Objective To investigate the vitamin content in common 48 kinds of desserts in Gansu Province based on the requirements of national nutritional monitoring. Methods A total of 48 kinds of common desserts from Gansu Province were analyzed. The content of vitamin A, carotene, vitamin B1, vitamin B2, vitamin C, vitamin E and vitamin E fractions, i.e. (α-tocopherol, β-tocopherol, γ-tocopherol and δ-tocopherol) were determined by high performance liquid chromatography method as specifiedin the national standard for food safety. Results The results showed that among the 48 kinds of desserts, except for vitamin B1, the detection rates of other vitamins ranged from high to low: Vitamin E>vitamin B2>vitamin A>carotene>vitamin C; the content of vitamin A in the sample is ND-549.000 μg/100 g, median of 20.000 μg/100 g; the content of carotene was ND-142.650 μg/100 g, median of 0.250 μg/100 g. The content of vitamin B2 was ND-0.226 mg/100 g, median of 0.090 mg/100 g; The content of vitamin C was ND-4.740 mg/100 g, median of 0.250 mg/100 g. The detected content of vitamin E (sum of various subtypes) ranges was ND-549.000 mg/100 g, median of 21.650 mg/100 g; the content of α-tocopherol was ND-2.690 mg/100 g, median of 1.015 mg/100 g; the content of β-tocopherol was ND-1.240 mg/100 g, median of 0.020 mg/100 g; the content of γ-tocopherol was ND-11.810 mg/100 g, median of 1.135 mg/100 g; the content of δ-tocopherol was ND-2.040 mg/100 g, median of 0.020 mg/100 g. Conclusion The fat-soluble vitamins are generally detected at higher levels in the samples from Gansu Province, while the water-soluble vitamins are detected at lower levels.

目的 基于国家营养监测要求对甘肃省内常见48种甜点中维生素含量情况进行检测。方法 以甘肃省内常见的48种甜品为研究对象, 采用食品安全国家标准中规定的高效液相色谱法对样品中维生素A、胡萝卜素、维生素B1、维生素B2、维生素C、维生素E以及维生素E(包括α-生育酚、β-生育酚、γ-生育酚和δ-生育酚)的含量进行检测。