Eight polyacetylenes were isolated from the extract of the stems and leaves of Chrysanthemum morifolium by various chromatographic methods. Their structures were determined as 2E, 4E, 12Z-tetradecatriene-1-pyrrolidine-1-oxo-8,10-diynoic(1), tetradeca-2E, 4E, 12E-trien-8,10-diynoic acid pyrrolidide(2), tetradeca-2E, 4E-dien-8,10-diynoic acid pyrrolidide(3), tetradeca-2E, 4E, 10Z-trien-8-ynoic acid pyrrolidide(4), 2E, 4E, 12E-tetradecatriene-8,10-diynoic acid isobutylamide(5), 2E, 4E-undecyldiene-8,10-diynoic acid isobutylamide(6), 2E, 4E, 10E-N-isobutyl-2,4,10-tetradecatrien-8-ynoic acid amide(7), and undeca-2E, 4E-diene-8,10-diynoic acid phenylethylamide(8) by spectroscopic methods, including UV, IR, ESI-MS, HR-ESI-MS, 1D and 2D NMR spectra.Among them, compound 1 is a new polyacetylene, and compounds 2-8 were isolated from this plant for the first time. Compounds 5-8 inhibited the proliferation of A549 cell significantly at certain concentration, showing potent antitumor activity.

采用多种色谱技术从怀菊花茎叶中分离得到8个聚炔类化合物。通过UV、IR、ESI-MS、HR-ESI-MS及NMR等波谱技术对其进行结构鉴定, 分别为菊花新炔C (2E, 4E, 12Z-十四碳三烯-1-吡咯烷基-1-酮-8,10-二炔)(1)、tetradeca-2E, 4E, 12E-trien-8,10-diynoic acid pyrrolidide (2)、tetradeca-2E, 4E-dien-8,10-diynoic acid pyrrolidide (3)、tetradeca-2E, 4E, 10Z-trien-8-ynoic acid pyrrolidide (4)、2E, 4E, 12E-tetradecatriene-8,10-diynoic acid isobutylamide (5)、2E, 4E-undecyldiene-8,10-diynoic acid isobutylamide (6)、2E, 4E, 10E-N-isobutyl-2,4,10-tetradecatrien-8-ynoic acid amide(7)和十一-2E, 4E-二烯-8,10-二炔酸苯乙胺(8)。其中化合物1为新化合物, 化合物2~8为首次从植物菊花中分离得到。化合物5~8在一定浓度下对A549细胞具有显著的抑制活性, 说明其具有潜在的抗癌活性。

采用多种色谱技术从怀菊花茎叶中分离得到8个聚炔类化合物。通过UV、IR、ESI-MS、HR-ESI-MS及NMR等波谱技术对其进行结构鉴定, 分别为菊花新炔C (2E, 4E, 12Z-十四碳三烯-1-吡咯烷基-1-酮-8,10-二炔)(1)、tetradeca-2E, 4E, 12E-trien-8,10-diynoic acid pyrrolidide (2)、tetradeca-2E, 4E-dien-8,10-diynoic acid pyrrolidide (3)、tetradeca-2E, 4E, 10Z-trien-8-ynoic acid pyrrolidide (4)、2E, 4E, 12E-tetradecatriene-8,10-diynoic acid isobutylamide (5)、2E, 4E-undecyldiene-8,10-diynoic acid isobutylamide (6)、2E, 4E, 10E-N-isobutyl-2,4,10-tetradecatrien-8-ynoic acid amide(7)和十一-2E, 4E-二烯-8,10-二炔酸苯乙胺(8)。其中化合物1为新化合物, 化合物2~8为首次从植物菊花中分离得到。化合物5~8在一定浓度下对A549细胞具有显著的抑制活性, 说明其具有潜在的抗癌活性。

Eight polyacetylenes were isolated from the extract of the stems and leaves of Chrysanthemum morifolium by various chromatographic methods. Their structures were determined as 2E, 4E, 12Z-tetradecatriene-1-pyrrolidine-1-oxo-8,10-diynoic(1), tetradeca-2E, 4E, 12E-trien-8,10-diynoic acid pyrrolidide(2), tetradeca-2E, 4E-dien-8,10-diynoic acid pyrrolidide(3), tetradeca-2E, 4E, 10Z-trien-8-ynoic acid pyrrolidide(4), 2E, 4E, 12E-tetradecatriene-8,10-diynoic acid isobutylamide(5), 2E, 4E-undecyldiene-8,10-diynoic acid isobutylamide(6), 2E, 4E, 10E-N-isobutyl-2,4,10-tetradecatrien-8-ynoic acid amide(7), and undeca-2E, 4E-diene-8,10-diynoic acid phenylethylamide(8) by spectroscopic methods, including UV, IR, ESI-MS, HR-ESI-MS, 1D and 2D NMR spectra.Among them, compound 1 is a new polyacetylene, and compounds 2-8 were isolated from this plant for the first time. Compounds 5-8 inhibited the proliferation of A549 cell significantly at certain concentration, showing potent antitumor activity.

