Objective To study the expression profile and possible roles of ferroptosis-related genes in a bleomycin-induced murine model of pulmonary fibrosis. Methods Twelve 6-7-week-old male C57BL/6 mice were randomly divided into model group and control group, 6 in each group. The model group received nasal inhalation of bleomycin, while the control group was given an equal volume of normal saline. Lung tissues were collected 3 weeks after modeling. Pathological changes and collagen deposition in lungs were observed by HE and Masson staining. Prussian blue staining was used to observe the level of iron accumulation in lung tissue. Total RNA was extracted for PCR Array to identify ferroptosis-related differentially expressed genes (DEGs). Functional analysis for DEGs was performed by Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis. Expression levels of DEGs were verified by RT-qPCR analysis. Results Compared with control group, fibrosis and obvious iron deposition occured in mouse lung tissue of model group. The PCR array identified five ferroptosis-related genes that expression significantly decreased in model group compared with control group including carbonic anhydrase 9 (CA9), cysteinyl-tRNA synthetase 1 (CARS1), heat shock transcription factor 1 (HSF1), NADPH oxidase 3 (NOX3) and mitochondrial ferritin (FTMT)(P<0.05). GO functional enrichment analysis and KEGG pathway analysis showed that the DEGs were mainly related to temperature homeostasis, NADPH oxidase complex, and carbonate dehydratase activity, and were involved in nitrogen metabolism, aminoacyl-tRNA biosynthesis and legionellosis signaling pathways. RT-qPCR verification confirmed that the expression levels of CA9, CARS1, HSF1 and FTMT were significantly decreased in model group than those in control group (P<0.05). Conclusions Change of the expression of genes related to ferroptosis has been confirmed in a murine model of pulmonary fibrosis induced by bleomycin. The result indicates that ferroptosis is involved in the process of idiopathic pulmonary fibrosis, ferroptosis-associated DEGs might provide potential targets for clinical treatment of it.

目的 分析博来霉素诱导的肺纤维化模型小鼠肺组织铁死亡相关基因的表达谱并探讨其可能的作用机制。方法 取12只6~7周龄雄性C57BL/6小鼠,随机分为模型组与对照组,每组6只,分别构建肺纤维化模型及进行对照处理。3周后处死小鼠,采用HE与Masson染色评估其肺部病理改变和胶原纤维堆积状况,普鲁士蓝染色观察肺组织铁聚集水平。应用小鼠铁死亡通路PCR Array筛选差异表达基因,采用生物信息学分析方法对差异基因进行GO功能富集与KEGG信号通路预测。运用实时定量PCR验证模型组与对照组肺组织铁死亡相关差异基因的表达水平。结果 与对照组比较,模型组小鼠肺组织出现纤维化改变及明显的铁沉积。PCR Array检测结果显示,与对照组比较,模型组碳酸酐酶9(CA9)、半胱氨酰-tRNA合成酶1(CARS1)、热休克转录因子(HSF1)、NADPH氧化酶3(NOX3)、线粒体铁蛋白(FTMT)等5个铁死亡相关基因的表达量明显降低(P<0.05)。GO功能富集分析与KEGG信号通路预测结果显示,肺纤维化模型小鼠肺组织的差异表达基因主要与体温内稳态、NADPH氧化酶复合体、碳酸脱氢酶活性等功能有关,并在氮素代谢、氨酰合成、军团菌病等通路中富集。实时定量PCR验证结果显示,模型组小鼠肺组织中CA9CARS1HSF1FTMT的表达量均明显低于对照组(P<0.05)。结论 博来霉素诱导的肺纤维化模型小鼠肺组织中多个铁死亡相关基因的表达水平发生明显变化,提示铁死亡在特发性肺纤维化中发挥重要作用,其中的差异基因或可为临床治疗提供潜在靶点。

