Objective To investigate the differences in amino acid content and nutritional value of Elaeagnus angustifolia from different regions in Xinjiang. Methods The amino acid content in the pulp of 18 Elaeagnus angustifolia samples from 9 different counties in Xinjiang were evaluated by automatic amino acid analyzer at wavelengths of 570 nm and 440 nm by post-column derivative ion exchange chromatography, and by comparing with Food and Agriculture Organization of the United Nations/World Health Organization (FAO/WHO) amino acid pattern spectrum and ideal protein standard, the amino acid ratio (RAA), ratio coefficient of amino acid (RC) and score of ratio coefficient (SRC) were calculated. The nutritional value of amino acids in the pulp of Elaeagnus angustifolia was evaluated. Results There were 17 kinds of amino acids in the pulp of 18 Elaeagnus angustifolia samples. The essential amino acids accounted for 13.87%-21.96%, and the pharmacodynamic amino acids accounted for 73.03%-80.21%. The amino acid composition of Elaeagnus angustifolia were quite different among regions, and the content of total amino acids, essential amino acids and pharmacodynamic amino acids were all the highest in Qira Country, with values of 3.69, 2.73, and 0.81 g/100 g, respectively. According to the comparison of FAO/WHO amino acid pattern spectrum and whole egg reference standard, Elaeagnus angustifolia in Yen gisar County had the highest SRC of 66.92% and 72.27%, respectively. Conclusion As a plant-based food source, Elaeagnus angustifolia pulp is rich in amino acids and has high medicinal value. The amino acid content of them sold in different regions of Xinjiang is different, so the subsequent study can further explore the breeding and processing of the dominant varieties of Elaeagnus angustifolia.

目的 研究新疆不同地区大果沙枣果肉中氨基酸含量差异及营养价值。方法 采用全自动氨基酸分析仪, 在波长分别为570 nm和440 nm处采用茚三酮柱后衍生离子交换色谱法对来自新疆9个不同县市、18个大果沙枣果肉中氨基酸含量进行测定, 并通过与联合国粮农组织/世界卫生组织(Food and Agriculture Organization of the United Nations/World Health Organization, FAO/WHO)氨基酸模式谱和理想蛋白质(全鸡蛋模式)标准比较, 计算氨基酸比值(amino acid ratio, RAA)、氨基酸比值系数(ratio coefficient of amino acid, RC)、氨基酸比值系数分(score of ratio coefficient, SRC), 对沙枣果肉中的氨基酸进行了营养价值评价。结果 18份大果沙枣样品果肉中均含有17种氨基酸, 必需氨基酸占比13.87%~21.96%, 药用氨基酸占比73.03%~80.21%。不同区域大果沙枣氨基酸组成差异较大, 但策勒县沙枣中氨基酸总量、药用氨基酸总量及必需氨基酸含量均为最高, 其值分别为3.69、2.73和0.81 g/100g。通过与FAO/WHO氨基酸模式谱和全鸡蛋模式标准比较, 英吉沙县沙枣的SRC值最高, 分别为66.92%和72.27%。结论 作为植物基食物来源, 沙枣的氨基酸具有较高的药用价值; 新疆不同地区沙枣的氨基酸含量存在差异, 后续研究可更进一步探索沙枣优势品种的选育和加工。

