Objective To explore the effects and mechanism of long non-coding RNA HLA complex group 18 (HCG18)on human ovarian cancer cell SKOV3 and its cisplatin resistance cell line SKOV3/DDP. Methods RT-PCR was used to detect the expression levels of HCG18 and miR-449 in SKOV3 and SKOV3/DDP cells. Cells were transfected with HCG18 siRNA and treated with DDP (12.5 μg/ml). After 48 h, we evaluated cell viability using the MTT assay, examined cell apoptosis and cell cycle through flow cytometry, and confirmed targeting binding of HCG18, miR-449, and Wnt1 with luciferase reporter assay. Results Compared with the human normal ovarian epithelial cell line NOEC, the HCG18 level was significantly increased (P<0.05), and the miR-449 level was significantly decreased in SKOV3 (P<0.05). Compared with the transfection control group, in the DDP-treated cells (both SKOV3 and SKOV3/DDP), we observed significantly reduced cell viability, enhanced cell apoptosis, and cell cycle G2/M phase arrest (P<0.05). Transfection of MiR-449 inhibitor partly reversed the effects of HCG18 siRNA on cell viability, cell apoptosis, and cell cycle in both SKOV3 and SKOV3/DDP cells (P<0.05). The luciferase reporter results showed that HCG18 could bind to the 3'-UTR of miR-449, and miR-449 could bind to the 3'-UTR of Wnt1. Transfection of HCG18 siRNA significantly increased the levels of miR-449 (P<0.05). Transfection of HCG18 siRNA or miR-449 mimic significantly decreased the mRNA levels of Wnt1(P<0.05). Conclusions HCG18 knockdown could inhibit cell viability, promote cell apoptosis and cell cycle G2/M phase arrest through miR-449/Wnt1 signaling pathway, and thus promote cisplatin sensitivity of human ovarian cancer cells.

目的 探讨长链非编码RNA人类白细胞抗原复合体18(HCG18)对人卵巢癌细胞SKOV3及顺铂耐药细胞SKOV3/DDP耐药性的影响及其作用机制。方法 采用RT-PCR检测SKOV3及SKOV3/DDP细胞中HCG18和miR-449的表达水平。采用HCG18 siRNA转染细胞,经12.5 μg/ml顺铂处理48 h后,MTT法检测细胞活性,流式细胞术检测细胞凋亡情况及细胞周期,荧光素酶报告基因实验检测HCG18、miR-449和Wnt1基因之间的靶向作用。结果 与人正常卵巢上皮细胞NOEC比较,SKOV3中HCG18表达明显增高(P<0.05),而miR-449的表达明显降低(P<0.05)。HCG18 siRNA转染后,顺铂处理后的SKOV3和SKOV3/DDP细胞活性均明显低于对照组(P<0.05),细胞凋亡率和G2/M期细胞占比均明显高于对照组(P<0.05)。miR-449 inhibitor转染SKOV3及SKOV3/DDP细胞后,HCG18 siRNA对细胞活性、凋亡和细胞周期的不利影响均被部分逆转(P<0.05)。荧光素酶报告基因实验结果显示,HCG18可靶向结合miR-449的3'-UTR区,miR-449可靶向结合Wnt1基因的3'-UTR区。转染HCG18 siRNA 可明显上调miR-449的表达水平(P<0.05)。单独转染HCG18 siRNA或miR-449 mimic均可明显下调Wnt1的mRNA表达水平(P<0.05)。结论 下调HCG18可通过miR-449/Wnt1信号通路抑制人卵巢癌细胞活性,促进癌细胞凋亡,诱导细胞周期G2/M期阻滞,进而提高卵巢癌细胞对顺铂的敏感性。