菊花是菊科植物菊[Chrysanthemum morifolium Ramat]的干燥头状花序, 始载于《神农本草经》, 列为上品, 2015版《中国药典》收录“怀菊”、“亳菊”、“滁菊”、“贡菊”、“杭菊”共同作为“菊花”入药[1]。怀菊花与怀地黄、怀山药、怀牛膝共同作为河南四大怀药, 具有极高的应用价值。课题组前期对怀菊花的化学成分和药理活性进行了相关研究[2-7]。长期已来, 由于怀菊花需求量大, 而在采摘过程中, 仅使用花序, 大量茎、叶作为非药用部位被丢弃, 由此产生的资源浪费, 已成为行业发展面临的棘手问题, 同时也给生态环境带来了巨大的负担。立足于资源节约和环境友好原则, 减少资源消耗, 提升资源利用效益[8], 本课题对怀菊花茎叶化学成分进行研究。本研究采用硅胶、凝胶、聚酰胺色谱柱以及半制备高效液相等多种色谱方法, 从怀菊花茎叶中分离得到8个聚炔类化合物, 分别为菊花新炔C (2E, 4E, 12Z-十四碳三烯-1-吡咯烷基-1-酮-8,10-二炔)(1)、tetradeca-2E, 4E, 12E-trien-8,10-diynoic acid pyrrolidide (2)、tetradeca-2E, 4E-dien-8,10-diynoic acid pyrrolidide (3)、tetradeca-2E, 4E, 10Z-trien-8-ynoic acid pyrrolidide (4)、2E, 4E, 12E-tetradecatriene-8,10-diynoic acid isobutylamide (5)、2E, 4E-undecyldiene-8,10-diynoic acid isobutylamide (6)、2E, 4E, 10E-N-isobutyl-2,4,10-tetradecatrien-8-ynoic acid amide (7)和十一-2E, 4E-二烯-8,10-二炔酸苯乙胺(8)(图 1)。其中化合物1为新化合物, 化合物2~8为首次从植物菊花中分离得到。
化合物1  白色粉末, HR-ESI-MS m/z: 290.150 1[M+Na]+(Calcd.for C18H21NONa, 290.151 5), 提示化合物的分子式为C18H21NO, 不饱和度为9。UV λmax(CH3OH)/nm (logε): 205 (0.98)、211(1.15)、253 (0.86)、267 (1.12)、282 (0.82), 提示该化合物存在共轭烯烃和聚乙炔基团; IR光谱中给出了-C≡C-(2 230 cm-1), -CH=CH-CH=CH-(1 623、1 593和990 cm-1), > N-C=O(3 353、1 653和1 438 cm-1)的特征吸收峰[9]1H NMR(500 MHz, CD3OD)中, δH7.23 (1H, dd, J=11.0, 15.0 Hz, H-3), 6.39 (1H, dd, J=11.0, 15.0 Hz, H-4), 6.32 (1H, d, J=15.0 Hz, H-2), 6.18 (1H, m, H-5), 5.53 (1H, d, J=10.8 Hz, H-12), 6.15 (1H, m, H-13)提示该化合物有3个双键, 其中1个顺式双键, 两个反式双键; δH3.62(2H, t, J=6.9 Hz, H-2'), 2.02 (2H, dd, J=6.9, 13.6 Hz H-3'), 1.94 (2H, dd, J=6.9, 13.6 Hz, H-4'), 3.50 (2H, t J=6.9 Hz, H-5')提示该化合物含有一个四氢吡咯环的结构片段; δH2.45 (2H, dd, J=6.9, 13.7 Hz, H-6), 2.53(2H, dd, J=6.9, 13.7 Hz, H-7), 1.89 (3H, dd, J=1.46.9 Hz, H-14)提示化合物含有两个亚甲基和一个甲基的结构片段。