Objective To investigate the expression of the histone deacetylase SIRT7 in glioma cells and its impact on epithelial-mesenchymal transformation (EMT), as well as its effects on proliferative, migratory and invasive capabilities of glioma cells. Methods Bioinformatics analysis was conducted on data from glioma patients in the Cancer Genome Atlas (TCGA) and the Chinese glioma Genome Atlas (CGGA) databases to explore the expression of SIRT7 gene in gliomas and its correlation with tumor grading, molecular characteristics and patient clinical prognosis. Glioma cells were randomly divided into control, SIRT7 knockdown, SIRT7 overexpression, drug treatment (10 μmol/L hydrochlorothiazide) and drug (10 μmol/L hydrochlorothiazide)+SIRT7 overexpression groups. The CCK-8 assay, cell scratch assay and Transwell assay were used to observe the effects of upregulating and downregulating SIRT7 expression on glioma cell proliferation, migration and invasion. RT-qPCR and Western blotting were employed to detect the effects of SIRT7 on the expression of neural cadherin (N-cadherin), Vimentin, E-cadherin, transforming growth factor-β (TGF-β), Ki-67, and Smad3 protein in glioma cells. Nude mouse tumor-bearing experiments were conducted to observe the effect of SIRT7 knockdown on glioma growth. Results Higher expression levels of SIRT7 gene were associated with poorer clinical prognosis (P<0.0001). SIRT7 expression levels were significantly correlated with tumor grading and 1p19q coding status (P<0.01). Compared with normal HA cells, glioma cells showed significantly increased SIRT7 expression levels (P<0.01). CCK-8 assay results indicated that, compared with control group, the proliferation activity of glioma cells in SIRT7 knockout group was significantly decreased (P<0.01), while SIRT7 overexpression group showed significantly increased proliferation activity (P<0.01). EdU assay results showed that, compared with control group, the proportion of glioma cells in the proliferative stage was significantly decreased in SIRT7 knockdown group (P<0.01), and significantly increased in SIRT7 overexpression group (P<0.01). Western blotting results revealed that, compared with control group, the protein expression levels of TGF-β, Smad3, N-cadherin and Vimentin were significantly decreased in SIRT7 knockdown group (P<0.01), while the expression level of E-cadherin protein was significantly increased (P<0.05). SIRT7 overexpression group showed significantly increased protein expression levels of TGF‑β, Smad3, N-cadherin and Vimentin (P<0.05), and a significantly decrease in E-cadherin protein expression level (P<0.05). Scratch assay results indicated that, compared with control group, the migration ability of cells in SIRT7 knockdown group and drug group was significantly decreased (P<0.01), and SIRT7 overexpression group showed significantly increased cell migration ability (P<0.05). Compared with drug group, drug+SIRT7 overexpression group exhibited significantly increased cell migration ability (P<0.01). Transwell assay results showed that, compared with control group, the migration and invasion abilities of cells in SIRT7 knockdown group and drug group were significantly decreased (P<0.01), and SIRT7 overexpression group exhibited significantly increased migration and invasion abilities (P<0.01). Compared with drug group, drug+SIRT7 overexpression group showed significantly increased migration and invasion abilities (P<0.01). Nude mouse tumor-bearing assay results indicated that the volume and weight of glioma in SIRT7 knockdown group were significantly reduced compared with control group (P<0.01). Conclusions Glioma patients with high SIRT7 expression have poorer clinical prognosis. SIRT7 can regulate the TGF-β/Smad3 pathway to mediate EMT, promoting the proliferation and migration of glioma cells. SIRT7 knockdown can inhibit the growth of transplanted gliomas in nude mice.