目的 探讨长链非编码RNA人类白细胞抗原复合体18(HCG18)对人卵巢癌细胞SKOV3及顺铂耐药细胞SKOV3/DDP耐药性的影响及其作用机制。方法 采用RT-PCR检测SKOV3及SKOV3/DDP细胞中HCG18和miR-449的表达水平。采用HCG18 siRNA转染细胞,经12.5 μg/ml顺铂处理48 h后,MTT法检测细胞活性,流式细胞术检测细胞凋亡情况及细胞周期,荧光素酶报告基因实验检测HCG18、miR-449和Wnt1基因之间的靶向作用。结果 与人正常卵巢上皮细胞NOEC比较,SKOV3中HCG18表达明显增高(P<0.05),而miR-449的表达明显降低(P<0.05)。HCG18 siRNA转染后,顺铂处理后的SKOV3和SKOV3/DDP细胞活性均明显低于对照组(P<0.05),细胞凋亡率和G2/M期细胞占比均明显高于对照组(P<0.05)。miR-449 inhibitor转染SKOV3及SKOV3/DDP细胞后,HCG18 siRNA对细胞活性、凋亡和细胞周期的不利影响均被部分逆转(P<0.05)。荧光素酶报告基因实验结果显示,HCG18可靶向结合miR-449的3'-UTR区,miR-449可靶向结合Wnt1基因的3'-UTR区。转染HCG18 siRNA 可明显上调miR-449的表达水平(P<0.05)。单独转染HCG18 siRNA或miR-449 mimic均可明显下调Wnt1的mRNA表达水平(P<0.05)。结论 下调HCG18可通过miR-449/Wnt1信号通路抑制人卵巢癌细胞活性,促进癌细胞凋亡,诱导细胞周期G2/M期阻滞,进而提高卵巢癌细胞对顺铂的敏感性。

Objective To explore the effects and mechanism of long non-coding RNA HLA complex group 18 (HCG18)on human ovarian cancer cell SKOV3 and its cisplatin resistance cell line SKOV3/DDP. Methods RT-PCR was used to detect the expression levels of HCG18 and miR-449 in SKOV3 and SKOV3/DDP cells. Cells were transfected with HCG18 siRNA and treated with DDP (12.5 μg/ml). After 48 h, we evaluated cell viability using the MTT assay, examined cell apoptosis and cell cycle through flow cytometry, and confirmed targeting binding of HCG18, miR-449, and Wnt1 with luciferase reporter assay. Results Compared with the human normal ovarian epithelial cell line NOEC, the HCG18 level was significantly increased (P<0.05), and the miR-449 level was significantly decreased in SKOV3 (P<0.05). Compared with the transfection control group, in the DDP-treated cells (both SKOV3 and SKOV3/DDP), we observed significantly reduced cell viability, enhanced cell apoptosis, and cell cycle G2/M phase arrest (P<0.05). Transfection of MiR-449 inhibitor partly reversed the effects of HCG18 siRNA on cell viability, cell apoptosis, and cell cycle in both SKOV3 and SKOV3/DDP cells (P<0.05). The luciferase reporter results showed that HCG18 could bind to the 3'-UTR of miR-449, and miR-449 could bind to the 3'-UTR of Wnt1. Transfection of HCG18 siRNA significantly increased the levels of miR-449 (P<0.05). Transfection of HCG18 siRNA or miR-449 mimic significantly decreased the mRNA levels of Wnt1(P<0.05). Conclusions HCG18 knockdown could inhibit cell viability, promote cell apoptosis and cell cycle G2/M phase arrest through miR-449/Wnt1 signaling pathway, and thus promote cisplatin sensitivity of human ovarian cancer cells.

