Objective To establish the detection of 172 kinds of pesticide residues in brown sugar by QuEChERS-gas chromatography-tandem mass spectrometry (GC-MS/MS). Methods The samples were fully dissolved in ultrapure water and extracted by high-speed homogenization of n-hexane, nitrogen blowing and concentration, and purified by QuEChERS method, and the data was collected in selective reaction monitoring mode by GC-MS/MS detection technology, and quantitatively detected by matrix curve and solvent curve. Results The 172 kinds of pesticides had a good linear relationship in the linear range of 10-200 ng/mL, and the linear correlation coefficient was between 0.9962 and 0.9999, the limit of quantification of the method was 0.01 mg/kg, performed 6 repeated spiked experiments at concentrations 1, 2, and 10 times the limit of quantification each, the recovery rate of spikes was between 71.0%-113.0%, and the relative standard deviation (n=6) was 0.49%-17.00%. Conclusion This method can detect hundreds of pesticides in one injection, which is fast, simple, high-throughput and economical, and is suitable for the detection of a variety of pesticide residues in brown sugar.

目的 建立QuEChERS-气相色谱-串联质谱法(gas chromatography-tandem mass spectrometry, GC-MS/MS)检测红糖中172种农药残留的方法。方法 样品加入超纯水充分溶解后经正己烷高速匀浆提取, 氮吹浓缩, QuEChERS法净化, 采用GC-MS/MS检测技术, 以分段时间选择反应监测模式(selective reaction monitoring, SRM)模式进行数据采集, 配制基质匹配曲线及溶剂曲线进行定量检测。结果 172种农药在10~200 ng/mL质量浓度的线性范围内线性关系良好, 线性相关系数在0.9962~0.9999之间, 定量限为0.01 mg/kg, 在1、2、10倍定量限的浓度下各进行6次重复的加标实验, 平均加标回收率在71.0%~113.0%之间, 相对标准偏差为0.49%~17.00% (n=6)。结论 该方法利用QuEChERS法净化, 一次进样可以检测上百种农药, 具有快速、简便、高通量、经济等特点, 适用于红糖中多种农药残留的检测。

目的 建立QuEChERS-气相色谱-串联质谱法(gas chromatography-tandem mass spectrometry, GC-MS/MS)检测红糖中172种农药残留的方法。方法 样品加入超纯水充分溶解后经正己烷高速匀浆提取, 氮吹浓缩, QuEChERS法净化, 采用GC-MS/MS检测技术, 以分段时间选择反应监测模式(selective reaction monitoring, SRM)模式进行数据采集, 配制基质匹配曲线及溶剂曲线进行定量检测。结果 172种农药在10~200 ng/mL质量浓度的线性范围内线性关系良好, 线性相关系数在0.9962~0.9999之间, 定量限为0.01 mg/kg, 在1、2、10倍定量限的浓度下各进行6次重复的加标实验, 平均加标回收率在71.0%~113.0%之间, 相对标准偏差为0.49%~17.00% (n=6)。结论 该方法利用QuEChERS法净化, 一次进样可以检测上百种农药, 具有快速、简便、高通量、经济等特点, 适用于红糖中多种农药残留的检测。

Objective To establish the detection of 172 kinds of pesticide residues in brown sugar by QuEChERS-gas chromatography-tandem mass spectrometry (GC-MS/MS). Methods The samples were fully dissolved in ultrapure water and extracted by high-speed homogenization of n-hexane, nitrogen blowing and concentration, and purified by QuEChERS method, and the data was collected in selective reaction monitoring mode by GC-MS/MS detection technology, and quantitatively detected by matrix curve and solvent curve. Results The 172 kinds of pesticides had a good linear relationship in the linear range of 10-200 ng/mL, and the linear correlation coefficient was between 0.9962 and 0.9999, the limit of quantification of the method was 0.01 mg/kg, performed 6 repeated spiked experiments at concentrations 1, 2, and 10 times the limit of quantification each, the recovery rate of spikes was between 71.0%-113.0%, and the relative standard deviation (n=6) was 0.49%-17.00%. Conclusion This method can detect hundreds of pesticides in one injection, which is fast, simple, high-throughput and economical, and is suitable for the detection of a variety of pesticide residues in brown sugar.

