The absorption of selenium by tartary buckwheat to form selenium-enriched characteristics is of great significance for promoting the high-quality development of the tartary buckwheat industry. In this study, 30 representative tartary buckwheat germplasm resources were selected to measure their agronomic traits, dry matter accumulation and translocation characteristics, as well as yield and grain trait indicators. Foliar application of selenium fertilizer was set as a treatment. Principal component analysis (PCA), cluster analysis, and GGE biplot analysis were comprehensively utilized to conduct a thorough evaluation of the growth, development, and selenium response characteristics of different tartary buckwheat germplasms. The results showed that significant variations existed among different tartary buckwheat germplasm resources in terms of agronomic traits, dry matter accumulation and translocation characteristics, yield and its components, and selenium accumulation, distribution, and enrichment traits. Furthermore, foliar selenium application exhibited a promoting effect on all measured growth and development indicators of the germplasm resources. Through cluster analysis, ten selenium-sensitive, 12 selenium-intermediate, and eight selenium-insensitive germplasm resources were screened. GGE biplot analysis identified YYMY002 as a germplasm with high yield, high selenium enrichment, and relative comprehensive superior traits.

苦荞吸收硒元素形成富硒特性,对推动苦荞产业高质量发展具有重要意义。选择30份具有代表性的苦荞种质资源,测定其植株的农艺性状、干物质积累与转运特性以及产量与籽粒性状指标,设置叶片喷施硒肥处理,综合运用主成分分析、聚类分析和GGE双标图分析法,对不同苦荞种质的生长发育及硒响应特征进行全面评价。研究表明,不同苦荞种质资源在农艺性状、干物质积累与转运特性、产量及其构成因素、硒积累分配富集性状等方面存在较大变异,且喷施硒肥对苦荞种质资源的生长发育性状各指标均表现出促进作用。通过聚类分析,筛选出10份硒响应敏感型、12份硒响应中间型和8份硒响应迟钝型的苦荞种质资源,GGE双标图分析筛选出相对综合性状最优、高产且富硒的种质YYMY002。

苦荞吸收硒元素形成富硒特性,对推动苦荞产业高质量发展具有重要意义。选择30份具有代表性的苦荞种质资源,测定其植株的农艺性状、干物质积累与转运特性以及产量与籽粒性状指标,设置叶片喷施硒肥处理,综合运用主成分分析、聚类分析和GGE双标图分析法,对不同苦荞种质的生长发育及硒响应特征进行全面评价。研究表明,不同苦荞种质资源在农艺性状、干物质积累与转运特性、产量及其构成因素、硒积累分配富集性状等方面存在较大变异,且喷施硒肥对苦荞种质资源的生长发育性状各指标均表现出促进作用。通过聚类分析,筛选出10份硒响应敏感型、12份硒响应中间型和8份硒响应迟钝型的苦荞种质资源,GGE双标图分析筛选出相对综合性状最优、高产且富硒的种质YYMY002。

The absorption of selenium by tartary buckwheat to form selenium-enriched characteristics is of great significance for promoting the high-quality development of the tartary buckwheat industry. In this study, 30 representative tartary buckwheat germplasm resources were selected to measure their agronomic traits, dry matter accumulation and translocation characteristics, as well as yield and grain trait indicators. Foliar application of selenium fertilizer was set as a treatment. Principal component analysis (PCA), cluster analysis, and GGE biplot analysis were comprehensively utilized to conduct a thorough evaluation of the growth, development, and selenium response characteristics of different tartary buckwheat germplasms. The results showed that significant variations existed among different tartary buckwheat germplasm resources in terms of agronomic traits, dry matter accumulation and translocation characteristics, yield and its components, and selenium accumulation, distribution, and enrichment traits. Furthermore, foliar selenium application exhibited a promoting effect on all measured growth and development indicators of the germplasm resources. Through cluster analysis, ten selenium-sensitive, 12 selenium-intermediate, and eight selenium-insensitive germplasm resources were screened. GGE biplot analysis identified YYMY002 as a germplasm with high yield, high selenium enrichment, and relative comprehensive superior traits.