结果 48种甜品中除去维生素B1均未检出外, 其余维生素检出率从高到低为维生素E>维生素B2>维生素A>胡萝卜素>维生素C; 样品中维生素A含量为未检出~549.000 μg/100 g,中位数为20.000 μg/100 g; 胡萝卜素检出含量为未检出~142.650 μg/100 g, 中位数为0.250 μg/100 g; 维生素B2检出含量为未检出~0.226 mg/100 g,中位数为0.090 mg/100 g; 维生素C检出为未检出~4.740 mg/100 g, 中位数为0.250 mg/100 g; 维生素E(各分型之和)检出含量为未检出~549.000 mg/100 g,中位数为21.650 mg/100 g; α-生育酚检出含量为未检出~2.690 mg/100 g, 中位数为1.015 mg/100 g; β-生育酚检出含量为未检出~1.240 mg/100 g, 中位数为0.02 mg/100 g; γ-生育酚检出含量为未检出~11.810 mg/100 g, 中位数为1.135 mg/100 g; δ-生育酚检出含量为未检出~2.040 mg/100 g, 中位数为0.020 mg/100 g。结论 脂溶性维生素在甘肃省内甜点样品中检出含量普遍较高, 水溶性维生素检出均较低。

目的 基于国家营养监测要求对甘肃省内常见48种甜点中维生素含量情况进行检测。方法 以甘肃省内常见的48种甜品为研究对象, 采用食品安全国家标准中规定的高效液相色谱法对样品中维生素A、胡萝卜素、维生素B1、维生素B2、维生素C、维生素E以及维生素E(包括α-生育酚、β-生育酚、γ-生育酚和δ-生育酚)的含量进行检测。结果 48种甜品中除去维生素B1均未检出外, 其余维生素检出率从高到低为维生素E>维生素B2>维生素A>胡萝卜素>维生素C; 样品中维生素A含量为未检出~549.000 μg/100 g,中位数为20.000 μg/100 g; 胡萝卜素检出含量为未检出~142.650 μg/100 g, 中位数为0.250 μg/100 g; 维生素B2检出含量为未检出~0.226 mg/100 g,中位数为0.090 mg/100 g; 维生素C检出为未检出~4.740 mg/100 g, 中位数为0.250 mg/100 g; 维生素E(各分型之和)检出含量为未检出~549.000 mg/100 g,中位数为21.650 mg/100 g; α-生育酚检出含量为未检出~2.690 mg/100 g, 中位数为1.015 mg/100 g; β-生育酚检出含量为未检出~1.240 mg/100 g, 中位数为0.02 mg/100 g; γ-生育酚检出含量为未检出~11.810 mg/100 g, 中位数为1.135 mg/100 g; δ-生育酚检出含量为未检出~2.040 mg/100 g, 中位数为0.020 mg/100 g。结论 脂溶性维生素在甘肃省内甜点样品中检出含量普遍较高, 水溶性维生素检出均较低。

Objective To investigate the vitamin content in common 48 kinds of desserts in Gansu Province based on the requirements of national nutritional monitoring. Methods A total of 48 kinds of common desserts from Gansu Province were analyzed. The content of vitamin A, carotene, vitamin B1, vitamin B2, vitamin C, vitamin E and vitamin E fractions, i.e. (α-tocopherol, β-tocopherol, γ-tocopherol and δ-tocopherol) were determined by high performance liquid chromatography method as specifiedin the national standard for food safety. Results The results showed that among the 48 kinds of desserts, except for vitamin B1, the detection rates of other vitamins ranged from high to low: Vitamin E>vitamin B2>vitamin A>carotene>vitamin C; the content of vitamin A in the sample is ND-549.000 μg/100 g, median of 20.000 μg/100 g; the content of carotene was ND-142.650 μg/100 g, median