Objective To investigate the effect and the underlying molecular mechanism of zinc finger protein Zpr1 on differentiation and the insulin signaling pathway of pre-adipocytes. Methods A high-fat diet-induced insulin resistance mouse model (IR group, n=7) was established by feeding 8-week-old C57BL/6J male mice using high-fat feed for 16 weeks, with conventional diet feeding mice as control group (n=7). Glucose tolerance and insulin tolerance tests were used to observe the blood glucose values of mice in each group and verify the insulin resistance phenotype of mice in the induction group. Western blotting and RT-PCR were performed to detect the protein and mRNA expression of Zpr1 in subcutaneous and visceral adipose tissue from high-fat diet induced insulin resistance mice and regular diet mice. Bioinformatic prediction combined with dual-luciferase reporter gene system were employed to detect the influence of single nucleotide polymorphism (SNP) rs964184 GG genotype and CC genotype in human ZPR1 against miR-4286 binding to the 3'UTR of ZPR1. After mouse preadipocyte 3T3-L1 transfected with miR-4286 simulant to observe the effect of miR-4286 regulating Zpr1 expression on adipocyte differentiation and insulin signaling pathway. Results The protein and mRNA expression levels of Zpr1 decreased obviously in the subcutaneous and especially in the visceral adipose tissues of high-fat diet induced insulin resistant (IR) mice compared with the mice in control group (P<0.05, P<0.01).The rs964184 SNP locus of ZPR1 made it difficult that ZPR1 3'UTR forms a multi-hairpin structure, but may promote miR-4286 to recognize and bind to ZPR1 3'UTR. The miR-4286 level increased obviously in subcutaneous and especially in visceral adipose tissues of IR model group mice (P<0.05); meanwhile, the protein expression level decreased of Zpr1 in mouse pre-adipose cells 3T3-L1 regulated by miR-4286 simulacrum (P<0.05). At the same time, after transfection of miR-4286 simulacrum, the expression levels of fat cell differentiation and insulin signaling pathway-related proteins PPAR-γ, Perilipin A, pAkt, and IRS-1 in 3T3-L1 cells were reduced markedly (P<0.05). Conclusions The GG genotype rs964184 locus can promote the post-transcriptional regulation of Zpr1 by miR-4286. The decreased expression of Zpr1 can inhibit the differentiation of 3T3-L1 preadipocytes and interfere the insulin signaling pathway.

目的 探讨锌指蛋白ZPR1对前脂肪细胞分化及胰岛素信号通路的影响及潜在的分子机制。方法 采用高脂饲料喂养8周龄C57BL/6J雄性小鼠16周建立高脂饮食诱导胰岛素抵抗小鼠模型(IR模型组,n=7),以常规饲料喂养小鼠为对照组(n=7)。采用葡萄糖耐量及胰岛素耐量实验,检测各组小鼠血糖值,验证诱导组小鼠胰岛素抵抗表型。采用Western blotting及RT-PCR检测模型组及对照组小鼠皮下和内脏脂肪组织中Zpr1蛋白及mRNA表达的变化。采用生物信息学预测联合双荧光素酶报告基因系统检测人ZPR1的单核苷酸多态性(SNP)rs964184位点分别出现GG基因型和CC基因型时对miR-4286结合ZPR1 3'UTR的影响。小鼠前脂肪细胞3T3-L1转染miR-4286模拟物后,观察miR-4286调控Zpr1表达对脂肪细胞分化及胰岛素信号通路的影响。结果 与对照组比较,IR模型组小鼠皮下及内脏脂肪组织中Zpr1蛋白及mRNA表达水平明显降低(P<0.05),且内脏脂肪组织降低更明显(P<0.01)。ZPR1的rs964184 SNP位点导致ZPR1 3'UTR不易形成多重发卡结构,可促进miR-4286识别并结合于ZPR1 3'UTR。IR模型组小鼠皮下及内脏脂肪组织中miR-4286表达水平明显升高,且内脏脂肪组织中增高更明显(P<0.05)。小鼠前脂肪细胞3T3-L1中Zpr1蛋白的表达水平受miR-4286模拟物调控降低(P<0.05);而转染miR-4286模拟物后,3T3-L1细胞中脂肪细胞分化及胰岛素信号通路相关蛋白PPAR-γ、Perilipin A、pAkt、IRS-1表达水平明显降低(P<0.05)。结论 GG型rs964184位点可促进miR-4286对Zpr1的转录后调控,Zpr1低表达可抑制3T3-L1前脂肪细胞的分化及胰岛素信号通路。

