In this study, eleven containing gallic acid moiety derivatives of Matijin-Su(MTS)were synthesized and evaluated for their anti-HBV activities in HepG2 2.2.15 cells in an effort to find novel effective anti-hepatitis B virus(HBV)agents. Compounds 5c, 6c and 6d exhibited significant anti-HBV activity with IC50 values of 6.12, 8.44 and 9.86 μmol·L-1, respectively. Incorporation of gallic acid moiety into MTS derivatives can lead to profound changes in their anti-HBV activity, and could be worth of further research.

本文以马蹄金素(Matijin-Su,MTS)为先导化合物进行结构优化,通过加入没食子酸结构单元,设计并合成了11个新的衍生物。对所合成的目标化合物以HepG2 2.2.15细胞进行体外抗HBV活性测试,结果表明:除4a4b外,其余目标化合物均有一定的体外抗HBV活性,其中5c6c6d显示较强的抗HBV活性,其IC50达到了6.12、8.44、9.86 μmol·L-1,说明含有没食子酸结构的马蹄金素衍生物具有进一步研究的价值。

本文以马蹄金素(Matijin-Su,MTS)为先导化合物进行结构优化,通过加入没食子酸结构单元,设计并合成了11个新的衍生物。对所合成的目标化合物以HepG2 2.2.15细胞进行体外抗HBV活性测试,结果表明:除4a4b外,其余目标化合物均有一定的体外抗HBV活性,其中5c6c6d显示较强的抗HBV活性,其IC50达到了6.12、8.44、9.86 μmol·L-1,说明含有没食子酸结构的马蹄金素衍生物具有进一步研究的价值。

In this study, eleven containing gallic acid moiety derivatives of Matijin-Su(MTS)were synthesized and evaluated for their anti-HBV activities in HepG2 2.2.15 cells in an effort to find novel effective anti-hepatitis B virus(HBV)agents. Compounds 5c, 6c and 6d exhibited significant anti-HBV activity with IC50 values of 6.12, 8.44 and 9.86 μmol·L-1, respectively. Incorporation of gallic acid moiety into MTS derivatives can lead to profound changes in their anti-HBV activity, and could be worth of further research.

乙肝病毒(hepatitis B virus, HBV)的感染可引起急、慢性的乙型肝炎, 世界卫生组织2016年6月公布的数据显示全球约有2.4亿人感染慢性乙型肝炎(简称乙肝, 乙肝表面抗原阳性持续至少6个月)[1], 乙肝患者还可能会逐渐发展为肝硬化、肝癌等[2, 3]
