Three 2,3-diketoquinoxaline alkaloids were isolated from Heterosmilax yunnanensis Gagnep. Their structures were determined through 1D and 2D NMR, HR-ESI-MS, UV, and IR as 1-[5′-(3″-hydroxy-3″-methyl) glutaryl] ribityl-2,3-diketo-1,2,3,4-tetrahydro-6,7-dimethylquinoxaline (1), 1-[2′-(3″-hydroxy-3″-methyl) glutaryl]ribityl-2,3-diketo-1,2,3,4-tetrahydro-6,7-dimethylquinoxaline (2), and 1-ribityl-2,3-diketo-1,2,3,4-tetrahydro-6,7-dimethylquinoxaline (3). Compounds 1 and 2 are novel compounds, and 3 was isolated from H. yunnanensis for the first time. The hepatoprotective activity of these three compounds was evaluated, with compound 3 showing promising hepatoprotective activity.

从短柱肖菝葜(Heterosmilax yunnanensis Gagnep.) 中分离得到3个2,3-二酮喹喔啉类生物碱, 通过一维和二维核磁共振、高分辨质谱、紫外光谱和红外光谱确定其化学结构, 分别鉴定为1-[5′-(3″-羟基-3″-甲基) 戊二酸单酰基] 核糖醇基-2,3-二酮-1,2,3,4-四氢-6,7-二甲基喹喔啉(1)、1-[2′-(3″-羟基-3″-甲基) 戊二酸单酰基] 核糖醇基-2,3-二酮-1,2,3,4-四氢-6,7-二甲基喹喔啉(2) 和1-核糖醇基-2,3-二酮-1,2,3,4-四氢-6,7-二甲基喹喔啉(3)。其中化合物12是新化合物, 化合物3是首次从短柱肖菝葜中分离得到, 通过肝保护活性筛选, 发现化合物3具有良好的肝保护活性。

从短柱肖菝葜(Heterosmilax yunnanensis Gagnep.) 中分离得到3个2,3-二酮喹喔啉类生物碱, 通过一维和二维核磁共振、高分辨质谱、紫外光谱和红外光谱确定其化学结构, 分别鉴定为1-[5′-(3″-羟基-3″-甲基) 戊二酸单酰基] 核糖醇基-2,3-二酮-1,2,3,4-四氢-6,7-二甲基喹喔啉(1)、1-[2′-(3″-羟基-3″-甲基) 戊二酸单酰基] 核糖醇基-2,3-二酮-1,2,3,4-四氢-6,7-二甲基喹喔啉(2) 和1-核糖醇基-2,3-二酮-1,2,3,4-四氢-6,7-二甲基喹喔啉(3)。其中化合物12是新化合物, 化合物3是首次从短柱肖菝葜中分离得到, 通过肝保护活性筛选, 发现化合物3具有良好的肝保护活性。

Three 2,3-diketoquinoxaline alkaloids were isolated from Heterosmilax yunnanensis Gagnep. Their structures were determined through 1D and 2D NMR, HR-ESI-MS, UV, and IR as 1-[5′-(3″-hydroxy-3″-methyl) glutaryl] ribityl-2,3-diketo-1,2,3,4-tetrahydro-6,7-dimethylquinoxaline (1), 1-[2′-(3″-hydroxy-3″-methyl) glutaryl]ribityl-2,3-diketo-1,2,3,4-tetrahydro-6,7-dimethylquinoxaline (2), and 1-ribityl-2,3-diketo-1,2,3,4-tetrahydro-6,7-dimethylquinoxaline (3). Compounds 1 and 2 are novel compounds, and 3 was isolated from H. yunnanensis for the first time. The hepatoprotective activity of these three compounds was evaluated, with compound 3 showing promising hepatoprotective activity.

