从巴戟天中克隆MEP途径中的1-脱氧-D-木酮糖5-磷酸还原异构酶基因MoDXR及其启动子序列，并进行生物信息学分析、启动子区顺式作用元件分析，以及进行原核表达分析。根据巴戟天转录组中DXR基因原始序列和NCBI-ORFfinder分析，设计特异性引物，并进行RT-PCR扩增和生物信息学分析；采用染色体步移克隆MoDXR基因的5'端启动子序列；通过亚细胞定位分析MoDXR基因在细胞中的位置；构建原核表达载体pET-28a-MoDXR，导入BL21（DE3）表达感受态细胞后，在IPTG诱导下表达。从巴戟天中克隆的MoDXR基因，其cDNA全长2 015 bp，预测的阅读框大小为1 425 bp，编码474个氨基酸，分子质量为51.27 kDa；BlastP序列比对分析表明MoDXR基因与其他植物的DXR基因具有高度同源性，如：咖啡树DXR（CaDXR）、萝芙木DXR（RvDXR）；系统进化发育树分析显示，巴戟天MoDXR蛋白与小粒咖啡和栀子的DXR蛋白聚为一类，其亲缘关系最近；由亚细胞定位可知MoDXR基因所编码的蛋白定位于叶绿体上；MoDXR基因5'端启动子序列长度为1 493 bp，包含了与光响应、胁迫响应和激素响应有关的多种调控元件；SDS-PAGE结果表明pET-28a-MoDXR重组蛋白的大小与预期相符，但是以不容性的包涵体的形式表达。成功克隆了MoDXR基因及其启动子序列并对其进行了生物信息学分析和启动子区顺式作用元件分析；后期需要优化MoDXR原核表达体系，为进一步纯化MoDXR蛋白，研究其结构和功能奠定基础。

The objective of this research was to clone 1-deoxy-D-xylulose 5-phosphate reductoisomerase gene (MoDXR) and its promoter sequence from Morinda officinalis and carry out bioinformatic analysis, cis-acting elements analysis, and prokaryotic expression. On the basis of the MoDXR gene sequence obtained from the M. officinalis transcriptome and with NCBI-ORFfinder analysis, a pair of specific primers were designed, and used for RT-PCR amplification. The promoter region sequence at the 5' end of MoDXR gene was isolated by the genome walking technique. Localization of MoDXR was carried out by subcellular analysis. The prokaryotic expression plasmid pET-28a-MoDXR was constructed and transfected into Escherichia coli BL21(DE3) chemically-competent cells; the recombiant plasmid expressed fusion protein after the induction by IPTG. The full-length cDNA of MoDXR was 2 015 bp, and open reading frame (ORF) size was 1 425 bp, and it encoded 474 amino acid residues and had a molecular mass of 51.27 kD. Sequence comparison with BlastP to the NCBI database revealed that MoDXR had high sequence similarity with many other DXRs, such as Coffea arabica DXR (CaDXR) and Rauvolfia verticillata DXR (RvDXR). A phylogenetic tree revealed that MoDXR had its closest relationship with DXR from Coffea arabica and Gardenia jasminoides. The subcellular localization revealed that MoDXR protein was located on the chloroplast. Plantcare analysis indicated that the promoter region sequence of MoDXR was 1 493 bp, covering multiple light, stress, and hormone-responsive cis-regulatory elements; protein electrophoresis showed that the expressed protein was the anticipated size. This research lays the foundation for further purification and structural and functional characterization of the MoDXR protein.

