A field experiment was conducted to investigate the effects of planting densities and nitrogen application levels on the growth, biomass allocation and yields of Salvia prionitis Hance. Three planting density levels, high density (666 666 plant/ha), medium density (200 000 plant/ha), and low density (95 238 plant/ha); and three fertilization levels [no nitrogen application, low nitrogen (22.5 g/m2) and high nitrogen (45 g/m2)] were set. The results showed that both the density and nitrogen level significantly affected plant size, biomass allocation and yield of S.prionitis. However, the interaction effect only extremely significantly affected the parameters related to biomass allocation. In general, nitrogen application increased the plant height, number of flower stalks, and number of primary branches flower stalks of S.prionitis under all planting densities, and leaf length, leaf width, and plant height of the two low density treatment were greater than those of the high density treatment under all nitrogen levels. Nitrogen level did not significantly affect the root biomass ratio and root- shoot ratio of S.prionitis at medium density treatment, while the root biomass ratio and root-shoot ratio at low and high densities were significantly reduced with the increasing of nitrogen application. Under each densities, the root biomass, supporting structure biomass, total biomass, whole plant yield and root yield of S.prionitis were the greatest in the low nitrogen treatment, then followed by the high nitrogen treatment. under every nitrogen application levels, the root biomass, supporting structure biomass and total biomass of S.prionitis increased significantly with the decreasing of density, However, the whole plant yield and root yield increased significantly with the decreasing of density. In all treatments, whole plant yield and root yield of S.prionitis were the greatest at high density (666 666 plant/ha) with low nitrogen level (22.5 g/m2 pure nitrogen, urea).

采用大田试验,研究不同密度和施氮水平对红根草生长、生物量分配和产量的影响。试验设置3个密度水平:高密度(666 666株/hm2)、中密度(200 000株/hm2)和低密度(95 238株/hm2);各密度设3个施肥水平:不施氮、低氮(22.5 g/m2)和高氮(45.0 g/m2)。结果表明,密度和施氮水平均显著影响红根草的植株大小、生物量分配和产量相关参数,但其交互作用仅对生物量分配参数影响极显著。总体而言,施氮会增大各密度下红根草的株高、花苔数和花苔一级分枝数,各氮水平下2个低密度植株的叶长、叶宽和株高大于高密度。施氮量对中密度红根草的根生物量比和根冠比无显著影响,低、高密度的根生物量比和根冠比均随施氮量增高而显著降低。低氮处理下各密度红根草的根生物量、支持结构生物量、总生物量、整株产量与根产量最大,高氮处理次之;各施氮水平下红根草根生物量、支持结构生物量和总生物量均随密度降低而显著增大,而整株产量与根产量却随密度降低而显著增大。所有处理中,密度666 666株/hm2、施纯氮22.5 g/m2(尿素)处理红根草的根产量和整株产量最大。

Three planting density levels, high density (666 666 plant/ha), medium density (200 000 plant/ha), and low density (95 238 plant/ha); and three fertilization levels [no nitrogen application, low nitrogen (22.5 g/m2) and high nitrogen (45 g/m2)] were set. The results showed that both the density and nitrogen level significantly affected plant size, biomass allocation and yield of S.prionitis. However, the interaction effect only extremely significantly affected the parameters related to biomass allocation. In general, nitrogen application increased the plant height, number of flower stalks, and number of primary branches flower stalks of S.prionitis under all planting densities, and leaf length, leaf width, and plant height of the two low density treatment were greater than those of the high density treatment under all nitrogen levels. Nitrogen level did not significantly affect the root biomass ratio and root- shoot ratio of S.prionitis at medium density treatment, while the root biomass ratio and root-shoot ratio at low and high densities were significantly reduced with the increasing of nitrogen application. Under each densities, the root biomass, supporting structure biomass, total biomass, whole plant yield and root yield of S.prionitis were the greatest in the low nitrogen treatment, then followed by the high nitrogen treatment. under every nitrogen application levels, the root biomass, supporting structure biomass and total biomass of S.prionitis increased significantly with the decreasing of density, However, the whole plant yield and root yield increased significantly with the decreasing of density. In all treatments, whole plant yield and root yield of S.prionitis were the greatest at high density (666 666 plant/ha) with low nitrogen level (22.5 g/m2 pure nitrogen, urea).