目的 探讨锌指蛋白ZPR1对前脂肪细胞分化及胰岛素信号通路的影响及潜在的分子机制。方法 采用高脂饲料喂养8周龄C57BL/6J雄性小鼠16周建立高脂饮食诱导胰岛素抵抗小鼠模型(IR模型组,n=7),以常规饲料喂养小鼠为对照组(n=7)。采用葡萄糖耐量及胰岛素耐量实验,检测各组小鼠血糖值,验证诱导组小鼠胰岛素抵抗表型。采用Western blotting及RT-PCR检测模型组及对照组小鼠皮下和内脏脂肪组织中Zpr1蛋白及mRNA表达的变化。采用生物信息学预测联合双荧光素酶报告基因系统检测人ZPR1的单核苷酸多态性(SNP)rs964184位点分别出现GG基因型和CC基因型时对miR-4286结合ZPR1 3'UTR的影响。小鼠前脂肪细胞3T3-L1转染miR-4286模拟物后,观察miR-4286调控Zpr1表达对脂肪细胞分化及胰岛素信号通路的影响。结果 与对照组比较,IR模型组小鼠皮下及内脏脂肪组织中Zpr1蛋白及mRNA表达水平明显降低(P<0.05),且内脏脂肪组织降低更明显(P<0.01)。ZPR1的rs964184 SNP位点导致ZPR1 3'UTR不易形成多重发卡结构,可促进miR-4286识别并结合于ZPR1 3'UTR。IR模型组小鼠皮下及内脏脂肪组织中miR-4286表达水平明显升高,且内脏脂肪组织中增高更明显(P<0.05)。小鼠前脂肪细胞3T3-L1中Zpr1蛋白的表达水平受miR-4286模拟物调控降低(P<0.05);而转染miR-4286模拟物后,3T3-L1细胞中脂肪细胞分化及胰岛素信号通路相关蛋白PPAR-γ、Perilipin A、pAkt、IRS-1表达水平明显降低(P<0.05)。结论 GG型rs964184位点可促进miR-4286对Zpr1的转录后调控,Zpr1低表达可抑制3T3-L1前脂肪细胞的分化及胰岛素信号通路。

Objective To investigate the effect and the underlying molecular mechanism of zinc finger protein Zpr1 on differentiation and the insulin signaling pathway of pre-adipocytes. Methods A high-fat diet-induced insulin resistance mouse model (IR group, n=7) was established by feeding 8-week-old C57BL/6J male mice using high-fat feed for 16 weeks, with conventional diet feeding mice as control group (n=7). Glucose tolerance and insulin tolerance tests were used to observe the blood glucose values of mice in each group and verify the insulin resistance phenotype of mice in the induction group. Western blotting and RT-PCR were performed to detect the protein and mRNA expression of Zpr1 in subcutaneous and visceral adipose tissue from high-fat diet induced insulin resistance mice and regular diet mice. Bioinformatic prediction combined with dual-luciferase reporter gene system were employed to detect the influence of single nucleotide polymorphism (SNP) rs964184 GG genotype and CC genotype in human ZPR1 against miR-4286 binding to the 3'UTR of ZPR1. After mouse preadipocyte 3T3-L1 transfected with miR-4286 simulant to observe the effect of miR-4286 regulating Zpr1 expression on adipocyte differentiation and insulin signaling pathway. Results The protein and mRNA expression levels of Zpr1 decreased obviously in the subcutaneous and especially in the visceral adipose tissues of high-fat diet induced insulin resistant (IR) mice compared with the mice in control group (P<0.05, P<0.01).The rs964184 SNP locus of ZPR1 made it difficult that ZPR1 3'UTR forms a multi-hairpin structure, but may promote miR-4286 to recognize and bind to ZPR1 3'UTR. The miR-4286 level increased obviously in subcutaneous and especially in visceral adipose tissues of IR model group mice (P<0.05); meanwhile, the protein expression level decreased of Zpr1 in mouse pre-adipose cells 3T3-L1 regulated by miR-4286 simulacrum (P<0.05). At the same time, after transfection of miR-4286 simulacrum, the expression levels of fat cell differentiation and insulin signaling pathway-related proteins PPAR-γ, Perilipin A, pAkt, and IRS-1 in 3T3-L1 cells were reduced markedly (P<0.05). Conclusions The GG genotype rs964184 locus can promote the post-transcriptional regulation of Zpr1 by miR-4286. The decreased expression of Zpr1 can inhibit the differentiation of 3T3-L1 preadipocytes and interfere the insulin signaling pathway.

