To determine a suitable nutrient solution for rapid propagation of virus-free sweet potato seedlings, four nutrient solutions at two concentrations each were tested using virus-free seedlings of Longshu No.9 in substrate culture to explore the effects of different treatments on the growth of virus-free seedlings. The results showed that both nutrient solution type and electrical conductivity significantly affected morphology and physiology of the virus-free seedling. The Longjiu treatments showed the best growth performance and root vitality, particularly Longjiu 2 resulted in superior stem vine length, stem node number, and aboveground fresh and dry weights compared with other treatments. These results indicated that Longjiu 2 was a suitable nutrient solution for rapid propagation of virus-free Longshu No.9 sweet potato seedlings.

为确定适宜甘薯脱毒苗快繁的基质培营养液,以龙薯9号脱毒苗为试验材料,选用4种营养液并各设2个浓度,探讨了不同处理对脱毒苗生长的影响。结果表明,营养液种类和溶液电导率均对脱毒苗的形态和生理产生影响,以“龙九”处理脱毒苗生长最好且维持根系活力最佳,尤其龙九2处理的脱毒苗茎蔓长、茎节数、地上部鲜重和干重均优于其他处理。表明龙九2可作为甘薯龙薯9号脱毒苗快繁适宜的培养液。

为确定适宜甘薯脱毒苗快繁的基质培营养液,以龙薯9号脱毒苗为试验材料,选用4种营养液并各设2个浓度,探讨了不同处理对脱毒苗生长的影响。结果表明,营养液种类和溶液电导率均对脱毒苗的形态和生理产生影响,以“龙九”处理脱毒苗生长最好且维持根系活力最佳,尤其龙九2处理的脱毒苗茎蔓长、茎节数、地上部鲜重和干重均优于其他处理。表明龙九2可作为甘薯龙薯9号脱毒苗快繁适宜的培养液。

To determine a suitable nutrient solution for rapid propagation of virus-free sweet potato seedlings, four nutrient solutions at two concentrations each were tested using virus-free seedlings of Longshu No.9 in substrate culture to explore the effects of different treatments on the growth of virus-free seedlings. The results showed that both nutrient solution type and electrical conductivity significantly affected morphology and physiology of the virus-free seedling. The Longjiu treatments showed the best growth performance and root vitality, particularly Longjiu 2 resulted in superior stem vine length, stem node number, and aboveground fresh and dry weights compared with other treatments. These results indicated that Longjiu 2 was a suitable nutrient solution for rapid propagation of virus-free Longshu No.9 sweet potato seedlings.