目的 研究新疆不同地区大果沙枣果肉中氨基酸含量差异及营养价值。方法 采用全自动氨基酸分析仪, 在波长分别为570 nm和440 nm处采用茚三酮柱后衍生离子交换色谱法对来自新疆9个不同县市、18个大果沙枣果肉中氨基酸含量进行测定, 并通过与联合国粮农组织/世界卫生组织(Food and Agriculture Organization of the United Nations/World Health Organization, FAO/WHO)氨基酸模式谱和理想蛋白质(全鸡蛋模式)标准比较, 计算氨基酸比值(amino acid ratio, RAA)、氨基酸比值系数(ratio coefficient of amino acid, RC)、氨基酸比值系数分(score of ratio coefficient, SRC), 对沙枣果肉中的氨基酸进行了营养价值评价。结果 18份大果沙枣样品果肉中均含有17种氨基酸, 必需氨基酸占比13.87%~21.96%, 药用氨基酸占比73.03%~80.21%。不同区域大果沙枣氨基酸组成差异较大, 但策勒县沙枣中氨基酸总量、药用氨基酸总量及必需氨基酸含量均为最高, 其值分别为3.69、2.73和0.81 g/100g。通过与FAO/WHO氨基酸模式谱和全鸡蛋模式标准比较, 英吉沙县沙枣的SRC值最高, 分别为66.92%和72.27%。结论 作为植物基食物来源, 沙枣的氨基酸具有较高的药用价值; 新疆不同地区沙枣的氨基酸含量存在差异, 后续研究可更进一步探索沙枣优势品种的选育和加工。

Objective To investigate the differences in amino acid content and nutritional value of Elaeagnus angustifolia from different regions in Xinjiang. Methods The amino acid content in the pulp of 18 Elaeagnus angustifolia samples from 9 different counties in Xinjiang were evaluated by automatic amino acid analyzer at wavelengths of 570 nm and 440 nm by post-column derivative ion exchange chromatography, and by comparing with Food and Agriculture Organization of the United Nations/World Health Organization (FAO/WHO) amino acid pattern spectrum and ideal protein standard, the amino acid ratio (RAA), ratio coefficient of amino acid (RC) and score of ratio coefficient (SRC) were calculated. The nutritional value of amino acids in the pulp of Elaeagnus angustifolia was evaluated. Results There were 17 kinds of amino acids in the pulp of 18 Elaeagnus angustifolia samples. The essential amino acids accounted for 13.87%-21.96%, and the pharmacodynamic amino acids accounted for 73.03%-80.21%. The amino acid composition of Elaeagnus angustifolia were quite different among regions, and the content of total amino acids, essential amino acids and pharmacodynamic