HPLC-RID and HPIC-CD methods were established for the determination of sucrose octasulfate content in irinotecan hydrochloride liposomes for injection. HPLC-RID:This method was performed on a Kromasil 100-5-NH2 column (250 mm×4.6 mm, 5 μm) with a mobile phase of 0.8 mol·L-1 ammonium sulfate buffer (pH 3.5)-acetonitrile (83:17). The flow rate, column temperature and detector temperature were maintained at 1 mL·min-1, 30℃ and 30℃ respectively. HPIC-CD:This method was performed on an anion exchange column Dionex InPacTM AS11-HC (250 mm×4 mm, 9 μm) with an eluent of 30 mmol·L-1 sodium hydroxide solution. The flow rate was 1.5 mL·min-1, the column temperature was 30℃ and the detector temperature was 35℃. The HPLC-RID method and HPIC-CD method were validated with respects to specificity, limit of detection, limit of quantitation, linearity, precision, accuracy, stability and robustness and met the validation requirements. There were no significant differences between the HPIC-CD and HPLC-RID methods according to T-test analysis, both of which were applicable for the measurement of sucrose octasulfate concentration in irinotecan hydrochloride liposomes for injection. However, the HPLC-CD was better at the following aspects:higher detection sensitivity, simpler sample pretreatment, lower time and money spent, better environmental protection.

采用高效液相色谱-示差折光检测器（HPLC-RID）和高压离子色谱-电导检测器（HPIC-CD）对盐酸伊立替康脂质体注射液中蔗糖八硫酸酯进行含量测定并比较，为脂质体制剂提供科学有效的分析方法。HPLC-RID法：采用Kromasil 100-5-NH2柱（250 mm×4.6 mm，5 μm），以0.8 mol·L-1硫酸铵溶液（pH 3.5）-乙腈（83：17）为流动相，柱温30℃，检测器温度30℃，流速1.0 mL·min-1。HPIC-CD法：采用Dionex InPacTM AS11-HC阴离子交换柱（250 mm×4 mm，9 μm），流速1.5 mL·min-1，柱温30℃，检测器温度35℃，淋洗液30 mmol·L-1氢氧化钠溶液。结果表明HPLC-RID法和HPIC-CD法的专属性、检测限、定量限、线性、精密度、准确度、稳定性以及耐用性验证均符合要求。两种方法均可准确测定盐酸伊立替康脂质体注射液中蔗糖八硫酸酯的含量，经过独立样本T-检验表明两种方法测得结果无显著性差异。与HPLC-RID法相比，HPIC-CD法具有专属性强、灵敏度高、分析速度快、操作简便、成本低、环保等优点。

采用高效液相色谱-示差折光检测器（HPLC-RID）和高压离子色谱-电导检测器（HPIC-CD）对盐酸伊立替康脂质体注射液中蔗糖八硫酸酯进行含量测定并比较，为脂质体制剂提供科学有效的分析方法。HPLC-RID法：采用Kromasil 100-5-NH2柱（250 mm×4.6 mm，5 μm），以0.8 mol·L-1硫酸铵溶液（pH 3.5）-乙腈（83：17）为流动相，柱温30℃，检测器温度30℃，流速1.0 mL·min-1。HPIC-CD法：采用Dionex InPacTM AS11-HC阴离子交换柱（250 mm×4 mm，9 μm），流速1.5 mL·min-1，柱温30℃，检测器温度35℃，淋洗液30 mmol·L-1氢氧化钠溶液。结果表明HPLC-RID法和HPIC-CD法的专属性、检测限、定量限、线性、精密度、准确度、稳定性以及耐用性验证均符合要求。两种方法均可准确测定盐酸伊立替康脂质体注射液中蔗糖八硫酸酯的含量，经过独立样本T-检验表明两种方法测得结果无显著性差异。与HPLC-RID法相比，HPIC-CD法具有专属性强、灵敏度高、分析速度快、操作简便、成本低、环保等优点。