随着人们生活水平提升及粮食产量稳步增长,农业生产正由单一高产向优质高产方向转型。农产品中的植物性食品是人体必需营养物质的重要来源,农产品质量特别是营养与健康质量日益受到重视。生命元素硒在生物体内具有多种不可替代的生物学功能[1]。在人体中,硒主要以硒代甲硫氨酸(Se-Met)和硒代半胱氨酸(Se-Cys)的形式存在,占比超过人体硒总量的50%,它们以渗入形式与蛋白质构成硒蛋白[2]。硒缺乏或过量均会引发植物和人畜生长发育相关病症,如婴幼儿缺硒会导致抵抗力降低和营养发育受阻,孕妇缺硒则可能造成胎儿畸形及妊娠高血压[3]。植物通过吸收并富集外源无机态硒,将其转化为安全有效的生物有机态硒,这是人体摄入硒的唯一途径[4]。现有研究[5]表明,人体无法自行合成硒,必须通过食物摄取,因而需要终身补硒。富硒农产品是补充人体硒元素的优质食物来源[6],有助于保障硒的充足供应,维持人体健康[7]
相较于主要粮食作物,苦荞含有更为丰富的人体必需氨基酸、蛋白质、生物类黄酮、维生素及多种天然矿物质[8]。苦荞在我国分布广泛,适应性强,能耐严寒天气与贫瘠土壤,适宜在干旱丘陵地区及凉爽气候中生长[9],主要种植于四川、云南和贵州等西南省份。四川省凉山彝族自治州作为我国最大的苦荞主产区,被誉为“苦荞麦之乡”和“中国苦荞之都”,亦是世界苦荞麦的核心起源地与遗传多样性中心[10]。凉山苦荞资源丰富、种类多样、种植广泛、产量高且品质优良,在我国独占鳌头[11]。凉山州光、热、水资源丰富,昼夜温差大,特殊的立体气候使得凉山州不同区域四季皆可种植苦荞;当地土壤质地疏松、通透性好、耕作层深厚且养分含量高,在此种植生产的苦荞成为一种稀有、天然无污染的绿色食品。苦荞在当地居民饮食结构中占据重要地位,对人体硒营养状况影响重大,苦荞中的硒在预防慢性疾病、防癌抗癌、抗衰老、抗氧化、预防心血管疾病、增强免疫力和排毒解毒等方面具有重要作用[12]。开发富硒荞麦可实现生物补硒,安全有效地预防因低硒或缺硒引发的各类病症。
目前,硒是否为植物必需营养元素尚无定论,但其在各类作物中的重要作用与潜在价值正不断被挖掘,一直备受关注。大量研究[13-14]表明,硒是植物生长发育过程中的有益元素。适量摄入硒元素,能够提高作物种子活力,刺激植株生长发育,促进新陈代谢,激发其抗氧化潜力,增强抵抗环境压力的能力,还可促进光合作用[15],同时增加荞麦的硒含量,改善荞麦籽粒的产量与质量[6]。对于低硒作物,通常采用施硒肥或喷施硒气雾剂的方法,提升粮食作物、茶叶、蔬菜和水果等植物的硒含量,这也是改善机体低硒状态的另一有效途径[16]。本研究选取具有代表性的苦荞种质资源进行叶片喷施硒肥处理,测定其农艺性状、干物质积累与运转特性、产量及其构成因素和籽粒性状等指标,通过主成分分析、聚类分析和GGE双标图分析,探究不同苦荞种质的性状指标及其对硒响应的差异。基于各苦荞种质生长发育及硒响应特性进行综合评价,以期筛选出适宜凉山州种植的富硒苦荞种质资源,为苦荞农艺性状及硒响应机制的研究提供材料基础,同时为凉山州富硒苦荞产业开发及地方经济提升提供理论依据。
30份供试苦荞种质资源由攀西特色作物研究与利用四川省重点实验室分别收集自四川、云南、贵州、内蒙古、山西和江西等省区,包括27份审定品种与3份地方种质资源,具体信息见表1。外源硒肥使用分析纯亚硒酸钠(Na2SeO3)配制,购自默克生命科学有限公司。
试验于2023年在西昌学院农业科学学院实验农场基地(27.94° N,102.21° E)进行,该地海拔1515.0 m,年均气温17.2 ℃,年均日照时数2432.1 h,年均降水量1087.5 mm,蒸发量1945.0 mm,无霜期273 d,属亚热带季风气候。供试土壤取自紫色冲积性水稻土,其质地较为黏重,土层深厚,前茬作物为花生。耕作层土壤基础理化性质为速效氮40.45 mg/kg、速效磷12.34 mg/kg、速效钾50.76 mg/kg、全硒0.02 mg/kg、有机质15.21 g/kg、pH 7.19。
