Fungal toxins are a class of toxic secondary metabolites produced by filamentous fungi under specific environmental conditions, with nephrotoxicity, hepatotoxicity, neurotoxicity, teratogenicity, carcinogenesis, and mutagenicity. These toxins can enter the food chain by contaminating agricultural products such as grains, fruits, vegetables, nuts, and their processed products, posing a serious threat to human health. Therefore, developing efficient and sensitive detection techniques for fungal toxins in food is particularly important. This article provided a systematic review of common fungal toxin detection technologies in food, focusing on the technical characteristics of different detection technologies in sensitivity, specificity, multi-component synchronous detection, and on-site rapid screening, analyzed the key technical bottlenecks such as complex food matrix interference elimination, precise quantification of trace toxins, and development of detection standard substances, providing reference for in-depth research in this field and the updating and optimization of national and other levels of detection standards, in order to further improve the accuracy and efficiency of fungal toxin detection in food and provide strong support for food safety assurance.

真菌毒素是一类由丝状真菌在特定环境条件下产生的有毒次生代谢产物, 具有肾毒性、肝毒性、神经毒性和致畸、致癌以及致突变性, 这类毒素可通过污染谷物、果蔬、坚果等农产品及其加工制品进入食物链, 对人体健康构成严重威胁。因此, 开发高效、灵敏的食品中真菌毒素检测技术显得尤为重要。本文针对目前食品中常见的真菌毒素检测技术进行了系统综述, 重点探讨了不同检测技术在灵敏度、特异性、多组分同步检测以及现场快速筛查方面的技术特征, 剖析了复杂食品基质干扰消除、痕量毒素精准定量、检测标准物质研制等关键技术瓶颈, 为该领域内的深入研究以及国家等各级检测标准的更新优化提供参考, 以期进一步提升食品中真菌毒素检测的准确性和效率, 为食品安全保障提供有力支撑。

Fungal toxins are a class of toxic secondary metabolites produced by filamentous fungi under specific environmental conditions, with nephrotoxicity, hepatotoxicity, neurotoxicity, teratogenicity, carcinogenesis, and mutagenicity. These toxins can enter the food chain by contaminating agricultural products such as grains, fruits, vegetables, nuts, and their processed products, posing