amino acids were all the highest in Qira Country, with values of 3.69, 2.73, and 0.81 g/100 g, respectively. According to the comparison of FAO/WHO amino acid pattern spectrum and whole egg reference standard, Elaeagnus angustifolia in Yen gisar County had the highest SRC of 66.92% and 72.27%, respectively. Conclusion As a plant-based food source, Elaeagnus angustifolia pulp is rich in amino acids and has high medicinal value. The amino acid content of them sold in different regions of Xinjiang is different, so the subsequent study can further explore the breeding and processing of the dominant varieties of Elaeagnus angustifolia.

沙枣(Elaeagnus angustifolia), 胡颓子属植物。在新疆, 沙枣因耐寒、耐旱、耐盐碱、防风固沙、固氮和快速生长等特点[1], 被广泛种植于塔里木盆地[2]、吐鲁番盆地[3]、哈密地区[4]、克孜勒苏柯尔克孜自治州[5]、阿克苏地区[6]、喀什以及和田等地区, 主要用作防风固沙的防护林。沙枣可以分为尖果沙枣和大果沙枣[4]。与尖果沙枣相比, 大果沙枣具有果实大, 可食率高和口感好的特点。大果沙枣直径在1.5~2.0 cm之间, 可食率超过70%[7], 新疆林科院选育的新品种“红铃”大果沙枣可食率高达81.80%[8]
大果沙枣中具有丰富的营养物质, 包括糖类、脂肪、蛋白质、氨基酸、矿物质等。以往的研究表明, 不同产地及品种的沙枣营养成分差异较大, 但在果肉中的主要营养物质仍为蛋白质和碳水化合物[9]。新疆大果沙枣果肉中还含有多种生物活性成分, 是西北地区极具加工和利用前景的重要资源[10-12]。王天等[13]综述了沙枣果实丰富的生物活性物质具有增加机体免疫能力、降糖、降低胆固醇、抗炎、抗菌、抗氧化、抗疲劳、抗腹泻等多种药理作用, 目前被加工制作成饮料、果酱和饼干等多种产品。氨基酸作为构成生物体蛋白的基本物质, 是蛋白质的分解产物, 也是评价食品营养价值的重要指标, 食物的营养价值能够从氨基酸组成及营养评价反映出来[14], 它不仅可以作为复方氨基酸输液, 可以作为钾、钙等无机离子的补充剂, 还可以作为疲劳恢复剂, 氨解毒剂, 肝机能促进剂等[15]。氨基酸类物质具有抗肿瘤性[16], 已经成为人们关注的热点。
目前对于大果沙枣的研究重点多为宏观营养素、活性成分含量及提取方面, 对其氨基酸营养价值评价的研究鲜有报道[17-19]。本研究分别收集新疆9个地区18个大果沙枣样本, 对果肉中氨基酸差异及营养价值进行评价; 采用全自动氨基酸分析仪, 结合联合国粮农组织/世界卫生组织(Food and Agriculture Organization of the United Nations/World Health Organization, FAO/WHO)氨基酸模式谱和理想蛋白质(全鸡蛋模式)标准开展了相关试验与数据比对, 探明新疆不同区域大果沙枣果肉中氨基酸差异及营养价值, 探讨不同地区沙枣果肉中的氨基酸组成及含量差异, 旨在为大果沙枣的营养成分分析和开发利用提供科学依据, 并为提升新疆大果沙枣的市场价值和推广应用提供理论支持。
材料详见表1, 新疆不同区域的样本共计18份, 取样量均为5 kg/份, 样本于2022年12月收集, 均在完熟期采收, 于自然干燥环境中保存对样本随机进行抽取。其中, 新疆喀什地区8份(编号为SZ-01~SZ-04、SZ-06、SZ-14~ SZ-16); 新疆克孜勒苏柯尔克孜自治州2份(编号为SZ-08、SZ-09); 新疆和田地区5份(编号为SZ-07、SZ-12、SZ-13、SZ-17、SZ-18); 新疆阿克苏地区3份(编号为SZ-05、SZ-10、SZ-11)。
BS124S电子天平(精度0.1 mg, 德国赛多利斯公司); FOSS 8400全自动凯氏定氮仪(丹麦福斯FOSS集团公司); S-433D氨基酸分析仪(德国SYLAM赛卡姆公司); R-12平行蒸发定量浓缩仪(瑞士Syncore BUCHI公司)。
甲醇(色谱纯, 美国赛默飞世尔科技公司); 盐酸(优级纯)、硫酸(分析纯)(成都市科隆化学品有限公司); 柠檬酸三钠(二水)(优级纯, 天津市鑫铂特化工有限公司); 茚三酮(色谱纯)、LCA K06/Na色谱柱(4.6 mm×150 mm)、混合氨基酸标准溶液(浓度2.5 µmoL/mL)[赛卡姆(北京)科学仪器有限公司]。