于2023年4月12日进行机械开沟人工条播,采用随机区组排列。每份材料设置未喷施硒肥(C)和喷施硒肥(P)2个处理,每个处理种植1个小区,小区面积3.6 m2(1.2 m×3 m),行距40 cm,3行区,播种量为20 kg/hm2,重复3次,共计180个小区,试验区域周边设置保护行。P处理各小区均匀喷施0.1 L浓度为100 mg/L的Na2SeO3溶液,C处理各小区均匀喷施0.1 L清水,选择苦荞种质达到开花期后的晴天,于17:00进行叶面喷施,喷施过程中做好小区间隔离。种植前施足基肥,苗期定苗,适时中耕除草并追施尿素。其他田间管理参照当地常规苦荞栽培措施。
待不同处理下的苦荞籽粒达到80%以上成熟时进行收获,依据《荞麦种质资源描述规范和数据标准》[17],随机选取10株测定农艺性状指标(株高、茎粗、主茎节数、主茎叶数、一级分枝数和二级分枝数)。
于苦荞成熟期,采用烘干称重法在每个小区选取具有代表性的5株,按器官分为根、茎、叶、花和籽粒,将样品杀青后烘干至恒重,测量各个器官的干物质积累量。借鉴小麦干物质积累模型[18]建立苦荞生长期营养体和籽粒干物质积累量的拟合方程,涉及指标包括营养体最大生物量、收获期营养体生物量、总干物质转运量和总干物质转运效率,其中,总干物质转运量(g/株)=营养体最大生物量-收获期营养体生物量;总干物质转运效率(%)=总干物质转运量/营养体最大生物量×100。
于苦荞达到成熟期后进行小区收获,风干脱粒处理后测产,使用TPKZ-1智能考种分析系统(浙江托普云农科技股份有限公司)测定单株粒重、单株粒数和千粒重。
参考《食品安全国家标准 食品中硒的测定》(GB 5009.93-2017)[19]中氢化物原子荧光光谱法测定苦荞硒含量;参考侯非凡等[20]和陈锦平等[21]的方法计算硒积累量与转运量:硒利用系数(%)=籽粒硒积累量/植株硒积累量×100,硒分配系数(%)=籽粒硒积累量/营养器官硒积累量×100,硒吸收系数(%)=(P处理植株硒积累量-C处理植株硒积累量)/施用硒肥量×100,硒富集系数(%)=(P处理籽粒硒积累量-C处理籽粒硒积累量)/施用硒肥量×100。
采用Excel 2019软件进行数据处理和描述性统计。采用SPSS 21.0软件进行相关性分析、主成分分析、聚类分析及GGE双图标分析等。采用Excel 2019、Origin 8.0和R 4.5.0软件绘制表达数据分析结果图表。
不同苦荞种质的株高、茎粗、主茎节数、主茎叶数、一级分枝数和二级分枝数存在较大差异(表2),变异系数均高于20%,P处理仅对苦荞株高影响显著,对其他农艺性状无显著影响。C处理下30份苦荞种质的株高和茎粗分别在38.90~ 100.60 cm和1.64~4.40 mm,P处理下则为45.94~ 102.66 cm和1.87~4.88 mm。C处理下主茎节数和主茎叶数分别在10.50~21.60和4.70~16.80,P处理下则为10.60~22.00和5.20~17.60。C处理下一级分枝数和二级分枝数分别在1.60~6.10和1.60~ 6.50,P处理下则为1.00~6.60和2.20~6.90。
30份供试苦荞种质的产量在不同处理间均存在较大差异(表3)。P处理下,编号16(黔苦5号)产量最高,编号25(昭苦2号)产量最低。C处理下,各种质产量在322.85~1674.10 kg/hm2,平均产量857.18 kg/hm2,变异系数59.86%。P处理下,各种质产量在341.55~1759.20 kg/hm2,变异系数64.02%,平均产量944.90 kg/hm2,平均增产10.23%。30份种质资源施用硒肥后的增产率在-8.63%~61.31%,其中26份种质产量增高,编号12(迪苦2号)增产率最高,4份种质产量降低,编号17(六苦2号)降幅最大,处理间差异不显著。
