Two new labdane diterpenoids were isolated from 95% ethanol extract of the leaves of Callicarpa formosana Rolfe by using silica gel column, MCI column, ODS column and HPLC. Their structures were elucidated by HR-ESI-MS, NMR and ECD spectral data. All of them are new compounds, named 13E-6β-hydroxylabda-8(17), 13-dien-15-oic acid (1) and 13E-7α-hydroxylabda-8(17), 13-dien-15-oic acid (2). Compounds 1 and 2 were tested for antioxidant activity, and none of them had obvious activity.

采用硅胶柱色谱、MCI柱色谱、ODS柱色谱及高效液相色谱等色谱技术, 从杜虹花叶95%乙醇提取物中分离得到2个新半日花烷型二萜类化合物, 并通过质谱、核磁共振和ECD等波谱数据鉴定其结构, 分别为13E-6β-hydroxylabda-8(17), 13-dien-15-oic acid (1) 和13E-7α-hydroxylabda-8(17), 13-dien-15-oic acid (2)。对化合物12进行了抗氧化活性测试, 均无明显活性。

Two new labdane diterpenoids were isolated from 95% ethanol extract of the leaves of Callicarpa formosana Rolfe by using silica gel column, MCI column, ODS column and HPLC. Their structures were elucidated by HR-ESI-MS, NMR and ECD spectral data. All of them are new compounds, named 13E-6β-hydroxylabda-8(17), 13-dien-15-oic acid (1) and 13E-7α-hydroxylabda-8(17), 13-dien-15-oic acid (2). Compounds 1 and 2 were tested for antioxidant activity, and none of them had obvious activity.

杜虹花(Callicarpa formosana Rolfe) 是马鞭草科(Verbenaceae) 紫珠属灌木。在我国江西、浙江、台湾、福建、云南等地均有分布。其叶入药, 有散瘀消肿, 止血镇痛的功效[1], 是紫珠叶的基源植物, 收载于2020版中国药典一部[2]。临床上常用来治疗喉炎、结膜炎等炎症[1]以及各种出血症[3]。杜虹花主要含有黄酮、萜类以及苯丙素类化学成分[4, 5], 具有镇痛、止血、抑菌、抗肿瘤、抗炎等活性[6-9]。二萜类成分是紫珠属植物的特征性化学成分之一, 表现了广泛的生物活性, 如抗炎活性[10]、抗结核活性[11]、抗癌活性[12]和免疫调节活性[13]。杜虹花二萜类成分的研究报道相对较少, 为了进一步明确杜虹花的药效物质基础, 本研究对杜虹花叶萜类富集部位展开研究, 利用多种色谱分离手段从杜虹花叶的95%乙醇提取物中分离得到2个新的半日花烷型二萜, 分别为13E-6β-hydroxylabda-8(17), 13-dien-15-oic acid (1) 和13E-7α-hydroxylabda-8(17), 13-dien-15-oic acid (2), 结构见图 1。并对化合物12进行了抗氧化活性测试, 实验结果显示均无明显活性。