HPLC-RID and HPIC-CD methods were established for the determination of sucrose octasulfate content in irinotecan hydrochloride liposomes for injection. HPLC-RID:This method was performed on a Kromasil 100-5-NH2 column (250 mm×4.6 mm, 5 μm) with a mobile phase of 0.8 mol·L-1 ammonium sulfate buffer (pH 3.5)-acetonitrile (83:17). The flow rate, column temperature and detector temperature were maintained at 1 mL·min-1, 30℃ and 30℃ respectively. HPIC-CD:This method was performed on an anion exchange column Dionex InPacTM AS11-HC (250 mm×4 mm, 9 μm) with an eluent of 30 mmol·L-1 sodium hydroxide solution. The flow rate was 1.5 mL·min-1, the column temperature was 30℃ and the detector temperature was 35℃. The HPLC-RID method and HPIC-CD method were validated with respects to specificity, limit of detection, limit of quantitation, linearity, precision, accuracy, stability and robustness and met the validation requirements. There were no significant differences between the HPIC-CD and HPLC-RID methods according to T-test analysis, both of which were applicable for the measurement of sucrose octasulfate concentration in irinotecan hydrochloride liposomes for injection. However, the HPLC-CD was better at the following aspects:higher detection sensitivity, simpler sample pretreatment, lower time and money spent, better environmental protection.

蔗糖八硫酸酯是以C12H14O35S8-R8为结构的一组化合物(图 1), 其中R可代表K、Na、Al等元素, 该化合物早期应用在治疗溃疡为主的药物中。2015年随着盐酸伊立替康脂质体在美国上市[1], 蔗糖八硫酸酯作为新型辅料引起药界同仁的关注。本文采用蔗糖八硫酸酯三乙胺(triethylammonium sucrose octasulfate, TEA8 SOS)梯度法制备盐酸伊立替康脂质体注射液, 可提高盐酸伊立替康疗效和缓释作用, 降低不良反应, 增加靶向性和生物利用度。在仿制药物的开发中, 蔗糖八硫酸酯的定量测定作为关键参数用于工艺和生产中的质量控制。例如, 蔗糖八硫酸酯三乙胺形成的pH梯度影响脂质体的包封率[2]。此外, 采用蔗糖八硫酸酯作为新型非活性辅料时, 应关注其带来的不良影响和体内活性, 以及类似生长因子的生理活性[3], 鉴于蔗糖八硫酸酯的含量测定对脂质体注射液的制备和评价起到关键作用, 因此有必要对其进行分析和质量控制。
国内外文献对蔗糖八硫酸酯的研究报道较少, 根据美国药典[4]、欧洲药典[5]、日本药典[6]收载以及Wang[7]、Sun等[8]相关文献报道, 均采用HPLC-RID法测定硫糖铝片中蔗糖八硫酸酯的含量, 重现上述方法后并不理想, 因此优化前处理方法和色谱条件。优化后HPLC-RID法前处理较为复杂且耗时长, 为寻找简单快速的分析方法应用于脂质体制剂, 本文同时建立离子色谱的分析方法。离子色谱是分析离子的一种液相色谱方法, 以离子交换机制为主[9]。经发展, 高压离子色谱可降低工作量, 提高分析效率[10], 应用的范围从分析水中常见阴阳离子与有机酸, 到分析极性有机化合物以及生物样品中的糖(单糖、寡糖)、氨基酸、肽、蛋白质等更加广阔的领域[11]。根据蔗糖八硫酸酯以离子形式存在于脂质体中等特点[12], 同时采用HPIC-CD法对其进行定量测定。结果表明两种方法均可用于测定盐酸伊立替康脂质体中蔗糖八硫酸酯的含量, 但HPIC-CD法灵敏度更高, 操作更为简便。
材料与方法

