Objective To investigate the protective effects of secretomes released by three-dimensional cultured mesenchymal stem cells (MSCs) on neurons subjected to seawater immersion (SW) and stretch injury (SI), and to provide new insights into neuronal repair following SW combined with traumatic brain injury (TBI). Methods MSCs were cultured using the hanging drop method, and the conditioned medium (CM) containing MSCs secretomes was collected. A cellular model combining SW with SI was established using mouse hippocampal neuronal cells (HT22 cells). HT22 cells were randomly assigned to five groups: control, SI, SI+SW, SI+CM, and SI+SW+CM groups. Cell viability was assessed using the CCK-8 assay, apoptosis rate was measured by flow cytometry, cell migration ability was evaluated by scratch assay, and the expression levels of apoptosis-related proteins Bcl-2 and Bcl-2-associated protein (Bax), and ferroptosis-related proteins long-chain acyl-CoA synthetase 4 (ACSL4) and cyclooxygenase-2 (COX-2) were detected by Western blotting. Results Immersion in 15% seawater for 12 h significantly decreased HT22 cell viability (P<0.05). The CCK-8 assay indicated that cell viability in both the SI and SI+SW groups was significantly lower than that in control group after 12 h of treatment (P<0.05). Treatment with CM containing MSCs secretomes significantly increased cell viability in SI+CM group compared to SI group (P<0.0001), and in SI+SW+CM group compared to SI+SW group (P<0.001). Flow cytometry results revealed that the apoptosis rate in SI and SI+SW groups was significantly higher than that in control group (P<0.05 or P<0.001), while in SI+CM group was lower than that in SI group (P<0.05), and in SI+SW+CM group was lower than that in SI+SW group (P<0.05). Western blotting showed that compared to control group, SI and SI+SW groups exhibited reduced Bcl-2 expression level (P<0.01 or P<0.0001) and increased expression levels of Bax, ACSL4, and COX-2 (P<0.01 or P<0.0001). Compared to SI group, the SI+CM group displayed increased Bcl-2 expression level (P<0.05) and decreased expression levels of Bax, ACSL4, and COX-2 (P<0.05). Compared to SI+SW group, SI+SW+CM group exhibited increased Bcl-2 expression level (P<0.01) and decreased expression levels of Bax, ACSL4, and COX-2 (P<0.01 or P<0.001). Scratch assay results