Four alkaloids were isolated from the total alkaloids of the twigs and leaves of Alstonia yunnanensis (Apocynaceae) by using silica gel, ODS, Sephadex LH-20, and HPLC chromatography. Structures were determined by physical, chemical and spectroscopic methods and identified as N4-methylpseudoakuammigine (1), pseudoakuammigine (2), vinorine (3), picraline (4). Among them, compound 1 is a new monoterpenoid indole alkaloid.

采用硅胶、碳十八烷基反相键合硅胶(ODS)、SephadexLH-20、HPLC等色谱方法进行分离纯化,从夹竹桃科植物鸡骨常山枝叶的总生物碱部位中分离并鉴定了4个生物碱类化合物,根据理化性质与波谱数据分别鉴定为N4-甲基脱氧阿枯明(N4-methylpseudoakuammigine,1)、脱氧阿枯明(pseudoakuammigine,2)、维诺林(vinorine,3)、匹克拉林碱(picraline,4)。其中化合物1为一个新的生物碱类化合物。

采用硅胶、碳十八烷基反相键合硅胶(ODS)、SephadexLH-20、HPLC等色谱方法进行分离纯化,从夹竹桃科植物鸡骨常山枝叶的总生物碱部位中分离并鉴定了4个生物碱类化合物,根据理化性质与波谱数据分别鉴定为N4-甲基脱氧阿枯明(N4-methylpseudoakuammigine,1)、脱氧阿枯明(pseudoakuammigine,2)、维诺林(vinorine,3)、匹克拉林碱(picraline,4)。其中化合物1为一个新的生物碱类化合物。

Four alkaloids were isolated from the total alkaloids of the twigs and leaves of Alstonia yunnanensis (Apocynaceae) by using silica gel, ODS, Sephadex LH-20, and HPLC chromatography. Structures were determined by physical, chemical and spectroscopic methods and identified as N4-methylpseudoakuammigine (1), pseudoakuammigine (2), vinorine (3), picraline (4). Among them, compound 1 is a new monoterpenoid indole alkaloid.

