Objective To study the tissue distribution and metabolic regulation of diazepam (DZP) in Carassius auratus after feeding with positive bait containing DZP. Methods Carassius auratus was selected as the study object and exposed to DZP-enriched bait through gavage, and tissue samples including scales, skin, muscle, plasma, gills, intestines, liver, gallbladder, gonads, and brain were collected at specific intervals ranging from 1 h to 456 h post-exposure. High performance liquid chromatography coupled with tandem high-resolution mass spectrometry was employed to detect DZP and its metabolites—nordazepam (NZP), temazepam (TZP), and oxazepam (OZP)—in these tissues, analyze their distribution and metabolic patterns. Results Following exposure to DZP-enriched bait, DZP rapidly accumulated in Carassius auratus tissues, with substantial residues persisting for up to 456 h. The highest concentrations were primarily found in the gonads, liver and gallbladder, with the gonads consistently showing elevated levels. NZP and TZP emerged as the primary metabolites. Conclusion DZP predominantly exists in its prototype form in Carassius auratus and tends to accumulate in the gonads. The prolonged metabolic cycle of DZP and its metabolites in Carassius auratus indicates potential food safety and health risks for consumers of aquatic products caught with DZP-containing bait.

目的 研究鲫鱼摄食含地西泮(diazepam, DZP)的阳性饵料后, DZP在其体内的组织分布及代谢规律。方法 选择鲫鱼作为研究对象, 使用含DZP的阳性饵料进行灌胃给药, 并在给药后1 h至456 h内, 定时对鱼鳞、鱼皮、肌肉、血浆、鳃、肠、肝脏、胆、性腺及脑等组织进行取样。采用高效液相色谱-串联高分辨质谱技术, 检测各组织中DZP及其代谢物去甲地西泮(nordazepam, NZP)、替马西泮(temazepam, TZP)、奥沙西泮(oxazepam, OZP)的残留量, 并分析其在组织中的分布特征和代谢规律。结果 摄食含DZP的阳性饵料后, DZP在鲫鱼体内迅速达到较高的残留量, 并在456 h后仍维持在高水平, 主要集中在性腺、肝脏和胆中, 尤其在性腺中残留量显著, 其主要代谢物为NZP和TZP。结论 DZP在鲫鱼体内主要以原型物形态存在, 并且容易在性腺中富集。DZP及其主要代谢物在鲫鱼体内的代谢周期较长, 消费者食用使用含DZP饵料捕获的水产品可能存在食品安全问题和健康风险。

目的 研究鲫鱼摄食含地西泮(diazepam, DZP)的阳性饵料后, DZP在其体内的组织分布及代谢规律。方法 选择鲫鱼作为研究对象, 使用含DZP的阳性饵料进行灌胃给药, 并在给药后1 h至456 h内, 定时对鱼鳞、鱼皮、肌肉、血浆、鳃、肠、肝脏、胆、性腺及脑等组织进行取样。采用高效液相色谱-串联高分辨质谱技术, 检测各组织中DZP及其代谢物去甲地西泮(nordazepam, NZP)、替马西泮(temazepam, TZP)、奥沙西泮(oxazepam, OZP)的残留量, 并分析其在组织中的分布特征和代谢规律。结果 摄食含DZP的阳性饵料后, DZP在鲫鱼体内迅速达到较高的残留量, 并在456 h后仍维持在高水平, 主要集中在性腺、肝脏和胆中, 尤其在性腺中残留量显著, 其主要代谢物为NZP和TZP。结论 DZP在鲫鱼体内主要以原型物形态存在, 并且容易在性腺中富集。DZP及其主要代谢物在鲫鱼体内的代谢周期较长, 消费者食用使用含DZP饵料捕获的水产品可能存在食品安全问题和健康风险。

Objective To study the tissue distribution and metabolic regulation of diazepam (DZP) in Carassius auratus after feeding with positive bait containing DZP. Methods Carassius auratus was selected as the study object and exposed to DZP-enriched bait through gavage, and tissue samples including scales, skin, muscle, plasma, gills, intestines, liver, gallbladder, gonads, and brain were collected at specific intervals ranging from 1 h to 456 h post-exposure. High performance liquid chromatography coupled with tandem high-resolution mass spectrometry was employed to detect DZP and its metabolites—nordazepam (NZP), temazepam (TZP), and oxazepam (OZP)—in these tissues, analyze their distribution and metabolic patterns. Results Following exposure to DZP-enriched bait, DZP rapidly accumulated in Carassius auratus tissues, with substantial residues persisting for up to 456 h. The highest concentrations were primarily found in the gonads, liver and gallbladder, with the gonads consistently showing elevated levels. NZP and TZP emerged as the primary metabolites. Conclusion DZP predominantly exists in its prototype form in Carassius auratus and tends to accumulate in the gonads. The prolonged metabolic cycle of DZP and its metabolites in Carassius auratus indicates potential food safety and health risks for consumers of aquatic products caught with DZP-containing bait.