demonstrated that at both 12 h and 24 h, the cell migration rate in SI and SI+SW groups was significantly lower than that in control group (P<0.01 or P<0.0001), while the migration rate in SI+CM group was significantly higher than that in SI group (P<0.0001 or P<0.01), and the migration rate in SI+SW+CM group was significantly higher than that in SI+SW group (P<0.0001). Conclusion Secretomes derived from MSCs cultured using the hanging drop method can alleviate neuronal damage caused by SW and TBI, potentially offering a therapeutic approach for SW combined with TBI.

目的 探究三维培养的间充质干细胞(MSCs)来源的分泌组物对海水浸泡(SW)合并拉伸损伤(SI)小鼠海马神经元的保护作用,为SW合并创伤性脑损伤(TBI)后的神经元修复提供新思路。方法 采用悬滴法培养MSCs,收集含有MSCs来源的分泌组物的条件培养基(CM)。采用小鼠海马神经元(HT22细胞)建立SW合并SI细胞模型。将HT22细胞随机分为对照组、SI组、SI+SW组、SI+CM组与SI+SW+CM组。采用CCK-8法检测各组细胞活力,流式细胞术检测细胞凋亡率,细胞划痕实验检测细胞迁移能力,Western blotting检测凋亡相关蛋白Bcl-2、Bcl-2关联X蛋白(Bax)、铁死亡相关蛋白长链酰基辅酶A合酶4(ACSL4)、环氧合酶-2(COX-2)的表达水平。结果 15%浓度海水浸泡12 h后,HT22细胞活力明显下降(P<0.05)。CCK-8法检测结果显示,处理12 h后,与对照组比较,SI组和SI+SW组HT22细胞活力均明显下降(P<0.0001);经MSCs分泌组物的CM处理后,SI+CM组细胞活力明显高于SI组(P<0.0001),且SW+SI+CM组细胞活力明显高于SI+SW组(P<0.001)。流式细胞术检测结果显示,SI组和SI+SW组细胞凋亡率明显高于对照组(P<0.05或P<0.001),而SI+CM组细胞凋亡率明显低于SI组(P<0.05),SI+SW+CM组细胞凋亡率明显低于SI+SW组(P<0.05)。Western blotting检测结果显示,与对照组比较,SI组和SI+SW组Bcl-2表达水平明显降低(P<0.01或P<0.0001),Bax、ACSL4和COX-2表达水平明显升高(P<0.01或P<0.001或P<0.0001);与SI组比较,SI+CM组Bcl-2蛋白表达水平明显升高(P<0.05),而Bax、ACSL4和COX-2蛋白表达水平明显降低(P<0.05);与SI+SW组比较,SI+SW+CM组Bcl-2表达水平明显升高(P<0.01),Bax、ACSL4和COX-2蛋白表达水平明显降低(P<0.01或P<0.001)。细胞划痕实验结果显示,12 h和24 h时,SI组和SI+SW组细胞迁移率明显低于对照组(P<0.01或P<0.0001),而SI+CM组细胞迁移率明显高于SI组(P<0.0001或P<0.01),SI+SW+CM组细胞迁移率明显高于SI+SW组 (P<0.0001)。结论 悬滴法培养的MSCs分泌组物可减轻SW合并SI神经元的损伤,有望成为SW合并TBI的潜在治疗措施。

目的 探究三维培养的间充质干细胞(MSCs)来源的分泌组物对海水浸泡(SW)合并拉伸损伤(SI)小鼠海马神经元的保护作用,为SW合并创伤性脑损伤(TBI)后的神经元修复提供新思路。方法 采用悬滴法培养MSCs,收集含有MSCs来源的分泌组物的条件培养基(CM)。采用小鼠海马神经元(HT22细胞)建立SW合并SI细胞模型。将HT22细胞随机分为对照组、SI组、SI+SW组、SI+CM组与SI+SW+CM组。采用CCK-8法检测各组细胞活力,流式细胞术检测细胞凋亡率,细胞划痕实验检测细胞迁移能力,Western blotting检测凋亡相关蛋白Bcl-2、Bcl-2关联X蛋白(Bax)、铁死亡相关蛋白长链酰基辅酶A合酶4(ACSL4)、环氧合酶-2(COX-2)的表达水平。结果 15%浓度海水浸泡12 h后,HT22细胞活力明显下降(P<0.05)。CCK-8法检测结果显示,处理12 h后,与对照组比较,SI组和SI+SW组HT22细胞活力均明显下降(P<0.0001);经MSCs分泌组物的CM处理后,SI+CM组细胞活力明显高于SI组(P<0.0001),且SW+SI+CM组细胞活力明显高于SI+SW组(P<0.001)。流式细胞术检测结果显示,SI组和SI+SW组细胞凋亡率明显高于对照组(P<0.05或P<0.001),而SI+CM组细胞凋亡率明显低于SI组(P<0.05),SI+SW+CM组细胞凋亡率明显低于SI+SW组(P<0.05)。