Objective To analyze the differences in mineral elements in wild rice from different regions and establish a method for tracing the origin of wild rice based on mineral element fingerprint characteristics. Methods Inductively coupled plasma mass spectrometry (ICP-MS) was used to analyze the content of 24 kinds of mineral elements in 40 wild rice samples from the United States, Canada, and China. The data were subjected to variance analysis, multiple analysis, principal component analysis, and discriminant analysis to explore the feasibility of using mineral element fingerprint characteristics for tracing the origin of wild rice. Results The content of 24 kinds of elements showed significant differences among the regions. Principal component analysis extracted 3 principal components, with a cumulative variance contribution rate of 93.1%. Principal component analysis successfully distinguished wild rice from the United States and Canada. Discriminant analysis correctly identified 97.7% of the wild rice samples. Conclusion This study demonstrates the feasibility and accuracy of classifying wild rice samples from different origins.

目的 分析不同地区野米中矿物元素的差异, 建立基于矿物质元素指纹特征的野米产地溯源方法。方法 应用电感耦合等离子体质谱法(inductively coupled plasma mass spectrometry, ICP-MS)分析来源于美国、加拿大和中国3个地域40个野米样品中24种矿物质元素含量, 并对数据进行方差分析、多重分析、主成分分析和判别分析。探索矿物元素指纹特征对野米产地溯源的可行性。结果 24种元素含量在产地间差异显著, 主成分分析得到3个主成分, 其累计方差贡献率达到93.1%。主成分分析可将产自美国和加拿大的野米区分开; 判别分析中有97.7%的野米被正确识别。结论 该研究对不同产地野米样品的分类具有可行性和准确性。

Objective To analyze the differences in mineral elements in wild rice from different regions and establish a method for tracing the origin of wild rice based on mineral element fingerprint characteristics. Methods Inductively coupled plasma mass spectrometry (ICP-MS) was used to analyze the content of 24 kinds of mineral elements in 40 wild rice samples from the United States, Canada, and China. The data were subjected to variance analysis, multiple analysis, principal component analysis, and discriminant analysis to explore the feasibility of using mineral element fingerprint characteristics for tracing the origin