白土苓是百合科肖菝葜属植物华肖菝葜(Heterosmilax chinensis Wang.)、短柱肖菝葜(H. yunnanensis Gagnep.) 和肖菝葜(H. japonica Kunth.) 的根茎, 主要分布于东亚的热带和亚热带地区, 具有清热、除湿和解毒的作用[1], 但在四川、湖北、山东、宁夏等地, 常与土茯苓(Smilax glabra Roxb.) 混淆使用。本文研究的白土苓是短柱肖菝葜(H. yunnanensis) 的干燥根茎, 是复方苦参注射液中的唯一臣药, 临床用于抗肿瘤和癌性疼痛治疗[2]。药理学研究表明, 短柱肖菝葜的粗提取物具有止痛和止血作用[3]。但目前对于短柱肖菝葜的化学成分研究较少, 只报道了约十余种化合物, 包括大泽明素[4]、芒果苷、丁香酸葡萄糖苷和β-谷甾醇等[5-8], 因此, 本研究对短柱肖菝葜进行系统的化学成分研究, 从中得到3个2,3-二酮喹喔啉类的生物碱结构(图 1), 其中化合物12是新化合物, 化合物3是首次从短柱肖菝葜中分离得到, 同时采用扑热息痛引起体外肝细胞损伤的保护模型对这3个化合物进行肝保护活性测定, 结果表明化合物3在10 μmol·L-1下具有良好的肝保护活性。
化合物1为白色无定形粉末; 难溶于水和甲醇, 根据高分辨质谱的准分子离子峰m/z: 469.181 8 [M+H]+ (计算值469.181 7, C21H29O10N2) 和13C NMR数据确定其分子式是C21H28O10N2, 不饱和度是9, 红外光谱显示该化合物具有羟基(3 370 cm-1)、苯环(1 627、1 514和1 449 cm-1) 和羰基(1 694 cm-1); 1H NMR谱(表 1) 显示两个芳香氢信号[δH 7.29 (1H, s) 和6.91 (1H, s)], 3个甲基氢信号[δH 2.22 (3H, s), 2.18 (3H, s) 和1.25 (3H, s)], 结合HSQC谱, 还观察到两个连氧(氮) 亚甲基氢信号[δH 4.46 (1H, dd, J = 10.0, 14.0 Hz) 与4.03 (1H, m)] 和[δH 4.22 (1H, dd, J = 2.5, 12.0 Hz) 与3.99 (1H, dd, J = 7.0, 12.0 Hz)], 两个亚甲基氢信号[δH 2.61 (1H, d, J = 14.0 Hz) 和2.54 (1H, d, J = 14.0 Hz)] 和[δH 2.43 (2H, s)], 3个连氧次甲基氢信号[δH 4.04 (1H, m), 3.79 (1H, td, J = 2.5, 7.0 Hz) 和3.58 (1H, dd, J = 5.0, 7.0 Hz)]; 13C NMR (表 1) 结合HSQC谱观察到21个碳信号, 包括8个芳香族的碳信号(δC 156.2, 154.2, 131.8, 131.5, 125.4, 123.8, 117.0和116.5), 剩余13个碳信号, 包括3个甲基碳信号(δC 28.0, 19.8和19.2), 两个羰基碳信号(δC 173.7和171.3), 8个脂肪族碳信号(δC 73.9, 70.0, 69.5, 68.3, 66.6, 46.2×2和45.2), 结合HMBC谱中(图 2) H-1′与C-2 (δC 156.2) 和C-9 (δC 125.4) 相关, H-3′与C-1′ (δC 45.2) 相关, H-4′与C-2′ (δC 68.3) 相关, H-5′与C-3′ (δC 73.9) 相关, H-11与C-7 (δC 131.8) 相关, H-12与C-6 (δC 131.5) 相关, 与文献[9-12]比对, 发现与1-核糖醇基-2,3-二酮-1,2,3,4-四氢-6,7-二甲基喹喔啉(3) 数据相似, 剩余信号与3-羟基-3-甲基戊二酸单酰基(HMG) 片段数据吻合[13], 且H-5′与C-1″ (δC 171.3) 相关。因此, 1-核糖醇基-2,3-二酮-1,2,3,4-四氢-6,7-二甲基喹喔啉与HMG通过C-5′-O-C-1″酯化连接。
化合物1的相对构型通过耦合常数J确定[14], H-2′和H-3′之间的小耦合常数(J = 5.0 Hz) 与H-3′和H-4′之间大耦合常数(J = 7.0 Hz), 表明2′和3′, 3′和4′之间是赤式构象。通过碱水解酯键, 得到化合物1中的1-核糖醇基-2,3-二酮-1,2,3,4-四氢-6,7-二甲基喹喔啉片段, 其比旋光度符号也为负, 因此2′, 3′, 4′绝对构型分别为S, S, R[12]。HMG中C-3″的绝对构型确定为S, 因为自然界中的HMG酯均由(S)-HMG-CoA形成[13, 15, 16]。综上所述, 该化合物的结构被确定为1-[5′-(3″-羟基-3″-甲基) 戊二酸单酰基] 核糖醇基-2,3-二酮-1,2,3,4-四氢-6,7-二甲基喹喔啉。
