Objective:

An HPLC-MS/MS method was established for the simultaneous determination of 15 components [betaine, agaritol, bergamolide, 2-(2-phenylidene) chromone, dehydrodiisoeugenol, D-(-)-quinic acid, gallic acid, protocatechuic acid, hydroxysafflor yellow A, rutin, isoquercetin, ellagic acid, luteolin and apigenin] in Qingxin Chenxiang Bawei powder (pills). The quantitative method, combined with chemometrics and entropy weight-grey correlation degree method to analyze the comprehensive quality, provides a reference for the quality control and evaluation of this preparation.

Methods:

The analysis was conducted using a Shim-pack GIST-HP C18 chromatographic column, with the mobile phase consisting of methanol (A) and a 0,1% formic acid aqueous solution (B). A gradient elution was employed at a column temperature of 35 ℃, with an injection column of 3 μL. Weisenxin platform was used for cluster analysis (CA), SIMCA 14.1 software was used for principal component analysis (PCA) and orthogonal partial least squares discriminant analysis (OPLS-DA), and the compounds with variable important in projection (VIP) value greater than 1.0 were used as the criterion to screen quality differences. The entropy weight-correlation degree method was used to analyze the comprehensive quality.

Results:

The linear relationship of the 15 components was good in their respective ranges, and the linear correlation r was greater than or equal 0.999 0. Precision RSD were all lower than 3.00%. The stability and repeatability were good, RSD lower than 5.00%; The average recoveries were 93.62%~105.10%, and RSD was 1.18%~4.37%. CA and PCA analysis classified 14 batches of Qingxin Chenxiang Bawei powder (pills) into 2 categories: manufacturers D (D1~D4) were grouped into one group, manufacturers A and M (A1~A4, M1~M6) were grouped into one group. Six difference markers were screened in OPLS-DA mode, which were D-(-)-quinic acid, agaritol, 2-(2-phenylethyl) chromone, dehydrodiisoeugenol, bergamolactone and quercetin, respectively. A2 has the best comprehensive quality.

Conclusion:

The established HPLC-MS/MS method for the simultaneous determination of 15 components in Qingxin Chenxiang Bawei powder (pill) can be used for the comprehensive quality study of Qingxin Chenxiang Bawei powder (pill) combined with chemometrics and entropy weight correlation degree method.

目的:

建立HPLC-MS/MS同时测定清心沉香八味散(丸)中15个成分[甜菜碱、沉香四醇、佛手柑内酯、2-(2-苯乙基)色酮、去氢二异丁香酚、D-(-)-奎尼酸、没食子酸、原儿茶酸、羟基红花黄色素A、芦丁、异槲皮苷、鞣花酸、木犀草素、芹菜素]的含量,并结合化学计量学与熵权-灰色关联度法分析综合质量,为该制剂的质量控制和评价提供科学依据。

方法:

采用Shim-pack GIST-HP C18色谱柱;流动相为甲醇(A)-0.1%甲酸的水溶液(B),梯度洗脱;柱温35 ℃;进样量3 μL。微生信平台进行聚类分析(cluster analysis,CA),SIMCA 14.1软件进行主成分分析(principal component analysis,PCA)和正交偏最小二乘判别分析(orthogonal partial least squares discriminant analysis,OPLS-DA),以VIP值>1.0为标准筛选质量差异物;采用熵权-关联度法分析综合质量。

结果:

15个成分在各自范围内线性关系良好,r均≥0.999 0;精密度RSD均<3.00%;稳定性和重复性良好,RSD均<5.00%;平均加样回收率为93.62%~105.10%,RSD为1.18%~4.37%。CA和PCA分析将14批清心沉香八味散(丸)聚为两大类:D厂家(D1~D4)聚为一类,A厂家和M厂家(A1~A4,M1~M6)聚为一类;OPLS-DA模式下筛选出6个差异标志物,分别为D-(-)-奎尼酸、沉香四醇、2-(2-苯乙基)色酮、去氢二异丁香酚、佛手柑内酯和槲皮素。结果表明,A2厂家综合质量最好。

