Four pyrazines were isolated from the n-butanol fraction of Hypecoum erectum L. by using various chromatographic methods, including MCI gel, ODS, silica gel and semi-preparative HPLC. The structures of the isolated compounds were identified as hyperectpyrazin A (1), 1′S-(6-methylpyrazin-2-yl)-ethane-1′, 2′-diol (2), 2-hydroxymethyl-6-methylpyrazin (3) and pyrazine-2-carboxylic acid (4) by spectroscopy methods (1D NMR, 2D NMR, UV, IR, MS, etc.). The absolute configuration of compound 2 was determined by using the Mo2(OAc)4 induced CD analysis for the first time. Compound 1 was a new compound, compounds 2-4 were isolated from H. erectum for the first time. Compounds 1-4 were evaluated for their inhibition against acetylcholinesterase and nitric oxide generation induced by lipopolysaccharide-RAW264.7 macrophage cells. At a concentration of 50 μmol·L-1, compounds 2 and 4 displayed inhibitory effects on acetylcholinesterase with the inhibition rates of 44.40% and 43.99%, respectively.

采用硅胶、ODS、MCI等柱色谱以及半制备液相等现代色谱分离技术对角茴香(Hypecoum erectum L.) 正丁醇萃取部位的化学成分进行分离纯化, 得到4个吡嗪类生物碱。运用现代波谱学方法(1D NMR、2D NMR、UV、IR、MS等) 鉴定结构分别为hyperectpyrazin A (1)、1′S-(6-methylpyrazin-2-yl)-ethane-1′, 2′-diol (2)、2-hydroxymethyl-6-methylpyrazin (3) 和pyrazine-2-carboxylic acid (4)。化合物1为1个新的吡嗪类生物碱, 化合物2~4为首次从角茴香中分离得到, 并首次通过Mo2(OAc)4诱导的CD谱确定了化合物2的绝对构型。采用乙酰胆碱酯酶抑制模型和脂多糖诱导的RAW264.7巨噬细胞炎症模型, 对化合物1~4进行体外活性评价, 其中化合物24表现出一定的乙酰胆碱酯酶抑制活性, 在终浓度为50 μmol·L-1时对乙酰胆碱酯酶的抑制率分别为44.40%和43.99%。

采用硅胶、ODS、MCI等柱色谱以及半制备液相等现代色谱分离技术对角茴香(Hypecoum erectum L.) 正丁醇萃取部位的化学成分进行分离纯化, 得到4个吡嗪类生物碱。运用现代波谱学方法(1D NMR、2D NMR、UV、IR、MS等) 鉴定结构分别为hyperectpyrazin A (1)、1′S-(6-methylpyrazin-2-yl)-ethane-1′, 2′-diol (2)、2-hydroxymethyl-6-methylpyrazin (3) 和pyrazine-2-carboxylic acid (4)。化合物1为1个新的吡嗪类生物碱, 化合物2~4为首次从角茴香中分离得到, 并首次通过Mo2(OAc)4诱导的CD谱确定了化合物2的绝对构型。采用乙酰胆碱酯酶抑制模型和脂多糖诱导的RAW264.7巨噬细胞炎症模型, 对化合物1~4进行体外活性评价, 其中化合物24表现出一定的乙酰胆碱酯酶抑制活性, 在终浓度为50 μmol·L-1时对乙酰胆碱酯酶的抑制率分别为44.40%和43.99%。

Four pyrazines were isolated from the n-butanol fraction of Hypecoum erectum L. by using various chromatographic methods, including MCI gel, ODS, silica gel and semi-preparative HPLC. The structures of the isolated compounds were identified as hyperectpyrazin A (1), 1′S-(6-methylpyrazin-2-yl)-ethane-1′, 2′-diol (2), 2-hydroxymethyl-6-methylpyrazin (3) and pyrazine-2-carboxylic acid (4) by spectroscopy methods (1D NMR, 2D NMR, UV, IR, MS, etc.). The absolute configuration of compound 2 was determined by using the Mo2(OAc)4 induced CD analysis for the first time. Compound 1 was a new compound, compounds 2-4 were isolated from H. erectum for the first time. Compounds 1-4 were evaluated for their inhibition against acetylcholinesterase and nitric oxide generation induced by lipopolysaccharide-RAW264.7 macrophage cells. At a concentration of 50 μmol·L-1, compounds 2 and 4 displayed inhibitory effects on acetylcholinesterase with the inhibition rates of 44.40% and 43.99%, respectively.

