To investigate the resistance levels to currently prevalent races and new isolates, as well as the distribution of stripe rust resistance genes in released and candidate wheat varieties from Longnan, Gansu, 73 wheat varieties (lines) were evaluated for seedling resistance against the prevalent races CYR32, CYR34, and ZS asexual isolate and ZS sexual isolate. Meanwhile, molecular markers linked to all-stage resistance genes Yr5, Yr9, Yr10, Yr15, Yr26, and the adult-plant resistance gene Yr18 were utilized for molecular detection. The results indicated that among the 73 tested wheat varieties (lines), 11 (15.07%), 11 (15.07%), 13 (17.81%), and nine (12.33%) exhibited seedling resistance to CYR32, CYR34, and the asexual and sexual isolates of ZS, respectively. Among all tested materials, only Zhongliang 14 exhibited seedling resistance to all the provided isolates of stripe rust. Molecular detection results revealed that the number of wheat varieties (lines) carrying Yr5, Yr9, Yr10, Yr18, and Yr26 was 1, 27, 3, 3, and 16, respectively, accounting for 1.37%, 36.99%, 4.11%, 4.11%, and 21.92% of the total tested materials. Three materials carried two resistance genes, and Zhongliang 14 carried three resistance genes. The Yr15 gene was not detected in any of the tested varieties (lines), and 28 materials carried none of the six tested resistance genes.

为了解甘肃陇南小麦育成品种和后备品种对当前流行小种和新菌系的抗性水平及其抗条锈基因的分布情况,利用当前流行小种CYR32、CYR34及ZS无性菌系、ZS有性菌系对甘肃陇南73份小麦品种(系)进行苗期抗病性鉴定,并利用已知的全生育期抗条锈病基因Yr5Yr9Yr10Yr15Yr26和成株期抗条锈病基因Yr18的分子标记对其进行分子检测。结果表明,73份供试小麦品种(系)中,对CYR32、CYR34、ZS无性菌系及ZS有性菌系表现苗期抗性的分别有11(15.07%)、11(15.07%)、13(17.81%)和9份(12.33%),所有供试品种(系)中,仅有中梁14号对提供的所有条锈菌表现苗期抗性。分子检测结果表明,供试材料中携带Yr5Yr9Yr10Yr18Yr26的小麦品种(系)分别有1、27、3、3和16份,分别占供试材料总数的1.37%、36.99%、4.11%、4.11%和21.92%;有3份材料携带2个抗性基因,中梁14号携带3个抗性基因;所有供试品种(系)中均未检测到Yr15基因,有28份材料未检测到供测的6个抗条锈病基因。

为了解甘肃陇南小麦育成品种和后备品种对当前流行小种和新菌系的抗性水平及其抗条锈基因的分布情况,利用当前流行小种CYR32、CYR34及ZS无性菌系、ZS有性菌系对甘肃陇南73份小麦品种(系)进行苗期抗病性鉴定,并利用已知的全生育期抗条锈病基因Yr5Yr9Yr10Yr15Yr26和成株期抗条锈病基因Yr18的分子标记对其进行分子检测。结果表明,73份供试小麦品种(系)中,对CYR32、CYR34、ZS无性菌系及ZS有性菌系表现苗期抗性的分别有11(15.07%)、11(15.07%)、13(17.81%)和9份(12.33%),所有供试品种(系)中,仅有中梁14号对提供的所有条锈菌表现苗期抗性。分子检测结果表明,供试材料中携带Yr5Yr9Yr10Yr18Yr26的小麦品种(系)分别有1、27、3、3和16份,分别占供试材料总数的1.37%、36.99%、4.11%、4.11%和21.92%;有3份材料携带2个抗性基因,中梁14号携带3个抗性基因;所有供试品种(系)中均未检测到Yr15基因,有28份材料未检测到供测的6个抗条锈病基因。

小麦品种(系)  /  条锈病  /  抗条锈基因  /  分子检测

To investigate the resistance levels to currently prevalent races and new isolates, as well as the distribution of stripe rust resistance genes in released and candidate wheat varieties from Longnan, Gansu, 73 wheat varieties (lines) were evaluated for seedling resistance against the prevalent races CYR32, CYR34, and ZS asexual isolate and ZS sexual isolate. Meanwhile, molecular markers linked to all-stage resistance genes Yr5, Yr9, Yr10, Yr15, Yr26, and the adult-plant resistance gene Yr18 were utilized for molecular detection. The results indicated that among the 73 tested wheat varieties (lines), 11 (15.07%), 11 (15.07%), 13 (17.81%), and nine (12.33%) exhibited seedling resistance to CYR32, CYR34, and the asexual and sexual isolates of ZS, respectively. Among all tested materials, only Zhongliang 14 exhibited seedling resistance to all the provided isolates of stripe rust. Molecular detection results revealed that the number of wheat varieties (lines) carrying Yr5, Yr9, Yr10, Yr18, and Yr26 was 1, 27, 3, 3, and 16, respectively, accounting for 1.37%, 36.99%, 4.11%, 4.11%, and 21.92% of the total tested materials. Three materials carried two resistance genes, and Zhongliang 14 carried three resistance genes. The Yr15 gene was not detected in any of the tested varieties (lines), and 28 materials carried none of the six tested resistance genes.