结论:

建立的HPLC-MS/MS法可同时测定清心沉香八味散(丸)中15个成分,结合化学计量学与熵权-灰色关联度法可用于清心沉香八味散(丸)综合质量研究。

目的:

建立HPLC-MS/MS同时测定清心沉香八味散(丸)中15个成分[甜菜碱、沉香四醇、佛手柑内酯、2-(2-苯乙基)色酮、去氢二异丁香酚、D-(-)-奎尼酸、没食子酸、原儿茶酸、羟基红花黄色素A、芦丁、异槲皮苷、鞣花酸、木犀草素、芹菜素]的含量,并结合化学计量学与熵权-灰色关联度法分析综合质量,为该制剂的质量控制和评价提供科学依据。

方法:

采用Shim-pack GIST-HP C18色谱柱;流动相为甲醇(A)-0.1%甲酸的水溶液(B),梯度洗脱;柱温35 ℃;进样量3 μL。微生信平台进行聚类分析(cluster analysis,CA),SIMCA 14.1软件进行主成分分析(principal component analysis,PCA)和正交偏最小二乘判别分析(orthogonal partial least squares discriminant analysis,OPLS-DA),以VIP值>1.0为标准筛选质量差异物;采用熵权-关联度法分析综合质量。

结果:

15个成分在各自范围内线性关系良好,r均≥0.999 0;精密度RSD均<3.00%;稳定性和重复性良好,RSD均<5.00%;平均加样回收率为93.62%~105.10%,RSD为1.18%~4.37%。CA和PCA分析将14批清心沉香八味散(丸)聚为两大类:D厂家(D1~D4)聚为一类,A厂家和M厂家(A1~A4,M1~M6)聚为一类;OPLS-DA模式下筛选出6个差异标志物,分别为D-(-)-奎尼酸、沉香四醇、2-(2-苯乙基)色酮、去氢二异丁香酚、佛手柑内酯和槲皮素。结果表明,A2厂家综合质量最好。

结论:

建立的HPLC-MS/MS法可同时测定清心沉香八味散(丸)中15个成分,结合化学计量学与熵权-灰色关联度法可用于清心沉香八味散(丸)综合质量研究。

Objective:

An HPLC-MS/MS method was established for the simultaneous determination of 15 components [betaine, agaritol, bergamolide, 2-(2-phenylidene) chromone, dehydrodiisoeugenol, D-(-)-quinic acid, gallic acid, protocatechuic acid, hydroxysafflor yellow A, rutin, isoquercetin, ellagic acid, luteolin and apigenin] in Qingxin Chenxiang Bawei powder (pills). The quantitative method, combined with chemometrics and entropy weight-grey correlation degree method to analyze the comprehensive quality, provides a reference for the quality control and evaluation of this preparation.

Methods:

The analysis was conducted using a Shim-pack GIST-HP C18 chromatographic column, with the mobile phase consisting of methanol (A) and a 0,1% formic acid aqueous solution (B). A gradient elution was employed at a column temperature of 35 ℃, with an injection column of 3 μL. Weisenxin platform was used for cluster analysis (CA), SIMCA 14.1 software was used for principal component analysis (PCA) and orthogonal partial least squares discriminant analysis (OPLS-DA), and the compounds with variable important in projection (VIP) value greater than 1.0 were used as the criterion to screen quality differences. The entropy weight-correlation degree method was used to analyze the comprehensive quality.