角茴香为罂粟科(Papaveraceae) 角茴香属(Hypecoum) 植物角茴香(Hypecoum erectum L.) 的全草, 别名咽喉草、麦黄草、黄花草等, 在我国华北、西北和东北等地大量分布[1, 2]。据《河南中草药手册》中记载: “角茴香, 味苦, 性寒, 具有清热解毒, 镇咳止痛的功效”, 主治咽喉肿痛、咳嗽、气管炎、肝炎、细菌性痢疾和目赤肿痛等症。现代药理活性研究表明角茴香主要表现为抗炎、保肝、退热和镇痛等作用[3]。化学成分研究表明角茴香主要含有生物碱类成分, 尤以螺苄基异喹啉类生物碱研究最多[4-6]。本课题组前期从角茴香脂溶性较强的二氯甲烷萃取部位分离得到2个骨架新颖的生物碱化合物[7]。为进一步丰富角茴香不同萃取部位的药效物质基础研究, 本实验从角茴香正丁醇萃取部位分离得到4个吡嗪类生物碱(图 1), 包括1个新的吡嗪类生物碱hyperectpyrazin A (1), 以及3个首次从角茴香中分离得到的吡嗪类生物碱1′S-(6-methylpyrazin-2-yl)-ethane-1′, 2′-diol (2)、2-hydroxymethyl-6-methylpyrazin (3) 和pyrazine-2-carboxylic acid (4)。
化合物1为淡黄色油状, HR-ESI-MS给出准分子离子峰m/z 214.095 2 [M+Na]+, 计算值为214.095 0 [M+Na]+, 推测分子式为C10H13N3O, 不饱和度为6。化合物1的IR光谱数据显示其在1 685、1 423 cm-1存在较强吸收, 表明化合物1的结构中可能存在吡咯烷酮片段[8]。化合物11H NMR (500 MHz, CD3OD) 数据(表 1) 显示存在2个芳香氢信号[δH 8.41 (1H, s, H-5), 8.35 (1H, s, H-3)], 4个亚甲基氢信号[δH 4.59 (2H, s, H-8), 3.50 (2H, t, J = 7.1, H-5′), 2.45(2H, t, J = 8.1, H-3′), 2.10 (2H, m, H-4′)] 和1个甲基氢信号[δH 2.55 (3H, s, H-7)]。其13C NMR (125 MHz, CD3OD) 谱共显示10个碳信号, 进一步结合DEPT谱发现化合物1中存在1个羰基碳信号[δC 178.1 (C-2′)], 4个芳香碳信号[δC 155.3 (C-6), 152.5 (C-2), 144.2 (C-5), 141.4 (C-3)], 4个亚甲基碳信号[δC 49.2 (C-5′), 46.9 (C-8), 31.6 (C-3′), 18.9 (C-4′)] 和1个甲基碳信号[δC 21.4 (C-7)]。通过对比化合物1和已知化合物3的1D和2D NMR数据, 推断化合物1存在与化合物3相同的吡嗪母核。
为了进一步确认化合物1的结构, 对化合物1进行了2D NMR测试。在1H-1H COSY谱中(图 2) 显示存在H-5′/H-4′、H-4′/H-3′的相关, 在HMBC谱中(图 2), 观察到H-3′/C-2′、H-4′/C-2′、H-5′/C-2′和C-8、H-8/C-2′, C-5′, C-2和C-3的相关信号, 证明化合物1中存在吡咯烷酮结构片段, 并通过亚甲基(C-8) 与吡嗪母核的C-2位相连。此外, 还观察到在HMBC谱中(图 2) 存在H-7/C-6和C-5、H-3/C-5的相关信号, 表明甲基连接在C-6位。综上所述, 化合物1的结构被确定为1′-((6-methylpyrazin-2-yl)methyl)-pyrrolidin-2′-one, 命名为hyperectpyrazin A。
化合物2为淡黄色油状, 其1D NMR数据在低场区与化合物13类似, 表明化合物2同样存在吡嗪母核。与化合物1相比, 其主要区别为化合物2的C-2位被乙二醇结构片段取代。其1H-1H COSY谱中(图 2) 存在H-1′/H-2′、H-1′/1′-OH、H-2′/2′-OH的相关, 在HMBC谱中(图 2), 观察到1′-OH/C-1′和C-2′、2′-OH/C-1′和C-2′的相关, 示为乙二醇结构片段的相关信号。此外, 在HMBC谱中(图 2) 还存在1′-OH/C-2、H-1′/C-2和C-3的相关信号, 证明化合物2中乙二醇结构片段连接在吡嗪环的C-2位。在HMBC谱中还观察到存在H-7/C-6和C-5、H-3/C-5的相关信号, 表明甲基连接在C-6位。化合物2的结构虽然可以在SCIFinder数据库中检索到[9], 但并未见有相关文献对其NMR数据进行详细归属, 因此本文对化合物2的NMR数据进行了归属(表 1) 并表征了其绝对构型。
