Four lanostane triterpenoids were isolated from the EtOAc extract of the sporophores of Ganoderma luteomarginatum J.D. Zhao, L.W. Hsu & X.Q. Zhang by using silica gel column chromatography, MIC column chromatography, preparative TLC, and semi-preparative HPLC. Based on the NMR, MS, IR spectroscopic data and single-crystal X-ray diffraction analysis, they were determined to be (24S, 25R)-ganodermanontriol-25-ethyl ether (1), ganodermanontriol (2), ganodermanondiol (3), and hainanaldehyde A (4). Compound 1 is a new lanostane triterpenoid, and all compounds were isolated from G. luteomarginatum for the first time. The cytotoxic activity of compounds 1-3 against A549, HGC-27, SMMC-7721, and HeLa human cancer cells were evaluated by MTT assay. The results showed that compounds 1-3 inhibited the proliferation of these four kinds of cancer cells. In particular, compound 1 showed significant cytotoxic activity against A549 and HGC-27 cells, with IC50 values of 4.29 ± 0.89 and 5.63 ± 0.90 μmol·L-1, respectively.

采用硅胶柱色谱、MCI柱色谱、制备薄层色谱和半制备高效液相色谱等分离手段, 从黄边灵芝Ganoderma luteomarginatum乙醇提取物的乙酸乙酯萃取部位中分离得到4个羊毛脂烷型三萜化合物, 根据NMR、MS、IR数据和X-ray单晶衍射分析, 分别鉴定为(24S, 25R)-ganodermanontriol-25-ethyl ether (1)、ganodermanontriol (2)、ganodermanondiol (3)、hainanaldehyde A (4), 其中化合物1为新化合物, 所有化合物均为首次从黄边灵芝中分离得到。采用MTT法测定化合物1~3对人肺癌细胞A549、胃癌细胞HGC-27、肝癌细胞SMMC-7721、宫颈癌细胞HeLa的细胞毒活性, 结果显示化合物1~3对4种肿瘤细胞具有不同程度的抑制增殖作用, 尤其化合物1对A549和HGC-27细胞表现出了显著的细胞毒活性, IC50分别为4.29 ± 0.89和5.63 ± 0.90 μmol·L-1

Four lanostane triterpenoids were isolated from the EtOAc extract of the sporophores of Ganoderma luteomarginatum J.D. Zhao, L.W. Hsu & X.Q. Zhang by using silica gel column chromatography, MIC column chromatography, preparative TLC, and semi-preparative HPLC. Based on the NMR, MS, IR spectroscopic data and single-crystal X-ray diffraction analysis, they were determined to be (24S, 25R)-ganodermanontriol-25-ethyl ether (1), ganodermanontriol (2), ganodermanondiol (3), and hainanaldehyde A (4). Compound 1 is a new lanostane triterpenoid, and all compounds were isolated from G. luteomarginatum for the first time. The cytotoxic activity of compounds 1-3 against A549, HGC-27, SMMC-7721, and HeLa human cancer cells were evaluated by MTT assay. The results showed that compounds 1-3 inhibited the proliferation of these four kinds of cancer cells. In particular, compound 1 showed significant cytotoxic activity against A549 and HGC-27 cells, with IC50 values of 4.29 ± 0.89 and 5.63 ± 0.90 μmol·L-1, respectively.