从免疫学角度看, 乙肝病毒携带者比非乙肝病毒携带者的肝癌发生率高100倍[4]。临床上用药治疗乙肝的药物主要有α-干扰素类药物和核苷类药物[5], 到目前为止, 还没有药物能够彻底清除体内HBV并最终治愈乙肝[6], 因此, 寻找和开发具有独特化学结构的创新药物显得尤为重要。本课题组前期从治疗肝炎的民族药马蹄金中发现了具有抗HBV活性的马蹄金素(Matijin-Su, MTS), 并以其为先导化合物设计合成了一系列衍生物。其中对1个优选的MTS衍生物(研发代号Y101, 结构见图 1)进行了深入系统的临床前研究, 获得了国家食品药品监督管理总局颁发的临床批件(原料药及其制剂批件号: 2013L02491, 2014L00070)[7, 8]
没食子酸酚羟基经过甲基化后的衍生物在药物中得到了广泛应用, 如治疗心率失常药物[9]、抗疲劳药物[10]等, 甲氧基都发挥了很重要的作用。很多药物结构中甲氧基也广泛存在[11-13], 如甲氧基化的黄酮在抗癌、抗炎活性上表现非常好, 且甲氧基的存在提高了化合物的脂溶性, 增加机体的药物吸收[14]。在前期研究基础上[15, 16], 本文以马蹄金素为母体, 设计合成了B环为3个羟基被甲基化的没食子酸马蹄金素衍生物, 并对其进行了体外抗HBV活性测试, 希望在拓宽马蹄金素结构修饰产物类型的同时, 获得更多具有深入研究价值的目标分子。
选择L-苯丙氨酸甲酯盐酸盐或L-酪氨酸甲酯盐酸盐为原料, 首先将其与3, 4, 5-三甲氧基苯甲酸进行缩合反应得到化合物1 (1a1b), 由化合物1水解得羧酸2 (2a2b), 再将所得羧酸2分别与L-苯丙氨酸甲酯盐酸盐、L-酪氨酸甲酯盐酸盐或L-苯丙氨醇进行缩合反应, 即得目标化合物3 (3a3b)和4 (4a4b), 最后通过对甲酯的水解和羟基的亲核取代, 即可得目标化合物5 (5a ~5c)和6 (6a ~6d)。见合成路线图 1
根据前期研究中积累的马蹄金素衍生物在抗HBV活性方面的构效关系数据, 选取不同的基团在R1、R2、R3三个位点进行取代, 或者通过化学方法换成不同的基团, 共合成目标化合物11个。其理化性质见表 1
衍生物体外抗HBV活性评价采用四氮唑盐(MTT)法测定其对细胞的毒性, 选择HepG2 2.2.15细胞为DNA复制的抑制率模型, 以拉米夫定为阳性对照药, 对所合成的11个化合物进行体外抗HBV活性评价。
活性测试结果(表 2)表明:马蹄金素的衍生物在测试浓度范围内除4a4b外均有抗HBV活性, 其中化合物5c6c6d表现出良好的抗HBV活性, IC50优于阳性对照药物拉米夫定, 并且其选择指数也较高。从结构来看, R3为COOH时化合物活性较好, 其中化合物5c (IC50: 6.12 μmol·L-1, SI = 73.05) 呈现出较好的抗HBV活性, 说明含有没食子酸结构的马蹄金素衍生物具有进一步研究的价值。
本文以L-苯丙氨酸甲酯盐酸盐、L-酪氨酸甲酯盐酸盐、L-苯丙氨醇、3, 4, 5-三甲氧基苯甲酸等为主要原料, 酰化、烷基化、水解、以及用IBCF和NMM等作为缩合剂形成酰胺(肽)键等反应合成了目标化合物11个, 用核磁共振、质谱对其进行了结构确证, 并以HepG2 2.2.15细胞为HBV载体, 对合成的11个衍生物进行了抗HBV活性研究, 活性筛选结果表明所合成的化合物对HBV DNA复制有一定的抑制作用。在测试浓度范围内, 除化合物4a4b无活性外, 5c的活性最好, 且抗HBV活性优于阳性对照药拉米夫定(IC50: 18.90 μmol·L-1, SI: 41.59), 值得进一步研究开发。
熔点用XT-4型熔点仪测定, 温度未校正; 核磁共振谱以TMS为内标, 用Inova-400 MHz型超导核磁共振仪(美国Varian公司)测定; 质谱用HP-5793质谱仪或HP1100 LC-MS液质联用仪测定(美国Hewlett-Packard公司); 柱色谱用硅胶(300~400目)及高效薄层板均为青岛海洋化工厂产品; 其余试剂均为市售分析纯或化学纯产品, 除特别说明外, 未经处理直接使用。