地西泮(diazepam, DZP)是一种苯二氮卓类镇定剂, 自20世纪60年代起广泛应用于人类和兽医临床中, 主要用于抗焦虑和镇静[1-2]。它也是目前应用最多的助眠类药物之一[3]。然而, 长期暴露于DZP会增加肝脏负担, 可能导致嗜睡、头痛和幻听等副作用[1,4], 并可能在新生儿中引起长期认知障碍[5]。2017年, 世界卫生组织国际癌症研究机构将DZP列为3类致癌物[6]。由于DZP使用量大且难以降解, 它可能长期广泛存在于环境中[7-9]。DZP及其主要代谢物具有较高的药理活性[10], 可能对生态环境构成潜在威胁。此外, DZP在生物体内具有富集作用, 可以通过食物链传递, 进而影响人体健康[11-14]
DZP在我国属于第二类精神药物, 在动物源性食品中不得检出, 目前也未被批准作为水产养殖兽药。然而, 近年来养殖水产品中DZP检出事件在全国各地频有发生, 在《农业农村部关于监督抽查发现的17批次问题农产品(不含豇豆)情况的通告》中有7批次不合格水产品, 其中有6批次样品检出DZP, 涉及大口黑鲈(Micropterus salmoides)、乌鳢(Channa argus)、鲤(Cyprinus carpio)、鳊(Parabramis pekinensis)等品种, 问题样品分布在辽宁、江苏、浙江、山东、贵州等地[15]。重庆市农业农村委员会关于2023年监督抽查发现的48批次问题农产品(不含豇豆)情况的通告中有19批次问题农产品属于水产品, 均为DZP检出[16]。这些报道表明由DZP残留导致的水产品质量安全问题较为突出, 并引起了社会和学者的强烈关注, 由此开发了一系列DZP残留量检测方法[17-20], 也有学者通过模拟养殖环境药浴暴露, 发现DZP在水产品中代谢周期较长, 需要经过较长的休药期来保证水产品质量安全[21]
水产品中DZP残留的污染来源可能是环境污染[22]、养殖或运输过程[23]中违规用药以及休闲垂钓使用DZP污染的垂钓饵料[18,24], 其中垂钓饵料引起的水产品DZP残留问题较为突出。前期的研究发现, DZP呈阳性的养殖水产品大多来自兼具休闲垂钓功能的养殖场[25], 且休闲垂钓饵料中DZP检出率较高[24]。通过休闲垂钓所得水产品多以食用为主, 或经过交易进入消费市场, 而摄食了受污染饵料的水产品体内可能残留较高浓度的DZP, 由此引发食品质量安全和消费者健康问题。因此有必要研究水产品摄食受污染饵料后, DZP在水产品体内的组织分布和消除代谢规律, 评估其对消费者的健康影响。
本研究以休闲垂钓场中常见水产品鲫鱼为研究对象, 用DZP阳性饵料进行灌胃给药, 分析不同时间点DZP及其主要代谢物去甲地西泮(nordazepam, NZP)、替马西泮(temazepam, TZP)、奥沙西泮(oxazepam, OZP)在鲫鱼鱼鳞、鱼皮、肌肉、血浆、鳃、肠、肝脏、胆、性腺、脑等组织的分布特征和代谢规律, 为保障水产品的食用安全和消费者健康风险评估提供数据支持。
甲醇、乙腈、甲酸、乙酸铵(质谱纯)、十八烷基硅烷(C18)分散固相萃取材料(40~60 μm, 60 Å)(上海安谱实验科技股份有限公司); 乙酸乙酯(色谱纯, 美国Baker公司); DZP、DZP-D5、OZP、NZP、TZP标准溶液(均为100 μg/mL, 天津阿尔塔科技有限公司); N-丙基乙二胺(N-propyl ethylenediamine, PSA) (40~63 μm, 60 Å, 天津博纳艾杰尔科技有限公司); 氯化钠(NaCl)(分析纯, 国药集团化学试剂有限公司); 聚四氟乙烯(polytetrafluoroethylene, PTFE)滤膜(0.22 μm, 天津市津腾实验设备有限公司); 实验室用水为Milli Q超纯水(电阻率18.2 mΩ)。
