To investigate the effects of priming treatments on the germination and physiological characteristics of wheat seeds and to screen the optimal priming method for wheat seeds with low germination ability, Jimai 325 seeds with a germination rate of 32.67% after artificial aging treatment were used as the research object. Nine priming agents from three categories, including PEG 6000, gibberellic acid (GA3), and Vitamin B1 (VB1), were applied at different concentrations for soaking durations of 8 h, 16 h, and 24 h. The study examined the effects of various priming treatments on germination indicators, such as germination potential, germination rate, germination index, electrical conductivity, and the activities of superoxide dismutase (SOD), peroxidase (POD), and catalase (CAT). The results showed that all priming treatments significantly enhanced the germination capacity of aged wheat seeds. The priming agents had significant impacts on germination potential, germination rate, and germination index, with VB1 and VB2 showing the best priming effects. As the priming time was extended, the germination potential, germination rate, and germination index of wheat seeds all significantly increased, with the effects of 16 h and 24 h being significantly better than those of 8 h. By measuring the leachate electrical conductivity and the activities of SOD, POD, and CAT for priming combinations that increased the germination rate to over 85%, it was determined that soaking in 0.08% VB1 for 16 h is the optimal priming method for Jimai 325 wheat seeds with an initial germination rate of 32.67%.

为探讨引发处理对小麦种子萌发及生理特性的影响,筛选低发芽力小麦种子引发的最佳方法,以人工老化处理后发芽率32.67%的冀麦325小麦种子为研究对象,利用PEG6000、赤霉素GA3和维生素B1等3类共9种引发剂,采用不同浓度浸种处理8、16和24 h,研究不同引发处理对小麦种子发芽势、发芽率、发芽指数、电导率、超氧化物歧化酶(SOD)、过氧化物酶(POD)和过氧化氢酶(CAT)活性等萌发指标的影响。结果表明,不同引发处理均可显著提高老化小麦种子的发芽力。引发剂对小麦种子发芽势、发芽率和发芽指数产生显著影响,其中维生素B1和维生素B2引发效果最佳;随着引发时间的延长,小麦种子发芽势、发芽率和发芽指数均有显著提高,引发16和24 h效果显著优于8 h。对引发后小麦种子发芽率提高至85%以上引发组合的浸出液电导率和SOD、POD、CAT活性进行测定,明确0.08% VB1浸泡16 h为初始发芽率32.67%冀麦325小麦种子的最佳引发方法。

To investigate the effects of priming treatments on the germination and physiological characteristics of wheat seeds and to screen the optimal priming method for wheat seeds with low germination ability, Jimai 325 seeds with a germination rate of 32.67% after artificial aging treatment were used as the research object. Nine priming agents from three categories, including PEG 6000, gibberellic acid (GA3), and Vitamin B1 (VB1), were applied at different concentrations for soaking durations of 8 h, 16 h, and 24 h. The study examined the effects of various priming treatments on germination indicators, such as germination potential, germination rate, germination index, electrical conductivity, and the activities of superoxide dismutase (SOD), peroxidase (POD), and catalase (CAT). The results showed that all priming treatments significantly enhanced the germination capacity of aged wheat seeds. The priming agents had significant impacts on germination potential, germination rate, and germination index, with VB1 and VB2 showing the best priming effects. As the priming time was extended, the germination potential, germination rate, and germination index of wheat seeds all significantly increased, with the effects of 16 h and 24 h being significantly better than those of 8 h. By measuring the leachate electrical conductivity and the activities of SOD, POD, and CAT for priming combinations that increased the germination rate to over 85%, it was determined that soaking in 0.08% VB1 for 16 h is the optimal priming method for Jimai 325 wheat seeds with an initial germination rate of 32.67%.

