The study was designed to test the role of 5, 2', 4'-trihydroxy-6, 7, 5'-trimethoxy flavone nanoparticle (TTF1-NP) on lipopoiysaccharide (LPS)-induced inflammatory response, and to explore the anti-inflammatory mechanism in human hepatocellular carcinoma cells. Inflammatory responses were induced in human hepato-cellular carcinoma HepG2 cells with LPS; Proliferation effect of TTF1-NP in LPS-stimulated HepG2 cells were detected by MTT assay; The expression of TLR4, AKT/mTOR signaling related proteins and IL-6 were detected by Western blot assay. The results showed that TTF1-NP inhibited the proliferation of HepG2 cells induced by LPS in a dose-dependent manner; TTF1-NP inhibited the expression of TLR4, the activation of AKT and mTOR, and expression of IL-6 in a dose-dependent manner; TTF1-NP inhibited the activation of AKT/mTOR signaling pathway and TLR4 proteins leading to suppression of IL-6 expression in HepG2 cells stimulated by insulin. These results suggest that TTF1-NP inhibited inflammatory responses from LPS treatment with a potential mechanisms in the inhibition of AKT/mTOR pathway.

探讨珍珠梅黄酮纳米粒(5,2',4'-trihydroxy-6,7,5'-trimethoxy flavone nanoparticle,TTF1-NP)抑制脂多糖(lipopoiysaccharide,LPS)诱发人肝癌HepG2细胞的炎性作用及调控机制。采用LPS诱发人肝癌HepG2细胞的炎症模型;MTT比色法检测TTF1-NP对人肝癌HepG2细胞增殖作用;Western blot法检测TTF1-NP对LPS诱发的HepG2细胞炎性反应的TLR4和IL-6及AKT/mTOR信号通路关键蛋白的表达作用。结果显示,TTF1-NP能够抑制LPS引起的人肝癌HepG2细胞增殖,抑制TLR4蛋白表达及AKT和mTOR蛋白的磷酸化,抑制炎症因子IL-6和IL-8表达;TTF1-NP抑制胰岛素引起的AKT/mTOR信号通路的活化和TLR4蛋白的表达,从而降低IL-6和IL-8蛋白水平。研究表明,TTF1-NP抑制LPS诱导的人肝癌HepG2细胞的炎性反应,其作用机制可能与AKT/mTOR信号通路有关。

探讨珍珠梅黄酮纳米粒(5,2',4'-trihydroxy-6,7,5'-trimethoxy flavone nanoparticle,TTF1-NP)抑制脂多糖(lipopoiysaccharide,LPS)诱发人肝癌HepG2细胞的炎性作用及调控机制。采用LPS诱发人肝癌HepG2细胞的炎症模型;MTT比色法检测TTF1-NP对人肝癌HepG2细胞增殖作用;Western blot法检测TTF1-NP对LPS诱发的HepG2细胞炎性反应的TLR4和IL-6及AKT/mTOR信号通路关键蛋白的表达作用。结果显示,TTF1-NP能够抑制LPS引起的人肝癌HepG2细胞增殖,抑制TLR4蛋白表达及AKT和mTOR蛋白的磷酸化,抑制炎症因子IL-6和IL-8表达;TTF1-NP抑制胰岛素引起的AKT/mTOR信号通路的活化和TLR4蛋白的表达,从而降低IL-6和IL-8蛋白水平。研究表明,TTF1-NP抑制LPS诱导的人肝癌HepG2细胞的炎性反应,其作用机制可能与AKT/mTOR信号通路有关。

The study was designed to test the role of 5, 2', 4'-trihydroxy-6, 7, 5'-trimethoxy flavone nanoparticle (TTF1-NP) on lipopoiysaccharide (LPS)-induced inflammatory response, and to explore the anti-inflammatory mechanism in human hepatocellular carcinoma cells. Inflammatory responses were induced in human hepato-cellular carcinoma HepG2 cells with LPS; Proliferation effect of TTF1-NP in LPS-stimulated HepG2 cells were detected by MTT assay; The expression of TLR4, AKT/mTOR signaling related proteins and IL-6 were detected by Western blot assay. The results showed that TTF1-NP inhibited the proliferation of HepG2 cells induced by LPS in a dose-dependent manner; TTF1-NP inhibited the expression of TLR4, the activation of AKT and mTOR, and expression of IL-6 in a dose-dependent manner; TTF1-NP inhibited the activation of AKT/mTOR signaling pathway and TLR4 proteins leading to suppression of IL-6 expression in HepG2 cells stimulated by insulin. These results suggest that TTF1-NP inhibited inflammatory responses from LPS treatment with a potential mechanisms in the inhibition of AKT/mTOR pathway.