目的 分析博来霉素诱导的肺纤维化模型小鼠肺组织铁死亡相关基因的表达谱并探讨其可能的作用机制。方法 取12只6~7周龄雄性C57BL/6小鼠,随机分为模型组与对照组,每组6只,分别构建肺纤维化模型及进行对照处理。3周后处死小鼠,采用HE与Masson染色评估其肺部病理改变和胶原纤维堆积状况,普鲁士蓝染色观察肺组织铁聚集水平。应用小鼠铁死亡通路PCR Array筛选差异表达基因,采用生物信息学分析方法对差异基因进行GO功能富集与KEGG信号通路预测。运用实时定量PCR验证模型组与对照组肺组织铁死亡相关差异基因的表达水平。结果 与对照组比较,模型组小鼠肺组织出现纤维化改变及明显的铁沉积。PCR Array检测结果显示,与对照组比较,模型组碳酸酐酶9(CA9)、半胱氨酰-tRNA合成酶1(CARS1)、热休克转录因子(HSF1)、NADPH氧化酶3(NOX3)、线粒体铁蛋白(FTMT)等5个铁死亡相关基因的表达量明显降低(P<0.05)。GO功能富集分析与KEGG信号通路预测结果显示,肺纤维化模型小鼠肺组织的差异表达基因主要与体温内稳态、NADPH氧化酶复合体、碳酸脱氢酶活性等功能有关,并在氮素代谢、氨酰合成、军团菌病等通路中富集。实时定量PCR验证结果显示,模型组小鼠肺组织中CA9CARS1HSF1FTMT的表达量均明显低于对照组(P<0.05)。结论 博来霉素诱导的肺纤维化模型小鼠肺组织中多个铁死亡相关基因的表达水平发生明显变化,提示铁死亡在特发性肺纤维化中发挥重要作用,其中的差异基因或可为临床治疗提供潜在靶点。

Objective To study the expression profile and possible roles of ferroptosis-related genes in a bleomycin-induced murine model of pulmonary fibrosis. Methods Twelve 6-7-week-old male C57BL/6 mice were randomly divided into model group and control group, 6 in each group. The model group received nasal inhalation of bleomycin, while the control group was given an equal volume of normal saline. Lung tissues were collected 3 weeks after modeling. Pathological changes and collagen deposition in lungs were observed by HE and Masson staining. Prussian blue staining was used to observe the level of iron accumulation in lung tissue. Total RNA was extracted for PCR Array to identify ferroptosis-related differentially expressed genes (DEGs). Functional analysis for DEGs was performed by Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis. Expression levels of DEGs were verified by RT-qPCR analysis. Results Compared with control group, fibrosis and obvious iron deposition occured in mouse lung tissue of model group. The PCR array identified five ferroptosis-related genes that expression significantly decreased in model group compared with control group including carbonic anhydrase 9 (CA9), cysteinyl-tRNA synthetase 1 (CARS1), heat shock transcription factor 1 (HSF1), NADPH oxidase 3 (NOX3) and mitochondrial ferritin (FTMT)(P<0.05). GO functional enrichment analysis and KEGG pathway analysis showed that the DEGs were mainly related to temperature homeostasis, NADPH oxidase complex, and carbonate dehydratase activity, and were involved in nitrogen metabolism, aminoacyl-tRNA biosynthesis and legionellosis signaling pathways. RT-qPCR verification confirmed that the expression levels of CA9, CARS1, HSF1 and FTMT were significantly decreased in model group than those in control group (P<0.05). Conclusions Change of the expression of genes related to ferroptosis has been confirmed in a murine model of pulmonary fibrosis induced by bleomycin. The result indicates that ferroptosis is involved in the process of idiopathic pulmonary fibrosis, ferroptosis-associated DEGs might provide potential targets for clinical treatment of it.