糖尿病威胁着全球近1/10人口的健康,其中约90%为2型糖尿病(type 2 diabetes mellitus,T2DM)[1]。全基因组关联研究(genome-wide association studies,GWAS)证实,T2DM是一种复杂的多基因变异代谢性疾病[2-4]。本课题组前期基于GWAS技术对比分析了本地区汉族人群中3505例T2DM患者与6911名健康无关个体,发现定位于锌指蛋白1(zinc finger protein 1,ZPR1)编码基因上的一个基因多态性位点(single nucleotide polymorphisms,SNPs),即rs964184位点的GG基因型与空腹血糖和糖化血红蛋白水平呈正相关,是T2DM的易感位点[5]ZPR1基因位于11号染色体长臂2区3带(11q23),其编码的ZPR1蛋白具有细胞质至细胞核的转位和信号转导功能,参与生理状态下机体的血糖平衡、糖脂代谢及细胞增殖等过程[5-8]。但当前关于ZPR1基因与糖脂代谢相关疾病的研究均未深入至分子机制层面,因此,深入探讨ZPR1基因促进T2DM发生发展的机制具有重要意义。rs964184位于ZPR1基因的3'UTR,可能会影响周围序列与微小非编码RNA分子(microRNA,miRNA)的靶向结合[9]。本课题组前期预实验通过生物信息学分析发现,miR-4286的潜在结合位点与rs964184距离最近。miR-4286可参与三酰甘油代谢的调控,既往研究证实,血浆中miR-4286与三酰甘油浓度呈明显正相关,并可增加发生急性冠脉综合征的风险[10]。上述结果提示,rs964184可能通过影响ZPR1的转录后调控来影响脂质代谢,进而诱发胰岛素抵抗及T2DM。本研究探讨了脂肪细胞中ZPR1的表达变化与胰岛素抵抗的相关性,并验证位于ZPR1基因3'UTR的SNP(即rs964184位点)GG/CC基因型影响miR-4286转录后调控ZPR1表达的具体机制,旨在从分子层面证实ZPR1在糖尿病发生发展中的作用机制。
人胚肾2 9 3 T细胞(HEK293T)和小鼠前脂肪细胞3T3-L1购自中国科学院细胞库(上海)。14只6周龄C57BL/6J雄性小鼠购自空军军医大学实验动物中心[动物许可证号:SYXK(陕)2017-001]。小鼠饲养于SPF级、12 h昼夜循环、环境温度22 ℃的标准动物房,自由饮水、进食。正式实验开始前适应性饲养2周。本研究获得西安交通大学医学部医学生物科研伦理委员会批准(批准号2017-588),实验过程符合国家及单位有关实验动物的管理及使用规定。
高脂饲料[含胆固醇(0.2%)、猪油(32.0%)、豆油(3.2%)、蛋白质(25.0%)、玉米淀粉(16.0%)、蔗糖(9.0%)、纤维素(6.0%)、其他(8.6%),货号D12492]购自美国Research Diets公司,常规小鼠饲料购自空军军医大学实验动物中心。高糖DMEM培养基、胎牛血清(FBS)和青-链霉素(100×)、0.25%胰蛋白酶购自美国Gibco公司,Trizol、第一链cDNA合成试剂盒(TransScript® Ⅱ First-Strand cDNA Synthesis SuperMix)、脂质体Lipofectamine 3000购自美国Invitrogen公司,5-羧基荧光素标记对照物、miR-4286 mimics及阴性对照(mimics NC)由上海吉玛基因公司合成。小鼠Zpr1正义及反义引物、内参基因β-actin正义及反义引物、miR-4286正义引物、内参基因U6正义引物均由上海生工生物工程股份有限公司合成。