不同苦荞种质的籽粒性状均存在较大差异。C处理下,千粒重、株粒重和株粒数分别在6.29~9.07 g、0.16~1.70 g和23.90~147.10,均值分别为8.01g、0.72g和76.42;P处理下则分别在5.11~10.33 g、0.23~1.90 g和19.20~158.60,均值分别为8.40 g、0.76 g和80.05。20份苦荞种质的千粒重在P处理下增加,其中3份种质资源(编号12、13和16)显著增加;10份种质千粒重降低,其中编号20(贵米苦)显著降低;不同处理间无显著差异。21份苦荞种质的株粒重在P处理下增加,6份种质降低,3份种质持平,处理间差异不显著。22份苦荞种质的株粒数在P处理下提高,6份种质下降,2份种质持平,处理间差异不显著。
分析不同生育阶段的干物质积累、分配与转运规律,有助于揭示苦荞种质的生长进程、营养状况及产量潜力。由图1a~c可知,苦荞种质的总干物质积累量随生育期推进均呈上升趋势。C处理下,各种质盛花期总干物质积累量的变异系数达52.56%,均值4.72 g/株;灌浆期变异系数为46.40%,均值9.36 g/株;成熟期变异系数为49.34%,均值13.52 g/株。P处理下,各种质盛花期总干物质积累量变异系数为52.56%,均值3.72 g/株;灌浆期变异系数为47.89%,均值9.36 g;成熟期变异系数为51.7%,均值14.90 g/株。
分析苦荞种质在各生育时期的总干物质量积累量及硒响应差异(图1d),P处理对苦荞种质各生育时期的总干物质积累量均无显著影响,但在不同生育时期其影响有一定差异。P处理下,70%的供试种质在盛花期总干物质积累量减少,在灌浆期则有53.33%的种质减少,成熟期仍表现降低的种质仅占26.67%。P处理下,全生育期内总干物质积累量较C处理始终呈增加趋势的包括编号1、17、23、27和28,始终呈降低趋势的则有编号4、5、7、12、20、21和25。
C处理下,各苦荞种质的营养体最大生物量、收获期营养体生物量、总干物质转运量和总转运效率存在较大差异,均值分别为9.34 g/株、6.75 g/株、2.59 g/株和29.29%,变异系数分别为48.45%、53.77%、56.20%和29.85%;P处理下,均值分别为10.31 g/株、7.25 g/株、3.07 g/株和31.28%,变异系数分别为49.10%、54.97%、56.86%和31.98%(表4)。综上,P处理可有效促进苦荞种质资源的干物质转运,对总干物质转运量影响极显著。
成熟期苦荞各器官的硒含量对其营养价值与保健作用至关重要,通过合理施用硒肥可有效提升苦荞的硒含量。C处理下,苦荞种质根部、茎秆、叶片和籽粒的硒含量均值分别为0.0086、0.0369、0.0495和0.0414 mg/kg;P处理下各器官硒含量均有所增加,硒含量均值分别增至0.0108、0.0489、0.0830和0.0727 mg/kg,其中对茎秆(P<0.05)、叶片(P<0.01)和籽粒(P<0.01)硒含量的提升效果更为显著(图2)。多数苦荞种质各器官硒含量总体表现为叶片>籽粒>茎秆>根部;C处理下叶片与籽粒硒含量基本持平,均略高于茎秆,根部硒含量最低;P处理下叶片和籽粒硒含量极显著增加,茎秆硒含量仅为叶片和籽粒的1/2,根部硒含量仍处于最低水平。不同处理下,各种质中编号14、15、22、24和28各器官硒含量均表现优良,而编号11和12表现较差。
