A tadalafil analogue was detected during routine screenings from two "fatigue reliever, immunity enhancer" dietary supplements by using UHPLC/Q-TOF HRMS. The MS2 spectrum of this compound was almost identical to that of 2-hydroxypropylnortadalafil. However, the retention time of this analogue was different from that of the 2-hydroxypropylnortadalafil isomers. The analogue was purified by using preparative HPLC and the structure was elucidated by mass spectrometric and NMR spectroscopic experiments. The spectral data suggested that the analogue bore a 3-hydroxypropyl group instead of the N-methyl group in tadalafil. The structure was further confirmed by comparison of the 1H NMR spectra data with those of the reference standard, and thus named as 3-hydroxypropylnortadalafil. The structure is first reported in China.

采用超高效液相色谱串联四级杆-飞行时间高分辨质谱(UHPLC/Q-TOF HRMS)法,从两种宣称具有缓解疲劳、增强免疫力的保健食品中发现了一种他达拉非衍生物。该化合物与2-羟丙基去甲他达拉非的质谱图特征非常相似,但保留时间却略有差异。采用制备液相色谱仪对该衍生物进行分离纯化,经质谱和氢核磁共振分析鉴定为3-羟丙基去甲他达拉非(3-hydroxypropylnortadalafil),为国内首次报道。

A tadalafil analogue was detected during routine screenings from two "fatigue reliever, immunity enhancer" dietary supplements by using UHPLC/Q-TOF HRMS. The MS2 spectrum of this compound was almost identical to that of 2-hydroxypropylnortadalafil. However, the retention time of this analogue was different from that of the 2-hydroxypropylnortadalafil isomers. The analogue was purified by using preparative HPLC and the structure was elucidated by mass spectrometric and NMR spectroscopic experiments. The spectral data suggested that the analogue bore a 3-hydroxypropyl group instead of the N-methyl group in tadalafil. The structure was further confirmed by comparison of the 1H NMR spectra data with those of the reference standard, and thus named as 3-hydroxypropylnortadalafil. The structure is first reported in China.