化合物2的绝对构型是通过Mo2(OAc)4诱导CD实验确定的, 化合物2经Mo2(OAc)4试剂诱导的CD谱在315 nm处显示的正Cotton效应(图 3), 根据其(Mo-O)-C-C-(O-Mo) 二面角扭角的符号与特定的310 nm附近谱带IV的符号相关联的螺旋规则[10], 最终将C-1′位的绝对构型指认为S。综上所述, 化合物2被鉴定为1′S-(6-methylpyrazin-2-yl)-ethane-1′, 2′-diol。
采用乙酰胆碱酯酶抑制模型, 对化合物1~4进行乙酰胆碱酯酶抑制活性评价, 在终浓度为50 μmol·L-1时, 化合物24表现出一定的乙酰胆碱酯酶抑制活性, 对乙酰胆碱酯酶的抑制率分别为44.40%和43.99%; 化合物13对乙酰胆碱酯酶的抑制率分别为28.04%和18.20%。
采用脂多糖诱导RAW264.7巨噬细胞炎症模型, 对化合物1~4进行抗炎活性评价, 在终浓度为50 μmol·L-1时, 化合物1~4的一氧化氮生成抑制率分别为(3.99 ± 2.09)%、(-0.29 ± 1.85)%、(4.25 ± 2.98)%和(4.99 ± 2.00)%, 结果表明化合物1~4均无抗炎活性。

角茴香药材于2019年9月购自河南省郑州市郑州药材市场, 经河南中医药大学药学院陈随清教授鉴定为罂粟科角茴香属植物角茴香Hypecoum erectum L.的干燥全草。凭证样品(CH20190912J) 存放于河南中医药大学中药化学研究室。
角茴香干燥全草50.00 kg, 95%乙醇回流提取2次, 滤过, 合并提取液, 减压浓缩, 得浸膏6.00 kg。浸膏加10倍量2%盐酸水溶液溶解, 过滤得到酸水溶液和不溶物。酸水溶液以石油醚萃取得到石油醚层和酸水层。酸水层加氨水调至pH 9~10得到碱水溶液。碱水溶液依次用氯仿和正丁醇萃取得到氯仿萃取部位约330.00 g和正丁醇萃取部位约280.00 g。正丁醇萃取部位经硅胶柱色谱分离, 依次用纯二氯甲烷、二氯甲烷-甲醇(50∶1、20∶1、10∶1、5∶1、2∶1、1∶1) 和纯甲醇进行梯度洗脱, 并经硅胶薄层检识, 合并相同馏分得到8个组分(Fr.1~Fr.8)。Fr.3 (12.25 g) 经中压ODS柱色谱分离依次用甲醇-水(10%→100%) 梯度洗脱并经硅胶薄层检识, 合并相同馏分得到10个组分(Fr.3-B1~Fr.3-B10)。Fr.3-B6组分经硅胶柱色谱分离, 依次用石油醚-乙酸乙酯(1∶0、20∶1、10∶1、5∶1、2∶1、1∶1、0∶1) 进行梯度洗脱, 并经硅胶薄层检识, 合并相同馏分得到9个组分(Fr.3-B6-b1~Fr.3-B6-b9)。Fr.3-B6-b9组分经制备型HPLC制备(乙腈∶水= 12∶88) 得到化合物1 (tR = 35.50 min; 20.01 mg)。Fr.3-B2组分经制备型HPLC制备(甲醇∶水= 4∶96) 得到化合物4 (tR = 19.85 min; 6.00 mg)。Fr.3-B3组分经制备型HPLC制备(甲醇∶水= 5∶95) 得到化合物3 (tR = 19.23 min; 10.05 mg)。Fr.4 (54.53 g) 经中压MCI柱色谱分离依次用甲醇-水(10%→100%) 梯度洗脱并经硅胶薄层检识, 合并相同馏分得到5个组分(Fr.4-C1~Fr.4-C5)。Fr.4-C1组分经硅胶柱色谱分离, 依次用二氯甲烷-甲醇(1∶0、30∶1、20∶1、10∶1、5∶1、2∶1、1∶1、0∶1) 进行梯度洗脱, 并经硅胶薄层检识, 合并相同馏分得到9个组分(Fr.4-C1-c1~Fr.4-C1-c9)。Fr.4-C1-c9组分经制备型HPLC制备(甲醇∶水= 5∶95) 得到化合物2 (tR = 36.12 min; 18.13 mg)。