在常见的妇科恶性肿瘤中,卵巢癌的发病率居第三位,病死率居首位[1]。化疗耐药是卵巢癌疗效不佳的重要原因[2-3]。据统计,约75%的卵巢癌患者在化疗过程中会产生耐药性[4]。因此,逆转耐药、提高药物敏感性是卵巢癌治疗中亟待解决的难题[5]。长链非编码RNA(long non-coding RNA,lncRNA)是一段没有固定开放阅读框的RNA序列,长度大于200个核苷酸。lncRNA在核苷酸序列、空间结构、表达水平及蛋白结合力等方面的异常会影响多种肿瘤的发生与发展[6-7]。lncRNA人类白细胞抗原复合体18(lncRNA HCG18)是一种免疫相关基因,在间变性胶质瘤中呈高表达并可作为预后标志物[8],但其在卵巢癌中的作用报道较少。微小RNA(microRNA,miRNA)可通过与下游靶基因3'-UTR靶向结合而抑制靶基因的表达,参与调控生物功能。异常表达的miRNA与肿瘤的发生、转移和耐药等过程密切相关,有望用于肿瘤的诊断、治疗及预后评估[9-10]。研究显示,miR-449在卵巢癌中表达下调,并能阻滞卵巢癌的细胞周期[11-12]。我们通过数据库预测发现,HCG18与miR-449存在靶向关系,因此,本研究旨在观察HCG18与miR-449在卵巢癌化疗耐药中的作用并探讨其机制。
人正常卵巢上皮细胞株NOEC、人卵巢癌细胞株SKOV3购自中国微生物菌种资源库,对顺铂(DDP)耐药的人卵巢癌细胞株SKOV3/DDP为本实验室前期建立。RPMI 1640培养基、胎牛血清(FBS)购自浙江天杭生物科技公司;脂质体LipofetamineTM 2000购自美国Invitrogen公司;MTT 试剂盒购自北京康为世纪公司;Trizol试剂盒购自美国 Sigma公司;RNA提取试剂盒购自德国QIAGEN公司;反转录酶和Taq酶购自美国Promega公司;Annexin V-FITC /PI双染细胞凋亡检测试剂盒购自北京索莱宝科技有限公司;重组载体质粒HCG18小干扰RNA(small interfering RNA,siRNA)、NC siRNA、mimic NC、miR-449 mimic及miR-449 inhibitor均购自上海吉凯基因化学技术有限公司;兔抗Wnt1多克隆抗体(27935-1-AP)、小鼠抗β-actin单克隆抗体(T0022)、辣根过氧化物酶(HRP)标记的山羊抗兔IgG(S0001)均购自Proteintech公司;HRP标记的兔抗小鼠IgG(ab6728)购自Abcam公司。
NOEC、SKOV3和SKOV3/DDP细胞分别常规培养于RPMI 1640培养基,内含10% FBS、100 U/ml青霉素及100 U/ml链霉素,置于湿度95%、5% CO2及37 ℃培养箱中恒温培养。每隔3 d以0.25%胰蛋白酶消化细胞并传代1次。实验采用活力良好的对数生长期细胞。转染前1 d分别将SKOV3和SKOV3/DDP细胞用胰蛋白酶消化成单细胞悬液,接种于6孔板中,密度为3×105个/孔,加入不含抗生素、含体积分数10% FBS的RPMI 1640 培养基培养过夜,待细胞生长至40%融合时,将特异性沉默lncRNA HCG18基因的重组载体质粒HCG18 siRNA和阴性对照NC siRNA分别转染至SKOV3和SKOV3/DDP细胞。采用同样的方法将mimic NC、miR-449 mimic或miR-449 inhibitor转染进细胞。
以Trizol试剂盒提取细胞中总RNA。为检测miR-449的表达水平,采用特异性反转录引物5'-CTCAACTGGTGTCGTGGAG TCGGCAATTCAGTTGAGACCAGCTA-3'进行反转录合成cDNA。为检测HCG18、Wnt1β-actinU6 snRNA的表达水平,采用Oligo dT Primer反转录合成cDNA。以反转录合成的cDNA为模板,采用RT-PCR试剂盒SYBR@ Premix Ex TaqTM进行PCR扩增,反应总体系25 μl;反应条件为:95 ℃ 10 min预变性;95 ℃ 15 s变性,56 ℃ 30 s退火,72 ℃ 45 s延伸,进行40个循环。引物均由上海生工生物公司合成(表1)。每个样本检测均重复3次,采用2–ΔΔCt法计算目的基因mRNA的相对表达量。