Western blotting检测结果显示,与对照组比较,SI组和SI+SW组Bcl-2表达水平明显降低(P<0.01或P<0.0001),Bax、ACSL4和COX-2表达水平明显升高(P<0.01或P<0.001或P<0.0001);与SI组比较,SI+CM组Bcl-2蛋白表达水平明显升高(P<0.05),而Bax、ACSL4和COX-2蛋白表达水平明显降低(P<0.05);与SI+SW组比较,SI+SW+CM组Bcl-2表达水平明显升高(P<0.01),Bax、ACSL4和COX-2蛋白表达水平明显降低(P<0.01或P<0.001)。细胞划痕实验结果显示,12 h和24 h时,SI组和SI+SW组细胞迁移率明显低于对照组(P<0.01或P<0.0001),而SI+CM组细胞迁移率明显高于SI组(P<0.0001或P<0.01),SI+SW+CM组细胞迁移率明显高于SI+SW组 (P<0.0001)。结论 悬滴法培养的MSCs分泌组物可减轻SW合并SI神经元的损伤,有望成为SW合并TBI的潜在治疗措施。

Objective To investigate the protective effects of secretomes released by three-dimensional cultured mesenchymal stem cells (MSCs) on neurons subjected to seawater immersion (SW) and stretch injury (SI), and to provide new insights into neuronal repair following SW combined with traumatic brain injury (TBI). Methods MSCs were cultured using the hanging drop method, and the conditioned medium (CM) containing MSCs secretomes was collected. A cellular model combining SW with SI was established using mouse hippocampal neuronal cells (HT22 cells). HT22 cells were randomly assigned to five groups: control, SI, SI+SW, SI+CM, and SI+SW+CM groups. Cell viability was assessed using the CCK-8 assay, apoptosis rate was measured by flow cytometry, cell migration ability was evaluated by scratch assay, and the expression levels of apoptosis-related proteins Bcl-2 and Bcl-2-associated protein (Bax), and ferroptosis-related proteins long-chain acyl-CoA synthetase 4 (ACSL4) and cyclooxygenase-2 (COX-2) were detected by Western blotting. Results Immersion in 15% seawater for 12 h significantly decreased HT22 cell viability (P<0.05). The CCK-8 assay indicated that cell viability in both the SI and SI+SW groups was significantly lower than that in control group after 12 h of treatment (P<0.05). Treatment with CM containing MSCs secretomes significantly increased cell viability in SI+CM group compared to SI group (P<0.0001), and in SI+SW+CM group compared to SI+SW group (P<0.001). Flow cytometry results revealed that the apoptosis rate in SI and SI+SW groups was significantly higher than that in control group (P<0.05 or P<0.001), while in SI+CM group was lower than that in SI group (P<0.05), and in SI+SW+CM group was lower than that in SI+SW group (P<0.05). Western blotting showed that compared to control group, SI and SI+SW groups exhibited reduced Bcl-2 expression level (P<0.01 or P<0.0001) and