近年来, 中成药和保健食品中非法添加壮阳作用的磷酸二酯酶(phosphodiesterase 5, PDE-5)抑制剂屡见报道[1]。针对这种现象, 国家食品药品监督管理局先后颁布了多项法规, 2017年以前主要针对西地那非、豪莫西地那非、伐地那非、他达拉非、氨基他达拉非等13种PDE-5抑制剂检测。不法生产厂商为了逃避监管, 非法添加上述PDE-5抑制剂的衍生物, 其中最常见的是在西地那非、他达拉非、艾迪地非、伐地那非等化合物母核的基础上进行结构修饰, 如文献报道中成药或保健食品中发现了用乙酰氨基[2]、羟乙基[3]等取代甲基结构的他达拉非类衍生物。这些衍生物的药理毒理学作用尚不明确, 严重危害消费者健康。同时, 层出不穷的衍生物也给食品药品监管工作带来了一定的挑战。为全面监测和打击食品中添加PDE-5抑制剂衍生物和其他具备抗疲劳的非法药物, 国家食品药品监督管理局2018年颁布了本课题组制定的《食品中那非类物质的测定》 (BJS201805), 对那非类物质的监管增加至了90种, 但是新的PDE-5抑制剂衍生物仍然不断的出现。
本课题组前期已经建立了中成药和保健食品中102种PDE-5抑制剂的快速定性定量分析方法[4]。在日常检验中, 从两批声称具有缓解体力疲劳、提高免疫力的咖啡饮品中发现了一种新型他达拉非衍生物。采用制备液相色谱方法从样品中分离该未知化合物, 通过质谱信息与核磁数据分析, 对其进行了准确结构鉴定。
仪器与试药  超高效液相色谱仪串联四级杆-飞行时间高分辨质谱仪(Agilent 1290/Agilent 6530, 美国Agilent公司), 分析天平(CP 224S, 德国Sartorius赛多利斯公司)。制备液相色谱仪(Agilent 1260 InfinityⅡ, 美国Agilent公司), Varian核磁共振仪(500 MHz, 美国瓦里安公司)。乙腈(MS级)、甲醇、甲酸、甲酸铵(色谱纯)均购自德国默克公司; 去离子水(Milli-Q超纯水); 2-羟丙基去甲他达拉非(加拿大TLC公司, 批号1516-091A7, 异构体混合物)。两种咖啡饮品购自网络平台。
供试品溶液制备  分别取两种咖啡饮品1 g, 加甲醇10 mL, 超声30 min, 过滤, 稀释1 000倍, 作为供试品溶液。
色谱条件  Agilent ZORBAX RRHD Eclipse Plus C18色谱柱(100 mm×3 mm, 1.8 μm); 柱温: 30 ℃; 进样体积: 1 μL; 流动相: A为0.1%甲酸-水溶液, B为0.1%甲酸-乙腈溶液; 梯度洗脱程序: 0~2 min, 20%~30% B; 2~5 min, 30% B; 5~6 min, 30%~45% B; 6~12 min, 45%~70% B; 12~14 min, 70%~98% B; 14~16 min, 98% B; 16~1 min, 98%~20% B; 17~20 min, 20% B。流速: 0.4 mL·min-1
质谱条件  采用电喷雾离子源(ESI)正离子模式检测, 扫描范围m/z: 100~1 000。离子源参数:雾化气(N2): 35 psi (1 psi ≈ 6.9 kPa); 干燥气(N2)流速15 L·min-1, 温度200 ℃; 鞘气(N2)流速12 L·min-1, 温度350 ℃; 毛细管电压3.5 kV, 碎裂电压250 V, 二级谱碰撞池(CID)电压5~40 eV。
分离纯化  分离纯化采用Varian Prep Star制备液相色谱仪和岛津Shim-pack PREP-ODS色谱柱(20 mm×250 mm, 15 μm); 流动相为甲醇-水(65:35), 流速为20 mL·min-1, 检测器为紫外检测器(检测波长222 nm)。分别取咖啡饮品12 g, 加甲醇120 mL, 振摇30 min, 过滤, 滤液浓缩至20 mL进样。
采用UHPLC/Q-TOF HRMS法对试样中非法添加物筛查时, 在两个咖啡饮品中同时发现一个m/z为434.172 0的未知化合物, 保留时间约为10.20 min, 与2-羟丙基去甲他达拉非具有相同的元素组成(C24H23N3O5), 但保留时间略有差别, 2-羟丙基去甲他达拉非为一对光学异构体, 保留时间分别为10.93和11.56 min (图 1), 二级质谱图相似(图 2), 主要碎片离子包括m/z 135.043 8、169.075 5、197.069 0、262.086 0、284.138 6和312.135 5, 推断其可能为2-羟丙基去甲他达拉非的同分异构体。
采用制备液相色谱仪对两个咖啡饮品中未知化合物进行分离纯化, 收集主色谱峰(波长220 nm)成分, 减压浓缩除去溶剂, 干燥后分别得未知化合物8.2 mg、13.3 mg。