特发性肺纤维化(idiopathic pulmonary fibrosis,IPF)是一种威胁生命的肺部疾病[1],其病死率超过多种肿瘤,发病群体主要为老年人[2]。IPF的危险因素主要包括年龄、吸烟及环境污染等[3]。在早期研究中,炎症被认为是IPF的主要发病机制,因此,类固醇疗法曾长期被用于IPF的治疗。近期研究显示,肺泡细胞的反复损伤与异常修复导致肺间质纤维化沉积的理论,可更为合理地阐释IPF的发病机制[4]。抗纤维化药物尼达尼布与吡非尼酮由于可延缓疾病进程[5],已获批应用于IPF的临床治疗。然而,除肺移植外,目前仍然缺乏治愈IPF的有效措施[6-7]。铁死亡是近年发现的可调控性细胞死亡[8-9],与凋亡、坏死、自噬等形式的细胞死亡不同,其主要特征为铁依赖的危害性脂质活性氧积聚[10-11]。越来越多的证据显示,铁死亡与包括肺纤维化在内的多种肺部疾病密切相关。例如,铁代谢通路中的重要分子谷胱甘肽过氧化物酶4(glutathione peroxidase 4,GPx4)表达缺陷型小鼠对博来霉素诱导的肺纤维化更为易感;而在采用GPx4过表达小鼠构建的IPF模型中,肺纤维化程度明显减轻[12]。关于铁死亡与IPF的多数文献报道,仅明确肺纤维化中存在铁死亡这一现象,其相关机制仍有待进一步阐明。本研究采用博来霉素诱导建立肺纤维化小鼠模型,运用PCR Array技术筛选该模型中与铁代谢相关的差异表达基因,探讨铁死亡在IPF发生中可能的作用机制。
博来霉素购自日本化药株式会社;4%多聚甲醛固定液、琼脂糖、HE染色试剂盒购自上海碧云天生物技术有限公司;Masson染色试剂盒购自南京建成生物工程研究所;普鲁士蓝染色试剂盒购自北京索莱宝科技有限公司;Trizol试剂购自美国Sigma公司;反转录试剂盒、定量PCR试剂盒和小鼠铁死亡相关基因PCR Array plate试剂盒购自上海沃吉基因科技有限公司。PCR引物由上海生工生物工程股份有限公司合成。石蜡包埋机购自常州派斯杰医疗设备有限公司;病理切片机(型号RM2235)购自德国Leica公司;核酸凝胶电泳系统购自上海天能科技有限公司;超微量紫外分光光度计(型号NanoDrop 2000C)购自美国Thermo公司;光学显微镜购自日本奥林巴斯公司;实时荧光定量PCR仪(型号ABI7500)购自美国ABI公司。
6~7周龄的SPF级健康雄性C57BL/6小鼠12只,体重20~25 g,购自常州卡文斯实验动物有限公司[实验动物生产许可证编号:SCXK(苏)2016-0010]。小鼠饲养于屏障环境下、通风良好的南京医科大学附属无锡市人民医院动物实验中心小鼠房内,可自由饮水、进食;饲养环境温度与湿度适宜,照明12 h、黑暗12 h交替进行。适应性喂养一周后,将小鼠随机分为对照组和模型组,每组6只。所有动物护理与实验均符合南京医科大学实验动物福利伦理管理委员会规范,全部操作及处理均遵循美国国立卫生研究院出版的《实验动物护理和使用指南》。
参照文献[13]的方法构建博来霉素诱导的肺纤维化小鼠模型。小鼠腹腔注射1%戊巴比妥(45 mg/kg)麻醉满意后,左手揪住其颈部皮肤,使其仰卧于手心,小指夹住尾巴固定小鼠,使用微量移液器通过鼻腔给药方式对模型组小鼠给予5 mg/kg的博来霉素,对照组给予等量生理盐水。给药3周后,使用1%戊巴比妥(100 mg/kg)过量麻醉对两组小鼠实施安乐死,收集其肺组织。根据解剖学特征将小鼠肺组织分为五叶,取其中一叶用于组织病理学染色,以评估肺纤维化模型构建是否成功,剩余肺组织存放于-80℃超低温冰箱,用于后续差异基因筛选分析等操作。
将新鲜取下的小鼠肺组织浸没于4%多聚甲醛液中固定24 h,梯度乙醇脱水,二甲苯透明,浸蜡,包埋。包好的蜡块以5 μm厚度切片,后续用于HE染色、Masson染色及普鲁士蓝染色,具体操作步骤按相应试剂盒说明书进行。封片后置于光学显微镜下观察,HE染色切片胞质被染成粉红色或红色,胞核呈蓝色;Masson染色切片胞质与肌纤维被染成红色,胶原纤维呈蓝色;普鲁士蓝染色切片胞质含铁血黄素或三价铁时被染成蓝色,细胞核被染成红色,其余呈浅红色或不着色。