Lam.]属于旋花科甘薯属,是具有很高食用价值的世界第七大作物,同时在食品加工和工业生产中发挥着重要作用,近年来,市场对甘薯的需求量越来越大[1-2]。然而,甘薯在连年种植过程中,易出现品种退化、品质变劣和产量降低等现象,严重限制甘薯产业的发展[3]。利用甘薯脱毒技术能恢复种性,可有效提高甘薯产量及品质[4-5]。目前,生产中快繁甘薯脱毒苗的方式有多种,但市场上脱毒苗仍供不应求,其中基质培快繁方式生产效率较高,所使用的营养液是生产优质脱毒苗的关键。因此,寻求适宜甘薯脱毒苗快繁的基质培营养液成为研究和生产上关注的重点。王永江等[6]选用3种常规营养液并配制成不同的浓度,对商薯19脱毒苗进行处理,认为1/2Hoaland营养液是适合商薯19脱毒苗静置水培的最佳营养液;李润根等[7]以菜用甘薯品种台农71为试验材料,选用5种常规营养液配方进行水培处理,认为华南农大A较适宜台农71水培培养。周全卢等[8]利用改进的MS营养液配制成一种甘薯脱毒苗使用的水培营养液MA,取得了较好的经济效益[9-11]。为进一步优化营养液,避免肥料浪费和节省成本,本试验前期采用仅富含大量元素与微量元素的MS液体培养基作为培养液,依据脱毒苗对元素吸收的需求,研发出新型营养液龙九1。在此基础上,将大量元素浓度提升至2倍,得到龙九2,以下将龙九1与龙九2统称为龙九。
研究表明,适当的水肥浓度可促进植物生长、改善苗木品质和增强抗病能力[12-13],但浓度过高会产生负面影响[14-17]。本研究旨在通过开展龙九营养液与其他常规营养液的效果对比试验,筛选出脱毒苗快繁的最佳营养液。探寻适宜的营养液浓度对于高效生产具有关键意义,故而针对营养液种类和浓度研究,能够为优质脱毒苗的生产提供重要的理论支撑,进而推动甘薯产业化的发展。
试验于2018年6-8月在四川省成都市四川农业大学农学院(30°42′20″ N,103°51′42″ E)温室大棚内进行,棚内控制温度20~30 ℃,空气相对湿度60%~70%,光强500~1000 µmol/(m2·s)。以龙薯9号甘薯脱毒苗为试验材料,所用栽培基质为珍珠岩,营养液配制所需药品均购自成都浩搏优科技有限公司。
基质培营养液种类选用4种:Hoaland营养液、循环水生菜营养液(以下简称循环)、仅含有大量与微量元素的MS液体培养基和龙九新型营养液,4种营养液分别设2个浓度:1/2Hoaland、Hoaland、1/2循环、循环、1/2MS、MS、龙九1和龙九2,共8个处理(表12),每个处理设3次重复,基质培框作为试验装置,框内放置4个花盆,盆内分别装满各处理营养液浸泡充分的珍珠岩,框内倒入各处理营养液,并将黑色地膜覆盖整个塑料框口,模拟根系环境,经过生根处理后的脱毒苗茎段定植于以上各处理的基质培装置中。每个框作为1次重复,每个重复16株脱毒苗。试验期间每天观察水分蒸发情况并用纯水及时补充至原始体积,将基质培营养液的pH控制在6.0~6.5。
选取每个装置内4个点,使用SG23电导仪测定电导率,每7 d测定1次,共测定8次。
每个重复内随机选取4株具有代表性的植株,利用直尺采用常规方法进行测定,每15 d测定1次,4次重复。
每个重复内选取向阳健康生长的第4、5叶位的5个成熟叶片,分别在每个叶片的叶基、叶中和叶尖处使用SPAD-502叶绿素仪测定SPAD值,每15 d测定1次,3次重复。
基质培第50天时采用万分之一电子天平(奥豪斯/CP114)测定鲜重与干重。每个重复内选择4株代表性植株,将脱毒苗地上部与地下部分分开,称其鲜重,然后放于烘箱105 ℃杀青30 min,最后75 ℃烘干至恒重,称其干重。
基质培至第25天和50天时测定,每个重复内选择4株具有代表性的植株,使用TTC法[18]测定根系活力。
采用Excel 2016进行汇总及整理试验数据,使用DPS 7.05软件进行统计分析,采用Duncan法进行多重比较,利用Origin 2021作图。
营养液EC值对于无土栽培植物发育及产量有着重要的影响。如表3所示,MS和龙九2营养液原始EC值分别为5.513和4.963 mS/cm。8个处理在6月20日-27日基质培过程中脱毒苗根系小,吸收营养少,相同时间内营养液的水分蒸发速度高于根系吸收同等水量内营养元素的速度,而导致各处理EC值变化小,甚至个别处理出现略有升高的现象;在7月3日-7月18日,随着脱毒苗根系的不断生长,各处理对营养液中元素不断吸收,EC值呈明显下降趋势,表明各处理营养液中总离子浓度配制合理,基质培营养液酸碱度、溶氧量和水温等均能有效维持植物细胞内外水分运输平衡及正常生理活动,促进生长快繁,在此期间各处理EC值均表现为MS>龙九2>1/2MS>循环>龙九1>Hoaland>1/2循环>1/2Hoaland;在7月25日-8月9日基质培过程中MS和龙九2的EC值显著高于其他处理,且除MS和龙九2外,其他处理均已下降低至0.5 mS/cm以下,8月9日时MS和龙九2的EC值分别为1.447和1.120 mS/cm,对根系活力的维持仍然具有一定的效用。1/2循环和1/2Hoagland EC值分别降至0.013和0.076 mS/cm,已明显无法满足脱毒苗正常生长的需求。