红根草(Salvia prionitis Hance)为唇形科鼠尾草属多年生草本植物,高20~43cm,属较矮小的草本植物,野外多分布在阳处草丛,植株寿命一般为3年左右,春季种植,第2年5月下旬进入开花盛期,6月上旬进入开花末期,在开花后期基生叶和花茎枯萎倒伏后会降低药材质量和产量,因此在夏秋季枝叶茂盛时采收较为合适[1]。红根草主要种植于广西、广东、江西、浙江及安徽等地,为治疗急性咽喉炎和扁桃体炎中成药的主要原料。在《中国植物志》[2]中记载广西以全株入药,《广西壮族自治区壮药质量标准》[3]第二卷亦记载红根草含有二萜醌类化合物,目前已分离鉴定出20多个二萜醌类化合物和新二萜类化合物,这些化合物具有抗菌、抗肿瘤及抗凝血等作用。近几年,国内学者[4-5]还从红根草中分离出齐墩果酸等多个三萜类化合物,但尚未确定其具体的指标性成分。目前有关红根草的研究主要集中在化学成分[6]和药理作用[7-8]等方面,栽培方面的研究较少。近年来,由于市场需求增大,再加上产区民众的过度采挖,以及生态环境的不断恶化,导致野生红根草根资源濒临枯竭,为拓展其药用来源,开展人工驯化和高效栽培技术研究势在必行。
在一定的生态条件下,种植密度和施肥量对农作物的生长和产量影响较大,是作物栽培的重要人工管理措施。种植密度会引起植物个体之间的互作效应,在一定范围内,随着密度的增大,单位面积的群体产量呈上升趋势,当密度达到一定程度时,产量可达最高值,但继续增大密度,作物会因光照和养分等资源不足而造成产量降低[9];种植密度的增加还会引起种间竞争,导致个体的生长与生物量发生变化,进而影响植物对资源的利用与分配[10-11]。氮素营养是影响植物生长最重要的元素之一,适量施用氮肥可以显著提高叶片光合能力,加速CO2同化速度,随之提高净光合速率,促进作物生长和产量增加,但过量施用氮肥时,不仅会导致叶片的光合速率下降,影响作物产量[12],还会导致环境污染和土壤退化等问题[13]。在生产过程中,密度和氮肥的增产效应不仅取决于各自单因素作用,还在于两者间的互作效应[14]。相关研究[15-17]表明,在一定范围内,施肥量与种植密度之间存在互补关系,在地力水平较差或养分不足时,可适当提高种植密度加以补偿;反之,当种植密度较大,则可适当增加施肥量以促进产量的提高。由上述可知,适宜的密度与氮素营养组合不仅能够促进作物的生长发育并提高产量[18],也能降低生产成本、增加效益,还能降低化肥施用量,减少环境污染。但目前国内外尚无种植密度、施氮水平及两者互作效应对红根草生长和产量影响方面的研究。本试验通过施氮水平和密度两因素三水平完全随机设计方案,研究不同种植密度和施氮量对红根草生长、生物量分配及产量的影响,为红根草的高产高效栽培提供理论依据。
试验于广西桂林雁山区广西植物研究所(25°04′ N,110°18′ E)实施,该地位于广西壮族自治区东北部,属亚热带季风气候,冬无严寒,夏无酷暑,降水量充沛,气候温和。试验地土壤为红壤,pH 5.8,0~15 cm土壤层的有机质、全氮、全磷和全钾含量分别为15.70 g/kg、2.27 g/kg、1.10 g/kg和11.10 g/kg。
试验采用施肥和密度两因素完全随机设计。于2022年2月15日将红根草种子播种于苗床,幼苗4片真叶时(4月15日),选择大小一致的裸根幼苗移栽。施肥因素设置3个水平:不施氮(N0)、低氮(N1,22.5 g/m2)和高氮(N2,45.0 g/m2);密度因子设置3个水平:高密度10 cm×15 cm(666 666株/hm2)、中密度20 cm×25 cm(200 000株/hm2)和低密度30 cm×35 cm(95 238株/hm2)。9个处理,3次重复,共27个小区,随机排列,每小区面积9 m2。本试验选用含氮量为46%的尿素,以追肥形式施入,结合除草苗期和生长盛期各施50%。各处理底肥各施有效磷和有效钾30 g/m2(钙镁磷肥和硫酸钾)。