我国是原发性肝癌高发大国, 每年约有47万人死于原发性肝癌, 占全球死亡人数的1/2[1], 严重威胁人类的健康。应用化疗药物是肝癌患者重要的治疗手段之一, 但大部分的化疗药物毒副作用较大[2, 3], 因此从天然植物中寻找高效低毒的分子靶向治疗药物, 已经成为现阶段抗肝癌药物研究的重要方向。5, 2', 4'-三羟基-6, 7, 5'-三甲氧基黄酮纳米粒(5, 2', 4'-trihydroxy-6, 7, 5'-trimethoxy flavone nanoparticle, TTF1-NP)是以体内可生物降解材料硬脂酸为载体, 制备的生物体可降级吸收的抗肿瘤单体化合物[4]。课题组前期研究发现, TTF1-NP具有抗肿瘤生物学活性, 其作用机制与诱导细胞凋亡和抑制肿瘤血管生成有关[5, 6], 此外, TTF1-NP对二乙基亚硝胺诱发大鼠原发性肝癌形成过程中的炎症因子也有调节作用[7]
炎症反应是生物体常见的病理生理现象, 慢性炎症反应能够促进肿瘤的形成, 与肿瘤的恶性程度和预后有关。炎症反应在肝癌的形成和发展过程中也起着关键的作用[8], 一方面长期的慢性肝炎能够促进原发性肝癌的形成, 另一方面肝病患者血清中的脂多糖(lipopoiysaccharide, LPS)浓度较高[9], LPS能够通过与细胞表面特异性受体Toll样受体4 (TLR4) 结合, 活化TLR4介导的肿瘤促增殖信号转导通路, 促进肝癌细胞增殖[10], 加快肿瘤的恶性化进程。蛋白激酶B/哺乳动物雷帕霉素靶蛋白(protein kinase B/the mammalian target of Rapamycin, AKT/mTOR)信号通路是调控肿瘤细胞增殖和细胞凋亡的经典通路[11, 12], 近几年研究发现, 该通路不仅参与调控肿瘤细胞生长, 还能够通过与转录因子的相互作用调节肿瘤细胞炎性微环境, 影响肿瘤-炎症反应过程[13, 14]
本研究利用LPS刺激建立人肝癌HepG2细胞炎性反应模型, 检测TTF1-NP的抗炎作用, 明确其分子调控机制。为原发性肝癌患者分子靶向药物的研究提供新的探索方向, 也为TTF1-NP的开发应用提供理论依据。
实验细胞  人肝癌HepG2细胞购于南京凯基生物制品有限公司。
实验药品与试剂  TTF1-NP由延边大学药学院关丽萍教授惠赠。SDS-PAGE凝胶制备试剂盒购自北京索莱宝试剂公司; ECL发光试剂盒购自Merck Millipore公司; 小牛血清和青链霉素购自美国Gibco公司; DMEM培养液和胰酶均购自Invitrogen公司; IL-6、IL-8、TLR4、AKT、p-AKT、mTOR和p-mTOR抗体均购自英国Abcom公司; β-Actin购自北京博奥森生物技术有限公司; 羊抗兔lgG、羊抗小鼠lgG购自中杉金桥; MTT粉购自美国Sigma公司; pg-LPS购自法国InvivoGen公司; 胰岛素(insulin)溶液购于碧云天生物技术有限公司。
实验仪器  电泳仪(DYY-7C)、转膜仪(DYCZ-40D) (北京市六一仪器厂产品); UVP凝胶成像仪(Biospectrum, 美国UVP凝胶成像系统有限公司); 超净工作台(C1109C)、CO2培养箱(COR-1150) (上海智城分析仪器制造有限公司); 光镜(日本Olypus公司); 酶标仪(日本TECAN公司)。
细胞培养及分组  人肝癌HepG2细胞培养于DMEM培养液中(含10%胎牛血清、100 u∙mL-1青霉素及0.1 g∙L-1链霉素), 培养条件为37 ℃、5% CO2、湿度95%, 细胞生长对数期进行传代、备用。细胞分为未用任何药物干预的空白对照组(NT)、LPS (10 g∙L-1)处理组(LPS), 不同浓度的TTF1-NP (5、10和20 μmol∙L-1)加药组(LPS+TTF1-NP)、AKT/mTOR信号通路激活剂insulin (1 g∙L-1)组。