13E-6β-hydroxylabda-8(17), 13-dien-15-oic acid (1)  白色无定型粉末, 易溶于甲醇, [α]20 +23 (c 0.1 MeOH)。高分辨质谱HR-ESI-MS m/z 319.227 7 [M-H]- (C20H31O3, 计算值319.227 9) 提示其分子式为C20H32O3, 不饱和度为5。化合物1的氢谱显示出4个甲基、1个环外双键和1个连氧氢信号, 初步表明化合物1可能是半日花烷型二萜类化合物[14]。通过化合物11H NMR、13C NMR、DEPT和HSQC谱对化合物1的碳氢信号进行归属(表 1)。通过分析化合物1的HMBC以及1H-1H COSY谱确定了1的平面结构。在HMBC谱(图 2) 中H-19与C-18、C-4、C-3、C-5相关, H-20与C-10、C-9、C-5、C-1相关, 以及H-17与C-9、C-7的相关, 揭示了化合物1中半日花烷双环母核以及C-8和C-17环外双键的存在。HMBC谱中H-16与C-12、C-14、C-13、C-15相关, H-14与C-15、C-16、C-12相关, 结合1H-1H COSY谱(图 2) 中H-11/H-12的相关, 提示结构中存在-CH2-CH2-C(CH3)=CH-COOH侧链。该侧链C-9-C-11的连接方式是通过H-9与C-11、C-12的HMBC相关, 以及1H-1HCOSY谱中H-9/H-11的相关确定的。此外, HMBC谱中H-6与C-8、C-10的相关, 以及1H-1H COSY谱中H-5/H-6/H-7的相关, 说明羟基连接在C-6位, 从而化合物1的平面结构得以确定。NOESY谱显示6-OH与H-19相关, H-5和H-6相关表明6-OH为β取向, H-14和H-12相关, 表明侧链双键为E构型, 因此确定该化合物的构型为1a (5S, 6R, 9S, 10R) 或1b (5R, 6S, 9R, 10S)。经过文献[15]数据比对, 化合物1与ent-labd-6α-ol-8(17), 13-dien-15-oic acid结构相似, 区别仅为6-OH构型不同。为了进一步确定化合物1的绝对构型, 采取量子化学计算的方法[B3LYP/6-311G (d, p), MeOH] 计算了1a1b的ECD谱, 结果表明1a的计算ECD曲线与化合物1的实验曲线吻合较好(图 3), 因此确定化合物1的构型为5S, 6R, 9S, 10R, 结构如图 1所示。
13E-7α-hydroxylabda-8(17), 13-dien-15-oic acid (2)  白色无定型粉末, 易溶于甲醇, [α]20 -2 (c 0.045 MeOH)。高分辨质谱HR-ESI-MS: m/z 343.223 8 [M+Na]+ (C20H32O3Na, 计算值343.224 4) 提示其分子式为C20H32O3, 不饱和度为5。化合物2与化合物1的核磁数据相似, 表明其为结构相似的半日花烷型二萜类化合物。化合物2与化合物1核磁数据的主要不同是连氧次甲基的化学位移, 提示结构中羟基的取代位置可能不同。在HMBC谱中H-7与C-5、C-8、C-9、C-17相关, 说明化合物2的羟基连接在C-7位。化合物2的NOESY谱显示, H-7与H-17b相关, H-17a与H-20相关, 表明H-7与H-20同侧为β取向, H-14和H-12相关, 表明侧链双键为E构型, 因此化合物2的构型为2a (5R, 7S, 9S, 10R) 或2b (5S, 7R, 9R, 10S)。为了进一步确定化合物2的绝对构型, 采取量子化学计算的方法[B3LYP/6-311G (d, p), MeOH] 计算了2a2b的ECD谱, 结果表明2b的计算ECD曲线与化合物2的实验曲线吻合较好(图 3), 确定化合物2的绝对构型为5S, 7R, 9R, 10S, 结构如图 1所示。
通过测试化合物在肝微粒体氧化抑制模型中对丙二醛(MDA) 释放量的抑制作用进行化合物的抗氧化活性评价, 药理结果显示化合物12均未表现出明显的MDA抑制活性。
紫外可见分光光度仪(JASCO V-650型); 旋光测定仪(JASCO P-2000型, 日本); 质谱仪(Agilent1100 LC/MSD)、高效液相色谱仪(Aglient1100) (安捷伦科技有限公司); 核磁共振仪(Bruker AV-ш400、Bruker AV-ш500); 中压制备液相色谱系统(Büchi Gradient Former B-687, RP C18), 制备液相色谱仪(LC-6AD, 日本岛津公司); 反相填料(YMC-ODS C18 50 μm, 日本YMC公司); MCI填料(CHP20/P120, 日本三菱公司); 制备色谱柱(YMC-pack, ODS-A C18, 250 mm × 20 mm, 日本YMC公司); 薄层色谱用硅胶GF254和柱色谱用硅胶(100~200目, 200~300目, 青岛海洋化工有限公司)。