miRcute microRNA cDNA第一链合成试剂盒、miRcute microRNA荧光定量检测试剂盒(SYBR Green)、miRNA通用反义引物(货号CD109,具体序列未告知)购自北京天根科技生化有限公司,FastStart Universal SYBR Green Master(ROX)荧光定量检测试剂盒购自美国Roche公司,双荧光素酶报告基因检测试剂盒购自美国Promega公司,RIPA细胞裂解液购自上海碧云天公司,Western blotting试剂盒购自北京鼎国生物科技公司,ZPR1抗体(货号2014S)、β-actin抗体(货号3700S)、PPAR-γ抗体(货号95128S)、Perilipin A抗体(货号9349)、磷酸化蛋白激酶B(pAkt)抗体(货号80722)、Akt抗体(货号4691)、IRS1抗体(货号2382)、辣根过氧化物酶(HPR)标记山羊抗兔二抗(货号7074S)、HPR标记马抗小鼠二抗(货号7076S)均购自美国Cell Signaling Tech公司。ZPR1基因3'UTR第734位常见基因CC型载体[pGL3-WT(1-741)]及rs964184位点GG型载体[pGL3-rs964184(GG型)]均由南京金斯瑞公司构建。小鼠前脂肪细胞3T3-L1购自中国科学院细胞库(上海),倒置荧光显微镜购自日本Olympus公司,荧光素酶活性检测使用美国Promega公司GLOMA Luminometer仪。
14只6周龄C57BL/6J雄性小鼠,饲养至8周龄时采用随机数字表法将其分为IR模型组(n=7,饲喂高脂饲料16周)与对照组(n=7,饲喂常规饲料)。
两组小鼠禁食过夜,正常饮水,第2天早上进行实验。空腹血糖检测:待测小鼠固定于小鼠固定器中,以采血针刺破鼠尾尖端侧面的静脉血管,待血滴积累至直径2~3 mm时,立即用血糖试纸的检测区沾取血滴,放入血糖仪中读取血糖值。检测小鼠空腹血糖后,按照2 g/kg的标准进行葡萄糖灌胃,随后检测葡萄糖灌胃后1、30、60、90、120 min的血糖值,并绘制小鼠血糖变化曲线,纵坐标单位为mg/dl,故需换算血糖值,方法:血糖值(mmol/L)×18=血糖值(mg/dl)。
两组小鼠禁食4 h,正常饮水。注射胰岛素前检测随机血糖,胰岛素注射量为1 U/kg,血糖测定时间点为注射胰岛素后30、60、90、120 min。小鼠采血及血糖检测方法同前。
将各组细胞样品用Trizol法提取总RNA,使用Nano-drop2000核酸定量仪测定总RNA浓度,随后用第一链cDNA合成试剂盒或miRcute microRNA cDNA第一链合成试剂盒将RNA反转录为cDNA;使用FastStart Universal SYBR Green Master(ROX)或miRcute microRNA(SYBR Green)荧光定量检测试剂盒进行RT-PCR检测Zpr1β-actin的mRNA表达,并检测miR-4286、内参基因U6的表达。具体引物序列信息见表1
取两组小鼠皮下及内脏脂肪组织,其中皮下脂肪组织选取小鼠双侧腹股沟脂肪,内脏脂肪组织选取小鼠双侧附睾脂肪。将脂肪组织样本加入RIPA裂解液于冰上充分研磨后,静止待脂质分层,弃去上层脂质,下层细胞蛋白裂解液用于后续实验。将各组细胞样品以PBS洗涤3次后,用RIPA细胞裂解液裂解20 min,提取总蛋白。BCA试剂盒定量后,加入上样缓冲液,100 ℃煮沸10 min。取30 μg总蛋白进行SDS-PAGE电泳,随后将总蛋白电转于硝酸纤维膜(NC)。一抗(ZPR1抗体、β-actin抗体)使用1×TBS稀释(1:1000),孵育14 h,然后加入二抗(HPR标记山羊抗兔抗体、HPR标记马抗小鼠抗体,稀释比例均为1:4000)孵育1 h。采用天能Tanon 5200全自动化学发光成像分析系统进行ECL发光成像。
HEK293T和3T3-L1细胞在DMEM培养基中培养,培养基均加入10%胎牛血清、青-链霉素。培养条件为37 ℃、湿度95%、5% CO2。每天观察细胞生长状态,间隔2~3 d换液;细胞生长约至80%融合时,使用0.25%胰蛋白酶消化传代,用于后续实验。
3T3-L1细胞处于对数生长期时铺种于6孔板,待细胞密度达80%时,按照LipofectamineTM 3000转染试剂说明书,转染5-羧基荧光素标记对照物,转染48 h后使用倒置荧光显微镜观察绿色荧光表达情况。