仪器  岛津LC-20A高效液相色谱仪(配备LC-20AD二元泵、SIL-20AD自动进样器、CBM-20A控制器、CTO-20AC柱温箱、RID-20A视差折光检测器及LabSolutions工作站Version 6.72 SP2), Thermo Dionex ICS-5000+SP离子色谱色谱仪[配有带抑制器(AERS, 4 mm)的电导检测器, Chromeleon 7.2.8工作站)], FE28 pH计(瑞士Mettler Toledo公司), CPA225D电子天平(德国Sartorius公司), IQ7000超纯水系统(美国Millipore公司), XTM0061多管涡旋混匀仪、MD200-2氮吹仪(杭州奥盛仪器有限公司)。
试剂与样品  蔗糖八硫酸酯钠对照品(批号: 20190320, 含量以99.80%计)购自上海赢瑞生物医药科技有限公司, 盐酸伊立替康脂质体注射液样品(批号: No.01、No.02、No.03)由玻思韬控释药业有限公司自制, 乙腈(色谱纯)购自上海星可高纯溶剂有限公司, 硫酸铵(分析纯)、Triton X-100 (分析纯)均购自MACKLIN公司, 磷酸(分析纯)购自天津市大茂化学试剂厂, 氢氧化钠溶液(质量分数为50%)购自赛默飞世尔科技(中国)有限公司, 纯化水由玻思韬控释药业有限公司自制。
色谱条件
HPLC-RID法分析条件  采用Kromasil 100-5-NH2柱(250 mm×4.6 mm, 5 μm), 以0.8 mol·L-1硫酸铵溶液(用磷酸调节pH值至3.5)-乙腈(83:17)为流动相, 柱温30 ℃, 检测器为示差折光检测器, 温度30 ℃, 流速1.0 mL·min-1, 进样量100 μL。
HPIC-CD法分析条件  采用Dionex InPacTM AS11-HC阴离子交换柱(250 mm×4 mm, 9 μm), 淋洗液30 mmol·L-1氢氧化钠溶液, 柱温30 ℃, 检测器为电导检测器(抑制器AERS, 4 mm), 温度35 ℃, 流速1.5 mL·min-1, 进样量25 μL。
对照溶液的制备
对照品溶液Ⅰ的制备  精密称取蔗糖八硫酸酯钠对照品100 mg, 置10 mL量瓶中, 加流动相溶解并稀释至刻度, 摇匀, 作为对照储备液Ⅰ。精密量取对照品储备液Ⅰ适量, 流动相稀释摇匀后, 精密量取上述溶液1 mL加95%甲醇溶液5 mL, 经氮气流下挥干溶剂, 加流动相溶解并稀释制成每1 mL中含0.26 mg的蔗糖八硫酸酯钠溶液, 涡旋5 min, 即得, 作为HPLC-RID法对照溶液。
对照品溶液Ⅱ的制备  精密称取蔗糖八硫酸酯钠对照品100 mg, 置10 mL量瓶中, 加超纯水溶解并稀释至刻度, 摇匀, 作为对照储备液Ⅱ。精密量取对照品储备液Ⅱ适量, 加超纯水溶解并稀释制成每1 mL中含0.26 mg的蔗糖八硫酸酯钠溶液, 摇匀即得, 作为HPIC-CD法对照溶液。
供试品溶液的制备
供试品溶液Ⅰ的制备  精密量取盐酸伊立替康脂质体注射液0.1 mL, 加95%甲醇溶液5 mL破乳, 振摇, 经氮气流下挥干溶剂, 加流动相1 mL复溶, 涡旋5 min, 滤过, 取续滤液即得, 作为HPLC-RID法供试品溶液。
供试品溶液Ⅱ的制备  精密量取盐酸伊立替康脂质体注射液0.1 mL, 置于5 mL量瓶中, 加1% Triton X-100溶液破乳, 并稀释至刻度, 振摇, 滤过, 取续滤液即得, 作为HPIC-CD法供试品溶液。
分析方法
专属性  分别取空白溶剂、对照品溶液和供试品溶液, 按“HPLC-RID法分析条件”和“HPIC-CD法分析条件”考察专属性, 记录色谱图。
仪器精密度  精密量取对照溶液Ⅰ和Ⅱ, 按“HPLC-RID法分析条件”和“HPIC-CD法分析条件”连续进样6次, 记录色谱图。
检测限和定量限  取对照储备液Ⅰ和Ⅱ适量逐级稀释, 以信噪比10:1作为定量限、3:1作为检测限进行测定。
线性与范围  取对照储备液Ⅰ和Ⅱ, 分别制备线性溶液, 稀释浓度范围为0.121 5、0.151 8、0.455 5、0.759 2、1.215 mg·mL-1和0.003 807、0.038 07、0.053 30、0.076 14、0.152 3 mg·mL-1。按“HPLC-RID法分析条件”和“HPIC-CD法分析条件”进样分析。
回收率  取已知含量的盐酸伊立替康脂质体注射液9份, 每份精密量取0.1 mL, 分别按已知含量的80%、100%和120% 3个水平加入对照品, 按“供试品溶液的制备”项下制备低、中、高3种不同浓度的供试品溶液, 每一种浓度平行制备3份, 按“HPLC-RID法分析条件”和“HPIC-CD法分析条件”进样测定, 计算回收率。
重复性  取同一批盐酸伊立替康脂质体注射液, 分别平行配制6份供试品溶液Ⅰ和Ⅱ, 按“HPLC-RID法分析条件”和“HPIC-CD法分析条件”进样分析。
稳定性  精密量取同一批盐酸伊立替康脂质体注射液0.1 mL, 制备供试品溶液Ⅰ和Ⅱ, 分别按“HPLC-RID法分析条件”和“HPIC-CD法分析条件”在0、4、8、12和24 h进样分析。
耐用性  分别考察流速变化±0.2 mL·min-1, 柱温变化±5 ℃, 流动相比例变化±5%对蔗糖八硫酸酯含量测定的影响。

