In order to clarify the pathogenicity and the potential risk of major corn pathogens to soybean root rot in corn-soybean inter planting, this study artificially inoculated non-co-occurring pathogens onto soybean roots and observed the symptoms in pots, and identified the infection grades of pathogens on soybean root rot. The results showed that Curvularia lunata causing corn curvularia leaf spot, Bipolaris maydis causing southern corn leaf blight, and Exserohilum turcicum causing northern corn leaf blight were non-co-occurring pathogens and could cause soybean root rot. Isolation and identification confirmed that the isolated strains were identical to the inoculated pathogens; C.lunata could cause soybean root rot up to Grade 5, B.maydis and E.turcicum could lead to soybean root rot up to Grade 3. The above results indicated that all three pathogens were causal agents of soybean root rot, and there was a potential risk of causing soybean root rot.

为明确玉米-大豆复合种植过程中,玉米主要病原菌对引起大豆根腐病的致病力及潜在风险,通过人工接种非共发性病原菌至大豆根部并观察其症状,鉴定病原菌对大豆根腐病的侵染级别。结果表明,引起玉米弯孢叶斑病的新月弯孢、引起玉米小斑病的玉蜀黍平脐蠕孢和引起玉米大斑病的大斑突脐蠕孢为非共发性病原菌,均可引起大豆根腐病。经分离鉴定,分离的菌株与接种菌株为同一病原菌;新月弯孢可导致大豆根腐病病级达5级,玉蜀黍平脐蠕孢和大斑突脐蠕孢菌可导致大豆根腐病病级达3级。以上结果说明3种病原菌均为大豆根腐病的致病菌,存在引起大豆根腐病的潜在风险。

为明确玉米-大豆复合种植过程中,玉米主要病原菌对引起大豆根腐病的致病力及潜在风险,通过人工接种非共发性病原菌至大豆根部并观察其症状,鉴定病原菌对大豆根腐病的侵染级别。结果表明,引起玉米弯孢叶斑病的新月弯孢、引起玉米小斑病的玉蜀黍平脐蠕孢和引起玉米大斑病的大斑突脐蠕孢为非共发性病原菌,均可引起大豆根腐病。经分离鉴定,分离的菌株与接种菌株为同一病原菌;新月弯孢可导致大豆根腐病病级达5级,玉蜀黍平脐蠕孢和大斑突脐蠕孢菌可导致大豆根腐病病级达3级。以上结果说明3种病原菌均为大豆根腐病的致病菌,存在引起大豆根腐病的潜在风险。

In order to clarify the pathogenicity and the potential risk of major corn pathogens to soybean root rot in corn-soybean inter planting, this study artificially inoculated non-co-occurring pathogens onto soybean roots and observed the symptoms in pots, and identified the infection grades of pathogens on soybean root rot. The results showed that Curvularia lunata causing corn curvularia leaf spot, Bipolaris maydis causing southern corn leaf blight, and Exserohilum turcicum causing northern corn leaf blight were non-co-occurring pathogens and could cause soybean root rot. Isolation and identification confirmed that the isolated strains were identical to the inoculated pathogens; C.lunata could cause soybean root rot up to Grade 5, B.maydis and E.turcicum could lead to soybean root rot up to Grade 3. The above results indicated that all three pathogens were causal agents of soybean root rot, and there was a potential risk of causing soybean root rot.