Results:

The linear relationship of the 15 components was good in their respective ranges, and the linear correlation r was greater than or equal 0.999 0. Precision RSD were all lower than 3.00%. The stability and repeatability were good, RSD lower than 5.00%; The average recoveries were 93.62%~105.10%, and RSD was 1.18%~4.37%. CA and PCA analysis classified 14 batches of Qingxin Chenxiang Bawei powder (pills) into 2 categories: manufacturers D (D1~D4) were grouped into one group, manufacturers A and M (A1~A4, M1~M6) were grouped into one group. Six difference markers were screened in OPLS-DA mode, which were D-(-)-quinic acid, agaritol, 2-(2-phenylethyl) chromone, dehydrodiisoeugenol, bergamolactone and quercetin, respectively. A2 has the best comprehensive quality.

Conclusion:

The established HPLC-MS/MS method for the simultaneous determination of 15 components in Qingxin Chenxiang Bawei powder (pill) can be used for the comprehensive quality study of Qingxin Chenxiang Bawei powder (pill) combined with chemometrics and entropy weight correlation degree method.

清心沉香八味制剂目前在市面上有散剂和丸剂(水丸)2种剂型,原方收载于《中华人民共和国卫生部药品标准》(蒙药分册),由沉香、广枣、檀香、紫檀香、红花、肉豆蔻、天竺黄和北沙参八味药组成,是蒙医临床常用药物之一[1],具有清肺、理气、镇静安神等功效,主要适用于治疗心肺火盛、胸闷不适、心慌气短等“赫依”性病症[2]。《中国医学百科全书·蒙医学》认为“赫依”是诱发疾病的主要原因,“赫依性疾病”是赫依偏盛导致功能紊乱引起的诸病总称。方中沉香,味辛、盖,性凉,清热止痛、利呼吸,以抑赫依为主要功效[3];现代研究表明,沉香具有镇静催眠、镇痛、抗心肌缺血等药理活性[4],为主药。广枣具有行气活血、安神养心之功,可镇赫依[5];紫檀香具有消肿止痛、清热血之功,以调节赫依血相讧[6];红花具有清热、活血止痛、滋养正精之功,可清赫依之热[7];肉豆蔻则有调胃火、消食开胃之功,可祛赫依[8];四药皆为辅药,共辅主药纠正赫依偏盛。檀香能清肺热,北沙参可止咳化痰[9],二药共为助药。诸药合用共同发挥清肺理气、镇静安神之功,此方为治疗赫依性疾病的良药。
目前,关于清心沉香八味制剂的质量研究主要以单一成分研究为主[10-11]。复方药物具有多成分、多靶点和多作用途径的特点[12],因此有必要建立一个客观多成分的综合质量标准,不仅有利于对该制剂深入研究,亦有助于临床用药安全和推广。
HPLC-MS/MS在多反应监测模式(multiple reaction monitoring,MRM)下具有简便快捷、灵敏度高、选择性高等特点[13]。化学计量学能够建立复杂模型,综合分析对质量控制具有显著贡献的主要成分[14]。熵权-灰色关联分析可对各指标的综合评判值进行直观表达,避免主观判断影响数据客观性[15]。基于此,本研究采用HPLC-MS/MS技术同时测定清心沉香八味制剂中甜菜碱、沉香四醇、佛手柑内酯、2-(2-苯乙基)色酮、去氢二异丁香酚、D-(-)-奎尼酸、没食子酸、原儿茶酸、羟基红花黄色素A、芦丁、异槲皮苷、鞣花酸、槲皮素、木犀草素和芹菜素15个成分的含量,并结合化学计量学与熵权-灰色关联度法对14批清心沉香八味制剂进行综合质量分析,旨在为该制剂的质量控制与评价提供科学依据。
LC—MS 8045型高效液相色谱-三重四极杆质谱联用仪,配备电喷雾离子源和LC—20AD型输液泵(日本岛津公司);AUW120D型十万分之一电子天平(日本岛津公司);BSA224S型万分之一天平(赛多利斯科学仪器有限公司);KQ5200E型超声波清洗器(广州博勒泰科有限公司)。
收集3个厂家共14批次清心沉香八味散(丸),分别为A厂家散剂(批号:210930,210739,230709,230506,编号:A1~A4)、D厂家丸剂(批号:220507,220706,220814,221101,编号:D1~D4)、M厂家散剂(批号:2306053,2304007,2210003,2211010,2112012,2303024,编号:M1~M6),A厂家和M厂家散剂规格均为3 g·袋-1,D厂家丸剂规格为1 g·8粒-1。以上样品制法根据《中华人民共和国卫生部药品标准》(蒙药分册),将处方中八味药粉碎成细粉,过筛,混匀(散剂按剂量分装即可),用凉开水泛丸,打光,干燥,约400粒·50 g-1