甘蔗是我国重要的糖料作物, 蔗糖产量占食糖总产量的90%以上。广西则是我国最大的甘蔗生产基地, 蔗糖产业作为广西经济发展的重要支柱产业, 也是广西特色优势产业[1-2]。目前, 甘蔗的深加工产品主要有红糖、白糖、冰糖等制品。红糖产品在中国不仅作甜味剂, 而且从古到今都被认为是保健品。在注重养生的当代社会, 红糖产品更是颇受欢迎, 所以红糖类产品的食品安全性也很受关注。在甘蔗的种植过程中, 为控制杂草与病虫害、提高甘蔗产量, 农药产品被广泛应用。然而农药的高频次使用, 或在喷洒过程中没有关注到安全间隔期、安全用量等, 则会导致甘蔗中存在农药污染风险, 因此甘蔗及其深加工产品的农药残留问题非常值得被本研究重视[3-5]。目前文献中针对甘蔗中上百种农药残留检测的高通量方法研究较少[6-7], 更是缺乏对甘蔗深加工产品的农药残留检测方法的研究。因此, 建立红糖中农药残留的高通量快速检测方法, 对于保证红糖产品的质量安全具有重要作用。
目前农药残留检测方法主要有高效液相色谱法[8-10]、气相色谱法[11-13]、气相色谱-串联质谱法[14-17]、液相色谱-串联质谱法[18-21]、酶联免疫吸附分析法[22-24]、胶体金快速检测试纸条[25]等。 酶联免疫吸附分析法、胶体金快速检测试纸条适用于农残检测的定性分析; 气相色谱、液相色谱没有质谱的鉴别力, 一般适用于样品基质干净, 基质干扰相对小的样品, 容易有干扰峰影响, 影响物质精准判定; 液相色谱-串联质谱法适用于非挥发性或热不稳定的农药残留检测, 具有很高的灵敏度和很宽的检测范围, 但仪器费用高同时不适合含糖量高的基质样品检测。现存基于气相色谱-质谱法的植物源性食品中农药残留检测的国标GB 23200.113—2018《食品安全国家标准 植物源性食品中208种农药及其代谢物残留量的测定 气相色谱-质谱联用法》, 只针对水果蔬菜等植物源性食品中的农药残留进行检测, 对于糖类产品并没有涉及。红糖类产品基质复杂且具有特殊性, 本研究针对红糖产品, 选择最合适的提取剂, 并对净化填料用量进行实验, 优化了仪器条件, 同时对基质效应(matrix effects, ME)进行探讨, 建立了气相色谱-串联质谱法(gas chromatography- tandem mass spectrometry, GC-MS/MS)结合QuEChERS法测定红糖的高通量检测方法, 一次进样即可完成对红糖类产品172种农药残留项目的检测。该方法具有前处理简单、检测周期短、成本低、高效稳定和安全的特点, 可为蔗糖产业的生产及贸易互通提供切实可靠的检测技术手段。
本研究所用样品来自华润blt超市、人人乐超市、华润万家超市、沃尔玛超市货架上售卖的各品牌红糖。
208种农药及其代谢产物混合标准品(GB 23200.113—2018, 上海安谱璀世公司); 乙酸乙酯、正己烷、二氯甲烷(色谱纯, 美国TEDIA公司); N-丙基乙二胺(primary secondary amine, PSA, 40~63 μm)、石墨化炭黑(graphitized carbon black, GCB, 120~400 mesh)(德国CNW公司)。
Thermo Scientific TRACE 1300-TSQ 9000气相色谱串联质谱仪(美国Thermo Fisher Scientific公司); FA25型均质机(上海FLUKO公司); ML802T型电子天平(精密度0.01 g, 美国梅特勒-托利多公司); Milli-Q型超纯水系统(法国默克公司); TurbovapLV型氮吹仪(瑞典Biotage公司); 3-18Ks型落地式冷冻高速离心机(美国SIGMA公司); VORTEX GENIE2涡旋振荡器(美国Scientific Industries公司); DB-5MS UI毛细管柱(30 m×0.25 mm, 0.25 μm, 美国Agilent公司)。