MTT比色法检测细胞增殖  对数生长期的人肝癌HepG2细胞, 取适量胰酶进行消化, 制成细胞悬液, 以细胞数1×104个/孔接种于96孔板中, 每孔细胞悬液200 µL。待细胞贴壁后, 弃去原孔内的培养液, 分别加入不同剂量的LPS或TTF1-NP分别培养12、24、48和72 h, 每孔内分别加入MTT溶液(5 g∙L-1) 20 μL, 继续培养4 h, 弃去孔内溶液, 每孔内加入二甲基亚砜(DMSO) 180 μL, 避光移至酶标仪内, 充分震荡10 min后, 测定490 nm处各孔内吸光度OD值, 计算细胞生长抑制率, 细胞生长抑制率(%) = (1 -OD样本 / OD阴性对照组) × 100%。
Western blot检测  提取各实验组全蛋白。根据BSA法测定各个实验组蛋白样品浓度。制备凝胶, 每孔依次上样, 电泳(80 V, 60~90 min), 转膜(100 V, 30~60 min)。取出PVDF膜浸泡在5%脱脂奶粉中, 室温封闭2 h。分别加入抗体(IL-6、IL-8、TLR4、AKT、p-AKT、mTOR、p-mTOR和β-actin) 4 ℃过夜, TBST清洗PVDF膜, 滴加二抗, 室温2 h, TBST清洗, ECL显影液孵育5 min, 显影并拍照。应用Image J软件对所得条带进行灰度分析。
数据处理  所有数据均采用$\overline{x}±s$表示, 采用单因素方差分析统计, P < 0.05有显著性统计意义, P < 0.01有极显著性统计意义。
MTT检测结果显示, 加入不同浓度LPS (0.1、1和10 g∙L-1)刺激人肝癌HepG2细胞后, 细胞相对活力上升, 在0.1~10 g∙L-1 LPS作用范围内表现出浓度依赖性(图 1A), 100 g∙L-1 LPS可促进HepG2细胞生长, 但作用不明显。在12、24和48 h内不同浓度的LPS刺激后HepG2细胞相对活力明显升高(图 1A), 在12~48 h内LPS促进人肝癌HepG2细胞增殖, 具有时间依赖性。由于10 g∙L-1 LPS作用48 h对HepG2细胞的促增殖作用最明显, 因此以10 g∙L-1 LPS刺激48 h为实验用模型诱导剂量。
不同浓度TTF1-NP (5、10和20 μmol∙L-1)对LPS (10 g∙L-1)刺激后人肝癌HepG2生长有抑制作用, 其生长抑制率随TTF1-NP浓度升高逐渐上升, 有剂量依赖性(图 1B)。
Western blot检测发现, TTF1-NP对HepG2细胞炎性受体蛋白TLR4和炎症因子IL-6表达有抑制作用(图 2A), 但与NT组比较差异不显著; TTF1-NP对LPS刺激后TLR4和IL-6表达有显著抑制作用(图 2A), 与LPS组比较有统计学意义(图 2B); TTF1-NP明显抑制LPS刺激后HepG2炎性受体蛋白TLR4和炎症因子IL-6表达(图 2C), 表现为剂量依赖性。与LPS组比较, 差异显著, 有统计学意义(P < 0.05或P < 0.01) (图 2D)。
Western blot检测发现, LPS (10 g∙L-1)可促进p-AKT和p-mTOR蛋白的表达(图 3A), 与NT组比较, 有统计学意义(P < 0.01) (图 3B)。不同浓度的TTF1-NP可以抑制LPS刺激后的人肝癌HepG2细胞p-AKT和p-mTOR蛋白表达(图 3A), 有剂量依赖性。
与LPS组比较, 有统计学意义(P < 0.05或P < 0.01) (图 3B)。
