Objective To optimize the preparation process of Brassica rapa L. polysaccharide oral liquid and test its quality. Methods Graded alcohol precipitation was used to study the yield of crude polysaccharides of Brassica rapa L. at different ethanol concentrations. The optimal formulation of oral solution of Brassica rapa L. polysaccharide was screened by one-way and orthogonal tests, and the quality was tested according to the pharmacopoeia. Results The extraction rates of crude polysaccharide from Brassica rapa L. were 12.8%, 9.2%, 23.2% and 24.8% for the alcohol precipitation group with ethanol concentration of 20%, 40%, 60% and 80%, respectively. The optimal preparation process conditions of oral solution of Brassica rapa L. refined polysaccharide were: 2% addition of Brassica rapa L. crude polysaccharide, 15% addition of honey, 0.6% addition of citric acid and 0.6% addition of sodium citrate. The best preparation process conditions for oral liquid of Brassica rapa L. crude polysaccharide were: 2% addition of Brassica rapa L. crude polysaccharide, 15% addition of honey, 0.6% addition of citric acid, 0.6% addition of sodium citrate. The pH of the oral solution of refined polysaccharide of Brassica rapa L. and the oral solution of crude polysaccharide of Brassica rapa L. were 4-5. The relative density of the oral solution of refined polysaccharide of Brassica rapa L. was 1.106 g/mL. The relative density of the oral solution of crude polysaccharide of Brassica rapa L. was 1.100 g/mL. Conclusion The optimal preparation process conditions for refined polysaccharides from Brassica rapa L. and crude polysaccharides from Brassica rapa L. are the same, but the taste, odor, and appearance of the former are superior to those of crude polysaccharides from Brassica rapa L.. However, the extraction process for refined polysaccharides from Brassica rapa L. is slightly more complex and not suitable for mass production in factories.