用丙酮将100 mg/L的208种混合标准品母液稀释10倍, 配制成质量浓度为10 mg/L的混合标准储备液, 置于-20 ℃冰箱中进行保存。用1:1的丙酮正己烷对混合标准储备液进行稀释, 分别配制成质量浓度为10、20、40、100、150、200 ng/mL的混合标准工作液。
(1)气相色谱条件
色谱柱: DB-5MS UI毛细管柱(30 m×0.25 mm, 0.25 μm); 载气: 氦气(纯度99.999%); 恒流模式, 流速: 1.0 mL/min; 进样量: 1 μL; 进样方式: 不分流进样; 不分流时间: 1.0 min; 进样口温度: 270 ℃; 传输线温度: 280 ℃; 柱温箱升温程序: 初始温度定为40 ℃保持1 min, 然后以25 ℃/min程序升温至90 ℃, 保持1.5 min, 再以25 ℃/min升温至180 ℃, 再以5 ℃/min升温至280 ℃, 最后以10 ℃/min升温至300 ℃保持5 min。
(2)质谱条件
离子源: 电子轰击离子(electronic ignition, EI)源; 电子能量: 70 eV; 离子源温度: 300 ℃; 四极杆温度: 150 ℃; 碰撞气: 氩气(纯度≥99.999%); 溶剂延迟时间: 5 min; 本研究采用分段时间选择反应监测模式(selective reaction monitoring, SRM)进行定量。
首先采用全扫模式对农药混合标准品进行扫描, 确定208种农药所对应的保留时间和母离子, 接着对母离子进行产物离子的扫描, 得到目标化合物的特征碎片离子对, 本研究选择其中响应值高的一对作为定量离子对, 而次强的则作为定性离子对。建立好离子碎片数据库后采用SRM, 根据各农药的保留时间及碎片离子质荷比(m/z)进行分时段检测, 适当调整驻留时间, 确保离子峰在对应时间内完全出峰。最后对碰撞电压等质谱参数进行优化, 使得定量离子对灵敏度处于最优, 检测结果更为可靠、准确。
本研究选用3种在气相色谱中常用的有机溶剂, 乙酸乙酯、正己烷、二氯甲烷进行不同提取剂的回收率对比实验。取空白红糖样品进行加标实验, 除提取溶剂不同, 其余操作步骤如下。称取5 g试样(精确至0.01 g)于50 mL塑料离心管中, 加入5 mL超纯水并涡旋混匀, 静置30 min。再加入15 mL提取剂(正己烷、乙酸乙酯、二氯甲烷), 均质2 min。放入高速离心机中, 以10000 r/min的转速离心7 min。吸取7.5 mL上清液于15 mL离心管中, 40 ℃水浴中氮气吹至近干。加入2 mL 1:1的丙酮正己烷混合溶液, 涡旋复溶。再将复溶液完全吸取至装有150 mg PSA+15 mg GCB净化填料的塑料离心管中进行净化。涡旋混匀1 min, 4000 r/min离心5 min, 过0.22 μm有机滤膜后装瓶, 用于测定。
本研究采用两因素六水平的正交实验以探讨净化填料PSA与GCB的合适比例。首先将PSA的添加量固定为150 mg, 添加不同含量的GCB (5、10、15、20、30、40 mg)于氮吹复溶后的溶液中, 涡旋混匀1 min, 4000 r/min离心5 min, 过0.22 μm有机滤膜后装瓶。计算不同GCB添加量的加标回收率。
接着将GCB添加量固定为15 mg, 添加不同含量的PSA (50、100、150、200、300、400 mg)于复溶后的溶液中, 涡旋混匀1 min, 4000 r/min离心5 min, 过0.22 μm有机滤膜后装瓶。同上对不同PSA添加量的加标回收率进行计算。