of wild rice. Results The content of 24 kinds of elements showed significant differences among the regions. Principal component analysis extracted 3 principal components, with a cumulative variance contribution rate of 93.1%. Principal component analysis successfully distinguished wild rice from the United States and Canada. Discriminant analysis correctly identified 97.7% of the wild rice samples. Conclusion This study demonstrates the feasibility and accuracy of classifying wild rice samples from different origins.

随着生活水平的提高, 公众健康养生意识的增强, 对饮食的要求已经由一般性的满足传统的营养需求转向更加科学健康、营养均衡的膳食需求, 尤其是天然健康的原料备受青睐。野米, 又称为菰米, 具有很高的营养价值, 不仅含有丰富的蛋白质、必需氨基酸、脂肪酸、维生素以及多种微量元素[1-3], 还含有大量生物活性物质, 用于预防消渴症和胃肠疾患[4]以及心血管疾病的发生[5-9]。野米主要产地在北美地区, 尤其是生长在世界上最大淡水湖区域的冰湖野米, 由于其生长环境无污染, 并具有特殊的地质结构从而保证它奇特的营养成份及药效, 被誉为“谷物中的鱼子酱”, 价格高昂[10-12]。在经济利益的驱使下, 市面上的野米产品鱼龙混杂, 存在着以假乱真、以次充好的情况, 严重损害消费者权益。因此, 如何确定产品产地来源是亟需解决的问题。
近年来针对产地溯源的研究越来越深入, 目前的研究技术主要是基于矿物元素指纹图谱和稳定同位素质谱检测的方法, 同时结合以近红外光谱、电子舌等现代无损分析模式, 可针对地理环境形成的矿物元素组成、同位素分布以及产品品质差异等特征性指标来判定产品的地理来源[13-14]。而野米作为植物源食品, 其植物生长过程所需要的矿质养分主要是从其产地土壤中吸取[15], 所以某一产区的野米中矿质元素含量与其产地土壤的矿质元素含量存在极强的相关性, 植物矿质元素的含量和种类可以作为一个重要的溯源指标。电感耦合等离子体质谱法(inductively coupled plasma mass spectrometry, ICP-MS)由于具有线性范围宽、检出限低、快速的多元素检测能力以及能提供同位素比信息而得到了广泛应用[16-23]。主成分分析(principal component analysis, PCA)是通过原始变量的少数几个线性组合来解释原始变量的绝大部分信息, 并寻求主成分来研究样本的一种方法, 是通过综合的方式把样品中多种元素的信息直观地表达出来, 目前在农产品溯源技术中被广泛应用[24-25]。为探讨矿物质元素对野米产地辨别的影响, 以此建立辨别模型。PCA得分图可以将对照组和实验组样本区分。在PCA图中, 如果样本数据聚集在一起, 说明样本间差异性小, 如果样本之间呈分散状, 距离较远, 说明样本之间差异性较大。因此, 本研究利用ICP-MS测定不同产地野米种常量和微量元素[26], 并结合化学计量学中的方差分析(analysis of variance, ANOVA)及PCA, 建立基于矿物元素指纹分析的加拿大冰湖野米产地溯源技术, 并为野米进口监管提供技术支撑。
通过实验室留存样品、市场及其他购买渠道, 收集国产野米(G#) 10份、加拿大冰湖野米(1#) 15份和美国产野米(3#) 15份, 共计40份样品。
7900型电感耦合等离子体质谱仪(美国安捷伦公司); TOPEX+型微波消解系统(PreeKEM科学仪器有限公司); AE240电子天平(精度0.01 mg, 瑞士梅特勒-托利多公司)。
硝酸(优级纯, 国药集团化学试剂有限公司); 氩气(Ar, 纯度99.995%以上, 大连光明特别气体有限公司); 纯水(电阻率为18.2 MΩ·cm, sartorius arium pro VF-T实验室超纯水系统); 混合元素标准储备液、质谱调谐液标准混合溶液、混合内标储备溶液(质量浓度为10 μg/mL, 美国安捷伦公司)。
(1)前处理方法
分别称取0.5 g(精确至0.001 g)样品置于微波消解管中, 加入6 mL 65%硝酸, 室温条件下预消解1 h, 再加入2 mL 30%的过氧化氢预消解0.5 h, 然后放入微波消解仪中进行消解。