大豆是我国重要的粮食作物[1],为保证农业可持续发展的种植模式,解决日益增长的人口、环境和资源问题[2],同时缓解土地资源利用、改善生态环境和降低病虫草害,近年来,大豆玉米带状复合种植技术和轮作的种植模式被积极推广和实施[3]。该技术以实现大豆和玉米协同共生,以及缓解大豆供应不足等方面发挥着重要的作用。
我国大豆种植面积呈逐渐扩大趋势,2022年大豆种植面积1024万hm2,比2021年增加18万hm2[4],2023年大豆种植面积1047万hm2,比2022年增加23万hm2[5]。在带状复合种植、轮作和邻作种植模式的三重压力下,大豆和玉米病虫害防控面临巨大的挑战。研究[6]表明,玉米和大豆病虫草害种类累计达到50余种,如玉米病害有小斑病、大斑病、弯孢叶斑病和根腐病等,大豆病害有根腐病、病毒病、叶斑病和锈病等。根腐病作为玉米和大豆2种作物的共发性病害,是一种破坏性极强的土传病害。据报道[7-10],镰孢菌作为多种作物根腐病的主要病原菌,可引起玉米和大豆的根腐病,还可造成玉米根腐病和大豆根腐病相互侵染。李红菊等[11]发现大豆根腐病病原菌层出镰孢、禾谷镰孢和尖孢镰孢等均可侵染玉米根部,引起玉米根腐病;玉米禾谷镰孢菌可侵染大豆根部,引起大豆根腐病,造成相互侵染。在套作、轮作以及邻作模式下,病原菌能够通过寄生于田间不同的作物、借助秸秆还田、病残体以及土壤越冬等方式进行循环传播,因此应关注2种作物根腐病的潜在发生风险。目前,尚未见玉米其他主要病原菌如新月弯孢、玉蜀黍平脐蠕孢、大斑突脐蠕孢和腐霉菌等病原菌对大豆根部的侵染致病情况及潜在风险的研究。本研究将玉米病原菌新月弯孢、玉蜀黍平脐蠕孢和大斑突脐蠕孢人工接种于大豆根部,观察其侵染情况,分析潜在风险,旨在解决大豆玉米带状复合和轮作种植模式下玉米和大豆非共发性病原菌对共发性病害带来的潜在风险,为玉米和大豆相关病害防控提供理论依据。
供试品种:冀豆30;供试菌株:玉米病原菌为新月弯孢(Curvularia lunata)、玉蜀黍平脐蠕孢(Bipolaris maydis)和大斑突脐蠕孢(Exserohilum turcicum),均来自本实验室保存菌种。
将冀豆30种子进行表面消毒,顺序为酒精消毒30 s,2.0% NaClO消毒1 min,无菌水清洗3次,晾干以备种植使用。
参考王晓艳等[10]的方法,将所需的3种病原菌活化至25 ℃培养箱中培养7 d,取直径8 mm的菌饼接种至高粱培养基中,每个培养袋中接种10个菌饼,25 ℃培养10 d左右,待菌丝布满每个高粱籽粒表面,即可用于接种。
将无菌盆栽土装入高11 cm,直径12 cm的花盆中,容量为花盆的3/4,浇足水后将带菌的高粱籽粒接种体接种于花盆中,每盆30粒,覆上约1.5 cm厚度无菌盆栽土后播种,每盆播种5粒,播种后覆盖1 cm厚度的无菌盆栽土。温室培养,正常管理,以不接种病原菌为对照,每处理3次重复,试验重复2次。
参考辛惠普等[12]定级标准将大豆根腐病病情严重度划分为5个级别。0级:植株正常生长;1级:主根未发病或轻微变成褐色,须根生长点发病,植株正常生长;3级:主根发病变黑,能继续生长,须根根尖变黑,植株正常生长;5级:主根严重变黑,不能继续生长,须根少或不长,植株生长不良;7级:根部腐烂,植株不能继续生长甚至死亡。
取发病部位病健交界处,剪成5 mm×5 mm小块,分别用75%酒精处理30 s,2.0% NaClO处理1 min,无菌水冲洗3次,用无菌滤纸吸干叶片表面水分,置于PDA培养基上25 ℃黑暗培养,待长出菌丝后将菌丝转到新的PDA培养基上培养,之后进行形态学鉴定。
采用Excel 2010整理试验数据。
图1可知,接种新月弯孢病原菌后,该菌可侵染大豆的整个茎基部及根系,主根部产生大面积黑色病斑,甚至整体坏死,须根很少或无,多为黑色(图1a~d),严重者病斑扩大可引起根部缢缩、凹陷或腐烂(图1e),剖开根部可发现皮层和中柱部变黑(图1f),根部生长受抑制。将发病部位进行病原菌分离与鉴定,发现分离到的菌株与接种菌株为同一病原菌(图1g~h),说明新月弯孢可引起大豆根腐病。