DZP阳性饵料由实验室前期采样筛查得到, DZP含量为75 mg/kg。使用的健康鲫鱼(体重300 g±10 g, 体长13~15 cm)购自上海市江阳水产批发市场, 通过随机抽样确认鲫鱼体内不含DZP及其代谢物。实验前, 鲫鱼被暂养在实验室养殖水箱中, 水温控制在(20±1) ℃, 并持续曝气。
Ultimate 3000超高效液相色谱-Q-Exactive静电场轨道离子阱质谱联用仪(美国赛默飞世尔科技公司); MS1涡旋混匀器(德国IKA公司); CF16RXII高速冷冻离心机(日本Hitachi公司); N-EVAP氮吹仪(美国Organomation公司); BT 423S电子天平(精度0.00001 g, 德国Sartorius公司); RRHD Eclipse Plus 95Å C18 (3.0 mm×150 mm, 1.8 μm, 美国安捷伦公司)。
实验将健康鲫鱼随机分为两组: 对照组和实验组, 每组鱼分别饲养在容量为500 L的养殖水箱中。实验组鲫鱼通过灌胃方式给予DZP阳性饵料, 每尾鱼的剂量为0.8 g, 其中含有60 μg的DZP。给药后, 在1、4、8、12、24、48、96、168、264、360、456 h分别随机选取3尾鱼。从每尾鱼的尾静脉采集1.5~2.0 mL的血浆样本。随后进行解剖, 分别取出鱼鳞、鱼皮、肌肉、鳃、肠、肝脏、胆、性腺和脑等组织样本。所有采集的样本均在-40 ℃下冷冻保存, 以备后续分析。
根据参考文献[20]中的方法, 对鱼鳞、鱼皮、肌肉、鳃、肠、肝脏、胆、性腺和脑等组织以及血浆中的DZP及其代谢物OZP、TZP、NZP进行提取和净化。首先, 准确称取2 g组织样品(血浆取1 mL)放入50 mL具塞离心管中, 加入50 μL DZP-D5内标溶液(质量浓度为100 ng/mL)和5 mL乙酸乙酯。之后进行涡旋振荡混合10 min, 室温静置。接着加入1 g NaCl, 再次涡旋振荡1 min, 然后在4 ℃和4000 r/min的条件下离心10 min。将上清液转移到新的50 mL离心管中, 并重复上述提取过程一次, 将两次提取的上清液合并。准确移取5 mL上清液至10 mL玻璃管中, 在40 ℃下温和的氮气气流中吹干, 随后加入1 mL 80%乙腈-水溶液, 充分涡旋复溶。然后, 分别向复溶液中加入50 mg的C18和50 mg的PSA, 涡旋混合10 min后静置。取上清液, 过0.22 μm PTFE滤膜, 样品即可用于后续分析。
(1)色谱条件
色谱柱: RRHD Eclipse Plus 95Å C18 (3.0 mm×150 mm, 1.8 μm); 流动相A为水(5 mmol/L乙酸铵和0.2%甲酸), B为乙腈; 梯度洗脱程序: 0~1.5 min, 1% B; 1.5~4.0 min, 1%~90% B; 4.0~6.0 min, 90%~95% B; 6.0~8.5 min, 95%~99% B; 8.5~8.6 min, 99%~1% B; 8.6~10 min, 1% B。柱温: 35 ℃; 进样体积: 10 μL; 流速: 0.5 mL/min。
