The chemical constituents of Litsea coreana were investigated using chromatographic methods, including column chromatography on silica gel, MCI, and semi-preparation HPLC. Two compounds were isolated and identified as hawktealignan A (1) and cinnamophilin (2) by NMR analyses as well as their physical and chemical properties. Compound 1 is a new lignan and compound 2 was isolated from this plant for the first time.

采用硅胶、MCI、半制备高效液相色谱等柱色谱方法对贵州老鹰茶乙酸乙酯部位的化学成分进行分离纯化,根据NMR数据以及理化性质鉴定了化合物的结构,分离得到2个木脂素类化合物分别鉴定为老鹰茶木脂素A(1)和cinnamophilin(2)。化合物1为新化合物,化合物2为首次从该植物中分离得到。

采用硅胶、MCI、半制备高效液相色谱等柱色谱方法对贵州老鹰茶乙酸乙酯部位的化学成分进行分离纯化,根据NMR数据以及理化性质鉴定了化合物的结构,分离得到2个木脂素类化合物分别鉴定为老鹰茶木脂素A(1)和cinnamophilin(2)。化合物1为新化合物,化合物2为首次从该植物中分离得到。

The chemical constituents of Litsea coreana were investigated using chromatographic methods, including column chromatography on silica gel, MCI, and semi-preparation HPLC. Two compounds were isolated and identified as hawktealignan A (1) and cinnamophilin (2) by NMR analyses as well as their physical and chemical properties. Compound 1 is a new lignan and compound 2 was isolated from this plant for the first time.

