The chemical constituents from Brassica rapa were identified by various chromatographic techniques including silica gel, reversed-phase silica gel, macroporous resin and Sephadex LH-20 column chromatography. Eight compounds were isolated from this plant. The isolated compounds were elucidated by physicochemical properties and spectroscopic methods, including extensive 1D, 2D-NMR, and HR-ESI-MS techniques. Compounds 1, 2 are new triterpenoids and 3-7 were isolated for the first time from Brassica rapa. The cytotoxic effect of compounds 1 and 2 were tested by MTT assay against five cancer cell lines. The result showed that all compounds exhibit growth inhibition for the cancer lines. Compound 1 has an IC50 value of 5.87 μmol·L-1 for growth inhibition of leukemia cell line HL-60, and IC50 value for compound 2 was 10.32 μmol·L-1.

综合运用大孔吸附树脂、硅胶柱色谱、半制备型高效液相及Sephadex LH-20凝胶柱色谱等各种色谱技术,对蔓菁Brassicarapa L.乙醇提取物的化学成分进行系统研究,共分离得到4个三萜、3个黄酮和1个甾体类化合物。根据其理化性质和高分辨及二维核磁等波谱数据鉴定化合物的结构,其中12为新的三萜类化合物,3~7首次从该植物中分离得到。细胞毒活性显示12对5种常见的癌细胞均显示一定的抑制作用,尤其是对HL-60具有显著的细胞毒活性,其IC50分别为5.87和10.32 μmol·L-1

综合运用大孔吸附树脂、硅胶柱色谱、半制备型高效液相及Sephadex LH-20凝胶柱色谱等各种色谱技术,对蔓菁Brassicarapa L.乙醇提取物的化学成分进行系统研究,共分离得到4个三萜、3个黄酮和1个甾体类化合物。根据其理化性质和高分辨及二维核磁等波谱数据鉴定化合物的结构,其中12为新的三萜类化合物,3~7首次从该植物中分离得到。细胞毒活性显示12对5种常见的癌细胞均显示一定的抑制作用,尤其是对HL-60具有显著的细胞毒活性,其IC50分别为5.87和10.32 μmol·L-1

The chemical constituents from Brassica rapa were identified by various chromatographic techniques including silica gel, reversed-phase silica gel, macroporous resin and Sephadex LH-20 column chromatography. Eight compounds were isolated from this plant. The isolated compounds were elucidated by physicochemical properties and spectroscopic methods, including extensive 1D, 2D-NMR, and HR-ESI-MS techniques. Compounds 1, 2 are new triterpenoids and 3-7 were isolated for the first time from Brassica rapa. The cytotoxic effect of compounds 1 and 2 were tested by MTT assay against five cancer cell lines. The result showed that all compounds exhibit growth inhibition for the cancer lines. Compound 1 has an IC50 value of 5.87 μmol·L-1 for growth inhibition of leukemia cell line HL-60, and IC50 value for compound 2 was 10.32 μmol·L-1.