于现蕾期(2023年5月28日)收割。采收前,在每个小区中随机抽取10株,测量其株高、平均冠幅、叶长和叶宽,再计算花苔数和花苔一级分枝数(各处理组合30株),之后每个小区从测量的10株中随机选8株整株连根挖出(各处理组合24株),洗净后测定叶、支持结构(主茎与分枝)和根干重(105 ℃杀青30 min,80 ℃烘干至恒重)。求出根生物量比、叶生物量比、支持结构生物量比及根冠比。之后,各小区全部挖根收获,烘干测定单位面积根产量与整株产量,将取样测定单株生物量的8株计入小区总量。
采用双因素方差分析法比较密度与施氮水平对红根草各变量的影响,因素间的交互作用由软件自动取舍。若交互作用显著,为找到最佳的处理组合,把不同的处理组合视作平行的比较组进行单因素方差分析;若交互作用不显著时,为分析各组数据之间的显著性,对数据进行多次单因素方差分析比较红根草不同密度和氮水平间的差异显著性,用邓肯多重比较法进行多重比较,检验的显著性界限水平α=0.05。采用Excel和SPSS22.0进行数据处理,用SigmaPlot 14.0绘图。
除密度对红根草花苔数影响不显著及施氮水平对红根草叶长和叶生物量比影响不显著外,密度和施氮水平均显著或极显著影响了红根草多数的植株大小、生物量分配和产量相关参数,但其交互作用仅对生物量分配参数影响极显著,对其他参数影响不显著(表1)。
图1可知,高、低密度下,2个施氮处理红根草的株高均显著高于N0处理;中密度下,各施氮处理的株高无显著差异。N0处理下,中、低密度红根草的株高无显著差异,但均显著高于高密度;N1和N2处理下,高、中密度红根草的株高无显著差异,均显著低于低密度。各氮处理下,密度对红根草的花苔数影响均不显著;各密度下,2个施氮处理的花苔数均显著高于N0处理。各密度下,2个施氮处理的花苔分枝数均显著高于N0处理;各氮处理下,中、低密度红根草的花苔分枝数无显著差异,但均高于高密度。各密度2个施氮处理的平均冠幅均显著高于N0处理;各氮处理下,平均冠幅随密度的降低而显著增大。除低密度下高氮处理的叶宽、叶长显著高于N1和N0处理外,高、中密度下各氮处理的叶宽和叶长均无显著差异;除N0处理下中密度的叶长和叶宽均显著高于低、高密度外,各氮处理,中、低密度的叶宽和叶长均无显著差异,但均显著高于高密度。
各密度下,N1处理红根草的根生物量、支持结构生物量、叶生物量和总生物量均显著大于N2与N0处理,而N2处理又显著大于N0处理;各施氮处理下,红根草根生物量、支持结构生物量和总生物量均随密度的降低而显著增大(图2)。N0处理下,中、低密度红根草的叶生物量无显著差异,但均显著大于高密度;N1处理下,中、高密度红根草的叶生物量无显著差异,但均显著小于低密度;N2处理下,高密度红根草的叶生物量显著大于中密度,但显著小于低密度。
双因素方差分析表明密度与施氮交互作用极显著影响红根草的生物量分配参数(表1),不同处理组合的单因素方差分析结果如图3所示,其中,红根草的根生物量比和根冠比的变化趋势一致,低、高密度的根生物量比和根冠比均随施氮量的增高而显著降低,中密度下不同氮处理间无显著差异;N0处理下,中、低密度的根生物量比和根冠比均无显著差异,但均显著大于高密度;N1和N2处理下,中密度的根生物量比和根冠比均最高,高密度最低;高、低密度下,N0处理红根草的支持结构生物量比显著高于2个施氮处理,中密度下反之;N0处理下,低密度的支持结构生物量比最高;N2处理,中密度的支持结构生物量比最高,不同处理组合中,中密度N2处理的支持结构生物量比最高,高密度高氮处理的支持结构生物量比最低。高、低密度下,红根草的叶生物量比均随施氮量的增大而显著增大,中密度下反之;各氮处理下,高密度的叶生物量比均显著大于低密度;不同处理组合中,高密度高氮处理的叶生物量比最高,中密度高氮处理的叶生物量比最低。