13C NMR (125 MHz, CD3OD), 共给出18个碳信号, 结合DEPT 135谱, 提示化合物中含有1个甲基[δC15.0 (C-14)]、6个亚甲基[δC31.3 (C-6), 18.5(C-7), 46.5 (C-2'), 25.5 (C-3'), 23.9 (C-4'), 45.8 (C-5')]、6个次甲基[δC120.6 (C-2), 141.9 (C-3), 130.0 (C-4)140.1 (C-5), 108.6 (C-12), 141.8 (C-13)]和5个季碳信号[δC165.9 (C-1), 83.0 (C-8), 65.2 (C-9), 78.0 (C-10)71.4 (C-11)]。其中δC46.5 (C-2'), 25.5 (C-3'), 23.9 (C-4'), 45.8 (C-5')进一步确定化合物中存在四氢吡咯环结构, δC165.9 (C=O)为羰基碳信号, δC83.0 (C-8)65.2 (C-9), 78.0 (C-10), 71.4 (C-11)为一组共轭的聚乙炔碳信号。
1H-1H COSY谱中, H-3与H-2和H-4有相关信号, H-5与H-4和H-6相关, H-6与H-7相关, 说明化合物中存在-CH=CH-CH=CH-CH2-CH2-结构片段(片段1);H-13与H-12和H-14相关, 说明化合物中存在-CH=CH-CH3结构片段(片段2)。在HMBC谱中, H-2'和H-5'与C-1相关, 提示四氢吡咯环与C-1相连(图 2); 另外, H-2与C-1有相关信号, 提示片段1与C-1相连; H-7与C-8、C-9、C-10、C-11有相关信号, 提示片段1与C-8相连; H-14与C-11相关, 提示C-11与片段2相连(图 2)。与化合物2相比, 仅在与H-12和H-13之间的构型不同。化合物1中, 3JH-12,H-13的偶合常数为10.8,说明其双键构型为顺式, 化合物23JH-12,H-13的偶合常数为15.8,说明其双键构型为反式。
综上所述, 并结合参考文献[9], 确定该化合物为2E, 4E, 12Z-十四碳三烯-1-吡咯烷基-1-酮-8,10-二炔, 命名为菊花新炔C。其1H NMR和13C NMR数据见表 1
采用MTT法检测化合物1~8对A549细胞活力的影响。结果表明, 化合物5~8均可以显著抑制A549的细胞活力, 提示其可能具有潜在的抗癌活性(表 2)。
本研究从怀菊花茎叶中分离并鉴定了8个聚炔类生物碱, 其中化合物1为新化合物, 化合物2~8为首次从怀菊花中分离得到。天然聚炔类化合物广泛存在于蔬菜和中药中, 多分布于菊科、伞形科、五加科、桔梗科等[10], 具有抗癌、抗炎、杀菌、抗过敏等作用[11, 12]。本研究发现, 化合物5~8对A549细胞具有显著的抑制活性, 这对聚炔类化合物以及怀菊花茎叶的进一步研究提供良好的理论基础。
LC50型高压制备液相色谱仪[赛谱锐思(北京)科技有限公司], 色谱柱为(YMC-Pack ODS-A, 250 mm×10 mm.D.S-5 m, 12 m), Bruker AVANCEⅢ500核磁共振仪(TMS为内标), Bruker maxis HD型飞行时间质谱仪(德国), Thermo Nicolet IS10红外光谱仪(Thermo Scientific, USA), Thermo EVO300紫外分光光度计(Thermo Scientific, USA), 