of 0.250 μg/100 g. The content of vitamin B2 was ND-0.226 mg/100 g, median of 0.090 mg/100 g; The content of vitamin C was ND-4.740 mg/100 g, median of 0.250 mg/100 g. The detected content of vitamin E (sum of various subtypes) ranges was ND-549.000 mg/100 g, median of 21.650 mg/100 g; the content of α-tocopherol was ND-2.690 mg/100 g, median of 1.015 mg/100 g; the content of β-tocopherol was ND-1.240 mg/100 g, median of 0.020 mg/100 g; the content of γ-tocopherol was ND-11.810 mg/100 g, median of 1.135 mg/100 g; the content of δ-tocopherol was ND-2.040 mg/100 g, median of 0.020 mg/100 g. Conclusion The fat-soluble vitamins are generally detected at higher levels in the samples from Gansu Province, while the water-soluble vitamins are detected at lower levels.

随着社会经济的发展和居民生活水平的提高, 甜品消费逐渐成为日常饮食的重要组成部分。甜品因其独特的风味和多样化的形式深受消费者青睐, 尤其在年轻群体及城市居民中消费需求持续增长。然而, 传统甜品多以高糖、高脂、高热量的原料为主, 其营养价值常因维生素等微量营养素含量不足而受到质疑。维生素作为人体必需的微量营养素, 在维持代谢功能、增强免疫力及预防慢性疾病中具有重要作用。因此, 系统评估甜品中维生素含量, 对引导健康饮食具有重要意义。微量营养素缺乏对国民营养健康也造成了严重的负面影响[1]。为提高人们健康水平, 中共中央提出了《“健康中国2030”规划纲要》, 根据2015—2017年中国居民营养与健康状况监测数据, 我国80%以上居民存在维生素B1和维生素B2摄入不足风险, 近70%居民存在维生素C摄入不足风险[2]。如能促进甜品从高糖、高脂、高热量的“愉悦消费”转向强化营养素的“健康消费”, 如日本功能标示甜品添加维生素B1和维生素B2, 成为便利店热销商品, 既顺应现代人对营养的关注, 也为行业创新提供方向。其核心目的是在满足味蕾的同时, 提升甜品的功能性价值, 推动健康饮食文化与城市生活方式的融合。
维生素是参与维持人体正常生理活动必需的一类有机化合物, 其主要功能是作为辅酶的成分调节代谢[3]。大部分维生素在体内不能合成或合成量很少, 不能满足机体需要, 必须由食物来供给[4]。随着经济的不断发展, 人民生活水平不断提高, 甜品己成为社会大众休闲、娱乐的首选消费食品, 占人们消费支出的比重也越来越高[5]。有数据显示, 中国甜品店铺数量已经位居餐饮市场第3[6]。鉴于甜品现如今被大众喜爱的程度以及我国居民营养缺乏的现状, 对甜品中各种营养素的检测是至关重要的。
目前国外对维生素的研究主要集中于各类维生素单一或者联合后在疾病中的作用[7-9], 对甜品方面的研究通过检索近十年文献发现大多集中于摄入过量甜品对人体健康的影响[10-11]。国内对维生素研究一方面主要集中在对检测维生素方法上的改良[12], 另一方面也有部分研究是对于维生素在疾病治疗中的作用[13]。国内对甜品方面的研究主要在市场发展与营销策略上[14], 而对于甜品中营养元素含量的基础性研究较少。本研究通过以甘肃省内的48种常见甜品为研究对象, 采用食品安全国家标准中规定的高效液相色谱法对样品中维生素A、胡萝卜素、维生素B1、维生素B2、维生素C、维生素E以及维生素E(包括α-生育酚、β-生育酚、γ-生育酚和δ-生育酚)的含量进行检测, 对省内食品营养结构进行了解, 以便为食品营养评价和人们健康膳食提供合理建议。
为确保所采集的样品具有代表性, 在甘肃省内连锁店(连锁店选择标准: 数量大于3家)采取随机抽样的方法对常见甜点进行采样, 共采集到48份样品。48份样品可分为6大类, 分别为特色类(n=2)、糕点类(n=7)、面点类(n=8)、蛋糕类(n=15)、面包类(n=11)和饼干类(n=5)。