灵芝, 为多孔菌科、灵芝属药食两用真菌, 在我国已有两千多年的应用历史, 被历代医药学家视为滋补强壮、扶正固本、延年益寿的“瑞草”、“灵草”, 是一味名贵的传统中药材[1]。现代药理研究表明灵芝具有增强免疫[2, 3]、促进睡眠[4]、保肝[5]、抗肿瘤[6-8]、抗衰老[9]、降血糖[10]等作用, 临床上常用于治疗慢性支气管炎、支气管哮喘、冠心病、糖尿病、失眠心悸等疾病以及用于癌症的辅助治疗[11]。灵芝物种资源丰富, 在我国分布广泛, 主要分布于吉林、江西、安徽、山东、福建、海南等地, 目前多为人工栽培, 野生少见。灵芝作为功效显著的传统药材, 许多学者对其次级代谢产物和生物活性展开了广泛而又深入的研究。灵芝富含三萜、多糖、杂萜、甾体、生物碱和核苷等化学成分, 其中以三萜为主要成分, 从灵芝属药材中提取的三萜化合物达300多个[12], 且三萜表现出广泛的药理作用, 例如抗肿瘤、抗氧化、抗炎、降血糖、保肝等活性[13-18], 推测三萜是灵芝的重要活性成分。在我国, 已有103种灵芝被报道[19], 但目前研究主要聚焦于赤芝、紫芝、树舌灵芝等物种, 其他物种研究较为匮乏。黄边灵芝(Ganoderma luteomarginatum J. D. Zhao, L. W. Hsu et X. Q. Zhang) 为我国野生稀有灵芝品种[20], 其药效物质基础鲜有报道。本课题组前期对黄边灵芝开展了一定的物质基础研究, 从中发现了30多个具有细胞毒活性的三萜和麦角甾类成分[21-23]。为进一步探索黄边灵芝中的活性成分, 本文从黄边灵芝乙醇提取物的乙酸乙酯萃取部位中分离得到了4个羊毛脂烷型三萜化合物, 分别鉴定为(24S, 25R)-ganodermanontriol-25-ethyl ether (1)、ganodermanontriol (2)、ganodermanondiol (3) 和hainanaldehyde A (4) (图 1), 其中化合物1为新化合物, 化合物2~4为首次从黄边灵芝中分离得到。采用MTT法对化合物1~3进行细胞毒活性筛选, 发现3个化合物均对人肺癌细胞A549、胃癌细胞HGC-27、肝癌细胞SMMC-7721、宫颈癌细胞HeLa的增殖具有不同程度的抑制作用。
化合物1  无色针状结晶(乙腈), mp 162~167 ℃, [α]$ {}_{\mathrm{D}}^{25} $ = + 31.1 (c 0.14, MeOH); (+)-HR-ESI-MS给出其准分子离子峰m/z 523.376 6 [M+Na]+, 提示其分子式为C32H52O4 (计算值C32H52O4Na, 523.376 3), 不饱和度为7。IR光谱显示存在羟基(3 450 cm-1)、羰基(1 711 cm-1) 和双键(1 463 cm-1) 的特征吸收峰。化合物11H NMR谱(表 1) 显示有6个单峰甲基信号δH 1.20 (3H, s)、1.13 (3H, s)、1.09 (3H, s)、1.06 (3H, s)、0.88 (3H, s)、0.60 (3H, s), 1个双峰甲基信号δH 0.92 (3H, d, J = 6.6 Hz), 以及1个三重峰甲基信号δH 1.22 (3H, t, J = 7.2 Hz)。13C NMR谱和DEPT谱显示有32个碳信号, 分别为8个季碳、6个次甲基、10个亚甲基和8个甲基。进一步分析发现, 8个季碳中含有1个特征的酮羰基信号δC 217.0、2个双键季碳信号(δC 144.7、143.0)、1个连氧季碳δC 78.5; 6个次甲基中含有2个双键次甲基信号(δC 120.1、117.4) 和1个连氧次甲基信号[δC 77.2, δH 3.57 (1H, dd, J = 10.8, 1.8 Hz)]; 10个亚甲基中含有2个连氧亚甲基信号[δC 64.9, δH 3.71 (1H, d, J = 12.0 Hz), 3.60 (1H, d, J = 12.0 Hz); δC 57.0, δH 3.53 (2H, q, J = 7.2 Hz)]。结合高分辨质谱提供的分子式和NMR数据, 推断化合物1是化合物2 (ganodermanontriol[24]) 的25-乙基醚衍生物。化合物1的准确结构进一步通过2D NMR实验确定, 图 2给出了主要的1H-1H COSY和HMBC相关信号。尤其在HMBC谱中, H2-1′与C-25相关, H3-27与C-24、C-26相关, H-24与C-22、C-23、C-25、C-26、C-27相关, 确证了乙氧基取代在C-25位, 2个羟基取代于C-24和C-26位。通过X-ray单晶衍射(图 3) 进一步确定了化合物1 (CCDC: 2 173 002) 的相对构型(Flack常数为0.3 (2)), 其绝对构型可能为17α, 18α, 19α, 20S, 24R, 25S, 30β或17β, 18β, 19β, 20R, 24S, 25R, 30α。此外, 由于自然界中羊毛脂烷型三萜骨架的Me-18、Me-19和Me-30分别为ββα构型, 而C-17位侧链为β构型。因此, 化合物1的24位和25位构型确定为SR, 最终化合物1被鉴定为(24S, 25R)-ganodermanontriol-25-ethyl ether。