a serious threat to human health. Therefore, developing efficient and sensitive detection techniques for fungal toxins in food is particularly important. This article provided a systematic review of common fungal toxin detection technologies in food, focusing on the technical characteristics of different detection technologies in sensitivity, specificity, multi-component synchronous detection, and on-site rapid screening, analyzed the key technical bottlenecks such as complex food matrix interference elimination, precise quantification of trace toxins, and development of detection standard substances, providing reference for in-depth research in this field and the updating and optimization of national and other levels of detection standards, in order to further improve the accuracy and efficiency of fungal toxin detection in food and provide strong support for food safety assurance.

真菌毒素是一类由产毒丝状真菌在食品中生长繁殖过程中产生的次生有毒代谢产物, 主要是由一些子囊菌属的真菌产生, 微量的真菌毒素即可对人体或动物的肾脏、肝脏等器官构成严重的威胁[1]。目前真菌毒素污染仍然是造成食品污染的主要原因之一, 其化学性质稳定, 加工、储存均难以去除[2], 这些毒素具有不同程度的致癌、致畸、致突变、抑制免疫系统效应, 一旦经食物链进入人体会干扰正常新陈代谢, 造成氧化应激诱导细胞癌变, 损害生命健康[3]
食品中含有真菌毒素, 不仅会影响食品的口感和营养价值, 还会降低食品的安全性, 影响消费者的健康。同时, 含有真菌毒素的食品在农产品贸易中也会受到限制, 影响农产品的出口[4]。因此多国对真菌毒素在食品中的含量制定了卫生标准并进行严格的监管[5], 我国GB 2761—2017《食品安全国家标准 食品中真菌毒素限量》基于风险评估原则, 筛选出可能对公众健康构成较大风险的真菌毒素, 包括黄曲霉毒素B1 (aflatoxin B1, AFB1)、黄曲霉毒素M1 (aflatoxin, AFM1)、脱氧雪腐镰刀菌烯醇(vomitoxin, DON)、展青霉素、赭曲霉毒素A (ochratoxin A, OTA)及玉米赤霉烯酮(zearalenone, ZEN)[6-12], 其中针对消费者膳食暴露量产生较大影响的食品设定了限量要求, 且国家市场监督管理总局在《全国食品安全监督抽检实施细则》中也根据食品分类体系, 对各品类真菌毒素作出具体要求(见表1), 全力保障食品安全。
为了预防和减少真菌毒素带来的危害, 保障人民食品安全, 在做好食品仓储、运输以防止霉菌滋生的同时, 进行真菌毒素的检测尤为重要, 将其对人类的潜在风险和资源的浪费降到最低。本文对近年来食品中常见真菌毒素检测过程中的前处理技术和检测技术进行总结和比对, 以期为真菌毒素的检测及研究提供理论参考。
在真菌毒素检测过程中, 由于实际样品中毒素含量较低且样品基质复杂, 易对检测过程产生干扰, 因此在仪器分析之前需对毒素进行高效的富集、分离、净化, 从而保障检测结果的准确性和可靠性。目前, 国内外针对真菌毒素最常见的样品前处理净化技术主要有免疫亲和柱(immunoaffinity column, IAC)净化技术、固相萃取(solid phase extraction, SPE)技术、液-液萃取(liquid-liquid extraction, LLE)技术、QuEChERS (quick, easy, cheap, effective, rugged, safe)等技术。