视黄醇、α-胡萝卜素、β-胡萝卜素(纯度≥95%, 坛墨质检标准物质中心); α-生育酚、β-生育酚、γ-生育酚、δ-生育酚(纯度≥95%, 天津阿尔塔科技有限公司); 维生素B1(纯度≥99%)、维生素B2(纯度≥98%, 北京曼哈格生物科技有限公司); L(+)-抗坏血酸标准品、D(-)-抗坏血酸(异抗坏血酸)(纯度≥99%, 北京振翔科技有限公司); 木瓜蛋白酶(酶活力≥800 U/mg)、淀粉酶(酶活力≥3700 U/g)、木瓜蛋白酶(酶活力≥10 U/mg)、高峰淀粉酶(酶活力≥100 U/mg)、淀粉酶(酶活力≥100 U/mg)(上海安谱实验科技股份有限公司); 2,6-二叔丁基对甲酚(纯度≥99.99%)、乙酸钠(三水)(纯度≥99.99%)(上海阿拉丁生化科技股份有限公司); 乙腈、甲醇、正己烷(色谱纯, 德国默克公司); 十六烷基三甲基溴化铵(分析纯, 沪试国药西陇科学股份有限公司); 三氯甲烷、甲基叔丁基醚、正丁醇(色谱纯, 上海麦克林生化科技股份有限公司); 二氯甲烷(色谱纯, 诺尔施成都市科隆化学品有限公司); 盐酸、冰乙酸、乙酸钠、氢氧化钠、氢氧化钾、抗坏血酸、铁氰化钾、L-半胱氨酸、无水乙醇、无水乙醚、无水硫酸钠、磷酸、磷酸二氢钾、偏磷酸、磷酸三钠(优级纯, 天津市科密欧化学试剂有限公司); 五氧化二磷(分析纯, 安徽金粤冠新材料科技有限公司); 石油醚(分析纯, 天津市富宁精细化工有限公司)。
Agilent 1200高效液相色谱仪(带荧光、紫外检测器,美国安捷伦公司); PL303电子天平(精度0.01 mg, 瑞士梅特勒托利多仪器有限公司); SS-1022高速多功能粉碎机(武义海纳电器有限公司); Milli-Q IQ 7000纯水机[默克化工技术(上海)有限公司]; R-215旋转蒸发仪(香港步琪有限公司); DHG-9240A电热恒温鼓风干燥箱(江苏正基仪器有限公司); TARGINVX-Ⅲ涡旋振荡器(北京踏棉科技有限公司); N-EVAP112氮吹仪(美国Orangaomation公司); 3k30高速离心机(德国Sigma公司); PHSJ-5酸度计(深圳市卡迪亚科技有限公司); SHA-B水浴恒温振荡器(江苏正基仪器有限公司); C18反相色谱柱(250 mm× 4.6 mm, 5 μm)、C30柱(250 mm×4.6 mm, 3 μm)(北京迪马科技公司)。
在本研究中均采用GB 5009系列中不同维生素相应的检测进行测定, 样品处理均严格按照食品国家安全标准中规定的步骤进行相应的操作。为了保持样品的稳定性和防止可能的质量变化, 其中在测定维生素A、E与维生素B2中样品需避光检测; 维生素C要尽快测定。样品中维生素A、E测定用GB 5009.82—2016《食品安全国家标准 食品中维生素A、D、E的测定》; 胡萝卜素测定用GB 5009.83—2016《食品安全国家标准 食品中胡萝卜素的测定》; 维生素B1测定用GB 5009.84—2016《食品安全国家标准 食品中维生素B1的测定》; 维生素B2测定用GB 5009.85—2016《食品安全国家标准 食品中维生素B2的测定》; 维生素C测定用GB 5009.86—2016《食品安全国家标准 食品中抗坏血酸的测定》。
采用Excel 2010软件进行原始数据记录及分类整理, 使用SPSS 25.0软件对样品中维生素含量进行多个独立样本非参数检验分析, 检验水准α=0.05, P<0.05为差异有统计学意义。
维生素可分为脂溶性与水溶性两大类, 其中脂溶性维生素包括维生素A、胡萝卜素和维生素E等, 水溶性维生素包括维生素B族、维生素C等[15]。通过使用GB 5009中各维生素检测第一方法对甘肃省48份样品中维生素含量进行检测分析, 整体检出率见表1。结果显示, 维生素E在48份样品中检出率最高, 为72.9%。维生素B1在48份样品中均未检出。各类维生素检出率从高到低依次为: 维生素E>维生素B2>维生素A>胡萝卜素>维生素C>维生素B1。根据表3中各类维生素检出情况得到图1, 发现48份样品中脂溶性维生素检出率较高, 占全部检出维生素的67%, 水溶性维生素检出率相对较少, 占33%。
对原始数据进行整理, 发现同一种样品中不同维生素含量检出差异较大。表2是对样品中各类维生素含量检测得到的原始数据通过中位数与四分位间距进行的统计描述, 结果发现, 不同样品中均未检出维生素B1(检出含量低于检测方法的最低检出限), 而同一种维生素在不同样品中检出的差异较大。将48份甜点样品按照传统分类进行归类, 可分为特色类、糕点类、面点类、蛋糕类、面包类和饼干类6大类。表3是样品经分类后, 各类别样品不同维生素具体含量。