化合物2为白色无定形粉末; 难溶于水, 可溶于甲醇, 高分辨质谱的准分子离子峰m/z为469.181 8 [M+H]+ (计算值469.181 7, C21H29O10N2), 确定其分子式是C21H28O10N2, 不饱和度是9, 红外光谱显示该化合物含有羟基、苯环和羰基; 该化合物与化合物1的分子式、紫外、红外等信息均一致, 仔细观察其核磁共振数据, 发现HMG的连接位点不同, 根据HMBC谱(图 2) 中H-2′与C-1″ (δC 172.1) 相关, 因此确定1-核糖醇基-2,3-二酮- 1,2,3,4-四氢-6,7-二甲基喹喔啉与HMG通过C-2′-O-C-1″酯化连接, 绝对构型确定方法和化合物1相同, 因此, 该化合物结构确定为1-[2′-(3″-羟基-3″-甲基) 戊二酸单酰基] 核糖醇基-2,3-二酮-1,2,3,4-四氢-6,7-二甲基喹喔啉。
将化合物1~3在10 μmol·L-1浓度作用HepG2细胞24 h, 结果显示细胞存活率均大于90% (表 2), 无明显毒性, 因此以10 μmol·L-1无毒浓度用于后续实验。采用MTT方法检测化合物1~3对APAP损伤的HepG2细胞保护的作用, 结果表明(表 3), 化合物3在10 μmol·L-1的浓度下对细胞的存活率明显提高, 且比阳性对照的作用更好, 表明化合物3具有良好的肝保护活性。
JASCO P-2000型旋光仪、JASCO V-650型紫外光谱仪(JASCO, USA); Nicolet is50 FT-IR ATR红外光谱仪(Thermo Scientific, USA); Bruker AVANCE Ⅲ-500光谱仪(Bruker Biospin, USA); Agilent 6520 HPLC-Q-TOF质谱仪(Agilent Technologies, Germany); Agilent 1260 series高效液相, Apollo C18 column (250 mm × 4.6 mm, 5 μm; Alltech Corp., USA); 溶剂为色谱纯(Fisher Scientific); Shimadzu LC-20AT制备型高效液相, SPD-10A紫外检测器, YMC-Pack ODS-A色谱柱(250 mm × 10 mm, 5 μm), 溶剂为色谱纯(北京市通广精细化工公司); EYELA N-1100D-WD型旋转蒸发仪、EYELA FDU-1100型冷冻干燥机(日本东京理化株式会社); SQP型十万分之一分析天平(德国Sartorius公司); 大孔树脂(Diaion HP-20, Mitsubishi Chemical Corp., Japan); RP-C18 (50 μmol·L-1, YMC Corp., Japan); Sephadex LH-20型凝胶(Pharmacia Fine Chemicals, Sweden); 聚酰胺(60~80目, RHAWN, China); 色谱纯试剂: HPLC级的甲醇和乙腈来自Fisher公司, 制备型甲醇和乙腈购自北京市通广精细化工公司; 分析纯试剂: 甲醇、乙腈、乙醇购自北京市通广精细化工公司。实验用药材由山西振东制药股份有限公司购买, 产地: 贵州安顺, 经中国医学科学院药用植物研究所郭宝林研究员鉴定为百合科肖菝葜属植物短柱肖菝葜(Heterosmilax yunnanensis Gagnep.), 植物标本保藏号: ID-S-3000。
短柱肖菝葜的干燥块茎100 kg, 粉碎后依次用95%、85%、75%乙醇和水回流提取3次, 每次2 h, 过滤后收集上清液, 合并3次的提取液, 减压回收溶剂得到乙醇水总提取液约24.5 kg。将总提取液中的乙醇除去, 然后用等体积的乙酸乙酯萃取3次, 减压浓缩乙酸乙酯部位后得到520 g浸膏, 又对萃取剩下的水部位用85%的醇沉除去大分子的糖、蛋白等物质, 浓缩后得到浸膏约6 142 g。对该浸膏用3 L水溶解, 分两次进行大孔树脂柱色谱分离(d = 14.5 cm, h = 82 cm, 柱体积13.5 L), 依次用0、5%、15%、30%、50%、70%、95%的乙醇洗脱, 获得7个亚部位依次命名为BTL-A~BTL-G。
将BTL-D部分(共约110 g) 用250 mL水溶解, 湿法上样, 进行凝胶柱色谱分离(d = 11.7 cm, h = 67.0 cm), 依次用0、5%、10%、15%、20%、25%、30%、40%、50%、60%、70%、95%的乙醇洗脱, 浓缩得Fra.D-1~Fra.D-343共343瓶流分。Fra.D-9和Fra.D-10合并后进行聚酰胺柱色谱分离(乙醇∶水= 0~95%梯度洗脱), 得JXA4.1~JXA4.46共46个组分; 对JXA4.46进行C18反相半制备液相色谱柱纯化(MeCN/H2O/AcOH, 17∶83∶0.1, v/v/v) 得到化合物1 (3.1 mg) 和2 (1.8 mg)。对JXA4.8~JXA4.15合并后进行Flash反相ODS加压柱色谱分离(甲醇∶水= 0~100%梯度洗脱), 记为F9柱, 得F9.1~F9.7共7个流分; 对F9.3用C18反相半制备液相色谱柱纯化(MeCN/H2O/AcOH, 13∶87∶0.1, v/v/v) 得到化合物3 (2.3 mg)。