目的 观察组蛋白去乙酰化酶SIRT7在胶质瘤细胞中的表达情况及其对上皮-间质转化(EMT)的影响,探讨SIRT7对胶质瘤细胞增殖、迁移和侵袭能力的影响。方法 对癌症基因组图谱(TCGA)数据库和中国胶质瘤基因组图谱(CGGA)数据库中胶质瘤患者的信息进行生物信息学分析,探究SIRT7基因在胶质瘤中的表达情况及其与肿瘤分级、分子特征和患者临床预后之间的相关性。采用RT-qPCR和Western blotting检测正常HA细胞和胶质瘤细胞SIRT7表达水平。将胶质瘤细胞随机分为对照组、SIRT7敲低组、SIRT7过表达组、药物组(10 μmol/L氢氯噻嗪)和药物(10 μmol/L氢氯噻嗪)+SIRT7过表达组。采用CCK-8法、细胞划痕实验及Transwell实验观察上调及下调SIRT7表达对胶质瘤细胞增殖、迁移和侵袭的影响。采用RT-qPCR和Western blotting检测SIRT7对胶质瘤细胞中神经钙黏蛋白(N-cadherin)、波形蛋白(Vimentin)、E-钙黏蛋白(E-cadherin)、转化生长因子-β(TGF-β)、Ki-67、Smad3蛋白表达的影响。采用裸鼠荷瘤实验观察SIRT7敲低对胶质瘤生长的影响。结果 胶质瘤患者SIRT7基因高表达时临床预后更差(P<0.0001);SIRT7表达水平与肿瘤分级、1p19q编码状态明显相关(P<0.01)。与正常HA细胞比较,胶质瘤细胞SIRT7表达水平明显升高(P<0.01)。CCK-8法检测结果显示,与对照组比较,SIRT7敲低组胶质瘤细胞增殖活性明显降低(P<0.01),SIRT7过表达组胶质瘤细胞增殖活性明显增高(P<0.01)。EdU实验结果显示,与对照组比较,SIRT7敲低组胶质瘤细胞处于增殖期的比例明显降低(P<0.01),SIRT7过表达组胶质瘤细胞处于增殖期的占比明显增高(P<0.01)。Western blotting检测结果显示,与对照组比较,SIRT7敲低组TGF-β、Smad3、N-cadherin、Vimentin蛋白表达水平明显降低(P<0.01),E-cadherin蛋白表达水平明显增高(P<0.05);SIRT7过表达组TGF-β、Smad3、N-cadherin、Vimentin蛋白表达水平明显增高(P<0.05),E-cadherin蛋白表达水平明显降低(P<0.05)。划痕实验结果显示,与对照组比较,SIRT7敲低组和药物组细胞迁移能力明显降低(P<0.01),SIRT7过表达组细胞迁移能力明显增强(P<0.05);与药物组比较,药物+SIRT7过表达组细胞迁移能力明显增强(P<0.01)。Transwell实验结果显示,与对照组比较,SIRT7敲低组及药物组细胞的迁移和侵袭能力明显降低(P<0.01),SIRT7过表达组细胞的迁移和侵袭能力明显增强(P<0.01);与药物组比较,药物+SIRT7过表达组细胞的迁移和侵袭能力明显增强(P<0.01)。裸鼠荷瘤实验结果显示,SIRT7敲低后,裸鼠胶质瘤体积、重量明显减小或降低(P<0.01)。结论 高表达SIRT7的胶质瘤患者预后较差;SIRT7可调控TGF-β/Smad3通路介导的EMT,促进胶质瘤细胞的增殖和迁移;敲低SIRT7可抑制裸鼠移植胶质瘤的生长。

Objective To investigate the expression of the histone deacetylase SIRT7 in glioma cells and its impact on epithelial-mesenchymal transformation (EMT), as well as its effects on proliferative, migratory and invasive capabilities of glioma cells. Methods Bioinformatics analysis was conducted on data from glioma patients in the Cancer Genome Atlas (TCGA) and the Chinese glioma Genome Atlas (CGGA) databases to explore the expression of SIRT7 gene in gliomas and its correlation with tumor grading, molecular characteristics and patient clinical prognosis. Glioma cells were randomly divided into control, SIRT7 knockdown, SIRT7 overexpression, drug treatment (10 μmol/L hydrochlorothiazide) and drug (10 μmol/L hydrochlorothiazide)+SIRT7 