48种样品中维生素A检出含量为未检出(低于检测方法最低检出限)~549.000 μg/100 g, 中位数为20.000 μg/100 g, 其中样品名为蔓越莓曲奇饼干的检出含量最高, 为549.000 μg/100 g。对各类别甜点的维生素A含量进行非参数检验发现差异无统计学意义(P>0.05)。
在48份样品中检出胡萝卜素的有11份, 检出含量为未检出(低于检测方法最低检出限)~142.650 μg/100 g, 中位数为0.250 μg/100 g, 极差为142.650 μg/100 g, 表明得到的样品中胡萝卜素含量数据离散程度较大, 其中含量最高的甜点是名为黄金唱片的面包。对各类别甜点的胡萝卜素含量进行非参数检验发现差异无统计学意义(P>0.05)。
48份样品中维生素B2检出含量未检出(低于检测方法最低检出限)~0.226 mg/100 g, 占NRV百分比范围在Tr~18.8%, 中位数为0.090 mg/100 g。各样品中检出含量均低于《中国居民膳食营养素参考摄入量》中参考含量。对各类别甜点的维生素B2含量进行非参数检验发现差异有统计学意义(P<0.001), 不同类别两两比较, 发现就维生素B2而言, 饮品类与蛋糕类两者之间差异有统计学意义(P=0.036); 面包类与面点类两者之间差异有统计学意义(P=0.035); 面点类与蛋糕类两者之间有统计学意义(P<0.001)。
48份样品中维生素C检出为未检出(低于检测方法最低检出限)~4.740 mg/100 g, 占NRV百分比范围在Tr~5.6%, 中位数为0.250 mg/100 g, 其中茶心芝士检出含量最多, 占NRV百分比为5.6%。样品中检测出有维生素C的仅有6种, 且检出含量均远低于《中国居民膳食营养素参考摄入量》中参考含量。对各类别甜点的维生素C含量进行非参数检验发现差异无统计学意义(P>0.05)。
48份甜品中维生素E(各分型之和)检出含量为未检出(低于检测方法最低检出限)~549.000 mg/100 g, 中位数为21.650 mg/100 g, 蔓越莓曲奇饼干和杏愿蛋糕检出含量分别为549.000 mg/100 g与488.000 mg/100 g, 对各类别甜点的维生素E(各分型之和)含量进行非参数检验发现差异无统计学意义(P>0.05)。
根据刘文芳[16]在研究中提到的维生素E中较为重要的分型有αβγδ 4类, 对6大类48份甜品分别进行检测, α-生育酚检出含量为未检出~2.690 mg/100 g, 中位数为1.015 mg/100 g; β-生育酚检出含量为未检出~1.240 mg/100 g, 中位数为0.020 mg/100 g; γ-生育酚检出含量为未检出~11.810 mg/100 g, 中位数为1.135 mg/100 g; δ-生育酚检出含量为未检出~2.040 mg/100 g, 中位数为0.020 mg/100 g。得到检测含量见表2。对各类别甜点的α-生育酚、β-生育酚、γ-生育酚、δ-生育酚含量进行非参数检验发现其中α-生育酚与β-生育酚均无统计学意义(P>0.05); γ-生育酚有统计学意义(P=0.004); δ-生育酚有统计学意义(P=0.006)。
表4是对不同维生素含量进行Spearman相关分析。由表4可知, 在P=0.01情况下, 维生素E(各分型之和)与维生素A相关系数为0.983, 呈显著正相关; 维生素C与胡萝卜素相关系数为0.464, 呈显著正相关; α-生育酚与γ-生育酚相关系数为0.619, 呈显著正相关; γ-生育酚与δ-生育酚相关系数为0.720, 呈显著相关。表明不同种维生素间有相互促进作用。
维生素A的生物活性分子形式主要为视黄醇、维甲酸、视黄素等[17], 以视黄醇为核心, 所以本研究以视黄醇含量指证维生素A, 本研究中维生素A检出率与检出含量都较高。维生素A主要存在于动物性食物中, 以动物肝脏、牛奶、鸡蛋、鱼肝油和奶油等含量较高[17]。维生素A较稳定, 耐酸、碱与热。维生素A经一般的烹调加工不会被破坏, 但易被氧化, 食物中的维生素C、磷脂等抗氧化剂有助于维生素A的稳定[18]。由此对研究结果做出推测为: 一方面是由于甜品原料中本身就含有较高的维生素A, 因为甜品制作都会加入较高含量的牛奶、鸡蛋及奶油, 尤其是蛋糕类甜点中会用到大量奶油, 另一方面是由于维生素E、胡萝卜素等抗氧化维生素的存在使维生素A在制作过程中免受氧化。
