Objective To establish an analytical method for the determination of selenium content in beef and offal (beef heart, beef liver, beef lung) using microwave digestion-hydride atomic fluorescence spectrometry (HG-AFS). Methods Based on 2 factors, namely the feeding amount and feeding time of the cattle, 80 simmental bulls weighing around 500 kg (±10 kg) were selected and randomly divided into 4 groups: A, B, C and D. Groups A, B and C served as experimental groups, while group D was the control group, with 20 cattle in each group. Selenium in beef and offal samples was digested using a nitric acid-hydrogen peroxide (HNO3-H2O2) microwave digestion method, with potassium borohydride as the reducing agent and 10% hydrochloric acid as the carrier stream, at a digestion temperature of 190 ℃. Results Selenium exhibited a good linear relationship within the mass concentration range of 0-20 ng/mL, with a correlation coefficient (r)>0.999. The average spike recovery rates (n=3) for samples A, B, C and D were 86.9%, 82.4%, 87.9% and 83.8%, respectively. Conclusion This study provides data support and a reference basis for the determination of selenium content in selenium-enriched cattle products and the development of selenium-enriched functional agriculture.

目的 建立微波消解-氢化物原子荧光光谱法(microwave digestion-hydride atomic fluorescence spectrometry, HG-AFS)测定牛肉及内脏(牛心、牛肝、牛肺)中硒含量的分析方法。方法 根据牛的饲喂量和饲喂时间两个因素, 选择体重在500 kg左右(±10 kg)西门塔尔公牛80头, 随机分成A、B、C、D 4组, A、B、C 3组作为实验组, D组为对照组, 每组20头, 采用硝酸-过氧化氢(HNO3-H2O2)微波消解牛肉及内脏样本中的硒, 硼氢化钾为还原剂, 10%盐酸为载流, 190 ℃消解。结果 硒在0~20 ng/mL质量浓度范围内呈现良好的线性关系, 相关系数(r)>0.999, A、B、C、D 4组样本的加标回收率(n=3)平均值分别为86.9%、82.4%、87.9%、83.8%。结论 该研究为富硒牛产品中硒含量的测定以及富硒功能农业的发展提供了数据支撑和参考依据。

目的 建立微波消解-氢化物原子荧光光谱法(microwave digestion-hydride atomic fluorescence spectrometry, HG-AFS)测定牛肉及内脏(牛心、牛肝、牛肺)中硒含量的分析方法。方法 根据牛的饲喂量和饲喂时间两个因素, 选择体重在500 kg左右(±10 kg)西门塔尔公牛80头, 随机分成A、B、C、D 4组, A、B、C 3组作为实验组, D组为对照组, 每组20头, 采用硝酸-过氧化氢(HNO3-H2O2)微波消解牛肉及内脏样本中的硒, 硼氢化钾为还原剂, 10%盐酸为载流, 190 ℃消解。结果 硒在0~20 ng/mL质量浓度范围内呈现良好的线性关系, 相关系数(r)>0.999, A、B、C、D 4组样本的加标回收率(n=3)平均值分别为86.9%、82.4%、87.9%、83.8%。结论 该研究为富硒牛产品中硒含量的测定以及富硒功能农业的发展提供了数据支撑和参考依据。

Objective To establish an analytical method for the determination of selenium content in beef and offal (beef heart, beef liver, beef lung) using microwave digestion-hydride atomic fluorescence spectrometry (HG-AFS). Methods Based on 2 factors, namely the feeding amount and feeding time of the cattle, 80 simmental bulls weighing around 500 kg (±10 kg) were selected and randomly divided into 4 groups: A, B, C and D. Groups A, B and C served as experimental groups, while group D was the control group, with 20 cattle in each group. Selenium in beef and offal samples was digested using a nitric acid-hydrogen peroxide (HNO3-H2O2) microwave digestion method, with potassium borohydride as the reducing agent and 10% hydrochloric acid as the carrier stream, at a digestion temperature of 190 ℃. Results Selenium exhibited a good linear relationship within the mass concentration range of 0-20 ng/mL, with a correlation coefficient (r)>0.999. The average spike recovery rates (n=3) for samples A, B, C and D were 86.9%, 82.4%, 87.9% and 83.8%, respectively. Conclusion This study provides data support and a reference basis for the determination of selenium content in selenium-enriched cattle products and the development of selenium-enriched functional agriculture.