目的 优化芜菁多糖口服液的制备工艺, 并对其进行质量检测。方法 采用分级醇沉研究不同乙醇浓度下芜菁粗多糖的得率。通过单因素实验和正交实验筛选出芜菁多糖口服液的最佳配方, 并根据药典规定进行质量检测。结果 乙醇浓度为20%、40%、60%和80%的醇沉组的芜菁粗多糖得率分别为12.8%、9.2%、23.2%和24.8%。芜菁精制多糖口服液最佳制备工艺条件为: 芜菁精制多糖添加量2%, 蜂蜜添加量15%, 柠檬酸添加量0.6%, 柠檬酸钠添加量0.6%。芜菁粗多糖口服液最佳制备工艺条件为: 芜菁粗多糖添加量2%, 蜂蜜添加量15%, 柠檬酸添加量0.6%, 柠檬酸钠添加量0.6%。芜菁精制多糖口服液pH和芜菁粗多糖口服液pH均在4~5。芜菁精制多糖口服液其相对密度为1.106 g/mL, 芜菁粗多糖口服液其相对密度为1.100 g/mL。结论 芜菁精制多糖和芜菁粗多糖口服液的最佳制备工艺条件相同, 前者的口味、气味以及外观均优于芜菁粗多糖口服液, 但是芜菁精制多糖的提取工艺略显复杂, 不适用于工厂批量生产。

目的 优化芜菁多糖口服液的制备工艺, 并对其进行质量检测。方法 采用分级醇沉研究不同乙醇浓度下芜菁粗多糖的得率。通过单因素实验和正交实验筛选出芜菁多糖口服液的最佳配方, 并根据药典规定进行质量检测。结果 乙醇浓度为20%、40%、60%和80%的醇沉组的芜菁粗多糖得率分别为12.8%、9.2%、23.2%和24.8%。芜菁精制多糖口服液最佳制备工艺条件为: 芜菁精制多糖添加量2%, 蜂蜜添加量15%, 柠檬酸添加量0.6%, 柠檬酸钠添加量0.6%。芜菁粗多糖口服液最佳制备工艺条件为: 芜菁粗多糖添加量2%, 蜂蜜添加量15%, 柠檬酸添加量0.6%, 柠檬酸钠添加量0.6%。芜菁精制多糖口服液pH和芜菁粗多糖口服液pH均在4~5。芜菁精制多糖口服液其相对密度为1.106 g/mL, 芜菁粗多糖口服液其相对密度为1.100 g/mL。结论 芜菁精制多糖和芜菁粗多糖口服液的最佳制备工艺条件相同, 前者的口味、气味以及外观均优于芜菁粗多糖口服液, 但是芜菁精制多糖的提取工艺略显复杂, 不适用于工厂批量生产。

Objective To optimize the preparation process of Brassica rapa L. polysaccharide oral liquid and test its quality. Methods Graded alcohol precipitation was used to study the yield of crude polysaccharides of Brassica rapa L. at different ethanol concentrations. The optimal formulation of oral solution of Brassica rapa L. polysaccharide was screened by one-way and orthogonal tests, and the quality was tested according to the pharmacopoeia. Results The extraction rates of crude polysaccharide from Brassica rapa L. were 12.8%, 9.2%, 23.2% and 24.8% for the alcohol precipitation group with ethanol concentration of 20%, 40%, 60% and 80%, respectively. The optimal preparation process conditions of oral solution of Brassica rapa L. refined polysaccharide were: 2% addition of Brassica rapa L. crude polysaccharide, 15% addition of honey, 0.6% addition of citric acid and 0.6% addition of sodium citrate. The best preparation process conditions for oral liquid of Brassica rapa L. crude polysaccharide were: 2% addition of Brassica rapa L. crude polysaccharide, 15% addition of honey, 0.6% addition of citric acid, 0.6% addition of sodium citrate. The pH of the oral solution of refined polysaccharide of Brassica rapa L. and the oral solution