IAC是利用生物大分子具有对某一类生物大分子特异识别和可逆结合的特性而制成, 用以分析检测抗体或抗原的亲和层析柱。IAC净化过程包括上样、淋洗与洗脱3个步骤, 以实现抗体对被测物的特异性识别, 其具有高选择性、高亲和力及易于操作的优势, 因此广泛应用于食品安全、蛋白质组学及生物医药等领域中的分离和纯化[13-14]。在食品安全领域, IAC技术的创新应用, 推动了多毒素同步检测技术的发展。孟春杨等[15]针对火锅底料基质复杂、多毒素共存的检测难点, 开发了五联复合IAC用于提取液净化, 可同时定性及定量检测火锅底料中的AFB1、黄曲霉毒素B2 (aflatoxin B2, AFB2)、黄曲霉毒素G1 (aflatoxin G1, AFG1)、黄曲霉毒素G2 (aflatoxin G2, AFG2)和OTA, 为火锅底料中真菌毒素暴露的风险评估提供了技术支撑。邵亮亮等[16]通过建立DON、ZEN、AFB1、OTA的四合一复合IAC同步净化、柱后光化学衍生、高效液相色谱二极管阵列检测器和荧光检测器同时检测, 实现小麦中这4种真菌毒素的同时快速测定。目前IAC技术在食品安全检测中的发展趋势: (1)通过改造, 提升多目标物同步捕获能力; (2)与新型检测技术联用形成痕量分析体系。
SPE柱是从层析柱发展而来的, 基于液-固相色谱理论的样品前处理技术[17]。采用固体吸附剂吸附、溶剂洗脱的方式富集净化目标物, 具有富集倍数和回收率高、试剂用量少、操作简便等优点, 主要应用于食品、生物样品以及环境样品中目标化合物的样品前处理[18-19]。LIU等[20]开发了一种基于腐植酸的SPE净化方法, 结合液相色谱-串联质谱法对食用油中AFB1、AFB2、AFG1、AFG2进行了分析。腐植酸经简单处理后可直接用作SPE吸附剂处理稀释后的食用油, 该方法具有、适应性强、处理便捷及分析快速等优点。BAYRAM等[21]以AFB1、AFB2、AFG1、AFG2为模板, 采用本体聚合法制备了分子印迹聚合物, 并将其用作SPE柱填料, 实现了农产品中4种黄曲霉毒素的同步分离富集。然而, SPE技术也存在批次间的重复性差, 样品处理时间长, 成本较高等问题。
LLE又称溶剂萃取或抽提, 是利用有机物在两种互不相溶的溶剂中的溶解性不同, 将有机物从一种溶剂转移到另一种溶剂的过程。该技术依赖于目标分析物在两种不同极性溶剂之间的分布情况。目前有传统LLE[22]、均相LLE[23]、分散液液微萃取等种类[24-26]。POCHIVALOV等[27]使用共晶溶剂(deep eutectic solvent, DES)与玉米赤霉烯酮的酯酮和羟基形成氢键, 达到其与固体样品的高效分离, 加快了萃取过程并提高了提取效率样本中玉米赤霉烯酮的提取, 该方法回收率达到93%。CHEN等[28]采用基于天然低共熔溶剂的分散液液微萃取技术, 实现了对花生油和葵花籽油中AFB1、AFB2和AFG2的高效萃取, 该前处理技术, 相较于传统的LLE, 产生的废液量少、溶剂与样品消耗量低, 且对环境更为友好, 体现了绿色分析化学的理念与研究趋势。
QuEChERS是近年来国际上最新发展起来的一种用于农产品检测的快速样品前处理技术, 由美国农业部ANASTASSIADES教授等于2003年开发, 该方法集快速、简单、便宜、有效、可靠和安全于一身, 是利用吸附剂填料与基质中的杂质相互作用, 吸附杂质从而达到净化的目的, 实现多种目标物同时提取分析[29-30]
QuEChERS技术主要包括乙腈提取、盐析分层、吸附除杂3个过程。RAUSCH等[31]利用QuEChERS前处理提取方法, 富集谷物中38种真菌毒素用于后续的检测, 该方法回收率达到61%~120%, 实现了多毒素的同步检测。QuEChERS方法结合高效液相色谱-串联质谱法, 可富集牛奶基质中6种玉米赤霉烯酮类化合物, 使6种玉米赤霉烯酮类化合物的检出限和定量限范围分别达到0.05~0.10 μg/kg和0.25~0.50 μg/kg, 回收率达到91.8%~114.5%[32]。QuEChERS技术在不同基质中可针对多种目标物的检测分析, 具有良好的适用性和可靠性, 为相关物质的检测提供了有效的技术手段。
食品中真菌毒素样品前处理技术详见表2
色谱法是利用物质的溶解性、吸附性等特性的物理化学分离方法, 其分离原理是根据混合物的各组分在互不相溶的两相(称为固定相和流动相)作用的差异作为分离依据的。