1 方法学考察
1.1 专属性
通过空白溶剂、对照品溶液和供试品溶液的色谱图(图 2)考察专属性, 实验结果表明两种方法的溶剂和样品基质均对蔗糖八硫酸酯的测定无干扰, 专属性良好。
1.2 仪器精密度
HPLC-RID法和HPIC-CD法测得蔗糖八硫酸酯含量的RSD (n = 6)分别为0.70%和0.69%, 结果表明仪器精密度良好。
1.3 检测限和定量限
结果表明两种方法灵敏度良好, HPIC-CD法灵敏度高于HPLC-RID法, 结果见表 1。
1.4 线性与范围
以质量浓度x (mg·mL-1)为横坐标, 以峰面积y为纵坐标绘制标准曲线, 回归方程及相关系数见表 1。结果表明两种方法在各自线性范围内线性关系良好。
1.5 重复性
HPLC-RID法和HPIC-CD法测得蔗糖八硫酸酯的含量分别为2.653 ± 0.013、2.649 ± 0.032 mg·mL-1, RSD (n = 6)均小于5.0%, 结果表明方法重复性良好。
1.6 准确度
HPLC-RID法和HPIC-CD法测得蔗糖八硫酸酯的平均回收率(n= 9)分别为99.2 ± 3.3%、99.0 ± 2.7%, RSD (n = 9)均小于5.0%, 方法准确度良好。
1.7 稳定性
HPLC-RID法和HPIC-CD法测得蔗糖八硫酸酯含量的RSD分别为1.67%、2.01%, 表明供试品溶液在24 h内稳定。
1.8 耐用性
HPLC-RID法和HPIC-CD法在不同条件下测得蔗糖八硫酸酯含量的RSD (n = 6)分别为1.56%和1.94%, 结果表明两种方法耐用性良好。
2 统计学分析