of crude polysaccharide of Brassica rapa L. were 4-5. The relative density of the oral solution of refined polysaccharide of Brassica rapa L. was 1.106 g/mL. The relative density of the oral solution of crude polysaccharide of Brassica rapa L. was 1.100 g/mL. Conclusion The optimal preparation process conditions for refined polysaccharides from Brassica rapa L. and crude polysaccharides from Brassica rapa L. are the same, but the taste, odor, and appearance of the former are superior to those of crude polysaccharides from Brassica rapa L.. However, the extraction process for refined polysaccharides from Brassica rapa L. is slightly more complex and not suitable for mass production in factories.

芜菁(Brassica rapa L.), 又叫蔓菁, 新疆人称恰玛古。它是十字花科芸苔属二年生植物[1], 常用来治疗食欲不振、腹泻、疮疖肿毒等疾病[2-4]。其含有多糖、黄酮、硫代葡萄糖苷等, 这些化合物表现出抗炎、抗肿瘤、抗氧化、抗疲劳、降血糖和调节免疫功能等多种作用[5-8]
芜菁多糖具有多种药理活性和医疗保健作用[9-10]。已有学者对芜菁多糖的化学结构、组成、理化性质, 以及不同提取工艺对其化学成分的影响等方面[11]进行了研究。研究发现芜菁多糖对生物体的生理作用和药理作用, 如抗氧化、抗菌、降血糖、降血脂、免疫调节等[12]。同时在医学领域研究发现芜菁多糖在预防与治疗多种疾病中均有效果, 如癌症、心血管疾病、肝病、糖尿病、肥胖症等[13-17]
芜菁多糖提取方法多样[18-20], 其中水提醇沉法是中药及食用菌多糖常用的提取方法, 具有操作简单, 成本低廉, 准确度高等优点, 但该方法得率相对较低, 本研究主要基于芜菁多糖水提法优化的基础上, 对醇沉最佳条件进行探究, 以期提高水提醇沉法的得率。
近年来随着芜菁多糖的深入研究, 关于芜菁多糖各类制剂的研究逐渐增多[21], 但芜菁多糖口服液的研究仍有欠缺。在制备口服液的过程中, 使用水提醇沉法对芜菁多糖进行提取和分离, 得到高纯度的芜菁多糖。结合制剂工艺技术, 根据芜菁多糖的物理化学性质和药效特点, 设计制剂工艺制备出稳定、有效、口味良好的口服液。
本研究在确定了芜菁多糖的最佳醇沉条件后, 将通过单因素和正交实验筛选出芜菁多糖口服溶液的最佳配方, 并比较芜菁粗多糖口服溶液和芜菁精制多糖口服溶液的口味差异, 为芜菁多糖的应用与开发奠定基础。
UV-2700紫外可见分光光度计(岛津仪器苏州有限公司); SHB-III循环水式真空多用泵(郑州长城科工贸有限公司); N-1001旋转蒸发仪、SB-1000水浴锅(上海爱朗仪器有限公司); 透析袋MD34-7000(北京索莱宝科技有限公司)。
芜菁脱脂片于2023年3月由新疆维吾尔自治区阿克苏地区柯坪县艾丽努尔公司提供, 经北京大学药学院张英涛副教授鉴定为十字花科芸苔属植物芜菁(Brassica rapa L.)的地下茎。
95%乙醇(批号20221125, 天津市凯通化学试剂有限公司); 氢氧化钠(批号20160308, 天津市盛奥化学试剂有限公司); 浓盐酸(批号2021719, 新疆医科大学中心实验室); 柠檬酸(批号C10576601, 上海麦克林生化科技股份有限公司); 柠檬酸钠(批号20220514, 西安拉维亚生物科技有限公司)。
本研究操作步骤如图1所示。
取芜菁干燥药材浸泡于95%乙醇中脱脂24 h, 取出晾干后用粉碎机粉碎成粉末, 过80目筛后备用。
参考阿曼妮萨·麦提如则等[22]的方法采用回流提取法进行提取。称取芜菁粉末, 加入蒸馏水, 按照料液比1: 30 (g/mL)放入圆底烧瓶, 装上并打开冷凝装置, 加热套加热提取, 温度为90 ℃, 单次回流时间为2 h, 回流3次。趁热过滤, 收集3次滤液, 蒸发浓缩备用, 该法芜菁多糖得率为11.56%。
