度, 记为A, 定义稳定系数F, F值越大则表明黄精枸杞固体饮料的冲调稳定性越高, 即黄精枸杞固体饮料的冲调稳定性与其沉淀速度呈反比, F按公式(2)计算。

全部数据分析利用Microsoft Office Excel 2010进行, 包括数据的录入、整理、统计学分析和图表的绘制。
结合不同文献[34-35]对于提取率和提取温度的选择, 可以看出提取温度大约在70~80 ℃可有效提取多糖, 当提取温度超过80 ℃, 多糖发生损失, 提取率反而下降, 因此本研究的温度固定为80 ℃。本研究对药材比例(黄精:枸杞子)、提取时间、液料比及超声功率4个因素进行单因素实验考察, 结果见图1
图1可知最佳提取条件为: 药材比为4:1 (m:m)、提取时间为30 min、液料比为25:1 (V:m)、超声功率为350 W。
根据表1中各因素水平数据, 结合L9(34)的正交实验设计表进行实验, 正交结果见表6, 方差分析见表7
表6分析可知, 以上4个因素对多糖提取率影响的主次顺序为: 超声功率(D)>液料比(C)>提取时间(B)>药材比例(A), 其最佳提取方案为A2B2C3D1, 即黄精和枸杞子的药材比例为4:1 (m:m), 提取时间为30 min, 液料比为30:1 (V:m), 超声功率为250 W。对表6中的正交实验结果进行方差分析(表7), 利用F值检验各因素对黄精枸杞多糖提取率的影响程度, 可得出在黄精枸杞多糖提取率方面, 4个因素均无显著影响。
本研究对95%乙醇添加量、麦芽糊精添加量及提取液稠膏添加量3个因素进行了单因素实验考察, 结果见图2
图2可知, 最佳制粒条件为: 95%乙醇15 mL、麦芽糊精添加量250 g、提取液稠膏添加量70 mL。
根据表2中各因素水平数据, 结合L9(33)的正交实验设计表进行实验, 结果见表8, 方差分析见表9
表6分析可知, 对于黄精枸杞多糖颗粒制备条件的影响主次顺序为: 提取液稠膏添加量(G)>95%乙醇添加量(E)>麦芽糊精添加量(F), 其颗粒的最佳制备条件为E2F2G2, 即加入95%乙醇15 mL, 麦芽糊精250 g, 提取液稠膏70 mL的条件下进行制粒。对表8中的正交实验结果进行方差分析(表9), 利用F值检验各因素对颗粒制备的影响程度, 可知对于黄精枸杞多糖颗粒的制备各因素均无显著影响。
本研究对提取物颗粒添加量、DL-苹果酸添加量、蔗糖粉添加量及β-环状糊精添加量4个因素进行了单因素实验考察, 结果见图3
图3可以看出, 最佳调配工艺为: 提取物颗粒添加量20 g、DL-苹果酸添加量0.20%、蔗糖粉添加量5 g (25%)、β-环状糊精添加量0.06%。
根据表5中各因素水平数据, 结合L9(34)的正交实验设计表进行实验, 正交结果见表10, 方差分析见表11
表10中的极差进行分析可知, 提取物颗粒、DL-苹果酸、蔗糖粉和β-环状糊精的添加量4个因素对感官评分的影响大小为: 提取物颗粒添加量(H)>DL-苹果酸添加量(I)>蔗糖粉添加量(J)>β-环状糊精添加量(K), 最佳调配工艺为H2I2J2K2, 即提取物颗粒添加量20 g、DL-苹果酸添加量0.20%、蔗糖粉添加量5 g (25%)、β-环状糊精添加量0.06%, 并结合正交结果的方差分析表(表11)得出, 在感官评分方面4个因素之间没有显著性影响。
综合上述提取工艺、制粒工艺、调配工艺的最佳工艺参数, 得出黄精枸杞固体饮料的最佳制备工艺如图4
按照优化后的制备工艺(2.4项下方法), 进行3次平行实验, 结果见表12
RSD是一种衡量实验结果稳定性和可靠性的指标, 其值越小, 说明实验结果的重复性和准确性越高。表12结果表明, 多糖提取率的RSD为0.235%, 颗粒评分的RSD为0.344%, 感官评分的RSD为0.408%, 说明此制备方法重复性高、稳定性好。
黄精与枸杞子作为兼具药用与食用价值的天然药材, 在功能性食品领域具有广阔的开发前景。本研究采用两者为原料, 通过系统的提取、制粒与调配工艺研究, 结合单因素与正交实验优化, 成功开发出一款新型固体饮料。该产品不仅色泽亮丽、酸甜适口, 更兼具黄精与枸杞的独特香气, 具有较高的营养价值和市场潜力。基于中医推荐的黄精(9~15 g/d)和枸杞子(3~5 g/d)摄入量标准, 结合产品规格(25 g/袋), 建议成人每日食用量为2袋, 每袋以500 mL水冲泡为佳。
固体饮料作为现代食品工业的重要品类, 其易冲泡性、独特风味、便捷携带和长保质期等特点, 在国际市场上已获得广泛认可。特别是美国、欧洲等发达经济体, 固体饮料已成为大众消费的常规品类。相较而言, 我国固体饮料产业起步较晚, 但发展势头强劲。据《中国固体饮料行业产销需求与投资预测分析报告前瞻》统计显示, 近年来国内固体饮料行业规模快速扩张, 市场接受度持续攀升。在国家政策层面, 2023年国民经济和社会发展执行计划明确提出将实施中医药振兴发展重大工程, 这为药食同源产品的研发提供了有力支持。基于中医药“药食同源”的核心理念, 将传统中药材如黄精、枸杞子与现代化固体饮料生产技术有机结合, 开发具有保健功能的创新型食品, 已成为我国食品行业转型升级的重要战略方向。本研究正是这一思路的积极探索与有益实践, 为传统中药材的现代化应用提供了新的技术路径和市场可能。