分别取适量甜菜碱、沉香四醇、佛手柑内酯、2-(2-苯乙基)色酮、去氢二异丁香酚、D-(-)-奎尼酸、没食子酸、原儿茶酸、羟基红花黄色素A、芦丁、异槲皮苷、鞣花酸、槲皮素、木犀草素、芹菜素,精密称定,置于不同10 mL量瓶中,用甲醇溶解定容至刻度,配制成浓度分别为0.549,0.285,0.360,0.294,0.300,0.717,0.323,0.275,0.495,0.565,0.333,0.685,0.326,0.320,0.316 mg·mL-1的对照品母液,分别精密量取各对照品母液适量置于同一10 mL量瓶中,用甲醇稀释至刻度,得到质量浓度分别为713.70,3 990.00,4.32,735.00,300.00,14 340.00,1 162.80,467.50,6 682.50,113.00,66.60,548.00,27.97,6.40,13.27 ng·mL-1的混合对照品溶液。
取样品粉末适量(过3号筛),称取约0.5 g,精密称定,置具塞锥形瓶中,精密加入75%甲醇8 mL,称定质量,超声处理(功率250 W,频率90 kHz) 55 min,放冷,再称定质量,用75%甲醇补足减失质量,摇匀,0.22 μm有机滤膜过滤,滤液稀释50倍,即得。
Shim-pack GIST-HP C18色谱柱(100 mm×2.1 mm,3 μm);流动相为甲醇(A)-0.1%甲酸的水溶液(B),梯度洗脱(0.01~0.9 min,10%~22%A;0.9~1.2 min,22%~45%A;1.2~3 min,45%~60%A;3.0~5.5 min,60%~78%A;5.5~6.2 min,78%~95%A;6.2~6.7 min,95%A;6.7~7.51 min,95%~10%A;7.51~9.5 min,10%A);体积流量0.25 mL·min-1;柱温35 ℃;进样量3 μL。质谱采用电喷雾离子源(electrospray ionization,ESI),正、负离子扫描;MRM模式,分段采集;喷雾电压为3.80 kV(+)和3.50 kV(-),雾化气体积流量为3 L·min-1;加热气体积流量为10 L·min-1;接口温度为300 ℃;脱溶剂温度为526 ℃;加热块温度为400 ℃。各成分质谱参数见表1
精密量取上述混合对照品溶液250,500,1 000,2 000,3 000和4 000 μL,分别置于不同5 mL容量瓶中,甲醇稀释至刻度线,再精密量取混合对照品溶液适量,于“3”项下条件进样分析。以对照品进样浓度为横坐标(x),峰面积为纵坐标(y)进行回归分析,15个成分在各自范围内线性关系良好,见表2
精密量取混合液3 000 μL,置于5 mL容量瓶中,甲醇稀释至刻度线,于“3”项下条件分别测定6次,计算得上述15个成分峰面积RSD分别为0.94%,1.61%,1.13%,1.40%,1.22%,0.58%,2.82%,0.63%,0.63%,2.54%,2.64%,1.97%,1.70%,2.89%和2.65%,表明仪器精密度良好。
取M1号样品(批号:2306053)6份,按“2”项下方法制备供试品溶液,于“3”项下条件分别进样分析,计算得上述15个成分平均含量分别为42.43,1 572.99,0.19,203.40,82.95,3 979.67,150.08,90.21,1 786.16,16.47,5.10,100.41,3.82,0.95和0.97 μg·g-1,RSD分别为0.82%,0.41%,1.51%,1.00%,1.97%,0.88%,1.78%,1.26%,1.71%,2.22%,1.94%,2.88%,3.99%,2.11%和2.99%,表明该方法具有良好的重复性。