老鹰茶也称“白茶”, 为我国南方各民族民间长期饮用的一种植物代用茶, 尤以重庆及贵州遵义地区少数民族饮用历史悠久。其汤色金黄带红, 具有强烈的樟科植物芳香味, 性甘甜, 有明目解渴和消食去胀之功效, 为药食同源植物[1]。老鹰茶主要是以樟科木姜子属植物毛豹皮樟[Litsea coreana Lévl. var. lanuginosa (Migo) Yang et P. H. Huang]的嫩梢和叶片为原料[2], 其主要生长于云南、贵州、四川、安徽等地[3], 现代药理研究表明老鹰茶具有抑菌、保肝、抗癌、抗氧化、降血糖、降血脂、抗炎、免疫调节等功效[4-7]。为进一步了解老鹰茶的活性物质基础, 本项目对其化学成分进行了深入研究, 从中分离得到2个木脂素类化合物, 分别鉴定为老鹰茶木脂素A (1)和cinnamophilin (2), 其中化合物1为新化合物, 化合物2为首次从该植物中分离得到(图 1)。
化合物1为黄色油状物。[α]D20+22.0 (c 0.18, CHCl3)。HR-ESI-MS给出其准分子离子峰[M+H]+m/z 357.141 6 (计算值: 357.133 8), 确定其分子式为C20H20O6, 有11个不饱和度。1H NMR (400 MHz, CDCl3) (表 1)结合HSQC谱显示有2组ABX芳环信号δH 6.89 (1H, d, J = 1.7 Hz)、6.84 (1H, d, J = 8.1 Hz)、6.82 (1H, dd, J = 8.1, 1.7 Hz)、δH 7.43 (1H, d, J = 1.7 Hz)、6.84 (1H, d, J = 8.2 Hz)和7.52 (1H, dd, J = 8.2, 1.7 Hz), 1个亚甲二氧基信号δH 6.04 (2H, s), 1个甲氧基信号δH 3.89 (3H, s), 3个次甲基信号δH 4.16 (1H, m)、4.48 (1H, d, J = 7.4 Hz)和2.47 (1H, m), 1个亚甲基信号δH 4.35 (1H, dd, J = 8.8, 7.3 Hz)、4.29 (1H, dd, J = 8.8, 7.3 Hz), 1个甲基信号δH 0.84 (3H, d, J = 6.9 Hz)。13C NMR (100 MHz, CDCl3)谱结合HSQC谱显示有1个甲基碳信号δC 13.5, 3个次甲基碳信号δC 46.2、49.1、87.8, 1个甲氧基碳信号δC 56.2, 1个亚甲基碳信号δC 69.8, 1个亚甲二氧基碳信号δC 102.2, 1个酮羰基碳信号δC 198.3和12个芳香碳信号δC 152.2、148.6、146.8、145.4、133.2、132.7、124.8、119.4、114.4、108.7、108.2和108.1, 通过数据分析和文献对比推测化合物1可能为木脂素类化合物[8,9]。HMBC谱(图 2)中H-2 (δH 7.43)和C-3 (δC 148.6)、C-4 (δC 152.2)、C-6 (δC 124.8)有相关, H-6 (δH 7.52)和C-2 (δC 108.1)、C-4 (δC 152.2)有相关, 亚甲二氧基(δH 6.04)和C-3 (δC 148.6)、C-4 (δC 152.2)有相关, 可以确定亚甲二氧基处于一个苯环的C-3和C-4上; HMBC谱中H-2和C-7 (δC 198.3)有相关, H-6和C-7有相关, 说明羰基连在C-1上。另外一个苯环体系, HMBC谱中H-2′ (δH 6.89)和C-3′ (δC 146.8)、C-4′ (δC 145.4)、C-6′ (δC 119.4)有相关, H-5′ (δH 6.84)和C-3′ (δC 146.8)有相关, 甲氧基(δH 3.89)和C-3′ (δC 146.8)有相关, 可以确定甲氧基处在苯环的C-3′上, 羟基处于苯环的C-4′上。HMBC谱中甲基(δH 0.84)和3个次甲基碳都有相关, 亚甲基(δH 4.35)和羰基有相关, 说明木脂素的两个C6C3单元通过C-8和C-8′相连, 进一步亚甲基(δH 4.35)和C-7′ (δC 87.8)有相关, 说明C-9和C-7′之间有氧桥相连。至此化合物1的平面结构得到确证。NOESY谱(图 2)中, H-7′和H-9′有相关, 说明H-7′和甲基处于同面, 进一步通过H-7′和H-8′的偶合常数J = 7.4 Hz推测出H-7′和H-8′处于反式[10]。NOESY谱中H-8和H-8′有相关, 但是H-8和H-9′没有相关, 说明H-8和H-8′处于同面, 即H-8和甲基处于反面, 至此推测出化合物1的相对构型。通过对比实测ECD和计算ECD曲线[11] (图 3), 将化合物1确定为(8S, 7′R, 8′R)-老鹰茶木脂素A。
Agilent DD2 400-MR型核磁共振仪测定, TMS为内标(美国Agilent公司); 紫外光谱测试用UV3600型紫外可见近红外分光光度计(日本岛津公司); Xevo G2-XS QTof超高液相色谱-质谱联用仪(美国Waters公司); JASCO J-810圆二色光谱仪(日本分光公司); 反相C18填料(ODS) (日本富士硅化学株式会社); MCI树脂(日本Mitsubishi公司); 薄层色谱硅胶GF254和柱色谱硅胶(300~400目, 中国青岛海洋化工有限公司); LC3000型半制备高效液相色谱仪(中国北京创新通恒公司), HPLC柱C18, 5 μm, 10 mm×250 mm (日本YMC公司)。
老鹰茶于2017年采自贵州遵义绥阳县, 由遵义医科大学药学院生药学教研室聂绪强副教授鉴定为毛豹皮樟[Litsea coreana Lévl. var. lanuginosa (Migo) Yang et P. H. Huang], 植物标本(No.20171022)现存放在遵义医科大学药学院。
毛豹皮樟叶干重4.6 kg, 粉碎后用90%甲醇80 ℃回流提取3次, 每次3 h, 提取液过滤, MCI柱色谱(60 mm×460 mm, 甲醇-水, 90:10, 体积比)除叶绿素, 减压浓缩得甲醇浸膏843 g, 将其分散于2 L水中, 依次用石油醚(3×2 L)、乙酸乙酯(3×2 L)、正丁醇(3×2 L)萃取, 减压浓缩得乙酸乙酯萃取物258 g, 该部位浸膏经硅胶柱色谱(80 mm×600 mm, 石油醚-乙酸乙酯梯度洗脱)分为7个部分: Fr.1~7。Fr.4经半制备HPLC分离(甲醇-水, 72:28, 体积比, 6.0 mL·min-1)得到化合物1 (11.6 mg, tR = 10.7 min)和化合物2 (16.3 mg, tR = 16.9 min)。
化合物1  黄色油状物。[α]D20 +22.0 (c 0.18, CHCl3)。HR-ESI-MS m/z [M+H]+ 357.141 6 (计算值: 357.133 8); m/z [M+Na]+ 379.120 6 (计算值: 379.115 8)。1H NMR (400 MHz, CDCl3)和13C NMR (100 MHz, CDCl3)数据见表 1。鉴定化合物1为老鹰茶木脂素A。
化合物2  黄色油状物。1H NMR (400 MHz, CDCl3) δH 0.85 (3H, d, J = 6.8 Hz, H-9′), l.l3 (3H, d, J = 6.7 Hz, H-9), 2.22 (1H, m, H-8′), 2.44 (1H, dd, J = 13.5, 7.0 Hz, H-7′), 2.56 (lH, dd, J = 13.5, 7.7 Hz, H-7′), 3.34 (1H, m, H-8), 3.84 (3H, s, OMe), 3.88 (3H, s, OMe), 6.67 (2H, m, H-2′, H-6′), 6.84 (2H, d, J = 8.1 Hz, H-5, H-5′), 7.22 (lH, dd, J = 8.1, 1.8 Hz, H-6), 7.40 (lH, d, J = 1.8 Hz, H-2);13C NMR (100 MHz, CDCl3) δC 203.0 (C-7), 150.3 (C-4), 146.8 (C-3), 146.6 (C-3′), 144.1 (C-4′), 132.8 (C-l′), 129.7 (C-l), 123.5 (C-5), 122.2 (C-2′), 114.3 (C-6), 113.8 (C-6′), 111.8 (C-5′), 110.5 (C-2), 56.2 (OMe), 56.1 (OMe), 42.9 (C-8), 41.6 (C-8′), 38.0 (C-7′), 15.5 (C-9), 11.6 (C-9′)。以上数据与文献报道基本一致[12], 故鉴定化合物2为cinnamophilin。
