Seven compounds were isolated from the alcohol extract of Edgeworthia gardneri by various technologies, including silica gel, Sephadex LH-20 and high performance liquid chromatography, and were identified as edgeworthiaside A (1), 2, 4, 6-trichlorol-3-methyl-5-methoxy-phenol 1-O-β-D-glucopyranosyl-(1-6)-β-D-glucopyranoside (2), 2, 6-dimethoxy-4-(2-propen-1-yl)phenyl 6-O-(6-deoxy-α-L-mannopyranosyl)-β-D-glucopyranoside (3), eugenol rutinoside (4), tiliroside (5), edgeworoside C (6), and salicylic acid (7). Compound 1 is a new chlorophenyl glycoside and 2-4 were isolated for the first time from Edgeworthia gardneri. The in vitro inhibition of α-glucosidase showed that the inhibition rate of compounds 1 and 2 were similar to acarbose.

采用硅胶柱色谱、凝胶色谱及高压制备色谱等方法对绿萝花的成分进行分离纯化,通过其理化性质及波谱数据对分离得到的化合物进行结构表征。从绿萝花醇提取物中共分离得到7个化合物:绿萝花苷A(1)、2,4,6-trichlorol-3-methyl-5-methoxy-phenol 1-O-β-D-glucopyranosyl-(1-6)-β-D-glucopyranoside(2)、2,6-dimethoxy-4-(2-propen-1-yl)phenyl 6-O-(6-deoxy-α-L-mannopyranosyl)-β-D-glucopyranoside(3)、丁香酚芸香糖苷(4)、银锻苷(5)、结香苷C(6)和水杨酸(7)。其中1为新化合物,2~4首次从该植物中分离得到。药理活性表明,化合物12均对α-葡萄糖苷酶具有显著的抑制活性,其抑制率与阿卡波糖相当。

采用硅胶柱色谱、凝胶色谱及高压制备色谱等方法对绿萝花的成分进行分离纯化,通过其理化性质及波谱数据对分离得到的化合物进行结构表征。从绿萝花醇提取物中共分离得到7个化合物:绿萝花苷A(1)、2,4,6-trichlorol-3-methyl-5-methoxy-phenol 1-O-β-D-glucopyranosyl-(1-6)-β-D-glucopyranoside(2)、2,6-dimethoxy-4-(2-propen-1-yl)phenyl 6-O-(6-deoxy-α-L-mannopyranosyl)-β-D-glucopyranoside(3)、丁香酚芸香糖苷(4)、银锻苷(5)、结香苷C(6)和水杨酸(7)。其中1为新化合物,2~4首次从该植物中分离得到。药理活性表明,化合物12均对α-葡萄糖苷酶具有显著的抑制活性,其抑制率与阿卡波糖相当。

Seven compounds were isolated from the alcohol extract of Edgeworthia gardneri by various technologies, including silica gel, Sephadex LH-20 and high performance liquid chromatography, and were identified as edgeworthiaside A (1), 2, 4, 6-trichlorol-3-methyl-5-methoxy-phenol 1-O-β-D-glucopyranosyl-(1-6)-β-D-glucopyranoside (2), 2, 6-dimethoxy-4-(2-propen-1-yl)phenyl 6-O-(6-deoxy-α-L-mannopyranosyl)-β-D-glucopyranoside (3), eugenol rutinoside (4), tiliroside (5), edgeworoside C (6), and salicylic acid (7). Compound 1 is a new chlorophenyl glycoside and 2-4 were isolated for the first time from Edgeworthia gardneri. The in vitro inhibition of α-glucosidase showed that the inhibition rate of compounds 1 and 2 were similar to acarbose.