化合物1  淡黄色油状, 易溶于甲醇; UV (MeOH) λmax (log ε) 201 (3.49), 270 (3.18) nm; IR νmax 2 945, 1 685, 1 535, 1 494, 1 464, 1 423 cm-1; 1H NMR (CD3OD, 500 MHz) 和13C NMR (CD3OD, 125 MHz) 数据见表 1, HR-ESI-MS m/z 214.095 2 [M+Na]+ (计算值为C10H13N3ONa+, 214.095 0)。
化合物2  淡黄色油状, 易溶于甲醇; UV (MeOH) λmax (log ε) 203 (3.77), 271 (3.78) nm; IR νmax 3 336, 2 944, 1 537, 1 428, 1 162, 1 106, 1 027 cm-1; 1H NMR (DMSO-d6, 500 MHz) 和13C NMR (DMSO-d6, 125 MHz) 数据见表 1, HR-ESI-MS m/z 177.063 3 [M+Na]+ (计算值为C7H10N2O2Na+, 177.063 4)。
化合物3  黄色油状, 易溶于甲醇, ESI-MS m/z 125 [M+H]+, 分子式为C6H8N2O。其1H NMR (CD3OD, 500 MHz) δH 8.51 (1H, s, H-3), 8.39 (1H, s, H-5), 4.72 (2H, s, H-1′), 2.53 (3H, s, H-7); 13C NMR (CD3OD, 125 MHz) δC 156.9 (C-2), 154.2 (C-6), 143.9 (C-5), 140.4 (C-3), 64.0 (C-1′), 22.2 (C-7)。以上数据与文献[11]报道基本一致, 并进一步经HMBC图谱证实(图 2), 确定化合物为2-hydroxymethyl-6-methylpyrazin。
化合物4  无色透明晶体, 易溶于甲醇, ESI-MS m/z 125 [M+H]+, 分子式为C5H4N2O2。其1H NMR (DMSO-d6, 500 MHz) δH 9.18 (1H, d, J = 1.5 Hz, H-3), 8.85 (1H, s, H-5), 8.72 (1H, dd, J = 2.5, 1.5 Hz, H-6); 13C NMR (DMSO-d6, 125 MHz) δC 165.1 (C-1′), 147.4 (C-5), 145.1 (C-3), 143.6 (C-6), 143.4 (C-2)。以上数据与文献[12]报道基本一致, 确定化合物为pyrazine-2-carboxylic acid。
将0.9 mg化合物2加入到1.8 mL 0.7 mg·mL-1 [Mo2(OAc)4] 的DMSO溶液中。混合后立即测定并记录第一张图谱, 并监测其演变直至稳定(30~60 min), 以得到稳定的诱导CD图谱。根据Mo2(OAc)4诱导CD的螺旋规则分析得到的诱导CD图谱, 最终确定化合物2的构型。
采用Ellman等[13]改进的分光光度法测定了化合物的乙酰胆碱酯酶抑制活性。将待测的样品加二甲基亚砜溶解后加入pH 8的0.1 mol·L-1磷酸缓冲液, 配制成50 μmol·L-1的待测溶液。阴性对照采用二甲基亚砜, 阳性对照采用他克林, 空白对照采用磷酸缓冲液。测定时首先在96孔板上加入用待测样品10 μL, 再依次加入110 μL磷酸缓冲液、20 μL 2.5 mmol·L-1的5, 5-二硫代双(2-硝基苯甲酸)、40 μL乙酰胆碱酯酶溶液, 振荡混匀, 37 ℃预孵10 min, 然后加入10 mmol·L-1碘化硫代乙酰胆碱20 μL, 37 ℃反应10 min, 酶标仪测定405 nm吸收度(OD) 值并计算样品对乙酰胆碱酯酶的抑制活性, 计算公式: 抑制率(%) = [(OD阴性对照 - OD反应本底) - (OD待测样品 - OD反应本底)] / (OD阴性对照 - OD反应本底) × 100%。
体外抗炎活性筛选采用小鼠单核巨噬细胞RAW 264.7模型, 将RAW264.7细胞接种至96孔板, 用1 μg·mL-1脂多糖进行诱导刺激, 同时加入待测样品(终浓度50 μmol·L-1), 设置不含药物组和L-NMMA阳性药物组做对照。细胞过夜培养后取培养基检测一氧化氮生成, 在570 nm处测定OD值, 并计算样品对一氧化氮生成的抑制活性。在剩余培养基中加入MTS进行细胞存活率检测, 排除化合物对细胞的毒性影响。抑制率(%) = (OD非药物处理组 - OD待测样品组) / OD非药物处理组 × 100%。