小麦(Triticum aestivum L.)是重要的粮食作物,全球小麦种质资源约85万份,我国长期保存的有4.9万份[1]。丰富的种质资源为新品种培育提供了重要的物质基础。然而,目前仍有部分小麦资源分散保存于农户、育种团队和种子企业等。在种质资源收集保护过程中,存在种子量少、存放年份久和发芽率低等问题,常导致珍贵资源流失或损耗。
种子引发(seed priming)是基于种子萌发的生物学机制提出的种子处理方法。该处理是将种子置于具有一定渗透势的溶液中,使其缓慢吸水,以减少吸胀损伤,促进种子萌发,提高萌发时间的稳定性和萌发整齐度[2]。对低发芽力种子进行引发处理,可提升种子萌发速度和质量,进而增强种子活力,改善后期田间生产幼苗品质,提高苗期抗逆性。将引发种子用于大田直播,出苗快且增产显著,可替代苗期移栽,节省繁种和生产成本[3],使种子具备明显的生长优势和生产潜力[4]
种子引发方法包括渗透调节、基质引发和滚筒引发[5]。其中,渗透引发是利用渗透调节物质控制介质渗透压以调节种子吸水的方法,也是最为常用的引发方法[6]。常用的渗透调节种子引发剂有聚乙二醇(PEG)、无机盐溶液、植物生长调节剂和维生素等[7]
PEG作为一种高分子渗透调节剂,可促进植物细胞膜修复,加快种子萌发速度并提高种子发芽率,且对种子无毒害作用[8]。相关研究[8]表明,PEG引发处理能够提升野菊花、白菜、远志、燕麦、紫苏、大麦和玉米等种子的发芽率,降低种子电解质泄漏率,同时提高超氧化物歧化酶(SOD)、过氧化物酶(POD)和过氧化氢酶(CAT)等酶的活性[9-11]。在植物生长调节剂中,吲哚乙酸(IAA)、赤霉素(GA3)和6-苄基腺嘌呤(6-BA)等对种子萌发具有显著影响。适宜浓度的IAA浸种处理可提高香椿种子的发芽率,增加幼苗生长量和干物质积累[12-13]。GA3引发处理可提高种子发芽率[14-16],保护细胞膜的完整性[16],增强种子内SOD和POD的活性,减轻质膜氧化损伤[17]。6-BA可促进种子出苗和分蘖芽的产生[14],提高植株幼苗的抗逆能力[15]。维生素是植物体内酶促反应的辅酶或辅酶的重要组成成分,用维生素处理植物种子能够显著提高种子的发芽势和发芽率[18-20]。上述研究多采用单个引发剂、多个浓度或多个引发剂、单一浓度对单一作物种子进行引发处理,而利用多种引发剂、不同引发浓度和引发时间对种子进行综合引发的研究鲜见报道。
本研究以低发芽力的冀麦325(发芽率33.67%)种子作为研究对象,选用渗透调节剂PEG 6000、植物生长调节剂(GA3、IAA和6-BA)以及维生素(VB1、VB2、VB6、VB12和Vc)这3类共9种引发剂溶液,对小麦种子开展渗透处理。通过分析不同引发剂、不同浓度及不同浸泡时间对低发芽力小麦种子萌发和生理指标的影响,筛选出适宜低发芽力小麦种子的最佳引发方法,并对比种子引发处理前后小麦种子萌发及生理生化指标的变化,旨在为小麦种质资源中濒危种子的拯救、安全保存与创新利用提供理论依据。
供试材料为小麦品种冀麦325,由河北省农林科学院粮油作物研究所小麦育种研究室提供。
试验于2023年4-7月在河北省农林科学院粮油作物研究所试验室进行。选取籽粒饱满、大小、形状和成熟度一致的种子分置于纱袋中,保持透气,于SRG-500A人工气候箱(杭州硕联仪器有限公司)中老化,老化条件为40 ℃、相对湿度75%,经30 d老化处理后,获得发芽率为32.67%的种子作为引发试验的初始材料。
试验设置引发剂、引发浓度和引发时间3个因素的小麦引发试验。3类9种引发剂,包括PEG 6000、GA3、IAA、6-BA、VB1、VB2、VB6、VB12和Vc;设置低、中和高浓度3种引发浓度,各引发剂和引发浓度见表1;引发时间设为8、16和24 h。共81个处理,不经引发剂处理的自然生长小麦种子为对照(CK),每处理设3次重复,每个重复设50粒小麦种子进行引发。引发处理后,将种子置于CTB870D种子干燥箱(河北正大怡和科技有限公司)中进行干燥,种子含水量达到7%~8%后进行发芽率检测和生理指标测定。
按照国际种子检验规程[21]进行标准发芽试验,发芽试验在ZLC-1500A种子发芽箱(杭州硕联仪器有限公司)中进行,发芽条件为20 ℃,无光照,每重复50粒种子,3次重复,计算发芽势、发芽率和发芽指数:发芽势(%)=(第3天正常发芽种子数/供试种子粒数)× 100;发芽率(%)=(第7天正常发芽种子数/供试种子粒数)×100;发芽指数(GI)=∑(Gt/Dt),式中,Dt为发芽日数,Gt为与Dt相对应的每天发芽种子数。
参照朱银等[22]的方法测定电导率,每个样品设3次重复,每次重复随机选取50粒大小均匀且无损伤的种子,用去离子水冲洗3次,用滤纸吸干浮水,将种子置于洁净的150 mL烧杯中,加入100 mL去离子水,于20 ℃恒温条件下浸泡24 h,用SG3电导率仪(梅特勒-托利多,瑞士)测定浸泡液的电导率。以去离子水为对照,每重复实际电导率=重复电导率读数-对照读数。
采用氮蓝四唑(NBT)还原法[19]测定SOD活性,以反应被抑制50%时的酶液用量作为1个酶活性单位;采用愈创木酚法[23]测定POD活性;采用紫外吸收法[24]测定CAT活性。
数据为3次重复的平均值±标准误;使用SPSS 23.0一般线性模型进行多因素邓肯方差分析;使用Excel 2016软件进行计算和制图。