参考林丽芹等[23]的方法并加以改进, 调整浓缩液乙醇浓度分别为20%、40%、60%、80%, 搅拌均匀, 4 ℃, 24 h, 弃上清液, 取沉淀晾干, 得干燥芜菁粗多糖, 干燥后分别称量不同浓度乙醇溶液醇沉出的芜菁粗多糖的重量, 计算不同浓度乙醇的得率。多糖得率计算见公式(1):
多糖得率/%=(m1/m)×100%
式中: m1为芜菁粗多糖质量, mg; m为芜菁干燥饮片质量, mg。
参考古丽米拉·卡德尔等[24]的方法制备芜菁精制多糖。准备7000 Da的透析袋, 将部分粗多糖溶于水, 制成芜菁粗多糖溶液, 将芜菁粗多糖溶液倒入透析袋中。将透析袋置于容器中, 加蒸馏水没过透析袋, 每6 h换一次水, 透析48 h。将透析液倒至离心管中, 4000 r/min离心5 min, 倾倒出上清液, 浓缩, 留存备用。准备AB-8大孔吸附树脂, 置于9%乙醇溶液中, 浸泡24 h, 滤出, 用蒸馏水冲洗。用5%的HCl溶液浸泡树脂3 h, 蒸馏水冲洗至洗出液为中性, 再倒入2% NaOH溶液浸泡3 h, 蒸馏水冲洗直至洗出液为中性, 放置备用。将预处理好的大孔吸附树脂湿法装柱, 芜菁粗多糖溶液上样, 静态吸附15 min, 流速为2 mL/min, 收集流出液并用7000 Da的透析袋进行透析, 蒸发浓缩, 冻干保存, 得芜菁精制多糖。
(1)单因素实验设计
结合舒文莉等[25]与耿春叶等[26]的方法并加以改进。①芜菁粗多糖与精制多糖添加量单因素实验: 固定柠檬酸添加比例0.6%, 蜂蜜添加比例为15%, 柠檬酸钠添加比例为0.6%, 分别加入芜菁精制多糖或芜菁粗多糖(2%、4%、6%、8%、10%), 蒸馏水定容至10 mL, 制备芜菁多糖口服溶液。②蜂蜜添加量单因素实验: 固定芜菁精制多糖或芜菁粗多糖比例为6%, 柠檬酸添加比例为0.6%, 柠檬酸钠添加比例为0.6%, 分别加入蜂蜜(5%、10%、15%、20%、25%), 蒸馏水定容至10 mL, 制备芜菁多糖口服溶液。③柠檬酸添加量单因素实验: 固定芜菁精制多糖或芜菁粗多糖比例为6%, 蜂蜜添加比例为15%, 柠檬酸钠添加比例为0.6%, 分别加入柠檬酸(0.2%、0.4%、0.6%、0.8%、1.0%), 蒸馏水定容至10 mL, 制备芜菁多糖口服溶液。④柠檬酸钠添加量单因素实验: 固定芜菁精制多糖或芜菁粗多糖比例为6%, 蜂蜜添加比例为15%, 柠檬酸添加比例为0.6%, 分别加入柠檬酸钠(0.2%、0.4%、0.6%、0.8%、1.0%), 蒸馏水定容至10 mL, 制备芜菁多糖口服溶液。柠檬酸、蜂蜜、柠檬酸钠添加量单因素试验中由于多糖添加比例相同, 最终只呈现一种结果。
(2)正交实验设计
基于单因素实验并结合李倩等[27]与马利平等[28]方法设计正交实验, 采用L9(34)的正交优化芜菁多糖口服溶液的配方, 实验设计水平见表12
(3)感官评价
参考吴伊莉等[29]与彭中旭等[30]的方法设计芜菁多糖铁口服液感官评价标准, 见表3。感官评价标准总分为100分, 实验结果以随机抽取10位大学生志愿者各自评分总和的平均值表示。
(4)最佳合成工艺下芜菁多糖口服溶液品质分析
相对密度测定: 相对密度的检测方法参照《药典2020版》通则0601中规定的方法(比重瓶法)测定。
pH测定: 参照《药典2020版》通则0631 pH测定法测定。
外观指标检验方法: 将芜菁多糖口服液置于光线明亮处, 用肉眼观察其是否澄清。
每组实验重复3次, 使用Excel 2021软件做数据统计与计算, 结果用平均值和±标准偏差表示; 使用SPSS 25软件进行方差分析, P<0.05认为具有统计学意义, 使用Origin 2018软件作图。
图2所示, 经称量计算, 醇沉时乙醇浓度为20%的组芜菁多糖得率为12.8%, 乙醇浓度为40%的粗多糖得率为9.2%, 乙醇浓度为60%的粗多糖得率为23.2%, 乙醇浓度为80%的粗多糖的得率为24.8%, 增长率逐渐降低。结果显示, 当乙醇浓度从60%增长到80%时, 芜菁粗多糖得率的增长速度变低且达到最高, 此时P<0.05数据结果有显著性差异。
图3A、B可知, 感官评分与芜菁多糖(精制多糖、粗多糖)添加量呈负相关, 当芜菁多糖添加量为2%时, 评分达到最高值, 这可能归因于此时多糖的添加量在体系中达到了最佳平衡状态, 既能改善产品的质构特性, 又不会对溶液的澄明度和口感产生负面影响。然而, 当添加量超过2%后, 感官评分显著下降, 推测是由于多糖分子间的相互作用增强, 导致不溶物增多, 溶液澄明度降低; 同时, 过量的多糖可能改变了体系的流变特性, 进而影响口感、气味和组织状态。因此, 2%的添加量可能是多糖在体系中溶解性和功能性的临界点, 超过这一比例后, 多糖的负面效应逐渐显现。