鸡骨常山(Alstonia yunnanensis)为夹竹桃科(Apocynaceae)鸡骨常山属植物, 为直立灌木, 主要产于云南、贵州和广西等地区的海拔1 100~2 400米的山坡或沟谷地带。其根可供药用, 用于治疗高血压、发热、发炎、头痛等症[1]。生物碱类成分为该种植物特征性成分且具有多种药理活性, 其中化合物vinorine和vellosimine对大鼠W256、小鼠Lewis肺癌和B16黑色素瘤有一定的活性[2, 3]; 化合物alstiyunnanenine D和E以及alstoniascholarine I对骨肉瘤细胞株表现出明显的细胞毒活性[4]; perakine N4-oxide、raucaffrinoline N4-oxide和vinorine N4-oxide化合物对Cox-2具有选择性抑制(> 85%)作用[5], 以及抗神经阻断作用[6]。为寻找结构新颖、活性良好的生物碱类成分, 本实验利用正、反相硅胶柱以及HPLC等多种分离手段从鸡骨常山枝叶中分离并鉴定了4个生物碱类化合物(图 1), 分别为: N4-甲基脱氧阿枯明(N4-methylpseudoakuammigine, 1)、脱氧阿枯明(pseudoakuammigine, 2)、维诺林(vinorine, 3)、匹克拉林碱(picraline, 4)。其中化合物1为新的柯南因-士的宁型生物碱类化合物。
化合物1   淡黄色粉末。[α]D25+148.5 (c 0.66, CH3OH), UV λmax (CH3OH): 205、239、286 nm有最大吸收峰; IR (KBr)光谱中显示含羰基(1 675 cm-1)和苯环(1 582和1 470 cm-1)的特征吸收峰; HR-ESI-MS m/z 367.201 0 [M+H]+ (Calcd. for C22H27N2O3, 367.201 6), 可确定该化合物的分子式为C22H26N2O3, 不饱和度为11。
1H NMR谱中, 在低场区有1组芳香氢的自旋耦合质子信号[δH 7.57 (1H, d, J = 7.7 Hz, H-9), 7.15 (1H, t, J = 7.7 Hz, H-11), 6.81 (1H, t, J = 7.7 Hz, H-10), 6.74 (1H, d, J = 7.7 Hz, H-12)], 1个烯烃氢信号δH 5.71 (1H, q, J = 7.0 Hz, H-19);在高场区给出2个氮甲基质子信号[δH 3.22 (1H, s, N4-CH3), 2.85 (1H, s, N1-CH3)], 以及1个甲基质子信号[δH 1.82 (3H, dd, J = 7.0, 2.2 Hz, H-18)]。13C NMR中, 显示有22个碳信号, 提示其中包括1个羰基碳信号[δC 176.6 (COO-)]; 1组邻二取代苯环碳信号[δC 152.1 (C-13), 140.2 (C-8), 128.9 (C-11), 124.9 (C-9), 122.1 (C-10)和111.9 (C-12)]; 1组双键碳信号[δC 133.0 (C-20), 126.0 (C-19)]; 2个氮甲基碳信号[δC 57.9 (N4-CH3), 29.9 (N1-CH3)]; 1个甲基碳信号[δC 14.1 (C-18)]。化合物1的NMR、MS数据与已知化合物2比较只是少了1个甲氧基碳信号, 多了1个氮甲基碳信号, 同时HMBC谱中可见δH 3.22 (3H, s)与δC 68.1 (C-3)、62.8 (C-5)有远程相关(图 2), 从而可知化合物1N4位被甲基取代。借助于1D和2D NMR数据, 对化合物11H NMR和13C NMR信号进行了全归属(表 1)。在化合物1的NOESY谱中可观察到H-3与H-21α, H-21α与H-15有NOE相关, 确定H-3和H-15为α-H; 通过H-15与H-18的NOE相关确定C-19~C-20位双键为E构型。因此可以确定化合物1的相对构型。
为了进一步确定化合物1的绝对构型, 对化合物进行ECD测试并与已知化合物2进行比较分析, 两者的ECD图谱非常相似(图 3)。从而确定了化合物1的绝对构型为2S, 3S, 7S, 15S, 16R
综上所述, 确定化合物1为一个新的单萜吲哚类生物碱, 命名为N4-甲基脱氧阿枯明(N4-methylpseudoakuammigine), NMR数据见表 1
JASCO P-2000旋光仪、Jasco V-550紫外/可见光谱仪和Jasco FI/IR-480 Plus Fourier Transform型红外光谱仪(KBr压片) (日本Jasco公司); Chirascan分光仪(英国应用光物理公司); Finnigan LCQ Advantage MAX型质谱仪(Thermo Finnigan公司); Agilent 6210 ESI/TOF质谱仪、Agilent 1260分析型高效液相色谱仪和Agilent 1260制备型高效液相色谱仪(美国安捷伦公司); Bruker AV-600/400型核磁共振仪(美国Bruker公司); 柱色谱用硅胶(100~200, 200~300)目, 青岛海洋化工厂); 硅胶GF254薄层预制板(烟台化学工业研究所); ODS柱色谱材料(Merck公司); Sephadex LH-20 (美国Amersham Biosciences Swede公司); 所用试剂均为市售分析纯或色谱纯。
实验用药材鸡骨常山枝叶于2017年8月采自云南昆明, 由暨南大学周光雄教授鉴定为鸡骨常山(Alstonia yunnanensis)干燥枝叶。标本(No. CP2017082001)保存于暨南大学药学院。
鸡骨常山干燥枝叶20.0 kg, 粉碎成粗粉, 用95%乙醇在室温下渗漉提取3次, 减压浓缩得总浸膏2.0 kg, 加水混悬后, 用10% HCl调pH值到2~3, 氯仿萃取, 酸水层用氨水调pH值到9~10, 氯仿萃取, 萃取液减压浓缩得到粗总碱55.3 g。粗总碱部位经硅胶柱色谱, 氯仿-甲醇系统(100:0~0:100)梯度洗脱, 得到14个流份(Fr. 1~14)。Fr. 9经凝胶柱色谱(氯仿-甲醇1:1)分离得到6个部分(Fr. 9A~F), 其中Fr. 9B经Sephadex LH-20 (甲醇)、及HPLC分析制备得到化合物3 (5.9 mg)、4 (21.0 mg)。Fr.10经Sephadex LH-20 (氯仿-甲醇1:1)、ODS (甲醇-水, 20:80~0:100), 以及HPLC分析制备得到化合物1 (7.38 mg)、2 (10.6 mg)。
化合物1   淡黄色粉末, 改良碘化铋钾反应显阳性。[α]D25+148.48 (c 0.66, CH3OH); HR-ESI-MS m/z 367.201 0 [M+H]+ (Calcd. for C22H27N2O3, 367.201 6), 可确定该化合物的分子式为C22H26N2O3; UV λmax (CH3OH): 205、239、286 nm; IR (KBr): 3 429、2 963、1 675、1 582、1 470、1 455、1 384、765 cm-1; 1H NMR (600 MHz, CD3OD)和13C NMR (150 MHz, CD3OD)数据见表 1
化合物2   淡黄色粉末, 改良碘化铋钾反应显阳性。HR-ESI-MS m/z: 367.200 9 [M+H]+ (Calcd. for C22H27N2O3, 367.201 6); 1H NMR (400 MHz, CDCl3)和13C NMR (100 MHz, CDCl3)数据见表 1。以上数据与文献[7]报道基本一致, 故鉴定化合物2为脱氧阿枯明(pseudoakuammigine)。