蔓菁(Brassica rapa)是一种药食同源植物, 属于十字花科, 在我国西藏、新疆等高海拔地区均有分布, 秋季块根及种子在成熟时采收。在藏药典籍中, 蔓菁又名“妞玛”、“圆根”、或者“芫根”[1], 而在维药中则称为“恰玛古”, 药用部分均为种子或其根部[2]。传统理论认为, 其味甘、性温, 具有清热解毒, 滋补增氧的特性, 临床用于治疗各种中毒症及身体虚弱者[3-5]。化学成分主要包括:皂苷类、黄酮类、多糖类、生物碱类及挥发油, Zhou等[6-8]从蔓菁叶中分离得到了槲皮素、山柰酚、异鼠李素等黄酮类成分, 但未见关于其块根具体的化学成分研究报道。现代药理研究表明, 蔓菁皂苷具有提高小鼠体内谷胱苷肽过氧化酶(GSH-PX)活性的作用[9], 同时具有抗疲劳、抗缺氧、抗肿瘤及降血糖等药理活性[8]。本课题组在前期研究中发现, 蔓菁的80%醇提取物对白血病细胞HL-60表现出较为显著的细胞毒活性。为了寻找具有更高活性单体化合物, 探明蔓菁的药理活性物质基础, 本研究对蔓菁块根的80%乙醇提取物的化学成分进行系统研究, 分离得到4个三萜、3个黄酮以及1个甾体类化合物。分别鉴定为:蔓菁皂苷A (1)、蔓菁酸(2)、桔梗皂苷D (3)、桔梗皂苷E (4)、异鼠李素-3-O-β-D-吡喃葡萄糖基-7-O-β-D-龙胆双糖苷(5)、异鼠李素-3, 7-二-O-β-D-吡喃葡萄糖苷(6)、胡萝卜苷(7)和异鼠李素(8) (图 1)。其中12为新的三萜类化合物, 3~7为首次从蔓菁中分离得到。同时, 采用MTT方式对化合物12进行了体外细胞毒活性进行评价, 结果表明化合物12对HL-60、A549、DU145、CEM、HeLa均表现出一定的抑制作用, 尤其是对HL-60能够显著抑制肿瘤细胞株的生长, 其IC50分别为5.87和10.32 μmol·L-1。本文旨在报道这些化合物的提取分离、结构鉴定以及它们的药理活性。
化合物1  白色无定形粉末(甲醇), [α]D25 -22.3 (c 0.1, MeOH); mp 218~220 ℃; Libermann-Berchard和Molish反应均呈阳性, TLC薄层色谱喷5%的硫酸乙醇溶液, 105 ℃加热显色, 呈紫红色, 提示其可能为三萜皂苷类化合物。ESI-MS m/z 941.5 [M-H]-、943.5 [M+H]+、965.5 [M+Na]+, 推测相对分子质量为942; HR-ESI-MS m/z 965.462 55 [M+Na]+ (计算值: 965.462 78, C48H78NaO18), 确定其分子式为C48H78O18, 不饱和度为10;在1H NMR谱中, 在高场区域出现7个角甲基信号δH 0.72、0.96、1.00、1.23、1.28、1.30、1.44 (s, each 3H)和1个糖上甲基信号δH 1.79 (d, J = 6.0 Hz, 3H); 同时谱图中间区域出现1个齐墩果烷型烯氢信号δH 5.31 (br s, 1H)以及3个糖的端基氢信号δH 4.99 (d, J = 7.8 Hz, 1H)、δH 5.73 (d, J = 7.6 Hz, 1H)、δH 6.23 (s, 1H)。在13C NMR (表 1)中, 低场部分除了出现齐墩果烷型烯烃双键信号δC 144.5、122.1以外, 还有1个羰基碳信号δC 172.2。分析HSQC、HMBC图谱, 除糖基碳信号外, 还有3个连氧碳信号δC 91.0 (d)、69.0 (d)、63.3 (t), 说明除了齐墩果酸母核C-3与氧相连外还有1个亚甲基和次甲基与氧相连。通过DEPT 135°、HSQC、HMBC对化合物的碳氢信号进行全归属, 与文献报道的已知化合物spartitrioside核磁数据比较一致[10], 化合物1的C-17位向低场移动3.2 ppm, C-22的则向高场移动了47.2 ppm, 说明22位没有连接羟基, 同时C-28位向低场移动4.2 ppm。HMBC谱进一步确证了发生具体变化的位置, δH 1.28 (H-28)与C-16 (δC 69.0)、C-17 (δC 39.6)、C-22 (δC 28.4)存在远程相关; 在1H-1H COSY图谱中δH 1.65 (H-21)与δH 0.84 (H-22)存在相关; 此外, 化合物1与spartitrioside的糖部分1H NMR和13C NMR基本一致, 同时HMBC也证实3位碳与糖基端基氢具有远程相关, 说明其糖部分与母核3位碳相连形成糖苷键。