overexpression groups. The CCK-8 assay, cell scratch assay and Transwell assay were used to observe the effects of upregulating and downregulating SIRT7 expression on glioma cell proliferation, migration and invasion. RT-qPCR and Western blotting were employed to detect the effects of SIRT7 on the expression of neural cadherin (N-cadherin), Vimentin, E-cadherin, transforming growth factor-β (TGF-β), Ki-67, and Smad3 protein in glioma cells. Nude mouse tumor-bearing experiments were conducted to observe the effect of SIRT7 knockdown on glioma growth. Results Higher expression levels of SIRT7 gene were associated with poorer clinical prognosis (P<0.0001). SIRT7 expression levels were significantly correlated with tumor grading and 1p19q coding status (P<0.01). Compared with normal HA cells, glioma cells showed significantly increased SIRT7 expression levels (P<0.01). CCK-8 assay results indicated that, compared with control group, the proliferation activity of glioma cells in SIRT7 knockout group was significantly decreased (P<0.01), while SIRT7 overexpression group showed significantly increased proliferation activity (P<0.01). EdU assay results showed that, compared with control group, the proportion of glioma cells in the proliferative stage was significantly decreased in SIRT7 knockdown group (P<0.01), and significantly increased in SIRT7 overexpression group (P<0.01). Western blotting results revealed that, compared with control group, the protein expression levels of TGF-β, Smad3, N-cadherin and Vimentin were significantly decreased in SIRT7 knockdown group (P<0.01), while the expression level of E-cadherin protein was significantly increased (P<0.05). SIRT7 overexpression group showed significantly increased protein expression levels of TGF‑β, Smad3, N-cadherin and Vimentin (P<0.05), and a significantly decrease in E-cadherin protein expression level (P<0.05). Scratch assay results indicated that, compared with control group, the migration ability of cells in SIRT7 knockdown group and drug group was significantly decreased (P<0.01), and SIRT7 overexpression