茜草科的巴戟天(Morinda officinalis How)为我国四大南药之一, 收录于中国药典, 以干燥根入药, 其生物活性成分以蒽醌类、环烯醚萜类和寡糖类为主, 具有补肾阳, 强筋骨, 祛风湿的功效^[1-5]。除此之外, 也有文献显示巴戟天中的蒽醌类物质对肝癌细胞有一定的抑制作用^[6]。巴戟天的生物活性成分中的茜草素型蒽醌类有机物的合成需要莽草酸途径和产生活性异戊二烯的甲基赤藓醇磷酸途径(MEP)或甲羟戊酸途径(MVA)的相互结合^[7]。莽草酸途径主要是形成茜草素型蒽醌的A环和B环, 而关于C环的形成前人有研究认为以MEP途径为主^[8]。1-脱氧-D-木酮糖5-磷酸还原异构酶基因(DXR)作为MEP途径的限速酶, 能够使1-脱氧-D-木酮糖5-磷酸(DXP)异构化为MEP^{[9, 10]}。关于植物中DXR基因的研究从最早的拟南芥^[11]、玉米^[12]、番茄^[13]到药用植物香樟^[14]、杜仲^[15]、长春花^[16]、丹参^[17]、雷公藤^[18]等物种已多有文献报道。茜草科的栀子^[19]、小粒咖啡^[20]、茜草^[21]的DXR基因也在近几年陆续被研究。在烟草中, 利用叶绿体转基因技术, 过表达DXR基因, 可以显著增加细胞中类异戊二烯的含量, 如:叶绿素a、胡萝卜素、叶黄素、玉米黄质及β-谷甾醇等^[22]。利用CRISPR/Cas9基因编辑技术, 敲除烟草中的DXR基因后, 转基因烟草呈现白化表型, 这也证明了DXR基因参与到了烟草的叶绿素合成的过程中^[23]。

现阶段巴戟天的研究主要集中在化学成分、提纯工艺、药理及栽培种植等方面, 对于巴戟天重要活性成分的生物合成途径相关基因的研究鲜有报道。本研究根据巴戟天的DXR原始转录组数据, 设计MoDXR基因的特异扩增引物, 利用RT-PCR技术从巴戟天根部组织中成功扩增到了MoDXR基因及其启动子序列, 并对其进行了生物信息学分析、启动子区顺式作用元件分析和原核表达分析。这为下一阶段通过转基因方法大幅提高巴戟天中茜草素型蒽醌类的含量提供了理论的可能, 并且丰富巴戟天的分子生物理论方面的研究。

材料与方法

材料巴戟天植株栽植于福建农林大学园林学院温室, 实验材料为2年生巴戟天的幼苗。经福建农林大学林学院邹小兴教授鉴定为茜草科植物巴戟天Morinda officinalis How。

试剂总RNA提取试剂盒(DP441)和质粒提取试剂盒均购自天根生化科技有限公司, 反转录试剂盒(6210A)、无缝克隆(in-fusion HD cloning kit)、TB Green Premix Ex TaqII、Genome Walking Kit试剂盒购自TaKaRa公司, 胶回收试剂盒购自OMEGA公司, DNA polymerase分别购自TOYOBO公司的KOD-Plus-Neo酶和TaKaRa公司的primestar max DNA Polymerase (045A), FastDigest EcoRI限制性内切酶购自ThermoFisher Scientific公司, GV3101根癌农杆菌化学感受态细胞、DH5α克隆感受态细胞和BL21(DE3)表达感受态细胞均购自上海唯地生物有限公司, 1 kb plus DNA marker和卡那霉素均购自索莱宝公司, pCAMBIA2300-35s-eGFP双元表达载体和原核表达载体pET-28a由福建农林大学原料林研究所分子实验室保存, pEASY-T5 Zero Cloning Kit载体和Trans1-T1感受态均购自北京全式金生物有限公司。

总RNA、DNA的提取和反转录cDNA第一链合成 按照总RNA提取试剂盒说明书和反转录试剂盒说明书的操作步骤分别提取巴戟天根、茎、叶3个组织部位的总RNA, 并反转录合成相应的cDNA第一链, 后将原始浓度cDNA浓度调整至100 ng·μL^-1左右。采用CTAB法提取巴戟天的DNA, 并进行浓度测定和琼脂糖凝胶电泳检测。