取M1号样品,按“2”项下方法制备供试品溶液,于“3”项下条件在0,2,4,8,12,24 h时进样分析,计算得上述15个成分峰面积RSD分别为0.68%,0.26%,2.00%,1.17%,3.43%,1.04%,1.93%,1.11%,0.82%,2.33%,1.59%,4.23%,2.39%,3.49%和3.04%,表明样品溶液在24 h内具有良好稳定性。
称取已知成分含量的M1号样品6份,每份约0.250 0 g,分别按样品中成分浓度质量约100%的比例加入对照品,按“2”项下方法制备供试品溶液,于“3”项下条件分别进样测定,计算得上述15个成分平均加样回收率分别为99.47%,96.41%,103.56%,95.88%,92.41%,93.90%,97.58%,101.23%,98.43%,94.4%,102.23%,102.10%,93.62%,103.71%和105.1%,RSD分别为3.32%,1.81%,4.17%,3.39%,3.64%,1.56%,2.63%,2.37%,1.76%,3.23%,3.48%,3.33%,4.37%,4.11%和3.34%。
取14批清心沉香八味散(丸),按“2”项下方法制备供试品溶液,于“3”项下条件进样测定,计算上述15个成分含量,结果见表3
将14批样品中15个成分的含量数据导入微生信平台进行CA分析,采用完全聚类法,距离度量方法设定为欧氏距离[16],结果见图1。以成分含量表达量为指标可以聚为两大类:A厂家和M厂家(A1~A4,M1~M4)聚为第一类,D厂家(D1~D4)聚为第二类,说明A厂家和M厂家样品间质量具有一致性,三厂家整体质量具有一定差异。
为探究质量差异,以14批清心沉香八味散(丸)15个成分的含量为变量,导入SIMCA 14.1软件中进行无监督判别PCA分析,结果见图2。样品A1~A4,M1~M4与样品D1~D4分离明显,A厂家内部样品间明显离散性较大,表明A厂家质量差异较大。
为了降低与组别无关变量的影响,并能更好地获得样品组间的差异来源信息,采用SIMCA 14.1软件的有监督模式进行OPLS-DA,进一步分析14批样品质量,构建的模型累计解释能力参数R2X为0.937,R2Y为0.964,预测能力参数Q2(cum)为0.768,以上3个参数均>0.5,表明此模型具有良好的解释能力[17],见图3。结果表明,A1~A4和M1~M4,D1~D4分别聚为一类,且聚类效果更明显,其结果与CA和PCA分析结果相同,进一步说明样品间存在质量差异。为筛选出导致样品差异的主要成分,以变量权重值(variable importance in projection,VIP)>1为标准筛选质量差异物[18]。由图4可知,共筛选出6个质量差异物,分别为D-(-)-奎尼酸、沉香四醇、2-(2-苯乙基)色酮、去氢二异丁香酚、佛手柑内酯和槲皮素,即该6个成分是造成14批样品质量差异的主要成分。
熵权法可通过衡量每个指标的信息熵(Ej)自动确定各指标Wi,从而避免主观赋权的偏差。其中Ej衡量指标的“信息不确定性”或“离散性”,Ej越大,指标的变化越分散,意味着该指标对评估贡献越小,Wi越小;Ej越小,指标变化越集中,贡献越大,Wi越大。计算方法如下。
假设有a个评价对象,b个评价指标(本研究中a=14,b=15),xij为第i个评价对象下的第j个评价指标所对应的数据(i=1,2,…,a; j=1,2,…,b),按如下公式进行分析。
按如下公式计算Ej值和Wi值(结果见表4),其中,若Pij=0,则定义lnPij=0。
由于不同性质的指标可能对结果产生显著影响,因此在计算过程中需要去除量纲的影响,对数据进行规范化处理。按如下公式进行计算,其中Xik为原始数据,Xk为样品第k个指标均值。
在计算母序列与特征序列之间的关联度时,首先选取最优参考序列(即所有样品中每个指标的最大值Ysk)和最差参考序列(即所有样品中每个指标的最小值Ytk),则最优参考序列关联系数计算公式如下,式中ρ为分辨系数,本研究ρ值取表4中相关指标Wi值。