小麦条锈病是由条形柄锈菌小麦专化型(Puccinia striiformis f. sp. tritici)引起的气传性真菌病害,其在我国发生面积广且流行成灾率高,危害十分严重。据统计[1-2],1950-2020年小麦条锈病在全国发生8次大规模流行,发生面积在333.3万~ 1000万hm2,防治后造成小麦减产共计约140亿kg。甘肃陇南(包括天水市和陇南市)因特殊地理环境和气候因素,成为我国小麦条锈病发生流行最重要的越夏区和病菌新毒性小种的“策源地”,小麦条锈菌在该区既能越夏又能越冬,完成周年循环[3-4]。已有研究[5]发现,甘肃陇南地区的野生小檗是小麦条锈菌的转主寄主,为新小种的产生和条锈菌的有性生殖提供了有利条件,并且在条锈病的发生中起到提供初始菌源的作用。
种植抗病品种是防治小麦条锈病最经济、有效和环保的措施,由于条锈菌毒性变异速度快,抗病品种一般种植3~5年便会丧失抗病性,自2009年以来,由于CYR34的出现并逐渐上升为第一优势小种[6],导致甘肃陇南育种中广泛利用的携带Yr10Yr26的贵农系和南农92R系等抗源材料和衍生系品种逐渐丧失抗病性[7]。甚至在2021年,由于CYR34、CYR32及新菌系ZS(中四)的联合作用,导致甘肃陇南小麦产量损失严重[8]
国际已正式命名的86个抗条锈基因[9]中,仅有全生育期基因Yr5Yr15Yr61及成株期基因Yr18Yr29Yr46等仍保持抗性且持续稳定[10-11]。甘肃陇南是我国重要的小麦条锈菌越夏菌源基地,条锈菌有性生殖和无性生殖并存[12],尽管前人在小麦抗条锈性评价方面开展了相关研究,但利用当前优势小种CYR34、CYR32及新菌系ZS(无性和有性)对甘肃陇南生产品种及后备品种进行抗病性研究的报道较少。基于此,本研究对甘肃陇南生产品种及后备品种进行抗条锈性鉴定及抗条锈基因的分子检测,旨在明确供试材料抗条锈性及抗病基因的分布,为小麦抗条锈育种和品种合理布局提供理论依据。
供试材料为甘肃陇南育成的冬小麦品种(系),共计73份,由天水市农业科学研究所收集与繁存。阳性对照材料为Yr5/6*Avocet S、Yr9/6*Avocet S、Yr10/6*Avocet S、Yr15/6*Avocet S、Yr18/6*Avocet S和Yr26/6*Avocet S,阴性对照材料为Avocet S,均由甘肃农业大学提供,感病对照品种铭贤169由甘肃省农业科学院植物保护研究所收集和繁殖。供试病原菌为小麦条锈菌生理小种CYR32、CYR34及条锈菌ZS无性和有性菌系,由甘肃省农业科学院植物保护研究所小麦病害课题组提供。
苗期抗性鉴定于2022年3-5月在甘肃省农业科学院植物保护研究所兰州温室完成。每个供试品种(系)挑选籽粒饱满的8~10粒种子,将其播种于直径8 cm、高10 cm的圆形黑色塑料盒中,每个盒子种植4份材料,待幼苗长至2叶1心期时,采用抖孢子粉法[13]接种预先繁殖好的条锈菌生理小种CYR32、CYR34及ZS无性菌系和ZS有性菌系,接种后的幼苗置于10~15 ℃黑暗条件下保湿24 h,之后置于一定温度、光照和光强的温室诱导发病,待感病对照品种铭贤169充分发病后,按照国家农业行业标准(NY/T 1443.1-2007)[14]调查记录病害的侵染型,即0(免疫)、0;(近免疫)、1(高抗)、2(中抗)、3(中感)和4级(高感),0~2为抗病,3~4为感病。