increased expression levels of Bax, ACSL4, and COX-2 (P<0.01 or P<0.0001). Compared to SI group, the SI+CM group displayed increased Bcl-2 expression level (P<0.05) and decreased expression levels of Bax, ACSL4, and COX-2 (P<0.05). Compared to SI+SW group, SI+SW+CM group exhibited increased Bcl-2 expression level (P<0.01) and decreased expression levels of Bax, ACSL4, and COX-2 (P<0.01 or P<0.001). Scratch assay results demonstrated that at both 12 h and 24 h, the cell migration rate in SI and SI+SW groups was significantly lower than that in control group (P<0.01 or P<0.0001), while the migration rate in SI+CM group was significantly higher than that in SI group (P<0.0001 or P<0.01), and the migration rate in SI+SW+CM group was significantly higher than that in SI+SW group (P<0.0001). Conclusion Secretomes derived from MSCs cultured using the hanging drop method can alleviate neuronal damage caused by SW and TBI, potentially offering a therapeutic approach for SW combined with TBI.

创伤性脑损伤(traumatic brain injury,TBI)是指由外部物理力量引发的脑功能紊乱或脑部病变,全球每年影响超5000万人,半数人口一生中或遭遇一次以上[1-2]。因海水的理化性质(低温、高渗、高盐、碱性及可能的高细菌量等)可能加剧损伤,TBI合并海水浸泡(seawater immersion,SW)则更为复杂,致使救治难度明显增加[3-5]。随着人类海洋活动的增多和TBI风险的上升,因救治受限,导致TBI的残疾率和病死率增高,海上作业者安全保障需求增加[6-8]。研究显示,SW可加重TBI的后果,动物实验亦显示其具有明显危害[6,9]。TBI的现有治疗方案多为缓解症状,对神经损伤的疗效有限,探索更有效的治疗方法迫在眉睫[10]。间充质干细胞(mesenchymal stem cells,MSCs)的旁分泌功能使其具有潜在的修复神经系统损伤的能力,且其分泌组物作为天然保护剂对细胞应激与损伤具有明显的保护作用[11-12]。3D培养的MSCs球体能增强分泌组物的释放,提升治疗效果[13-15],但其对SW合并TBI的作用尚不清楚。本研究建立模拟SW与拉伸损伤(stretch injury,SI)的小鼠海马神经元模型,并用含3D MSCs分泌组物的条件培养基(conditional medium,CM)治疗,探究其可能的保护作用,旨在为TBI治疗研究提供新的策略。
细胞损伤控制仪购自世联博研(北京)科技有限公司;酶标仪购自瑞士TECAN公司;倒置荧光显微镜购自德国Carl Zeiss公司;离心机购自山东博科控股集团有限公司;流式细胞仪、PE偶联Annexin-V凋亡检测试剂盒购自美国BD公司;KCl、CaCl2、NaCl、MgSO4、MgCl2、NaBr、NaHCO3(分析纯)、0.25% TE购自美国Sigma公司;MEM-α培养基、DMEM高糖培养基购自美国Gibco公司;胎牛血清(FBS)购自以色列BI公司;CCK-8试剂购自北京索莱宝科技有限公司;抗Bcl-2蛋白、Bcl-2关联X蛋白(Bcl-2-associated X protein,Bax)、长链酰基辅酶A合酶4(long-chain acyl-CoA synthetase 4,ACSL4)、环氧合酶-2(cyclooxygenase-2,COX-2)、β-肌动蛋白(β-actin)抗体购自英国Abcam公司;辣根过氧化物酶(HRP)-山羊抗兔二抗购自美国Jackson Immuno-Research公司。
小鼠海马神经元细胞系(HT22)由军事医学研究院军事认知与脑科学研究所提供;脐带血来源的MSCs由解放军总医院第六医学中心儿科实验室馈赠。
HT22细胞在37 ℃水浴锅中解冻,然后1000 r/min离心3 min,弃上清,用含10% FBS和1%青霉素-链霉素的DMEM完全培养基重悬,用移液枪接种至细胞培养皿中,并补充完全培养基至10 ml,置于37 ℃、5% CO2的恒温培养箱中孵育,待细胞丰度长至70%~90%时即可处理细胞。