高效液相色谱法(high performance liquid chromatography, HPLC)以液体为流动相, 采用高压输液系统, 将具有不同极性的单溶剂或不同比例的混合溶剂、缓冲液等流动相泵入装有固定相的色谱柱, 在柱内各成分被分离后, 进入检测器进行检测, 从而实现对试样的分析。通过优化前处理、色谱柱选择及检测条件, 可以显著提高检测的准确性和可靠性, 为食品安全提供科学依据[33-37]。韩祎陟等[38]采用基于免疫磁珠净化的HPLC技术测定植物油中AFB1、AFB2、AFG1和AFG2含量, 4种毒素在5种商品植物油基质中的平均回收率为74.0%~96.0%, 该方法准确、灵敏且成本低, 适用于植物油中4种黄曲霉毒素的同时定性、定量分析。
高效液相色谱-质谱法(high performance liquid chromatography-mass spectrometry, HPLC-MS)是将HPLC的分离能力和质谱(mass spectrum, MS)的鉴定能力相结合, 利用液相色谱对复杂样品的高效分离能力, 并发挥质谱对化合物的高灵敏度检测和准确结构鉴定的特点, 其具有高准确性、高灵敏度和高通量等优势[39-41]。严晓贤等[42]选取AFM1等17种乳及乳制品中常检真菌毒素作为研究对象, 通过QuEChERS净化结合超高效液相色谱-串联三重四极杆质谱法建立了酥油中同时检测17种真菌毒素的分析方法, 检出限为0.15~3.00 μg/kg, 定量限为0.50~10.00 μg/kg。该方法灵敏度好, 准确度高, 可作为日常监测酥油中真菌毒素的的有效检测方法。类似的, 黄忠亮等[43]建立自动SPE前处理技术结合HPLC-MS法检测药食同源杂粮中14种真菌毒素, 进行定性定量分析, 该方法简单高效, 灵敏准确, 为杂粮基质中真菌毒素监测提供技术手段。但HPLC-MS技术因仪器设备昂贵、对进样液净化程度要求高等问题, 一定程度上限制了该技术的广泛应用。
薄层色谱法(thin layer chromatography, TLC)是利用真菌毒素在紫外光线产生蓝紫色荧光的特性以及荧光的强弱和斑点的大小, 进行判定毒素类别和含量, 其优点是无需专业设备、操作简便、分离速度快, 可同时分离多个样品, 对样品前处理要求低等, 在定性鉴定、半定量分析中发挥着重要作用。SALISU等[44]针对粮食基质的复杂性, 对传统TLC方法进行系统性优化, 以含5%氯化钠的甲醇/水(3:1, V:V)为提取液, 以乙腈和二氯甲烷(3:17, V:V)为展开剂, 建立了TLC法检测黄曲霉毒素, 并将该方法成功应用于84批次粮食样品的黄曲霉毒素检测。但TLC法仍存在斑点显色稳定性差和定量精度有限的局限。
免疫学原理是利用抗原与抗体之间的特异性结合反应, 将其与一些可检测的信号如荧光、酶促反应等结合, 通过检测信号进行目标物质的定性及定量分析。具有灵敏度高、特异性强、快速简便、仪器设备成本低等优势。目前, 常用于真菌毒素的免疫学检测方法主要有酶联免疫吸附法(enzyme-linked immunosorbent assay, ELISA)、免疫层析分析技术(immuochromatography assay, ICA), 其中ELISA适合大量样本初次筛查, 而ICA操作简单, 耗时短, 适合现场快速检测。
ELISA是一种基于抗原-抗体特异性结合的高灵敏度检测技术, 通过酶标记的抗体或抗原, 将免疫反应转化为可定量分析的信号, 从而实现对毒素的定性和定量检测。ELISA灵敏度高、操作简便, 成本低, 适用于大批量样品的快速筛查[45-47]
由于真菌毒素作为小分子毒素, 具有较低的免疫原性, 难以得到相应的优异抗体[48]。蔡冲[49]制备建立AFM1抗独特型纳米抗体替代AFM1标准品, 抗独特型纳米抗体不仅可以作为替代抗原进行免疫学检测, 同时还可以替代真菌毒素标准品, 建立无毒、环境友好的检测方法, 可以最大程度地减小对操作人员的危害还能够避免环境的二次污染, 采用该方法对收集的20份实际样品进行测定, 结果与高效液相色谱-串联质谱法结果具有较好的一致性。表明建立的纳米抗体替代标准品ELISA免疫检测方法可应用于乳制品中AFM1的污染监测。但目前纳米抗体的规模化生产仍存在稳定性较低, 且对结构类似物存在交叉反应等劣势。
ICA是一种基于特异性免疫反应的快速检测方法, 结合了色谱层析技术和免疫标记技术, 具有操作简便、快捷, 无需任何特殊设备, 肉眼即可观察结果等优势[50-51]