称取5 g试样(精确至0.01 g)于50 mL塑料离心管中, 加入5 mL超纯水并涡旋混匀, 静置30 min。再加入15 mL正己烷作为提取剂, 均质2 min。放入高速离心机中, 以10000 r/min的转速离心7 min。吸取7.5 mL上清液于15 mL离心管中, 40 ℃水浴中氮气吹至近干。加入2 mL 1:1的丙酮正己烷混合溶液, 涡旋复溶。再将红糖试样的复溶液完全吸取至含有150 mg PSA和15 mg GCB的塑料离心管中进行净化。涡旋混匀1 min, 4000 r/min离心5 min, 过0.22 μm有机滤膜后装瓶, 用于测定。
实验采用配制基质匹配标准曲线和溶剂外标曲线, 将两条曲线的斜率进行对比, 以探究基质的影响。按照上述1.3.6的步骤分别制备红糖样品基质空白管, 氮吹至近干后加入混合标准储备液, 配制成质量浓度为10、20、40、100、150、200 ng/mL的基质匹配标准曲线, 同时配制相同浓度梯度的溶剂外标曲线。采用ME/%=(B-A)/A×100% (AB分别为溶剂和基质标准曲线的线性方程斜率)对基质效应进行评估。|ME|<10%表示基质效应较弱, 无需采取措施, 可采用溶剂外标曲线进行定量; 10%≤|ME|≤50%为中等基质效应, 需要采用基质匹配标准曲线或加入内标进行校正以消除基质效应的影响; |ME|>50%为强基质效应, 需要重新对前处理方法进行优化。
采用基质匹配标准曲线(10、20、40、100、150、200 ng/mL)进行定量分析。将满足信噪比≥10时的最小加标浓度定为该方法的定量限。参考GB/T 27404—2008《实验室质量控制规范 食品理化检测》选择低、中、高3个浓度进行加标试验。将加标水平定为0.01、0.02、0.10 mg/kg, 每个加标水平做6个平行, 其余步骤按照1.3.6的前处理方法进行操作。利用公式(样品平均浓度/加标浓度)×100%计算得到样品的平均加标回收率; 以6次平行实验结果计算方法精密度=(标准偏差/样品浓度平均值)×100%。
本研究应用赛默飞Trace Finder 4.1定量软件进行数据分析, 采用外标法进行定量; 应用软件Excel 2016、OriginPro 2018进行制表、绘图; 本研究在1、2、10倍定量限的浓度下, 各进行6次重复的加标实验。
首先在每次进样前做好仪器日常维护, 更换气相色谱仪进样口处的衬管隔垫, 对质谱仪进行真空度测试并进行自动调谐, 使仪器达到最佳状态。在数据分析处理时利用保留时间和特征离子进行双定性, 能帮助定性结果更加的准确可靠并有效排除干扰峰的影响, 排除假阳性结果。本研究建立了208种农药及其代谢产物的质谱数据库, 但是在实际实验过程中剔除掉了一些加标回收率低于70%、峰形较差以及基质效应≥50%的化合物, 最终选取出172种化合物作为此次研究讨论的目标化合物。采用按时间分段选择SRM检测208种农药及其代谢产物的混合标准溶液得到的总离子流色谱峰图见图1。172种目标化合物的保留时间、离子对信息、碰撞能等质谱参数见表1
分别统计3种提取剂中加标回收率满足70%~120%的目标化合物数量所占比例(图2)。当正己烷作为提取剂时, 加标回收率满足70%~120%要求的目标化合物数量最多, 为172种, 比例为100%; 二氯甲烷次之, 为162种, 占比94.2%; 乙酸乙酯满足要求的化合物所占比例最低, 为90%。
选择不同的提取剂, 对糖制品中农药提取的效率不同。QuEChERS前处理方法中常用的提取剂有乙腈、丙酮、乙酸乙酯、甲醇等有机溶剂[26-28]。文献中的提取剂大多采用乙腈体系, 但如果样品基质中含糖量较高则会产生包覆现象, 阻碍乙腈渗透而影响提取效率, 进而影响检测结果的准确性[29]。所以本研究选用正己烷、乙酸乙酯、二氯甲烷进行回收率对比实验。从实验过程中发现, 正己烷作为提取剂时的红糖样品在经过均质处理后, 会与加入的水产生乳化现象, 高速离心后分成3层。由于二氯甲烷的密度比水大, 所以均质离心后有机层在下层, 分离时容易吸取到上层的水层。故综合考虑后选用正己烷作为提取剂。