硒是一种天然的非金属元素, 也是人体必需的微量元素之一。近些年来, 富硒农产品受到追捧和热议[1-2], 宁夏中卫作为我国代表性富硒特色区域, 有着天然的地理优势, 成为“全国硒资源变硒产业十佳地区”之一。肉牛产业是中卫市农业特色优势产业之一, 在国家、自治区和市政府的大力支持下, 肉牛养殖逐步成为全市农业农村经济增长和农民增收致富的支柱产业, 培育打造了“夏华肥牛”“穆和春”等“中国名牌农产品”。但从总体发展质量水平上看, 初级加工品比重大、品牌影响力弱、市场竞争力低、产品附加值少等问题, 依然是制约肉牛产业高质量发展的瓶颈难题[3-4]。因此, 借助国家、自治区政府扶持发展富硒功能农业的政策机遇, 通过科学的技术手段培育打造富硒“产、供、销”一体化产业链, 促进肉牛产业高质量发展具有重要的意义。
目前, 关于硒形态分析及营养品质的研究报道较多[5-9], 但是针对富硒牛肉及内脏中硒含量的分析研究较少。常用的硒元素检测方法有氢化物发生-原子荧光光谱法[10-12]、电感耦合等离子体发射光谱法[13]、电感耦合等离子体质谱法[14]、原子吸收光谱法[15-16]、荧光分光光度法[17]、电化学分析法[18]等。氢化物原子荧光光谱法因其灵敏度高、操作简单、运行成本低等优势, 广泛应用于总硒的测定[19-21], 随着科学研究的不断发展, 硒检测的方法越来越成熟与先进, 一些质谱-色谱、光谱-质谱-色谱等的联用方法也不断涌现[22]
本研究在肉牛日粮中合理添加外源性含硒添加剂, 建立微波消解-氢化物原子荧光光谱法(microwave digestion-hydride atomic fluorescence spectrometry, HG-AFS)测定牛肉及内脏中硒含量[23-25], 为探索建立富硒肉牛生产技术规程和技术标准, 以及产品的生产开发提供一定的技术依据。
选择中卫市阜华肉牛羊繁育示范基地有限责任公司(宁夏夏华集团下属全资子公司)为富硒牛肉生产技术实验示范点。在肉牛出栏前的特定时间段(90~120 d), 通过在饲料中添加植物性硒饲料添加剂, 经60 d后饲养试验结束时肉牛活重达到600 kg左右进行屠宰。胴体置于0~4 ℃冷库排酸7 d后, 分别取牛肉及内脏(牛心、牛肝、牛肺)样本, 检测总硒的含量。
硒标准储备液(质量浓度1000 mg/L, 国家有色金属及电子材料分析测试中心); 硝酸(HNO3)、盐酸(HCl)、铁氰化钾[K3Fe(CN)6]、过氧化氢(H2O2)、硼氢化钾(KBH4)(优级纯, 国药集团化学试剂有限公司); 实验用水为一级水。
AFS-933双道原子荧光光度计(北京吉天仪器有限公司); Multiwave PRO 24位微波消解仪[安东帕(上海)商贸有限公司]; SECURA 224-1CN万分之一电子分析天平(德国赛多利斯公司); UPU-III-90HW-20纯水/超纯水机(四川优普科技有限公司); GST24-2280赶酸板(天津市莱玻特瑞仪器设备有限公司)。