图4所示,各密度下,N1处理红根草的整株产量与根产量均显著大于N2和N0处理,而N2处理又显著大于N0处理;各施氮处理下,高密度红根草的整株产量与根产量均显著大于中密度和低密度,而中密度又显著大于低密度。
种植密度会直接影响植株个体空间、光和养分等资源的可用性,从而影响植株生长。当植株密度较高时,由于养分、水分、光和物理空间的不足,植物会调整自身形态结构来适应环境[19-20]。郑伟等[21]研究发现,种群密度增大能抑制猪毛菜的生长;朱仕明等[22]也发现,密度增加导致乐昌含笑的苗高及各器官干重降低。本研究中,红根草的平均冠幅、株高、花苔数和花苔一级分枝数均随密度的增大呈显著降低趋势,根生物量、支持结构生物量、叶生物量和总生物量均随密度的增大而显著降低,表明种植密度的变化显著影响了红根草的形态结构和各部位生物量积累。低密度下养分、水分、光和物理空间资源竞争较小,红根草的个体大小和生物量较大;高密度下养分、水分、光和物理空间资源竞争较大,植株个体所获得的资源有限,红根草不得不将更多的资源用于维持生存,将较少的资源用于生长和繁殖,这与郑伟等[21]和朱仕明等[22]的研究结果相似。范高华等[23]研究亦表明,大果虫实的株高、分枝数及分枝长随着密度增大而减小,其原因是密度变化迫使大果虫实调整自身形态结构来适应环境,通过影响茎顶端分生组织的活动来决定茎秆的形成与发育,进而调控大果虫实侧生器官的数目和形态。尽管各施氮水平下,低密度红根草的单株生物量最大,但由于单位种植面积的植株数减少,低密度处理的单位面积整株产量和根产量显著低于中、高密度。
氮素是植物体内多种有机化合物的重要组成成分,其在光合作用、细胞生长和基因变异等过程中发挥着重要的作用,因此,氮对植物的生长和产量具有决定性作用[24]。虽然外源氮的施入可以促进植物生长发育,但植物对氮素需求有一定阈值,当氮的施入超过阈值时,植物生长将会受到损害,且还会引起氮流失和土壤酸化板结[25]。李韦柳等[26]研究发现,狼尾草的株高、茎粗和单位面积有效茎数随着施氮量的增加而逐步提高,但叶片叶绿素含量和鲜、干草产量的提高程度在施氮量超过225.0 kg N/hm2后有饱和趋势。翟凤强等[27]研究发现,在较低施氮量下,藜麦品种幼苗的植株生物量积累明显比不施氮处理增加,但在较高施氮下,藜麦叶片光合速率出现降低趋势,生物量积累减少。红根草亦是如此,本研究中,施氮显著增加了各密度下红根草的花苔数、花苔一级分枝数、冠幅及高、低密度株高,但根生物量、叶生物量、支持结构生物量、总生物量、根产量和整株产量均随施氮量增加表现出先增后减的趋势,这与前人[25-27]的研究结果基本一致,说明低氮(施氮22.5 g/m2)为红根草的施氮量阈值,达该施氮水平后,如果继续增加施氮量,植株生长将会受损、产量降低。
生物量的分配是植物获得资源、竞争和繁殖的关键因子,同时也是衡量植物竞争力的一个重要标志[19,28]。最优分配假说认为,植物在光照、水分和养分等资源受限的情况下,会把更多的生物量分配给有效获取有限资源的器官中,以维持生长所需营养,保持生长速率[29]。一般来说,植物地上生物量分配越高植物对光照的竞争能力越强[30];而地下生物量分配越高植物对土壤养分的竞争力越强[31]。本研究表明,各施氮水平,红根草中、低密度的根生物量比和根冠比均显著大于高密度,高密度叶生物量比显著大于中、低密度,可能是植株密度较高时,植株个体间的竞争关系逐渐增加,红根草植株为增强个体截留光能的能力,将更多的生物量分配到茎和叶,增强光合作用,积累有机物,这一分配行为的结果使根冠比减小[32]。这与前人对稗草[33]和菊芋[34]等物种的研究结果一致。