L-苯丙氨酸甲酯盐酸盐25.88 g (120 mmol)与3, 4, 5-三甲氧基苯甲酸21.22 g (100 mmol)混合置于反应瓶内, 于70 ℃真空干燥1 h, 冷却至室温后溶解于500 mL干燥的CH2Cl2 (二氯甲烷)和50 mL DMF (N, N-二甲基甲酰胺)中, 再加入NMM (N-甲基吗啡啉) 24.71 mL (225 mmol), 氩气保护, 冰水浴冷却30 min后开始滴加IBCF (氯甲酸异丁酯) 12.65 mL (100 mmol)和80 mL CH2Cl2混合液, 控制其流速使其在50 min内滴加完毕, 体系自然升温至室温, 反应8 h。TLC (薄层色谱) (氯仿/甲醇= 30:1, Rf = 0.5, 磷钼酸显色)检测反应完毕; 减压回收CH2Cl2, 加入乙酸乙酯和水, 萃取, 所得有机层依次以稀盐酸、饱和碳酸氢钠溶液及饱和氯化钠溶液洗涤, 无水硫酸钠干燥, 减压回收乙酸乙酯至干得中间体1a。将以L-酪氨酸甲酯盐酸盐替换起始原料L-苯丙氨酸甲酯盐酸盐, 按照以上合成方法可制备得中间体1b
2a为例:将中间体1a溶解于50 mL DMF中, 加入100 mL氢氧化钠溶液(1.0 mol·L-1), 室温搅拌反应2.5 h。TLC (氯仿/甲醇= 10:1, Rf = 0.2, 磷钼酸显色)检测反应完毕; 浓盐酸调pH 2~3, 乙酸乙酯萃取, 依次以蒸馏水、饱和氯化钠溶液洗涤, 无水硫酸钠干燥, 过滤, 减压回收乙酸乙酯至干得粗品2a
3a为例:将上述合成的中间体2a (其合成见1.1.2) 35.94 g (100 mmol)与L-酪氨酸甲酯盐酸盐27.80 g (120 mmol)混合置于反应瓶内, 于70 ℃真空干燥1 h, 自然冷却后溶解于350 mL干燥的CH2Cl2和80 mL DMF中, 加入NMM 24.71 mL (225 mmol), 取IBCF (氯甲酸异丁酯) 12.65 mL (100 mmol)与50 mL CH2Cl2混合置于滴液漏斗中, 氩气保护, 冰水浴(0 ℃)冷却30 min后开始滴加, 控制其流速使其在50 min内滴加完毕, 反应8 h。TLC (氯仿/甲醇= 30:1, Rf = 0.4, 磷钼酸显色)检测反应完毕; 加水少许终止反应, 减压回收部分CH2Cl2, 乙酸乙酯萃取, 依次用蒸馏水、稀盐酸、饱和碳酸氢钠溶液及饱和氯化钠溶液洗涤, 再用无水硫酸钠干燥, 过滤, 减压回收部分乙酸乙酯, 析出白色固体经纯化后得白色结晶3a 16.65 g (收率: 31.03%)。3b4a4b用类似方法合成。
3b 0.43 g (0.81 mmol, 其合成见1.1.3)、碳酸钾0.67 g (4.9 mmol)与二甲氨基氯乙烷盐酸盐0.087 g (0.81 mmol)置于反应瓶内, 加入1, 4-二氧六环(20 mL)与水(0.3 mL), 于内温86 ℃反应3.5 h。TLC (氯仿/甲醇= 10:1, Rf = 0.5, 磷钼酸显色)检测反应完毕; 乙酸乙酯萃取, 水洗使其呈中性, 1.0 mol·L-1盐酸溶液反萃取, 再以水洗有机层, 合并酸水层。酸水层用1.0 mol·L-1氢氧化钠溶液调pH 9~10, 析出白色固体, 乙酸乙酯萃取, 无水硫酸钠干燥, 过滤, 减压蒸去乙酸乙酯, 纯化后得白色结晶5a 0.16 g (收率: 32.50%)。
取化合物3b 0.42 g (0.81 mmol, 其合成见1.1.3) 与碳酸钾1.12 g (8.1 mmol)置于反应瓶内, 加入DMF (20 mL)溶解, 再加入碘代正丁烷0.1 mL (0.88 mmol), 35 ℃水浴搅拌8 h。TLC (氯仿, Rf = 0.5, 磷钼酸显色)检测反应完毕; 乙酸乙酯萃取, 依次以水、饱和氯化钠洗涤, 无水硫酸钠干燥, 过滤, 减压蒸去部分乙酸乙酯, 析出白色固体以氯仿/甲醇重结晶即得白色结晶5b 0.23 g (收率: 47.91%)。