化合物1: 白色无定形粉末; 比旋光度[α]$ {}_{\mathrm{D}}^{20} $ 4.8 (c 0.1, MeOH); UV (MeOH) λmax (log ε) 355 sh (3.36), 337 sh (3.69), 322 (3.76), 309 sh (3.66), 241 sh (3.73), 235 sh (3.79), 215 (4.38) nm; IR νmax 3 370, 2 925, 1 694, 1 627, 1 601, 1 514, 1 449, 1 398, 1 318, 1 271, 1 201, 1 078, 1 021, 953, 901, 873, 706, 665 cm-1; 1H和13C NMR数据见表 1; HR-ESI-MS m/z 469.181 8 [M+H]+ (C21H29O10N2计算值469.181 7)。
化合物2: 白色无定形粉末; 比旋光度[α]$ {}_{\mathrm{D}}^{20} $ 4.0 (c 0.1, MeOH); UV (MeOH) λmax (log ε) 355 sh (3.33), 338 sh (3.64), 322 (3.70), 309 sh (3.61), 241 sh (3.68), 235 sh (3.74), 215 (4.32) nm; IR νmax 3 391, 2 924, 1 694, 1 627, 1 601, 1 515, 1 449, 1 398, 1 332, 1 271, 1 202, 1 088, 1 021, 953, 874, 707, 669 cm-1; 1H和13C NMR数据见表 1; HR-ESI-MS m/z 469.181 8 [M+H]+ (C21H29O10N2计算值469.181 7)。
化合物3: 白色无定形粉末; 比旋光度[α]$ {}_{\mathrm{D}}^{25} $ -11.2 (c 0.05, MeOH∶H2O = 1∶1); HR-ESI-MS m/z 325.139 5 [M+H]+ (C15H21O6N2计算值325.139 4); 1H NMR (500 MHz, DMSO-d6): δH 7.33 (1H, s, H-5), 6.92 (1H, s, H-8), 4.52 (1H, dd, J = 10.0, 14.0 Hz, H-1′a), 4.06 (1H, d, J = 10.5 Hz, H-2′), 3.99 (1H, d, J = 14.0 Hz, H-1′b), 3.61 (1H, d, J = 12.0 Hz, H-5′a), 3.56 (2H, overlap, H-3′, 4′), 3.42 (1H, dd, J = 5.5, 11.0 Hz, H-5′b), 2.22 (3H, s, H-11), 2.18 (3H, s, H-12); 13C NMR (125 MHz, DMSO-d6): δC 155.8 (C-2), 153.9 (C-3), 131.3 (C-7), 131.0 (C-6), 124.9 (C-9), 123.4 (C-10), 116.6 (C-8), 116.1 (C-5), 73.6 (C-4′), 72.8 (C-3′), 68.1 (C-2′), 63.5 (C-5′), 44.6 (C-1′), 19.4 (C-11), 18.8 (C-12)。通过与文献[11, 12]对比核磁数据和比旋光度, 确定该化合物为已知化合物1-核糖醇基-2,3-二酮-1,2,3,4-四氢-6,7-二甲基喹喔啉。
HepG2细胞接种于96孔细胞培养板中, 培养24 h后, 加入不同浓度的待测化合物, 同时设溶剂对照组, 每个药物浓度设3个平行孔。药物作用细胞24 h后, 弃去培养液, 每孔加入MTT (0.5 mg·mL-1) 液100 μL, 继续培养4 h, 弃去MTT液, 每孔加入DMSO 150 μL, 混和振荡器振荡, 在酶标仪570 nm波长处测定吸光度值。细胞存活率(%) = (给药细胞OD平均值/溶剂对照细胞OD平均值) × 100%。
HepG2细胞接种于96孔细胞培养板中, 培养24 h后, 加入待测化合物及扑热息痛(APAP, 终浓度8 mmol·L-1), 同时设阳性药物对照组(谷胱甘肽GSH)、溶剂空白对照组及模型组。继续作用细胞24 h。弃去培养液, 每孔加入MTT (0.5 mg·mL-1) 液100 μL, 继续培养4 h, 弃去MTT液, 每孔加入DMSO 150 μL, 混和振荡器振荡, 在酶标仪570 nm波长处测定吸光度值。细胞存活率(%) = (给药组OD平均值/溶剂对照组OD平均值) × 100%。
所有数据均以平均值± SD表示。两两组比较采用t-检验分析。P < 0.05或P < 0.01或P < 0.001的值被认为具有统计学意义, ns被认为不具有统计学意义。