MoDXR基因ORF区克隆由巴戟天转录组数据分析得到关于MoDXR基因的核心片段, 使用Primer Premier5软件分别设计5′-RACE和3′-RACE特异性扩增引物, 经PCR扩增的产物纯化后与pEASY-T5 Zero Cloning Kit载体连接后转化涂板, 菌液PCR验证后送福州铂尚测序公司测序, 测序后的序列使用DNAman软件进行拼接, 最终得到MoDXR基因的全长cDNA序列。使用NCBI-ORFfinder预测MoDXR基因的开放阅读框。根据预测的MoDXR基因的ORF区序列和原核表达载体PET-28a限制性酶切位点EcoR I序列设计MoDXR基因的一对特异性扩增引物(表 1), KOD酶反应体系(50 μL): ① KOD-Plus-Neo 1 μL, MgSO₄ 4 μL, dNTPs 5 μL, 10×PCR Buffer 5 μL, 引物各1.5 μL, cDNA模板2 μL, ddH₂O 30 μL。PCR扩增反应程序: 94 ℃ 2 min; 98 ℃ 10 s, 59 ℃ 30 s, 68 ℃ 90 s, 共30个循环; 4 ℃保存。② TaKaRa primeSTAR max反应体系(50 μL): primeSTAR max Premix 25 μL, 引物各2 μL, cDNA模板2 μL, ddH₂O 19 μL。PCR扩增反应程序: 98 ℃ 10 s, 55 ℃ 15 s, 72 ℃ 100 s, 共35个循环。扩增产物经1.0%琼脂糖凝胶电泳检验合格后, 用OMEGA的DNA胶回收试剂盒纯化PCR扩增产物。

MoDXR基因生物信息学分析 NCBI的ORFfinder分析预测基因的开放阅读框; 使用NCBI-BlastP、DNAMAN、NCBI-CDD和InterPro软件进行蛋白序列的同源比对和保守结构域分析; 利用MEGA7.0软件的Neighbor-Joining法对巴戟天和其他植物DXR蛋白进行聚类分析; 运用在线工具Rare Codon Caltor进行稀有密码子预测; 运用在线处理软件ExPASy-Protparam、ExPASy-ProtScale和NetPhos3.1server分析预测MoDXR基因的所编码的氨基酸序列组成、理化性质、亲疏水性及磷酸化修饰位点; 分别用在线工具TMHMMserver 2.0、SignalP分析和预测跨膜结构域及信号肽分析; 运用WoLF PSORT在线工具进行MoDXR蛋白的亚细胞定位分析; 运用ChloroP在线工具预测叶绿体转运肽存在与否; 分别用在线工具SOPMA和SWISS-MODEL预测MoDXR蛋白的二级和三级结构; 运用plantCARE在线分析工具进行基因启动子区顺式作用元件分析。

MoDXR亚细胞定位分析将MoDXR基因序列的终止密码子除去并进行克隆, 后将克隆片段与线性化质粒载体pCAMBIA2300-35s-eGFP连接。将pCAMBIA2300-35s-MoDXR-eGFP和pCAMBIA2300-35s-eGFP (对照)分别侵染烟草幼苗, 用ZEISS Observer A1倒置荧光显微镜分别在Fs 14HE (激发光: 510~560 nm)通道观察叶绿体荧光信号, 和用GFP (激发光: 470 nm)通道观察GFP荧光信号。

MoDXR基因启动子的分离依据MoDXR基因的CDS序列, 利用Primer primer5.0软件从ATG起始密码子下游600 bp内设计3条退火温度较高的特异性引物(SP1、SP2和SP3)进行巢式PCR反应(表 1)。SP3在SP2内侧, SP2在SP1内侧, 与试剂盒提供的4种退火温度较低的兼并性引物(AP1、AP2、AP3和AP4)进行3次热不对称PCR反应。取上述3次PCR反应液5 μL进行琼脂糖凝胶电泳检测, 切胶回收第3次PCR扩增产物, 将回收的PCR产物与pEASY-T5 Zero载体连接并测序。