杜虹花叶购于河北安国药材批发市场, 经中国医学科学院药物研究所马林教授鉴定为杜虹花Callicarpa formosana Rolfe的叶, 标本(ID-S-2985) 现存放于中国医学科学院药物研究所的植物标本室。
杜虹花(10 kg) 用95%乙醇加热回流提取2次, 减压回收乙醇, 得到乙醇提取物(8.53 kg)。将提取物加水混悬, 用体积为1/2的二氯甲烷萃取, 得到二氯甲烷层和水层。将二氯甲烷层减压回收溶剂, 得到1.075 kg浸膏, 该浸膏溶解后用等量的100~200目硅胶拌样, 通过硅胶柱色谱进行初步分离, 用石油醚-乙酸乙酯(5∶1~1∶1)、乙酸乙酯、乙酸乙酯-甲醇(9∶1~7∶3)、甲醇梯度洗脱, 得到43个洗脱部A1~A43。将流分A4~A8合并得45 g, 与MCI拌样, 进行MCI柱色谱分离, 用50%~100%的甲醇梯度洗脱, 得到28个洗脱部分(B1~B28)。将流分B8、B9合并(7 g), 进行中压液相色谱分离, 用75%~100%的甲醇梯度洗脱, 得到26个洗脱部分: C1~C26。流分C15通过反相HPLC制备(55%乙腈水溶液洗脱, 0~120 min) 进行纯化, 得到化合物1 (tR = 83.5 min, 17 mg)。流分C18通过反相HPLC制备(55%乙腈水溶液洗脱, 0~100 min) 进行纯化, 得到化合物2 (tR = 42.7 min, 8 mg)。
化合物1  白色无定形粉末; [α]20+23 (c 0.1 MeOH); UV (MeOH) λmax (logε)/nm: 205 (4.18)、220 (4.01)、280 (2.79); IR/cm-1: 3 521、2 927、1 705、1 158、858; HR-ESI-MS: m/z 319.227 7 [M-H]- (C20H31O3, 计算值319.227 9), 确定其分子式为C20H32O3; 1H NMR (DMSO-d6, 500 MHz) 和13C NMR (DMSO-d6, 100 MHz) 数据见表 1
化合物2  白色无定形粉末; [α]20 -2 (c 0.045 MeOH); UV (MeOH) λmax (logε)/nm: 204 (5.85)、218 (5.89); IR/cm-1: 3 339、2 926、1 688、1 205、802; HR-ESI-MS: m/z 343.223 8 [M+Na]+ (C20H32O3Na, 计算值343.224 4), 确定其分子式为C20H32O3; 1H NMR (DMSO-d6, 500 MHz) 和13C NMR (DMSO-d6, 100 MHz) 数据见表 1
设置空白组(C组)、模型组(M组)、阳性药组(J1、J2) 和加药组, 每组设置3个复孔(即平行3次)。每管各加PBS缓冲液155 μL、肝微粒体30 μL、半胱氨酸50 μL, 即混合体系每管共235 μL。C组和M组每管加DMSO 2.5 μL, 阳性药组每管加姜黄素(1×10-3 mol·L-1)2.5 μL (终浓度1×10-5 mol·L-1), 加药组每管加待筛化合物(1×10-3 mol·L-1) 2.5 μL (终浓度1×10-5 mol·L-1)。于37 ℃水浴震荡15 min。阳性药组、M组和加药组每管加1×10-2 mol·L-1 FeSO4溶液13 μL, C组加PBS缓冲液13 μL。于37 ℃水浴震荡15 min。每管各加TCA溶液250 μL、TBA溶液500 μL。于100 ℃水浴10 min。每管取600 μL于1.5 mL EP管中, 8 000 r·min-1离心10 min。取上清液200 μL于96孔板中, 于532 nm处(选择MDA) 测吸光度(OD)。MDA浓度C = (OD - 0.006) / 0.07 × 10 nmol·mL-1。抑制率(IR) (%) = (C模型 - C化合物) / (C模型 - C空白) × 100%