图3可知,C处理下苦荞种质的全株硒积累量在0.0964~1.8201 μg/株,P处理下在0.2008~ 2.7160 μg/株,P处理较C处理显著提高。苦荞种质中编号28、14、6、7、13、29、27、16和15在C处理下全株硒积累量均高于0.6000 μg/株,P处理下均高于1.1000 μg/株;而编号1、21、26和12在C处理下全株硒积累量均低于0.2000 μg/株,P处理下有所提高,但仍低于0.3000 μg/株。C处理下,苦荞地上部(茎+叶+籽粒)硒积累量在0.0960~1.7851 μg/株,P处理下显著提高至0.2507~2.6541 μg/株;不同处理下,各种质中均以编号28(YYMY002)的地上部硒积累量最高,编号11(迪苦1号)最低。各种质中,编号26、21、19、23、18、25和24的籽粒硒积累量占比较高,可作为富硒苦荞种质资源。
C处理下,苦荞硒利用和分配系数均值分别为30.09%和45.12%,编号26(晋苦12号)表现最佳,编号20(贵米苦)最差;P处理下极显著提高至38.13%和65.17%,编号21(凤苦2号)表现最佳,编号29(DCAN003)最差;不同处理下,编号21、19和23的硒利用和分配系数均较高(表5)。各种质间硒吸收和富集系数差异较大,均值分别为25.14%和11.89%。编号28、27、26、6和29的硒吸收系数较高,编号13、16、14、9和7的硒富集系数较高。
对30份苦荞种质的21个性状进行主成分分析(图4),保留前5个主成分,累计贡献率达85.881%。农艺性状指标中株高、茎粗、主茎节数和主茎叶数,干物质积累指标中营养体最大生物量、收获期营养体生物量,以及硒积累量、硒吸收量与富集指标在PC1中具有较高载荷;籽粒性状指标中千粒重、株粒重和株粒数,以及硒利用与分配指标在PC2中具有较高载荷;分枝数在PC3中载荷较高;总干物质转运量和总干物质转运率在PC4中载荷较高;产量在PC5中载荷较高。
基于主成分分析特征值和载荷值计算不同苦荞种质各性状指标的隶属函数值和权重,进而获得综合评价值D,根据D值将参试种质分为3类,其中编号28、6、13、19、14、30、15、29、16和27共10个种质为类群Ⅰ,其D值最大,属于硒响应敏感型群体,占比33.33%;编号7、24、2、10、3、4、11、18、20、23、5和8共12个种质为类群Ⅱ,属于硒响应中间型群体,占比40.00%;编号17、9、12、26、1、21、22和25共8个种质为类群Ⅲ,其D值最小,属于硒响应迟钝型群体,占比26.67%(图5)。
基于3个苦荞类群的产量、干物质总转运率、硒吸收系数、硒富集系数及籽粒硒积累量进行聚类分析的反向验证。其中,类群Ⅰ的产量(1044.86 kg/hm2)显著高于类群Ⅱ(852.10 kg/hm2)和类群Ⅲ(598.49 kg/hm2),其干物质总转运率(41.95%)同样显著高于类群Ⅱ(40.10%)和类群Ⅲ(40.29%),硒吸收系数、硒富集系数及籽粒硒积累量均表现为类群Ⅰ极显著高于其他类群。
GGE双标图选用主成分分析的前2个主成分作为横纵坐标,二者分别解释47.60%和15.54%的变异(图6)。不同苦荞种质各性状硒响应双标图共划分为5个分区,21个相关性状位于4个分区;一级分枝数、二级分枝数、株粒数、株粒重、硒富集系数、硒吸收系数、籽粒硒积累量、地上部硒积累量和植株硒积累量为Ⅰ区,可反映苦荞生产及硒响应性能,该区域种质在上述指标中综合表现较优,其中编号28(YYMY002)和13(云荞2号)表现最优;株高、茎粗、主茎节数、主茎叶数、营养体最大生物量、收获期营养体生物量和总干物质转运量为Ⅱ区,其中编号6(西荞4号)综合表现较好;产量、千粒重、硒利用系数和硒分配系数为Ⅲ区,其中编号15(黔苦2号)和24(凉6A)综合表现较好;总干物质转运效率则单独为Ⅳ区,编号20(贵米苦)在该区域表现较好,但其硒响应性能表现不佳;Ⅴ区无性状指标呈现,该区域种质所测定指标的综合表现相对较差,编号25(昭苦2号)表现最差。