对化合物1进行酸水解, 通过薄层色谱的方式与对照品进行比较分析, 该化合物含有D-葡萄糖醛酸、L-鼠李糖及D-葡萄糖; 结合HSQC、13C NMR谱中出现3个糖基端基碳信号δC 105.2 (d)、102.1 (d)、101.4 (d), 以及1H NMR谱图端基氢的偶合常数, 表明3个糖基分别为β-D-葡萄糖醛酸吡喃糖基、α-L-鼠李糖基以及β-D-吡喃葡萄糖基[11]
糖的连接顺序由HBMC谱确定:在3-O糖链中, β-D-吡喃葡萄糖端基氢(δH 4.99)与母核3位碳(δC 91.0)具有远程相关, β-D-吡喃葡萄糖端基氢(δH 5.73)和葡萄糖醛酸的C-2' (δC 78.0)具有远程相关, 而α-L-鼠李糖端基氢(δH 6.23)与葡萄糖的C-2″ (δC 76.3)远程相关; 在NOESY图谱中H-27 (δH 1.30)与H-23 (δH 1.44)相关, 说明3位羟基为β构型, 而H-16 (δH 5.01)与H-28 (δH 1.28)相关, 说明16位羟基为α构型(图 2)。综合以上分析, 确认该化合物结构为: 3-O-α-L-吡喃鼠李糖基-(1→2)-β-D-吡喃葡萄糖基-(1→2)-β-D-葡萄糖醛酸吡喃糖基-3β, 16α, 24-三羟基-齐墩果烷, 命名为蔓菁皂苷A (brasside A)。
化合物2  白色针状晶体(甲醇), [α]20D +37.8 (c 0.1, MeOH); mp 267~269 ℃; Libermann-Berchard和Molish反应均呈阳性, TLC薄层色谱喷5%的硫酸乙醇溶液, 105 ℃加热显色, 呈紫红色, 提示其可能为三萜类化合物。ESI-MS m/z 503.3 [M-H]-、527.3 [M+Na]+, 推测相对分子质量为504; HR-ESI-MS m/z 527.329 20 [M+Na]+ (计算值: 527.329 35, C30H48NaO6), 确定其分子式为C30H48O6, 不饱和度为7;从1H NMR谱图可以看出, 信号主要集中在高场区域, 除了6个角甲基氢信号[δH 0.97、1.01、1.31、1.34、1.56、1.59 (s, each 3H)]以外, 还有5个连氧氢信号(δH 3.70~4.93)。结合13C NMR及DEPT 135°, 发现该化合物共有30个碳信号, 包括9个季碳、5个叔碳、10个仲碳、6个伯碳。低场部分有3个季碳信号, 包括1个羰基(δC 179.9)及1个烯烃碳信号(δC 137.7、128.0), 但在氢谱中无相应的烯信号, 说明该烯烃氢全部被取代基取代。同时还有4个连氧碳信号δC 73.8 (d)、72.6 (d)、71.6(d)、68.4 (t)。结合HSQC及HMBC, 对化合物的碳氢信号进行全归属, 核磁数据与centellasapogenol A进行比较, 数据基本一致[12], 同时化合物2的分子质量比centellasapogenol A多16 Da, 推测化合物2比centellasapogenol A多一个羟基, 这与核磁数据推测结果相吻合。利用HMBC判断羟基的连接位置, δH 4.90与C-14 (δC 26.9)、C-17 (δC 55.2)、C-18 (δC 128.0)、C-28 (δC 179.9)存在远程相关, 说明16位连接有羟基。同时通过NOESY确认羟基的相对构型, δH 4.57 (H-2)\4.29 (H-3)\1.73 (H-5)\1.55 (H-27)相关, δH 4.90 (H-16)\1.30 (H-26)相关, 说明2、3位羟基为β构型, 而16位羟基为α构型(图 3)。
综上所述, 化合物2结构鉴定为2β, 3β, 16α, 24-四羟基-13(18)-烯-齐墩果酸, 经Scifinder结构检索, 未见相关报道, 命名为蔓菁酸(brassica acid)。
化合物3~8为已知化合物, 经核磁分析并与文献数据进行比较, 鉴定为桔梗皂苷D (3)[13]、桔梗皂苷E (4)[14]、异鼠李素-3-O-β-D-吡喃葡萄糖基-7-O-β-D-龙苷(5)[15]、异鼠李素-3, 7-二-O-β-D-吡喃葡萄糖苷(6)[16]、异鼠李素(7)[16]、胡萝卜苷(8)[14]