向3T3-L1细胞中转染mimics NC、miR-4286 mimics,将细胞分为空白对照组、mimics NC组、miR-4286 mimics组,转染方法同1.4.2。向各组细胞中分别转染对应小片段RNA,空白对照组仅加入LipofectamineTM 3000转染试剂。转染48 h后,采用Western blotting检测Zpr1、β-actin蛋白的表达水平。采用miRcute microRNA(SYBR Green)荧光定量检测试剂盒进行RT-PCR实验,检测miR-4286的表达水平。mimics NC、miR-4286 mimics序列信息见表1
将HEK293T细胞铺种于24孔细胞培养板,密度为5×104/孔,正常培养24 h后分为pGL3-WT(1-741)共转染mimics NC组、pGL3-WT(1-741)共转染miR-4286 mimics组、pGL3-rs964184(GG型)共转染mimics NC组、pGL3-rs964184(GG型)共转染miR-4286 mimics组。采用LipofectamineTM 3000转染试剂共转染上述各组质粒、小片段RNA及pRL-TK内参载体,每组设3个复孔。转染24 h后,PBS洗涤细胞,然后用Progema试剂盒裂解细胞,于4 ℃下12 000 r/min离心5 min,取4 μl上清,检测荧光素酶活性。
小鼠前脂肪细胞3T3-L1转染方法同1.4.2,将细胞分为mimics NC组与miR-4286 mimics组。为了维持细胞内mimics的表达量,mimics NC与miR-4286 mimics共需转染3次,即在两组细胞分化诱导前2 d,按照上述方法在3T3-L1细胞中转染mimics NC或miR-4286 mimics,此时计为第0天,转染6 h后,更换培养基为含有0.5 mmol/L异丁基甲基黄嘌呤(IBMX)、1 μmol/L地塞米松(DEX)、1 mg/L胰岛素的完全培养基,诱导2 d;再次进行mimics NC、miR-4286 mimics转染,并更换培养基为含有1 mg/L胰岛素的完全培养基,培养2 d;最后在完全培养基中继续培养两组细胞,每2 d换液一次,并于诱导分化第6天进行第3次mimics NC、miR-4286 mimics转染,共分化8 d,90%以上可分化为成熟脂肪细胞,用于后续实验。
将实验1.4.5中的两组细胞弃去培养基,4%多聚甲醛室温固定20 min,PBS轻柔冲洗,然后用新配制的油红O染液孵育30 min,弃去染液后用60%乙醇分色至背景透明,PBS轻柔冲洗3次,置于显微镜下观察并拍照。随后每孔用200 μl异丙醇孵育10 min萃取油红O,并于510 nm波长处测定吸光度值。
采用Western blotting检测1.4.5中两组因的3'UTR,发现miR-4286可识别并结合于ZPR1 3'UTR rs964184的上游19 nt处(697~704 nt),位点相距最近且预测结合评分最高(图3A、B)。通过RNAfold网站对ZPR1的3'UTR进行最小自由能二级结构预测,结果显示,CC基因型3'UTR具有更复杂的空间结构,并导致miR-4286的结合位点被折叠包裹在多重发卡结构内部,而GG基因型可避免3'UTR多重发卡结构的出现,利于miR-4286结合位点的暴露(图3B)。荧光成像结果显示,3T3-L1细细胞脂肪分化及胰岛素应答信号通路相关蛋白表达的变化,蛋白电泳及转膜方法同上,一抗(PPAR-γ抗体、Perilipin A抗体、pAkt抗体、Akt抗体、IRS1抗体)使用1×TBS稀释(1:1000),孵育14 h,二抗(HPR标记山羊抗兔抗体、HPR标记马抗小鼠抗体,1:4000)孵育1 h。采用天能Tanon 5200全自动化学发光成像分析系统进行ECL发光成像。