图3所示, 当PSA的添加量固定为150 mg时, 改变GCB的含量(5、10、15、20、30、40 mg), 红糖组样品的加标回收率先减少后增加, 随后又减少, 当GCB添加量为15 mg时回收率达到最大, 表明此时净化效果最好, 所以将红糖的GCB最优添加量定为15 mg。随后将GCB的添加量固定为15 mg, 改变PSA的含量(50、100、150、200、300、400 mg)进行实验。实验结果(图3)表明, 加标回收率均是先减少后增加, 当红糖组PSA添加量为150 mg时加标回收率达到最大。
常用的QuEChERS净化填料由PSA、GCB、C18 3种组成, PSA用于去除碳水化合物、有机酸、脂肪酸; GCB对色素、甾醇等影响因素可以有效去除; C18则用于去除脂肪。考虑到待测组分的特点以及上述吸附剂的特性, 红糖中不含有过高的脂肪, 故不使用C18作为净化填料的组成成分, 只需要探讨PSA和GCB之间的合适比例。实验数据显示, 当PSA和GCB添加量较多时, 不仅会选择性的修饰某些种类的化合物, 还会提高实际应用的成本。综合两组实验数据分析, 最终选择15 mg GCB+150 mg PSA为红糖的净化方案。
以172种化合物的序号(X)对ME (Y)作图, 得到图4。从图中可以看出, NO.6、NO.51(烯丙菊酯、溴氰菊酯)表现为基质抑制效应, 但|ME|<20%; 其余目标化合物均表现为基质增强效应, 且|ME|<50%, 实验数据表明本方法中的172种化合物均可以用基质匹配标准曲线进行定量, 以消除ME的影响。
ME是食品检测中普遍存在的一种干扰现象, 指的是样品中存在除了目标化合物以外会影响检测结果的基质成分[30]。蔗糖在经过深加工程序制成红糖、白糖、姜糖、绵白糖等制品后其中的多糖、氨基酸、色素等成分增加, 使得基质成分变得复杂, 若在前处理时未能很好的净化, 则会对定性定量结果带来影响。
实验结果以目标物峰面积为纵坐标(Y), 质量浓度为横坐标(X, ng/mL), 绘制标准曲线得到线性相关系数(r2)。通过加标实验数据计算得到样品的平均加标回收率、相对标准偏差, 结果见表1。在10~200 ng/mL的线性范围内各目标化合物均具有较好的线性关系, r2在0.9962~0.9999之间, 加标回收率在71.0%~113.0%范围, 相对标准偏差为0.49%~17.00%, 该结果表明本方法的准确度和精密度均能满足国家标准GB/T 27404—2008《实验室质量控制规范 食品理化检测》的要求, 具有较好的重现性和稳定性。
本研究采用空白基质加标的方法考察方法的定量下限, 在加标回收实验中, 将满足信噪比≥10时的最小加标浓度定为该方法的定量限。经过加标实验得到的数据表明, 本方法中的所有目标化合物在0.01 mg/kg加标水平的信噪比(S/N)均大于10, 因此本方法中172种目标化合物的定量限均为0.01 mg/kg。
本研究从广西各大超市购买20余种品牌的红糖产品, 并对这些红糖产品采用本研究建立的方法进行检测, 其检测结果均为未检出。说明目前广西市场上流通的红糖产品农药污染的风险低, 产品质量安全有一定的保障。
本研究建立了一种GC-MS/MS结合QuEChERS对红糖类产品农药残留高通量检测的方法, 一次进样可同时检测并分析红糖中172种农药残留。该方法具有较好的回收率、重现性及稳定性, 同时前处理简单, 大大提高了实验效率。更为重要的是, 该方法解决了红糖类产品农药残留项目检测无标准、无方法的难题, 适用并满足红糖中多种农药残留的检测, 对于保证红糖产品的质量安全具有重要作用, 可为蔗糖产业的生产及贸易互通提供切实可靠的技术支持。