(2)质谱条件
电喷雾离子源(electrospray ionization, ESI), 喷雾电压4000 V; 鞘气15 L/min, 辅气5 L/min, 辅气温度300 ℃; 离子传输管温度350 ℃。采用平行反应监测(parallel reaction monitoring, PRM)正离子模式采集质谱数据, DZP、DZP-D5、OZP、TZP、NZP的定性离子对、定量离子对和碰撞能量见表1
为构建定量分析标准曲线, 使用80%乙腈-水溶液配制目标化合物的混合标准工作液。具体质量浓度设定为0.1、0.2、0.5、1.0、2.0、5.0、10.0、20.0、50.0、100.0和200.0 ng/mL, 同时每个标准溶液中的内标DZP-D5质量浓度固定为5 ng/mL。在液相色谱-质谱仪上对这些标准工作液进行检测, 并将外标与内标的特征峰面积比值作为纵坐标, 标准物质的浓度作为横坐标, 构建标准曲线。
采用Xcalibur 4.1软件和Excel 2016软件对DZP及其代谢物进行定量分析。利用Origin 2018绘制DZP及其代谢物的浓度-时间曲线以及残留物分布图。实验中的结果为3次重复实验的平均值。采用SPSS Statistics 26进行显著性分析。
本研究采用内标法对DZP及其代谢物OZP、TZP、NZP进行定量分析。通过将内标和外标的峰面积比作为纵坐标(Y), 目标化合物的质量浓度作为横坐标(X, ng/mL), 构建了标准曲线。结果显示, 在0.1~200.0 ng/mL的质量浓度范围内, DZP及其代谢物OZP、TZP、NZP的标准曲线线性方程分别为: DZP: Y=0.029309+0.274873X, OZP: Y=-0.00306887+0.0115645X, NZP: Y=0.0202551+0.116911X和TZP: Y=-0.000235352+0.0515858X, 相关系数(r²)均不小于0.999。4种化合物在鲫鱼各种组织中的检出限均为0.1 μg/kg。DZP的回收率为85.7%~108.0%, OZP的回收率为87.2%~104.0%, TZP的回收率为85.3%~102.0%, 而NZP的回收率为86.3%~104.0%。所有化合物的相对标准偏差均在15%以下。结果表明, 本研究所采用的分析方法精确度和准确度均表现良好, 可用于复杂生物样本中的DZP及其代谢物的定量分析。
本研究对经过阳性饵料灌胃暴露的鲫鱼各组织样品中的DZP残留量进行测定。如图1所示, DZP在鲫鱼体内的残留量迅速增加, 在1 h后各主要组织中已达到较高浓度水平, 并随着时间的推移呈现不同的代谢趋势, 且由于肝肠循环、多位吸收等因素的影响, DZP在鲫鱼组织中的含量呈现双峰或多峰的波动现象[26-27]。其中鱼鳞、血液、肌肉、鳃、鱼皮中DZP的残留量在1 h达到最高值, 分别为36.606、45.033、85.878、87.125和118.977 μg/kg。随后, 这些组织中的DZP残留量开始逐渐下降, 并在168 h后趋于稳定。相反, 性腺、脑、肠、肝脏和胆等组织的DZP残留量则呈现出先升后降的趋势。特别是肠和肝脏的DZP残留量, 在12 h后达到峰值, 分别为236.754 μg/kg和112.162 μg/kg, 此后至48 h迅速下降, 随后肠中的DZP下降速度放缓, 而肝脏中的DZP含量则趋于稳定。