MoDXR基因原核表达载体构建挑取已导入DH5α的pET-28a质粒载体单克隆, 37 ℃、220 r·min^-1摇菌, 经菌落PCR验证后抽提质粒。用FastDigest EcoRI限制性内切酶单酶切PET-28a环状质粒载体, 反应体系(20 μL):质粒17 μL (≤1 μg), FastDigest EcoRI 1 μL, 10× FastDigest Buffer 2 μL。酶切反应程序: 37 ℃ 1 h, 80 ℃ 5 min。酶切产物经1.0%琼脂糖凝胶电泳检验合格后, 用OMEGA的DNA胶回收试剂盒纯化酶切产物。将纯化的MoDXR基因扩增产物与线性化的载体pET-28a用in-fusion的方法重组连接并转化到DH5α克隆感受态细胞中, 涂板并挑单克隆。37 ℃、220 r·min^-1摇菌后, 经菌落PCR验证后, 送至铂尚生物技术公司测序, 最终将测序正确的pET-28a-MoDXR原核表达载体提取质粒后导入至BL21(DE3)表达感受态细胞中, 挑取部分单克隆加到含5 mL 2×YT (含kan)培养基中的50 mL离心管中, 在37 ℃、250 r·min^-1条件下, 大约3 h后将菌液吸光度(A)值控制在0.8左右。1 500 r·min^-1离心5 min, 倒掉上清, 重新加入含kan的5 mL 2×YT培养基, 再加入100 mmol·L^-1的IPTG, 使其终浓度为1 mmol·L^-1。在22 ℃、180 r·min^-1条件下分别诱导1~5 h后提取蛋白, 将处理的样品各取20 μL经SDS-PAGE电泳和考马斯亮兰染色, 检验MoDXR蛋白表达情况。

MoDXR基因表达量分析根据巴戟天转录组数据用primer primer5软件设计巴戟天内参基因和MoDXR-qRT-PCR的引物, 经过巴戟天内参基因筛选验证, 最终选择elongation factor 1-alpha (ef1α)作为巴戟天的内参基因, 其扩增效率为97.836%, 以及线性相关系数R²=0.995, 扩增片段大小为109 bp。MoDXR-qRT-PCR引物扩增片段大小131 bp (表 1)。两对引物熔解曲线为单峰, 引物无二聚体, 特异性良好。qRT-PCR扩增体系(20 μL): TB Green Premix Ex Taq 10 μL, 正反引物各0.8 μL, RoxII 0.4 μL, cDNA 2 μL (100 ng·μL^-1), ddH₂O 6 μL。扩增程序: 95 ℃ 30 s; 95 ℃ 5 s, 60 ℃ 34 s, 共40个循环; 熔解曲线: 95 ℃ 15 s, 60 ℃ 1 min, 95 ℃ 15 s。每个样品做3个平行实验, 并做3个生物学重复, 后用Ct(2^-ΔΔCt)法计算MoDXR基因在巴戟天根、茎、叶3个不同组织中的相对表达量。

1 巴戟天MoDXR基因克隆

按照SMARTer RACE 5′/3′kit试剂盒说明书操作步骤对MoDXR基因分别进行5′/3′-RACE扩增, 经测序和拼接后得到片段长度为2 015 bp的全长cDNA序列, 后经Blastp比对分析, 确认所获得的片段为DXR基因, NCBI的ORFfinder分析预测MoDXR基因的cDNA开放阅读框大小为1 425 bp, 编码424个氨基酸。以巴戟天的cDNA为模板, 对MoDXR基因的ORF区进行克隆, 经两种PCR聚合酶扩增和琼脂糖凝胶电泳检测, 条带出现在1 500 bp Marker左右, 大小与预期相符(图 1)。将两种方法纯化的克隆片段连接到pET-28a载体上, 挑取部分阳性单克隆测序, 最终验证MoDXR基因ORF序列与转录组数据相同。

2 巴戟天MoDXR生物信息学分析

根据巴戟天MoDXR基因的测序碱基序列用NCBI的ORFfinder分析预测MoDXR基因的cDNA开放阅读框大小为1 425 bp, 编码424个氨基酸(图 2)。