采用MTT法对所得的新化合物进行了5种常见癌细胞的细胞毒活性实验, 并以依托泊苷作为阳性对照进行评价, 化合物12均显示出一定的药理活性(表 2), 特别是对于HL-60能够显著抑制肿瘤细胞的生产, 这对于进一步深度开发该药用资源提供参考依据。
Waters Vion® IMS QTof型高分辨质谱仪(Waters公司, 美国), Bruker AV 600型核磁共振波谱仪(TMS为内标), 中压制备色谱仪(MPLC) FS-9200T (天津博纳艾杰尔科技有限公司), 半制备型高效液相色谱仪Waters 2545, 2487检测器(美国Waters公司), 酶标仪及培养箱(Thermo公司, 美国), 细胞株(DU145、CEM、HeLa、A549、HL-60, ATCC公司, 美国), 柱色谱硅胶、薄层色谱硅胶(青岛海洋化工), 乙腈、甲醇(色谱纯), 其余试剂为国产分析纯。
实验所用蔓菁药材2018年5月采购于西藏拉萨市, 经中国科学院成都生物研究所赵佐成研究员鉴定为蔓菁(Brassica rapa)的地下部分, 标本保存于成都师范学院化学与生命科学学院标本室(标本号: 20180510)。
将新鲜的蔓菁20 kg切片, 阴干, 加10倍体积的80%乙醇, 加热回流提取3次, 每次2 h。将提取液合并, 减压回收至基本无醇味, 再加等体积的水进行分散稀释。中速滤纸过滤, 将滤液上D-101大孔吸附树脂, 水洗至流出液基本无色, 直接用95%乙醇进行洗脱, 浓缩洗脱液至干, 得95%乙醇洗脱部分55 g。将95%乙醇洗脱部分(50 g)用甲醇溶解, 60~80目硅胶拌样, 进行硅胶柱色谱分离。洗脱剂为二氯甲烷-甲醇-水(20:1:0.01→1:1:0.01), 约1 L收集一份, 共收集36份。用TLC对各流分进行检测, 显色剂为5%硫酸-乙醇溶液, 合并相同部分, 得到Fr. A~F。对具有明显斑点的Fr. B (1.1 g)进行再次硅胶柱色谱分离, TLC过程监控, 收集较为单一部分Fr. B1和Fr. B6; Fr. B1用甲醇重结晶, 得化合物7 (32 mg), Fr. B6用半制备液相色谱进行分离纯化, 流动相为甲醇-水(86:14), 检测波长为205 nm, 得到化合物2 (17 mg); 将Fr. C (2.6 g)用常压反相硅胶柱色谱进行再次粗分, 以甲醇-水(60:40→90:10)进行梯度洗脱, TLC过程监控, 得到Fr. C2、Fr. C5和Fr. C8三个较单一部分, 对得到的每一部分样品再用半制备高效液相进行精制, 流动相为乙腈-水(65:35), 检测波长为205 nm, 得到化合物3 (28 mg)、化合物4 (16 mg), 化合物8 (25 mg)。Fr. D (0.8 g)再次进行硅胶柱色谱分离, TLC过程检测, 得到一个较为单一的组分Fr. D1, 将Fr. D1进行半制备反相色谱分离, 流动相为乙腈-水(68:32), 检测波长为205 nm, 收集主要目标化合物, 减压浓缩至干, 甲醇溶解, 过滤, 滤液上Sephadex LH-20再次精分, 甲醇为洗脱剂, 得到化合物1 (14 mg); Fr. F (0.8 g)再次进行硅胶柱色谱分离, TLC进行过程检测, 得到一个较为单一的组分Fr. F1, 将Fr. F1进行半制备反相色谱分离, 流动相为乙腈-0.2%磷酸水(25:75), 检测波长为254 nm, 收集主要目标峰, 减压浓缩至析出固体, 得到化合物5 (17 mg)、6 (10 mg)。

化合物12, 使用二甲基亚砜(DMSO)溶解。配成浓度为100 mmol·L-1的母液, 储存于-20 ℃。临用时用相应的培养液将其稀释为100、10、1及0.1 μmol·L-1进行实验。DMSO配置的样品进行实验时, DMSO的终浓度为0.1%。阳性药依托泊苷注射乳浓度为100、10、1和0.1 μmol·L-1
取对数生长期的细胞, 调整适当的细胞密度, 接种于96孔板内, 每空100 μL, 培养于37 ℃、5% CO2的培养箱内。培养24 h后加药, 作用72 h。分设空白组、给药组和阳性对照组, 每组设4个复孔。将细胞与0.25 mg·mL-1 MTT于37 ℃共同抚育3~4 h, 离心吸除上清液, 加入DMSO 100 μL, 完全溶解后使用酶标仪于492 nm测定其OD值。最后以空白组OD值为100%, 计算细胞抑制率。数据采用SPSS (16.0)统计软件包进行检验分析。