胆中的DZP残留量在12 h达到180.775 μg/kg的峰值, 然后快速下降, 但在264 h再次达到190.857 μg/kg的另一个峰值, 随后缓慢下降, 表现出明显的双峰现象。在脑组织中, DZP残留量在1 h内迅速达到高点, 24 h内略有下降, 但之后逐渐上升, 并在168 h达到140.983 μg/kg的峰值。性腺中的DZP残留量则表现出持续上升的趋势, 至360 h达到263.415 μg/kg的最高值, 且在456 h仍维持在较高水平。
图2所示, 经过456 h的代谢, DZP在鲫鱼各组织中的残留量由低至高依次为: 鳞(9.624 μg/kg)、肌肉(13.613 μg/kg)、血液(20.178 μg/kg)、鳃(21.688 μg/kg)、肠(31.496 μg/kg)、鱼皮(38.46 μg/kg)、脑(43.491 μg/kg)、肝脏(64.304 μg/kg)、胆(153.133 μg/kg)和性腺(225.678 μg/kg)。与暴露最大残留量相比, 所有组织中的DZP含量均显著下降(P<0.05)。特别地, 肠中的DZP残留量降低程度最大。这可能是因为灌胃暴露后, DZP被消化系统迅速吸收并通过血液循环输送至各组织, 导致早期即出现高浓度残留。肌肉、鱼鳞、鳃、血液和皮肤在暴露1 h后达到最大残留量, 而到了456 h, 这些组织中的DZP残留量相比峰值有较高的消除率。在脑组织中, 虽然DZP含量下降较多, 但在该组织中有一个短暂的富集过程(24~168 h), 这一现象和文献[21]中的结果一致, 而脑组织中高浓度DZP的存在可能会对神经系统产生一定的影响。在肝脏、胆和性腺中, DZP的代谢程度较低, 在456 h仍有较高的残留量, 尤其是在性腺中, DZP含量随着时间的增加而持续升高, 残留量远高于鲫鱼其他组织(P<0.05), 这表明DZP在肝脏、胆和性腺中容易富集, 且性腺可能是DZP在鲫鱼体内的靶器官, 对鲫鱼具有潜在的生殖和发育功能影响。随着代谢时间的增加, 虽然DZP在鲫鱼各组织中的含量有不同程度的减少, 但各组织中最终残留量依然较高, 表明其在鲫鱼体内代谢较慢, 代谢周期较长。
DZP在人体内主要通过细胞色素P450 CYP2C19和CYP3A酶的作用去甲基生成其代谢物NZP, 并在CYP3A酶的介导下生成羟基化产物TZP。NZP和TZP进一步代谢生成OZP, 最终以葡萄糖醛酸结合物的形式通过尿液排出[28-29]。这些代谢物保持与DZP相似的药理活性[30-31]。因此, 本研究考察DZP在鲫鱼各组织中代谢规律的同时, 也考察了其代谢物NZP、TMZ和OZP在鲫鱼各组织中的残留及消除规律。
鲫鱼经DZP阳性饵料灌胃暴露后, DZP在各组织中的代谢物为NZP和TZP, 而OZP在整个代谢实验中未检出, 且NZP和TZP的残留量较低, ALVAREZ-MORA等[32]在DZP暴露的鳗鱼中也仅筛查出了NZP, 这表明DZP在鲫鱼体内主要以原型物的形式存在, 仅有少部分DZP代谢为去甲基代谢产物NZP和羟基化产物TZP。