将2D培养的MSCs用0.25%胰酶消化至细胞变圆,加入适量的MSCs完全培养基中和、停止消化,轻轻吹打混匀,将细胞上清转移至离心管中,离心,计数,配制成含8000个细胞的细胞悬液。用移液枪将细胞悬液滴至细胞培养皿盖内侧,50 μl/滴,在培养皿中加入适量PBS防止蒸发,没过皿底即可。翻转皿盖盖于细胞皿上,置于细胞培养箱中培养3 d;用细胞刮轻轻将细胞球和条件培养基刮至一侧皿盖,用移液枪收集细胞条件培养基至离心管中,1000 r/min离心3 min,收集细胞上清,用0.22 μm过滤器过滤,即收集完CM,置于-20 ℃冰箱中储存。
参照国家海洋局第三海洋研究所提供的配方配制。人工海水含NaCl 26.50 g/L、KCl 0.70 g/L、MgCl2 2.50 g/L、NaHCO3 0.20 g/L、MgSO4 3.30 g/L、NaBr 0.08 g/L、CaCl2 1.14 g/L,浓度为3.0%~3.6%,利用酸碱中和反应将pH值调至8.2。
HT22细胞达到处理时间后,在细胞培养板中加入培养基总体积1/10量的CCK-8试剂,然后置于细胞培养箱中孵育至少0.5 h。孵育完成后,将含10% CCK-8试剂的培养基用移液枪转移至96孔板中,100 μl/孔,采用酶标仪检测450 nm波长处的吸光度。
将HT22细胞以1×106/ml的密度接种于48孔板,当细胞融合达80%~90%时,加入0%、15%、25%、50%、75%和100%浓度的海水,细胞培养0、3、6、9和12 h后,采用倒置显微镜镜观察细胞形态和数量的变化并拍照记录,结合CCK-8实验结果,筛选出建立SI+SW细胞模型适宜的海水浓度和浸泡时间。
将HT22细胞以1×106/ml的密度接种至胶原拉伸细胞培养板中,待细胞丰度长至70%~90%,随机分为对照组、SI组、SI+SW组、SI+CM组和SI+SW+CM组。SW组、SI+SW组和SI+SW+CM组细胞加入15%浓度海水浸泡,SI组、SI+SW组和SI+SW+CM组细胞进行SI,SI+CM组和SI+SW+CM组给予含20% 3D MSCs分泌组物的CM处理,培养12 h后,采用CCK-8法检测各组HT22细胞活力。
HT22细胞损伤及治疗12 h后,将细胞培养板中原有的培养基收集至离心管,加入胰酶室温下孵育,消化完成后,加入收集的培养基中和以终止消化,收集细胞悬液,预冷PBS洗涤2次,加入适量1×缓冲液重悬,加入PE Annexin V和7-AAD染色,室温下避光孵育15 min,1 h内上机进行检测。
取干预后的HT22细胞,加入细胞裂解液,冰上静置30 min,4 ℃下12 000 r/min离心5 min;取蛋白上清液用BCA法测定各组总蛋白浓度,加入适量6× 蛋白上样缓冲液,99 ℃金属浴10 min使蛋白变性;将30 μg变性蛋白进行SDS-PAGE电泳1.5 h,转膜2 h,封闭1 h,加入兔抗Bcl-2、Bax、ACSL4、COX-2和β-actin一抗(均为1:1000)4 °C孵育过夜,次日加入HRP标记的二抗(1:5000)室温孵育1 h,用ECL化学发光法显影。采用ImageJ软件进行定量分析,以与内参蛋白吸光度的比值表示目的蛋白的表达水平。
HT22细胞以1×106/ml的密度接种至6孔胶原拉伸培养板中,待细胞丰度长至70%~90%,加入海水浸泡,进行SI,加入3D CM处理,用枪头在培养板每孔中央划痕,然后于12、24 h拍照,观察细胞愈合程度。
采用GraphPad Prism 9软件进行统计分析并作图。计量资料均符合正态分布,以$\bar{x}±s$表示,多组间比较采用方差分析,进一步两两比较采用Turkey检验。所有实验重复3次。P<0.05为差异有统计学意义。
显微镜下观察显示,培养3 h,使用完全培养基(0%浓度海水)的HT22细胞生长良好;50%浓度海水浸泡的HT22细胞形态发生了可辨识的变化;75%浓度海水浸泡的HT22细胞数量减少、形态改变明显;100%浓度海水浸泡的HT22细胞可观察到大面积细胞膜破裂、细胞体积缩小,细胞形态难以辨识,数量明显减少。培养12 h,使用完全培养基的HT22细胞生长正常;而100%海水浸泡的细胞几乎完全死亡,仅留下少量细胞碎片。15%浓度海水浸泡的HT22细胞,培养3、12 h后细胞形态基本保持稳定(图1A)。CCK-8法检测结果显示,HT22细胞活力受浓度和时间梯度的双重影响;经单纯海水浸泡12 h后,仅15%浓度海水能够维持约80%的细胞活力(图1B)。后续实验采用15%浓度海水浸泡HT22细胞12 h建立SI+SW细胞模型。
CCK-8法检测结果显示,处理12 h后,与对照组(100.00%±3.28%)比较,SW组(81.57%±1.49%)、SI组(70.43%±3.29%)和SI+SW组(61.88%±0.31%)HT22细胞活力均明显下降(P<0.0001);经MSCs分泌组物处理后,与SI组比较,SI+CM组的细胞活力明显增强(P<0.0001);与SI+SW组相比,SW+SI+CM组细胞活力明显增强(P<0.001)(图2)。
流式细胞术检测结果显示,SI组和SI+SW组HT22细胞凋亡率明显高于对照组(14.02%±0.87% vs. 16.99%±1.21% vs. 9.77%±1.94%),且SI+SW组明显高于SI组,差异均有统计学意义(P<0.05);经MSCs分泌组物处理后,SI+CM组HT22细胞凋亡率(11.13%±1.24%)明显低于SI组,SI+SW+CM组HT22细胞凋亡率(13.67%±1.51%)明显低于SI+SW组,差异均有统计学意义(P<0.05)(图3)。
