Northern Japonica rice region is susceptible to cold damage, and breeding cold-tolerant rice varieties is the most direct and effective means to address this issue. In this study, 45 accessions of northern Japonica rice were used as experimental materials. Under 14 °C cold stress, seven indicators including germination potential, germination rate, germination index, vigor index of shoot, vigor index of root, average germination days, and germination coefficient were measured. Multivariate statistical methods such as correlation analysis, principal component analysis (PCA), membership function comprehensive analysis, and cluster analysis, were employed to evaluate cold tolerance. The results showed that all germination indicators exhibited varying degrees of decline under cold stress, with the vigor index of root showing the most obvious decrease. By calculating the membership function values of each indicator, the seven indicators were transformed into two comprehensive indicators through PCA. Cluster analysis was then used to categorize the 45 accessions into four groups, successfully screening 13 cold-tolerant materials. Furthermore, a regression equation was established, which identified that germination potential, germination rate, vigor index of root, and germination coefficient are the key indicators for evaluating cold tolerance in northern Japonica rice at the germination stage.

北方粳稻区易受冷害,选育耐冷水稻品种是应对冷害最直接有效的手段。以45份北方粳稻为试验对象,设定14 ℃为冷胁迫温度,测定各材料的发芽势、发芽率、发芽指数、芽活力指数、根活力指数、平均发芽天数及发芽系数等7个指标,采用相关性分析、主成分分析、隶属函数综合分析和聚类分析等多元统计方法进行耐冷性评价。结果表明,冷胁迫下各项萌发指标均呈不同程度的下降趋势,根活力指数降幅最为明显。通过计算各指标隶属函数值,经主成分分析将7个指标转化为2个综合指标,利用聚类分析将45份北方粳稻划分为4类,成功筛选出13份耐冷材料。进一步通过回归分析构建方程,明确发芽势、发芽率、根活力指数和发芽系数是鉴定北方粳稻萌发期耐冷性的关键指标。

北方粳稻区易受冷害,选育耐冷水稻品种是应对冷害最直接有效的手段。以45份北方粳稻为试验对象,设定14 ℃为冷胁迫温度,测定各材料的发芽势、发芽率、发芽指数、芽活力指数、根活力指数、平均发芽天数及发芽系数等7个指标,采用相关性分析、主成分分析、隶属函数综合分析和聚类分析等多元统计方法进行耐冷性评价。结果表明,冷胁迫下各项萌发指标均呈不同程度的下降趋势,根活力指数降幅最为明显。通过计算各指标隶属函数值,经主成分分析将7个指标转化为2个综合指标,利用聚类分析将45份北方粳稻划分为4类,成功筛选出13份耐冷材料。进一步通过回归分析构建方程,明确发芽势、发芽率、根活力指数和发芽系数是鉴定北方粳稻萌发期耐冷性的关键指标。

Northern Japonica rice region is susceptible to cold damage, and breeding cold-tolerant rice varieties is the most direct and effective means to address this issue. In this study, 45 accessions of northern Japonica rice were used as experimental materials. Under 14 °C cold stress, seven indicators including germination potential, germination rate, germination index, vigor index of shoot, vigor index of root, average germination days, and germination coefficient were measured. Multivariate statistical methods such as correlation analysis, principal component analysis (PCA), membership function comprehensive analysis, and cluster analysis, were employed to evaluate cold tolerance. The results showed that all germination indicators exhibited varying degrees of decline under cold stress, with the vigor index of root showing the most obvious decrease. By calculating the membership function values of each indicator, the seven indicators were transformed into two comprehensive indicators through PCA. Cluster analysis was then used to categorize the 45 accessions into four groups, successfully screening 13 cold-tolerant materials. Furthermore, a regression equation was established, which identified that germination potential, germination rate, vigor index of root, and germination coefficient are the key indicators for evaluating cold tolerance in northern Japonica rice at the germination stage.