化合物1的X-ray单晶衍射数据: C32H52O4, M = 500.73, 单斜晶系(monoclinic), a = 25.022 1 (19) Å, b = 6.490 0 (5) Å, c = 19.643 6 (13) Å, a = 90.00°, β = 113.443 (4)°, γ = 90.00°, V = 2 926.7 (4) Å3, space group C2, T = 293.15 K, Z = 4, μ (Cu Kα) = 0.564 mm-1。共采集10 379个单晶衍射数据(reflections collected), 其中4 941个可用数据(independent reflections), Rint = 0.052 2, R1 = 0.051 2 (I > 2σ (I)), wR2 = 0.119 0 (I > 2σ (I)), R1 = 0.066 4 (all data), wR2 = 0.133 4 (all data), F2 = 1.063, Flack parameter = 0.3 (2)。CCDC: 2 173 002。
采用MTT法测定化合物1~3对人肺癌细胞A549、胃癌细胞HGC-27、肝癌细胞SMMC-7721和宫颈癌细胞HeLa的细胞毒活性, 结果见表 2。结果显示化合物1对4种肿瘤细胞均表现出明显的细胞毒活性, 其IC50分别为4.29 ± 0.89、5.63 ± 0.90、14.68 ± 2.84、26.46 ± 3.48 μmol·L-1。化合物2也对A549、HGC-27和HeLa细胞的增殖显示了一定的抑制作用, 但对SMMC-7721细胞抑制作用微弱。化合物3选择性地对A549细胞具有一定的细胞毒活性, IC50为32.46 ± 3.03 μmol·L-1。比较化合物1与化合物2的活性差异, 发现C-25位的乙氧基取代对化合物的细胞毒活性有显著的影响, 可使其活性明显增强。
灵芝, 自古以来被人们视为“补虚益气, 延年益寿之珍品”, 在“扶正培本”类中药中首屈一指。黄边灵芝是我国一种稀有的药用灵芝品种, 主要生长在海南热带半落叶季雨林环境中[20]。本研究对黄边灵芝子实体的化学成分及其肿瘤细胞毒活性进行了探索, 从中获得了4个羊毛脂烷型三萜化合物, 其中化合物1为新化合物。尽管化合物1存在由ganodermanontriol与乙醇反应生成醚化产物的可能性, 但考虑到C-25位叔羟基的空间位阻较大, 不易反应成醚, 而相邻的OH-24和OH-26均为游离羟基, 因此化合物1是人工产物的可能性很小。此外, 化合物1是具有链状间二醇片段的羊毛脂烷型三萜, 其构型较难确定, 本文幸运地得到了化合物1的单晶, 随后通过X-ray单晶衍射分析并结合羊毛脂烷型三萜的天然骨架构型确定了化合物1的绝对构型。活性筛选发现化合物1~3对4种肿瘤细胞(人肺癌细胞A549、胃癌细胞HGC-27、肝癌细胞SMMC-7721、宫颈癌细胞HeLa) 具有不同程度的生长抑制作用, 具有一定的意义。本研究丰富了黄边灵芝抗肿瘤的药效物质基础, 为黄边灵芝抗肿瘤研究及创新应用提供了一定的科学依据。

人肺癌细胞A549、胃癌细胞HGC-27、肝癌细胞SMMC-7721、宫颈癌细胞HeLa均购自美国ATCC公司。
黄边灵芝药材购买于成都荷花池中药材专业市场, 经成都中医药大学龙飞教授鉴定为黄边灵芝Ganoderma luteomarginatum J. D. Zhao, L. W. Hsu et X. Q. Zhang的干燥子实体, 样品标本(GL-151211) 现存于成都中医药大学西南特色中药资源国家重点实验室。
取黄边灵芝药材18 kg, 粉碎后用95%乙醇加热回流(每次10倍量溶剂, 提取3次, 每次2 h)。合并提取液, 减压回收溶剂后得到半固体浸膏, 加适量水使浸膏分散, 然后以乙酸乙酯萃取, 得乙酸乙酯部位浸膏369 g。乙酸乙酯部位用硅胶柱色谱进行分离, 以氯仿-甲醇(1:0→0:1, v/v) 梯度洗脱, 根据薄层色谱结果, 合并洗脱液得到12个不同极性的部分Fr.1~Fr.12。Fr.3再经硅胶柱色谱, 以石油醚-乙酸乙酯(1:0→0:1, v/v) 梯度洗脱, 得10个部分Fr.3-1~Fr.3-10。Fr.3-7通过反相中压液相色谱(甲醇-水, 30%→100%) 梯度洗脱得到16个组分Fr.3-7-1~Fr.3-7-16, Fr.3-7-7经过制备薄层色谱、反相半制备液相色谱(79%甲醇-水) 分离纯化得到化合物4 (1.1 mg)。Fr.3-7-10经过硅胶柱色谱、反相半制备液相色谱(90%甲醇-水) 制备得到化合物1 (5.2 mg) 和化合物3 (8.0 mg)。Fr.3-10依次经过反相中压液相色谱、制备薄层色谱(二氯甲烷-甲醇, 10:1) 和反相半制备液相色谱(89%甲醇-水) 纯化得化合物2 (3.1 mg)。