采用改良的CTAB法[15-16]对73份小麦品种(系)、6份阳性对照品系及阴性对照品系的幼叶提取全基因组DNA,选用国内外现已开发的全生育期抗条锈病基因Yr5Yr9(1BL/1RS)、Yr10Yr15Yr26及成株期抗条锈病基因Yr18的SSR、SCAR或STS标记分别对供试材料进行分子检测,所用引物由北京赛百盛基因技术有限公司合成,PCR反应体系为10 μL,包括:5 μL 2×Master Mix、2 μL ddH2O、上下游引物各1 μL、模板DNA 1 μL(50 ng/μL)。扩增程序为:94 ℃预变性4 min;94 ℃变性50 s,58~62 ℃退火50 s(依据引物的退火温度),72 ℃延伸50 s,共35个循环;最后72 ℃延伸10 min。扩增产物经1.8%~2.0%的琼脂糖凝胶电泳后在胶片观察灯上拍照检测。标记类型、基因名称及引物序列见表1
采用Excel 2010进行数据分析与作图。
利用条锈菌流行小种CYR32、CYR34及条锈菌ZS无性菌系和ZS有性菌系对73份甘肃陇南小麦品种(系)进行苗期抗病性鉴定,结果见图1表2。对CYR32表现抗病的有11份,占15.07%,其中表现免疫、近免疫和中抗的分别有9、1和1份,表现感病的有62份,占84.93%。对CYR34表现抗病的有11份,占15.07%,其中表现免疫和中抗的分别有10和1份,表现感病的也有62份。对ZS无性菌系表现抗病的有13份,占17.81%,其中表现免疫、近免疫和中抗的分别有7、2和4份,表现感病的有60份,占82.19%。对ZS有性菌系表现抗病的有9份,占12.33%,其中表现免疫和近免疫的分别有8和1份,其余64份材料表现感病,占87.67%。在供试品种(系)中,中梁14号、中梁28号、兰天17号和兰天37号对CYR32和CYR34均表现抗性,占5.48%,中梁14号和天选81号对ZS无性和有性菌系均表现抗性,占2.74%,仅中梁14号对CYR32、CYR34、ZS无性菌系和ZS有性菌系均表现抗性,占1.37%。
利用Xwmc175、AF1/AF4、SC200、Xbarc8、csLV34及We173标记对73份品种(系)进行抗条锈病基因分子检测,结果表明,中梁14号可能携带Yr5基因(图2),占供试材料的1.37%;中梁21号、天选39号和兰天10号等27份材料可能携带Yr9基因(图3),占供试材料的36.99%;中梁12号、中梁16号和中梁18号3份材料可能携带Yr10基因(图4),占供试材料的4.11%;中梁14号、中梁16号和中梁28号3份材料可能携带Yr18基因(图5),占供试材料的4.11%;中梁29号、天选72号和兰天17号等16份材料可能携带Yr26基因(图6),占供试材料的21.92%。3份材料可能携带2个抗条锈病基因,其中中梁16号携带Yr10Yr18基因,天选39号和兰天61号携带Yr9Yr26基因;中梁14号携带3个抗条锈病基因Yr5Yr9Yr18。28份材料(系)未检测到上述6个抗条锈病基因。所有供试材料抗条锈基因分子检测结果见表2
甘肃陇南越夏区在我国小麦条锈病流行体系中起着重要的作用,多年来,陇南越夏区推广种植的兰天系、中梁系和天选系小麦品种在甘肃陇南及黄淮麦区小麦条锈病的控制与保障粮食安全中发挥了重要作用。由于甘肃陇南条锈菌无性繁殖与有性繁殖长期共存,使得条锈菌毒性变异速度快,品种抗条锈病性丧失速度更快。