水稻作为广泛种植的谷物作物,是多数人口的主要食物来源[1],其对寒冷敏感,早期生长易受冷害影响[2-3]。种子萌发是作物正常生长的首要环节,萌发期遭受冷害会导致种子萌发受阻,甚至丧失发芽能力[4-6],进而严重制约水稻产量潜力的发挥[7-8]
我国北方地区是主要的农业产区,但其热量条件有限,春播期和幼苗生长期常遭遇大规模低温冷害,抑制作物生长发育[8-10]。北方粳稻区纬度跨度大,涵盖东北早熟粳稻区、华北单季粳稻区和西北干燥粳稻区,种植面积约占我国粳稻总面积的60%[11]。该区域春季温度波动大,清晨气温低、升温慢,水稻冷害频发,减产现象时有发生,严重制约了北方粳稻区的稻作发展[12]
选育耐冷品种是解决水稻冷害问题最直接有效的手段[13],通过遗传改良培育新品种对北方粳稻区水稻生产至关重要,而构建耐冷综合评价体系是筛选耐冷材料的前提。目前,学者已对谷子[4]、糜子[14]、棉花[15]、高粱[16]和花生[17]等多种作物开展了萌发期耐冷性综合评价,但针对北方粳稻萌发期耐冷性综合评价的相关研究较少。本试验利用人工培养箱模拟冷胁迫环境,对45份北方粳稻的萌发期耐冷性进行鉴定,分析耐冷材料的性状特征,明确萌发期耐冷性的关键指标,旨在为北方粳稻区耐冷水稻选育提供优质种质资源与理论依据。
以45份粳稻类型水稻为供试材料,包括来源中国的43份(表1)和来源日本的2份(秋光,J44;秋田小町,J45),均由铁岭市农业科学院提供。
参照金铭路等[18]的方法,每份材料称取30 g种子置于装有0.5% NaClO溶液的烧杯中充分搅拌,消毒20 min后将种子取出,用蒸馏水冲洗干净,随后在蒸馏水中浸种12 h。每份材料挑选100粒饱满的种子,均匀放置于垫有滤纸的培养皿中,置于培养箱中,设置26 ℃(对照组,CK)和14 ℃(处理组,T)2个萌发温度[19],期间每天更换培养皿中的蒸馏水,每个处理3次重复。
以种子露白记为发芽,逐日观察并记录CK和T处理的发芽情况;以当日发芽粒数无新增的时间记为规定期限,CK处理为5 d,T处理为15 d,用直尺测量并记录芽长和根长。参照黄贺等[20]的方法,计算发芽势(germination potential,GP)、发芽率(germination rate,GR)、发芽指数(germination index,GI)、芽活力指数(vigor index of shoot,VIS)、根活力指数(vigor index of root,VIR)、平均发芽天数(average germination days,AGD)和发芽系数(germination coefficient,GC)7个指标。其中,在1/3规定期限时统计发芽势(CK处理于第2天测定,T处理于第5天测定),此时种子出芽旺盛,考察各品种出芽整齐度和种子活力;在规定期限时统计发芽率,此时种子已基本出芽完毕,考察各品种出芽能力。各指标计算公式为:

采用Microsoft Excel 2016进行数据整理,利用R 4.1.3的corrplot包进行Pearson相关性分析,利用SPSS 23进行主成分分析,利用R 4.1.3的ggplot2包、ComplexHeatmap包、circlize包和dendextend包进行聚类分析,采用ward.D方法层次聚类,采用Euclidean方法计算遗传距离,利用SPSS 23的线性回归分析对综合耐冷性评价值(D)进行回归方程分析。相关计算公式[21]如下:

其中,CRC1~CRC7分别表示发芽势、发芽率、发芽指数、芽活力指数、根活力指数、平均发芽天数和发芽系数的耐冷系数。

式中,U(CI)表示各综合指标的隶属函数值,CIj表示第j个综合指标值,CImax表示第j个综合指标的最大值,CImin表示第j个综合指标的最小值。

式中,Wj表示第j个综合指标的权重;Pj表示第j个综合指标的贡献率。

45份水稻材料在T处理下的平均发芽天数与CK处理相比增加了219.90%(表2)。与CK处理相比,发芽势、发芽率、发芽指数、芽活力指数、根活力指数和发芽系数均降低,降幅分别为39.47%、15.31%、71.23%、80.66%、89.48%和71.43%,其中根活力指数的降幅最大,发芽率降幅最小。此外,CK处理各指标的变异系数在1.95%~ 35.13%,T处理在23.57%~84.83%,2个处理均存在较大差异,由此计算其耐冷系数进行后续分析。
表3可知,各指标耐冷系数的变异系数在22.99%~74.85%,根活力指数的耐冷系数变异最大,发芽率的耐冷系数最小,表明供试材料间存在广泛的遗传多样性,故对其进行综合评价。
45份水稻材料萌发期各耐冷系数之间存在不同程度的相关关系(图1)。发芽势、发芽率、发芽指数、芽活力指数、根活力指数和发芽系数的耐冷系数之间均呈极显著正相关,其中芽活力指数与根活力指数耐冷系数的相关系数高达0.94。平均发芽天数与其余指标的耐冷系数均呈负相关,其中与发芽势、发芽指数、芽活力指数、根活力指数和发芽系数的耐冷系数呈显著负相关,与发芽系数耐冷系数的相关系数绝对值最大(-0.73)。各单项指标间存在不同程度的相关性,使信息产生重叠,因此,直接利用单项指标评价北方粳稻萌发期耐冷性是不全面的,需要进行多元统计分析。
对45份水稻材料萌发期的耐冷系数进行主成分分析,将原有的7个指标转化为2个独立的综合指标CI1和CI2(表4)。2个主成分特征值均大于1,累计贡献率为89.866%,以贡献率占比作为权重,主成分CI1权重占比83.7%,主成分CI2权重占比16.3%。这2个主成分可以较好地替代7个性状指标,对萌发期耐冷性进行评价。
将45份水稻材料进行聚类分析(图2),分为4种类型,其中J23、J16、J5、J20、J35、J32、J41、J21、J34、J29、J12、J22和J27归为一类,共13份材料,为耐冷型;J17、J8、J31、J40、J19、J33、J15、J42和J14归为一类,共9份材料,为较耐冷型;J4、J18、J45、J13、J11、J3、J37和J30归为一类,共8份材料,为冷敏感型;其余15份归为一类,为冷较敏感型。
45份水稻材料萌发期综合耐冷性评价值D的分布范围在0.024~0.890,其中J29最大,表明其萌发期耐冷性最好;J30最小,表明其萌发期耐冷性最差(表5)。为进一步分析水稻萌发期评价指标与耐冷性之间的关系,以耐冷系数为自变量、D值为因变量进行回归分析,构建最优回归方程:D1=−0.104+0.178X1+0.292X2+1.034X5+0.552X7R2= 0.994,P<0.05),式中,D1为耐冷评价预测值,X1X2X5X7分别为发芽势、发芽率、根活力指数和发芽系数的耐冷系数,可以解释D1值的99.4%,说明此方程可用于萌发期耐冷性评价。方程估计精度较高,利用该回归方程估算D值,可提高水稻萌发期耐冷性评价的工作效率。
近年来,低温对农作物生产及粮食安全产生了诸多影响[22]。种子萌发作为作物生长发育过程中的重要环节,对产量具有决定性作用,若作物在萌发期具有较强的耐冷性,则能为后期生长奠定良好基础[2]。随着农村劳动力外流,劳动力与生产物资成本提高,直播稻因其省工节本的特点愈发受到农民青睐,然而,直播稻早春播种时易遭受低温冷害[23-24]。因此,筛选并培育耐冷品种既契合北方粳稻区的生产需求,也满足了耐冷直播水稻品种的选育要求,还可推广至其他高纬度、高海拔低温地区,有助于扩大水稻的低温适应性,降低气候变化对水稻产量的影响[2,25]
由于各指标间差异显著,为更直观地呈现冷胁迫对各指标的影响,采用T处理与CK处理的相对值,即耐冷系数,可消除不同品种间基本性状的差异[4]。本试验的45份北方粳稻中,各指标的变化趋势与幅度存在差异,且各耐冷系数间存在不同程度的相关性。由于指标间信息重叠,仅依靠单一指标的耐冷系数难以全面阐释耐冷实质,从而导致无法准确评价不同材料萌发期的耐冷性[26-27]。隶属函数分析与主成分分析已广泛应用于多种作物的抗逆性评价中[28-31]。本研究运用主成分分析法,将萌发期7个耐冷性指标转化为2个主成分,可解释89.866%的变异,能更准确地反映萌发期的耐冷信息。通过计算综合隶属函数值并结合聚类分析,对萌发期耐冷性进行分级评价,将45份北方粳稻划分为耐冷型、较耐冷型、冷较敏感型和冷敏感型4种类型,可有效筛选出对低温胁迫反应极端的材料。
构建回归方程并确定关键指标,可进一步提高筛选效率[32-33]。本研究构建回归方程:D1=−0.104+ 0.178X1+0.292X2+1.034X5+0.552X7R2=0.994,P<0.05),明确发芽势、发芽率、根活力指数和发芽系数可用于萌发期耐冷性评价。同时,各材料的估算精度较高,表明该回归方程能够较为准确地预测不同北方粳稻品种的耐冷性强弱。
通过多元统计方法进行萌发期耐冷性鉴定,45份北方粳稻材料可划分为4类。铁粳香3号、松粮850、辽粳9号、铁粳1603、盘粳202、北粳K13、铁粳96、东研稻27、天隆优717、铁粳1743、港优9号、铁粳1808和天隆优619共13份材料耐冷性最强,可作为耐冷种质资源加以利用。通过回归分析构建方程,确定发芽势、发芽率、根活力指数和发芽系数为北方粳稻萌发期耐冷性评价的关键指标,可据此进行耐冷品种鉴选。