每份沙枣样品随机选取500 g, 去除果核, 果肉粉碎, 采用高微粉碎机制备成粉状, 备用。
分别称取制备好的沙枣粉末0.2 g(精确至0.1 mg)于水解管中, 在水解管中分别加入10 mL 6 mol/L盐酸溶液。使用酒精喷灯将水解管管口进行拉管操作, 使其便于抽真空。冷却后将水解管接到真空泵的抽气管上, 抽真空, 使用酒精灯对水解管进行封口。将已封口的水解管放在110 ℃±1 ℃的恒温干燥箱中水解22 h后取出, 冷却至室温, 将水解液用漏斗放置中性滤纸干过滤至50 mL容量瓶中, 用少量水多次冲洗水解管, 水洗液合并转移入50 mL容量瓶中, 定容至刻度, 振荡混匀, 备用。
依据GB 5009.124—2016《食品安全国家标准 食品中氨基酸的测定》。采用全自动氨基酸分析仪测定沙枣果肉的17种氨基酸含量。
准确吸取1.0 mL 1.3.2中制备滤液移入25 mL浓缩试管中, 将试管置于平行蒸发仪上, 在40~45 ℃加热环境下减压干燥至蒸干。吸取1.0 mL pH 2.2柠檬酸钠缓冲溶液(浓度为0.2 moL/L)于干燥后试管内溶解, 涡旋振荡混匀后, 通过0.22 µm滤膜, 转移至进样瓶中, 为样品待测液, 供仪器测定用。氨基酸分析测定条件如表2所示。
使用混合氨基酸标准工作液注入氨基酸自动分析仪。色谱参考条件色谱柱为磺酸型阳离子树脂, 检测波长为570 nm(除脯氨酸以外的氨基酸)和440 nm(用于测定脯氨酸)。混合氨基酸标准工作液和样品测定液分别以30 µL体积注入氨基酸分析仪, 以外标法通过峰面积计算样品测定液中氨基酸的浓度。
根据氨基酸含量测定数据换算沙枣样品中必需氨基酸的相关比例, 参考FAO/WHO 1973年建议的氨基酸评分标准模式和中国预防医学科学营养与食品卫生研究所提出的鸡蛋模式, 并根据氨基酸平衡理论, 利用标准模式谱计算必需氨基酸的氨基酸比值(amino acid ratio, RAA)、氨基酸比值系数(ratio coefficient of amino acid, RC)、变异系数(coefficient of variation, CV)、氨基酸比值系数分(score of ratio coefficient, SRC)值[20-21], 相关计算如公式(1)~(4):
RAA=样品中必需氨基酸含量/模式谱中相应必需氨基酸含量
RC=RAA/RAA的平均值
CV=RC的标准差/RC的均值
SRC=100-CV×100
采用SPSS Statistics 25进行数据分析, 采用Kruskal- Wallis检验显著性差异, 当P<0.05时, 表明具有显著性差异。采用Excel 2021软件制表绘图。
采用GB 5009.5—2016《食品安全国家标准 食品中蛋白质的测定》全自动凯氏定氮法对不同区域18个沙枣果肉样本中蛋白质进行检测分析, 结果见表3。来自和田地区策勒县的沙枣蛋白质含量最低(4.67±0.55) g/100 g, 麦盖提县沙枣的蛋白质含量最高(5.68±0.06) g/100 g, 但各地区间沙枣蛋白含量均无显著性差异(P>0.05)。江发寿等[22]研究发现新疆沙枣果实可食部分蛋白质含量为7.03%, 与本研究的结果较为接近。
利用氨基酸分析仪对9个地区沙枣中的氨基酸含量进行测定, 结果见表4。结果显示, 每个产地沙枣中均检出17种氨基酸, 包括7种EAA, 2种CE和8种NEAA。以往的研究结果表明, 沙枣果肉至少含有18种氨基酸, 其总含量可达3.40%, 必需氨基酸含量达1.12%[23]。而在本研究中, TAA在2.63~3.69 g/100 g之间。9个产地沙枣中氨基酸总量由高到低排列顺序为: 策勒县>麦盖提县>英吉沙县>皮山县>喀什市>阿瓦提县>伽师县>阿图什市>莎车县。此外, 苏氨酸、丙氨酸、胱氨酸、谷氨酸、蛋氨酸和异亮氨酸含量在各地区沙枣间无显著性差异(P>0.05)。除脯氨酸、天门冬氨酸及胱氨酸外, 策勒县沙枣的氨基酸含量均为最高, 天门冬氨酸在英吉沙县沙枣中含量最高, 为1.69 g/100 g, 而脯氨酸则在麦盖提县沙枣中含量最高, 为0.42 g/100 g。有研究表明, 谷氨酸、天冬氨酸、胱氨酸等氨基酸可单独用以治疗某些疾病, 如肝脏疾病、消化道疾病、心脑血管疾病、呼吸道疾病以及用于提高肌肉活力、儿科营养和解毒等[24]
EAA是指人体不能合成或合成速度远不能适应机体需要, 必须由食物蛋白质供给的氨基酸[25]。本研究中, 9个产地的EAA在0.41~0.81 g/100 g之间, 其中, 策勒县的EAA总量最高, 莎车县和阿图什市的EAA总量最低。EAA含量由高到低排列顺序为: 策勒县>皮山县>麦盖提县>伽师县>英吉沙县>阿瓦提县>喀什市>阿图什市=莎车县。蛋氨酸含量在所有地区沙枣中含量均最低, 天门冬氨酸含量在所有地区沙枣中较高。对于缬氨酸和赖氨酸含量, 除麦盖提县, 策勒县均显著高于其他各县, 莎车县赖氨酸显著低于英吉沙县和皮山县(P<0.05)。对于亮氨酸含量, 除阿图什市外, 策勒县显著高于其他各县(P<0.05)。对于苯丙氨酸, 策勒县含量显著高于其他各县(P<0.05)。综上所述, 策勒县的沙枣EAA含量最高, 为(0.81±0.07) g/100 g, 营养价值高。