从同一批盐酸伊立替康脂质体注射液中随机取样, 分别按照上述两种方法各制备8份供试品溶液并测定其含量。利用SPSS Statistical 17.0版软件对两种方法的测定结果进行统计学分析。Shapiro-Wilk检验两组数据的P值分别为0.137和0.421, 均大于0.05, 表明数据呈正态分布。两组数据相互独立, 因此采用独立样本t检验对其进行分析, 置信区间设定为95%。经F检验, P值为0.225大于0.05, 由此可知两总体方差无显著性差异。t检验得到的t统计量为-1.27, 对应的双尾概率P值为0.225, 大于显著性水平(α = 0.05), 表明两组数据差异无统计学意义。判断两种方法测定盐酸伊立替康脂质体注射液中蔗糖八硫酸酯的含量无显著性差异, 统计分析结果见表 2。
3 样品含量测定

取3批盐酸伊立替康脂质体注射液分别制备供试品溶液并测定其含量。HPLC-RID法和HPIC-CD法测得3批盐酸伊立替康脂质体中蔗糖八硫酸酯的含量分别为2.630 ± 0.029、2.718 ± 0.043、2.593 ± 0.054 mg·mL-1和2.675 ± 0.069、2.768 ± 0.088、2.538 ± 0.074 mg·mL-1。两种方法同批测得值相互比较, 其差异均小于5.0%。

HPLC-RID法: USP 42版以1 mol·L-1的硫酸铵为流动相, 使用氨基柱测定蔗糖八硫酸酯的含量, 要求蔗糖八硫酸酯拖尾因子在4以内, 理论塔板数大于400。但重现上述方法后并不理想, 原因如下:脂质体中蔗糖八硫酸酯的检测先经过破乳剂破乳, 破乳剂使蔗糖八硫酸酯从脂质体内部释放出来的同时造成其浓度被稀释, 较难达到示差折光检测器的定量限, 且破乳剂的峰展宽严重影响蔗糖八硫酸酯出峰。针对上述问题, 本文优化了相关前处理方法与分析条件, 最终采用高浓度有机试剂破乳, 氮气流下将溶剂挥干, 少量流动相复溶, 过滤得到供试品溶液, 此过程既减少蔗糖八硫酸酯稀释倍数又避免脂质体基质的干扰。优化色谱条件后蔗糖八硫酸酯的拖尾因子在1.00~1.50之间, 理论塔板数大于3 000。
HPIC-CD法:为开发简单快速的分析方法, 根据蔗糖八硫酸酯的结构与性质, 建立HPIC-CD法。现有文献报道未见采用HPIC-CD法测定蔗糖八硫酸酯含量, HPIC-CD法提供了与HPLC-RID法不同的前处理方式和检测方式, 根据离子色谱的检测原理避免了样品基质干扰, 可直接进样分析, 极大缩短了前处理时间、操作简便, 且分析过程未使用有机试剂, 环保。通过对破乳剂、淋洗液种类和浓度以及色谱条件的考察, 最终选取1% Triton X-100为破乳剂, 30 mmol·L-1氢氧化钠溶液为淋洗液等度洗脱。优化色谱条件, 蔗糖八硫酸酯的保留时间约为4 min, 拖尾因子在0.95~1.10之间, 理论塔板数大于5 000。
作者通过优化前处理和色谱条件重现HPLC-RID法, 同时建立了HPIC-CD法测定盐酸伊立替康脂质体中蔗糖八硫酸酯含量。两种方法均采用外标法以峰面积计算含量, 利用SPSS软件对两种方法测得的结果进行统计学分析, 结果表明两种检测方法没有显著性差异。与HPLC-RID法相比, HPIC-CD法优势在于极大缩短了前处理时间, 提高分析速度、避免脂质体基质干扰、灵敏度高、成本低、环保。采用HPIC-CD法不仅为蔗糖八硫酸酯的含量测定提供新的思路, 也为盐酸伊立替康脂质体注射液质量标准和质量控制的完善提供依据。