在本次研究中胡萝卜素整体检出率相对其余脂溶性维生素较低, 推测是由于胡萝卜素本身化学稳定性差[19], 而且类胡萝卜素最主要的来源是蔬菜和水果[20], 甜点制作中所用的含胡萝卜素原料较少且制作过程中由于稳定性差被分解故而检出较低。值得一提的是在名为黄金唱片与乳香包的甜点中胡萝卜素检出含量较高, 根据张同等[20]提到在食品工业中类胡萝卜素被大量用作着色剂, 推测是在制作过程中加入了人工合成和提取的胡萝卜素作为着色剂故而检出含量较高。在相关性分析中胡萝卜素与维生素C在P=0.01情况下呈显著相关(r=0.464), 经过对相关文献的查阅发现在胃肠道中维生素C可以增加类胡萝卜素的稳定性[21]。本研究维生素C与胡萝卜素呈显著相关, 推测是由于维生素C增加了胡萝卜素的化学稳定性。
在本次研究中维生素B1均未检出; 维生素B2检出率相对较高。B族维生素是维持机体正常功能与代谢活动必须的水溶性维生素, 人体无法自行制造合成[22]。维生素B1在大多数食物中含量较少, 主要富含于谷类中谷粒的外皮[23], 另一方面维生素B1在加工过程中极易降解[24]。在本研究中检测的样品使用原料均为已经过加工的面粉, 不含谷粒外皮, 且甜品制作有多道工序, 维生素B1在制作过程中被降解, 故维生素B1未检出。在人体内维生素B1是糖代谢过程中重要的辅酶[25], 当人体摄入较多甜品时便需要一定量的维生素B1的去参与糖代谢, 而甜品中又未检测到维生素B1, 故而推荐居民尤其喜爱甜品的居民在日常生活中注意摄入适当含维生素B1较丰富的食物。奶类制品中含有较多的维生素B2, 有国外研究表明西方国家膳食维生素B2的主要来源就是奶和奶制品[26]。甜品制作中一般会用到牛奶、炼乳和奶粉等一系列奶类制品, 故而样品中维生素B2整体检出率较高。但是对数据进行进一步分析观察到能检出维生素B2的样品中检出含量均远低于推荐摄入量, 根据2015—2017年中国居民营养与健康状况监测数据显示我国有近80%以上居民存在维生素B2摄入低于推荐摄入量[2], 这与本研究结果相符。
在本研究中维生素C检出率仅为12.5%, 且检出的各样品种含量均低于中国居民膳食营养素参考摄入量中的参考摄入量。维生素C, 又称为抗坏血酸, 属于水溶性维生素, 有较强的抗氧化能力。维生素C主要存在于新鲜蔬菜和水果中[4]。由此对本研究结果做出推测: 一方面是因为甜品中较少的使用富含维生素C的原材料, 另一方面是维生素C在加工烹调过程中易损失, 且烹调时间越长损失越多[23], 甜点在制作过程中往往较为复杂, 故而检出率与检出含量都较低。根据2015—2017年中国居民营养与健康状况监测数据显示我国有近70%的居民存在维生素C摄入低于推荐摄入量[2], 这与本研究结果相符。鉴于研究结果与维生素C在烹饪过程中易损失的特点, 推荐居民在日常生活中注意摄入新鲜水果与蔬菜, 以确保摄入足够机体所需的维生素C。
在本研究中维生素E检出率与检出含量都居高。维生素E是一种脂溶性维生素化合物, 具有抗氧化和抗癌等多种活性[27]。维生素E多存在于麦胚油、豆油和玉米油等植物油及人造奶油、坚果和各种谷类中[16]。对维生素E检出率与检出含量均高这一研究结果做出推测是由于在甜点制作过程中会运用植物油及奶油等, 且维生素E本身稳定性就较强, 也有可能是甜点制作过程中为了防氧化而加入了维生素E。本研究结果2.3中提到维生素E与维生素A在P=0.01情况下呈现高度相关(r=0.983), 推测是由于维生素E有较强的抗氧化能力[28], 在制作过程中维生素E保护维生素A免受氧化, 故而两者含量呈高度相关。
48份常见甜点中脂溶性维生素检出率与检出含量都较高, 水溶性维生素检出均偏低。其中脂溶性维生素A、E检出率分别高达64.6%与72.9%, 水溶性维生素C检出率仅为12.5%。对于同一种甜品检出的不同种类维生素含量差异较大。对不同种类维生素通过Spearman相关分析, 发现维生素A与维生素E, 胡萝卜素与维生素C都存在显著性相关, 其中维生素A与E在P=0.01的情况下, 相关性高达0.983。《中国食品网》统计数据显示, 烘焙食品的销售额每年以超出10%的速度持续增长[5]。近年来越来越多的国人对甜品喜爱程度增加, 更有研究提出中国消费者更有可能将甜品当成主餐一部分食用[29], 因此建议甘肃省内甜品店在甜品制作过程中在兼顾美味的情况下, 注意原材料的合理搭配, 尝试添加天然果蔬成分或强化营养, 使甜品中可以富含更多人体所需的维生素, 消费者健康意识加强, 知晓营养强化食品而获取的健康益处, 而选择营养强化食品[30] 。与此同时, 相关部门应该加强居民营养健康教育宣传, 让大众自身意识到健康饮食的重要性, 从而改变不合理的饮食结构, 使我国的国民健康水平不断提高[31]