从两个样品中分离出来的化合物具有相似的1H NMR数据, 见表 1[5]δH6.75 (2H, m)和6.85 (1H, s)为苯并二噁唑苯环的1, 3, 4取代的H信号; δH5.9 (2H, s)为苯并二噁唑亚甲基的H信号; δH7.54 (1H, d, J = 8.0 Hz), 7.00 (1H, t, J = 7.5 Hz), 7.06 (1H, t, J = 7.5 Hz), 7.30 (1H, d, J = 8.0 Hz)是邻二取代苯的H信号; δH 4.52 (1H, s)和11.06 (1H, s)分别为O-H和N-H信号; δH 4.43 (1H, dd, J = 12, 4.5 Hz)为烯烃次甲基质子信号; δH 6.16 (1H, s)提示次甲基连接多个吸电子基; δH 3.93 (1H, d, J = 17 Hz)和4.23 (1H, d, J = 17 Hz)为亚甲基两个不等价氢相互偶合的H信号; 与2-羟丙基去甲他达拉非的母核H信号一致。但是, 未知化合物的1H NMR没有2-羟丙基去甲他达拉非的δH 3.88 (1H, br m)信号(连氧脂肪族次甲基H信号)和δH1.04 (3H, d, J = 6.3 Hz)信号(脂肪族甲基H信号), 而多出了δH 3.42 (2H, m)信号(与氧相连的亚甲基信号)和δH 1.66(2H, m)信号(脂肪族亚甲基H信号), 推测可能为2-羟丙基去甲他达拉非结构中2-羟丙基被3-羟丙基取代。
为了进一步确定未知化合物结构, 定制了构型为6R, 12aR的3-羟丙基去甲他达拉非对照品(加拿大TLC公司), 经比较发现, 未知化合物的保留时间、精确分子量、高分辨二级质谱图及1H NMR数据与对照品均一致。由此, 可确证未知物的结构为2-羟丙基去甲他达拉非的同分异构体3-羟丙基去甲他达拉非, 化合物结构见图 3。3-羟丙基去甲他达拉非的可能的质谱裂解途径如图 4所示。m/z 312.135 3为母离子失去苯并二噁唑基团的碎片, 且提示双酮哌嗪环结构上有一个C3H7O片段。m/z 169.075 5和m/z 197.069 0为他达拉非类化合物中吡啶并吲哚环系统部分的碎片, m/z 135.043 8为苯并二噁唑基团的碎片, 这三个离子是具有双酮哌嗪环的他达拉非类衍生物常见的离子碎片。
他达拉非及其衍生物具有较稳定的母核结构, 通过其质谱碎片信息, 可以快速对他达拉非衍生物进行结构推测。目前发现的他达拉非衍生物多数都是双酮哌嗪环上发生结构变化, 一类是在2-N的取代上有差异, 如N-ethyl nortadalafil[6]、cyclopentyl nortadalafil[7], 另外一类没有双酮哌嗪环, 如chloropretadalafil[8]、diethylaminopretadalafil[9]。比较这些化合物的二级质谱图, 一般含有m/z 135、169、197、262等碎片离子, 低质量区碎片分子量比较类似, 可以帮助此类化合物的快速推断。在他达拉非母核6、12a位有两个手性中心, 存在4种构型, 但顺式他达拉非衍生物(6R, 12aR)活性明显优于反式[10], 根据产品所描述的功效推断该非法添加的衍生物结构最大可能性为6R、12aR构型。经分离得到的非法添加的未知化合物与定制的6R、12aR构型3-羟丙基去甲他达拉非对照品的保留时间、一级二级质谱及1H NMR均能匹配, 故确定其为6R、12aR构型的3-羟丙基去甲他达拉非。
后期实验中进一步测定了两种咖啡饮品中3-羟丙基去甲他达拉非含量, 结果显示该化合物含量分别高达1.70和2.05 mg·g-1, 根据产品标签的用法用量, 每次服用量高达22和25 mg, 超过了同类上市药物他达拉非的每次服用量(10 mg), 对人体存在严重的潜在危险。该化合物在国内尚未被报道, 药理毒理作用尚不清楚, 对人体的潜在危害认知还较为有限, 值得引起关注和追踪。
采用UHPLC/Q-TOF HRMS从两种咖啡饮品中检测发现了2-羟丙基去甲他达拉非的同分异构体, 经分离纯化和1H NMR确证, 该化合物为2-羟丙基去甲他达拉非的同分异构体3-羟丙基去甲他达拉非, 该化合物化学名为(6R, 12aR)-6-(1, 3-苯并间二氧戊环-5-基)-2-(3-羟丙基)-2, 3, 6, 7, 12, 12a-六氢化吡嗪并[1', 2':1, 6]-吡啶并[3, 4-b]吲哚-1, 4-二酮。此化合物的发现提供了一种新型他达拉非衍生物发现的新思路, 有助于新型衍生物结构的发现, 保障监管工作及时有效。