表4所示,6月27日龙九1茎蔓长显著高于其他处理,7月11日龙九2处理最高,但与其他处理间差异小,7月25日龙九2处理显著高于其他处理,至8月8日,龙九2处理显著高于除1/2MS以外的处理,并且比1/2Hoagland处理提高101.37%,比MS提高43.37%,此时各处理茎蔓长依次为龙九2>1/2MS>龙九1>MS>循环>1/2循环>Hoagland>1/2Hoagland;如表5所示,6月27日龙九1处理的茎节数最多,但与除1/2MS以外的其他处理无显著性差异,在7月11日,龙九2处理最高,龙九1次之,且二者之间无显著性差异,7月25日龙九2处理显著高于其他处理,8月8日龙九2处理显著高于除1/2MS以外的其他处理,并且比1/2Hoagland提高85.48%,比MS提高27.53%,此时各处理茎节数依次为龙九2>1/2MS>龙九1>MS>1/2循环>循环>Hoagland>1/2Hoagland。
表6所示,龙九2与MS处理下脱毒苗地上鲜重显著高于其他处理,龙九2比1/2Hoagland处理提高158.82%,比MS提高4.21%;龙九2处理地上干重最高,显著高于1/2Hoagland、Hoagland、1/2循环、循环与MS处理,1/2Hoagland地上干重最低,龙九2比1/2Hoagland处理提高56.24%,比MS处理提高9.60%;在地下鲜重和干重指标中,1/2Hoagland处理显著高于其他处理,分别比龙九2提高31.29%和100.32%。并且1/2Hoagland、Hoagland、1/2循环和循环处理均有薯块产生(图1)。各处理地上部分干、鲜重在同种营养液中基本表现为高剂量浓度大于低剂量浓度,且地上干、鲜重与地下干、鲜重基本呈负相关。
测定脱毒苗叶片SPAD值可作为调节营养液中N元素的参考依据,由表7所示,不同处理对脱毒苗SPAD值的影响开始差异较小,越到培养后期差异变大,表现为浓度低的SPAD值变大。在基质培15 d时,龙九2处理的脱毒苗SPAD值为38.443,显著高于除龙九1以外的其他处理;基质培30 d时,1/2Hoagland和1/2循环处理的脱毒苗SPAD值分别为41.577和41.967,显著高于其他处理,分别比MS处理高9.90%和10.93%,其次为龙九2和龙九1;基质培45 d时,1/2Hoagland处理的SPAD值为47.693,显著高于其他处理,比最小值循环高9.31%,其次为Hoagland、1/2循环和龙九1。
表8所示,在不同时期各基质培营养液处理对根系活力有不同的影响。基质培25 d时,1/2Hoagland和龙九2根系活力最高,显著高于Hoagland和MS,但各处理间的差异不明显;基质培50 d时各处理根系活力排序为1/2MS>龙九1>龙九2>循环>MS>Hoagland>1/2Hoagland>1/2循环,并且越到后期龙九对延缓根系衰老的效果越好,而总离子浓度较低的处理尤其1/2Hoagland和1/2循环则维持根系活力效果相对较差,分别比25 d下降89.50%和86.45%,这也印证了2.1中的分析结果。
基质培快繁营养液作为植物根系吸收营养物质的主要来源,不同营养液中各营养元素含量的配比对于植物生长至关重要[19-20]。李平芳等[21]研究了不同N、P、K配比对甘薯产量和商品率的影响,结果得出N(0.8~1):P(0.8~1):K(2)时最优。柳洪鹃等[22]在研究中发现,甘薯北京553完成生长周期同样需要合理的元素含量配比,其中所需要的大量元素中N、K和Ca最多,所需微量元素中Fe最多。本研究表明,在脱毒苗茎蔓长、茎节数和地上鲜重与干重等指标均是龙九2处理优于其他处理,其原因可能为龙九2养分配比是根据脱毒苗生长过程中对各营养元素的吸收情况合理配制的,各营养元素间配比均衡,富含植物生长所必需的大量元素和微量元素,且新型营养液龙九2中大量元素N、K和Ca离子浓度最多,微量元素中的Fe含量相对较高,新型营养液龙九2的养分配比与柳洪鹃等[22]研究结果类似。
作物基质培营养液除了各元素配比对植物生长至关重要外,营养液EC值对作物生长发育同样具有重大影响,其可反映溶液的总离子浓度剂量大小。无土栽培管理中经常会根据EC值的大小确定营养液剂量,相反也可以根据剂量来计算得到EC值[23]。许多学者[24-25]曾对基质培营养液EC值与作物生长发育做过相关研究。