每组随机选取3只小鼠,Trizol法提取肺组织总RNA。使用RNA凝胶电泳评估其质量,超微量紫外分光光度仪检测其纯度与浓度。对质检合格的RNA进行反转录以合成cDNA模板,将获得的cDNA与qPCR预混液按说明书比例混合,以9 μl/孔的用量精准加入Ferroptosis PCR Array Plate (mouse)反应板各孔内,根据反应推荐程序对样品进行实时荧光定量PCR测定。
对于PCR Array检测获得的数据,采用2-ΔCt法计算倍数变化值(fold change,FC),以上调或下调倍数变化值∣FC∣≥2且P<0.05作为差异基因的筛选阈值。应用R语言程序包clusterProfiler(4.2.2)对筛选获得的差异基因进行基因本体(gene ontology,GO)富集分析及京都基因与基因组百科全书(Kyoto Encyclopedia of Genes and Genomes,KEGG)信号通路预测,其中GO分析涵盖了生物学中的生物过程、细胞组分和分子功能三个方面。
使用Trizol试剂提取对照组与模型组小鼠肺组织总RNA,利用反转录试剂盒将其反转录为cDNA,采用定量PCR试剂盒对合成的cDNA模板进行实时荧光定量PCR扩增,以β-actin为内参基因,以2-ΔΔCt法计算mRNA的相对表达量。引物序列见表1
通过美国国立生物技术信息中心创建并维护的基因表达综合数据库(gene expression omnibus,GEO,https://www.ncbi.nlm.nih.gov/geo),在“GEO Profiles”检索模式下分别以各差异基因名称与“idiopathic pulmonary fibrosis”作为关键词检索相关数据集,选取以正常对照与IPF患者肺组织样品为基础的高通量芯片数据集,分析各差异基因在相应数据集的表达情况。
采用GraphPad Prism 8软件进行统计分析。计量资料符合正态分布时以$\bar{x}±s$表示,两组间比较采用成组样本t检验。P<0.05为差异有统计学意义。
HE染色结果显示,对照组小鼠肺内结构清晰、肺泡间隔正常,无明显的炎症细胞浸润或纤维化形成;模型组小鼠肺部出现肺泡结构大面积破坏,肺泡腔萎缩消失,出现明显的肺间质纤维化及大量炎性细胞浸润。Masson染色结果显示,对照组小鼠肺泡结构正常、蓝染区域较少且着色浅,未见明显的胶原沉积;与对照组比较,模型组小鼠肺泡结构完全丢失,胶原纤维所对应的蓝染区域明显增加、着色加深,且部分肺泡被胶原覆盖(图1)。
普鲁士蓝染色结果显示,对照组小鼠肺泡结构清晰,肺泡腔与间隔内未见明显的蓝色颗粒,铁染色结果为阴性;与对照组比较,模型组小鼠肺组织结构紊乱,肺间质内出现一定数量的蓝染颗粒,铁染色结果呈阳性(图2)。
分别提取两组小鼠肺组织RNA,反转录后行PCR Array检测,将上调或下调倍数变化值∣FC∣≥2且P<0.05作为筛选条件对结果进行分析。热图可体现样本与差异表达基因的聚类分布情况。本次Array所检测的90个铁死亡相关基因中,红框标注的5个基因在对照组与模型组中各自积聚到一个集中区域(图3)。利用火山图观察两组样本铁死亡相关基因表达水平的差异分布,结果也同样显示,与对照组比较,模型组中有5个基因存在差异表达,即碳酸酐酶9(carbonic anhydrase 9CA9)、半胱氨酰-tRNA合成酶1(cysteinyl-tRNA synthetase 1CARS1)、热休克转录因子(heat shock transcription factor 1HSF1)、烟酰胺腺嘌呤二核苷酸磷酸氧化酶3(NADPH oxidase 3NOX3)和线粒体铁蛋白(mitochondrial ferritinFTMT)基因,其表达变化方向均为下调(P<0.05,图4)。