藏药绿萝花是瑞香科植物结香属滇结香[Edgeworthia gardneri (Wall.) Meisn.] 的干燥花蕾, 又名黄金葛、石柑子、马蹄金, 主要分布在西藏及云南寒冷的高海拔地区, 是西藏特色的民族习用药材。民间主要通过热水泡茶直接饮用的方式来治疗糖尿病、高血压等慢性疾病[1, 2], 现代药理研究表明, 其具有降血糖、抗氧化、激活PPARs、抑制DNA聚合酶P等多种药理活性[3-7]。绿萝花是西藏的珍贵药材, 现代药理学实验结果证实绿萝花的降血糖作用尤为明显, 但是其有效成分与作用机制并不明确, 一般降糖活性研究仅仅是对绿萝花粗提物进行, 对具体的药效物质基础缺乏深入、系统性研究。绿萝花的各种功效仅局限于当地人的认知, 有针对性、大规模的开发不足, 制约了绿萝花这一宝贵传统资源的利用[8]。因此, 为了进一步研究绿萝花的化学成分, 明确其降血糖活性的药效物质基础, 科学合理地开发该药用资源, 本实验从绿萝花中分离到了7个化合物(图 1), 分别为绿萝花苷A (1)、2, 4, 6-trichlorol-3-methyl-5-methoxy-phenol 1-O-β-D-glucopyranosyl-(1→6)-β-D-glucopyranoside (2)、2, 6-dimethoxy-4-(2-propen-1-yl)phenyl 6-O-(6-deoxy-α-L-mannopyranosyl)-β-D-glucopyranoside (3)、丁香酚芸香糖苷(4)、银锻苷(5)、结香苷C (6) 和水杨酸(7)。其中1为新化合物, 2~4首次从该植物中分离得到。药理活性表明, 化合物12均对α-葡萄糖苷酶具有显著的抑制活性, 其降糖效果与阿卡波糖相当。
化合物1  白色针状晶体(甲醇-水), [α]20D -38.2 (c 0.1, MeOH); mp 257~259 ℃; TLC薄层色谱在254 nm紫外灯下有暗斑, 喷5%的硫酸乙醇溶液, 105 ℃加热显色, 呈黄色。ESI-MS m/z 558.8 [M-H]-, 560.8 [M-H]-, 562.8 [M-H]-, 564.8 [M-H]-, 分子质量两两相差2.0 Da, 从质谱图的丰度显示可以分成两组, 且同组中两个分子质量丰度比例大约为3∶1, 推测该化合物含有两个氯原子[9], 相对分子质量为560; HR-ESI-MS (positive) m/z 583.096 2 [M+Na]+ (计算值: 583.096 1, C21H30O13Cl2Na); HR-ESI-MS (negative) m/z 559.098 0 [M-H]- (计算值: 559.098 5, C21H29O13Cl2) 确定其分子式为C21H30O13Cl2。在氢谱(表 1) 的低场区, 只有一个质子信号δH 7.39 (1H, s), 结合紫外吸收情况, 说明该化合物含有一个五-取代的苯环; 在中间区域显示有两个葡萄糖端基氢质子信号δH 4.29 (1H, d, J = 7.6 Hz)、4.26 (1H, d, J = 7.6 Hz) 及两个甲氧基质子信号δH 3.88 (3H, s)、3.82 (3H, s)。碳谱结合DEPT 135°分析, 该化合物共有21个碳原子, 包括5个季碳、11个叔碳、3个仲碳、2个伯碳; 低场部分显示一个5取代苯环及两个葡萄糖的端基碳信号δC 154.8、152.6、136.7、117.8、115.5、108.2、103.7、102.7, 中间场区主要是两个葡萄糖的碳信号δC 77.3、77.2、76.9、76.3、74.0、73.8、70.5、70.3、68.7、66.8、61.5, 从数据上看, 有一个葡萄糖的6位碳向低场移动, 说明有一个取代基连在葡萄糖的6位上, 除了两个葡萄糖以外, 该化合物还有一个连氧的亚甲基; 在高场部分显示有两个甲氧基的碳信号δC 60.8、57.0, 说明苯环上有两个甲氧基取代基。
通过HSQC对碳氢进行全归属, 由HMBC确定各基团的连接顺序, δH 7.39 (H-6)与δC 136.7 (C-1)、117.8 (C-2)、115.5 (C-4)、154.8 (C-5)、66.8 (C-7) 相关, 说明该化合物含有苄基的结构单元; δH 3.88 (H-3-OMe) 与δC 152.6 (C-3), δH 3.82 (H-5-OMe) 与δC 154.8 (C-5) 相关, 说明甲氧基连在苄基的3、5位上, 2、4位被氯取代; 同时, δH 4.26 (H-1') 与δC 66.8 (C-7) 相关, 提示一个葡萄糖基团连在苄基的7位上, δH 4.29 (H-1'') 与δC 68.7 (C-6') 相关, 则进一步证实第二个葡萄糖基团是连在第一个葡萄糖的6位上(图 2)。
对化合物1进行酸水解, 通过薄层色谱的方式与对照品进行比较分析, 同时将水解液与D-葡萄糖对照品通过HPLC进一步确认, 说明该化合物只含有D-葡萄糖, 结合1H NMR端基氢的耦合常数, 连接的糖为β-D-葡萄糖。通过以上分析, 确认该化合物结构为: 2, 4-二氯-3, 5-二甲氧基-苄基-7-O-β-D-葡萄糖(1→6)-O-β-D-葡萄糖苷。经Scifinder查询, 未有该化合物的报道, 为新化合物, 命名为绿萝花苷A。
依据文献[10]方法, 以PNPG为底物, 阿卡波糖为阳性对照, 对化合物1~6进行降糖活性筛选, 化合物12均显示良好的降糖活性(表 2), 这对于进一步深度开发该药用资源提供参考依据。
AB SCIEX Triple TOFTM 5600高分辨质谱仪(美国AB公司), Bruker AV 400型核磁共振波谱仪(TMS为内标), 中压制备色谱仪(MPLC) FS-9200T (天津博纳艾杰尔科技有限公司), 半制备型高效液相色谱仪Waters 2545, 2487检测器(美国Waters公司), 酶标仪及培养箱(美国Thermo公司), 阿卡波糖(上海源叶生物科技有限公司), α-葡萄糖苷酶(酿酒酵母) (上海源叶生物科技有限公司), 葡聚糖凝胶Sephadex LH-20 (瑞典Pharmacia公司), 柱色谱硅胶、薄层色谱硅胶(青岛海洋化工), 乙腈、甲醇(色谱纯), 其余试剂为国产分析纯均购自成都市科龙化工试剂厂。
实验所用藏药绿萝花于2019年9月购买于西藏拉萨市亚郎藏药, 经中国科学院成都生物研究所赵佐成研究员鉴定为瑞香科结香属植物滇结香(Edgeworthia gardneri Wall. Meisn.) 的干燥花蕾, 标本保存于成都师范学院化学与生命科学学院标本室(标本号: 20190903)。
将绿萝花药材4 kg粗粉, 加10倍体积的95%乙醇。加热回流提取3次, 第一、二次2 h, 第三次1.5 h, 将3次提取液合并, 减压回收至溶液基本无醇味, 再加2倍体积的水进行分散稀释。中速滤纸过滤, 滤渣用50%乙醇水洗涤两次, 合并滤液。先用石油醚萃取除去大部分叶绿素, 再用乙酸乙酯和正丁醇依次萃取, 减压回收溶剂, 得乙酸乙酯相86 g, 正丁醇相53 g。将正丁醇部分(50 g) 用甲醇溶解, 60~80目硅胶拌样, 200~300目硅胶湿法装柱, 进行硅胶柱色谱分离。洗脱剂为二氯甲烷-甲醇(20∶1→2∶1), 约500 mL收集一份, 共收集35份。用TLC对各流分进行检测, 显色剂为5%硫酸-香草醛溶液, 合并相同部分, 得到Fr. A~F。将Fr. B (0.8 g) 再次进行硅胶柱色谱, 洗脱剂为二氯甲烷-甲醇(15∶1), TLC进行过程监控, 收集主要斑点, 用半制备反相色谱对收集部分再次分离, 流动相为乙腈-水-磷酸(45∶55∶0.2), 检测波长为254 nm, 得到化合物5 (21 mg)、化合物6 (17 mg); 对具有明显斑点的Fr. C (1.3 g) 再次进行硅胶柱色谱分离, 洗脱剂为二氯甲烷-甲醇(10∶1), TLC进行过程监控, 收集较为单一部分Fr. C1, 用半制备反相色谱分离, 流动相为乙腈-水-磷酸(38∶62∶0.2), 得化合物3 (17 mg)、化合物4 (8 mg), 将Fr. D (1.5 g) 用常压反相硅胶柱色谱进行再次粗分, 以甲醇-水(50∶50→90∶10) 进行梯度洗脱, TLC过程监控, 得到Fr. D3、Fr. D4两个组分, 对得到的每一部分样品再用半制备高效液相进行精制, 流动相为乙腈-水-磷酸, 检测波长为254 nm, 得到化合物1 (13 mg)、化合物2 (16 mg)。Fr. F (0.7 g) 再次进行硅胶柱色谱分离, 洗脱剂为二氯甲烷-甲醇(6∶1), TLC进行过程检测, 收集合并主要斑点, 用半制备反相色谱分离, 流动相为乙腈-水-磷酸(18∶82∶0.2), 检测波长为254 nm, 收集主要目标化合物, 减压浓缩至干, 甲醇溶解, 过滤, 滤液上Sephadex LH-20再次精分, 纯甲醇为洗脱剂, 得到化合物7 (32 mg)。