植物生长深受养分因子的影响,因此研究植物对养分资源的可塑性,可了解物种对养分环境的适应机制和生态分布规律[35]。营养资源受限时,植株会向根部提供更多的养分,增大根的生物量比,促进根的生长,从而提高地下竞争力,当营养资源充足时则会增加对地上部分的投入,促进地上部分生长,提高地上竞争力[36]。黎蕾等[37]研究发现,当光强条件一致,催吐萝芙木根生物量比和根冠比均随着施氮量的增加而减小。尹丽等[38]研究发现,施氮对麻疯树地下部分的促进作用显著小于地上部分,施氮后根冠比显著降低。在本研究中,施氮水平对红根草的单株生物量及其分配影响显著,低、高密度红根草的根生物量比、根冠比均随施氮量的增高而显著降低,2个施氮处理的叶生物量比均显著大于不施氮处理,表明不施氮时,由于养分竞争激烈,红根草会增加吸收器官的生物量分配,增大根生物量分数和根冠比,提高地下竞争力,增强根对土壤养分的吸收;随着施氮量的增加,养分资源充足,植物生长不再受养分因子的限制,根生物量比和根冠比减小,叶生物量增大,同化产物更多投入到碳同化器官,提高植株的光合能力,以实现光合效益的最大化,这与前人[35-38]的研究结果一致。
调整种植密度和施氮水平是生产中最为常见的2个栽培措施,二者均影响作物的生长、产量和生物量分配,而且两因素间通常存在交互作用。王彦辉等[39]发现,当种植密度60万株/hm2、施氮水平120 kg/hm2时谷子产量最高,且和其他组合之间有显著差异,密度水平120万株/hm2、施氮水平120 kg/hm2的产量次之,因此,他们认为氮密互作对谷子产量的影响显著,适度增加氮肥可以减缓因种植密度增加造成养分不足而导致产量下降的趋势。段媛媛等[40]研究发现,氮密互作没有显著影响垂穗披碱草的根系体积和单株分蘖数,但显著影响了株高、生物量及根冠比;垂穗披碱草产量在植株密度为102株/m2和施氮量为200 mg/kg的组合产量最大,且生物量分配达到最优。本研究中,密度与施氮交互作用显著影响了红根草的根生物量比、支持结构生物量比、叶生物量比和根冠比。所有处理中,高密度低氮处理的单位面积根产量和整株产量最大,高密度高氮处理次之,这说明密度与施氮互作对红根草的生长产生了影响,且两者互作时存在最佳组合,其交互作用在统计学上的影响不显著可能与密度和施氮单因素对红根草植株大小、生物量和产量影响的F值远大于其交互作用F值有关。因此,种植红根草要采用适宜的种植密度和施氮处理,只有二者处于最佳的耦合状态时,红根草才能发挥最大生产潜能,实现高产目标,还能节约氮素,减少生产投资及环境污染,这与段媛媛等[40]的研究结果相似。本试验从生长指标和产量方面初步探讨了施氮量和种植密度对红根草的影响,下一步将结合对其药用成分含量的影响开展研究。
密度和施氮水平显著影响了红根草的植株大小、生物量分配以及产量。栽培密度和施氮水平互作虽然对植株大小、生物量和产量没有显著影响,但两者互作对生物量分配影响极显著。所有处理中,高密度低施氮水平的根产量和整株产量最大,其次是高密度高施氮水平;从整体收益的角度分析,高密度和低施氮水平的组合不仅能获得高产,而且肥料投入较少,既节约成本还降低了对环境的破坏。因此类似本试验土壤条件下,推荐红根草人工栽培采用666 666株/hm2、施纯氮22.5 g/m2(尿素)。