边瑞丽等[52]采用芳香添加剂介导的晶种法制备金纳米棒(AuNRs), 利用其独特的光学特性, 成功构建了免疫层析试纸条法检测玉米中的AFB1, 10 min即可完成定量检测, 检出限达到0.124 ng/mL。陈绅等[53]建立了基于金纳米花(AuNFs)标记物的免疫层析试纸条方法, 可同时检测大米中DON和AFB1含量。选用AuNFs作为标记物提高了试纸条灵敏度, 实现了谷物中DON和AFB1的多重定量检测, 可视化检出限分别为0.2 ng/mL、0.5 ng/mL, 满足日常检测需求, 为谷物等现场快速筛查、风险检测和预警提供了良好的检测方法。但当前ICA技术仍面临定量依赖主观判读、动态范围窄等局限。
生物传感器是一种利用生物识别元件(如酶、抗体、抗原等)与物理化学换能器相结合的仪器, 传感器能够通过生物识别元件与目标真菌毒素的特异性结合, 引发一系列物理或化学变化, 从而实现对真菌毒素的快速、准确检测[54-55]
祁兴普等[56]通过DNA碱基互补配对, 构建了一种新颖的比率比色传感器用于OTA的检测, 研究结果表明, 在0.01~1000 ng/mL质量浓度范围内, A400/A580比值和OTA浓度的对数之间呈现良好的线性关系, 检出限可达0.033 ng/mL, 成功地应用于实际样品中OTA的检测, 回收率达到93.9%~96.0%。方鹏等[57]建立了一种基于靶标诱导滚环扩增放大荧光信号的无标记适配体快速检测OTA的生物传感器检测方法, 该方法具有较高的特异性, 检出限低至6.6×10-2 nmol/L, 成功应用于稻谷样品的分析检测。该生物传感器无需复杂化学标记、操作简单、成本低廉, 为食品安全现场快速检测提供了参考。
然而食品中通常存在多种毒素污染的实际情况, 开发能够同时检测多种毒素的高效平台成为当前研究的必然趋势。李静芝[58]建立了一种基于磁分离和双色上转换的高灵敏免疫荧光传感器技术, 用于同时检测伏马毒素B1和ZEN, 磁分离后以480 nm和550 nm处的发射峰分别作为AFB1和ZEN浓度的监测信号, 实现多种毒素的同时检测。秦英凯等[59]建立磁控双色上转换荧光法同时检测玉米和燕麦粉中OTA与ZEN的含量, 对OTA的检出限为3.97×10-2 ng/mL, 对ZEN的检出限为3.11×10-2 ng/mL, 加标回收率为91.7%~109.4%, 该方法检测灵敏度高, 特异性好, 为多种真菌毒素的检测提供了可靠依据。但生物传感器方法在实际应用中, 可能受到样品基质复杂性的干扰, 影响检测的准确性和稳定性, 且该技术设备成本较高、操作流程复杂, 导致其在基层检测机构的难以普及。
光谱法是一种利用光谱信息进行物质分析的技术, 在食品真菌毒素检测方面, 常用的光学技术有近红外光谱法[60]、荧光光谱[61]、拉曼光谱[62]、光谱成像[63]等技术, 相比传统方法, 光谱分析技术具有快速、环保、无损等技术优势, 更符合现代食品检测的需求。
张悦湘等[64]构建了花生油中AFB1的表面增强拉曼光谱快速定量检测方法, 该方法利用表面增强拉曼光谱技术对AFB1的特征信号进行强化, 使得检出限低至0.02 mg/kg。王成宏等[65]采用多种化学计量学方法, 基于激光诱导荧光技术检测人工污染的花生中AFB1, 建立了对单粒花生AFB1浓度的分类和预测。结果显示, 建立的定性模型能够精准判别花生是否受AFB1污染, 预测正确率高达100.00%, 可应用花生中的AFB1的定性和定量检测。但该技术仍存在基质效应、模型泛化、荧光背景干扰等问题。

食品中真菌毒素污染作为全球食品安全治理的顽固性难题, 广泛影响各种食品与农产品, 尤其是在花生、玉米、坚果和豆类等食品中最为严重。真菌毒素的隐蔽性强, 无法通过感官识别, 理化性质稳定, 且多数产毒真菌会产生多种有毒代谢产物, 易造成食品中多种真菌毒素共污染, 即使微量存在, 也会威胁人类和动物健康, 因此开发精准、快速、普适的先进检测方法意义重大。
近年来, 食品真菌毒素检测研究进步显著。传统检测技术如色谱法、免疫分析法, 通过优化前处理流程和升级仪器性能, 提升了检测准确性与重现性。同时, 基于纳米材料、生物传感器和分子印迹技术的新型检测方法发展迅猛, 在快速检测、多毒素同步检测和现场快速筛查领域优势突出。CRISPR-CAS基因编辑技术及人工智能辅助的数据分析模型, 为真菌毒素高通量检测与风险预测提供新思路。然而, 现有技术仍面临复杂食品基质干扰检测结果、痕量毒素富集效率低、便携式设备成本较高等挑战, 亟需在标准化、实用性和跨平台整合方面实现突破。随着材料科学、生物技术与人工智能的深度融合, 真菌毒素检测将向更精准、更智能、更普适的方向发展, 如研发新型纳米复合材料, 提升对痕量毒素的吸附与富集能力; 借助人工算法, 实现对复杂检测数据的智能分析与处理, 有效克服基质干扰; 通过多学科交叉, 开发低成本、便携式的一体化检测设备, 推动检测技术的广泛应用, 为食品安全构筑起坚固防线, 全方位满足日益增长的食品安全检测需求。