group showed significantly increased cell migration ability (P<0.05). Compared with drug group, drug+SIRT7 overexpression group exhibited significantly increased cell migration ability (P<0.01). Transwell assay results showed that, compared with control group, the migration and invasion abilities of cells in SIRT7 knockdown group and drug group were significantly decreased (P<0.01), and SIRT7 overexpression group exhibited significantly increased migration and invasion abilities (P<0.01). Compared with drug group, drug+SIRT7 overexpression group showed significantly increased migration and invasion abilities (P<0.01). Nude mouse tumor-bearing assay results indicated that the volume and weight of glioma in SIRT7 knockdown group were significantly reduced compared with control group (P<0.01). Conclusions Glioma patients with high SIRT7 expression have poorer clinical prognosis. SIRT7 can regulate the TGF-β/Smad3 pathway to mediate EMT, promoting the proliferation and migration of glioma cells. SIRT7 knockdown can inhibit the growth of transplanted gliomas in nude mice.

胶质瘤是常见的原发性颅内肿瘤,其发病率、复发率、病死率均较高。流行病学调查显示,脑胶质瘤的年发病率为(5~8)/10万,约占全部颅脑肿瘤的2/5,所有恶性脑肿瘤的4/5,其5年病死率在全身肿瘤中高居第3位[1]。目前脑胶质瘤的治疗以手术为主,辅以放疗、化疗。胶质瘤细胞常呈浸润性生长,大部分胶质瘤难以被彻底切除,残余的肿瘤细胞成为复发的根源[2];胶质瘤的新生血管多、血管异型性显著,易发生转移。去乙酰化酶Sirtuins家族是NAD+依赖的组蛋白去乙酰化酶,在细胞的各种生命活动中发挥关键作用[3],已知的人类Sirtuins家族成员有7个(Sirtuins1-7,SIRT1-7)[4],其中SIRT7在核仁中富集,可促进RNA聚合酶Ⅰ依赖的核糖体RNA(ribosomal,rRNA)的转录[5]。文献报道SIRT7与黑色素瘤、结肠癌的细胞增殖和致癌活性有关[6-7],但其在胶质瘤中的作用鲜有报道。
上皮‑间质转化(epithelial-mesenchymal transition,EMT)是恶性肿瘤细胞获得迁移和侵袭能力的重要生物学过程[8]。识别靶向EMT的关键分子有助于调控EMT过程,而探索基于EMT的诊断方法及治疗手段是当前肿瘤转移机制研究的关键问题。本研究分析胶质瘤中SIRT7的表达情况,观察SIRT7在胶质瘤细胞中的表达及其对EMT过程的影响,探讨其表达变化对胶质瘤细胞增殖、迁移和侵袭能力的影响,以期为胶质瘤的分子机制研究提供证据。
HA细胞(人星形胶质细胞)和U251、LN229细胞(人胶质瘤细胞)购自武汉普诺赛生命科技有限公司,由解放军总医院第七医学中心脑科学实验室冻存;sh-SIRT7慢病毒购自上海吉凯基因医学科技股份有限公司;cDNA-SIRT7慢病毒购自上海和元生物技术股份有限公司;DMEM培养基、胎牛血清和青霉素-链霉素混合液购自美国Gibco公司;CCK-8检测试剂盒购自北京索莱宝科技有限公司;EdU-594细胞增殖检测试剂盒、蛋白质提取试剂盒和BCA蛋白质浓度检测试剂盒购自上海碧云天生物技术有限公司;Transwell小室、Matrigengel基质胶购自美国康宁公司;RNA提取试剂盒购自美国赛默飞世尔科技公司;抗SIRT7、Ki-67、甘油醛-3-磷酸脱氢酶(GAPDH)、转化生长因子-β(transforming growth factor-β,TGF-β)、Smad3、E-钙黏蛋白(E-cadherin)、神经钙黏蛋白(N-cadherin)、波形蛋白(Vimentin)抗体均购自美国CST公司;辣根过氧化物酶(HRP)-山羊抗兔重组二抗购自武汉三鹰生物技术有限公司。6只5周龄雄性BALB/c裸鼠购自北京斯贝福生物技术有限公司[实验动物生产许可证号:SCXK(京)2019-0004]。