根据指标相对关联度大小对评价对象进行排名,结果见表5。A厂家中A2综合质量最好,A厂家、D厂家、M厂家相对关联度分别为0.470~0.523,0.496~0.504和0.458~0.463,提示A厂家质量差异程度最大。
为保证研究的科学性,所选成分指标参考了《中华人民共和国药典》2020年版一部,其中明确规定了沉香、广枣、红花、肉豆蔻的含量测定指标分别为沉香四醇、没食子酸、羟基红花黄色素A和去氢二异丁香酚,其他成分指标则参考了相关文献研究[19-22]
本研究中15个待测成分在不同流动相条件下分离效果和色谱强度皆不相同,由此课题组考察了不同流动相(甲醇-0.1%甲酸水、甲醇-0.2%甲酸水)对实验结果的影响,结果表明当流动相为甲醇-0.1%甲酸水时各成分色谱峰峰形最优、分离度最好;在对待测成分质谱条件考察时,由于甜菜碱为叔胺碱类化合物带有较强的电负性,即有较强的接受质子能力[23],为了得到更高响应度的色谱峰选择正离子模式分析甜菜碱。为全面分析各化合物,本研究采用MRM和正负离子模式扫描,之后经电压优化,选择响应度更高的参数作为化合物最终质谱参数进行分析。
在清心沉香八味散(丸)综合质量研究中,以15个成分的含量为指标,首先采用CA,PCA和OPLS-DA对3个厂家14批次样品进行了全面质量分析。结果显示,14批次样品可以分为两大类(D厂家聚为第一类,A厂家和M厂家聚为第二类)。为了进一步探讨质量差异的来源,以OPLS-DA模式分析,VIP值>1作为标准,筛选出6个质量差异物,分别为D-(-)-奎尼酸、沉香四醇、2-(2-苯乙基)色酮、去氢二异丁香酚、佛手柑内酯、槲皮素。鉴于此制剂在《中华人民共和国卫生部药品标准》(蒙药分册)中并没有含量测定项,因此可将这6个质量差异物作为含量测定项指标,为该药的生产质量控制提供参考依据。在质量评价研究中,熵权法通过客观赋权避免了主观判断误差,灰色关联度分析则可利用已知信息预测和解释未知信息[24]。从结果可知,A厂家、D厂家、M厂家相对关联度分别在0.470~0.523,0.496~0.504和0.458~0.463,A厂家中A2综合质量最好,但该厂家的相对关联度范围较大,与PCA分析中样品间离散性结果一致,说明其质量均一性较差。相对关联度结果与化学计量分析中聚类结果大致相同,亦进一步验证了前者的准确性,若以剂型和厂家为单位,则D厂家质量均一性最好,综合质量最好,后续课题组将继续增大样本量以探究综合质量是否与剂型或不同厂家制备工艺有关。
综上所述,本研究采用HPLC-MS/MS法同时测定清心沉香八味散(丸)中15个成分的含量,具有快速分析、专属性强和稳定性强的特点,结合化学计量学与熵权-关联度法可用于清心沉香八味散(丸)综合质量分析,为其质量控制与评价提供科学依据。