Western blot检测发现, TTF1-NP显著抑制了insulin活化的p-AKT和p-mTOR蛋白表达(图 4A), 与insulin组比较, 具有显著性差异(P < 0.05) (图 4B); Insulin对炎性受体蛋白TLR4表达也有促进作用, TTF1-NP抑制以上作用(图 4C), 与insulin组比较, 具有显著性差异(P < 0.01) (图 4D); TTF1-NP能够显著抑制insulin引起的炎症因子IL-6和IL-8的表达(图 4E), 与insulin组比较, 具有显著性差异(P < 0.05) (图 4F)。
原发性肝癌是严重威胁人类生命健康的恶性肿瘤之一, 研发高效低毒的靶向分子治疗药物具有重要现实意义。慢性炎症参与肝癌的发生和发展过程, 本课题组[15]也在前期研究中发现, 间歇饲喂DEN诱发大鼠原发性肝癌形成过程中, 有大量炎性细胞浸润, 低氧诱导因子和炎症因子IL-10活化表达。炎症因子能够直接作用于免疫调节细胞, 影响免疫细胞正常功能, 又能够调节细胞内信号因子, 影响细胞内信号通路, 从而促进肝癌的恶性进展。因此, 通过抑制炎性反应抑制原发性肝癌的发展是新的研究方向。
LPS刺激是诱发体外炎症模型最常用的方法之一[16], 其诱导发生的炎症反应与TLRs家族调控有关。TLR4是TRLs家族重要成员, 也是与肿瘤-炎症反应关系密切的最主要调节因子, 广泛存在于多种细胞表面。有学者[17]报道, LPS通过识别特异性受体TLR4, 形成受体复合物, 触发细胞内信号级联反应, 调控细胞内信号转录因子, 最终诱导IL-6、IL-8和TNF-α等炎症因子释放[18-21]。本研究检测发现类似结果, TTF1-NP抑制LPS的促人肝癌HepG2细胞增殖作用, 抑制TLR4表达, 并能抑制炎性因子IL-6表达。
TLR4是LPS诱发炎症反应的重要识别受体, 其介导的炎症反应与AKT/mTOR信号通路密切相关。AKT/mTOR信号通路是调控肿瘤细胞增殖、生长和代谢的重要转导途径[22], 近几年研究发现, 该信号通路参与调节肿瘤细胞的炎性反应[23]。Sun等[24]研究发现, LPS促进胰腺癌细胞TLR4信号转导的活化, 促进AKT的磷酸化, 利用siRNA技术沉默TLR4和PI3K/AKT抑制剂LY294002均可抑制LPS诱发的肿瘤细胞的增殖和血管生成等生物学反应。本研究利用Western blot技术检测AKT/mTOR信号通路关键分子AKT、mTOR、p-AKT和p-mTOR蛋白的表达变化发现, LPS促进p-AKT和p-mTOR蛋白表达, TTF1-NP可逆转以上作用。因此推测, TTF1-NP通过抑制AKT/mTOR信号通路活化, 影响炎症因子释放。
为进一步研究TTF1-NP通过AKT/mTOR通路对炎性因子的调节作用, 本研究加入AKT/mTOR信号通路活化剂insulin。Insulin是生物体内重要的糖激素蛋白, 能够与细胞膜上特异受体结合诱发PI3K变构、激活PI3K, PI3K在功能上是AKT活化的首要调控者[25]。本研究发现, insulin促进AKT/mTOR信号通路蛋白表达, 促进TLR4蛋白表达和炎症因子释放, TTF1-NP抑制insulin活化的AKT/mTOR信号通路, 抑制肝癌细胞的炎性因子表达。
炎症反应在肿瘤的恶性进展中扮演重要角色, 参与了恶性肿瘤发生、发展、侵袭和转移等病理过程, 抑制炎症反应成为治疗肿瘤的新策略。本研究结果显示, TTF1-NP能够抑制LPS诱发人肝癌HepG2细胞的炎性反应, AKT/mTOR信号通路发挥了重要作用。今后将进一步结合体内实验深入研究TTF1-NP的抗炎作用机制, 为TTF1-NP的开发利用提供可靠的实验依据, 为抗肿瘤药物的研究提供新的方向。