GO富集结果显示,差异基因在生物过程方面主要参与体温内稳态、盐反应、微管结合调控、铁离子的细胞内固等进程,细胞组分方面主要涉及NADPH氧化酶复合体、前核、分子伴侣复合体、着丝粒等组分,分子功能方面主要与STAT家族蛋白结合、碳酸脱氢酶活性、三价铁结合、二价铁结合、氨酰连接酶活性等功能相关(图5)。KEGG信号通路预测结果显示,除铁死亡外,差异基因还参与氮素代谢、氨酰的生物合成、军团菌病通路等过程(图6)。
采用RT-qPCR检测5个差异基因在对照组与模型组小鼠肺组织中的表达情况,结果显示,与对照组比较,模型组CA9CARS1FTMTHSF1的表达量均明显降低(P<0.05),而两组NOX3表达水平差异无统计学意义(P>0.05,图7)。
检索GEO数据库,结果显示目标差异基因在两个满足筛选条件的数据集中有表达,其中CA9HSF1表达于GDS1252数据集,CARS1NOX3表达于GDS4279数据集,FTMT则未检索到符合样本条件的数据集。对上述差异基因在其相应数据集中的表达量进行分析,结果显示,CA9HSF1CARS1NOX3在IPF临床肺组织样本中的表达模式与本研究两组小鼠肺组织PCR Array检测结果中的表达模式一致,差异均无统计学意义(表2)。
IPF作为一种慢性、进展性、年龄相关的间质性肺部疾病,因其发病率较高、发病机制尚未明确、病程不可逆且预后不良而成为当前社会经济的潜在负担之一[14]。近年研究显示,IPF患者感染新冠肺炎病毒后,发展为重症新冠肺炎的风险较非IPF感染者增高[15]。鉴于上述特征,深入研究IPF发病机制、寻找有效的早期诊断与治疗靶点迫在眉睫。
有多种动物模型先后被用于IPF发病机制的研究[16],但由于疾病本身的独特性及其复杂的病理生理学机制,科学家们尚未发现能够完全复制IPF发生的体内模型。目前用于IPF研究的动物模型须具备肺部损伤、无休止的修复以及过量瘢痕形成等三要素[17]。常见的肺纤维化动物模型构建方式包括博来霉素、二氧化硅、石棉等药物诱导,辐射诱导,基因工程诱导等,其中气管滴注法给予博来霉素因其模型动物肺部发生强烈的纤维化反应、与IPF患者相似的组织学特征以及相对低廉的造模成本而被广泛应用于IPF发病机制研究[16,18]
铁死亡作为一种铁依赖型细胞死亡调控方式,由Dixon等[10]于2012年首次报道。研究显示,铁死亡可参与有机体的多种生理与病理进程,在卒中、脑损伤和肿瘤等多种疾病中起重要作用[19-20]。尽管铁元素为机体细胞正常生理过程所必需,但过量的铁累积会导致胞内脂质过氧化物含量增加,进而触发铁死亡[21]。一些被认定为铁死亡标志或调控因子的基因可参与慢性阻塞性肺疾病、急性肺损伤、哮喘、肺癌等多种良性与恶性肺部疾病的发生发展[22]。值得注意的是,IPF患者肺组织中铁含量升高,且铁元素的积聚与气道纤维化程度及肺功能密切相关[23]。本研究博来霉素诱导的肺纤维化模型小鼠肺组织铁染色呈阳性,提示模型组小鼠肺部有铁元素沉积,与IPF的上述特征相符。在转化生长因子β诱导的肺纤维化细胞模型中,铁死亡诱导剂可通过促进脂质过氧化以及降低GPX4表达而加速成纤维细胞向效应方向分化,而铁死亡抑制剂则可通过降低脂质过氧化和增强GPX4表达而抑制肺纤维化与铁死亡的发生[24]。在博来霉素诱导的肺纤维化模型中,抑制长链非编码RNA锌指反义链1的表达可显著减弱博来霉素引起的脂质过氧化作用,从而抑制铁死亡,缓解肺纤维化[25]。亦有研究显示,IPF患者的支气管肺泡灌洗液中,某些与铁代谢或纤维化相关的标志分子表达紊乱[26]。上述研究成果均提示铁死亡可能在IPF发病过程中发挥重要作用。