采用SPSS 22.0软件进行统计分析,并采用GraphPad Prism 8.0软件制图。计量资料符合正态分布且方差齐时以$\bar{x}±s$表示,两组间比较采用t检验,多组间比较采用单因素方差分析,进一步两两比较采用LSD-t检验;方差不齐时采用秩和检验。P<0.05为差异有统计学意义。
C57BL/6J小鼠喂养高脂饮食16周后,IR模型组小鼠的体重较对照组明显增加[(52.68± 2.541)g vs.(30.70±1.76)g,图1A],且血糖水平也明显高于对照组[(146.7±12.28)mg/dl vs.(126.8±14.87)mg/dl,图1 B]。进一步通过葡萄糖耐量实验发现,IR模型组小鼠经葡萄糖灌胃30 min后的血糖水平明显高于对照组[(333.9±75.26)mg/dl vs.(188.6±35.65)mg/dl];随时间延长,两组小鼠血糖水平均降低,至120 min后,对照组小鼠血糖水平[(106.65±25.12)mg/dl]已恢复至接近初始水平[(124.2±15.33)mg/dl],而IR模型组小鼠血糖水平[(216.45±18.55)mg/dl]仍明显高于对照组(图1C)。胰岛素耐量实验结果显示,IR模型小鼠注射胰岛素后,除90 min外,其余各时间点血糖水平均明显高于对照组(P<0.05,图1D),即出现胰岛素抵抗表型,提示建模成功。
Western blotting检测结果显示,对照组小鼠内脏脂肪中Zpr1蛋白表达水平明显高于皮下脂肪(P<0.05),而IR模型组小鼠差异无统计学意义(P>0.05);与对照组比较,IR模型组小鼠皮下及内脏脂肪组织中的Zpr1蛋白表达水平均明显降低(P<0.05),其中内脏脂肪中Zpr1蛋白的降低更为明显(P<0.01,图2A)。RT-PCR结果显示,与对照组比较,IR模型组小鼠皮下及内脏脂肪中Zpr1 mRNA表达水平均明显降低(P<0.05,图2B)。
rs964184位于ZPR1基因3'UTR第734位(常见基因型为CC)。TargetScan网站分析ZPR1基胞转染小片段RNA,即5-羧基荧光素标记阴性对照物,可出现明显绿色荧光(图3C)。RT-PCR结果显示,与mimics NC组比较,转染miR-4286 mimics可使脂肪前体细胞3T3-L1中miR-4286的表达量增加约5倍(P<0.01,图3D)。Western blotting结果显示,miR-4286 mimics组细胞内源性Zpr1蛋白表达水平明显低于mimics NC组(P<0.05,图3E)。双荧光素酶报告基因检测结果显示,miR-4286可识别并结合于ZPR1 3'UTR,且相较于常见的CC基因型,GG型rs964184可明显增强miR-4286与ZPR1 3'UTR的结合(P<0.05,图3F、G)。
RT-PCR检测结果显示,与对照组比较,IR模型组小鼠皮下及内脏脂肪中miR-4286表达明显升高(P<0.05,图4A)。本研究设计了在前脂肪细胞3T3-L1中转染miR-4286模拟物并诱导分化为成熟脂肪细胞的实验流程(图4B),并采用油红O染色进行检测,结果显示,3T3-L1细胞转染miR-4286 mimics后可诱导分化为成熟脂肪细胞,与mimics NC组比较,miR-4286 mimics组细胞中的脂滴明显减少(图4C),且油红O定量检测结果也显示miR-4286 mimics组脂滴内油红O含量明显低于mimics NC组(P<0.05,图4D)。Western blotting检测结果显示,与mimics NC组比较,miR-4286 mimics组Zpr1、PPAR-γ、Perilipin A、IRS1及pAkt蛋白表达水平均明显降低(P<0.01或P<0.05,图4E)。