化合物7   无色针状晶体, ESI-MS m/z: 139 [M+H]+, 137 [M-H]-1H NMR (400 MHz, DMSO-d6) δH: 7.80 (1H, dd, J = 6.0, 1.6 Hz, H-6), 7.51 (1H, m, H-4), 6.96 (1H, d, J = 7.6 Hz, H-3), 6.90 (1H, m, H-5); 13C NMR (100 MHz, DMSO-d6) δC: 113.5 (C-1), 161.6 (C-2), 117.7 (C-3), 136.2 (C-4), 119.8 (C-5), 130.7 (C-6), 172.3 (C-7)。以上波谱数据与文献报道的化合物[16]对照基本一致, 故鉴定化合物7为水杨酸。
以PNPG为底物, 阿卡波糖为阳性对照, 根据文献实验方法: 将实验分为样品空白组(待测化合物+PBS+酶)、酶空白组(PBS+酶)、酶活性组(PBS+PNPG+酶)、对照组(阿卡波糖+PBS+PNPG+酶) 和样品组(待测化合物+PBS+PNPG+酶)。操作步骤如下: 先在96微孔板中加入磷酸盐缓冲液(pH = 6.8) 20 μL, 加入α-葡萄糖苷酶液(0.6 U·mL-1) 溶液20 μL, 再加入浓度为50 mg·L-1的样品溶液40 μL, 低速震荡混匀, 于37 ℃恒温反应10 min后, 加入PNPG 20 μL, 低速震荡混匀, 37 ℃恒温条件下再反应15 min, 加入0.2 mol·L-1 Na2CO3 80 μL中止反应, 将反应液置于酶标仪405 nm波长下测定其吸光度值, 按以下公式计算化合物对α-葡萄糖苷酶液活性的抑制率: α-葡萄糖苷酶抑制率(%) = 1-[(A-A样空)·(A-A酶空)-1]×100%。每一组重复3次, 用Origin 9.1计算其IC50值。