图3C可知, 感官评分数据显示, 随着蜂蜜添加量的增加, 评分呈现先上升后下降的趋势。当添加量为15%和20%时, 感官评分达到最高值; 而当添加量增至25%时, 评分显著下降。这一变化趋势可能与蜂蜜的甜度及其对体系流变特性的影响有关。在添加量为15%时, 蜂蜜的香甜味能够有效提升产品的风味, 同时不会对体系的质地和口感产生明显负面影响。当添加量达到20%时, 尽管感官评分仍处于较高水平, 但溶液的黏稠度略有增加, 可能导致口感略微厚重。而当添加量进一步增加至25%时, 过高的甜度和黏稠度可能破坏了风味的协调性, 使口感过于甜腻, 从而降低了感官评分。因此, 综合考虑风味和质地的平衡, 15%的蜂蜜添加量为最优选择。
图3D可知, 当柠檬酸添加量为0.6%时, 评分达到最高值; 而当添加量增至0.8%时, 感官评分开始下降。这一变化趋势可能与柠檬酸对溶液酸度及风味平衡的影响有关。在添加量为0.6%时, 柠檬酸能够有效提升溶液的酸度, 增强口感的清爽度, 同时不会对整体风味的协调性产生负面影响。然而, 当添加量增加至0.8%时, 过高的酸度可能导致口感过于刺激, 破坏了溶液的风味平衡, 使整体口感显得不协调, 从而降低了感官评分。因此, 0.6%的柠檬酸添加量能够在提升口感的同时维持风味的协调性, 是最优选择。
图3E可知, 当柠檬酸钠添加量为0.6%时, 评分达到最高值; 添加量增至0.8%时, 感官评分显著下降。这一现象可能与柠檬酸钠对溶液酸碱平衡及离子强度的调控作用有关。当添加量为0.6%时, 柠檬酸钠能够有效中和体系中过量的酸性成分, 稳定溶液pH, 从而改善口感的柔和度与协调性。然而, 当添加量超过0.6%(如0.8%)时, 过高的钠离子浓度可能导致溶液离子强度增加, 引发轻微刺激性口感; 同时, 过量柠檬酸钠可能打破原有的酸碱平衡, 使溶液pH偏离适宜范围, 导致风味失衡。因此, 0.6%的添加量既能优化酸度调节效果, 又可避免因离子浓度或pH异常引起的感官负面影响, 是最佳选择。
表4表5可知4个因素的影响关系为A>C>B>D, 通过R值分析最佳的工艺参数为A1B2C2D2, 即芜菁精制多糖添加量为2%, 蜂蜜添加量为15%, 柠檬酸添加量为0.6%, 柠檬酸钠添加量为0.6%。在这个最佳条件下做3组平行验证实验, 芜菁精制多糖口服液感官评分均在85分以上, 平均分为87分。表5结果显示, 因素A影响显著: F=9.712, P<0.05, 偏方差和(472.667)远高于误差项(48.667), 效应量较大, 是主要影响因素。因素B (F=1.329)和因素C (F=1.411)虽显著, 但效应量较低, 实际影响有限。
表6表7可知4个因素的影响关系为A>B>C>D, 通过R值分析最佳的工艺参数为A1B2C2D2, 即芜菁粗多糖添加量为2%, 蜂蜜添加量为15%, 柠檬酸添加量为0.6%, 柠檬酸钠添加量为0.6%。在这个最佳条件下做3组平行验证实验, 芜菁粗多糖口服液感官评分均在80分以上, 平均分为82分。表7结果显示, 因素A对因变量的影响显著: F=39.000, P<0.05, 其偏方差和(546.000)远高于误差项(14.000), 效应量极大, 表明因素A是主要影响因素。因素B虽也显著(F=2.333, P<0.05), 但其效应量较低, 实际影响可能有限。
根据最佳配方配制的芜菁精制多糖口服液, 色泽: 口服液呈淡黄色, 颜色均一; 风味: 具有淡淡的芜菁独有的气味, 口味酸甜, 无异味; 组织状态: 溶液均匀, 稳定性良好, 无明显分层和沉淀现象。
根据最佳配方配制的芜菁粗多糖口服液, 色泽: 呈棕黄色, 颜色均一; 风味: 口感酸甜, 但是芜菁特有的气味稍浓; 组织状态: 无明显分层现象, 长时间放置后有少量沉淀生成, 不影响使用。
芜菁精制多糖口服液pH在4~5(平行测量3次), 芜菁粗多糖口服液pH在4~5(平行测量3次)。
芜菁精制多糖口服液, 其相对密度为1.106 g/mL。芜菁粗多糖口服液, 其相对密度为1.100 g/mL。
本研究以芜菁多糖为原料, 制备芜菁精制多糖口服液和芜菁粗多糖口服液, 并通过正交实验优化其制备工艺, 筛选了口服液的最佳配方。得到芜菁精制多糖口服液最佳制备工艺条件为: 芜菁精制多糖添加量2%, 蜂蜜添加量15%, 柠檬酸添加量0.6%, 柠檬酸钠添加量0.6%。该条件下芜菁精制多糖口服液感官评分为87分。此时制备的芜菁精制多糖口服液其感官指标、理化指标都符合或高于药典国家标准的要求。芜菁粗多糖口服液最佳制备工艺条件为: 芜菁粗多糖添加量2%, 蜂蜜添加量15%, 柠檬酸添加量0.6%, 柠檬酸钠添加量0.6%。该条件下芜菁粗多糖口服液感官评分为82分。此时制备的芜菁粗多糖口服液其感官指标、理化指标都符合或高于药典国家标准的要求。将两种口服液对比, 芜菁精制多糖口服液的口味, 气味以及外观均优于芜菁粗多糖口服液, 从批量化生产工艺来讲, 芜菁粗多糖口服液相较于芜菁精制多糖口服液工艺更为简便。