图2可知,玉蜀黍平脐蠕孢菌接种大豆根部后,初期在大豆主根部形成不规则长条形的病斑(图2a),后期病斑逐渐扩大连接成片或在茎基部形成长形病斑(图2f),严重时可造成主根部变黑、腐烂(图2b)或凹陷(图2c~d),须根变黑且减少或无,整个根系生长不良,剖开根部可发现内部组织变黑或褐色,侵染深入中柱部(图2e)。将发病部位进行病原菌分离、鉴定,发现分离到的菌株与接种菌株为同一病原菌(图2g~h),说明玉蜀黍平脐蠕孢可引起大豆根腐病。
图3可知,大斑突脐蠕孢菌接种大豆根部后,主要侵染大豆的主根、须根和茎基部,初期病斑呈褐色椭圆形(图3a),后逐渐扩大,使整个主根呈褐色(图3b),在大豆根部开裂病斑呈长梭形,黑褐色(图3c),严重时可造成根部腐烂和皱缩(图3d~e),与未接菌的根部比较可知主根变褐(图4),须根变褐或减少,剖开根部发现其仅侵染至表皮组织,表皮和中柱组织未被侵染(图3f)。将发病部位进行病原菌分离、鉴定,发现分离到的菌株与接种菌株为同一病原菌(图3g),说明大斑突脐蠕孢可引起大豆根腐病。
图5可知,新月弯孢引起的大豆根腐病与对照相比可使大豆植株变矮,主根严重变黑,须根变少或无,植株生长不良,达侵染病级5级;玉蜀黍平脐蠕孢引起的大豆根腐病可使大豆主根发病变黑,须根变少,植株正常生长,达侵染病级3级;大斑突脐蠕孢引起的大豆根腐病达侵染病级3级。
新月弯孢不仅是玉米叶斑病的主要病原菌,而且可以侵染小麦[13]、向日葵[14]、水稻[15]和高粱[16]等作物,还可侵染狗尾草、马唐、牛筋草和稗草等常见禾本科杂草[17]。有研究[18-19]表明,新月弯孢可存在于大豆种间和根际,Anwar等[18]在26份大豆种子样品中分离到了15株田间真菌和5株贮藏真菌,其中就有新月弯孢;Ismail等[19]在大豆种子中分离出新月弯孢,通过接种发现新月弯孢可造成大豆发芽率减少,生长出现异常;Wang等[20]在耐镉元素的大豆品种的根部分离到3种真菌,分别为卷枝毛霉(Mucor circinelloides)、新月弯孢和粉红枝穗霉(Clonostachys rosea)。以上研究结果表明,新月弯孢可存在于大豆根部或种子中。本研究在此基础上,将新月弯孢病原菌接种于大豆根部,研究其引起大豆根腐病的潜在风险,当大豆种子携带新月弯孢病或玉米田收获后,大量的病残体存在于田间时,应关注大豆根腐病的发生情况,加强新月弯孢引起的大豆根腐病的防治。
蠕孢菌的寄主范围较广,不仅能侵染玉米、水稻和小麦等植物的叶片和穗部,还能侵染多种作物的根部,引起如玉米[21]和高粱[22]等的根腐病,其中麦根腐蠕孢菌是小麦根腐病的主要病原菌,可造成小麦和玉米的交叉侵染致病[23],黄淮海地区上茬作物多以小麦为主,在麦茬地收获后,在小麦―大豆、小麦―玉米轮作田或玉米大豆条带种植田中应对大豆或玉米根部的侵染情况多加关注,有报道[24]称,平脐蠕孢菌可侵染植物的根、茎、叶和叶鞘,可通过种子、土壤和病残体传播或越冬,玉蜀黍平脐蠕孢菌是一种在大豆玉米条带种植中的一种非适应性病原体,但尚未有研究能证明大豆对玉蜀黍平脐蠕孢菌有免疫反应,Dong等[25]研究发现,在大豆玉米条带种植过程中,大豆中存在非共发性病原体的系统性非宿主抗性(NHR)分子机制,表明大豆和玉米之间存在着复杂的相互作用。因此,本研究将玉米上的玉蜀黍平脐蠕孢菌和大斑突脐蠕孢菌接种于大豆根部,证明其可引起大豆根腐病,在条带种植模式下非共发性病原菌有引起根腐病的潜在风险。
在套作、轮作和邻作模式下,玉米和大豆2种作物中非共发性病害存在演变成共发性病害的风险,由本研究结果可知,人工接种玉米病原菌新月弯孢、玉蜀黍平脐蠕孢和大斑突脐蠕孢菌能够侵染大豆根部和茎基部,影响植株生长,证明3种病原菌可以侵染大豆根部,引起大豆根腐病,存在交叉侵染致病性;通过病情定级可知,新月弯孢可造成大豆根腐病5级的病情,玉蜀黍平脐蠕孢和大斑突脐蠕孢可造成大豆根腐病3级的病情。在玉米弯孢叶斑病、小斑病和大斑病大发生年份,应关注大豆根腐病害的发生,加强防治。
通过人工接种玉米病原菌至大豆根部,发现玉米病原菌新月弯孢、玉蜀黍平脐蠕孢和大斑突脐蠕孢均可侵染大豆根部,引起大豆根腐病,是大豆根腐病的致病性病原菌,在玉米―大豆条带复合种植过程中,存在交叉侵染致病性及风险。