将巴戟天的MoDXR所预测的氨基酸序列用NCBI的BlastP进行同源序列比对, 结果(图 3)表明巴戟天的MoDXR基因编码的氨基酸序列与萝芙木(RvDXR)、咖啡树(CaDXR)、栀子(GjDXR)和桂花(OfDXR)的同源性都在90%以上, 其中与咖啡树的同源性高达91.81%, 而与桑树的同源性只有85.35%, 这说明MoDXR属于植物DXR蛋白家族。从不同物种的DXR氨基酸序列比对结果还可以看出DXR蛋白的C端相较于N端有高的保守性, DXR氨基酸序列同源性差异主要来自于N端, 而N端主要是大部分植物DXR蛋白的质体转运肽所在区域。由NCBI-CDD和InterPro保守结构域分析可知巴戟天的MoDXR蛋白含有3个结构域(图 3):第80~208位氨基酸残基组成的1-deoxy-D-xylulose 5-phosphate reductoisomerase, N-terminal结构域, 第222~305位氨基酸残基组成的1-deoxy-D-xylulose 5-phosphate reductoisomerase, C-terminal结构域, 第337~458位氨基酸残基组成的DXP reductoisomerase C-terminal domain。

利用MEGA7.0软件的Neighbor-Joining法对MoDXR氨基酸序列和其他具有较高同源性的DXR氨基酸序列的植物进行系统进化分析, 结果(图 4)显示, 巴戟天的DXR氨基酸序列与咖啡树和栀子聚为一类, 其亲缘关系最近, 都属于茜草科的植物, 而与葡萄、桑、可可、陆地棉、雷公藤、木薯和毛果杨的亲缘关系较远。

MoDXR基因氨基酸组成及其理化性质经由ExPASy-Protparam分析可得: MoDXR基因所编码的蛋白的分子质量为51.27 kDa, 理论等电点为5.93, 氨基酸组成中亮氨酸(Leu)、丙氨酸(Ala)、缬氨酸(Val)、异亮氨酸(Ile)的比例较高, 分别为10.3%、9.9%、8.0%和7.4%。带负电荷的氨基酸残基(Asp+Glu)占11.4% (54aa), 带正电荷的氨基酸残基(Arg+Lys+His)占12.3% (47aa)。其不稳定系数为34.97, 被划分为稳定的蛋白。总平均疏水系数(GRAVY)为0.026。具体到每个氨基酸残基的亲疏水性分析, 结合ProtScale在线工具的亲水性/疏水性的预测结果显示, MoDXR基因所编码的氨基酸序列的N端部分序列表现出亲水性, 而且整体的趋势是中心和靠近C端部分序列, 疏水性氨基酸的比例偏高, 其中疏水性和亲水性最强的位点分别是第174位和75位, 整体趋向疏水性。这为接下来的蛋白质折叠和蛋白质二级结构预测提供了参考。

经Rare Codon Caltor在线工具预测MoDXR基因所编码的氨基酸序列中稀有密码子的比例为8.6%, 选择BL21(DE3)表达感受态细胞用于MoDXR基因的原核表达分析。NetPhos3.1server分析MoDXR基因所编码的氨基酸序列发现, 有10个可信度高的磷酸化位点, 包含8个丝氨酸(Ser)、1个苏氨酸(Thr)和1个酪氨酸(Tyr)磷酸化位点。这10个可信度高的磷酸化位点都没有特定的修饰激酶, 这说明MoDXR基因所编码的氨基酸序列不仅有较多的磷酸化位点而且修饰类型丰富。

SignalP在线分析软件结果显示:预测MoDXR基因可能存在信号肽位点的C、S和Y值都小于0.2, 说明其不存在信号肽位点, 属于非分泌型蛋白。TMHMM跨膜分析软件结果显示MoDXR基因所编码的氨基酸序列全部在膜外, 没有跨膜结构域, 不属于跨膜蛋白。

WoLF PSORT在线工具亚细胞定位分析结果显示: MoDXR蛋白可能定位于叶绿体上, 而ChloroP在线工具预测MoDXR基因所编码的氨基酸序列中不存在叶绿体转运肽, 但仍预测前导肽的长度为66个氨基酸残基。