将转染后的各组SKOV3及SKOV3/DDP细胞分别以1×105个/孔接种于96孔板,每孔终体积为200 μl。待细胞贴壁后,每孔加入12.5 μg/ml 顺铂,各组细胞均设置3个复孔;48 h后,每孔加入10 μl MTT溶液(5 mg/ml),于37 ℃避光孵育4 h;每孔继续加入10 μl二甲基亚砜(DMSO),避光震荡10 min。应用酶标仪于570 nm波长下读取吸光度值(A值)。细胞活性(%)=实验组A值/对照组A值×100%。
(1)检测细胞凋亡:将转染后的各组SKOV3和SKOV3/DDP细胞分别以4×105个/孔接种于6孔板,待细胞贴壁后,分别加入终浓度为12.5 μg/ml的顺铂;药物处理48 h后,收集并重悬细胞,按照Annexin V-FITC /PI双染细胞凋亡检测试剂盒说明加入Annexin V-FITC和PI,避光孵育后于流式细胞仪中进行双色荧光细胞流式计数,计算凋亡细胞百分比。(2)检测细胞周期:将转染后的各组细胞分别以4×105个/孔接种于6孔板,待细胞贴壁后,分别以12.5 μg/ml 顺铂处理48 h;收集细胞,按照细胞周期检测试剂盒说明处理待测细胞,采用流式细胞仪检测细胞周期的变化并分析结果。
分别构建HCG18和Wnt1野生型质粒及突变型质粒(HCG18 3'-UTR wt/mut,Wnt1 3'-UTR wt/mut);分别以mimic NC/miR-449 mimic与HCG18 3'-UTR wt/mut或Wnt1 3'-UTR wt/mut共转染SKOV3和SKOV3/DDP细胞。转染48 h后,采用双荧光素酶报告基因检测试剂盒检测荧光素酶活性。各组独立实验均重复3次。
收集细胞,弃掉培养液后每孔加入1 ml RIPA裂解液,充分混悬,冰上裂解0.5 h,离心,取上清,采用BCA试剂盒检测蛋白浓度。每孔上样20 μg蛋白,进行10% SDS-PAGE凝胶电泳,结束后电转至PVDF膜上,随后用5%脱脂奶粉室温封闭1 h。TBST清洗,加入一抗Wnt1(1:1000)或β-actin(1:10 000),4 ℃过夜孵育。TBST清洗4次,加入HRP标记的二抗(1:10 000),室温孵育2 h,洗膜,显影。用ImageJ软件进行灰度分析。以目的条带和β-actin条带的灰度值比值作为蛋白相对表达量。实验重复3次。
采用SPSS 19.0软件进行统计分析。所有实验均重复进行3次,符合正态分布的计量资料以$\bar{x}±s$表示,多组间比较采用单因素方差分析,进一步两两比较采用SNK-q检验。P<0.05为差异有统计学意义。
RT-PCR检测结果显示,与正常细胞NOEC比较,卵巢癌SKOV3细胞中lncRNA HCG18相对表达水平明显增高(P<0.05,图1A),而miR-449相对表达水平明显降低(P<0.05,图1B)。
经12.5 μg/ml 顺铂处理48 h后,对HCG18 siRNA转染的SKOV3和SKOV3/DDP细胞进行RT-PCR检测显示,与对照组比较,HCG18的表达水平均明显降低(P<0.05,图2A);MTT法检测结果显示,与对照组比较,SKOV3和SKOV3/DDP细胞活性均明显降低(P<0.05,图2B)。
经12.5 μg/ml 顺铂处理48 h后,采用流式细胞术检测各组HCG18 siRNA转染后细胞凋亡情况,结果显示,与对照组比较,SKOV3和SKOV3/DDP细胞凋亡率均明显增高(P<0.05,图3)。
经12.5 μg/ml 顺铂处理48 h后,以流式细胞术检测HCG18 siRNA转染的各组细胞周期,结果显示,与对照组比较,SKOV3和SKOV3/DDP的G2/M期细胞占比均明显增多(28.7%vs. 19.8 %,23.4% vs. 10.6 %,P<0.05,图4)。