胶质瘤患者的基因表达和临床数据来自中国胶质瘤基因组图谱(CGGA,http://www.cgga.org.cn)数据库和癌症基因组图谱(TCGA,https://www.cancer.gov/ccg/research/genome-sequencing/tcga)数据库。从TCGA数据库获取了702例胶质瘤患者的RNA-seq和临床数据信息,从CGGA数据库获取了325例胶质瘤患者的RNA-seq和临床数据信息。本研究符合《赫尔辛基宣言》,并获解放军总医院伦理委员会授权。确保两个公共数据库信息均获得患者的知情同意。分析胶质瘤患者SIRT7基因表达水平与典型预后特征(如世界卫生组织分类、IDH野生型/突变型、1p/19q编码/非编码、MGMT甲基化/未甲基化)之间的相关性,以及SIRT7表达水平与患者预后之间的关系。
所有细胞均使用含10%胎牛血清和1%青霉素-链霉素的DMEM培养基培养,培养条件为37 ℃、5% CO2。将U251和LN229细胞随机分为对照组、SIRT7敲低组、SIRT7过表达组、药物组(10 μmol/L氢氯噻嗪)和药物(10 μmol/L氢氯噻嗪)+SIRT7过表达组。氢氯噻嗪是一种可抑制TGF-β/Smad3信号通路的药物[9]
由上海吉凯基因医学科技股份有限公司将sh-RNA质粒克隆到pMD2G和psPAX2载体质粒中,构建sh-SIRT7慢病毒;用sh-SIRT7慢病毒感染U251和LN229细胞,为SIRT7敲低组。将SIRT7编码区亚克隆到pcDNA3.1载体中,构建cDNA-SIRT7慢病毒;用cDNA-SIRT7慢病毒感染U251和LN229细胞,为SIRT7过表达组。为了便于观察,在慢病毒的质粒系统中添加绿色荧光蛋白(GFP)。待U251和LN229细胞在培养皿中生长至30%~50%融合时,根据试剂盒说明书进行慢病毒感染,37 ℃下培养48 h,更换新的培养基继续培养24 h,加入8 μg/ml嘌呤霉素筛选稳转株用于后续实验。
使用Trizol试剂从细胞中提取RNA。使用All-in-OneTM miRNA第一链反转录试剂盒2.0将RNA反转录为cDNA,然后在LightCycler480系统(瑞士罗氏)上使用2×Color SYBR Green qPCR Master Mix进行定量分析。采用GAPDH作为管家基因对SIRT7的表达进行归一化。引物序列:SIRT7,5'-AGCACGGCAGCGTCTATC-3'(正向),5'-CATGTGGGTGAGGGTTGG-3'(反向);GAPDH,5'-TCCACATGAAGTGTGACGT-3'(正向),5'-TACTCCTGCTTGCTGATCCAC-3'(反向);E-cadherin, 5'-GCCTCCTGAAAAGAGAGTGGAAG-3'(正向),5'-TGGCAGTGTCTCTCCAAATCCG-3'(反向);N-cadherin,5'-CCTCCAGAGTTTACTGCCATGAC-3'(正向),5'-GTAGGATCTCCGCCACTGATTC-3'(反向);Vimentin,5'-GACGCCATCAACACCGAGTT-3'(正向),5'-CTTTGTCGTTGGTTAGCTGGT-3'(反向);TGF-β,5'-TACCTGAACCCGTGTTGCTCTC-3'(正向)和5'-GTTGCTGAGGTATCGCCAGGAA-3'(反向);Smad3,5'-ACTACAGCCATTCCATCCC-3'(正向),5'-TAGGTTTGGAGAACCTGCG-3'(反向)。
用RIPA提取液和蛋白酶抑制剂混合液裂解细胞,BCA蛋白检测试剂盒测定蛋白浓度。用SDS-PAGE凝胶电泳分离30 μg蛋白,然后转移到PVDF膜上,5% BSA封闭后,用1:1000的Sirt7、TGF-β、Smad3、E-cadherin、N-cadherin、Vimentin和GAPDH一抗稀释液在4 ℃下孵育过夜。次日,TBST溶液清洗,室温下用1:5000的HRP标记的山羊抗兔二抗稀释液孵育2 h。TBST清洗,用超敏化学发光底物(Clarity Max Western ECL,美国Bio-Rad)对膜进行成像,并通过Quantity One软件(美国Bio-Rad)对蛋白条带进行定量分析。
采取CCK-8实验检测细胞的生长活力。将胶质瘤细胞平铺于96孔板中,每孔加入10 μl CCK-8溶液并培养2 h。然后分别在加入CCK-8 溶液后24、48、72、96 h测量光密度值。