根据苗期抗病性鉴定结果可知,73份小麦品种(系)中,对当前优势小种CYR32、CYR34及新菌系ZS无性和ZS有性表现苗期抗性的品种(系)较少,对CYR32和CYR34同时表现抗性的有中梁14号、中梁28号、兰天17号及兰天37号,对ZS无性和有性菌系均表现抗性的有中梁14号及天选81号,仅有中梁14号对提供的所有菌系均表现苗期抗性,这一研究结果也进一步证实了甘肃陇南育成品种(系)中,具有苗期和全生育期抗性的品种(系)较少,多数品种(系)表现成株抗性的特点[8,22]。条锈菌ZS菌系为近年发现的对鉴别寄主中四感染的新菌系,贾秋珍等[12]研究发现,甘肃陇南生产品种对ZS菌系的抵抗力弱,ZS菌系存在潜在风险。本研究发现,供试品种(系)对条锈菌ZS无性和有性菌系表现苗期抗病的有13和9份,大多数品种(系)对ZS新菌系表现苗期感病。因此,为控制新菌系在甘肃陇南麦区的持续发生和流行,确保小麦高产与稳产,新抗源材料的筛选及抗病新基因的挖掘与利用已迫在眉睫。
分子标记技术的发展为抗条锈基因的检测提供了可能与便利,是一种有效的检测方法,在小麦分子标记辅助育种中,利用与已知抗条锈基因紧密连锁的标记对检测抗条锈基因、明确目标基因的存在与否、客观且合理地利用抗病基因以及提高小麦育种效率均具有重要意义。
Yr5Yr15为小种专化型全生育期抗性基因,对我国当前流行小种仍具有抗性。目前陕西已发现对Yr5表现亲和的小种TSA-6和TSA-9,且2个小种的寄生适合度高于生理小种CYR31、CYR33和CYR34[23]。黄亮等[24]对我国79个小麦品种(系)进行分子检测发现,有4份材料携带Yr5基因;庞云星等[25]和王树和等[26]曾分别在河北和山西的89份小麦品种及四川的100份小麦品种(系)中均未检测到Yr5基因的存在。周警卫等[27]对国内外153份小麦种质进行条锈菌抗性鉴定与评价,未检测到Yr15基因;白斌等[28]对甘肃陇南和陇东的117份小麦品种进行检测,也未发现携带Yr15基因的材料。本研究检测到仅有中梁14号携带Yr5基因,没有检测到Yr15基因,说明Yr5Yr15基因目前在小麦育种中的利用率仍很低,应引起育种家们的高度重视。
Yr9来源于黑麦1BL/1RS易位系,因其易位片段携带Yr9Lr26Sr31Pm8基因而在育种中被广泛利用,随着CYR29的出现,导致生产上携带Yr9的材料因丧失抗病性而失去利用价值。黄亮等[24]对79份我国小麦品种(系)检测,发现35份材料携带Yr9,占44.3%;戴妙飞等[29]对来自ICARDA的203份小麦种质检测,发现Yr9的检出率为25.5%。本研究检测到携带Yr9的材料有27份,占比与上述结论相似。已有研究[30]表明,1BL/1RS易位导致小麦缺失了1BS上重要的低分子量麦谷蛋白和醇溶蛋白,进而被品质较差的黑麦碱取代,引起加工品质变劣,因此在育种中应谨慎使用携带Yr9基因的亲本材料。
CYR34的出现使得Yr10Yr26丧失抗病性,导致我国小麦生产再次受到条锈病的威胁。李峰奇等[31]对黄淮麦区126个小麦品种(系)抗条锈病的基因检测显示,仅4份材料可能含有Yr10。韩德俊等[32]对1980份小麦地方品种和国外种质的抗条锈性鉴定和分子标记检测表明,仅有Spanish D188和红茧儿麦可能含有Yr10。本研究检测到中梁12号、中梁16号和中梁18号3份材料携带Yr10,并且发现携带Yr10的材料为上世纪80、90年代育成品种,在之后的育成品种中未检测到Yr10的存在,说明Yr10在甘肃陇南育成品种中利用率低,与上述结论一致。黄苗苗等[33]通过分子检测发现,来自甘肃麦区的223份小麦地方品种携带Yr26的材料占比达50.22%。本研究检测到73份品种(系)中有16份材料携带Yr26,且在不同年份的育成品种中均检测到Yr26,由此可见,Yr26在育种中的利用率仍很高,需引起育种家的重视。