以理想种质为圆心做多层次同心圆,供试种质越接近同心圆中心则代表其综合表现越好。硒响应敏感型种质多数接近圆心点,硒响应中间型种质居中,硒响应迟钝型种质均远离圆心点,说明聚类分析与GGE双标图分析结果基本一致。其中,编号28(YYMY002)最接近圆心点,综合表现最佳,其次为编号6、13、14、15、16、19和27,而编号1、21、22和25则远离圆心点,综合表现最差。
本研究在筛选苦荞种质资源的农艺性状、干物质积累转运特性、产量及其构成因素和硒积累转运富集性状的基础上,通过叶片喷施硒肥处理,对比分析了不同苦荞种质资源在上述指标上的硒响应基础机理。结果表明,供试苦荞种质在农艺性状和产量构成因素上存在较大差异,无论是否喷施硒肥,株高、茎粗、主茎节数、主茎叶数、一级分枝数、二级分枝数、千粒重、株粒数、株粒重、产量及干物质积累量等指标的变异范围较广,变异系数均超过20%;盛花期、灌浆期和成熟期总干物质积累量的变异系数均在50%左右,与前人[22-25]研究结果一致,表明供试苦荞种质资源来源广泛,良种改良繁育潜力大,且农艺性状、生物积累量和产量构成因素的变异程度较高,可为高产优质苦荞选育提供了良好的材料基础。
目前,关于施用硒肥促进农作物生长发育及高产优质的研究[26-32]较多,多数结论表明施用硒肥对农作物农艺性状及产量构成因素等指标均有影响,但受施硒量、施硒方式、作物品种及试验环境条件等因素影响,各指标受影响的程度和表现存在差异。本研究结果表明,除个别情况外,喷施硒肥对苦荞种质资源的农艺性状、干物质积累转运、产量及其构成因素均具有促进作用。
苦荞器官的硒含量对其营养价值与保健功能具有关键作用。研究[7,15,33-36]通过合理施用硒肥及采用适宜的喷施方式,有效提升了苦荞对硒元素的吸收、积累、转运与富集能力,进而增强了富硒苦荞产品的经济效益与市场竞争力。前人[37-40]研究表明,适量、适时及适宜方法的硒肥施用可增加农作物的硒含量。本试验在探究苦荞种质资源对硒元素积累利用的基础上,通过叶片喷施硒肥处理,揭示了不同苦荞种质资源对硒元素的积累分配特性及其富集效果,旨在筛选出富硒能力强的苦荞种质资源。本研究结果显示,未施硒肥的苦荞种质在成熟期时,其根部、茎秆、叶片和籽粒的硒含量均值分别为0.0086、0.0369、0.0495和0.0414 mg/kg;而喷施硒肥可显著提高苦荞成熟期茎秆、叶片及籽粒的硒含量,并显著增加叶片和籽粒的硒积累量,但对根部和茎秆的硒积累量影响不显著,这一结果与刘铖[7]和董石峰等[41]的研究相似。此外,喷施硒肥处理还能显著提升硒的利用系数和分配系数。
前人[42-44]研究多采用主成分分析及聚类分析方法,从不同角度对农作物的农艺性状、产量及质量进行综合评价,对于苦荞而言,针对种质资源农艺性状的主成分分析和聚类分析研究较多,但针对农作物对硒元素利用、吸收和富集指标的研究则相对较少,尤其是应用GGE双标图法的研究更少。本研究发现,30份供试苦荞种质的21个性状指标间均存在不同程度和方向的相关性,通过隶属函数法进行主成分分析综合评价,获得5个主成分,其累计贡献率达85.881%;结合聚类分析,将30份苦荞种质资源划分为硒响应敏感型群体、硒响应中间型群体和硒响应迟钝型群体,占比分别为33.33%、40.00%和26.67%;在聚类分析的基础上,选取5个指标进行反向验证,证实了供试苦荞种质资源分类的客观性与可行性。本研究通过科学合理的综合评价方法,从总体水平和指标体系2个维度全面反映了供试种质资源的特性,且不同评价方法相互印证,确保了结果的客观性、真实性与有效性,为富硒苦荞新良种的培育提供了理论依据。
运用主成分分析与聚类分析法对30份苦荞种质资源的农艺性状、干物质积累转运特性、产量及其构成因素、硒积累转运富集利用性状和硒响应特性进行综合评估。筛选出10份硒响应敏感型、12份硒响应中间型和8份硒响应迟钝型苦荞种质资源。采用GGE双标图对供试苦荞及其性状指标进行分析,不同扇区内各种质表现存在差异,YYMY002综合性状表现最优。