糖尿病威胁着全球近1/10人口的健康[1],其中约90%为T2DM。脂肪组织作为机体最重要的脂质储存及胰岛素应答组织,与T2DM的发病及治疗密切相关[11]。在高脂、高糖饮食条件下,脂肪组织的脂代谢失衡,脂质储存能力过饱和,进而向血液中释放三酰甘油,导致脂质异位堆积在其他内脏器官中[12]。同时,血浆循环脂质的脂毒性可导致脂肪、肌肉和肝组织的胰岛素抵抗,是T2DM发病的主要原因[13]。在T2DM的临床治疗中,增强脂肪组织的三酰甘油储存能力,可明显降低机体的脂毒性,增加脂肪、肌肉、肝脏等关键代谢器官的胰岛素敏感性[13]。同时,作为一个重要的分泌器官,脂肪组织还可分泌脂联素、成纤维细胞生长因子1(FGF1)、瘦素(leptin)等多种脂肪因子,作用于肌肉和肝脏等代谢组织,进一步促进胰岛素应答、抑制恶性糖异生等[14-15]。因此,调节脂肪组织的功能是T2DM的常用治疗策略。本研究以高脂饮食诱导的胰岛素抵抗小鼠为模型,发现高脂饮食诱导胰岛素抵抗小鼠的皮下及内脏脂肪组织中,Zpr1蛋白表达水平较常规饮食对照组明显降低,且在内脏脂肪中降低更为明显,提示Zpr1可能参与了脂肪细胞脂质代谢失衡导致的胰岛素抵抗。
随着T2DM病因学研究的深入,近年来大量GWAS研究证实,T2DM是一种复杂的多基因变异代谢性疾病[16],基因水平改变介导的能量代谢异常已成为新的研究关键点。本课题组前期的GWAS研究证实,锌指蛋白ZPR1基因内的SNP位点rs964184(GG型)与空腹血糖和糖化血红蛋白水平呈正相关,是T2DM的易感位点[5],但具体分子机制尚不明确。既往关于ZPR1基因的研究多集中于脊髓性肌萎缩、神经退行性变及细胞增殖等方向[17-18],而近期的多个GWAS研究将ZPR1的功能拓展到了能量代谢性疾病[19],证实ZPR1基因及其所处的11号染色体长臂2区3带(11q23)与血脂异常密切相关,其中ZPR1基因的rs964184位点是该区段影响血浆三酰甘油水平的主要因素[20]
然而上述研究均未涉及分子层面的机制,为解析ZPR1在糖脂代谢中的作用机制,本研究以3T3-L1前脂肪细胞为模型,结合生物信息学分析手段,发现GG型rs964184位点可促进miR-4286与ZPR1 3'UTR的结合及mRNA的降解。miR-4286与三酰甘油代谢调控密切相关,有研究发现,血浆中miR-4286与三酰甘油浓度呈明显正相关,并可增加急性冠脉综合征的发生风险[10]。因此,推测脂肪细胞中的ZPR1可能受miR-4286调控参与脂肪细胞功能及胰岛素应答。为验证这一假说,笔者在前脂肪细胞3T3-L1中转染miR-4286,随后诱导细胞成脂分化,结果发现与预期一致:在分化成熟的脂肪细胞中检测到Zpr1蛋白水平明显降低,同时发现调控脂肪细胞分化的重要转录因子PPAR-γ及参与脂滴形成的关键蛋白Perilipin A在miR-4286转染组的表达水平均明显降低,提示Zpr1参与了脂肪细胞的分化成熟。在脂肪细胞的脂滴形成过程中,PPAR-γ转录调控Perilipin A的表达,后者可定位于脂滴表面,促进脂滴形成并包裹保护三酰甘油,避免基础状态下脂肪酶的脂解作用,同时还能在激素刺激时促进脂肪分解。Perilipin A的表达调控在肥胖相关的胰岛素抵抗及糖尿病等代谢性疾病中也发挥了重要作用[21]。此外,本研究还发现,胰岛素通路的关键蛋白IRS1及pAkt表达水平在miR-4286转染组均明显降低,提示胰岛素信号通路受到抑制,细胞呈胰岛素抵抗状态。
综上所述,本研究结果提示,锌指蛋白ZPR1基因内的SNP位点rs964184(GG型)可改变ZPR1 3'UTR空间构象,促进miR-4286对此3'UTR区段的结合及mRNA的降解,从而降低ZPR1的表达,进一步影响脂肪细胞的分化成熟,促进细胞产生胰岛素抵抗表型。但本研究未在脂肪细胞分化成熟过程中探讨ZPR1的rs964184位点对脂肪细胞成熟分化及胰岛素应答的影响。在后续研究中,可在人脂肪间充质干细胞中利用CRISPR/Cas9点突变系统,构建稳定携带ZPR1 GG型rs964184位点的细胞,进一步研究rs964184位点影响miR-4286调控ZPR1表达在脂肪细胞分化成熟和胰岛素应答中的作用。