用ICP-MS测定24种矿物元素浓度(B、Na、Mg、Al、K、Ca、V、Cr、Mn、Fe、Co、Ni、Cu、Zn、As、Sr、Mo、Ag、Cd、Sn、Sb、Ba、Hg、Pb)。
(2)测定仪器条件
仪器参数设置为: 射频功率为1550 W, 氦气流速为4.35 mL/min, 石英玻璃同心雾化器, 采样时间为0.1 s。
温度先上升到120 ℃, 保持2 min; 再上升至160 ℃, 保持5 min; 再上升至180 ℃, 保持15 min。将消化后的溶液转移到塑料样品管中, 并用超纯水定容至15 mL。每个样品进行两个平行实验。Rh、Re、In和Ge的内标溶液用于消除基质效应和仪器偏差。内标溶液为单元素标准溶液配制成质量浓度为10 µg/L的混合溶液。采用外标法进行定量分析, 标准曲线的线性回归系数大于0.99。
实验至少重复测定3次, 采用SPSS 26.0软件进行方差分析(analysis of variance, ANOVA)、多重比较分析和PCA。
通过对40份野米样品中24种矿质元素的含量进行多重比较分析, 结果表明(见表1), 只有K、Sn元素在中国、美国和加拿大产的野米中含量无显著差异(P>0.05)。Mg、Al、Ca和Ba元素含量在中国与另外两个产地之间的差异显著(P<0.05), 而Co和Ni元素含量在美国与另外两个产地之间的差异显著(P<0.05)。其余元素在3个不同产地野米中均存在显著的差异(P<0.05), 其中B、Na、Cu、Cd和Hg元素含量在3个产地中的大小顺序为美国>加拿大>中国; Cr、Fe、As和Ag元素含量在3个产地中的大小顺序为美国>中国>加拿大; V、Mn和Mo元素含量在3个产地中的大小顺序为中国>美国>加拿大; Zn元素含量在3个产地中的大小顺序为加拿大>美国>中国; Sb和Pb元素含量在3个产地中的大小顺序为加拿大>中国>美国。一般情况下, 土壤中的矿质元素种类及丰度具有地理性质特异性, 而土壤矿元素特性直接影响植物矿质元素含量, 即可以通过植物中的矿物元素对生长环境进行溯源。近年来, 利用不同产地来源的植物中矿质元素组成的差异性进行区域特色农产品或道地药材的产地溯源已被证明有广泛的应用价值。
本研究对加拿大和美国野米中24种元素进行主成分分析, 结果显示前3个主成分的累计方差贡献率为93.1%(表2), 第一个主成分的特征值为20.16, 特征贡献率为84.0%, 主要影响因子为Na、Co、Ni、Cu、As、Sr、Mo、Sb、Hg; 第二个主成分的特征值为1.22, 特征贡献率为5.1%, 主要影响因子为Zn; 第三个主成分的特征值为0.98, 特征贡献率为4.1%, 主要影响因子为Fe。在美国和加拿大野米矿物元素PCA分析图中(图1), 不同颜色代表不同产地的样品, PCA分析显示不同地区的矿物元素含量有明显区分, 可将产自不同地区的野米区分开。
为进一步明确矿物质元素指标在野米产地判别中的作用及效果, 利用逐步判别分析筛选重要的变量, 选择那些能够鉴别产地的变量因素。其可利用各类样品之间的相关性建立判别模型而对未知样品进行分类, 还可以采用交叉验证方法对剩余样品作为训练集, 用建立的判别模型对其进行判别, 从而极大地增加了判别模型的稳定性, 本研究对在主成分分析中差异显著的11种元素进行分析[27]。如表3所示, 在0.05的显著水平上, Na、Co、Ni、Cu、As、Sr、Mo、Sb、Hg、Zn、Fe 11种对野米产地判别作用显著的元素建立判别模型:

该模型的交叉验证结果如表4所示, 11种元素分析的原始整体判别率为100.0%; 交叉验证的分类结果中, 97.7%的野米被正确识别。所以, 该模型的建立对野米的产地具有有效的判断力, 为鉴别冰湖野米原产地指纹判别模型的构建提供理论基础。
本研究基于目前的产地溯源技术[28-29], 以来源于美国、加拿大和中国3个地区的野米为研究对象, 通过对样品中矿物质元素进行分析, 探究来自于加拿大的冰湖野米与其他野米的微量元素含量的区别, 从而可以有效区分不同产地的野米。研究结果显示,加拿大冰湖野米在Zn、As、Mo、Sb、Fe等元素与美国产野米以及国内市场收集的样品有显著差异; PCA结果明确了基于矿质元素含量可实现不同产地野米样品的分类。根据对野米产地判别作用显著的11种元素建立判别模型, 97.7%的样品可以正确识别, 表明建立的矿物元素溯源方法可以应用于冰湖野米的产地溯源。
野米的植物微量元素保证了其作为一种功能性食品的发展[30-32]。但由于产地不同微量元素含量差别较大, 加拿大冰湖野米因独特的地理优势使其成为野米中的“贵族”, 高昂的价格导致市面上存在以假乱真、以次充好的现象。因此, 如何对产品真实性进行判别势在必行。本研究建立的矿物元素指纹图谱技术为野米的产品溯源提供了有力的技术支撑。目前, 矿物元素指纹分析技术已经广泛应用到食品的多个领域, 包括粮食、肉制品、蔬菜、水果等。产地溯源技术的发展可以有效保障产品的真实性。