PCR Array作为一种以功能分类的PCR阵列芯片技术,结合了实时定量PCR准确灵敏的特性与微阵列芯片检测通量大的优势,可一次性检测多种基因的表达,是分析信号通路或某些生理功能、病理进程相关基因表达变化的首选工具之一[27]。本研究选用的小鼠铁死亡相关基因PCR Array包含了铁死亡通路中90个关键基因,涉及铁代谢、氨基酸与谷胱甘肽代谢、脂质代谢等参与调控铁死亡的生物过程。本研究PCR Array检测显示,与对照组比较,模型组共有5个差异基因,分别为CA9CARS1HSF1NOX3FTMT,其表达变化方向均为下调。本研究通过实时定量PCR实验与GEO数据库挖掘分别验证了5个差异基因在IPF模型小鼠与临床患者肺组织中的表达,结果显示差异基因在动物实验中的表达情况与PCR Array结果基本一致,在肺纤维化临床样本中的表达模式与PCR Array测得的模式也基本相符,但后者表达差异无统计学意义,这可能与各类基因芯片及高通量测序数据中检测铁死亡相关特异性基因的数据集较少,以及目前检索到的两个数据集中样本量有限等因素有关。
CA9是一种跨膜蛋白,属于功能庞大的碳酸酐酶家族,其主要成分为酸性氨基酸,在生理状态下具有维持细胞外基质酸碱平衡的重要作用[28];病理状态下,CA9参与调控实体肿瘤的微环境,且其表达量与多种肿瘤预后密切相关,已成为当下肿瘤研究领域的热点基因,是评估抗肿瘤治疗效果的潜在分子标志物[29]。FTMT是重要的线粒体铁储存蛋白,具有铁氧化酶活性,可通过催化二价铁向三价铁转化实现铁元素的存储[30-31]。现有研究提示,FTMT的主要功能为保护组织特异性细胞免受铁元素依赖的氧化损伤,而非直接调控细胞铁含量[32];Wang等[33]在体外研究中发现FTMT可通过降低细胞不稳定铁池储量及胞质活性氧生成而抑制铁死亡。尽管CA9及FTMT在IPF中的作用鲜有报道,但两者均被认为是铁死亡的抑制因子[33-34];而铁死亡通路在IPF中呈活化状态,故CA9FTMT基因在本研究IPF模型中的表达量降低与铁死亡通路的改变相一致,提示这两个基因可能通过铁死亡通路参与了IPF的疾病发生过程。HSF1作为一种在温度上升等应激状态下可被迅速诱导并表达的转录因子,在生物体生命活动的各阶段、各器官组织中广泛表达,具有调节生长发育、抗凋亡、保护心脏缺血性损伤等功能,并在对抗呼吸机导致的肺部机械损伤性炎症、炎性肠病、类风湿关节炎,以及革兰阴性菌感染导致的内毒素血症等炎症相关疾病中发挥重要作用[35-36]。Chen等[37]的研究显示,HSF1可促进人胚肺成纤维细胞的增殖和纤维化转变,敲低HSF1则可抑制细胞的纤维化进程,这与本研究HSF1在肺纤维化模型中表达量降低的结果并不一致;考虑到HSF1强大的抗炎作用以及炎症反应在肺纤维化中的重要地位,HSF1在本研究中表达量下降或可理解为其在肺纤维化中的抗炎作用大于促纤维化作用。然而影响疾病潜伏与发生的因素通常错综复杂,基因的表达也存在着时空特异性,故HSF1在IPF中的作用仍值得进一步探究。本研究筛选得到的另外两个差异基因CARS1NOX3均为铁死亡通路中的驱动因子,分别行使着抑制肿瘤发生和参与内耳石形成的重要功能[10,38],但两者与IPF的关系鲜有报道,加之对本次PCR Array的验证实验中NOX3表达量在模型组与对照组间的差异无统计学意义,提示CARS1NOX3在IPF中的表达变化及其作用有待进一步验证与探索。
为进一步探究铁死亡在IPF中的作用,本研究对筛选所得的差异基因开展了功能富集性分析,其结果或可为探究IPF的发生机制提供一定参考。GO与KEGG分析结果显示,差异基因主要与体温内稳态、NADPH氧化酶复合体、碳酸脱氢酶活性等功能有关,主要通过氮素代谢、氨酰的生物合成、军团菌病等信号通路参与肺纤维化的发生发展。已有研究显示氮素代谢参与调控IPF的发生[39-40],早年报道中亦有急性军团菌肺炎患者并发肺纤维化的案例[41]
综上所述,本研究通过建立博来霉素诱导的小鼠IPF模型,检测其肺组织铁死亡通路相关基因的表达变化,针对筛选所获的差异基因开展生物信息学分析及表达验证,并结合现有文献报道初步讨论了其在IPF中的潜在作用。本研究以铁死亡为切入点,为IPF的机制探索提供了新思路,也为IPF的早期诊断与治疗提供了潜在的分子标志物与用药靶点。下一步,我们将结合体外与体内实验研究各差异基因通过铁死亡通路调控IPF发生的具体机制。