3b (1 mmol)置于反应瓶内, 加入15 mL DMF溶解, 再加入2 mL 1.0 mol·L-1氢氧化钠溶液, 常温反应3 h。TLC (氯仿/甲醇=10:1, Rf = 0.2, 磷钼酸显色)检测反应完毕; 1.0 mol·L-1盐酸调pH 2~3, 析出白色固体, 过滤、烘干后得白色结晶5c 0.47 g (收率: 89.94%)。
以化合物6a为例:取化合物4a 0.41 g (0.81 mmol, 其合成见1.1.3) 与碳酸钾1.12 g (8.1 mmol)置于反应瓶内, 加入DMF (20 mL)溶解, 再加入碘代正丁烷0.1 mL (0.88 mmol), 35 ℃水浴搅拌8 h。TLC (氯仿/甲醇= 30:1, Rf = 0.6, 磷钼酸显色)检测反应完毕; 乙酸乙酯萃取, 依次以水、饱和氯化钠洗涤, 无水硫酸钠干燥, 过滤, 减压蒸去部分乙酸乙酯, 析出白色固体以氯仿/甲醇重结晶即得白色结晶6a 0.28 g (收率: 61.22%)。6b以类似方法合成。
将化合物4a 0.24 g (0.47 mmol, 其合成见1.1.3) 置于反应瓶内, 加入2.5 mL吡啶溶解, 缓慢滴加乙酸酐0. 4 mL (4.23 mmol), 室温继续搅拌反应8 h。TLC (氯仿/甲醇=30:1, Rf = 0.5, 磷钼酸显色)检测反应完毕; 将反应物倒入水中, 1.0 mol·L-1盐酸调pH 2~3, 乙酸乙酯萃取, 依次用蒸馏水、饱和氯化钠溶液洗涤, 以无水硫酸钠干燥, 过滤, 减压回收乙酸乙酯至干得白色结晶6c 0.24 g (收率: 86.16%)。6d以类似方法合成。
以HepG2 2.2.15细胞传到3到4代时培养瓶长满细胞后, 经0.25%胰酶溶液消化后用10% DMEM配制成每毫升约25万个的细胞悬液, 接种到96孔培养板内, 每孔100 μL, 摇匀后置37 ℃、5% CO2培养箱中培养24 h, 细胞贴壁后即可给药(配药:称取10 mg样品, 加1 mL DMSO溶解, 溶液经无菌微孔滤膜滤过即得10 mg·mL-1储存液, 取相应量储存液用2% DMEM稀释至所需给药浓度), 每孔给200 μL药液, 每个浓度设6个孔, 以2% DMEM代替药液作为阴性细胞对照组, 以拉米夫定药液代替药液作为阳性对照组。置于37 ℃、5% CO2培养箱培养3天, 每孔加入20 μL MTT溶液(5 mg·mL-1), 37 ℃、5% CO2继续培养6 h, 1 000 r·min-1离心10 min, 吸去孔内培养液, 每孔加入200 μL DMSO, 振荡10 min, 使形成的结晶充分溶解后通过酶联免疫检测仪测定其吸光度值(OD值), 采用改良寇氏法计算药物的半数有毒浓度(TC50)。改良寇氏法计算TC50, TC50 = Log-1 [Xm-i (ΣP-0.5)]。
以HepG2 2.2.15细胞传到3到4代时的细胞, 经0.25%胰酶溶液消化后用10% DMEM配制成每毫升约25万个的细胞悬液, 每孔500 μL接种到24孔细胞培养板, 每孔给700 μL药液, 共3个浓度, 每个浓度设3个孔, 以10% DMEM代替药液作为阴性细胞对照组, 以拉米夫定药液代替药液作为阳性对照组。置于37 ℃、5% CO2培养箱培养, 3天换原浓度药液或10% DMEM培养。培养6天后即可提取HBV DNA, 经荧光定量PCR测定其DNA含量, 通过含量测定结果计算样品对细胞中HBV DNA的抑制率, 采用改良寇氏法计算样品的半数抑制浓度(IC50)。分别计算得到的半数有毒浓度(TC50)和半数抑制浓度(IC50)即可计算出样品的选择指数(SI)[17]。改良寇氏法计算IC50, IC50 = Log-1[Xm-i (ΣP-0.5)], SI = TC50/IC50