CE是指儿童成长所需的具有促进生长激素分泌与骨骼发育等重要生理功能的氨基酸, 包括组氨酸和精氨酸[26]。9个地区沙枣的CE总量在0.35~0.59 g/100 g之间, 占TAA的比例均超过10%, 其中, 策勒县产沙枣中的CE含量最高为(0.59±0.05) g/100 g, 但伽师县产沙枣中的CE占总氨基酸比例最高, 达17.93%, 策勒县次之, 麦盖提县占比最低, 为10.65%。
9个产地沙枣中EAA/TAA在13.87%~21.96%之间, 其中, 策勒县沙枣中EAA/TAA最高, 为21.96%±0.61%, 皮山县和伽师县分居第二和第三, 英吉沙县沙枣中EAA/TAA最低。因此, 本研究中策勒县产沙枣是较为优质蛋白质的来源。
MEA包括天冬氨酸、谷氨酸、甘氨酸、蛋氨酸、亮氨酸、酪氨酸、苯丙氨酸、赖氨酸、精氨酸等9种氨基酸, 是维持机体平衡的重要氨基酸, 因此也成为评价食品和中药材品质的重要指标[27-28]。由表4可知, 不同产地间药用氨基酸含量在2.09~2.73 g/100 g之间, 9个产地中药用氨基酸总量从高到低排序为: 策勒县>英吉沙县>麦盖提县>皮山县>喀什市>阿瓦提县>阿图什市>伽师县>莎车县。天冬氨酸和谷氨酸在神经系统生长、发育方面起重要作用, 主要用于改善儿童智力发育, 以及治疗神经损伤、脑震荡, 预防高血压、心脏病等, 是重要的功能性氨基酸[29]。沙枣中天冬氨酸含量在所有地区均最高, 占比在41%~65%之间, 是沙枣中药用氨基酸的主要成分。沙枣中药用氨基酸含量占比次之的为精氨酸, 为11%~19%。精氨酸是儿童必需氨基酸, 能够调节体内激素分泌, 在儿童的成长发育中具有重要的生理功能[30]。赖氨酸通常在植物中含量较少, 能够提高智力、促进生长和钙吸收[31], 但沙枣中赖氨酸占比较低, 在3%~5%之间。而蛋氨酸含量在所有地区均为最低。谷氨酸的含量在不同地区间没有显著性差异(P>0.05)。
氨基酸是构成蛋白质的基本单位, 必需氨基酸的组成和含量与氨基酸的营养价值息息相关。由表5可知, 不同地区沙枣种氨基酸模式有较大差异。除了苯丙氨酸+酪氨酸含量所占比例高于或接近于模式谱外, 其他氨基酸百分含量均低于模式蛋白。
RC大于或小于1.00%, 说明该种氨基酸相对过剩或相对不足, RC等于1.00%表明其组成比例与模式谱一致。RC最小值对应的氨基酸为第1限制氨基酸, 以此类推[32]。从表6中可知, 不同产地沙枣中相对过剩和不足的氨基酸不尽相同。总体来说, 苯丙氨酸+酪氨酸在除英吉沙县以外地区的沙枣中均相对过剩, 蛋氨酸+胱氨酸在所有地区的沙枣中均相对不足且为第1限制氨基酸。苏氨酸为所有地区沙枣的第2限制氨基酸。此外, 缬氨酸在伽师县、英吉沙县、喀什市、麦盖提县沙枣中的组成比例与FAO/WHO模式(表6)一致; 赖氨酸在英吉沙县和麦盖提县沙枣中的组成比例与标准模式一致。相比FAO/WHO模式的氨基酸组成评价, 全鸡蛋模式(见表7)评价下的结果表明, 亮氨酸在伽师县、英吉沙县阿瓦提县沙枣中组成比例较好。其他两者间的结果基本一致。
SRC值越大, 蛋白质营养价值越高[33]。由图1可知, 不同地区沙枣样品的SRC值由大到小排序为: 英吉沙县>麦盖提县>伽师县>皮山县>阿瓦提县>喀什市>莎车县>阿图什市>策勒县。
本研究对新疆9个不同产地18份沙枣样品中的蛋白质及氨基酸含量进行了测定。结果表明, 不同产地间的蛋白质含量并无显著性差异。TAA在2.63~3.69 g/100 g之间, EAA含量在0.41~0.81 g/100 g之间, CE含量在0.35~0.59 g/100 g之间, MEA含量在2.09~2.73 g/100 g之间。蛋氨酸是所有地区沙枣中的第一限制性氨基酸。通过分别与FAO/WHO和全鸡蛋模式蛋白比较, 总体来说, 沙枣中的各种氨基酸含量大多相对不足。在9个产地中, 策勒县沙枣的TAA、EAA、MEA含量均排第一, CE含量位居第二。因此, 策勒县产沙枣在氨基酸含量上相对其他地区表现更好。新疆和田、喀什、阿克苏、阿图什等地区的大果沙枣品种繁多, 但尚未有统一的分类和品种命名, 多以外形统称大果沙枣。本研究分析评价了大果沙枣的蛋白质和氨基酸营养价值, 为进一步研究沙枣药理机制及产品开发提供数据支撑。随着大食物观的提出, 沙枣果实开发利用的进一步研究正应其时。新疆和田、喀什、阿克苏、阿图什等地是市场销售大果沙枣区主要和优势产区。由于大部分处于野生和半野生状态, 虽然大沙枣品种较多, 但尚未有统一的分类和品种命名, 多以外形统称大沙枣。本研究针对环塔里木盆地南疆四地州仅对9个县市的18个大果沙枣进行了氨基酸和蛋白质含量及营养价值进行分析评价, 未来应加强品种资源收集和保存、进行科学系统的品种选优和分类, 进一步研究大果沙枣果肉中的营养成分, 促进新疆沙枣产业的多元化发展。