图3所示, NZP在暴露1 h后在肝脏中检出, 4 h后在胆中检出。8 h后, 在除鱼鳞外的所有组织中均可检出NZP, 并随着时间增长, NZP在各组织中的残留量逐渐上升, 显示出明显的双峰或多峰现象。在168 h时, 肝脏、性腺、胆、脑、肠、鱼皮、鳃和血液中NZP残留量达到峰值, 之后逐渐下降, 并在360 h后再次上升。而肌肉和鱼鳞的NZP残留量则持续上升, 至456 h达到峰值。肝脏中NZP的峰值最高(13.022 μg/kg), 明显高于其他组织(P<0.05), 且经过456 h的代谢, 肝脏中NZP残留量最大(6.117 μg/kg), NZP在其他组织中的残留量由大到小分依次为脑、性腺、肠、鱼皮、血液、肌肉、鳃和鱼鳞。这些结果表明, 肝脏是DZP在鲫鱼体内发生去甲基代谢反应的主要器官, 其代谢产物NZP在鲫鱼各组织中长期存在, 代谢周期较长[33], 具有一定的富集作用。
图4所示, 在代谢初期, 肠中的TZP残留量迅速增加, 于12 h达到峰值, 明显高于其他组织, 但之后迅速下降。相比之下, 其他组织中的TZP残留量虽然也呈现出快速上升后下降的趋势, 但峰值普遍较低。12 h后, 仅性腺的TZP含量超过1 µg/kg, 而其他组织的含量均低于0.6 µg/kg。TZP在肠组织中的含量变化趋势与DZP在肠道中的变化类似, 在12 h时DZP含量也远高于其他组织, 随着DZP含量的下降, TZP的含量也快速下降, 并且远低于峰值。这可能是因为CYP3A酶是肠道中最丰富的酶, DZP在体内的羟基化代谢反应主要发生在肠道中, 羟基化产物TZP被肠道吸收后扩散到其他组织中。在其他组织中, TZP的残留量随着代谢时间的增加呈现多峰或双峰现象, 但相较于DZP和NZP, TZP在鲫鱼组织中的含量整体处于较低水平, 经过456 h的代谢, TZP仅在肝脏、胆和性腺中有检出, 这也说明鲫鱼体内, DZP的羟基化代谢反应相对有限, TZP并非DZP的主要代谢产物。
高浓度的DZP对鱼类可能具有一定的诱食作用, 因此不良商家为提高经济效益非法添加DZP致使饵料中DZP含量极高。鱼类摄食这些饵料后, DZP会在体内快速吸收和分布, 尤其是在肌肉、鱼皮、脑和血液等可食部分, 可在短时间内达到较高浓度。如果消费者食用了这些鱼类, 高浓度的DZP及其代谢物可能对人体健康造成严重威胁。这不仅引起即时的食品质量安全问题, 还可能导致长期的健康风险。此外, 由于DZP及其代谢物在鱼体内的残留周期较长, 即便鱼类经过一段时间的暂养, 其体内的化合物依然可能对消费者造成影响。此外, 垂钓活动中大量使用含DZP的饵料可能导致该化合物在水环境中扩散, 进而对生态系统和水生生物造成潜在的威胁。随着时间的推移, DZP可能通过食物链累积并最终影响人类健康。
本研究通过对鲫鱼进行DZP阳性饵料灌胃暴露实验, 分析了DZP及其代谢物(NZP、TZP、OZP)在鲫鱼各组织中的分布和代谢规律。结果显示, 鲫鱼摄食阳性饵料后, DZP在体内被迅速吸收, 短时间内在各组织中达到较高的残留量, 且具有较长的代谢周期。经过456 h的代谢, 各组织中的DZP残留量仍然较高, 尤其是在性腺、胆、肝脏和脑中。DZP在性腺中表现出显著的富集现象, 提示其对鲫鱼的生殖和发育功能可能具有潜在影响。在鲫鱼体内, DZP主要以原型物存在, 仅检出了去甲基代谢产物NZP和少量的羟基化代谢产物TZP, 而未检出OZP。这些代谢物在鲫鱼体内的残留时间较长, 其中NZP在鲫鱼体内也表现出富集现象; 研究表明, 使用含DZP的阳性饵料会导致DZP在鲫鱼体内长期残留, 可能对消费者的健康构成威胁, 引发食品安全和公共健康问题。需要对使用此类饵料的垂钓活动进行严格监控, 以减少DZP对环境和公众健康的潜在风险。