提前1 d将U251和LN229细胞种植在24孔板中,按照EdU-594细胞增殖检测试剂盒说明书加入预热的EdU工作液,37 °C孵育2 h进行EdU标记,再用4%多聚甲醛溶液固定。随后加入500 μl Click反应液,室温避光孵育30 min。最后加入1 ml Hoechst 33342溶液,室温避光孵育10 min。荧光显微镜下观察,计算各组处于增殖期的细胞所占百分比。
在6孔板中铺入约5×105个胶质瘤细胞,24 h后融合率接近100%。用无菌枪头以垂直细胞平面的方向从上往下在细胞间划出一道直线,划痕完成后,使用无菌PBS洗3次,将脱落的细胞洗掉,在显微镜下拍照后更换新鲜无血清培养基。将细胞放入37 ℃、5% CO2培养箱培养,在24、48 h节点取出细胞,在显微镜下观察拍照,使用ImageJ软件测量划痕面积。以对照组的面积变化为标准,对其他4组的面积变化进行标准化处理。
实验前1 d预先将生长状态良好的胶质瘤细胞用无血清培养基进行饥饿处理。第2天在冰上将基质胶用无血清培养基稀释至1 mg/ml,使用预冷的吸头混合至均匀状态。取60 μl基质胶垂直加入Transwell小室中,使其均匀平铺在底部,随后置于37 ℃、5% CO2培养箱孵育3 h,弃掉未结合的基质胶(如迁移实验则不需要这一步)。加入100 μl不含血清的培养液,将培养板置于37 ℃培养箱孵育30 min,进行水化。每个小室内加入1×105个细胞并用200 μl无血清培养基进行培养,在小室所在的24孔板内加入600 μl含10%胎牛血清的完全培养基,置于37 ℃、5% CO2培养箱内培养24 h。取出Transwell小室,去除培养液,用PBS浸湿的棉签去除小室的内基质胶和细胞。用4%多聚甲醛固定液固定30 min,以PBS洗涤后放入结晶紫染色液中染色20 min,PBS洗3次,最后在显微镜下观察,采用ImageJ软件进行细胞计数。
采用随机数字表法将6只5周龄雄性BALB/c裸鼠分为对照组与SIRT7敲低组,每组3只。每只裸鼠背部皮下注射5×106个胶质瘤U251细胞。每周测量裸鼠背部肿瘤体积,35 d后取出肿瘤并称重。本研究动物实验方案经解放军总医院动物保护与使用委员会审批(2018-66)。
用4%多聚甲醛溶液固定自裸鼠背部取出的胶质瘤组织,石蜡包埋,4 μm厚度切片,室温下用二甲苯脱蜡,无水乙醇脱水。用柠檬酸钠溶液在98 ℃热锅中修复抗原15 min,室温下加入山羊血清封闭剂阻断非特异性位点。加入抗SIRT7、Ki-67抗体,4 ℃过夜。第2天,切片在室温下放置30 min后洗去抗体,加入50 μl链霉素过氧化物酶溶液,室温下孵育10 min后洗去。在每张胶片上滴加100 μl DAB溶液,在显微镜下观察10 min。染色结束,酌情冲洗。用苏木精重染2 min后冲洗,切片干燥后镜下观察。
采用GraphPad Prism软件进行统计分析。所有实验至少独立重复3次。计量数据均符合正态分布,以$\bar{x}±s$表示,两组间比较采用t检验,多组间比较采用单因素方差分析,进一步两两比较采用LSD-t检验。P<0.05为差异有统计学意义。
利用TCGA和CGGA数据库中胶质瘤患者的数据进行生物信息学分析(图1A),将SIRT7的表达、是否甲基化(MGMTp)、1p/19q是否编码、IDH突变状态、年龄、性别和肿瘤分级等纳入分析,结果显示,与SIRT7高表达的患者比较,SIRT7低表达的患者临床预后较好(P<0.0001)(图1B)。在TCGA数据库患者队列中,SIRT7表达水平与肿瘤分级(P<0.0001)、1p19q编码状态(P<0.01)明显相关;而在CGGA数据库患者队列中,SIRT7表达水平与肿瘤分级、MGMTp、1p19q编码状态明显相关(P<0.0001)(图1C)。
(1)与正常HA细胞比较,U251和LN229细胞中SIRT7 mRNA及蛋白表达水平明显升高(P<0.01,图2A);用含sh-SIRT7基因片段的慢病毒感染的U251和LN229细胞中SIRT7 mRNA及蛋白质表达水平明显降低(P<0.01,图2B)。
(2)CCK-8法检测结果显示,与对照组比较,SIRT7敲低组细胞在24、48、72、96 h的OD值均明显降低(U251细胞:0.153 vs. 0.673,0.372 vs. 1.156,0.684 vs. 1.794,0.863 vs. 2.177,P<0.01;LN229细胞:0.125 vs. 0.454,0.264 vs. 0.765,0.462 vs. 1.186,0.697 vs. 1.479,P<0.01)(图2C)。
(3)EdU实验结果显示,与对照组比较,SIRT7敲低组处于增殖期的细胞比例明显降低(U251细胞:21% vs. 62%,P<0.05;LN229细胞:24% vs. 60%,P<0.01,图2D)。
(4)划痕实验结果显示,SIRT7敲低组U251和LN229细胞在24、48 h的相对迁移率分别为对照组的49%、66%和51%、52%,均明显低于对照组(P<0.01,图3A)。
