Transmembrane protein 16A (TMEM16A) is a Ca2+-activated Cl- channel that is overexpressed in various malignant tumors. The gene is located in chromosome 11q13 region, and its amplification is one of the main reasons for the high expression of this protein. In addition, transcriptional regulation, epigenetic regulation and microRNAs (miRNAs) can promote TMEM16A expression in many cancer cells. TMEM16A not only activates a variety of signaling pathways that promote tumor cell proliferation, migration and invasion, but also plays an important role in inhibiting tumor cell apoptosis. Therefore, the development of anticancer drugs targeting TMEM16A is of great clinical significance. This article reviews the mechanism of TMEM16A in malignant tumors, summarizes the signaling pathways of TMEM16A in promoting tumor development, and analyzes the key role of TMEM16A as a potential biomarker in tumor development. TMEM16A inhibitors may become a new treatment for cancers with high expression of TMEM16A.

TMEM16A plays a crucial role in tumor development through its involvement in multiple signaling pathways. TMEM16A overexpression can enhance EGFR phosphorylation and promote EGF secretion, thereby activating STAT3/ROCK1 signaling pathways and SOCE. This activation promotes tumor cell proliferation and migration. TMEM16A induces ERK activation through the PI3K/AKT signaling pathway, which promotes tumor cell-cycle progression. TMEM16A activates NF-κB signaling pathway, contributing to tumor cell proliferation, migration and invasion. Overexpression of TMEM16A can activate the TGF-β signaling pathway, promoting TGF-β secretion, downregulating E-cadherin expression, and facilitating EMT, thereby enhancing tumor cell migration and invasion. Knockdown of TMEM16A promotes tumor cell apoptosis by activating the ASK1/p38/JNK/JUN signaling pathway. Knockdown of TMEM16A activates TNF-α signaling pathway, leading to FADD phosphorylation and caspase activation, thereby promoting tumor cell apoptosis, figureFileSmall=N9aFYhhYZhmIvqnKQvu4Sw==, figureFileBig=H/B9AR8xKthFurrzcsV9HQ==, tableContent=null), ArticleFig(id=1308497859369074881, tenantId=1146029695717560320, journalId=1189982191388893191, articleId=1308455469509534195, language=EN, label=null, caption=null, figureFileSmall=null, figureFileBig=null, tableContent=

3 (signal transducer and activator of transcription 3, STAT3)、c-myc和核因子κB (nuclear factor‑κB, NF-κB) [29, 30, 40]。