根据饲喂量和饲喂时间两个因素, 实验设计3个实验组和1个对照组, 选择体重在500 kg左右(±10 kg)西门塔尔公牛80头, 随机分成A、B、C、D 4组, A、B、C 3组作为实验组, D组为对照组, 每组20头。在配制全混合日粮时分别将4、5、6 kg硒动物营养添加剂分两次(日饲喂2次)加入A、B、C 3个实验组日粮内, 对照组D不添加; 各组按正常的日均采食量给予饲喂。经60 d后, 肉牛活重达到600 kg左右, 饲养实验结束时进行屠宰, 各组分别取牛肉及牛心、牛肝、牛肺4个部位的样品共320个, 测定各样本中硒的含量。
分别称取牛肉及牛心、牛肝、牛肺试样0.5~5.0 g(精确至0.001 g)置于消化罐中, 加入8 mL硝酸预消解30 min以上, 加入2 mL过氧化氢, 振摇混匀并加盖密封, 于微波消解仪中消化, 消解参数见表1。消解结束待消解液冷却后, 设定赶酸温度160 ℃, 将消解罐放入赶酸板上进行赶酸至近干, 加入5 mL浓盐酸放置在赶酸板上加热煮5 min至溶液变为清亮无色, 冷却, 转移至25 mL比色管中, 用水定容至刻度, 混匀待测, 同时做试剂空白。
从1 μg/mL硒标准溶液中准确吸取1.00 mL置于10 mL容量瓶中, 用2.0 mol/L硝酸(HNO3)稀释至刻度线, 摇匀, 制得质量浓度为0.1 μg/mL硒标准溶液Ⅰ, 备用。将0.1 μg/mL硒标准溶液至于原子荧光分光光度计中逐级稀释, 最终制得质量浓度分别为0、4、8、12、16、20 ng/mL的硒标准使用溶液, 并测定荧光强度值。
AFS-933双道原子荧光光度计: 负高压340 V, 灯电流100 mA, 原子化温度800 ℃, 炉高8 mm, 载气流速500 mL/min, 屏蔽气1000 mL/min, 进样时间10.0 s, 进样体积2.0 mL, 测其荧光值强度。
每组样品平行检测2次, 加标回收率平行测定3次, 以平均值±标准偏差表示最终结果。实验数据通过SAS 9.4软件进行方差分析, 用Microsoft Excel (15.0, 2013)制表、绘图。
本研究参照GB 5009.93—2017《食品安全国家标准 食品中硒的测定》标准中第一法氢化物原子荧光光谱法, 对牛肉及牛心、牛肝、牛肺中硒含量进行检测, 采用微波消解法处理样品, 并对标准中的检测过程进行部分优化[26-27]。首先是标准中使用电热板加热赶酸, 本研究放置到赶酸板上赶酸, 使得赶酸更加均匀、彻底, 提升回收率。其次在加入5 mL盐酸继续赶酸前, 先加入1 mL的超纯水, 近干时再加入盐酸, 可以达到充分赶酸的目的, 溶液更加清亮。
根据仪器和试剂的条件, 选择0~20 ng/mL为线性范围, 现配现用0.1 μg/mL硒标准对照液, 由仪器自动稀释6次, 分别测定荧光强度值, 以质量浓度(X, ng/mL)为横坐标, 荧光强度值(Y)为纵坐标进行线性拟合。求得A、B、C、D 4组牛肉的线性回归方程分别为: Y=112.08X+23.381 (r2=0.9994)、Y=125.71X+16.324 (r2=0.9998)、Y=109.57X-25.389 (r2=0.9996)、Y=102.38X-22.528 (r2=0.9993)。结果表明, A、B、C、D 4组牛肉在0~20 ng/mL范围内线性关系良好。
按照1.4.4项仪器设备条件, 精密吸取硒标准对照品储备液Ⅰ 2.0 mL, 重复进样6次, 分别测定荧光强度值, 计算所得相对标准偏差(relative standard deviation, RSD)为1.83%, 说明仪器精密度良好。