(5)Transwell实验结果显示,与对照组比较,SIRT7敲低组U251和LN229细胞迁移率均明显降低(32% vs. 76%,34% vs. 79%,P<0.01),细胞侵袭能力也明显降低(21% vs. 48%,28% vs. 53%,P<0.01,图3B)。
(6)RT-qPCR检测结果显示,与对照组比较,SIRT7敲低组TGF-βSmad3N-cadherinVimentin mRNA表达水平均明显降低(P<0.05或P<0.01),而E-cadherin mRNA表达水平明显升高(P<0.05或P<0.01);Western blotting 检测结果显示,与对照组比较,SIRT7敲低组TGF-β、Smad3、N-cadherin、Vimentin蛋白表达水平均明显降低(P<0.01),而E-cadherin蛋白表达水平明显升高(P<0.01,图3C)。
(1)用含有cDNA-SIRT7基因片段的慢病毒感染U251和LN229细胞,SIRT7 mRNA和蛋白表达水平均明显高于对照组(P<0.01,图4A)。
(2)CCK-8法检测结果显示,与对照组比较,SIRT7过表达组胶质瘤细胞在24、48、72、96 h的OD值均明显增高(U251细胞:0.613 vs. 0.225,1.128 vs. 0.630,1.589 vs. 0.922,2.194 vs. 1.332,P<0.01;LN229细胞:0.653 vs. 0.324,1.123 vs. 0.632,1.689 vs. 0.923,2.214 vs. 1.334,P<0.01,图4B)。
(3)EdU实验结果显示,与对照组比较,SIRT7过表达组处于增殖期的细胞占比明显增高(U251细胞:84% vs. 56%,P<0.01;LN229细胞:76% vs. 50%,P<0.01,图4C)。
(4)划痕实验结果显示,SIRT7过表达组U251和LN229细胞在24、48 h的相对迁移率分别为对照组的150%、152%和153%、170%,均明显高于对照组 (P<0.05,图5A)。
(5)Transwell迁移和侵袭实验结果显示,与对照组比较,SIRT7过表达组U251和LN229细胞迁移率均明显增高(83% vs. 66%,80% vs. 60%,P<0.01),细胞侵袭能力也明显增高(66% vs. 48%,69% vs. 53%,P<0.01,图5B)。
(6)RT-qPCR检测结果显示,与对照组比较,SIRT7过表达组TGF‑βSmad3N-cadherinVimentin mRNA表达水平均明显升高(P<0.05或P<0.01),而E-cadherin mRNA表达水平明显降低(P<0.05或P<0.01);Western blotting 检测结果显示,与对照组比较,SIRT7过表达组TGF‑β、Smad3、N-cadherin、Vimentin蛋白表达水平明显升高(P<0.01),E-cadherin蛋白表达水平明显降低(P<0.01,图5C)。
(1)划痕实验结果显示,与对照组比较,在24、48 h,药物组细胞相对迁移率均明显降低(U251细胞:59% vs. 100%,50% vs. 100%,P<0.01;LN229细胞:61% vs. 100%,51% vs. 100%,P<0.01);SIRT7过表达组细胞相对迁移率均明显增高(U251细胞:138% vs. 100%,149% vs. 100%,P<0.01;LN229细胞:144% vs. 100%,149% vs. 100%,P<0.01)。与药物组比较,药物+SIRT7过表达组相对迁移率明显增高(U251细胞:87% vs. 59% ,80% vs. 50%,P<0.01;LN229细胞:91% vs. 61%,84% vs. 51%,P<0.01,图6A)。
(2)Transwell迁移实验结果显示,与对照组比较,SIRT7过表达组细胞迁移率明显增高(U251细胞:66% vs. 51%,P<0.01;LN229细胞:58% vs. 44%,P<0.01),药物组细胞侵袭能力明显降低(U251细胞:28% vs. 51%,P<0.01;LN229细胞:25% vs. 44%,P<0.01)。与药物组比较,药物+SIRT7过表达组细胞穿过多孔膜的比例明显增高(U251细胞:42% vs. 28%,P<0.01;LN229细胞:40% vs. 25%,P<0.01,图6B)。Transwell侵袭实验结果显示,与对照组比较,SIRT7过表达组细胞穿过基质胶和多孔膜的比例明显增高(U251细胞:56% vs. 37%,P<0.01;LN229细胞:51% vs. 40%,P<0.01),药物组细胞穿过基质胶和多孔膜的比例明显降低(U251细胞:17% vs. 37%,P<0.01;LN229细胞:27% vs. 40%,P<0.01)。与药物组比较,药物+SIRT7过表达组细胞穿过基质胶和多孔膜的比例明显增高(U251细胞:32% vs. 17%,P<0.01;LN229细胞:51% vs. 27%,P<0.05,图6C)。