柱色谱硅胶H (200~300目, 青岛海洋化工厂), 所用分析纯和色谱纯试剂为北京化工厂和天津第三化学试剂厂生产, 柱色谱填料Toyopearl HW-40 (日本三菱化学公司), 聚酰胺柱色谱(60~80目, 麦克林生化科技有限公司), 二氧化碳培养箱(上海STIK), 超净工作台(苏净集团), Arium 611 VF超级组合型超纯水器(SARTORIUS), 倒置显微镜(Nikon); A 549人非小细胞肺癌细胞株(中国科学院上海细胞库); 培养皿(Corning), 96孔板(Costar), 胰蛋白酶(Solarbio), DMEM高糖培养基(Gibco), 胎牛血清(浙江天杭生物科技有限公司), DMSO (Solarbio), 水为超纯水, PBS缓冲液等为自配。
本课题所用的怀菊花茎叶于2018年11月采自河南温县, 经河南中医药大学董诚明教授鉴定为菊科植物怀菊[Chrysanthemum morifolium Ramat.]的干燥茎叶; 留样标本[20181110]存放于河南省中药资源与中药化学重点实验室。
怀菊花茎叶35 kg, 用50%含水丙酮组织破碎提取2次, 提取液减压浓缩得总浸膏2.85 kg, 用水分散溶解, 依次用石油醚、乙酸乙酯、正丁醇萃取, 得到石油醚部位(16.5 g)、乙酸乙酯部位(90.0 g)、正丁醇部位(160.0 g)和水部位(1.00 kg)。将正丁醇部位加水溶解, 经聚酰胺柱色谱, 依次用水、20%乙醇、50%乙醇、70%乙醇、95%乙醇洗脱, 得到水部位、20%乙醇部位、50%乙醇部位、70%乙醇部位、95%乙醇部位。
50%乙醇部位经硅胶柱, 用二氯甲烷-甲醇(100∶1、50∶1、30∶1、10∶1)洗脱得到Fr.1~Fr.9。其中Fr.3经Toyopearl HW-40柱色谱, 以甲醇-水=70∶30为流动相等度洗脱, 得Fr.3.1~Fr.3.2。Fr.3.1经半制备高效液相, 流动相(乙腈-水=40∶60, 3 mL·min-1, λ=210 nm)等度洗脱, 得到化合物2 (8.8 mg, tR=26.23 min)、化合物5 (5.46 mg, tR=32.76 min)、化合物6 (1.92 mg, tR=46.56 min)。Fr.4经Toyopearl HW-40柱色谱, 甲醇洗脱, 得Fr.4.1~Fr.4.4。Fr.4.2经半制备高效液相, 流动相(甲醇-水=70∶30, 3 mL·min-1, λ=210 nm)等度洗脱, 得到化合物1 (9.67 mg, tR=28.57 min)、化合物3 (7.68 mg, tR=33.37 min)、化合物4 (7.39 mg, tR=42.30 min)。
70%乙醇部位经硅胶柱, 用二氯甲烷-甲醇(100∶1、50∶1、20∶1、10∶1)洗脱, 得到Fr.1~Fr.7。Fr.2经硅胶柱, 分别用石油醚-丙酮(20∶1)和二氯甲烷-甲醇(200∶1、100∶1)洗脱, 得到Fr.2.1~Fr.2.6。Fr.2.3经半制备高效液相, 流动相(甲醇-水=75∶25, 3 mL·min-1, λ=210 nm)等度洗脱, 得到化合物7 (4.53 mg, tR=32.29 min)。Fr.2.6经半制备高效液相, 流动相(甲醇-水=60∶40, 3 mL·min-1, λ=210 nm)等度洗脱, 得到化合物8 (6.48 mg, tR=42.10 min)。

取对数期生长的A549细胞, 以每毫升2×104个接种于96孔板中, 24 h后, 实验分为正常对照组和化合物1~8给药组。给药组每孔加入20μmol·L-1的化合物, 每孔200μL, 培养48 h, 采用MTT方法检测各组细胞活力。