在线工具SOPMA对MoDXR蛋白的二级结构预测结果显示:该蛋白的二级结构中α螺旋(H)、β折叠(E)、β转角(T)和无规则卷曲(C)的数量分别为: 168 (35.44%)、95 (20.04%)、38 (8.02%)和173 (36.50%)。SWISS-MODEL预测的三级结构的图 5中显示, 该蛋白主要是有α螺旋和无规则卷曲组成。

3 巴戟天MoDXR基因启动子区顺式作用元件分析

依据MoDXR基因的CDS序列设计3条特异性引物, 采用染色体步移的方法克隆得到了MoDXR基因5′端上游启动子区的长度为1 493 bp (图 6)。由plantCARE在线分析软件分析结果可知(表 2), 在MoDXR基因上游启动子区中含有基本启动子元件CAAT-box和TATA-box; 多个光响应顺式作用元件, 如: AE-box、Box 4、Box Ⅱ、G-box、GA-motif和TCT-motif; 与低温胁迫相关的响应元件LTR; 参与防御和胁迫响应的顺式作用元件TC-rich repeats; 厌氧诱导顺式作用元件ARE; 还存在多种激素响应元件, 如:脱落酸响应元件ABRE、茉莉酸甲酯诱导顺式作用元件CGTCA-motif、生长素响应元件TGA-element; 此外, 还有与转录因子MYB、MYB-like、Myb和MYC结合位点有关顺式作用元件, 说明上述顺式作用元件能够响应外界环境的变化, 从而调控MoDXR基因的表达变化。

4 MoDXR亚细胞定位分析

将pCAMBIA2300-35s-eGFP (对照)和构建的融合表达载体pCAMBIA2300-35s-MoDXR-eGFP分别侵染烟草幼苗, 验证含eGFP荧光信号的MoDXR蛋白在细胞中的定位。在470 nm激发光下可以观察到GFP绿色荧光信号, 在510~560 nm激发光下(Fs 14 HE通道)可以观察到叶绿体自发荧光信号。由图 7的A、B和C可知pCAMBIA2300-35s-eGFP空载含有的eGFP绿色荧光蛋白能够在470 nm激发光下产生绿色荧光信号, 但是没有定位功能, 在细胞质和细胞核中都能够观测到绿色荧光信号。由图 7D可知在470 nm激发光下pCAMBIA2300-35s-MoDXR-eGFP融合表达载体能够检测到绿色荧光信号, 而在510~560 nm激发光下可以观察到叶绿体自发的暗红色的荧光信号(图 7E), 将图 7的D和E融合后, 融合表达载体绿色荧光信号和叶绿体自发的暗红色的荧光信号相一致, 表明MoDXR蛋白定位于叶绿体上。

5 巴戟天MoDXR蛋白原核表达分析

经SDS-PAGE电泳和考马斯亮兰染色, pET-28a-MoDXR重组蛋白在第1~5 h的表达情况显示(图 8左):与未经IPTG诱导的对照(C)相比, pET-28a-MoDXR重组蛋白分别在1~5 h内成功表达, 蛋白条带大小(含His标签)位于55~72 kDa之间, 与实际大小相符。在22 ℃、180 r·min^-1条件下, 菌液经IPTG诱导1 h后超声破碎, 用免疫磁珠的方法纯化pET-28a-MoDXR重组蛋白, 结果显示重组蛋白在沉淀中表达, 上清液中没有检测到MoDXR重组蛋白, 说明pET-28a-MoDXR在上述条件下主要在细胞质中以不溶性的包涵体的形式表达(图 8右)。

6 巴戟天MoDXR基因相对表达量分析

以ef1α作为巴戟天的内参基因, 验证MoDXR基因在巴戟天根、茎、叶3个不同组织中的相对表达量, 结果显示: MoDXR基因在巴戟天幼苗的根、茎、叶3个不同组织的表达量有所不同, 其中以叶的表达量最高, 茎的表达量只有叶的0.711倍, 而根中MoDXR基因的表达量最低, 仅占叶的47.1%。