本试验中发现溶液原始EC值大约在0~5 mS/cm范围内脱毒苗生长量与溶液EC值呈正相关,其中龙九2处理原始EC值为4.963 mS/cm,在脱毒苗基质培适宜的离子浓度范围内,符合脱毒苗正常生长需求,MS处理原始EC值为5.513 mS/cm,EC值过高,李嘉炜等[26]研究了可溶性盐浓度对南瓜幼苗生长的影响,表明EC值过高,会形成反渗透压,造成根系失水,进而降低南瓜对养分的吸收和分配,使得生长速度变慢。本研究中MS处理的脱毒苗生长同样受到一定程度的影响。因此,在作物无土栽培过程中,营养液EC值应根据作物需求规律和目的,适当调节营养液EC值,促进作物生长的同时避免养料浪费。周庐萍等[27]研究还发现,高营养液EC值在一定条件下能够促进菊花对水分和营养元素的吸收,利于菊花健康生长,并且前人[26,28]研究还发现即使轻微过量的养分供应也可提高作物幼苗光能利用效率,促进作物生长。本试验中发现虽然MS处理EC值略高于适宜EC值范围,但对脱毒苗的生长依旧优于1/2循环、循环、Hoagland及1/2Hoagland处理,当然,这也可能是因为MS处理相对于其他处理含有更多的N元素,更利于脱毒苗地上部营养生长的原因所致。
另外,研究[29]表明N元素过多会与K、P、Ca和Mg元素产生拮抗作用,而这些元素对甘薯膨大具有重要支持作用,所以这可能就是虽然在龙九2和MS处理中这些元素足够多,但对脱毒苗根系膨大无明显作用的原因。且通过分析发现,各处理地上与地下部分的生长呈明显的负相关性,这是由于植物地上与地下部分的生长存在相互协调和制约的关系,这种关系通常受植物对营养、水分等需求的影响,李炎等[30]研究得出,当冬小麦受到水分、营养及水分和营养双重胁迫时,根冠比会增加。同理,在本研究中发现EC值高,营养元素含量较多的处理中(龙九1、龙九2、1/2MS和MS处理)N元素含量较多,大量光合产物用于蛋白质的合成,地上营养生长过旺,碳代谢失调,从而削弱了同化物向地下的转运,甘薯贮藏根膨大受到影响,根冠比减小,这同样也是对根系膨大无明显作用的原因,生产中可通过水肥管理和化学调控达到丰产目的。
本研究中还发现EC值低的处理不利于地上部分的生长,但促进了地下部分生长量的增加,同时还发现EC值较低的1/2Hoagland和1/2循环处理下的脱毒苗基质培后期SPAD值较高,根系活力较低。原因可能是由于无土栽培植物相对于土培植物生长周期较短[31-32],尤其EC值较低的1/2Hoagland和1/2循环处理的脱毒苗会最先出现养分缺乏而导致生长周期缩短[33-34],使结薯期提前,SPAD值上升,说明叶片对N素吸收率提高,叶片中的有效N对叶绿素的合成有重要作用。叶绿素是光合色素的主要组成成分,在光合作用中能够吸收光能,将CO2和水转化为有机物质,产生更多的干物质[35],进而促进结薯性能的提高,这与前人[36]的研究结果类似,当然,结薯性能提高也与EC值较低的营养液中溶氧量较高有关,这促使根系膨大,进而导致根系活力下降[37-39],这就解释了EC值较低的营养液对脱毒苗地下部分生长量的增多效果较好及基质培后期SPAD值较高及根系活力较低的原因,所以在配制营养液时应严格控制EC值。
根系活力影响植物对矿质元素的吸收能力,反映脱毒苗质量的好坏,好的培养液能够更持久地维持根系活力,本研究中发现各处理对根系活力的影响越到后期越显著,是因为在前期各处理溶液中的各种养分比较全面,均能够满足脱毒苗需求,根系活力差异较小,而到了50 d时,某些营养液处理中的一些养分不足,已不能满足脱毒苗正常生长需求。
大量元素中N、P和K对根系活力影响更大[40-41],除1/2MS和MS处理以外,龙九中N、P和K含量与其他处理相比相对较多,N、P和K含量相对较少的处理尤其1/2Hoagland和1/2循环基质培后期更容易缺乏营养,出现营养胁迫而导致根系活力下降较大。另外,龙九中微量元素除B和Cu以外,Mn、Mo、Fe、Zn和Cl的含量也相对较多(除1/2MS和MS以外),这些微量元素对根系活力同样具有重要影响,例如Mn可参与植物光合和酶促反应,促进根细胞的呼吸及养分吸收;Mo是固氮酶的重要部分,参与N的固定和转化;Fe参与植物光合和呼吸等代谢过程,缺Fe会导致植株生长发育异常,近而影响根系发育和活力;Zn对生长素的合成有着重要作用,促进根尖部分发育;Cl可维持根系细胞的渗透压和离子平衡,缺Cl会导致根系功能异常[42-43]。这也就证明了龙九相对于其他处理根系活力的维持效果较好并再一次解释说明了EC值较低的营养液后期根系活力较低的原因。
然而,本研究中1/2MS、MS及龙九溶液配方中含有Cl元素,这是因为配方中的Ca源于化学药品CaCl2,导致溶液中含有Cl元素,相关研究[41]已证明薯类作物为忌氯作物,Cl元素会影响薯类作物块根或者块茎中的淀粉等糖含量,影响口感,但本研究目的为快繁,发现Cl元素未对脱毒苗生长产生不利影响,正如戚冰洁等[44]通过研究发现,短期内低浓度外源氯对甘薯幼苗生长及养分吸收具有积极作用,并且相同纯度的含Ca化学药品中CaCl2价格较低,可有效减少成本。
新型营养液龙九2对甘薯脱毒苗快繁效率高,薯苗质量好,适宜甘薯脱毒苗基质培快繁生产,且相较于MS配方可降低肥料成本,可为甘薯脱毒苗专用营养液的研究提供参考;EC值较低的营养液(1/2Hoagland和1/2循环)有利于脱毒苗地下干物质积累,促进根系膨大。