Yr18是成株期抗性基因,具有抗病谱广且抗性持久的特点。徐默然等[34]对103份材料进行表型鉴定,结合分子标记检测,鉴定出19份材料含有Yr18。管方念等[35]对152份地方品种进行Yr18分子标记检测,发现131份材料携带该基因。杨文雄等[36]对231份我国小麦育成品种及422份农家品种进行Yr18分子检测,发现分别有14份和359份材料携带该基因。本研究检测到有中梁14号、中梁16号及中梁28号3份材料携带Yr18,这一结果进一步表明Yr18基因在育成品种中分布频率较低。
研究[10]表明,Yr9Yr18基因在苗期对当前流行小种CYR32及CYR34表现感病,Yr26基因对CYR34已失去抗性。但本研究发现,73份供试材料中有少数材料的苗期抗病性鉴定结果与抗条锈病基因分子检测结果不一致,其中中梁26号、兰天26号、兰天34号、兰天51号和兰天61号虽携带Yr9基因,但中梁26号、兰天26号和兰天51号苗期抗CYR34,兰天34号和兰天61号苗期抗CYR32;中梁28号虽携带Yr18基因,但苗期对CYR32和CYR34均表现抗病;中梁29号、中梁43号、天选72号及兰天37号虽携带Yr26基因,但苗期对CYR34表现抗性,推测这可能与苗期接菌鉴定中人为误差以及分子检测假阳性现象的发生有关,尚需进一步研究。
国内外的育种家在认识到单一的抗病基因抗性脆弱和难以持久的基础上,均在尝试通过基因聚合分子育种手段来提高品种的抗性程度和持久性[37]。白斌等[38]利用成株期抗病基因和全生育期基因育成成株抗性品种兰天132和兰天196,为甘肃陇南麦区减轻了致病小种定向选择压力,延长了抗病品种使用年限。本研究发现,中梁14号携带3个抗条锈病基因(Yr5Yr9Yr18),并且全生育期对所有供试菌种表现抗病[22],推测原因可能是其携带全生育期抗性基因Yr5、成株期抗性基因Yr18及未知的新抗病基因,其为上世纪90年代育成品种,多年来田间抗病性保持良好,在育种中可作抗源加以利用。
甘肃陇南麦区的小麦品种(系)对当前主要条锈菌流行小种及新菌系的苗期抗性水平低,73份供试材料中,对CYR32、CYR34、ZS无性及有性菌系表现苗期抗病的材料分别有11、11、13和9份。该地区小麦品种(系)以携带Yr9(36.99%)和Yr26(21.92%)为主,主效基因Yr5Yr18的出现频率达1.37%和4.11%,Yr10出现频率为4.11%,未检测到Yr15。3份材料同时携带2个抗条锈病基因,包含2种组合方式(Yr10+Yr18Yr9+Yr26),中梁14号对提供的所有条锈菌苗期均表现抗性,并且携带3个抗条锈病基因,组合方式为Yr5+ Yr9+Yr18。在今后的小麦育种工作中应减少对Yr9Yr10Yr26的使用,尽可能利用全生育期抗条锈病基因Yr5、Yr15和成株期抗条锈病基因Yr18与其他有效抗病基因聚合来培育持久抗条锈病品种,以便提高品种的持久抗性,保障小麦生产安全。