Starbase2.0预测结果显示,HCG18上存在miR-449的潜在结合位点(图5A);而TargetScan预测显示,Wnt1基因的3'-UTR也存在miR-449的潜在结合位点(图5B)。荧光素酶报告基因实验结果显示,miR-449 mimic与HCG18或Wnt1 3'-UTR的野生型质粒共转染后荧光强度均明显降低(P<0.05,图5C-D)。RT-PCR结果显示,HCG18 siRNA转染后miR-449表达水平明显增高(P<0.05,图5E);miR-449 mimic转染后miR-449表达水平明显增高(P<0.05),miR-449 inhibitor转染后miR-449表达水平明显降低(P<0.05,图5F)。RT-PCR和Western Blotting结果显示,HCG18 siRNA或miR-449 mimic单独转染均可显著下调Wnt1的mRNA和蛋白表达水平(P<0.05,图5G)。
与HCG18 siRNA单独转染比较,miR-449 inhibitor与HCG18 siRNA同时转染后,顺铂处理48 h后的SKOV3和SKOV3/DDP细胞中miR-449表达水平下降,细胞活性增高,细胞凋亡率下降,G2/M期细胞占比增高,差异均有统计学意义(P<0.05,图6)。
顺铂是目前治疗卵巢癌的一线化疗药物,其主要作用机制为抑制DNA复制和转录,阻滞细胞停滞于G2期,但其耐药性可影响治疗效果。这可能与抗肿瘤药物引起的细胞凋亡程序缺陷有关[13]。越来越多的研究显示,lncRNA-miRNA轴与卵巢癌耐药性相关[14-15]。有研究报道,lncRNA DANCR在卵巢癌中能够通过靶向调节miR-145促进肿瘤增长和血管生成[16];上调lncRNA ANRIL水平可以促进卵巢癌细胞的增殖、迁移和侵袭[17]。然而,许多异常表达的lncRNA在卵巢癌中的作用还不清楚[18]
lncRNA HCG18与多种疾病的病理过程相关,例如,在腰椎间盘退变中参与炎症反应和细胞增生过程,而在胃癌、肝癌中高表达,并与患者的不良预后显著相关[19-20]。另外,在结直肠癌中HCG18可通过靶向miR-1271增强MTDH/Wnt/β-catenin信号通路的作用,从而促进结直肠癌的发生发展[21];在胃癌细胞中敲除HCG18,胃癌细胞的增殖、迁移和侵袭均受到抑制[22]
本研究结果显示,lncRNA HCG18在卵巢癌细胞中的表达水平明显高于正常卵巢上皮细胞;下调HCG18在卵巢癌细胞及其耐药细胞中的表达水平,可明显降低两组细胞顺铂处理后的细胞活性,并可促进细胞凋亡和G2/M期阻滞。应用公共数据库Starbase2.0分析结果显示,HCG18下游靶向调控的miRNA中,miR-449在卵巢癌细胞中的表达水平降低;miR-449具有阻滞肿瘤细胞周期的作用。双荧光素酶报告基因实验结果显示,HCG18与miR-449存在靶向结合。以往研究显示,Wnt/β-catenin信号通路在细胞增殖和肿瘤发生过程中发挥重要作用,提示HCG18可能通过miR-449调控Wnt1,也提示miR-449与HCG18和Wnt1之间可能均存在靶向调控关系。有研究显示,miR-449可通过靶向调节Wnt/β-catenin通路抑制甲状腺乳头状癌细胞的增殖[23]。本研究经TargetScan网站预测miR-449可以靶向调节Wnt1,随后通过双荧光素酶报告基因实验、RT-PCR及Western blotting检测发现,miR-449可靶向抑制Wnt1的表达。由此推测,HCG18可能通过调控miR-449/Wnt1轴调节卵巢癌细胞的耐药性。为了验证这一推测,将miR-449 inhibitor转染入HCG18敲除的卵巢癌耐药细胞内,结果显示,miR-449 inhibitor可逆转HCG18敲除对顺铂处理后卵巢癌细胞活性、细胞凋亡和细胞周期的影响,降低卵巢癌细胞对顺铂的敏感性。
综上所述,本研究结果显示,下调HCG18可通过miR-449/Wnt1信号通路抑制卵巢癌细胞顺铂处理后的细胞活性并促进其凋亡,诱导细胞周期G2/M期阻滞,进而提高卵巢癌细胞对顺铂的敏感性。