采用U251细胞在裸鼠皮下构建异种移植胶质瘤模型,免疫组织化学染色结果显示,与对照组比较,SIRT7敲低组的SIRT7染色阳性细胞明显减少,且Ki-67着色明显减少,差异均有统计学意义(P<0.05,图7A、B)。与对照组比较,SIRT7敲低组裸鼠21、28、35 d的肿瘤体积均明显缩小(162 mm3 vs. 248 mm3,267 mm3 vs. 450 mm3,471mm3 vs. 760 mm3P<0.01);第35天时的肿瘤重量明显降低(0.72 g vs. 1.12 g,P<0.01,图7C、D)。
随着疾病研究从细胞水平转向分子水平,越来越多的分子生物标志物被发现。这些生物标志物不仅可提高疾病诊断和预后预测的准确性,还能促进对疾病发生发展过程的认识。脑胶质瘤是起源于神经胶质细胞的常见的原发性颅内肿瘤,传统的治疗方式以手术为主,辅以放化疗。由于胶质瘤浸润性生长的特点,手术往往难以完全切除,因此化疗成为常见的后续治疗手段[10-11];目前使用较多的化疗药物是替莫唑胺,但多数胶质瘤患者的治疗效果并不理想,复发率和耐药率较高。基于此,对胶质瘤治疗的探索也逐渐扩大到肿瘤分子标志物,越来越多的分子标志物被认识,同时更多的分子靶向方案被提出。
SIRTs是一类进化上高度保守的NAD+依赖性组蛋白去乙酰化酶,可通过调节多种关键信号通路参与一系列生物过程[12]。目前已知的人类SIRT家族成员有7个,分别以SIRT1~7命名。大量证据显示,SIRT蛋白家族可广泛参与多种肿瘤的发生发展过程,并可通过调节机体代谢及炎症反应影响肿瘤细胞的增殖、凋亡、自噬、迁移和侵袭等多种过程[13]。SIRT1~6在胶质瘤中的作用已有文献报道,但SIRT7在胶质瘤中的作用鲜见报道。目前仅有一篇报道指出抑制SIRT7的表达可抑制信号转导和转录激活蛋白3(signal transducer and activator of transcription,STAT3)的磷酸化,并抑制磷酸化胞外信号调节激酶(phosphorylated extracellular signal-regulated kinase,p-ERK)的激活,同时降低胶质瘤细胞中细胞周期蛋白依赖性激酶2的表达[14]。本研究结果显示,SIRT7在胶质瘤细胞中高度富集,SIRT7基因表达量高的胶质瘤患者临床预后较差;降低SIRT7表达可使胶质瘤细胞的增殖、迁移和侵袭能力下降。
癌细胞的扩散和转移是临床面临的一大难题。癌细胞发生转移时离开原发肿瘤区域,随着血液循环到达远处器官,这一过程常依赖EMT[15]。EMT是指黏附性强的上皮细胞通过特定程序,转化为黏附性差且具有间质表型细胞的生物学过程;上皮细胞与基底膜的连接松动,使肿瘤细胞获得较高的迁移和侵袭能力[8]。在EMT过程中,细胞黏附分子如E-cadherin表达减少,细胞骨架形态向波形蛋白形态转变,同时具有间充质细胞特征的N-cadherin表达增加[16]。EMT主要涉及3个信号通路:TGF-β信号通路、Wnt信号通路和Notch信号通路[17-19]。TGF-β被认为是诱导肿瘤细胞完成EMT过程最重要的因子。在一些细胞实验中,仅添加TGF-β即可诱导EMT的发生[20]。TGF-β介导EMT主要通过经典的Smad通路实现[21]。在TGF-β/Smad经典通路中,TGF-β信号激活Smad2和Smad3,并与Smad4结合形成Smad复合物,进而转移到细胞核内与转录因子一起介导靶基因的抑制或激活,也可在细胞核内诱导miRNA的表达,从而抑制上皮细胞的标志性蛋白表达,促进间充质细胞特性的蛋白表达[22]
EMT是肿瘤细胞获得迁移和侵袭能力的重要生物学过程,而TGF-β/Smad3通路是EMT过程中的经典通路。EMT使原发肿瘤细胞失去极性,转化为具有迁移和侵袭能力的间质细胞,许多转录因子、关键蛋白、细胞表面受体参与了这一过程,从细胞和分子水平上共同促进EMT[23-24]。因此,要干扰肿瘤细胞的转移就离不开对EMT过程的研究。TGF-β是EMT的诱导因子之一,也是目前研究最为清楚的信号通路。TGF‐β信号通路包括经典与非经典Smad通路,在经典Smad信号通路中,TGF‐β先与细胞膜表面的TGF‐β受体结合,进一步激活Smads(主要为Smad2和Smad3)磷酸化,磷酸化的Smad2和Smad3可与Smad4形成复合物,进入细胞核中调控靶基因的表达[25]。本研究发现,SIRT7可通过调节TGF-β/Smad3通路介导胶质瘤细胞的EMT过程,进而影响胶质瘤细胞的迁移和侵袭能力。
本研究仍存在一定的局限性,只验证了SIRT7可通过调节TGF-β/Smad3通路影响胶质瘤细胞的EMT过程,但未揭示相关的调节机制。后续研究需要进一步探究SIRT7调节TGF-β/Smad3通路的具体机制,以期为胶质瘤的诊断和治疗提供新靶点,为胶质瘤的分子机制研究提供新思路。