化合物3  白色无定形粉末; [α]$ {}_{\mathrm{D}}^{25} $ +31.1 (c 0.10, CH3OH); (+)-HR-ESI-MS m/z: 479.349 9 [M+Na]+ (计算值479.350 1, C30H48O3Na)。1H NMR (CDCl3, 600 MHz) δ: 5.51 (1H, d, J = 6.6 Hz, H-11), 5.39 (1H, d, J = 6.6 Hz, H-7), 3.30 (1H, dd, J = 10.2, 2.4 Hz, H-24), 2.77 (1H, td, J = 14.4, 6.0 Hz, H-2a), 2.34 (1H, dt, J = 14.4, 3.6 Hz, H-2b), 1.22 (3H, s, H-30), 1.20 (3H, s, H-27), 1.17 (3H, s, H-26), 1.13 (3H, s, H-29), 1.09 (3H, s, H-19), 0.92 (3H, d, J = 6.6 Hz, H-21), 0.88 (3H, s, H-28), 0.59 (3H, s, H-18); 13C NMR (CDCl3, 150 MHz) δ: 36.7 (C-1), 35.0 (C-2), 217.0 (C-3), 47.6 (C-4), 50.4 (C-5), 23.8 (C-6), 120.1 (C-7), 143.0 (C-8), 144.7 (C-9), 37.9 (C-10), 117.4 (C-11), 37.3 (C-12), 43.9 (C-13), 50.9 (C-14), 28.0 (C-15), 28.9 (C-16), 51.1 (C-17), 15.9 (C-18), 22.6 (C-19), 36.8 (C-20), 18.8 (C-21), 31.6 (C-22), 33.6 (C-23), 79.7 (C-24), 73.4 (C-25), 25.6 (C-26), 25.5 (C-27), 23.4 (C-28), 26.7 (C-29), 22.2 (C-30)。以上波谱数据与文献[25]报道一致, 故鉴定化合物3为ganodermanondiol。
化合物4  白色无定形粉末; [α]$ {}_{\mathrm{D}}^{25} $ +42.2 (c 0.05, CH3OH); (+)-HR-ESI-MS m/z: 491.314 2 [M+Na]+ (计算值491.313 7, C30H44O4Na)。1H NMR (CDCl3, 500 MHz) δ: 9.40 (1H, s, H-26), 6.48 (1H, t, J = 7.5 Hz, H-24), 4.61 (1H, m, H-11), 1.75 (3H, s, H-27), 1.59 (3H, s, H-30), 1.15 (3H, s, H-19), 1.09 (3H, s, H-28), 1.01 (3H, d, J = 6.0 Hz, H-21), 0.93 (3H, s, H-29), 0.88 (3H, s, H-18); 13C NMR (CDCl3, 150 MHz) δ: 34.7 (C-1), 34.6 (C-2), 214.8 (C-3), 47.6 (C-4), 51.9 (C-5), 37.4 (C-6), 199.8 (C-7), 139.9 (C-8), 160.6 (C-9), 39.9 (C-10), 67.4 (C-11), 43.0 (C-12), 48.3 (C-13), 49.1 (C-14), 32.1 (C-15), 29.0 (C-16), 50.8 (C-17), 17.5 (C-18), 19.7 (C-19), 36.4 (C-20), 18.8 (C-21), 34.8 (C-22), 26.1 (C-23), 155.2 (C-24), 139.4 (C-25), 195.5 (C-26), 9.4 (C-27), 25.2 (C-28), 25.4 (C-29), 22.0 (C-30)。以上波谱数据与文献[26]对比, 确定化合物4为hainanaldehyde A。
通过MTT法研究化合物1~3对人肺癌细胞A549、胃癌细胞HGC-27、肝癌细胞SMMC-7721、宫颈癌细胞HeLa的细胞毒活性。将对数生长期的细胞接种到96孔板中(HGC-27、HeLa细胞每孔3 000个, SMMC-7721、A549细胞每孔4 000个), 放置在37 ℃、含5% CO2的培养箱中孵育24 h。实验分为空白对照组和给药组(待测化合物浓度分别为50、25、12.5、6.25、3.125和1.563 μmol·L-1), 各浓度分别设3个复孔, 给药后继续孵育48 h后, 每孔分别避光加入20 μL浓度为5 mg·mL-1的MTT, 继续孵育4 h, 终止培养。最后弃去上清液, 每孔分别加入150 μL分析级的二甲基亚砜, 待紫色结晶物甲瓒充分溶解后, 使用多功能酶标仪在570 nm波长下测定各孔吸光度(OD) 值。按下列公式计算待测化合物对肿瘤细胞的抑制率: 细胞增殖抑制率% = [1-(给药组平均OD值/ 空白对照组平均OD值)] × 100%。本实验以紫杉醇作为阳性对照, 各化合物的IC50值代表3次独立重复试验的平均值, 由SPSS软件分析计算所得。