本实验首次克隆了MoDXR基因的cDNA序列及其上游启动子区域, 并对启动子区域的顺式作用元件进行了分析。证实了MoDXR基因在巴戟天的根、茎和叶3个组织部位都有表达。此外, MoDXR基因上游启动子区含有与环境因素应答和激素诱导有关的顺式作用元件, 今后可以通过非生物胁迫的方式调节MoDXR基因的表达。

通过查阅相关文献发现, 大部分植物中DXR基因只有一个单拷贝, 但部分植物中存在两个拷贝, 如:橡胶树^[24]、大豆^[25]。这些多拷贝的DXR基因有的在不同的组织中有不同的调控方式, 也有的在不同的环境条件下表达出时空动态性。从本试验的研究中已确定了巴戟天中存在一个DXR基因, 但是具体的拷贝数未来需要通过Southern blot或全基因组图谱的方法进行确认。通过NCBI-CDD和InterPro保守结构域分析可知MoDXR蛋白含有3个结构域, 这些保守结构域与其他植物的DXR蛋白的结构域高度同源。分析大部分DXR基因编码的氨基酸序列, N端有如下特性:保守性较差但富含丝氨酸, 有保守切割位点的质体转运肽, 有一个保守的Cys-Ser-X基序, 其中X代表了任何疏水性氨基酸但以丙氨酸(Ala)、缬氨酸(Val)和甲硫氨酸(Met)为主。除此之外, 还有一个富含脯氨酸(Pro)的保守区域^{[11, 26]}。还有一点不同的是, MoDXR蛋白N端保守的Cys-Ser-X基序中, X代表了亲水性的苏氨酸(Thr), 这与大部分植物DXR蛋白N端保守的Cys-Ser-X基序X代表疏水性的氨基酸不同。

本实验中用pET-28a-MoDXR构建的原核表达载体, 经22 ℃、190 r·min^-1和1 mmol·L^-1 IPTG诱导的条件下, 在BL21(DE3)表达感受态细胞成功表达, 但是将细胞超声破碎后, 检测到重组蛋白是以不溶性的包涵体的形式存在于离心的沉淀中, 而在上清液中没有检测到, 这为以后的蛋白纯化带来了困难。张晓东构建的原核表达载体pGEX-4T-1-GrDXR, 37 ℃、1 mmol·L^-1 IPTG诱导的条件下, 在Rosetta (DE3)表达感受态细胞也成功表达除了滇龙胆的DXR蛋白, 但是没有验证GrDXR重组蛋白是不是在上清液中表达^[27]。在接下来的实验中需要选择合适的载体, 和表达感受态, 以及优化培养条件, 尽量使MoDXR蛋白在细胞周质以正确的折叠形式表达, 为进一步纯化MoDXR蛋白, 研究其结构和功能奠定基础。

DXR基因作为调控萜类化合物合成的限制性步骤, 主要合成单萜、二萜(叶绿素、脱落酸及质体醌等)、四萜(类胡萝卜素)等萜类化合物, 通过DXR基因过表达可以显著提高相应萜类化合物的含量, 而敲除DXR基因后的拟南芥突变体除了能够降低相应萜类化合物的量外, 其表型发生了一系列变化, 如:植物矮小, 白化, 腺毛数量减少及大部分气孔不能正常闭合^{[22, 28]}。巴戟天DXR基因在其3个组织部位都有表达, 且叶中的相对表达量大于根和茎, 其他植物如丹参^[29]和高良姜^[30]中DXR基因在叶中的相对表达量大于根和茎, 而在地黄^[31]中DXR基因在根中的相对表达量大于叶和茎, 巴戟天、丹参及地黄以干燥根入药, 高良姜以根茎中的挥发油为有效成分, 这说明药用植物中含有主要有效成分的组织部位中DXR基因表达量不一定高, DXR基因参与的只是萜类生物合成途径中的MEP途径, MVA途径(甲羟戊酸途径)作为另外一条萜类(倍半萜、三萜及甾体等)合成方式也参与其中, 而且叶和茎组织中叶绿素的含量远大于根部, 这会造成萜类物质的分流。