Western blotting检测结果显示,与对照组比较,SI组和SI+SW组促凋亡蛋白Bax表达水平均明显升高(P<0.0001),且SI+SW组明显高于SI组(P<0.05);与对照组比较,SI组和SI+SW组抑凋亡蛋白Bcl-2表达水平均明显降低(P<0.01,P<0.0001),且SI+SW组明显低于SI组(P<0.01)。经MSCs分泌组物处理后,与SI组比较,SI+CM组Bax表达水平明显降低(P<0.05),Bcl-2表达水平明显升高(P<0.05);与SI+SW组比较,SI+SW+CM组Bax表达水平明显降低(P<0.001),Bcl-2表达水平明显升高(P<0.01)(图4)。
Western blotting检测结果显示,与对照组比较,SI组和SI+SW组ACSL4和COX-2蛋白表达水平均明显升高(P<0.01或P<0.001或P<0.01)。经MSCs分泌组物处理后,与SI组比较,SI+CM组ACSL4和COX-2蛋白表达水平均明显降低(P<0.05);与SI+SW组比较,SI+SW+CM组ACSL4和COX-2蛋白表达水平均明显降低(P<0.01)(图5)。
细胞划痕实验结果显示,与对照组比较,SI组和SI+SW组细胞迁移率明显降低(P<0.01或P<0.0001),且SI+SW组明显低于SI组(P<0.0001);经MSCs分泌组物处理12 h和24 h,SI+CM组细胞迁移率明显高于SI组(P<0.01,P<0.0001),SI+SW+CM组细胞迁移率明显高于SI+SW组(P<0.0001)(图6)。
本研究观察了含3D MSCs来源的分泌组物CM对SW合并SI神经元的保护作用,结果显示,MSCs分泌组物可促进SW合并SI神经元的损伤修复,在SW合并TBI治疗方面具有一定潜力。
动物体内环境较为复杂,为了排除体内其他因素对干细胞保护效能的干扰,本研究选用小鼠海马神经元细胞系HT22作为研究对象,在体外建立SW+SI细胞模型,观察含MSCs分泌组物CM对创伤神经元的保护效能。常见的TBI体外模型有细胞划痕模型、细胞拉伸模型、氧糖剥夺模型、过氧化氢模型、压迫损伤模型、加速模型等[16-18]。细胞拉伸模型目前被认为是模拟亚致死创伤性神经元损伤的“金标准”,具有简单性、多样性和可重复性等优势;研究者采用该模型可自主控制细胞的损伤程度及所处的外环境,从而模拟不同程度的损伤条件,为深入研究神经元损伤机制及探寻治疗策略提供了可用工具[14]。前期本研究团队建立了体外SW的TBI细胞模型,即SW合并SI细胞模型[19];本研究也选择该体外模型来模拟体内神经元的损伤情况,观察MSCs对SW合并损伤神经元的治疗作用,为探索MSCs在相关损伤治疗中的潜力提供实验依据。
本研究首先筛选出构建SW+SI细胞模型的适宜海水浓度为15%,浸泡时间为12 h,随后以这一条件构建了SW+SI神经元损伤模型。比较了单纯SW、单纯SI及SW+SI对HT22细胞活力的影响,结果显示,SW+SI的HT22细胞活力最低,而单纯SI的HT22细胞活力低于单纯SW,提示SW+SI HT22细胞模型建立成功。本研究在细胞模型中加入含MSCs分泌组物的CM进行处理,并观察到含MSCs分泌组物的CM对SW+SI神经元活性具有明显的保护效能。本团队前期研究显示,在体外和体内SW的TBI模型中均可观察到神经元凋亡,且SW后神经元凋亡加剧[19-20]。有研究显示,MSCs可通过其旁分泌作用减轻TBI后的细胞凋亡,在神经元损伤修复中存在潜在的应用价值[21-23]。本研究结果显示,与SI组相比,SI+SW组神经元凋亡率增高,促凋亡蛋白Bax表达水平升高,而抑凋亡蛋白Bcl-2表达水平降低,提示SW可加重损伤的神经元凋亡,与本研究团队的前期结果一致。然而,在加入3D MSCs来源的含有分泌组物的CM处理后,SI+SW+CM组细胞凋亡率低于SI+SW组,同时,促凋亡蛋白Bax表达水平降低,抑凋亡蛋白Bcl-2表达水平升高,提示富含MSCs分泌组物的CM可抑制SW+SI神经元凋亡。上述结果提示,通过抑制神经元凋亡,含有MSCs分泌组物的CM有望用于SW合并TBI的治疗。有研究发现,MSCs可抑制TBI诱导的铁死亡,有助于减轻TBI后的认知障碍,为治疗TBI提供了新的潜在策略[24]。本研究中,富含MSCs分泌组物的CM处理SW合并TBI细胞模型可降低促铁死亡蛋白ACSL4和COX-2的表达,表明CM可抑制SW合并SI的HT22细胞铁死亡,提示抑制铁死亡可能成为MSCs分泌组物治疗SW合并TBI的一种有效策略。多项研究显示,MSCs分泌组物在神经损伤修复方面具有潜在的应用前景[25-28]。在本研究中,SW+SI细胞模型在细胞划痕处理12 h和24 h后,含MSCs分泌组物的CM可明显促进HT22细胞损伤的修复,提示3D MSCs来源的分泌组物可能有助于修复SW合并TBI的神经元损伤。
综上所述,本研究结果显示,3D MSCs来源的分泌组物在SW合并TBI的治疗中具有潜在的应用价值。这些分泌组物可通过多重机制,如保护细胞活力、抑制细胞凋亡和铁死亡及促进细胞迁移等促进神经元损伤的修复,进而对SW合并TBI的神经元发挥保护作用。本研究仍存在一定的局限性,如仅在细胞层面观察了MSCs分泌组物对TBI损伤的短期作用,未考虑长期效应及其具体作用通路和分子机制,且使用的细胞系种类有限。未来的研究需要在动物模型中进一步验证这些分泌组物对TBI损伤的作用,并探讨其长期影响、具体作用机制及最佳治疗时间窗等。