为确定对小麦种子引发效果影响显著的因素,对不同引发处理后小麦种子的发芽势、发芽率和发芽指数进行显著性测验(表2)。不同引发剂和引发时间对发芽势、发芽率和发芽指数影响均达极显著水平;引发剂×引发时间对发芽势、发芽率和发芽指数的影响达极显著水平;提高小麦种子萌发效果最显著的因素为引发剂,引发时间次之。
9种引发剂对初始发芽率32.67%小麦种子的发芽势、发芽率和发芽指数均产生显著影响(图1)。其中,PEG 6000对小麦种子发芽势的提升最大,从10.00%提高至35.48%;VB2对发芽率提升最大,从32.67%提高至79.78%;VB1对发芽指数提升最大,从3.94提高至14.03。
3个引发时间对小麦种子发芽势、发芽率和发芽指数的影响显著(图2)。引发处理16 h对小麦种子发芽势、发芽率和发芽指数的提升最大,分别从10.00%提高至32.99%,从32.67%提高至76.54%,从3.94提高至12.74。
综上,VB1和VB2对小麦种子萌发的促进效果较好;引发16 h和24 h效果显著高于8 h,其中引发16 h效果最佳。
基于前文分析结果,对VB1和VB2各组合进一步分析(表3)可知,VB1和VB2各组合均可显著提高种子发芽力,促进种子萌发,其中利用0.04%和0.08%浓度的VB1浸泡24 h后发芽势提升效果最佳,从10.00%提高至77.33%;0.04% VB2浸泡16 h后发芽率提升效果最佳,从32.67%提高至90.00%;0.02% VB2浸泡16 h后发芽指数提升效果最佳,从3.94提高至17.13。
发芽率≥85%为合格小麦种子[25],经引发后种子发芽率≥85%的引发处理有4个,分别为T9、T10、T11和T12处理。
比较发芽率≥85%的4个引发处理的种子浸出液电导率,结果(图3)表明,引发处理后小麦种子电导率较CK均显著降低,各处理间差异不显著,其中T9处理引发后电导率降幅最大,从80.76 μS/cm降至23.46 μS/cm。
对引发处理后小麦种子发芽率≥85%的4个处理进行SOD、POD和CAT活性测定。经4个引发处理后,SOD活性均显著高于CK处理,各处理间影响不显著,其中T9处理引发后的SOD活性增幅最大,从109.33 U/g增至365.67 U/g(图4)。引发处理后小麦种子POD活性如图4b所示,各处理均较CK显著提高,其中T9、T10和T11处理提高效果优于T12处理,T10处理引发后POD活性最高,POD活性从322.00 U/g提高到2942.67 U/g。
各处理对小麦种子CAT活性的影响(图4c)表明,经4种引发处理后,CAT活性均显著提高,但4个处理间无显著差异,其中,T9处理引发后CAT活性为76.17 mg/g,T10处理引发后CAT活性为77.19 mg/g。
种子老化是贮藏过程中普遍存在的现象,种子引发可有效提升多种作物种子的活力[26]。在小麦种子引发方面,前人采用不同方法均取得了一定成效,利用10% PEG 6000浸泡24 h,可使小麦种子发芽率从72%提升至90%[27];用6 mmol/L氯化钙浸泡12 h,发芽率从83.33%提高到96.00%[28];0.04%壳聚糖浸泡1 h,发芽率从81%升至92%[29];0.08%氨苄青霉素浸泡24 h,发芽率从75%提高到88%[30]。且上述研究所用小麦材料的初始发芽率均高于70%。本试验采用0.08% VB1浸泡16 h,将初始发芽率仅为32.67%的小麦种子发芽率提高至90.67%,引发后老化种子发芽率达到健康种子水平(发芽率≥85%),引发效果较上述研究更为显著。
本研究发现,VB1和VB2的引发效果显著优于其他7种引发剂。程梓祺等[18]研究表明,0.01%VB1浸泡甜菜种子24 h后,其浸出液相对电导率从104.66 μS/cm降至53.33 μS/cm。而李洋等[31]研究显示,0.1 g/L VB1浸种24 h会抑制郑单958和先玉335玉米品种的萌发。此外,分别采用VB1和VB2对甜玉米[32]和菜豆[33]进行引发处理,结果显示VB1的引发效果优于VB2,且0.01% VB1浸种24 h后,甜玉米发芽率从60.00%提升至67.15%;10 mg/L VB1浸种24 h后,菜豆种子发芽率从60.00%提高到61.33%。由此可见,VB1和VB2的引发效果因作物种类不同而存在差异。本试验结果表明,经VB1和VB2引发处理的小麦种子,其发芽力得到提高,膜透性较对照显著降低,酶活性较对照明显增强。这一结果可能与VB1和VB2参与种子内部生理生化反应的独特机制有关,它们或许能够更有效地激活种子内部的代谢途径,加速种子萌发进程。不过,VB1和VB2具体的引发机制尚未完全明确,仍需进一步研究。
通过对比9种不同引发剂、3个浓度及3个引发时间对低发芽力小麦种子萌发的影响,发现各引发剂均能显著提升小麦种子的发芽力,其中VB1和VB2的引发效果优于其余7种引发剂;引发16 h和24 h的效果显著优于8 h的处理。为进一步探究VB1和VB2的引发效果,对发芽率≥85%的4个处理开展浸出液电导率以及SOD、POD、CAT活性测定,最终确定0.08% VB1浸泡16 h是初始发芽率为32.67%的冀麦325种子的最佳引发方法。