分别取A、B、C、D组中一个样本(耳标号00-211、0-510、00-260、000-49), 平行称取样品6个, 按照1.4.2项方法进行前处理操作, 在同样仪器设备条件下分别进样, 测定荧光强度值, 结果4组样本的RSDs分别为1.32%、1.61%、2.57%、1.91%, 表明重复性良好。
分别取A、B、C、D组中一个样本(耳标号0-508、11995、11568、11736), 平行称取样品3个, 加标水平为50 ng/g, 按照1.4.2项方法进行前处理操作, 测定含量并计算回收率, 各组平均回收率分别为86.9%、82.4%、87.9%、83.8%之间, 结果较为满意, 详见表2
牛肉是蛋白质的重要来源, 品质直接影响消费者的健康, 外源硒作为饲料添加剂, 能够有效提高牛肉中的硒含量, 通过外源硒的添加补充, 进入牛体内后, 主要经过吸收代谢、抗氧化作用、免疫调节等方式发挥作用, 可以直接或者间接改善牛肉的营养价值和自身品质[28]。本研究分别将A、B、C、D 4组中牛肉及牛心、牛肝、牛肺4个部位共320个样品按照1.4.2项下方法进行前处理操作, 并上机测定硒含量, 结果见表3
实验数据经SAS软件进行方差分析, 结果见表4
表4中可以看出, 横向对比分析, A、B、C、D组中牛肝、牛肺的硒含量均明显高于牛心和牛肉。
纵向对比分析, B组牛心部位的硒含量显著高于其他3组(P<0.05); A、B、C、D各组牛肝和牛肺的硒含量差异不显著(P>0.05); B组和C组牛肉的硒含量显著高于A组和D组(P<0.05)。总体来看, 牛肝中硒含量最高, 牛肺和牛心次之, 牛肉中硒含量相对较低。
依据GH/T 1135—2017《富硒农产品》规定, 肉类中总硒含量≥0.075 mg/kg为含有硒元素农产品, 总硒含量≥0.15 mg/kg为富含硒元素农产品。本研究中内脏总体硒含量较高, 牛肝达到富含硒元素农产品标准, B组的牛心、牛肺和C组的牛肺达到富含硒元素农产品标准; B组和C组牛肉达到含硒标准。
本研究发现日粮添加硒可以显著提高牛肉及牛心、牛肝、牛肺中硒含量。有研究称[29-30], 添加有机硒更易引起牛肌肉中硒含量的显著提高, 随着硒添加时间的延长, 牛肉硒含量呈现出先缓慢增加后快速增加的趋势。徐娥等[31]发现在日粮中添加有机硒, 不仅可以极显著提高牛肉中的硒含量, 钙、铁、镁的含量也得到了不同程度的提高。
本研究通过对4组牛肉及牛心、牛肝、牛肺样本中硒含量的检测, 得到一些富硒肉牛养殖经验, 在肉牛日粮中添加适量富硒营养添加剂后, 牛肉及牛心、牛肝、牛肺中硒含量与日粮中硒的添加量呈正相关, 且在肉牛出栏前2个月内日粮中添加适量富硒营养添加剂, 不会引起肉牛硒元素蓄积中毒。
本研究在肉牛养殖出栏前开展日粮添加外源硒对牛肉及内脏中硒含量吸收情况进行实验测定, 检测的是总硒含量, 但由于时间、人员和设备设施等客观因素限制, 没有针对硒的不同添加时间对肉牛生长性能及牛肉品质的影响进行研究, 未检测牛粪便和尿液中硒的含量[32-33]。因此, 针对加入富硒营养添加剂及加入量的范围, 肉牛吸收硒的数量或能力大小, 牛肉及内脏中硒形态等方面的内容, 有待进一步研究探讨。
